BACKGROUND: MicroRNA-21 (miR-21) is a regulator of osteoclastogenesis and a promoter of osteoclast differentiation, but its role in periodontitis remains unclear. OBJ ECTlVE: To investigate whether miR-21 is involved in bone destruction in periodontitis. METHODS: Real-time PCR was used to detect and analyze the differential expression of miR-21 in periodontitis samples. Using liposome transfection method, miR-21 mimics (up-regulating miR-21) or miR-21 inhibitor (down-regulating miR-21) was used to transfect osteoclasis. Expressions of miR-21 and bone destruction markers TRAP and CTSK were detected by real-time PCR. Cell counting kit-8 was used to detect the miR-21 effect on osteoclast proliferation. RESULTS AND CONCLUSION: (1) MiR-21 expression increased in periodontitis samples. (2) When miR-21 mimics was transfected into osteoclasis, miR-21, TRAP and CTSK mRNA expression increased; when miR-21 inhibitor was transfected into osteoclasis, miR-21, TRAP and CTSK mRNA expression decreased. (3) Transfection with miR-21 mimics promoted the proliferation of osteoclasis, while transfection with miR-21 inhibitor inhibited the proliferation of osteoclasis. Objective By establishing a special rats models of the multiple cerebral concussion (MCC) at different strike interval (that is, 24h, 48h and 72h), to observe the changes of space cognitive behavior by MWM (Morris water maze) after injury and to evaluate learning and memory function, so as to provide experimental data for clinic diagnosis and intervene. Methods (l) Groups: we selected 40 healthy adult SD rats, half male and half female, the average weight was 260-280g. The model rats were randomly divided into 3MCC 24 h, 3MCC 48 h, 3MCC 72 h (each group has 10 rats) after injury. And we set up a control group (10 rats). 2. The metal pendulum device was used at interval of 24 h, 48 h and 72 h to establish 3MCC 24 h, 3MCC 48 h and 3MCC 72 h injury group. (3) We evaluated the space cognitive function of the rats in each group through Morris water maze test. Results (l) In the underwater platform experiment, the escape latency (the time spent to find the platform) of the Control group appeared a rapid decline at first, and then tended to a steady state; the escape latency of each injury group all appeared a slow decline before a steady state. Compared with the Control group, the recovery after injury of the escape latency in 3MCC 24 h, 3MCC 48 h and 3MCC72h all had been prolonged significantly, and the 1-14 d of 3MCC 24 h and 3MCC 48 h group, the 12 days except the 7 d and 10 d of the 3MCC 72 h were statistically significant (P < 0.05). Compared with the 3MCC 24 h, other two 3MCC groups were no more delayed than the 3MCC 24 h, the 1-11d of the 3MCC 48 h and 3MCC 72 h were statistically significant (P < 0.05). (2) In the above water platform experiment, compared with the control group, the three 3MCC groups had been prolonged significantly, and the differences were statistically significant (P < 0.05), 3. In the no platform experiment, compared with the control group, the rats of the injury groups stayed less time in the first quadrant, and from light to heavy followed as 3MCC 72 h, 3MCC 48 h and 3MCC24h group. And the different of 3MCC 24 h and 3MCC 48 h groups were statistically significantly (P < 0.05). Conclusions (l) All 3MCC injury groups at different interval have obvious damage on spatial cognitive function and can not recover 14 days after injury. Objective To establish a rat model of the Pure Cerebral Concussion (PCC) and Multiple Cerebral Concussion (3MCC), so as to observe and make comparison of the reactions and changes in microglia in the saeptum coronary section of the early injured rats. Methods (l) Rats grouping: 30 healthy adult SD rats with weight of 260 ± 20g, were divided randomly into normal group, PCC model group and 3MCC model group. The model groups were regrouped into 1d group and 4d group for observation. (2) The Metal pendulum closed brain injury blow device was used to set up PCC model and MCC model in order to replicate 12 rats of PCC model and MCC model respectively. (3) The immunohistochemical S - P method was adopted to dye the CD 11b antibodies. After that, we observed the expressions of rats' CD11b in FrPa, cc, CPu and MS-LV in the septal coronary denomination parietal lobe. Then we measured and analyzed the reactant of the CD11b through Image Analysis Technology. Results (l) Under normal condition, the expression of the CD11b was rare; (2) The expression of CD11b in different brain areas which were partitioned in PCC model and MCC model increased gradually after injury. And compared with the normal group, they were significantly different (P<0.05). (3) The expression increase tendency of CD11b in 1d and 4d of MCC group after injure were higher than the 1d and 4d in PCC group. Conclusions (1) MG in the partition surface can be activated in the early injure of PCC and MCC, suggesting that MG is involved in the pathological changes of PCC and MCC after injury; (2) The expression of CD11b increases regularly with the exacerbation of injury severity.
