Objective To investigate the effect of Versican V0/V1 on biological behaviors of mouse dental papilla cells (mDPCs). Methods mDPCs were isolated from C57BL/6J mice at embryonic day 16.5 (E16.5). A small interfering RNA (siRNA) constructed specifically for Versican V0/V1 was transfected into mDPCs.The silencing efficiency was verified by quantitative real-time fluorescence polymerase chain reaction (qRT-PCR) and immunofluorescence staining. The proliferation rate of mDPCs was determined using EdU assay; the migration ability of mDPCs was evaluated by scratch test and transwell assay. Alkaline phosphatase (ALP) staining and alizarin red staining were used to assess the mineralization capability of mDPCs. The molecules related to odontogenic differentiation and mineralization at mRNA levels were measured by qRT-PCR. In order to clarify the influence of Lactiplantibacillus pentosus fermentation on the structure of peanut proteins, changes in the structure of peanut protein isolate (PPI), arachin and conarachin before and after fermentation were studied. The results showed that L. pentosus fermentation significantly increased the free sulfhydryl content, surface hydrophobicity and denaturation temperature, and significantly reduced the β-folding content and denaturation enthalpy of arachin (P < 0.05). It reduced the free sulfhydryl content, surface hydrophobicity, and denaturation enthalpy, and significantly increased the degeneration temperature and α-helix content of PPI and conarachin (P < 0.05). Through particle size, potential and spectral analysis, we found that L. pentosus fermentation significantly improved the particle size and zeta potential of each protein component, and resulted in a red shift in their fluorescence peaks. In contrast to PPI and conarachin, the maximum fluorescence emission wavelength of arachin showed a 1.3 and 2.4 nm red shift, and arachin exhibited a 19.54% and 14.75% higher increase in thermal denaturation temperature, respectively. Thermal properties and scanning electron microscopic (SEM) analysis showed that arachin had maximum structural unfolding and higher thermostability after fermentation, indicating that L. pentosus fermentation is more likely to promote the molecular modification of arachin, and contributes to the gel modification of peanut proteins.

Background Intellectual activities such as reading and playing puzzle games can slow the decline of cognitive function in the elderly, but the effects of specific types of such activities on cognitive function and cognitive domains need to be further studied. Objective To explore the influence of common types of intellectual activities on cognitive function and cognitive domains of the elderly in the community. Methods From May to August 2022, stratified convenience sampling was used to select elderly people from four communities in Nanjing, Changzhou, Nantong and Xuzhou of Jiangsu Province. A face-to-face survey was conducted with a general information questionnaire and the Montreal Cognitive Assessment (MoCA) Beijing edition to collect data regarding sociodemographics, frequency and types of intellectual activities, and cognitive function. Stepwise multiple regression analysis was used to explore the relationship between intellectual activities and different cognitive domains. Results In total, 782 cases attended the survey, and 758 of them (96.93%) who completed it were included for analysis, including123 from Nanjing, 197 from Changzhou, 240 from Nantong, and 198 from Xuzhou. The intellectual activities done by these older people include learning new knowledge (n=170), playing chess and cards (n=228), reading (n=228), singing (n=59), playing puzzle games (n=57), helping grand children with their homework (n=42), painting (n=16), playing a musical instrument (n=47), and practicing calligraphy (n=30).
