The three-point bending fatigue method was used to determine the S-N fatigue curves of smooth specimens and straight-notched specimens to study the fatigue properties of Stellite12 cobalt-based alloy, and the fatigue fracture processes of the cobalt-based alloy were investigated by the observation of fracture morphology.The results show that the fatigue limit of the smooth specimen is 545 MPa, which is about 25.4% of the original bending strength of 1552 MPa. The fatigue limit of the straight-notched specimen is about 101 MPa, which is about 19.1% of static bending strength of 517.6 MPa. The fatigue sensitivity of smooth specimens and straight-notched specimens is 397 and 31, respectively. In addition, it is found that the fatigue cracks can initiate in the carbides aggregated regions in the near surface. Surface defects can also induce the initiation of fatigue cracks. The transgranular fracture of the carbides dominates in the crack propagation. The cobalt base metal presents a lot of tearing ridges under the condition of stress ratio R=0.1, but it also shows a certain degree brittle fracture mode, so the fatigue crack propagation mode is a mixed mode of true fatigue and static fatigue.

The wetting of TA2 pure Ti by two industrial grade Al alloys (i.e.,6061 Al and 4043 Al alloys) was studied by using the modified sessile drop method at 600,650,700℃ under high vacuum. The results show Al/Ti system is a typical reactive wetting, the small amount of alloying element Si in the Al alloys can cause significant enrichment at liquid/solid interface and satisfy thermodynamic condition;the formation of the Si-rich phase (Ti7Al5Si12) strangling material exchange at triple line zone; Ti7Al5Si12 decomposition and Al3Ti formation can remove the oxide film and promote wetting;the spreading dynamics can be described by reaction product control model, further the whole wetting behavior can be divided into two stages:the first stage for the nonlinear spreading and the second stage forthe linear spreading;the activation energies which are 56kJ/mol, 47kJ/mol for nonlinear stages of 6061 Al and 4043 Al alloys, and 112kJ/mol for linear stage of 6061 Al alloys, respectively,Ti7Al5Si12 decomposition is corresponding to the nonlinear spreading.

The wetting behavior and interfacial microstructures of molten magnesium AZ61 alloy on the surface of two different Q235 and galvanized steel plates under the condition of cold metal transfer were investigated by using dynamic sessile drop method. The results show that the wetting behavior is closely related to the wire feed speed. Al-Fe intermetallic layer was observed whether the substrate is Q235 steel or galvanized steel, and the formation of Al-Fe intermetallic layer should satisfy the thermodynamic condition of such Mg-Al/Fe system. The wettability of molten AZ61 magnesium alloy is improved with the increase of wire feed speed whether on Q235 steel surface or on galvanized steel surface, good wettability on Q235 steel surface is due to severe interface reaction when wire feed speed increases, good wettability on galvanized steel surface is attributed to the aggravating zinc volatilization. When the wire feed speed is ≤10.5m·min-1, the wettability of Mg alloy on Q235 steel plate is better than on galvanized steel plate. However, Zn vapor will result in instability for metal transfer process.

Coarse grain heat affected zone (HAZ) of F460 steel was simulated by a Gleeble 3800 thermo-mechanical simulator. The microstructure, critical event of the HAZ formed at various heat inputs (E) were characterized and determined by optical microscopy (OM) and scanning electronic microscopy (SEM), and cleavage fracture stress σf was also calculated by ABAQUS software. Based on above systematic analysis, the intrinsic mechanism of ductile-brittle transition for F460 steel heat affected zones with different heat inputs were revealed. The results indicate that:with the improvement of heat input, the microstructures in sequence are a minority of lath martensite and massive fine lath bainite, more lath bainite with less granular bainite, more granular bainite with less lath bainite, bulky of granular bainite; and the maximum size of the original austenite grain and bainite packet becomes bigger with the improvement of heat input. The size of bainite packet is critical event of the cleavage fracture for coarse grain heat affected zone specimens with various heat inputs by comparing the relationships among residual crack length, original austenite grain size and bainite packet size. With the decreasing of the bainitic packet, the ductile to brittle transition temperature decreases. In addition, cleavage fracture stress σf is also calculated by ABAQUS software, σf gradually decreases with the increase of the heat input, which can explain the intrinsic mechanism of ductile to brittle transition temperature Tk with the change of the heat input.

In order to improve the cuttage propagation coefficient of fresh-eating peanut, the effect of different catting parts and ABT concentration on the cutting growth of fresh-eating peanut was studied by taking Jihuatian No. 1 plant as the test material. The results showed that the survival rate of cutting in different parts of peanut was in the order of the upper part > middle part > lower part. The upper cuttings of peanut plant were treated with five different concentrations of ABT rooting agents, among which the treatment with 150 mg ⋅ L-1 concentration had the best cutting effect and the most vigorous growth vigour. After 40 days of cutting, the survival rate was 83.3%, the average rooting was 12.7, the average root length was 2.85 cm, and the yield reached 3259.04 kg ⋅ hm², which was 148.48% higher than that of the control group with clear water, and the difference was very significant. In order to screen out the new peanut varieties with high yield, disease resistance and high quality suitable for being planted in the south of China, Zhanyou 75 was used as the control variety to identify the yield, quality and disease resistance of the 10 new peanut strains newly bred in the south of China. The results showed that the pod yield of Haikehua 181 and Fuhua 0945 respectively increased by 39. 66% and 23. 05% compared with that of Zhanyou 75, and the seed kernel yield respectively increased by 43. 79% and 24. 42% compared with that of Zhanyou 75, with extremely significant differences. Compared with Zhanyou 75, the pod yield of Quanhua 1478, Guihua 66 and Shany-ou 121 increased significantly, but the seed kernel yield did not increase significantly, while the pod yield and seed kernel yield of Qianyoufense and Zhanyou 103 did not increase significantly. Qionghua 1104 had higher oleic acid content (70. 12%). The protein content of Heyou 1079 was 29. 43%, which reached the national high protein standard (28%). The drought resistance and flood resistance of all the tested peanut strains were strong, and the disease resistance level was medium. In consideration of the indexes such as the yield, field growth and resistance to disease, Haikehua 181 and Fuhua 0945 showed better overall performance and could be used for the small-scale planting demonstration experimentally. Aiming at analyzing the effects of atmospheric parameters on gas turbine inlet air cooling technology (GTIAC) , the thermodynamic process, influencing factors and economic effects of GTIAC were studied. Based on the operating parameters of turbine and gas-steam coined cycle unit in a certain power plant, the system models of two kinds of power units were built in modular modeling method by GateCycle software, a thermal equilibrium simulation software, and GTIAC equipment was installed. The output power incase of these two kinds of power units after cooling inlet air (inlet air cooling effects) under different atmospheric parameters (temper, humidity and pressure) was analyzed quantitatively. The results indicate that the change of atmospheric parameters has a great effect on the gas turbine inlet air cooling effects, showing a certain numerical relationship between them. Straight line feature is the basis of linear target recognition. The existing heuristic search algorithms are suitable for straight line feature extraction in optical image while performing poor in SAR image. For this problem, a straight line extraction method for SAR image based on heuristic search is presented. Firstly, SAR image is filtered by Frost filter. Then, edges are detected by ratio of exponential weighted average (ROEWA) operator and non-maxima suppression location algorithm is adopted to obtain the binary edge map. Finally, considering the local information sufficiently, the strategy of search twice and the principle of "go straight" is utilized and the cost function and rules of heuristic search are designed. Then the straight line extraction is completed. The experiment results show good anti-noise performance and fracture resistance of the proposed method which can effectively describe the straight line feature of SAR images. [ABSTRACT FROM AUTHOR]

[ Objective ]The growth development characteristic and yield of peanut were studied under different mulches in order to provide references for extending high-yield cultivation techniques in peanut production of Fujian Province. [ Method ]The experiment was designed in randomized block with three replications, peanut varieties Quanhua 7 and Quanhua 8 were used for investigating the effects of different mulches viz., ordinary film, photolysis film, straw and bare land (control) on growth and development, and yield of peanut, etc. [ Result ] Ordinary film, photolysis film and straw mulching could shorten the growth duration of peanut, increase plant height and dry matter weight of stem and leaf. Ordinary film and photolysis film mulching could increase germination rate, productivity per plant of peanut, 100-pod weight, pods per plant, plump pods per plant. As compared with the control, the yield of peanut for photolysis film treatment was the highest, which significantly increased by 34.20% and 19.10% (P<0.01) of Quanhua 7 and Quanhua 8, respectively. The next was that of ordinary film treatment, the yield increased by 27.47% (P<0.01) and 16.54% (P<0.05). The yield of straw mulching treatment was decreased by 4.29% and 11.19%, respectively. As compared with the control, the economic benefit of Quanhua 7 and Quanhua 8 increased by 6455 and 11025 RMB Yuan/ha in photolysis film treatment, 5575 and 8790 RMB Yuan/ha in ordinary film treatment, while it decreased by 1640 and 4430 RMB Yuan in straw treatment. [ Conclusion ]Different mulches cultivating for peanut have different merits and demerits, respectively, so suitable cultivation model should be employed according to various environmental conditions to get high yield and benefit. Objective: To analyze the prevalence, genotype distribution and hematological characteristics of α,β-thalassaemia carriers in Huizhou area of Guangdong Province., Methods: 10 809 carriers of simple β-thalassaemia and 1 757 carriers of α,β-thalassaemia were enrolled as our study cohort. The hematological parameters were detected by automated blood cell counters and automatic capillary electrophoresis. Suspension array technology, gap-polymerase chain reaction (gap-PCR) and PCR-reverse dot blot were used for the genotyping of thalassaemia carriers., Results: The prevalence of α,β-thalassaemia in Huizhou area of Guangdong Province was 1.99%. A total of 62 genotypes were detected, and the most prevalent genotype was -- SEA / αα , β CD41-42 / β N (19.29%), the next was -- SEA / αα , β IVS-II-654 / β N (16.73%). Significant differences in mean corpuscular volume (MCV) and mean corpuscular hemoglobin (MCH) were found between different genotype groups for simple β-thalassaemia and α,β-thalassaemia. Violin plots showed that carriers with co-inheritance of β-thalassaemia and mild α-thalassaemia expressed the lightest anemia, and carriers with co-inheritance of β-thalassaemia and hemoglobin H (Hb H) disease expressed the most severe anemia., Conclusion: There is a high prevalence of α,β-thalassaemia in Huizhou area of Guangdong Province. Because of the lack of specific hematological makers for diagnosis of α,β-thalassaemia, it is necessary to distinguish it from simple β-thalassaemia by genotyping of α- and β-thalassaemia in order to correctly guide genetic counseling and prenatal disgnosis.

Determination of chloride salt solution by near infrared spectrum plays a very important role in Biomedicine. The near infrared spectrum analysis of Sodium chloride, potassium chloride, calcium chloride aqueous solution shows that the concentration change of chloride salt can affect hydrogen bond, resulting in the variation of near infrared spectrum of water. The temperature influence on NIR spectrum has been decreased by choosing reasonable wavelength range and the wavelength where the temperature effects are zero (isosbestic point). Chlorine salt prediction model was established based on partial least squares method and used for predicting the concentration of the chlorine ion. The impact on near infrared spectrum of the cation ionic radius, the number of ionic charge, the complex effect of ionic in water has also discussed in this article and the reason of every factor are analysed. Experimental results show that the temperature and concentration will affect the near-infrared spectrum of the solution, It is found that the effect of temperature plays the dominant role at low concentrations of chlorine salt; rather, the ionic dominates at high concentration. Chloride complexes are formed in aqueous solution, It has an effect on hydrogen bond of water combining with the cations in chlorine salt solution, Comparing different chloride solutions at the same concentration, the destruction effects of chloride complexes and catnions on the hydrogen bond of water increases in the sequences: CaCl2 >NaCl>KC. The modeling result shows that the determination coefficients (R2) = 99.97%, the root mean square error of cross validation (RM- SECV) = 4.51, and the residual prediction deviation (RPD) = 62.7, it meets the daily requirements of biochemical detection accuracy.

Adsorption tests of elemental mercury were carried out by using KI modified clay (bentonite) in simulated flue gas under different conditions. Brunauer-Emett-Teller measurement (BET), Fourier Transform Infraredspectroscopy (FTIR) and Thermogravimetric Analysis (TGA) were used to analyze the physical and chemical properties of the materials. Compared with the original bentonite, Hg(0) removal efficiency and Hg(0) adsorption capacity were drastically improved by the KI treatment. The experiment results also indicated that temperature could enhance the property of Hg(0) adsorption. Chemical adsorption was the dominant part in the process of Hg(0) adsorption. O2 was a beneficial factor for Hg(0) adsorption. SO2 was found to have a slight promotional effect on Hg(0) adsorption. The existence of H2O exhibited a dramatic inhibitory effect on Hg(0) adsorption.

The research on near infrared spectroscopy of sodium in biological and medicine is significant. Sodion is the main component of electrolytes in human blood and electrolytes help maintain the body's acid-base balance. In the present paper the concentration of sodium was determined with the use of NIR spectra. On the basis of NIR spectroscopic measurement mechanism of sodion, prediction models of the concentration of sodium were developed with linear regression using the absorbance at selected wavelengths. In order to reduce temperature perturbations to water bands with the measurement of sodium, Partial least squares regression (PLS) was adopted using select spectra area. The result shows that the determination coefficients (R2) = 99.82%, the root mean square error of cross validation (RMSECV) = 14.5, and the residual prediction deviation (RPD) = 23.7, for the calibration model. It meets the daily requirements of biochemical detection accuracy. This technique can be applied to quantitative analysis of sodion in the hospital laboratory.

Objective: To observe the clinical effects and complications in treating osteoporotic vertebral fractures with percutaneous kyphoplasty (PKP) and in order to found the cause and countermeasures of the complications., Methods: From March 2006 to March 2007, 31 patients with osteoporotic vertebral fractures were treated with unilateral percutaneous kyphoplasty. There were 11 males and 20 females, ranging in age from 54 to 91 years with the mean of 81 years. All patients were followed up for more than three years. At pre-treatment and postoperatively immediately, 1, 2, 3 years after PKP, the height of anterior vertebral body and thoracic-lumbar and back pain were respectively analyzed by imaging data and VAS scoring., Results: At pre-treatment and postoperatively immediately, 1, 2, 3 years after PKP, the height of anterior vertebral body were (0.9 +/- 0.2), (2.6 +/- 0.3), (2.6 +/- 0.2), (2.5 +/- 0.7), (2.5 +/- 0.4) cm, respectively; the VAS soring were (7.6 +/- 1.4), (2.3 +/- 0.7), (2.4 +/- 0.5), (2.8 +/- 0.3), (3.1 +/- 0.2) scores, respectively. The height of anterior vertebral body recoveried obviously after PKP (P < 0.05); following prolongation of time, the height of anterior vertebral body gradually reduced (P > 0.05). The thoracic-lumbar and back pain relieved obviously after PKP (P < 0.05); following prolongation of time, the pain gradually aggratated, but there was no significant difference (P > 0.05). At final follow up, reinforced vertebral re-fractures was found in 2 cases, adjacent vertebral fractures in 6 cases, distal vertebral fractures in 2 cases, asymptomatic degeneration of adjacent intervertebral in 5 cases., Conclusion: PKP have definite and early effects in treating osteoporotic vertebral fractures. But in mid-stage after PKP, the height of anterior vertebral body reduce and the pain aggravate gradually, especially degenerative adjacent vertebral fracture advent. Strict choosing the candidate of the precedure, improvement of materials of perfusion and reducing of volume of bone cement maybe can decrease incidence rate of complications.

Objective: To study the association between endometrioid uterine carcinomas and metabolic syndrome (MS)., Methods: A retrospective study was conducted on 123 patients who were admitted in Department of Gynecology Oncology, Zhejiang Cancer Hospital (study group) and 90 healthy women (control group) with matching age from Jan. 2005 to Mar. 2009. The general conditions [including age, whether menopausal, body mass index (BMI)]; the risk factors for MS [including waist circumference, fasting plasma glucose, triglycerides (TG), high-density lipoprotein (HDL) and systolic and diastolic blood pressure] were analyzed. The clinical stage, histological type, and pathology differentiated degree of study group with or without MS were also analyzed by univariate analysis and Cox proportional hazards models., Results: (1) The univariate survival analysis shown that there were no significant difference with age in two groups [(54.3 +/- 0.6) vs. (54.2 +/- 0.9) years; P > 0.05], while the rate of menopausal, BMI (> or = 25 kg/m(2)), the cases coupled with MS, the size of waist circumference (> 80 cm), the level of fasting plasma glucose (> or = 5.6 mmol/L), TG (> 1.7 mmol/L) and abnormal systolic and diastolic blood pressure in study group were higher than those in control group (67.5% vs. 48.9%, 45.5% vs. 23.3%, 43.9% vs. 18.9%, 50.4% vs. 27.8%, 53.7% vs. 21.1%, 40.7% vs. 21.1% and 40.7% vs. 25.6%, respectively, all P < 0.05). The percentage of HDL (< 1.30 mmol/L) was higher in study group than that in control group (63.4% vs. 32.2%, P < 0.05). (2) There were not significant difference for the clinical stage, pathological type, grades between patients with or without MS in study group (P > 0.05). (3) The Logistic multivariate survival analysis shown that central obesity, higher TG, lower HDL and abnormal plasma glucose were independent risk factors for endometrioid uterine carcinomas coupled with MS (P < 0.05)., Conclusion: Metabolic syndrome is marginally associated with an increased risk of endometrioid uterine carcinomas, which may be the new point to screen, prevention and treatment endometrioid uterine carcinomas.

Objective: To compare the effects of the treatment for the distal clavicle fracture with two different methods., Methods: There were 120 cases of the distal clavicle fractures involved in the study. Among them, 75 cases were treated by self-improved elastic band inclined "8" fixation (Manipulation group), 45 cases were treated by open reduction and internal fixation with clavicular hook plate (Operation group)., Results: There was statistically significant difference between manipulation group (6.71+/-2.35 weeks) and operation group (11.38+/-4.58 weeks) in the time of fracture union (P<0.01); but, there was no statistically significant difference between manipulation group (100%) and operation group (97.8%) in the rate of fracture union (P>0.05). According to the evaluation of shoulder-joint function, 56, 14 and 5 cases obtained excellent, good and bad result respectively in manipulation group; 38, 5 and 2 cases obtained excellent, good and bad respectively in operation group. There was no statistically significant difference between manipulation group (93.3%) and operation group (95.5%) in the rate of excellent and good (P>0.05)., Conclusion: Compared with open reduction and internal fixation with clavicular hook plate, treatment with self-improved elastic band inclined "8" fixation can shorten the union time of fracture and reduce therapeutic risks, so it is an economical, practical and safe method for the distal clavicle fracture.

As one of the major genes encoding plasma membrane H+ -ATPase, AHA1 gene plays an important role in regulating plant development and resistance to adverse stress. Taking AHA1 transgenic and wild type Arabidopsis thaliana as test plants, the nutrient uptake, resistance to oxidative stress, and organic acid secretion of the plants under aluminum (Al) stress were examined. The results showed that Al decreased the uptake of nitrogen (N), potassium (K), calcium (Ca) and magnesium (Mg), but increased the phosphorus (P) uptake by A. thaliana roots. AHA1 transgenic plant could accumulate more P and less Al than wild type plant. Al stress induced the increase of plant SOD and POD activities, but no significant difference was observed between AHA1 transgenic and wild type A. thaliana. Al triggered the secretion of organic acids significantly, and AHA1 transgenic plant secreted more organic acids than wild type plant. Vanadate, an inhibitor of plasma membrane H+ -ATPase, could inhibit the secretion of organic acids significantly, while Zn2+ and Mg2+ could promote the Al-induced secretion, and partially improve the inhibitory effects triggered by vanadate. It was suggested that AHA1 transgenic A. thaliana could increase its Al resistance via enhanced P uptake and organic acid secretion.

Objective: To explore effects of nordy on biological behaviors of human malignant glioblastoma cell line U87MG in vitro and transplanted tumor in vivo, and to identify the differential proteome upon Nordy induced differentiation., Methods: Glioblastoma U87MG cells were induced to differentiate by synthetic lipoxygenase inhibitor, Nordy. The drug was also given via peritoneal injection to nude mice (27 mg/kg body weight) bearing orthotopic transplanted tumors of U87MG cells in the brain. The tumor volumes and GFAP expression were measured. Total proteins of U87MG cells after Nordy treatment were analysed by two-dimensional gel electrophoresis. PDQuest 7.1 computer software was used to compare protein profiles of the treated cells with that of untreated control. Differentially expressed proteins were then selected and characterized by matrix assisted laser desorption ionization-time of flight-mass spectrometry. The functional aspects of these proteins were analyzed by bioinformatics., Results: Nordy suppressed both the proliferation of U87MG cells in vitro and the tumor growth of orthotopic transplanted tumors in vivo (P < 0.01). The differentially expressed proteins induced by Nordy included proliferation-associated gene A, alternative splicing factor ASF-3, eukaryotic translation initiation factor 5A, coffilin 1 (non-muscle), beta galactoside binding lectin, glyceraldehyde-3-phosphate dehydrogenase, enolase 1 and an unknown protein., Conclusions: Nordy promotes the differentiation of glioblastoma cells, by which it may serve as a therapeutic agent. Various proteins identified during Nordy-induced differentiation are involved in the cell proliferation, metabolism, differentiation, apoptosis and gene transcription.

Objective: To isolate, culture and identify glioma stem cells from human malignant glioma cell line U87, and investigate the changes of pro-angiogenic factors production by glioma stem cells followed by activation of CXCR4 and observe their tumorigenesis as well as the expression of vascular endothelial growth factor when implanted into nude mice., Methods: The ratio of CD133 positive cells was detected by flow cytometry. Magnetic separation of CD133 positive cells was carried out on the magnetic cell sorting system (MACS). Expression of nestin, glial fibrillary acidic protein (GFAP) and CXCR4 on tumorspheres was detected by indirect immunofluorescence under confocal laser scanning microscopy. The functional activation of CXCR4 was assessed by calcium mobilization experiments. ELISA was used to detect the production of vascular endothelial growth factor (VEGF) and interleukin-8 (IL-8) in conditioned medium. Glioma stem cells were implanted into nude mice to assess their tumorigenesis ability and the expression of VEGF., Results: The ratio of CD133 positive cells with stem cell property was 0.5% in U87 cells. Activation of CXCR4 on glioma stem cells induced calcium mobilization and increased VEGF and IL-8 protein secretion. CD133 positive cells secreted more VEGF and IL-8 than their negative counterparts in vitro. Tumors derived from CD133 positive cells grew more rapidly and expressed elevated level of VEGF than their negative counterparts., Conclusions: There are a small fraction of glioma stem cells in human glioblastoma cell line U87. Expressing functional CXCR4 and secreting more pro-angiogenic factors may be involved in tumor angiogenesis mediated by glioma stem cells.

Background & Objective: Chemoattractant receptors participate in essential pathophysiologic processes, including inflammation, wound healing, human immunodeficiency virus infection, and most interestingly in the progression of malignant tumor. This study was to explore the functional expression of formylpeptide receptor (FPR) in human glioblastoma cell line U87., Methods: The expression of FPR in U87 and FPR small interfering RNA (siRNA)-transfected U87 cells (FPR-siRNA-U87 cells) was detected with indirect immunofluorescent staining by confocal laser scanning microscopy. FPR was activated by its ligand formy-Met-Leu-Phe (fMLF). Cell proliferation was assessed by MTT assay. The production of vascular endothelial growth factor (VEGF) and interleukin-8 (IL-8) was measured by ELISA, and IL-8 mRNA was assessed by reverse transcription-polymerase chain reaction (RT-PCR)., Results: FPR was expressed on U87 cells, but not on FPR-siRNA-U87 cells. After activation of FPR by fMLF, the proliferation of U87 cells was enhanced markedly (P<0.05), but that of FPR-siRNA-U87 cells had no obvious change (P>0.05). fMLF (100 nmol/L) elicited a time-dependent increase in the secretion of IL-8 and VEGF. When stimulated with 100 nmol/L fMLF for 36 h, the protein levels of VEGF and IL-8 were significantly higher in stimulated U87 cells than in control cells [(3.13+/-0.23) ng/ml vs. (2.55+/-0.25) ng/ml, P<0.05û (7.54+/-0.53) ng/ml vs. (4.02+/-0.09) ng/ml, P<0.05], but those in FPR-siRNA-U87 cells had no obvious change; the mRNA level of IL-8 was significantly increased in U87 cells, but not changed in FPR-siRNA-U87 cells., Conclusion: Activating FPR can promote the proliferation of U87 cells and enhance the production of angiogenic factors.

Nordy is a synthesized chrial compound. To investigate the effects of nordy (25 - 100 micromol x L(-1)) on the function of formylpeptide receptor (FPR) of malignant human glioma cells, human glioblastoma cell line U87 was used to detect its proliferation, migration, calcium mobilization, vascular endothelial growth factor (VEGF) mRNA and protein levels after activation of FPR by its agonist N-formyl-methionyl-leucyl-phenylalanine (fMLF). Cell proliferation, migration ability, VEGF mRNA, VEGF protein and calcium mobilization were evaluated by cell counting, chemotaxis assay, RT-PCR, ELISA and spectrometry. Nordy (50 - 100 micromol x L(-1)) potently inhibited the proliferation, migration and calcium mobilization of U87 cells induced by fMLF (P < 0.05). Moreover, 100 micromol x L(-1) nordy showed a significantly impaired VEGF mRNA expression and protein secretion induced by fMLF (P < 0.05). Nordy could inhibit FPR functioning in glioma cell proliferation, migration and angiogenesis, which might be a possible mechanism of its anti-cancer effects.

Objective: To explore the effect of Nordy, a synthesized lipoxygenase Inhibitor, on the expression of formylpeptide receptor (FPR) and its functional activities after activation in human malignant glioma cells., Methods: Human malignant glioma cells of the line U87 were cultured and divided into 3 groups: N-formyl-methionyl-leucyl-phenylalanine (fMLF) group, added with 100 nmol/L fMLF; Nordy group, added with 100 micromol/L Nordy for 12 h and then added with 100 nmol/L fMLF; and control group. The expression of FPR I the U87 cells was detected with indirect immunofluorescence staining and confocal laser scanning microscopy. The effect of Nordy on the U87 cell proliferation induced by fMLF was assayed by MTT method. RT-PCR was used to detect the mRNA expression of vascular endothelial growth factor (VEGF) and interleukin-8 (IL-8) in the U87 cells. ELISA was used to detect protein secretion of VEGF and IL-8 in the supernatant., Results: FPR was expressed on the cytoplasmic membrane of U87 cells. The mean FPR absorbance of the Nordy group was significantly lower than that of the fMLF group. The U87 cell proliferation of the Nordy group was significantly inhibited in comparison with the other 2 groups. The mRNA expression of VEGF and mRNA expression of IL-8 in the U87 cells activated by fMLF were significantly higher than those of the control group (both P<0.05), and Nordy significantly attenuated the mRNA expression of VEGF and IL-8 at different time points (all P<0.05). ELISA showed that the VEGF and IL-8 protein levels in the supernatant were 4.14 ng/ml+/-0.28 ng/ml and 4.03 ng/ml+/-0.59 ng/ml respectively, and increased to 6.46 ng/ml+/-0.33 ng/ml and 7.54 ng/ml+/-0.73 ng/ml respectively after stimulation of fMLF, however, the VEGF and IL-8 protein levels in the supernatant of the Nordy group were 3.59 ng/ml+/-0.33 ng/ml and 3.13 ng/ml+/-0.48 ng/ml respectively., Conclusion: Nordy inhibits the FPR expression and the effects in promoting the cell proliferation and mRNA expression and protein secretion of VEGF and IL-8 after the activation of FPR in malignant human glioma cells, showing that Nordy has the effects of inhibition of tumor growth and angiogenesis.

Trans-translation is a mechanism that fixes stalled translation course. tmRNA is the core of trans-translation and it has dual function as a tRNA and a mRNA. tmRNA recognizes the ribosome carrying truncated mRNA with specificity under the help SmpB protein. It can be directed to A site by ribosomal protein S1. At first tmRNA prolongs the genetic message on the stalled mRNA, then termination codon stops peptide synthesis to form a nascent chain with the tag sequence. Finally, tmRNA-SmpB system frees stalled ribosomes and directs degradation of the products of these frustrated protein synthesis reactions. This paper introduces the most recent studies on trans-translation.

Objective: To investigate the association between atrial fibrillation and the single nucleotide polymorphism (SNP) of slow delayed rectifier K(+) channel (I(Ks)) genes in Han nationality Chinese., Methods: Three hundred and eighty of Han nationality Chinese (142 atrial fibrillation, 120 in-hospital and 118 out-hospital control) were enrolled in this study. Asian specific non-synonymous SNPs of KCNQ1 P448R, KCNQ1 R519H, KCNQ1 G643S, KCNE1 G38S and KCNE1 D85N were genotyped by restriction fragment length polymorphism analysis. A newly cloned KCNE4 gene was also screened for any possible SNP., Results: The minor allele frequency of KCNQ1 P448R, KCNQ1 R519H, KCNQ1 G643S, KCNE1 G38S and KCNE1 D85N in out-hospital subjects was 0.079, 0, 0.042, 0.317 and 0.004, respectively. None of these SNPs was relationed with any atrial fibrillation phenotype. A KCNE4 E145D was discovered and proven statistically to relation significantly to atrial fibrillation by logistic regression analysis (OR = 1.66, P = 0.044). The minor allele frequency of KCNE4 E145D was as high as 0.271 in out-hospital subjects., Conclusions: None of the SNPs of KCNQ1 P448R, KCNQ1 R519H, KCNQ1 G643S, KCNE1 G38S and KCNE1 D85N was associated with atrial fibrillation phenotype, but KCNE4 E145D may relation to atrial fibrillation. The effect of KCNE4 E145D variation on the function of I(Ks) channel is to be determined.

Objective: To remodel retinal neovascularization by overexpression of HESR-1 in vitro., Methods: Human retinal capillary endothelial cell (HRCEC) was isolated, then HRCECs were transfected with pcDNA3.1+HESR-1. The changes of expression of KDR and occluding in HRCECs were analyzed using ELISA assay, the regulation of occluding in HRCECs by HESR-1 was evaluated by western blot., Results: Over-expression of HESR-1 down-regulated KDR expression, but occludin expression was up-regulated in HRECEs, the result also showed that the vessels permeability was reduced by HESR-1., Conclusions: Overexpression of HESR-1 in HRCECs can inhibit the neovascularization and decrease the permeability of new vessels. It may be a promising gene therapy for intraocular neovascularization.

Objective: To analyze the correlation between drug exposure (AUC0-4) and blood concentration of different sample points during Neoral absorption phase in Chinese adult liver transplant recipients, and to evaluate the possibility of using post-dose 2 hour level (C2) as a surrogate marker of AUC0-4 for the therapeutic drug monitoring (TDM) of Neoral., Methods: Neoral levels at 0, 1, 2, 3, and 4 hours (C1, C2, C3, C4) after morning dose of 22 de novo Chinese adult liver transplant recipients were monitored during different posttransplant periods. Liner regression was used to analyze the correlation between CsA concentration at different time points and the AUC0-4., Results: The best correlation was found at C2, while the correlation of C0 was the lowest. During following-up, the correlation between C2 and AUC0-4 was very stable., Conclusion: C2 had the best correlation with drug exposure. This correlation is very stable. C2 may be used as a good surrogate marker of AUC0-4 for the TDM of Neoral.
