2. Simultaneous Analysis of 16 Polycyclic Aromatic Hydrocarbons in Cigarette Smoke Based on Matrix Solvent Correction Combined with High-Temperature Ion Source GC-MS/MS

An improved method was developed for the detection of 16 polycyclic aromatic hydrocarbons (PAHs) in mainstream cigarette smoke during combustion. The methodological approach involved matrix matching standard solution preparation, direct injection of the extraction solution, and measurement of standard linearity curves to quantitatively analyze the release of target compounds. Considering the diverse physical and chemical properties as well as distribution patterns of the 16 PAHs in cigarette smoke, the impact of room temperature and high-temperature ion sources on the response of target compounds was investigated. Additionally, the preparation methods of matrix solvents and the matrix effects on the quantitative results of target compounds were also explored. Furthermore, a systematic study was conducted on the smoke release distribution of commercially available cigarettes with different circumferences. The results demonstrated that direct injection of sample extract eliminates the need for sample purification, concentration, and other processing steps employed in current detection methods. This simplifies sample pretreatment procedures while reducing organic solvent usage and consumables consumption. Moreover, it resolves issues related to low operational efficiency when dealing with large quantities of samples. The use of matrix solvent to prepare standard solution effectively reduces the influence of matrix effect, and solves the technical problem of low recovery of target substance in flue gas caused by no purification treatment when using gas chromatography-tandem mass spectrometry (GC-MS/MS) to analyze 16 kinds of PAHs. Under the normal working parameters of the instrument, the electron impact ion source (EI) was used under high temperature to improve the ionization efficiency of the targets in complex matrix and to ensure the accurate analysis of targets with low concentrations. The correlation coefficients (R2) of the calibration curves for 16 PAHs are 0.996 4-0.999 8, the limits of dectection and the limits of quantification are 0.29-1.48, 0.95-4.94 ng/cigarette, respectively. Under different concentration levels, the average recovery is 87.71%-123.26%, and the precision is within 1.58%-9.45%. By analyzing the reference cigarette 3R4F, there is no significant difference between this method and those reported in the literature. The total amount of PAHs released from 58 brands of cigarette samples are 742.76-1 505.99 ng/cigarette. At the same time, with the decrease of tar content in cigarette, the release of PAHs will be reduced accordingly. This method is simple, practical, stable, and accurate, providing more technical means for the detection of trace PAHs in cigarette smoke.

3. Whole petroleum system in Jurassic coal measures of Taibei Sag in Tuha Basin, NW China

Based on the latest results of near-source exploration in the Middle and Lower Jurassic of the Tuha Basin, a new understanding of the source rocks, reservoir conditions, and source-reservoir-cap rock combinations of the Jurassic Shuixigou Group in the Taibei Sag is established using the concept of the whole petroleum system, and the coal-measure whole petroleum system is analyzed thoroughly. The results are obtained in three aspects. First, the coal-measure source rocks of the Badaowan Formation and Xishanyao Formation and the argillaceous source rocks of the Sangonghe Formation in the Shuixigou Group exhibit the characteristics of long-term hydrocarbon generation, multiple hydrocarbon generation peaks, and simultaneous oil and gas generation, providing sufficient oil and gas sources for the whole petroleum system in the Jurassic coal-bearing basin. Second, multi-phase shallow braided river delta–shallow lacustrine deposits contribute multiple types of reservoirs, e.g. sandstone, tight sandstone, shale and coal rock, in slope and depression areas, providing effective storage space for the petroleum reservoir formation in coal-measure strata. Third, three phases of hydrocarbon charging and structural evolution, as well as effective configuration of multiple types of reservoirs, result in the sequential accumulation of conventional-unconventional hydrocarbons. From high structural positions to depression, there are conventional structural and structural-lithological reservoirs far from the source, low-saturation structural-lithological reservoirs near the source, and tight sandstone gas, coal rock gas and shale oil accumulations within the source. Typically, the tight sandstone gas and coal rock gas are the key options for further exploration, and the shale oil and gas in the depression area is worth of more attention. The new understanding of the whole petroleum system in the coal measures could further enrich and improve the geological theory of the whole petroleum system, and provide new ideas for the overall exploration of oil and gas resources in the Tuha Basin.

4. Numerical simulation on the melting behaviors of steel scrap in a ladle with bottom argon blowing

More steel scrap consumption in the iron and steel industry can not only mitigate the shortage of iron ore resources but also greatly lower production costs and carbon emissions to the atmosphere, which is a crucial link in low-carbon metallurgy. Because of the drawbacks of different factors, the amount of steel scrap consumed in the current converter steelmaking process is restricted, and the technology of multipoint adding scrap comes into play. Thus, steel scrap addition into ladles has gradually garnered attention from metallurgical experts, but there are few reports on the three-dimensional melting behavior of steel scrap in a real ladle environment with argon blowing. To examine the melting law of steel scrap in a ladle, in this study, the flow field, temperature distribution, and melting behavior of steel scrap in a 70 t refining ladle at various argon blowing rates with steel scrap addition of different specific surface areas and preheating temperatures were numerically investigated and compared using a mutiphysical mathematical model. The results revealed that the melting rate of steel scrap increases with increasing specific surface area, and bottom argon blowing can accelerate the melting of steel scrap, while the promoting effect gradually decreases with increasing specific area. With bottom argon blowing, the core temperature of steel scrap with specific surface areas of 120, 130.22, and 160.81 m2·m–3 increased by 7.06, 6.51, and 3.73 K·s–1, the melting rate was increased by 0.92, 0.88, and 0.28 cm3·s–1, and the melting time was shortened by 17, 15, and 3 s, respectively, compared with those without bottom blowing. When the initial temperature of steel scrap increases from 300 to 1000 K, the melting rate increases from 2.97 to 3.26 cm2·s–1 and the melting time is shortened by 3 s accordingly. The argon blowing rate significantly influenced the melting rate of steel scrap. When the argon blowing rate increases from 100 to 200 L·min–1, the melting time of steel scrap with specific surface areas of 120, 130.22, and 160.81 m2·m–3 is reduced from 44 to 35 s, 42 to 34 s, and 34 s to 31 s, respectively. Thus, on the premise of smooth production, the melting speed of the steel scrap in the ladle can be significantly increased by increasing the argon blowing rate and adding the slab scrap at higher initial temperatures and specific surface areas. This work offers a reference for developing steel scrap rapid-melting technology for ladles in steel plants.

To study the effect of resting on the quality of frozen dough, the changes in the moisture state of the dough during the resting process (0, 30, 60, 90, 120 min), the moisture distribution of dough and their effects on ice crystal distribution, microstructure, and creep recovery characteristics after dough freezing were investigated. The results showed that the freezable water content in the dough was decreased by 0.96% with the extension of the resting time. The pores and number of ice crystals formed in the dough during the freezing process gradually decreased and the distribution of ice crystals became more and more uniform. The average pore area decreased from 22.65 to 11.80 with the extension of the resting time from 0 min to 120 min, and the number of pores decreased from 22.65 to 11.80 with the extension of the resting time. 7. Research Progress on the Stability of Edible Jiaosu Products

Edible jiaosu is one type of food with many bioactive substances, which is very beneficial to human health. The stability of edible jiaosu is the key and difficult point to the development to the new jiaosu products. Currently, many studies on edible jiaosu have been conducted both domestically and internationally. However, there has been no systematic induction and summary until now. Therefore, based on the existing research results, this article summarizes the detection models and evaluation indicators related to the stability of jiaosu products. It shows that the main models include the predicting shelf life dynamics models. The stability indicators mainly include the sensory evaluation, pH index, the functional evaluation, the content of activity, and the probiotic content, among which the sensory evaluation and the antioxidant activity are the main evaluation indicators for the most stability evaluation. The factors that affect the stability of edible jiaosu products include the production process conditions (breeding strain, and fermentation time), the post-processing and the storage environment. The summary shows that by selecting suitable strains or mixed strains for fermentation, reasonable adjustment of fermentation time, adopting appropriate sterilization and drying methods, other post-processing methods, and using the low temperature, dark, anaerobic conditions are all beneficial to improve the stability of jiaosu products. It is recommended to further explore the selection of advantageous strains, research on post processing techniques, and stability regulations for jiaosu processing in the future. In summary, this paper summarizes the evaluation models and indicators for the stability of edible jiaosu, pointed out that production the process conditions, the post-processing of edible enzyme products, and the storage environment have a significant impact on the stability of edible jiaosu, and proposed measures and suggestions to improve its stability. This work provides a theoretical reference basis and guidance for promoting the stability of edible jiaosu and the development of its related industries.

Objective: To elucidate the main components and mechanism of adjuvant prevention of type 2 diabetes of Citrus peel. Methods: The inhibitory activity of Citrus peel of α-glycosidase was determined in vitro. It were used to predict the material basis and potential mechanism of Citrus peel in the treatment of type 2 diabetes by network pharmacology, and verified the results by molecular docking. Results: The α-glucosidase was inhibited by 13 kinds of Citrus peel and showed a dose-effect relationship in the linear range. It was predicted that the active ingredients naringin, tangeretin and lauric acid might inhibit hyperglycemia through oxidative stress by network pharmacology and molecular docking. Citrus peel treatment of type Ⅱ diabetes was related to PI3K-Akt, Ras, HIF-1 signaling pathway in inflammation and vascular related VEGF signaling pathway. Tangeretin was the main component. Conclusion: Citrus peel had an inhibitory effect on α-glycosidase , which might play a role in the prevention of type 2 diabetes.

9. Development progress and outlook of deep and normal pressure shale gas of SINOPEC

In the past decade, with the improvement of shale gas exploration and development theory and techno-logy, the commercial development of marine shale gas from Ordovician Wufeng to Silurian Longmaxi formations in southern China has been achieved, and the target of shale gas exploration and development is gradually shifting towards shale gas of deep and complex structural areas and new formations. In order to further promote efficient exploration and development of shale gas, a systematic review was conducted on the recent exploration and deve-lopment progress of SINOPEC shale gas and an analysis was conducted on the development trend of shale gas in China. The research results indicate that: ① The high-quality Fuling National Shale Gas Demonstration Zone has been built and a key technology system for three-dimensional development of marine shale gas has been innovatively formed. ② The large-scale and efficient development of atmospheric shale gas in Dongsheng and Baima blocks of Nanchuan has been realized, and four shale gas accumulation and dispersion models have been constructed: anticline, monocline, reverse fault blocking, and residual syncline. A fine zoning and differentiated development model and a low-cost technology system for atmospheric shale gas have been preliminarily formed. ③ The deep large-scale fields of trillions of square meters in southeastern Sichuan basin edge have been preliminarily implemented, an "overpressure rich gas" model of marine deep shale gas has been innovatively formed, and a fracturing technology system of deep shale gas in the deep range of 4 000-4 500 m has been preliminarily formed. ④ Major breakthroughs have been made successively in Jurassic of Yuanba and Puguang, Permian of Hongxing and Puguang, and Cambrian of Jingyan and Qianwei. ⑤ The future breakthrough and development of shale gas cannot be achieved without innovative research on exploration and development theory and technology. The integration of exploration and development, geological engineering, and technology and economy must be adhered to. The complexity of deep, normal-pressured, and geological characteristics of new shale gas formations should be fully understood. In addition, policy support is also required.

10. Genetic Variations of AUF2 Gene and Their Association with Agronomic Traits in Flowering Chinese Cabbage

【Objective】By detecting the natural variation of Auxin Up-regulated F-box protein2 (AUF2) gene and exploring the elite alleles associated with agronomic traits in order to provide theoretical references for molecular marker assisted breeding of flowering Chinese cabbage.【Method】PCR amplification and sequencing was used to detect the natural variations in AUF2 gene among 156 flowering Chinese cabbage accessions. By using the mixed linear model (MLM) of Tassel 5.0 software, the correlation between the variation sites of AUF2 gene and six agronomic traits of flowering Chinese cabbage was analyzed, including plant height, single plant weight, maximum leaf length, maximum leaf width, maximum petiole length and chlorophyll content (SPAD), and the polymorphic sites significantly associated with agronomic traits would be identified. Then, elite alleles and excellent haplotypes were determined.【Result】Through sequencing analysis, a total of 34 mutation sites were detected in the 2 996 bp amplification region of AUF2 gene, including 36 haplotypes. The AUF2 gene showed high nucleotide diversity (π = 0.00503) and haplotype diversity (Hd = 0.780). Among them, 2 InDels and 26 SNPs were in the 1 498 bp upstream non-conding region, and 1 InDel and 5 SNPs were in the 960 bp coding region. No mutation sites were detected in the 538 bp downstream non-conding region. Neutral test analysis showed that Tajima's D value, Fu and Li's D* value, and Fu and Li's F* value of the AUF2 gene were positive, and deviated significantly from neutral evolutionary model. One plausible reason was that flowering Chinese cabbage population was subject to balancing selection. The association analysis showed that 4 SNPs were significantly associated with at least one of the four agronomic traits, including single plant weight, maximum leaf length, maximum leaf width and maximum petiole length. The phenotypic variation of a given trait explained by each associated SNP ranged from 5.04% to 6.52%. The haplotype Hap2, which contained elite alletes of the four SNPs mentioned above, could significantly raise the single plant weight, and remarkably increase the length of the maximum petiole.【Conclusion】There are abundant variations in the AUF2 gene, which has a significant impact on the agronomic traits of flowering Chinese cabbage. These elite alleles and excellent haplotypes of AUF2 gene will benefit the genetic improvement of flowering Chinese cabbage.

11. Effects of Ultrasonic Assisted Freezing on Physicochemical Properties and Molecular Structure of Wheat Starch in Dough

In this study, ultrasonication was applied in the whole process of dough freezing and in the maximum ice crystal formation zone, respectively, to explore the effects of ultrasonic-assisted freezing on the physicochemical and structural properties of wheat starch in dough. The results showed that compared with the frozen samples without ultrasonication, the peak viscosity of wheat starch in the dough decreased by 8.6% and 14.5%, the trough viscosity reduced by 17.1% and 13.7%, and the final viscosity increased by 11.3% and −3.6%, respectively, when ultrasonic-assisted freezing was applied in the maximum ice crystal formation zone and the whole process. The consistency coefficient and shear resistance of wheat starch system in dough were maintained to a certain extent by applying ultrasonic in the maximum ice crystal formation zone and the whole process. The consistency coefficient of starch decreased by 19.0% and 23.3%, and the shear structure resilience increased by 4.0% and 7.2%, respectively. Compared with the frozen samples without ultrasonication, the relative crystallinity of wheat starch in the sonicated dough was reduced, and the short-range ordered structure of starch molecules was reduced to a certain extent. In summary, the application of ultrasonic field refines the ice crystals and slows down the damage to the internal structure of wheat starch in the dough during the freezing process. A comparison between the two ultrasound processes, it can be speculated that the effect of ultrasonic application in the whole process of dough-freezing is significantly better than that in the maximum ice crystal formation zone.

12. Segmentation Method of Stroke Lesions Based on Bidirectional Recurrent U-Net Model

Stroke has a high disability rate and fatality rate. It is of great clinical significance to study the automatic recognition and segmentation of stroke lesions. Convolutional neural network can not make use of time sequence correlation of medical image data, and has the problem of low feature utilization. Therefore, a bidirectional recurrent U-Net (BIRU-Net) model was proposed for segmentation of lesions. First, a recurrent neural network is introduced to replace part of the convolutional layer in U-Net with an improved attention convolutional gate recursive unit (ACGRU), so that the segmentation model is not only suitable for small-scale annotated medical image data sets, but also has the characteristics of long-term memory. Second, a dual-channel fusion training mechanism is adopted to input the forward and reverse slice data of a single view into BIRU-Net at the same time, and realize bidirectional feature fusion in the process of model forward propagation, effectively utilizing the bidirectional dependence of slice sequence. Finally, the segmentation results of each single view are refused to effectively utilize the spatial context information of data. The experimental results of ATLAS data set show that the DSC value of the proposed method reaches 62.58%. Compared with other methods at the present stage, the proposed method can segment the lesion region more accurately.

Background and objective microRNAs play an important role in the development and biological phenotype of lung cancer. The present study was to investigate miR-367-3p level in non-small cell lung cancer (NSCLC) patients and its biological function of NSCLC cells. Methods Twenty-two patients with NSCLC (13 cases of adenocarcinoma and 9 cases of squamous carcinoma) admitted to our hospital and treated by surgery were included. During the operation, cancer tissue, paracancerous tissue and 5 mL peripheral blood were collected. Meanwhile, 22 healthy controls were selected and 5 mL peripheral blood was taken. Real-time PCR was applied to detected the expression of miR-367-3p in cancer tissues, peripheral blood of patients with NSCLC and healthy controls. miR-367-3p was detected in lung cancer cell lines (A549) and normal bronchial epithelial cells (BEAS-2B). The proliferation and invasion ability of A549 cells before and after infection were detected by MTT and Transwell assay after transfection with exogenous miR-367-3p. The downstream target gene of miR-367-3p was analyzed by bioinformatics. Zinc finger E-box binding homeobox 2 (ZEB2) was detected by Real-time PCR and Western blot. The general characteristics of karst collapse in Yunnan plateau is comprehensively influenced and determined by the specific geological environment, karst development, hydrogeology, hydrometeorology and human engineering activities. This article analyses these factors and its mechanism, deepen the understanding on the basic characteristics and development law of karst collapse. The karst collapse in Yunnan Plateau is sporadically distributed, showing "point" characteristics, and mainly develops in karst geological environment units such as karst faulted basin, karst trough valley (depression) land, karst platform or ancient plateau surface.The main reason is that most of the plateau is exposed bedrock mountainous area, with deep rivers, vertical karst zones and deep groundwater.In basin, valley, depression and other areas inhabited by human beings, the overburden of loose soil is thicker, the proportion of cohesive soil layer is higher, and the degree of soil consolidation is higher, except for the marginal zone.Karst development is not uniform, karst caves and tubes are developed, but the underground karst rate is not high in general, and the caves and tubes that cause karst collapse are isolated and scattered.

Network security is an important part of the national security, and network security threat intelligence work has become an important part of the network security protection.Aiming at the problems of power enterprise carrying out network security threat intelligence, such as many sources, many types, wide range, many vulnerability risks, many subordinate departments and personnel, the network security threat intelligence work plan of power enterprise was put forward.The scheme includes five aspects: information source, information research and judgment, information disposal, information platform and information performance.The evaluation method of network security vulnerabilities, information emergency disposal process and information performance evaluation method were puts forward.The preliminary application results show that the proposed scheme has important reference value for power enterprise to improve the emergency response ability of network security information and the level of network security protection ability.

Objective: In this study, Tuber liyuanum was used as the experimental material to optimize and determine the extraction process of polysaccharides, and analyze the monosaccharide components. Methods: On the basis of single factor experiment, L9(34) orthogonal experiment was used to design, optimize and determine the optimum technological conditions for extracting polysaccharides by water extraction and alcohol precipitation, and the monosaccharide components of T. liyuanum were analyzed by 1-phenyl-3-methyl-5-pyridine pre-column derivatization HPLC with oxazolinone. Results: The optimum process conditions of extracting polysaccharides from T. liyuanum were as follows: Extraction temperature was 90 ℃, extraction time was 60 min, and solid-to-liquid ratio was 1:25 g/mL. Under these conditions, the yield of polysaccharides was 10.57% ± 0.31%. The polysaccharides of T. liyuanum were mainly composed of D-glucose and a small amount of D-mannose and D-galactose. And the ratio of its substances was 1:0.023:0.006. Conclusion: The water extraction and alcohol precipitation method could obtain a higher yield of T. liyuanum polysaccharides under the optimal extraction process conditions, which was simple, stable and feasible. The determination of monosaccharides composition in T. liyuanum polysaccharides by pre-column derivatization HPLC was simple, reproducible and accurate, which could provide a theoretical basis for further research on T. liyuanum polysaccharides in pear orchards.

17. Corporate Effect of Vacuum Mixing and Freezing-thawing Treatment on Microstructure and Quality of Quick-frozen Youtiao Dough

In order to investigate the corporate effect of vacuum mixing and freezing-thawing treatment on microstructure and quality of quick-frozen Youtiao dough and fritters, frozen Youtiao dough was prepared under the vacuum mixing (0~0.08 MPa) and freezing-thawing treatment (0~3 times). Combined analytical methods covering multiple instruments (e.g., low field nuclear magnetic resonance, scanning electron microscopy, and fourier infrared spectrum analyzer) were used to investigate the water distribution, microstructure, and secondary structure of treated frozen Youtiao dough. Meanwhile, the oil content, water content, specific volume and textural quality of Youtiao were analyzed. Results showed that freezing-thawing treatment destroyed the structure of the dough, declined the strong bonding water content of dough and α-helical content in gluten (14.80%~13.61%), resulting in the loose and disordered protein structure. However, with the increase of vacuum degree, the strong bonding water content in quick-frozen Youtiao dough gradually increased, and the ordered degree of protein network structure in dough also improved. Furthermore, freezing-thawing treatment could also decrease the volume (1.82 cm3/g~1.47 cm3/g), increase the oil content (15.08%~16.88%) and reduce the textural quality of Youtiao, while vacuum mixing could effectively decrease the oil content (15.08%~12.64%), improve volume (1.82 cm3/g~2.38 cm3/g), reduce hardness and improve other qualities of Youtiao. These results indicated that vacuum mixing could reduce the quality deterioration caused by freezing-thawing treatment by regulating the water distribution and protein structure in quick-frozen Youtiao dough. Therefore, this study can provide theoretical basis and guidance for large-scale production of quick-frozen Youtiao.

Background and objective Malignant pleural effusion is one of the common clinical manifestations of patients with lung adenocarcinoma. Patients with pleural effusion at the initial diagnosis of lung adenocarcinoma usually indicate poor prognosis. Epidermal growth factor receptor (EGFR) mutations mainly occur in patients with lung adenocarcinoma. Patients with different mutant subtypes have different prognosis. The clinical characteristics and prognostic factors of patients with EGFR mutated lung adenocarcinoma of different molecular subtypes combined with pleural effusion at initial diagnosis are still unclear. This study was designed to explore the clinical characteristics and prognostic factors of these patients in order to provide management recommendations for them. Methods A retrospective analysis of the clinical characteristics, treatment, outcomes and progression-free survival (PFS) of first-line treatment in patients with EGFR mutated lung adenocarcinoma combined with pleural effusion at initial diagnosis admitted to Department of Medical Oncology and Radiation Sickness, Peking University Third Hospital from January 2012 to June 2021 was performed. Pearson's chi-square test or Fisher's exact test were performed for comparison between groups. Kaplan-Meier method was performed for survival analysis and Cox proportional risk regression model was performed for multivariate analysis. Results 76 patients met the inclusion criteria in this study. The incidences of EGFR classical mutations 19del, 21L858R and non-classical mutations were 46.0%, 38.2% and 15.8%, respectively among these patients. There was no significant difference between the three mutations in terms of gender, age, presence of dyspnea at presentation, whether other distant metastases were combined, site of pleural effusion, volume of pleural effusion, presence of other combined effusions, tumor-node-metastasis (TNM) stage, presence of other gene mutations, and treatment of pleural effusion (P>0.05). In patients with EGFR classical mutations 19del or 21L858R or non-classical mutations subtype, the proportion of chemotherapy in first-line regimens were 17.1%, 20.7% and 58.3%, respectively (P=0.001); and first-line disease control rates were 94.3%, 75.9% and 50%, respectively (P=0.003); pleural effusion control rates were 94.3%, 79.3% and 66.7%, respectively (P=0.04); PFS were 287 d, 327 d and 55 d, respectively (P=0.001). Background: The literature recommends that reduced dosage of CPT-11 should be applied in patients with UGT1A1 homozygous mutations, but the impact of UGT1A1 heterozygous mutations on the adverse reactions of CPT-11 is still not fully clear. Methods: A total of 107 patients with UGT1A1 heterozygous mutation or wild-type, who were treated with CPT-11 from January 2018 to September 2021 in Peking University Third Hospital, were retrospectively enrolled. The adverse reaction spectra of patients with UGT1A1*6 and UGT1A1*28 mutations were analyzed. Adverse reactions were evaluated according to National Cancer Institute Common Terminology Criteria for Adverse Events (NCI-CTCAE) 5.0. The efficacy was evaluated according to Response Evaluation Criteria in Solid Tumors (RECIST) 1.1. The genotypes of UGT1A1*6 and UGT1A1*28 were detected by digital fluorescence molecular hybridization. Results: There were 43 patients with UGT1A1*6 heterozygous mutation, 26 patients with UGT1A1*28 heterozygous mutation, 8 patients with UGT1A1*6 and UGT1A1*28 double heterozygous mutations, 61 patients with heterozygous mutation at any gene locus of UGT1A1*6 and UGT1A1*28. In order to enrich the functionalities of existing learned multidimensional indexes and improve the efficiency, the dynamic data segmentation algorithm DDSA was proposed, which could preserve the data distribution characteristics.A hybrid spatial index was constructed by combining the QuadTree and Z-order curve (QML).The range query algorithm were designed and KNN query algorithm respectively.The proposed index allowed flexible fast queries and updates with preserving the characteristics of data distribution.Experimental results show that QML optimizes the query efficiency on the premise of achieving rich functionalities, and the time complexity of data update is O(1).Compared with R*-tree, the storage consumption of QML is reduced by about 33%, and the update efficiency is improved by 40%~80% .The query efficiency is similar to the optimal tree Index.

Traditional small module spiral bevel gear milling needs to use a large number of cutting fluid,which will not only cause environmental pollution,but also increase production costs,the new technology of high speed dry cutting does not use cutting fluid,which reduces environmental pollution and improves production efficiency. Based on the theory of bevel cutting,combining the equivalent plane method of oblique cutting with the machining theory of spiral bevel gear,the high speed dry cutting technology of small module spiral bevel gear is studied. Aiming at the characteristics of high speed dry cutting technology of small module spiral bevel gear,the thermal mechanical coupling model of bevel cutting is established,and the influence of different process conditions on cutting force is simulated and analyzed by using Abaqus finite element software. The cemented carbide is selected as the tool material,and the high speed dry cutting technology process of small modulus spiral bevel gear is discussed. The feasibility of this technology is verified by machining test. The research provides a reference basis for the implementation of this technology.

The vulnerability detection model of Android application based on learning lacks semantic features.The extracted features contain noise data unrelated to vulnerabilities, which leads to the false positive of vulnerability detection model.A feature extraction method based on code information slice (CIS) was proposed.Compared with the abstract syntax tree (AST) feature method, the proposed method could extract the variable information directly related to vulnerabilities more accurately and avoid containing too much noise data.It contained semantic information of vulnerabilities.Based on CIS and BI-LSTM with attention mechanism, a context-aware Android application vulnerability detection model VulDGArcher was proposed.For the problem that the Android vulnerability data set was not easy to obtain, a data set containing 41 812 code fragments including the implicit Intent security vulnerability and the bypass PendingIntent permission audit vulnerability was built.There were 16 218 code fragments of vulnerability.On this data set, VulDGArcher’s detection accuracy can reach 96%, which is higher than the deep learning vulnerability detection model based on AST features and APP source code features.

The gear and pinion are generated normally by the method of duplex, which has high processing efficiency, but the meshing quality on their surface is controlled in difficulty, which fundamentally limits the improvement of its transmission performance. For the bad contact zone of spiral bevel gears with duplex processing, the gear machining parameters are modified and optimized by using the TCA technology, thus the contact condition and meshing quality of the gear pair are improved. This can not only ensure the production efficiency of common modulus spiral bevel gear which is machined by duplex method, but also improve the meshing quality of the gear pair.

The analysis method of tooth flank mismatch meshing is studied for the spiral bevel gear of automobile drive axle. Firstly,the mathematical model of tilt cutting method is established,and the cradle angle is derived according to the meshing equation,so the tooth flank equation can be simplified. Secondly,the gear meshing angle is derived based on the equality unit normal vector of the gear meshing mathematical model(TCA),so the TCA solving equations can be simplified. On this basis,the tooth flank mismatch values are calculated by calculating the deviations between the pinion actual tooth flank and pinion reference tooth flank which is fully conjugated with the gear,and the tooth flank Ease off topology is constructed by graphical expression. Compared with the traditional tooth flank meshing analysis method,the proposed method not only reduces the number of equations,but also makes the algorithm more stable. The Ease off topology can visually display the whole tooth flank mesh state and improve the current TCA output.

Aiming at the dual duplex spread blade method for cutting spiral bevel gears, the midpoint of the tooth space of pinion is treated as the reference point for the bilateral modification tooth surfaces. The target tooth surface is constructed to meet with the meshing performance by predetermining of the meshing parameters. The mathematical model and constraint function of the target tooth surface errors based on the optimization of machining tool setting are constructed. The nonlinear constrained optimization function is used to derive the machining parameters of the gear. The optimized method is verified by tooth contact analysis and finite element contact analysis. The simulation results show that the meshing performance meets the expected requirements after the tooth surface modification.

In order to solve the difficulty in climbing stairs for the elderly and disabled, a new type of combined climbing wheelchair with planet wheel track is proposed. The combined structure with planet wheel track is used to achieve the function of stair-climbing and obstacle-surmounting. The wheelchair is driven by a motor, the planet wheel adopts chain drive to climb stairs and drive on the flat ground. Combined with the track device, this structure effectively reduces the vibration of the wheelchair with planet chair and improves the obstacle-surmounting ability of traditional wheelchair. On the basis of comparing the structural characteristics of the existing climbing wheelchair, the scheme of designing a new type wheelchair robot with a combination of with the planet wheel and track is introduced, and its operating principle is analyzed, a mathematical model is established to make a kinematics and dynamics analysis on the stair-climbing process. The results show that, the wheelchairs can safely climb most stairs. The structure design of the wheelchair is reasonable, the combination of climbing stairs and moving on the flat ground is realized.

Immune checkpoint inhibitors (ICIs) are new anticancer agents in recent years, and are monoclonal autoantibodies specifically targeting T cell inhibitory receptors, may enhance the anti-tumor effect by blocking negative regulatory factors that inhibit T cell function. ICIs can generate peculiar immunotoxicity, called immune-related adverse events (irAEs), which can potentially affect many tissues, the most frequent toxic reactions to endocrine system include hypophysitis, thyroid dysfunction, adrenal insufficiency, and autoimmune diabetes mellitus. Objective To study the relationship between the level of serum triglyceride (TG) and the early onset and prognosis of acute pancreatitis (AP). Methods Retrospective analysis of 238 patients with primary AP admitted to the General Hospital of Eastern Theater Command from January 2012 to October 2019 was performed. Serum TG level was measured at 48 h after admission. According to the different TG levels, the patients were divided into normal TG group, mildly-high TG group, moderately-high TG group, severely-high TG group and extremely severely-high TG group. Serum inflammatory makers, organ injury, pancreatic necrosis, prognosis and mortality were observed. Results In the 238 cases, 78 had a normal TG level, 160 had a high-level TG. Objective To explore the role and mechanism of 17-allylamino-17-demethoxygeldanamycin (17-AAG) in neurons apoptosis induced by oxygen-glucose deprivation and recovery (OGD/R). Methods Primary rat neurons were cultivated in vitro, and OGD/R model was reproduced. Neurons were exposed to OGD for 0.5h, 1h and 2h, and then reperfusion for 24h, the effect of OGD/R on neurons apoptosis was detected by TUNEL assay. On OGD/R, neurons were treated with different concentrations of 17-AAG (0.5, 1.0 and 2.0μmol/L), and the effect of 17-AAG on OGD/R treated neurons apoptosis was detected by TUNEL assay. Western blotting was performed to detect the expression of heat shock protein 70 (HSP70). HSP70 interference lentivirus was then constructed. The effect of HSP70 interference on the neurons apoptosis treated with OGD/R and 17-AAG was detected by TUNEL assay. Results OGD/R significantly induced neurons apoptosis, and the rate of neurons apoptosis increased with the increase of OGD time. 17-AAG obviously inhibited the neurons apoptosis induced by OGD/R, and the higher the 17- AAG concentration, the more obvious the inhibition. OGD/R significantly suppressed the expression of HSP70 protein in neurons, and 17-AAG obviously reversed the inhibition of HSP70 protein expression induced by OGD/R. However, HSP70 lentivirus interference markedly reversed the protective effect of 17-AAG on OGD/R treated neurons. Objective To investigate the effects of vagus nerve electrostimulation (VNS) on the brain damage of rat middle cerebral artery occlusion (MCAO)/reperfusion model and its mechanism. Methods Twenty four adult male SD rats were randomly divided into two groups (12 each): MCAO/reperfusion group (MCAO group) and MCAO/reperfusion+VNS group (MCAO+VNS group). Subsequently, the neurological function deficit was determined by neurological scoring according to Zea Long scoring method 24h after MCAO/reperfusion. The cerebral infarction volume was determined by TTC assay. The cell apoptosis in brain damage zone was determined by TUNEL assay. Then, the effect of VNS on cAMP response element binding protein (CREB) and p-CREB protein expression was determined by Western blotting. The effect of VNS on Bcl-2 and Bax expression was determined by immunohistochemistry assay. Results Compared with MCAO group, VNS significantly inhibited MCAO-induced neurological deficit (P0.05). Conclusion VNS may ameliorate MCAO-induced neurological deficit and decrease brain infarct volume, which may be related to the promotion of p-CREB protein expression level. Objective To investigate the effects of alprostadil on expression of proinflammatory cytokines in patients with severe acute pancreatitis (SAP) and evaluate the clinical efficacy. Methods Seventy-three SAP patients were collected from January 2014 to May 2016, and then were randomly divided into control group (n=37) and experimental group (n=36). On the basis of routine treatment, the experimental group patient was given alprostadil at a dose of 15µg/d. The expression of C-reactive protein (CRP), white blood cell (WBC) count, amylase, alanine aminotransferase (ALT), aspartate aminotransferase (AST), creatinine, serum proinflammatory cytokine tumor necrosis factor alpha (TNF-α), interleukin -1 beta (IL-1β), interleukin -6 (IL-6) were detected in serum on the 1st, 3rd and 7th day. Results The biochemical indexes and expression of proinflammatory cytokines were significantly increased in the two groups on the 1st day, and decreased gradually, with a significant difference between the time points (P0.05). Objective To explore the relationship between lipid accumulation index (LAI) and hyperuricemia in females. Methods One hundred and seventy-four adult females were included in the cross-sectional study from Mar. to Sep. 2006 through a simple random sampling from a group of population subjected to routine physical examination in the Luohuang Community of Chongqing municipality. The anthropometric measurements, blood pressure (BP), uric acid (UA), lipid profile, glucose and insulin levels were measured. Lipid accumulation product (LAP), body mass index (BMI) and the homeostasis model assessment of insulin resistance (HOMA-IR) were calculated. Subjects were divided into normal group and hyperuricemia group based on their UA levels, and an independent-samples t test was performed for comparison between the two groups; one-factor analysis of variance was performed for multiple comparisons. The Student-Newman-Keuls test was performed for homogeneity of variance. The relationship between UA and other variables was analyzed by Pearson correlation analysis and multiple linear stepwise regression analysis. Logistic regression analysis was used to determine the odds ratio of hyperuricemia in the highest quartile of LAP level. The analysis of the areas under the receiver operating characteristic (ROC) curves was performed to identify the predictive ability of relevant variables to hyperuricemia. 38. Preliminary study on the efficacy and safety of atorvastatin in northern Han patients with acute ischemic cerebrovascular disease

Objective Depending on the stratification of risk factors, to observe the efficacy and safety of different doses of atorvastatin in patients of northern Han population with acute ischemic cerebrovascular disease. Methods One hundred and sixty patients with acute ischemic cerebrovascular disease, admitted from Oct. 2013 to Jan. 2014, were involved in present study, and they were divided into three groups according to the etiology and pathogenesis. In addition to routine treatment, three groups of patients were given 20mg (n=50), 40mg (n=50) and 60mg (n=60) atorvastatin, respectively. Lipids contents, liver function, renal function, muscle enzymes, high sensitivity C reactive protein (hCRP) levels, and NIH Stroke scale (NIHSS) score were determined respectively before treatment and before discharge from the hospital. Results Blood lipid levels were reduced in all the 3 groups, total cholesterol (TC), high density lipoprotein cholesterol-C (HDL-C) and low density lipoprotein cholesterol-C (LDL-C) lowered obviously with significant difference among the 3 groups (P0.05). The highest compliance rate of LDL-C was observed in 60mg group. Compared with those before treatment, the mean levels of alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were elevated, and that of total bilirubin (TBil) and direct bilirubin (DBil) declined after treatment (P0.05). The average level of creatine kinase (CK) was lowered (P0.05). NIHSS scores were decreased in a dose-dependent manner after treatment with no significant difference among the 3 groups (P=0.157). Conclusions Atorvastatin can safely and effectively reduce the blood lipid levels, improve the neurological deficits, and reduce the hCRP level to certain extent. Administration of 60mg of atorvastatin may result in highest compliance rate of LDL-C and the greatest degree of improvement of NIHSS. However, further study should be untaken to demonstrate if the long-term and high-dose medication of atorvastatin is safe and effective for ischemic stroke. DOI: 10.11855/j.issn.0577-7402.2015.07.02

Objective To investigate the correlation between the single nucleotide polymorphisms (SNPs) of rs3811951 at proprotein convertase subtilisin/kexin type 1 (PCSK1) and rs2021785 at PCSK2 gene and the genetic predisposition to newly diagnosed type 2 diabetes (T2DM) in Han population in Chongqing district. Methods A case-control study was performed in Han Chinese subjects with 152 cases of normal control and 227 cases of T2DM patients. Clinical data and blood samples were collected, blood glucose, lipids and other biochemical indexes were determined, and the SNPs of rs3811951 and rs2021785 were genotyped by matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS). Results The minor allele frequency of rs2021785 at PCSK2 was significantly lower in T2DM group than in control group (20.04% vs 26.64%, P=0.0335), and carriers of A allele showed a lower risk of suffering T2DM (OR=0.69, 95%CI 0.49-0.97, P=0.0335). The AA genotype frequency of rs2021785 at PCSK2 was significantly lower in T2DM group than in control group (6.6% vs 10.5%, P0.05). Conclusion SNP of rs2021785 at PCSK2 is significantly associated with T2DM in Han population in Chongqing district, however, there is no significant association between SNP of rs3811951 at PCSK1 and T2DM. Objective To study the distribution of genotypes 5'-AMP-activated protein kinase alpha-2 subunit gene PRKAA2 rs10889007 (301A>T) and rs2746338 (201A>G) polymorphism in Han population in Chongqing, China, and to explore the relationship between these two single nucleotide polymorphisms (SNPs) and type 2 diabetes mellitus (T2DM). Methods Five hundred and twenty-three patients with T2DM (case group) and 312 healthy subjects (control group) from Chongqing were enrolled in the present study. Genotyping was performed for all the subjects enrolled using PCR and restriction fragment length polymorphism (RFLP) to check the two single nucleotide polymorphisms (SNPs) rs10889007 (301A>T) in the intron 1 region and rs2746338 (201A>G) in the intron 2 region of PRKAA2. Linkage disequilibrium (LD) structure and haplotype were examined in two SNPs. Results The genotype frequency of AA, AT and TT of PRKAA2 rs10889007 (301A>T) showed no statistical difference between case group and control group (χ2=3.227, P>0.05); the frequencies of minor allele (T) showed no significant difference between the two groups (χ2=0.887, P>0.05). The genotype frequency of AA, AG and GG of PRKAA2 rs2746338 (201A>G) showed no statistical difference between case group and control group (χ2=3.346, P>0.05); the frequencies of minor allele (G) showed no significant difference between two groups (χ2=0.981, P>0.05). The frequencies of the 4 haplotypes constructed with rs10889007 (301A>T) and rs2746338 (201A>G) of PRKAA2 gene showed no statistical difference between case group and control group (P>0.05). Methods Forty SD rats were randomly divided into normal control group (n=8) and diabetes model group (n=32). Rats in diabetes model group were injected STZ (35 mg/kg). After the diabetes model was reproduced successfully, they were randomly divided into diabetes control group (n=8), diabetes sham-operation group (n=8) and diabetes operation group (n=14). Roux-en-Y gastric bypass was performed in rats of diabetes operation group. The fasting blood glucose (FBG), renal hypertrophy index (kidney weight/body weight), blood urea nitrogen (BUN), serum creatinine (Cr), activity of total superoxide dismutase (TSOD) and glutathione peroxidase (GSH-PX), content of malondialdehyde (MDA), activity of aldose reductase (AR) and AR mRNA expression in the kidney were determined 8 weeks after operation. Objective To investigate the effect of liraglutide on serum advanced glycosylated end products (AGEs), inflammatory factors and glycometabolism in OLETF rats with impaired glucose tolerance (IGT). Methods Thirty-two 12-weekold OLETF rats were randomly assigned into 4 groups (8 each) and received intraperitoneal injection of saline (OLETF-saline group) or liraglutide 50, 100 or 200μg/kg twice a day for 12 weeks (liraglutide intervention groups). Eight LETO rats served as the normal control group and received saline injection. Twelve weeks after the treatment, all the rats were examined for fasting and 2h postprandial plasma glucose after Oral Glucose Tolerance Test (OGTT), and serum AGEs and inflammatory factors were determined by enzyme linked immunosorbent assay (ELISA). Results Before liraglutide intervention, the levels of fasting plasma glucose (FPG), plasma glucose 2 hours after OGTT (2hPG), and fasting plasma insulin (FPI) were significantly higher in IGT-OLETF rats than in LETO rats. 44. Expression and significance of SOCS3 in liver tissue of rats with severe acute pancreatitis complicated by liver injury

Objective To investigate the expression and mechanism of action of suppressor of cytokine signaling 3 (SOCS3) in liver tissue of rats with experimental severe acute pancreatitis (SAP) concurring with liver injury. Methods The rat model of SAP was reproduced by retrograde injection of 4% sodium taurocholate into the biliopancreatic duct. Thirty-two male SD rats were randomly assigned into 4 groups (8 each): normal control group (NC), SAP 6h, 12h, and 18h groups. The levels of serum amylase (AMY), alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were measured dynamically. The concentrations of IL -6 and IL -18 were determined by ELISA. The localization and expression of SOCS3 protein in liver were determined by immunohistochemical staining and Western blotting. Results Compared with NC group, the serum levels of AMY, ALT and AST increased significantly in SAP groups (P < 0.05), and there was significant difference among SAP groups. The serum concentrations of IL-6 and IL-18 increased significantly in the SAP groups than in NC group (P < 0.05), and there was significant difference among SAP groups. Compared with NC group, the concentration of SOCS3 protein increased significantly in SAP groups, and increased gradually along with the increased duration of pancreatitis (P < 0.05). A minor expression of SOCS3 protein was found in NC group. The change in SOCS3 protein concentration was consistent with the severity of liver injury as well as the serum concentrations of IL-6 and IL-18. Conclusions The inflammatory action induced by SAP concurring with liver injury may induce the expression of SOCS3 in liver tissue, and it may increase in intensity along with the severity of liver injury and inflammatory reaction. The mechanism may be attributed to a negative feedback regulation of the inflammatory action mediated by JAK/STAT pathway.

Objective To further localize 5' distal enhancer-like region of the Toll-like receptor 4 (TLR4) gene, and identify the trans-acting factors involved in its transcriptional regulation. Methods The molecular cloning technique and reporter gene analysis were used to analyze the effects of different regions of TLR4 gene 5' flank on its transcriptional activity. The pGL-3 vectors with TLR4 gene 5' flank -1337 to -683bp from transcriptional start site and their deletants were constructed and transferred into K562 cell line by liposomal transfection method. The instant luciferase expression of different clones was detected and the effects of the deletants on the transcriptional activity of TLR4 gene were evaluated. The electrophoretic mobility shift analysis and transcription factor database search were used to locate and identify the trans-acting factors. Results A 245 bp enhancer-like region was further identified in the range of -923 to -679bp from the TLR4 gene transcriptional start site, and -923 to -742 and -721 to -679 regions were refined as two functional units. The activator protein 1 (AP1) was confirmed as a trans-acting factor in the -721 to -679 functional units, and lymphoid enhancer-binding factor (LEF-1) may be another one. Conclusion AP1 acts as a trans-acting factor interacting with -923 to -679 region of TLR4 5' flank, and plays an enhancer-like role together with LEF-1.

Objective To evaluate the expression of estrogen receptor (ER), progesterone receptor (PR) and human epidermis growth-factor receptor 2(HER-2), to determine the phenotype transformation of these receptors before and after recurrence and/or metastasis, and to explore the effects of expression and phenotype transformation of receptors on the treatment efficacy and clinical prognosis of patients with breast cancer. Methods Based on the phenotype transformation of ER, PR, and HER-2 receptor, 211 breast cancer patients were assigned to 3 groups. Twenty patients of Group A were with primary triple-negative breast cancer (TNBC, defined as lacking expression of ER, PR and HER2) which transformed into non-TNBC after recurrence and/ or metastasis, 73 of Group B were with primary non-TNBC which transformed into TNBC after recurrence and/or metastasis, and 118 of Group C were with primary TNBC which was still TNBC after recurrence and/or metastasis. The phenotype transformation of receptors, recurrence/metastasis, and efficacy and clinical prognosis were analyzed following collection of general information of the patients. Results The median age of 211 recurrent patients was 52 years (range, 22 to 78 years). Most of the patients exhibited solitary metastasis. The most common locations of the initial metastasis were lymph node, bone and skin. The median disease-free survival for Groups A, B, and C was 34.0, 25.0, and 20.0 months, respectively. The clinical effect of Groups B and C was better than that of Group A for first-line, second-line, and third-line rescuing therapy (P=0.030, 0.003, 0.001). However, the clinical benefit rate of Group A was higher than those of Groups B and C for rescuing endocrine therapy. The median follow-up time of the 211 patients was 68 months (range, 20 to 127 months), and the median survival after recurrence for Groups A, B, and C was 63.1, 33.7, and 25.8 months respectively (P=0.000). The median overall survival for Groups A, B, and C was 156.7, 67.8, and 47.4 months respectively (P=0.000). Conclusions The phenotype transformation of ER, PR and HER-2 may influence the clinical prognosis of patients with breast cancer. Therefore, the expression of ER, PR and HER-2 should be determined to ensure the efficacy of rescuing therapy.

47. Prospective study on the effect of omega-3 fish oil emulsion on inflammatory responses, immune function, and prognosis in patients with liver transplantation

Objective To investigate the effect of omega-3 fish oil emulsion on the inflammatory responses, immune function, and prognosis of patients after liver transplantation. Methods Thirty-six patients undergoing orthotopic liver transplantation at the Liver Transplantion Center of No. 81 Hospital of PLA from January 2009 to December 2010 were enrolled in this study. The patients were randomly divided into two study groups: the fish-oil group, in which patients were given omega-3 fish oil emulsion (n=18), and the control group (n=18). All the patients received isocaloric and isonitrogenous nutrition for six days, starting on the day after the operation. Blood samples were obtained pre-operatively and on the 2nd, 5th, and 8th days post-operation to measure cytokines (IL-1β, IL-2, IL-6, IL-10, TNF-α), C-reactive protein (CRP), and lymphocyte subpopulation (CD4, CD8, HLA-DR). The incidence of infection, and acute rejection, mortality, length of ICU stay, and postoperative length of hospital stay were also recorded. Results The cytokine levels after liver transplantation in all the patients were higher than those before operation. The IL-1β, IL-2, IL-6, and TNF-αlevels in the fish-oil group were significantly lower than those in the control group (PPPPP+ lymphocyte count in the fish-oil group was significantly lower than that in the control group on POD8 (PP=0.035). Moreover, the incidence rate of acute rejection and mortality showed no statistical difference between the two groups. However, patients in the fish-oil group had significantly shorter postoperative ICU and hospital stays (3.9±1.9d, 28.6±7.2d, respectively) than the control group (5.8±3.1d and 35.7±10.6d; P=0.034, P=0.024, respectively). Conclusion The administration of omega-3 fish oil emulsion to liver transplant patients can reduce excessive inflammatory responses, maintain the balance of pro-/antiinflammatory cytokines, modulate the immune function, lower the incidence of infection, and improve prognosis.

Objective To investigate the correlation between single nucleotide polymorphisms(SNPs) of rs7903146 and rs11196218 at TCF7L2 gene and the early phase insulin secretion of newly diagnosed patients with type 2 diabetes mellitus(T2DM).Methods A case-control study was conducted in 227 newly diagnosed patients with T2DM(T2DM group) and 152 normal people(control group) from Chongqing area.SNPs of rs7903146 and rs11196218 were genotyped by matrix assisted laser desorption/ionization time-of-flight mass spectrometry(MALDI-TOF MS).Results The minor allele frequencies(MAF) of rs7903146 in T2DM and control group were 5.73% and 4.28%,respectively,with no significant difference.Logistic regression analysis revealed that the SNPs of rs7903146 and rs11196218 were negatively associated with T2DM no matter with additive,dominant or recessive models(P > 0.05).The SNPs of rs7903146 were associated with ΔI30/ΔG30(OR=1.012,95%CI 1.000-1.025,P=0.05),the levels of ΔI30/ΔG30 in T-allele carriers were significantly higher than that in normal allele carriers(P=0.0385).Conclusion There is no significant correlation between SNPs of rs7903146 and rs11196218 at TCF7L2 gene and newly diagnosed T2DM,but SNPs of rs7903146 may associate with the early phase insulin secretion.

Objective To investigate the relationship between the occurrence of fatigue as a result of military operation and sleep quality of recruits during period of training. Methods The Modified Fatigue Rating Scale (MFIS) and Pittsburgh Sleep Quality Index (PSQI) were used to conduct the survey of over 500 recruits undergoing basic military training. The aim of the survey was to analyze the relationship between their sleep quality and the occurrence of military fatigue. Results The mean fatigue score of the 500 recruits was 23.55. The physical fatigue score was 12.52, which was obviously higher than that of mental fatigue, which was 11.02 (PPConclusions Poor sleep quality of the recruits may be one of causes of fatigue occurrence, and the aggravation of fatigue can affect sleep quality. Therefore, the management and regulation of sleep quality of the recruits should be emphasized in order to reduce military fatigue.

Objective To investigate the mechanism of action of JAK/STAT signaling pathways in the lung injury of experimental severe acute pancreatitis(SAP).Methods The rat model of SAP was reproduced by retrograde injection of 4% sodium taurocholate into the biliopancreatic duct.Thirty-two male SD rats were randomly assigned into 4 groups(8 each): normal control group(NC),SAP 6h,12h and 18h groups.The level of serum amylase(AMY) was measured dynamically.The pathological changes in pancreas and lung were observed under the light microscope,and the wet/dry weight ratios of lung were evaluated.The concentrations of IL-6 and IL-18 were determined by ELISA,and the expressions of JAK2 and STAT3 protein in lung were determined by Western blotting.Results Compared with NC group,serum level of AMY increased significantly in SAP groups(P < 0.01),and the pancreas and lung injuries under the light microscope were gradually aggravated with disease progression.The wet/dry weight ratio of lung in SAP groups increased significantly compared with that in NC group(P < 0.01).The serum concentrations of IL-6,IL-18 in the SAP groups increased significantly compared with those in NC group(P < 0.01).The concentrations of JAK2 and STAT3 protein in SAP groups increased significantly compared with that in NC group(P < 0.01) and reached the peak in 12-18 hours.Conclusion Activation of JAK2/STAT3 signaling pathways may induce the over-expressions of IL-6 and IL-18,and aggravate the inflammatory reaction and lung injury induced by SAP.
