Abstract [Objective] Ornamental crabapple is an excellent spring flowering tree species. Studying its flowering phenology characteristics and stability and change rules, and exploring its relationship with climate change are of great significance for the prediction and regulation of the flowering period of crabapple. [Methods] Based on the phenological observation data of 67 varieties for 8 years, the flowering date, length, and stability of crabapple were divided, and the adaptability of each variety to climate change was analyzed in combination with meteorological data. [Results] (1) The stability of the initial flowering time of the crabapple variety group was high, and the fluctuation range of the initial flowering time of most varieties (68.7%) was 0-0.04. (2) The flowering life of crabapple was highly sensitive to climate, and only some varieties (18%) had a relatively stable flowering length, with a fluctuation range of 1-2 months. (3) Ornamental crabapple flowering required a certain low temperature stimulation, and different varieties had different low temperature requirements for flowering. (4) When daily maximum temperature ≥25 ℃, daily precipitation≥5 mm, or daily relative humidity ≥80%, the flowering period of crabapple was shortened. [Conclusion] Low temperature before flowering significantly affects the stability of the initial flowering period of crabapple, and maintaining a suitable climate during flowering is of great significance for prolonging the flowering period of crabapple.

Objective To develop a teaching strategy which is suitable for training talents and improving teaching quality in field of clinical gynecological oncology. Methods Seventy-eight clinical students of grade 2020 in a medical university were divided into control group (n=38) and research group (n=40). The “3+2” teaching mode of team-based learning (TBL) and workshop (WS) were adopted, and the learning outcome was evaluated at preview preparation, knowledge application and the questionnaire survey of students, so as to promote the cultivation of clinical reasoning of medical students. Results Compared with traditional teaching method, TBL+WS teaching group had better academic performance.The difference of before class tests and final exams was more significant [17.53±6.15 points and (76.81±5.10)points, respectively, P＜0.001].However, among the eight dimensions of the classroom teaching questionnaire, the teaching quality dimension was the highest (97.5%). More students think that this model had a positive effect on cultivating clinical thinking and developing new knowledge. Conclusions This integrated teaching strategy improves the quality of obstetrics and gynecology teaching and suppots students' capacity building on clinical reasoning .

Objective To explore the effect of paclitaxel with carboplatin combined chemotherapy on hemocyte related indexes,vascular endothelial growth factor (VEGF) and tumor markers in ovarian cancer (OC) patients. Methods OC patients combined chemotherapy (CC) group and ovarian benign tumor control group with 50 cases in each. The pathological changes in the ovarian cells were observed by HE staining and the expression of VEGF was observed by immunohistochemical staining (IHC).Blood cell was counted by flow cytometry (FC). Serum carbohydrate antigen 125 (CA125) and human epididymal protein 4 (HE4) were measured by electrochemical luminescence (ECL). Results Compared with control group, the ovarian tissue structure was disordered and the cell atypia was obvious in OC.VEGF expression was significantly enhanced. Platelet count (PLT), CA125 and HE4 were significantly increased (P＜0.01). However,necrotic cells were observed in ovarian tissue of CC group.VEGF expression was inhibited.PLT count and the level of CA125 and HE4 were significantly lower than those of control group after chemotherapy(P＜0.01). Conclusions Combined chemotherapy may regulate the level of VEGF, CA125 and HE4 in OC patients.

This study aims to compare the prostate cancer detection rate between magnetic resonance imaging (MRI)-transrectal ultrasound (TRUS) cognitive fusion targeted biopsy and systematic biopsy. A total of 614 patients who underwent transrectal prostate biopsy during 2016-2018 with multiparametric magnetic resonance imaging (mpMRI) were included. All patients with a PI-RADS V2 score ≥ 3 accepted both targeted biopsy and systematic biopsy, and those with a PI-RADS V2 score ≤ 2 only accepted systematic biopsy. Overall prostate cancer detection rate between the two biopsies was compared. MRI-TRUS cognitive fusion targeted biopsy identified 342 cases (75.7%) of prostate cancer while systematic biopsy identified 358 cases (79.2%). There was no significant difference in the detection rate between the two groups ( χ 2 = 1.621, P = 0.203). Targeted biopsy had significant fewer biopsy cores compared with systematic biopsy, reducing (9.3 ± 0.11) cores ( P < 0.001) in average. Targeted biopsy had about 10.8% ( P < 0.001) more tumor tissues in positive cores compared with systematic biopsy. The results show that both MRI-TRUS cognitive fusion targeted biopsy and systematic biopsy have good detection rate on prostate cancer. Cognitive targeted biopsy may reduce biopsy cores and provide more tumor tissues in positive cores.

Objective: To clone higA gene from genome of Mycobacterium tuberculosis, express higA protein in E.coli and prepare polyclonal antibodies against higA protein of Mycobacterium tuberculosis., Methods: HigA gene was obtained by PCR from the genome of Mycobacterium tuberculosis H37Rv strain and inserted into E.coli plasmid pET-32a⁺. The recombinant expression vector pET-32a⁺-higA was then transformed into prokaryotic expression bacterium E.coli BL21, and the recombinant protein higA was expressed in E.coli BL21 under IPTG induction. The protein was identified by SDS-PAGE and Western blotting. Purified protein higA was used to immunize New Zealand rabbits to prepare anti-higA polyclonal antibody. Quality and quantity of the antibody were examined., Results: HigA gene was successfully amplified and inserted into pET-32a.⁺ DNA sequencing confirmed the positive recombinant plasmid pET-32a⁺-higA screened by PCR and restriction enzyme digestion analysis. After induction with IPTG, the recombinant protein higA with 36.63 kDa was successfully expressed in E.coli BL21. Rabbit anti-higA polyclonal antibody with a good specificity was obtained, and the titer was above 1:3200., Conclusion: The higA protein was successfully expressed in E.coli BL21. Rabbit anti-higA antibody with high titer and specificity was prepared.

We have assessed the relationships between immune trait (antibody titers of Sheep red blood cell, SRBC; Avian influenza, AI; Newcastle disease, ND) and varieties of MHC B-LBHII Gene in local chicken breeds (Wenshang Barred chicken, LH; Laiwu Black chicken, LWH; and Jining Bairi chicken, BR). We selected 300 chickens randomly from the three indigenous chicken populations. The variations of MHC B-L BII gene were detected by directly DNA sequencing and polymerase chain reaction-single strand conformation polymorphism (PCR-SSCP). The results indicated that there were about 19-22 nucleotide mutations in the three local breeds, which could affect 16-18 amino acid variations. Another results indicated that there was significantly relationship between seven to eight SNPs of the MHC B-LBII region and some immune traits (P < 0.05 or P < 0.01). Both locus G97A and locus T138A were found in the three species, which were significantly related to the antibodies of SRBC, ND and AI antibody titers (P < 0.05). Among them, the locus G97A was significantly associated with ND antibody titers (P < 0.05) in BR chicken, with SRBC antibody titers (P < 0.05) in LWH chicken, and with H9 antibody titers (P < 0.05) in LH chicken. Furthermore, locus T138A was significantly associated with H9 antibody titers in BR and LH chickens (P < 0.05). All those results suggest relationships among the different varieties of MHC B-LBII and immune traits in the three local breeds.
