2. PSMA and 18F-FDG PET Imaging in the Treatment of Metastatic Prostate Cancer with 223Ra: Initial Application and Experience

Objective To share our experience of the use of prostate specific membrane antigen (PSMA) and/or 18F-fluoroodexyglucose (18F-FDG) positron emission tomography (PET) in the setting of radium-223 dichloride (223Ra) in treatment of bone metastases of prostate cancer in order to broaden the know-ledge of application of 223Ra in China. Methods Retrospective analysis was conducted for the patients with advanced prostate cancer treated with 223Ra in Peking Union Medical College Hospital between September 2021 to January 2023.The treatment outcomes and survival status were recorded.Characteristics of bone scans and PET imaging (including PSMA PET and/or 18F-FDG PET) before and after treatment were summarized. Results Nine patients were enrolled (2 additional patients were excluded for the reason of visceral metastases confirmed by pretreatment PET imaging), among whom 8 patients had metastatic castration resistant prostate cancer and 1 had metastatic hormone-sensitive prostate cancer.In total the 9 patients received 36 doses of 223Ra, and the numbers of patients receiving 6, 5, 4, 2, and 1 doses were 3, 1, 2, 2, and 1, respectively.At the middle or end of treatment, there was 1 case of partial remission, 1 case of stable disease, 6 cases of progressive disease, and 1 case was switched to endocrine therapy after 1 shot of 223Ra treatment so that the efficacy was not evaluated.At the time of the last follow-up (March 15, 2023), among the 9 patients, 4 died (1 due to heart failure and 3 due to disease progression) and 5 survived.For patients who died or with progressive disease, there was an inconsistency between the metastases shown on baseline PET images and bone scans, with the former modality revealing more bone metastases.PET imaging especially PSMA PET was more accurate than bone scan in assessing response and could avoid bone scintillation. Conclusions PSMA PET and/or 18F-FDG PET could help detect visceral metastases and assist in patient screening.The inconsistency between baseline PET images and bone scan images suggests a worse outcome of 223Ra treatment.During the efficacy evaluation process, PSMA and/or 18F-FDG PET (especially PSMA PET) also outperforms bone scan.

Taking the grouting filling treatment project in the goaf of 8712 working face of Huoshizui Coal Mine as the background, based on the theory of mining subsidence and the method of combining borehole wave velocity test and pressure water test, the characteristics of fracture development in overburden strata after fully mechanized caving are expounded. The results show that during the overall movement of the overlying strata after fully mechanized caving mining, the integrity of the rock mass was damaged to varying degrees, and a tensile gap with a horizontal width of about 60 m was formed at the edge of the goaf. The comprehensive treatment technology of “seepage-proofing at the bottom of the reservoir, full-clay filling and grouting in the goaf, and monitoring the movement and deformation of the affected area” has achieved good governance effects.

4. Stability analysis of coal mining subsidence area of underlying fully mechanized top coal caving based on Rhino3D-FLAC3D coupling technology

In order to qualitatively and quantitatively analyze the degree of influence of the water load on the stability of fully mechanized caving coal subsidence area, the underground coal mining subsidence area of Hongyanhe Reservoir was taken as the research object, and a three-dimensional numerical model was quickly constructed through Rhino3D-FLAC3D coupling technology. Combined with field experiments, the development characteristics of overburden cracks in fully mechanized top coal caving mining subsidence area and the influence of water load on the stability of overburden rock in coal subsidence area were studied. The results show that: 8712 fully mechanized top coal caving mining subsidence area water-carrying fissure zone as a whole is “saddle” shape, the maximum development height is 138.24 m; the vertical displacement of the surface of coal mining subsidence area increased by 334.7 mm, the development height of the water fissure zone increases by 3.76 m.

7. Automatic extraction and classification of pole-like objects from vehicle LiDAR point cloud

Aiming at the poor quality of vehicle LiDAR point cloud data and the mutual concealment of various ground objects in urban road scenes, an automatic extraction and classification algorithm for pole-like objects was proposed. Firstly, ground points in point cloud data were removed by improving the mathematical morphology algorithm. According to the morphological characteristics of the pole-like objects, preliminary extraction of pole-like objects was carried out through the longitudinal grid template.Secondly, the extracted suspected pole-like objects were regularized with point cloud data and some noise was removed by statistical analysis. Finally, SVM classification model was trained according to the previously established pole-like object samples to classify the extracted pole-like objects. The experimental results showed that the method could effectively extract the pole-like objects in urban road scenes under the condition of poor data quality, and classified the extracted pole-like objects with high precision.

8. Fast algorithm to search for the minimum number of active S-boxes of block cipher.

- Abstract
To solve the problem of fast calculation of the minimum number of active S-boxes in cryptographic design, the difference and mask propagation of the diffusion layer were investigated, and a method was proposed to compute the difference (resp. mask) pattern distribution table of MDS (maximum distance separable) matrices and binary matrices. A lower bound on the computation complexity of the proposed method was also given. Based on the difference (resp. mask) pattern distribution table of diffusion matrix, a fast algorithm to search for the minimum number of active S-boxes of block cipher was proposed. The proposed algorithm is applied to some SPN (substitution permutation network) block ciphers, and finds the minimum number of active S-boxes for the full round of LED, SKINNY, CRAFT and FIDES. The program control valve of S Zorb lock hopper is used to isolate the high-pressure hydrogen and low-pressure oxygen containing environment. The large temperature difference and pressure difference, frequent switching between hydrogen and oxygen environment, severe operating conditions, and high switching action frequency, it easy to occur such as switch timeout, wear, jamming, internal leakage and other prob-lems. High failure rate, high maintenance cost and high risk have always been the thorny problems affecting the long-term operation of the unit. In this paper, the above problems are deeply analyzed and optimized. Through the reduction of the pressure relief hole of the instrument air, the transformation of the valve structure, the use of two position five-way switching mode and the use of fork cylinder valves, the frequent faults of the program control valves are effectively solved. The shortest service life of the valves is increased from 7 months in the previous cycle to 18 months in this cycle. Most valves have been in normal use for more than 31 months. It effectively prevents the internal leakage of the valve. Save about 6 tons adsorbent every year, and saved more than 1.2 million yuan of adsorbent cost every year. [ABSTRACT FROM AUTHOR]

10. Point Cloud Information Extraction for Streetlights with Vehicle-borne LiDAR

The acquisition of detailed information for the streetlights in a large scene remains a tough task since the streetlights are of great number and types. In this paper, a method is proposed to extract and classify the streetlights, with the aid of prior sample sets on the basis of skeleton-line-buffer discriminant algorithm. First, a model and a priori sample set for streetlights are established according to the expression characteristics of streetlights in vehicle-borne LiDAR point cloud. Secondly, with the theory and method of mathematical morphology, the rod-shaped objects are extracted in vehicle LiDAR point cloud scene, and the candidate streetlights are chosen under the constraint of streetlight model and semantic rules. Then, the candidate samples are selected from the sample sets according to the parameter information and the statistical information obtained from the selected streetlights. Finally, based on the matching algorithm of least squares theory, we select and match the priori samples of streetlights and the candidate streetlights. Based on the double buffer of streetlight skeleton information, we discriminate and analyze the candidate streetlights to achieve the extraction and identification of street lights. Finally, the priori samples of streetlights and the point cloud of the candidate streetlights are matched and screened with the matching algorithm of least square theory; and based on the double buffer of streetlight skeleton information, the candidate streetlights are discriminated and analyzed to achieve the extraction and identification of streetlights. Our experiment shows that the algorithm is efficient and robust for the extraction of detailed information of streetlights. For the streetlights with less occlusion and relatively complete data, the extraction accuracy is 0.952, and for those with serous occlusion, low point cloud density and poor data integrity, the extraction accuracy is 0.780. And the above results validate the robustness of the proposed algorithm for the extraction of intermediate streetlights from large scenes. The detailed information extracted by the algorithm can be used to serve the fine and dynamic management of streetlights in large scenes.

11. Amylase desizing process of cotton fabric for digital printing.

To improve the pretreatment quality of cotton fabric for digital printing, the a-amylase was used to desize cotton fine plain cloth by impregnation method. The effect of desizing temperature, enzyme concentration, I)H value, calcium ion concentration, osmotic concentration on desizing effect was analyzed. The weight loss of the fabric in one bath process of desizing, scouring and bleaching was characterized by subsection method. The results show that the optimum amylase desizing process is enzyme concentration 3 g/L, chlorination calcium 5 g/L, penetrant JFC 2 g/L, treatment solution pH value 6, temperature 90 "C, holding time 30 min, bath ratio 40 : 1.Amylase desizing and scouring-bleaching one bath cotton fabric can almost achieve the same effect as the alkali treated cotton fabric, and the fabric strength is not too much damaged. Pretreated cotton fabrics after amylase desizing and alkali desizing are applied to digital printing respectively, and the cotton fabric after amylase desizing has bright color, pure light, good color fastness to friction, with no significant influence on K/S value. 12. Resistance to chloride erosion of cement matrix composite materials under dry-wet cycling and their micro-structural changes.

In order to further reveal the mechanism of chloride erosion to cement matrix composite materials under dry-wet cycling, the resistance to chloride erosion of two kinds of water to cement ratio of cement concrete, mortar and paste specimens under dry-wet cycling were studied. It is found that dry-wet cycling can promote the intrusion speed of chloride ions and increase its invasion depth and concentration. With the increase of water to cement ratio, the above effect of dry-wet cycling gets more obvious. Furthermore, the incorporation of slag can increase the resistance to chloride erosion of cement matrix composite materials and reduce the invasive effect of dry-wet cycling. Meanwhile, the corresponding micro-structural changes before and after chloride erosion under dry-wet cycling show that Friedel's salt is obvious on the surface of paste samples of cement matrix composite materials and adding slag significantly increases the formation of Friedel's salt in the sample. - Abstract
Objective: To investigate the effect of oxygen therapy with Venturi combined with MR850 heating humidifier on patients without mechanical ventilation after tracheotomy., Methods: Eighty patients (≥ 18 years old) who had undergone tracheotomy and without mechanical ventilation admitted to Fenyang Hospital of Shanxi Province from June 2016 to December 2017 were enrolled, and they were divided into control group and observation group according to random number table method, with 40 patients in each group. The observation group was given Venturi (removed the mask) combined with MR850 device active warm and humid oxygen therapy; the control group was given one-off ordinary flow device and warm and humid exchanger (artificial nose, HME) passive humid oxygen therapy. Body temperature increased by 1 centigrade above basal body temperature, white blood cell count (WBC) increased 2×10
9 /L than the base value, oxygenation index (PaO2 /FiO2 ) < 300 mmHg (1 mmHg = 0.133 kPa), airway mucosal hemorrhage, pulmonary infection and sputum viscosity were observed in the two groups for 5 days after oxygen therapy., Results: Among the 80 patients, there were 46 males and 34 females, with an average age of (67.7±12.2) years. Compared with the control group, the incidence of increased body temperature (5.0% vs. 20.0%), the incidence of increased WBC (7.5% vs. 35.0%), the incidence of low PaO2 /FiO2 (2.5% vs. 7.5%), the incidence of airway mucosal bleeding (5.0 % vs. 15.0%) and the incidence of pulmonary infection (2.5% vs. 10.0%) were significantly decreased in the observation group (all P < 0.01), and the proportion of sputum viscosity I degree of patients was significantly increased (57.5% vs. 12.5%, P < 0.01)., Conclusions: Venturi combined with MR850 device can effectively control airway temperature and humidity, promote sputum dilution and conducive to drainage, reduce pulmonary infection in adults patients without mechanical ventilation after tracheotomy, thereby reducing postoperative complications of tracheotomy.- Published

- Abstract
CAR-T cell therapy that targets surface antigens to kill tumor cells specifically has recently become another cornerstone in tumor immunotherapy. In this study, a lentiviral expression plasmid of CAR targeting human epidermal growth factor receptor 2 (HER2) was constructed by genetic engineering. The recombinant plasmid was co-transfected with other packaging plasmids into HEK293T cells by calcium phosphate precipitation to generate lenti-car, which are CAR lentiviral particles. HER2-specific CAR-T cells were obtained by transducing human peripheral blood mononuclear cells with lenti-car. Their specific inhibitory effects on HER2-positive and HER2-negative tumor cells were analyzed in vitro. The constructed CAR-T cells were specifically activated by HER2-expressing tumor cells as indicated by secretion of IFN-γ and IL-2. The inhibitory rate on HER2-positive SK-OV-3 cell line was (58.47±1.72)%, significantly higher than that on the mock-treated control group (P<0.05). The inhibitory rate on HER2-negative K562 cell lines was (11.74±2.37)%, which was not significantly different from that on the control group (P>0.05). Furthermore, when we transfected a HER2-expressing vector into K562, the inhibitory rate increased to (30.41±7.59)%, which was higher than that on HER2-negative K562 (P<0.05). Thus, the constructed second-generation HER2-specific CAR-T cells specifically suppressed growth of tumor cells overexpressing HER2 protein, suggesting that HER2-specific CAR-T cells might prove useful for immunotherapy of HER2-positive cancer.

Objective: To investigate the value of serum IgA/C3 ratio in the diagnosis of IgA nephropathy and explore its relationship with the clinicopathological features of the patients., Methods: Sixty-six patients with IgA nephropathy, 111 with other glomerular diseases, and 40 healthy control subjects without kidney disease were tested for serum IgA and C3 levels using CRM470 adjusted standardized immune turbidimetric method, and the IgA/C3 ratio was calculated. According to Oxford and Lee's classification criteria, we analyzed the pathological grades of the renal biopsy samples from patients with IgA nephropathy. The ROC curve was used to assess the value of serum IgA and IgA/C3 ratio in predicting IgA nephropathy., Results: Patients with IgA nephropathy had an elevated serum IgA/C3 ratio than those with other glomerular diseases and the control subjects, with an area under the ROC curve of 0.776. An elevated serum IgA/C3 ratio was not found to significantly correlate with the pathological grade of renal biopsy samples in patients with IgA nephropathy., Conclusion: In the absence of renal biopsy findings, serum IgA/C3 ratio can help in the diagnosis of IgA nephropathy.

17. [Clinical analysis of renal cell carcinoma with brain metastases (report of 10 cases)].

Objective: To investigate the optimal treatment and prognostic factors of renal cell carcinoma with brain metastases (RCCBM) by the analysis of clinical features., Methods: The clinical data of 10 patients with RCCBM in our hospital from Jul. 2003 to Aug. 2011 were analyzed retrospectively. The age range of the patients was 48-80 years, and the mean age was 64 years old. Nine patients were male and one patient was female. Six patients had neurological symptoms. Brain metastasis and kidney cancer were found in the same period in 7 patients. Two patients had brain metastases alone, and 8 patients had metastasis in brain and other parts. There were 4 patients had more than 3 brain metastatic foci. Brain metastatic foci located in: frontal lobe in 6 patients, parietal lobe in 2 patients, occipital lobe in 2 patients, cerebellum in 2 patients, temporal lobe in 1 patient, thalamus and basal ganglia in 1 patient., Results: The overall survival (OS) of 10 patients with RCCBM was 0.8 to 62.6 months, with an average OS of 12.7 months. Two patients refused therapy and those average OS is only 1.4 months. Three patients were performed stereotactic radiotherapy (SRS). The tumor volume of 1 patient reduced >25% and 2 patients showed no progression in magnetic resonance images at 2-months after SRS. Two patients accepted sorafenib treatment and the average OS was 16.7 months. Patients with 3 or more brain metastases had an average OS of 1.9 months, and the average OS of those with less than 3 brain metastases was 20 months. Patients with synchronous or metachronous brain metastases of renal cell carcinoma had an average OS of 6.8 or 26.5 months, respectively. Patients with or without neurological symptoms had an average OS of 17.9 or 4.9 months, respectively., Conclusions: For RCCBM patients, SRS could effectively control the local brain metastases while sorafenib could extend the overall survival. With 3 or more metastases, synchronous brain metastases and no neurological symptoms were the adverse prognostic factors of RCCBM.

Objective: To explore clinical characteristics of sinonasal malignant melanoma and curative effect of the combined modality therapy., Method: Clinical data of 6 cases with sinonasal malignant melanoma was retrospectively analyzed. All patients received surgery and postoperative radiotherapy. In addition, 3 cases received postoperative chemotherapy which scheme was CDBT and bioimmunotherapy consisted of INF-α and IL-2 after surgery, of which, 2 cases received one cycle of preoperative chemotherapy., Result: Six cases were followed up. The survival time ranged from 15 months to 98 months. The average survival time was 62.7 months. Analyzed by direct method, the 1-year, 3-year and 5-year survival rates were 100%, 83% and 67% respectively. Three cases which received the combined modality therapy, of whch, 2 cases received preoperative chemotherapy have survived by now., Conclusion: The combined modality therapy should be adopted in case of sinonasal malignant melanoma with operation indication. For the patients who can not be operated recently Postbiopsy, it was beneficial to improve the efficacy of therapy that one cycle of preoperative chemotherapy and bioimmunotherapy should be implemented.

- Abstract
Objective: To explore the relationship between the major allergens of 6 common allergic foods and IgA nephropathy., Methods: A sensitive sandwich enzyme-linked immunosorbent assay (ELISA) was used to detect the serum levels of food-specific IgA1, IgG and IgE in 31 patients with IgA nephropathy and 80 healthy volunteers. All the patients were examined for a history of food allergy using a questionnaire., Results: Serum levels of IgA1 and IgG against the major allergens of the 6 common allergic foods were significantly higher in patients with IgA nephropathy than in healthy volunteers (P<0.05). There was no detectable food-specific IgE antibodies in the two groups. No patients had a clear history of food allergy. All the patients with increased IgG levels specific to 4 or more foods simultaneously had proteinuria., Conclusions: Some foods especially the highly allergic ones may participate in the pathogenesis and progression of IgA nephropathy.

- Abstract
Objective: To prepare anti-human retinoblastoma (Rb) monoclonal antibody (McAb) by immunization with solid tumor cells and preliminarily study the characteristics of its antigen., Methods: Three Balb/C mice were immunized by intraperitoneal injection of Rb solid tumor cells acquired from enucleation. Spleen lymphocytes were separated from them and fused with myeloma cell line SP2/0. The anti-human Rb McAb was selected by Rb solid tumor cell, SO-Rb(50), SO-Rb(70), etc as antigens. The characteristics of its antigen were preliminarily studied by immunofluorescence, immunohistochemistry and Western blot., Results: After repeatedly cloning them with micro-manipulation equipment, we successfully established 3 hybridoma cell lines secreting anti-human Rb McAb. Of all, 3C6 hybridoma cells could steadily secrete anti-human Rb McAb after they were frozen, resuscitated and passaged repeatedly for 2 years. Its subgroup was IgG(1). Both immunofluorescence and immunohistochemistry examinations demonstrated that the corresponding antigen of McAb 3C6 was specifically and highly expressed in Rb tumor cell membrane and cytoplasm. The other tumor tissues and human normal eye tissues were negative. Western blot analysis preliminarily demonstrated that McAb 3C6 could bind 25 000 protein band of Rb antigen., Conclusions: The anti-human Rb McAb 3C6 established by immunization of solid tumor cells is specifically and highly expressed in Rb tumor cells and has no cross-response with other tumor tissues and normal eyeballs. The antigen molecular weight bounded to this antibody is 25 000 or so, which further shows that it is a possibility for a new unidentified gene to be concerned with the tumor formation of Rb.
