Background Along with the acceleration of population aging and the increase in average life expectancy, how to improve the quality of life of residents has become a social focus. Cancer is a major factor that seriously affects the health, causing increased medical burden. Therefore, it is of great significance to analyze cancer incidence in Cenxi City, Guangxi. Objective To perform a statistical analysis of cancer incidence in Cenxi from 2016 to 2020. Methods Data on cancer incidence in Cenxi from 2016 to 2020 were obtained from the Cancer Registry System of Cenxi Center for Disease Control and Prevention. Cancers were classified by the Guideline for Chinese Cancer Registration and International Classification of Disease for Oncology, Third Edition (ICD-O). Sex-specific and age-specific crude incidence rates, standardized incidence rates, and cumulative incidence rates (0-74 years), as well as truncated age-standardized incidence rate (35-64 years) were calculated. The standardized incidence rate was calculated by the 2000 Population Census of China and Segi's world population structure (Chinese age-standardized incidence rate was referred to as ASIRC, and world age-standardized incidence rate as ASIRW) . Results There were 4 205 registered new cases of cancer from 2016 to 2020 in Cenxi. The crude cancer incidence rate was 98.77/105. The ASIRC, ASIRW, cumulative incidence rate (0-74 years) and the truncated age-standardized incidence rate (35-64 years) were 85.48/105, 82.54/105, 9.08%, and 165.08/105, respectively. Nasopharyngeal carcinoma (NPC) is a common malignant tumor in China. Radiotherapy is the first-line treatment. After appropriate radiotherapy, about 5%-15% patients experience recurrence. In view of the poor efficacy and high incidence of severe late toxicities associated with re-irradiation, salvage surgery by the transnasal endoscopic approach is recommended for recurrent NPC (rNPC). Compared with re-irradiation, endoscopic surgery can better prolong survival, improve the quality of life, and reduce complications and medical expenses of patients with rNPC. However, the complexity of the nasopharyngeal skull base enhances the difficulty and risk of surgery. Expanding the boundary of surgical resection remains a clinical challenge for otolaryngologists. In this regard, to help more advanced patients with rNPC, the surgical innovative system of NPC needs to be established by multi-disciplinary cooperation, involving skull base anatomy-based investigation, appropriate administration of the internal carotid artery (ICA), repair of skull base defect, and establishment of various types of endoscopic endonasal nasopharyngectomy.

Objectives Due to lower weight and volume under the same working conditions, permanent magnet equipment is extensively used by many kinds of ships. However, its measured vibration at high frequencies is much higher than traditional equipment, which will affect the ship's acoustic performance, and reduce the effectiveness of using such equipment in ships. A kind of compound rubber vibration isolator is therefore designed to solve this problem.Methods In order to isolate the vibrations of a given piece of permanent magnet equipment, a compound rubber vibration isolator is designed by inserting middle mass into a rubber isolating system. The transmission characteristics of traditional and compound rubber vibration isolators are compared, and experimental validation for the isolating design is carried out.Results The results show that the proposed compound vibration isolator is feasible and can improve the vibration isolation effect of a piece of permanent magnet equipment's machine leg at high frequency. This effect can improve 20 dB compared to traditional rubber vibration isolators.Conclusions As a method for vibration control, the designed compound rubber vibration isolator can effectively control high frequency vibration.

Significant results have been achieved in the application of distributed underground water reservoirs in Shendong area.Ningdong area has begun to demonstrate its feasibility.However, the mine water in Ningdong area is mostly of high turbidity and high salinity, so the pre-treatment of the reservoir water is required.The experiment compares the flocculation effect of fly ash-based composite flocculant and unfilled coal ash flocculant, and explores the influence factors of fly ash-based flocculant, and determines the optimal dosage and test conditions.In view of the characteristics of high turbidity and high salinity mine water, this experiment, using fly ash as raw material, to obtain high turbidity and high ore by mixing it with polyacrylamide(PAM), polyaluminum chloride(PAC) and chitosan, gets flocculants for treating high turbidity and high salinity mine water.The experiment compares the flocculation effect of fly ash-based composite flocculant and unfilled coal ash flocculant, and explores the influencing factors of fly ash-based flocculant, and determines the optimal dosage and test conditions.The experimental results show that:1.The treatment effect of mine water in the five treatment groups is:PAM+PAC+chitosan+fly ash > PAM+PAC+chitosan > PAM+PAC> PAM> PAC;2.The optimum condition of fly ash composite flocculant in temperature is 25° C, pH 7, stirring strength 220 r/min, stirring time 20 min;3.Under the optimal conditions, the removal rates of salinity, suspended solids, calcium ions, total hardness and sulfate by composite flocculants are 12.16%, 98.04%, 80.57%, 77.86% and 95.44%;4.It is feasible to use fly ash-based composite flocculant as pretreatment flocculant for the treatment of high salinity high suspended solid water in the groundwater reservoir.This article provides experimental theoretical basis and technical support for the pretreatment of flocculation in the groundwater reservoir.

[Objectives] We found a new component of propeller noises which is induced by the excitation of the low-frequency elastic mode of the propeller. In particular,the model where all the blades of a propeller vibrate in the same phase is stimulated will result in strong acoustic radiation. We have named the mode shape in which all the blades of a propeller vibrate in the same phase as "Jellyfish Mode". Also, the low-frequency narrow-band sound radiation of the propulsion system induced by the excitation of "Jellyfish Mode" is named by the "Jellyfish Effect". To reveal its mechanism,a study of the low frequency intrinsic modal characteristics of the propeller is carried out.[Methods] the refined mesh FEM method is utilized to study the dynamic feature of the propeller. The modal characteristics of the propeller both in air and water are obtained and the influence of the structural detuning of the propeller on the modal characteristics is analyzed. Then,the cyclic symmetric structure vibration theory is cited to validate and further study the low-frequency dynamic property of the propeller.[Results] The research revealed the grouping modal characteristics of the propeller both in air and water. Furthermore,the single frequency mode and repetition frequency mode characteristics in a mode group are also brought out. The research results showed that the structural detuning of the propeller has a great effect on the repetition frequency mode while it has little effect on the single frequency mode. The refined mesh FEM numerical analysis results are consistent with the theoretical results.[Conclusions] The study revealed the low frequency natural dynamic characteristics of the propeller and the research above can provide theoretical supports for propeller noise analysis and control, which is of great significance in theoretical and practical engineering.

Objective To investigate the effect of down-regulation of Notch1 expression mediated by lentivirus on the activation of caspase-9 and mitochondrial membrane potential of fibroblast from pathological scars(PSF). Methods PSFs were isolated and cultured and then infected by Notch1 siRNA lentivirus and negative control lentivirus respectively.The level of Notch1 mRNA and protein was detected by real-time PCR and Western blot,respectively.MTT assay was used to detect cell proliferation; PI monochrome staining was used to detect cell cycle changes. Annexin V-FITC/PI assay was used to detect apoptotic changes; Western blot was used to detect the level of cyclin D1, CDK4, c-caspase-3, c-caspase-9 and cytochrome C protein levels in cytoplasm and mitochondria; JC-1 method was used to detect the mitochondrial membrane potential. Results The expression of Notch1 in PSFs transfected with Notch1 siRNA lentivirus was significantly lower than that in the negative control lentiviruses and the non-transfected PSFs(P＜0.05).The proliferation of PSFs decreased after Notch1 was down-regulated (P＜0.05);the percen-tage of G0/G1 phase increased(P＜0.05); the apoptotic rate increased(P＜0.05); the expression of c-caspase-3 and c-caspase-9 protein increased(P＜0.05); the mitochondrial membrane potential of the cells decreased significantly(P＜0.05); the level of cyclin D1 and CDK4 protein in cells decreased(P＜0.05); cytochrome C protein increased in cytoplasm and decreased in mitochondria(P＜0.05). Conclusions Down-regulation of Notch1 inhibits proliferation and induces apoptosis of PSF.

Further evidence is needed to explore the impact of high-altitude environments on the neurologic function of neonates. Non-invasive techniques such as cerebral near-infrared spectroscopy and amplitude-integrated electroencephalography can provide data on cerebral oxygenation and brain electrical activity. This study will conduct multiple cerebral near-infrared spectroscopy and amplitude-integrated electroencephalography monitoring sessions at various time points within the first 3 days postpartum for healthy full-term neonates at different altitudes. The obtained data on cerebral oxygenation and brain electrical activity will be compared between different altitudes, and corresponding reference ranges will be established. The study involves 6 participating centers in the Chinese High Altitude Neonatal Medicine Alliance, with altitude gradients divided into 4 categories: 800 m, 1 900 m, 2 400 m, and 3 500 m, with an anticipated sample size of 170 neonates per altitude gradient. This multicenter prospective cohort study aims to provide evidence supporting the impact of high-altitude environments on early brain function and metabolism in neonates. [ABSTRACT FROM AUTHOR]

Objective: To investigate the role and mechanisms of SOCS3 protein in IL-10 inhibiting the expression of inflammatory factors in Chlamydia-infected cells. Methods: The activation of STAT3 protein were examined in Chlamydia-infected cells by Western blot, and the activation of p38 and ERK1/2 were also examined in infected cells treated with socs3 siRNA. The expression of socs3 gene was examined in Chlamydia-infected cells treated with IL-10 or Stattic by RT-PCR. IL-6 and IL-12 were measured in infect-ed cells treated with socs3 siRNA using ELISA kits. Results: Socs3 expression was up-regulated by IL-10 through activation of STAT3 protein. IL-6 and IL-12 induced by Chlamydia were down-regulated by IL-10 through induction of socs3. The activation of p38 and ERK1/2 signalling pathways were inhibited by SOCS3. Conclusion: IL-10 inhibits the production of pro-inflammatory cytokines through induction of SOCS3 and inhibition p38 and ERK1/2 signalling pathways. [ABSTRACT FROM AUTHOR]

Objective.To explore the clinical predictive value of skin cholesterol and electrocardiogram treadmill exercise (EET) for coronary heart disease (CHD) in patients with atypical chest pain. Methods.One hundred and twenty-eight patients with atypical chest pain admitted from April 2020 to April 2021 were enrolled. All patients did not take any lipid-lowering drugs or anti-myocardial ischemia drugs within 1 month before admis- sion. Skin cholesterol content test, EET test, coronary angiography, Gensini score and number of diseased vessels were completed during hospitalization. According to the results of coronary angiography, the patients were divided into CHD group and non-CHD group. General clinical data, skin cholesterol content and EET results of the two groups were analyzed. Correlation analysis was performed between skin cholesterol content and Gensini score. Multi- variate logistic regression was used to screen factors with predictive value for the diagnosis of CHD in patients with atypical chest pain. Results.The content of cholesterol in skin increased significantly in patients with CHD. With the increase of the number of coronary artery lesions, the content of cholesterol in skin also increased significantly. Skin cholesterol content was positively correlated with Gensini score. Logistic regression analysis showed that skin cholesterol content and EET positive were predictors of CHD. The area under ROC curve of skin cholesterol content and EET in predicting the diagnosis of CHD in patients with atypical chest pain was 0.879 and 0.617, the corre- sponding sensitivity 86.2% and 67.3%, and the specificity 74.3% and 49.3%, respectively. The area under ROC curve of the combination of the two in predicting the diagnosis of CHD in patients with atypical chest pain was 0.917, and the sensitivity and specificity were 91.5% and 82.3%, respectively. Conclusion.Skin cholesterol content is of good value in the diagnosis of CHD in patients with atypical chest pain, and EET can further improve the sensitivity and specificity of diagnosis. Evaluating possible negative effects of alien genes expressed in transgenic plants is essential in environmental safety assessment of genetically modified (GM) organisms. We compared bacterial communities in rhizosphere soil of Brassica napus overexpressing mEPSPS (JHX6) and the control line J9707. Results showed that Proteobacteria, Bacteroidetes, Acidobacteria and Actinobacteria were the dominant bacterias in the rhizosphere soil of B. napus. Neither alpha diversity analysis (e.g. Shannon and Simpson index), PCoA nor MRPP analysis had revealed significant differences between the same developmental stage of the GM rape and control line. Meanwhile, Planctomycetes was less enriched in rhizosphere soil of GM rape at flowering stage compared with that of wild type. The enrichments of few bacterial genera were significantly different at 3 developmental stages of the GM rape and wild type. The bacterial diversity and community structure were great difference among the 3 developmental stages, especially in comparison of seedling stage. In total, mEPSPS overexpression in B. napus plants caused no significant variation on bacterial community in rhizosphere soil. While long-term dynamic monitoring is still necessary. A concept of emergence was introduced, and the optimization of system was carried out through reconstruction by utilizing the inherent logical relationship between system elements and architecture and the external expression of architecture and form, so as to equip system with more positive emergence instead of negative emergence. A concept of reconstruction pole was put forward, and the method of comprehensive evaluation across systems was established through the comparison of reconstruction pole elements, thus the problem of complex giant system optimization was broken. Objectives: To explore the effect of electroacupuncture (EA) at "Baihui" (GV20) and "Shenting" (GV24) on the microvascular structure and related protein expression in the hippocampus of vascular dementia (VD) rat model, and to investigate the mechanism of EA in the treatment of VD., Methods: A total of 24 SD rats were randomly divided into sham operation, model, EA, and oxiracetam groups, with 6 rats in each group. Multiple cerebral infarction method was used to establish VD model. In the EA group, EA was applied to GV20 and GV24 for 30 min, once daily for 14 days. Rats in the oxiracetam group were treated with oxiracetam (50 mg/kg) by intraperitoneal injection, and the course of treatment was the same as that in the EA group. Learning and memory ability were evaluated by using Morris water maze test and new object recognition experiment. The cerebral blood flow was detected by laser doppler. The microvascular structure in the hippocampus was observed by transmission electron microscopy. The expression of vascular structure related proteins of platelet-derived growth factor receptor (PDGFR)-β, platelet endothelial cell adhesion molecule-1(CD31), neural cadherin N-Cadherin, Zonula occludens protein-1(ZO-1) in the hippocampus were measured by Western blot., Results: Compared with the sham operation group, the rats in the model group had a significant increase in time of first crossing the platform, a significant decrease in the number of crossing platform and the new object preference index ( P <0.05), a significant decrease in cerebral blood flow ( P <0.05), and a significant increase in the brain weight ( P <0.05). The structure boundary of pericyte and endothelial cells in the microvessels of the hippocampal CA1 area of model group was blurred, accompanied by obvious edema around the vessels and the reduction of tight junctions. The protein expression levels of PDGFR-β, CD31, N-Cadherin, ZO-1 were significantly decreased in the model group compared with those in the sham operation group ( P <0.05). Compared with the model group, the time of first crossing the platform of rats in the EA and oxiracetam group was shortened, the number of crossing platform were increased ( P <0.05), the cerebral blood flow was increased ( P <0.05), the brain weight was decreased ( P <0.05), the morphology and structure of pericyte and endothelial cells in the microvessels of hippocampal CA1 area were intact, accompanied by the increase of tight junctions. Additionally, Compared with the model group, the EA group had a significant increase in the new object preference index ( P <0.05), the protein expression levels of PDGFR-β, CD31, ZO-1 in the EA group were increased ( P <0.05), and the expression of PDGFR-β, N-Cadherin, ZO-1 in the oxiracetam group were increased ( P <0.05)., Conclusions: EA at GV20 and GV24 can improve the learning and memory ability of VD rats, and the mechanism may be related to the repair of microvascular structures and improvement of cerebral blood flow.

Histone lysine-specific demethylase 1(LSD1) has become a promising molecular target for lung cancer therapy. Upon the screening platform for LSD1 activity, some Chinese herbal extracts were screened for LSD1 activity inhibition, and the underlying mechanism was preliminarily investigated at both molecular and cellular levels. The results of LSD1 inhibition showed that Puerariae Lobatae Radix extract can effectively reduce LSD1 expression to elevate the expression of H3 K4 me2 and H3 K9 me2 substrates in H1975 and H1299 cells. Furthermore, Puerariae Lobatae Radix was evaluated for its anti-lung cancer activity. It had a potent inhibitory ability against the proliferation and colony formation of both H1975 and H1299 cells. Flow cytometry and DAPI staining assays indicated that Puerariae Lobatae Radix can induce the apoptosis of lung cancer cells. In addition, it can significantly suppress the migration and reverse the epithelial-mesenchymal transition(EMT) process of lung cancer cells by activating E-cadherin and suppressing the expression of N-cadherin, slug and vimentin. To sum up, Puerariae Lobatae Radix displayed a robust inhibitory activity against lung cancer, and the mechanism may be related to the down-regulation of LSD1 expression to induce the cell apoptosis and suppress the cell migration and EMT process. These findings will provide new insights into the action of Puerariae Lobatae Radix as an anti-lung cancer agent and offer new ideas for the study on the anti-cancer action of Chinese medicine based on the epigenetic modification.

To provide accurate information for registration and safety evaluation of surgical robot, the pose repeatability measurement method was proposed. According to the terminal instrument of the master-slave surgical robot (such as high-frequency electric knife, ultrasonic knife), a suitable target ball fixture was designed. The node data at 10%, 50% and 100% rated speed were measured respectively. Through data analysis, the pose repeatability property of the tested samples at different speeds was obtained. It has high applicability and repeatability, and can meet the requirements of data traceability and registration testing.

Objective: To investigate the application value of whole exome sequencing technology in fetuses with congenital structural abnormalities., Methods: The chromosomal abnormalities of 1147 families were analyzed. According to the follow-up results, the data of fetuses with new phenotypes in late pregnancy or after birth were reanalyzed. Subgroups were divided according to the organs involved and whether single malformation or not. The gene regulatory network map was drawn by using string database and Cytoscape software. Fisher exact probability method was used to compare the difference of the diagnostic rate of pathogenic genes among the groups., Results: A total of 160 fetal cases received positive molecular diagnosed, involving 178 variant sites of 125 pathogenic genes, including 8 cases (4.9%, 8/163) by data reanalysis, and the overall positive diagnosis rate was 13.9%. Diagnostic rate was highest in the group of skeletal malformation (31.5%, 39/124) and lowest in that with thoracic malformation (0, 0/32). The gene clusters of fetal edema and intrauterine growth restriction were independent, and were not associated with the major structural malformations. The probability of each parent carrying the same recessive gene variant was 0.03 (39/1146) and 0.08 (4/53) with positive family history., Conclusion: For fetuses with congenital structural abnormalities that are negative for conventional genetic tests, 13.9% of phenotypic associated pathogenic/likely pathogenic genetic variants can be detected by whole exome sequencing technology. Its application value for prenatal diagnosis varies in fetus with different organs involved. Reanalysis of sequencing data for cases with new phenotypes in late pregnancy or after birth can further improve the molecular diagnosis rate. Further investigations are needed to explore the related genetic mechanisms.

Objective: To assess the value of chromosomal microarray analysis (CMA) for fetal duodenal obstruction (DO)., Methods: Fifty-one fetuses with DO identified by prenatal ultrasound were divided into DO only group and DO with other anomaly group. CMA was carried out on amniotic fluid or umbilical blood samples, and the outcome of pregnancy of all cases were followed up., Results: Eight fetuses (15.7%) were found with genomic abnormalities, which included 3 chromosomal aneuploidies and 5 copy number variations (CNVs), including one 17q12 microduplication syndrome, one 13q21.33q31.1 microdeletion, one 13q21.32q22.3 deletion, one 13q21.2q31.1 deletion and one 1q43q44 duplication. EDNRB from 13q and HNF1B from 17q12 are candidate genes for fetal DO. No significant difference was found in the detection rate of pathogenic CNVs between the DO only and DO with other anomaly groups (9.5% vs.11.1%, P> 0.05). There were 39 live borns, 1 stillbirth, and 11 artificial abortions (8 with abnormal CMA results)., Conclusion: There is a correlation between fetal DO and abnormal copy number of the genome, for which prenatal diagnosis is necessary. CMA not only can detect microdeletions/microduplications, but also identify pathogenic genes, which can facilitate prenatal diagnosis, genetic counseling and prognosis for the fetus.

Objective: To study the mechanisms of curcumin alleviating oxidative stress and spleen apoptosis induced by overtraining in rats by regulating Kelch-like ECH-associated protein-1 (Keap1)-nuclear factor erythroid 2-related factor 2 (Nrf2)-antioxidant response element (ARE) signaling pathway., Methods: Male Wistar rats of 7 weeks old were divided into control group (C group, 12), overtraining group (OM group, 11), curcumin + overtraining group (COM group, 14). The C Group did not undergo any exercise intervention. The OM and COM group underwent 8-week incremental load swimming training. During the training, rats in the COM group were treated with curcumin at the dose of 200 mg/(kg·d) in the volume of 5 ml/kg by gavage, and rats in the other groups were given an equal volume of solvent, 0.5% sodium carboxymethylcellulose. Twenty-four hours after the last training, the spleen index was calculated by weighing, the pathological changes of the spleen were observed by light microscopy, and the biochemical indicators of blood and spleen were detected., Results: The spleen structure of C group was normal under light microscope; the spleen index of OM group was significantly lower than that of C group (P＜0.01) and pathological changes were obvious; the spleen index of COM group was significantly higher than that of OM group (P＜0.05) and histomorphological changes were relieved. Compared with C group, in OM group, serum corticosterone (Cor) level, spleen apoptosis level, malondialdehyde (MDA) concentration and the expression of proapoptotic Bcl-2 associated X protein (Bax) in spleen were increased (P＜0.05 or P＜0.01); the body weight, serum testosterone (T), superoxide dismutase (SOD) activity, the expressions of heme oxygenase-1 (HO-1) and anti-apoptotic B cell lymphoma-2 protein (Bcl-2) in spleen were decreased (P＜0.05 or P＜0.01); the expression of Nrf2 was not changed significantly (P＞ 0.05). Compared with OM group, in COM group, there were no significant changes in body weight (P＞0.05), serum T level, SOD activity, the expressions of Bcl-2, Nrf2 and HO-1 in spleen were increased (P＜0.05 or P＜0.01); serum Cor level, spleen apoptosis level, MDA concentration and the expression of Bax were decreased (P＜0.05 or P＜0.01). The change trend of T/Cor ratio between groups was consistent with the change of testosterone, and the change trend of Bcl-2/Bax ratio was consistent with the change of Bcl-2., Conclusion: The 8-week incremental load excessive swimming training aggravated spleen apoptosis, led to pathological changes and dysfunction of spleen. Curcumin can up-regulate expression of Nrf2 and HO-1, alleviate oxidative stress induced by overtraining, enhance Bcl-2 expression and attenuate Bax expression, thereby inhibiting excessive spleen apoptosis of rats, protecting the structure and function of spleen.

The MS8.0 Wenchuan earthquake in 2008 led to huge damage to land covers in northwest Sichuan, one of the critical fragile eco-regions in China which can be divided into Semi-arid dry hot climate zone (SDHC) and Subtropical humid monsoon climate zone (SHMC). Using the method of Bilog-ECO-microplate technique, this paper aimed to determine the functional diversity of soil microbial community in the earthquake-affected areas which can be divided into undamaged area (U), recover area (R) and damaged area without recovery (D) under different climate types, in order to provide scientific basis for ecological recovery. The results indicated that the average-well-color-development (AWCD) in undamaged area and recovery area showed SDHC > SHMC, which was contrary to the AWCD in the damaged area without recovery. The AWCD of damaged area without recovery was the lowest in both climate zones. The number of carbon source utilization types of soil microbial in SHMC zone was significantly higher than that in SDHC zone. The carbon source utilization types in both climate zones presented a trend of recover area > undamaged area > damaged area without recovery. The carbon source metabolic diversity characteristic of soil microbial community was significantly different in different climate zones. The diversity index and evenness index both showed a ranking of undamaged area > recover area > damaged area without recovery. In addition, the recovery area had the highest richness index. The soil microbial carbon sources metabolism characteristic was affected by soil nutrient, aboveground vegetation biomass and vegetation coverage to some extent. In conclusion, earthquake and its secondary disasters influenced the carbon source metabolic diversity characteristic of soil microbial community mainly through the change of aboveground vegetation and soil environmental factors.

Objective: To explore the expression of PI3K in esophageal squamous cell carcinoma (ESCC) and its clinical significance., Methods: The expression of PI3K proteins was detected by immunohistochemical SP staining in 91 ESCC tissues and normal mucosa of 10 cases. RT-PCR was used to detect the expression of PI3K mRNA in 30 ESCC tissues and adjacent normal mucosa. The relation between PI3K expression and the clinicopathological features and prognosis was analyzed., Results: The positive rate of expression of PI3K protein in ESCC was 86.8%, significantly higher than that in normal esophageal mucosa (10.0%, P<0.001). There was a positive correlation between P13K expression and the degrees of differentiation, invasion depth, and clinical stage. No positive correlation was found between the expression of PI3K protein and age, gender and lymph node metastasis (P>0.05). The poorer is the differentiation of ESCC, the stronger is expression of PI3K protein (r = 0.351, P<0.05); the deeper is the invasion depth of ESCC, the stronger is expression of PI3K protein (r = 0.210, P<0.05); and the higher is the clinical stage of ESCC, the stronger expression of PI3 K protein (r = 0.240, P<0.05). RT-PCR results demonstrated that the level of PI3K mRNA expression in ESCC was 0.675 +/- 0.029, significantly higher than that in the adjacent normal mucosa (0.349 +/- 0.073, P<0.05). The high expression of PI3K mRNA was correlated with poor differentiation, deep invasion and clinical stage of ESCC (P<0.05). The result was consistent with SP staining immunohistochemistry. By Kaplan-Meier analyses, following factors were observed to be significantly associated with prognosis: PI3K expression, the degrees of differentiation, invasion depth, lymph node metastasis, clinical stage, tumor embolus and stump invaded (all P<0.05). The multivariate analysis showed that clinical stage (P<0.05), invasion depth (P<0.05) and stump invaded (P<0.05) were significantly correlated with the prognosis. The multivariate analysis of Cox model showed that PI3K was not an independent prognostic indicator of prognosis (chi2 = 1.235, P = 0.266)., Conclusions: The expression levels of PI3K protein and mRNA in ESCC tissues are significantly increased and PI3K plays a role in the carcinogenesis and development of ESCC. The enhanced PI3K expression may serve as a reference index indicating a poor prognosis in ESCC.
