Objective To analyze the incidence trend of cancer from 2015 to 2020 in Sichuan Province and provide a reference for prevention and control of cancer. Methods Based on the cancer-related case information data on the Homepage of medical records in Sichuan Province from 2015 to 2020, the crude incidence rate was calculated, and the average annual change percentage (AAPC) of incidence rates was analyzed with Joinpoint 4.2.0.1. Results The new cancer cases in Sichuan Province increased from 181 490 in 2015 to 276 770 in 2020, and the incidence rate increased from 198. 57/105 in 2015 to 304.76/105 in 2020. From 2015 to 2020, the average annual percentage change (AAPC) was 7.514%, 6.751%, 8.611%, 4.713%, and 6.838%, respectively. The incidence of cancer in the Chengdu Plain Economic Zone and the Northwest Sichuan Ecological Economic Zone showed a rising trend, AAPC values with statistical significancebeing 7.549% and 9. 668%, respectively. From 2015 to 2020, the incidence rate of lung cancer, colorectal cancer, breast cancer, cervical cancer and prostate cancer in Sichuan Province showeda rising trend, with AAPC values of 11.220% (95% CI; 5.866-16.845), 6.606% (95% CI: l.395-13.202), 6.322% (95% CI: 1.411-11.472), 5.765% (95% CI: l.431-10.284) and 13. 222% (95% CI : 7.396-19.364), respectively. Conclusion The incidence of cancer in Sichuan Province shows a rising trend, and the prevention and control situation is grim. Lung cancer, digestive system cancer, breast cancer, cervical cancer and prostate cancer are the dominant cancer types in Sichuan Province, and specific prevention, control and screening should be strengthened. [Objectives] To investigate the effects of temperature and humidity on the population dynamics of Grapholita molesta, a pest that causes significant losses to pear crops worldwide. [Methods] A monitoring site was set up in a pear orchard in Daxing, Beijing municipality where sex pheromone traps were used to monitor the population dynamics of adult G. molesta over three years (2018-2021). The data obtained were combined with local meteorological data collected over the same period to analyze the relationship between temperature, humidity and population dynamics. [Results] G. molesta began to appear in the pear orchard in mid to late March. Four population peaks occurred each year; in mid to late April, mid-June, mid to late July and early to mid-September. There was a significant relationship between the average and maximum temperature and population density, which can be described by the equation Y= - 5.62 - 0.16X1+0.72X2+0.52X3+0.18X4, where X3 is the highest temperature, X1 is the average temperature, X2 is the lowest temperature and X4 is the average relative humidity. The relative importance of these factors was: X3> X1> X2> X4. [Conclusion] There is a significant relationship between the occurrence of G. molesta in pear orchards in Daxing, Beijing municipality, and local temperature and humidity data. Three kinds of potted one-year-old pear rootstocks (Pyrus calleryana, P. pashia, and P. xerophila) and 15N trace technique were used to examine the effects of different phosphorus application rates (P0, P1, P2, P3 and P4 equivalent to 0,50, 100, 150 and 200 kg ⋅ hm-2 P2O5, respectively) on plant growth and the characteristics of N -urea absorption and utilization. The results showed that, with the increases of phosphorus levels, plant height, ground diameter, dry mass, root surface area, root length, number of root tips, root activity, root respiration rate, Ndff values, and nitrogen use efficiency of rootstocks first increased and then decreased. However, the range of rise and fall of different rootstocks were distinct, and each index reached the highest level at different phosphorus levels. The plant height, diameter, dry mass of P. xerophila were the highest under the same phosphorus level, followed by P. pashia, and P. calleryana was the lowest. The root architecture parameters and root respiration rate showed the same trend, but Ndff values and nitrogen use efficiency performed different. Under different phosphorus levels, each index of P. xerophila reached the highest level at P3, but those of P. pashia and P. calleryana appeared at P2 and P1 rates, respectively. The Ndff values in stem were the highest among different organs of rootstocks at different phosphorus levels, and the highest nitrogen use efficiency of P. xerophila, P. pashia and P. calleryana was 9.6%, 8.9% and 8.3%, respectively. The variations of plant growth and N absorption and utilization of different pear rootstocks across different phosphorus levels indicated that phosphorus fertilizer should be carry out reasonably in practice and give full consideration of phosphorus demands of plants. To explore the effects of different fulvic acid application rates on 15N-urea absorption, utilization, residue amount, loss and fruit yield and quality, fifteen-year-old ' Red General' / Malus micromalus Makino was selected as experiment material by using the 15N-labeled tracer technique. Five treatments were established: single urea (CK), combination of urea and fulvic acid with varied rates (NF1, NF2, NF3 and NF4 represented 75, 150, 300 and 450 kg ⋅ hm-2 fulvic acid application rates, respectively). The main results were as follows: the 15N derived from fertilizer (Ndff) in root, one-year-old branch and leaf were displayed with an order of NF3>NF4>NF2>NF1 >CK at the fruit maturity stage, and significant difference was observed in Ndff value among the five treatments. The total N content and 15N absorption amount were highest in NF3, followed by NF4, and lowest in CK. Compared with CK, the 15N utilization rate in NF1, NF2, NF3 and NF4 was increased by 14.2%, 33.5%, 64.2% and 50.0%, and the 15N loss rate was decreased by 9.1%, 18.5%, 37.1% and 28.7% respectively. The residue amount of N -urea varied across different treatments, and that in 0" 60 cm soil layer under combination of urea and fulvic acid was significantly higher than CK, with the highest value occurred in NF3, while it was lower than CK in 60-100 cm soil layer. In comparison with CK treatment, the largest increment of yield per plant and net income appeared in NF3, with an enhancement of 15.8% and 20.2%, respectively. In addition, the fruit hardness, soluble solid and sugar-acid ratio reached the highest value in NF3. Considering the responses of fruit yield and N utilization efficiency, the suitable fulvic acid application rate was 326.41-350.61 kg ⋅ hm-2. [ABSTRACT FROM AUTHOR]

[Objective] The present study was carried out to explore the effects of oligochitosan treatment on sugarcane physiological indices and drought resistant mechanism under drought stress in order to provide references for oligochitosan application in drought resistance of crops. [Method] Sugarcane variety ROC22 was used as tested material. Three treatments viz., mormal watering (CK), keeping drought stress, and spraying 100 mg/L oligochitosan solution at the early elongation stage under drought stress were designed, and the +1 leaves of all treatments were taken on 2, 4, 6, 8 days after spraying to determine the endopeptidase, MDA, soluble sugar, Fv/Fm and ΦPS II. [Result] Under normal watering, the endopeptidase activity, MDA content, soluble sugar content, Fv/ Fm and ΦPS II, and RWC of sugarcane leaves kept at certain level without significant change. With the elongation of drought stress time, the endopeptidase activity, MDA and soluble sugar contents of sugarcane leaves in drought stress and oligochitosan treatments presented increasing trend. While the Fv/Fm and ΦPS II of drought stress treatment, and the relative water content (RWC) of drought stress and oligochitosan treatments showed decreasing trends. The endopeptidase activity of leaves after 2-8 days of oligochitosan treating was higher than that of drought stress treatment to varying degrees, while the MDA content of drought stress on 4-8 days was higher than that of oligochitosan treatment. The soluble sugar and RWC of leaves after 2-6 days of oligochitosan treating were lower than those of drought stress treatment, while they were significantly higher than the drought stress treatment on the 8th days, which were 28.1 mg/g and 66.7%. Meanwhile, the Fv/Fm and ΦPS II of oligochitosan treatment showed increasing trends and reached the highest on the 8th days, which were 1.28 and 1.58 times higher than drought stress treatment. [Conclusion] It concluded that applying oligochitosan could regulate the process of physiological metabolism in sugarcane leaves under drought stres.s, improve adaptablity of sugarcane leaves to drought stress, and enhance drought resistance of sugarcane. Based on the dynamic analysis of insect community structure characteristics, an investigation was conducted on the composition, abundance, and stability of wheat aphids and their major natural enemies in the wheat field adjacent to rape field in Handan City, Hebei Province in 2008-2009. The wheat and rape fields had the similar composition of insect communities. The main pests were aphids, their predatory natural enemies included ladybirds, green lacewings, spiders, and hover flies, and their parasitic natural enemies were wasps. In 2008-2009, the composition of insect communities in the wheat and rape fields had definite changes, but the common features were the lowest abundance of aphids and the highest abundance of predatory and parasitic natural enemies in wheat field adjacent to rape field. The insect community, aphid sub-community, and predatory natural enemy's sub-community had a high stability in the wheat field adjacent to rape field and a low stability far from the rape field, suggesting that planting wheat adjacent to rape could effectively protect and make use of the natural enemies and promote their control ability on wheat aphids in wheat field. The allelochemicals released by Eupatorium adenophorum roots is an important factor inducing the changes of the weed soil environment,which provides favorable conditions for the successful invasion of the weed. By using GC/MS technique,the components and their relative contents of phenolic allelochemicals in aphid-infested and non-infested E. adenophorum root zone soils were analyzed, with unplanted soil as the control. Less difference was observed in the components of phenolic allelochemicals among the test soils,but their relative contents differed significantly. The relative contents of benzoic acid and 4-hydroxybenzoic acid in aphid-infested E. adenophorum root zone soil were significantly higher than the control; whereas that of 4-hydroxylcinnamic acid was in reverse.In the test three soils, the proportions of benzoic acid, 4-hydroxybenzoic acid,and 4-hydroxylcinnamic acid were different. The bacteriostatic test with the three acids and their combinations based on their proportions in test soils showed that at lower concentrations(50-150 mg∙L-1), the three acids had significant inhibitory effects on five kinds of soil-borne pathogens, but their combinations had different inhibitory effects on the five pathogens, with the effects being significantly higher for the combinations with the proportions in non-infested E. adenophorum root zone soil and the control. Based on the investigation of arthropods biodiversity and insect pests controlling effect in the paddy fields of demonstration area of organic rice production in Guangzhou, a comprehensive assessment on the economic, social, and ecological profits and costs of organic rice production in study area was conducted by using analytic hierarchy process (AHP). The results indicated that among all insect pests controlling measures, organic rice planting had the best comprehensive benefits, with a weight value of 0.5355. Ecological cost played a dominant role in comprehensive costs, and that of traditional pesticide-based insect pests control was most cruel, with a weight value of comprehensive costs being 0.6252. From the viewpoint of the ratio of comprehensive profits to comprehensive costs (RPC), organic rice planting was better than any other insect pests controlling modes, whose RPC was 2.4776, being accorded with the requirement of sustainable development of agriculture. This study constructed a nano-drug delivery system, A3@GMH, by co-delivering the stapled anoplin peptide(Ano-3, A3) with the light-harvesting material graphene oxide(GO), and evaluated its oncolytic immunotherapy effect on triple-negative breast cancer(TNBC). A3@GMH was prepared using an emulsion template method and its physicochemical properties were characterized. The in vivo and in vitro photothermal conversion abilities of A3@GMH were investigated using an infrared thermal imager. The oncoly-tic activity of A3@GMH against TNBC 4T1 cells was evaluated through cell counting kit-8(CCK-8), lactate dehydrogenase(LDH) release, live/dead cell staining, and super-resolution microscopy. The targeting properties of A3@GMH on 4T1 cells were assessed using a high-content imaging system and flow cytometry. In vitro and in vivo studies were conducted to investigate the antitumor mechanism of A3@GMH in combination with photothermal therapy(PTT) through inducing immunogenic cell death(ICD) in 4T1 cells. The results showed that the prepared A3@GMH exhibited distinct mesoporous and coated structures with an average particle size of(308.9±7.5) nm and a surface potential of(-6.79±0.58) mV. The encapsulation efficiency and drug loading of A3 were 23.9%±0.6% and 20.5%±0.5%, respectively. A3@GMH demonstrated excellent photothermal conversion ability and biological safety. A3@GMH actively mediated oncolytic features such as 4T1 cell lysis and LDH release, as well as ICD effects, and showed enhanced in vitro antitumor activity when combined with PTT. In vivo, A3@GMH efficiently induced ICD effects with two rounds of PTT, activated the host's antitumor immune response, and effectively suppressed tumor growth in 4T1 tumor-bearing mice, achieving an 88.9% tumor inhibition rate with no apparent toxic side effects. This study suggests that the combination of stapled anoplin peptide and PTT significantly enhances the oncolytic immunotherapy for TNBC and provides a basis for the innovative application of anti-tumor peptides derived from TCM in TNBC treatment.

We examined nitrogen use efficiency of 15 N-urea of 15-year-old 'Gala' apple trees by broadcast fertilization (T 1 ), nitrogen fertilization with drip irrigation (T 2 ) and nitrogen fertilization with subsurface irrigation (T 3 ), to further improve the water and fertilizer integration technology for apple orchard and to improve nitrogen utilization efficiency. The results showed that leaf area, chlorophyll, and leaf nitrogen content of leaves were significantly higher in T 3 treatment than those in T 1 and T 2 treatments. Soil mineral nitrogen content (20-40 cm) in each period followed the order of T 3 >T 2 >T 1 , while that in 0-20 cm followed an order of T 2 >T 3 >T 1 . The Ndff value at organ level (the contribution rate of the 15 N amount absorbed by various organs of the tree to the total nitrogen content of the organ) was the highest in T 3 treatment in each period, followed by T 2 and T 1 treatments. The utilization rate of 15 N in the fruit ripening period followed an order of T 3 >T 2 >T 1 . The 15 N utilization rate of T 3 reached 24.2%, being 1.19 and 1.65 times of T 2 and T 1 , respectively. The 15 N distribution rate in the fruits during the fruit maturity stage was the highest in T 1 treatment, while that in the storage organs was the highest in T 2 treatment and that in the reproduction organs was the highest in T 3 treatment. The single fruit weight, yield, soluble solids, hardness, soluble sugar and sugar-acid ratio were the highest in T 3 treatment, followed by T 2 and T 1 treatments. In summary, nitrogen application by percolation irrigation (subsurface application) significantly promoted leaf growth and nitrogen utilization of apple tree, and improved fruit yield and quality.

To study the effect of ginseng saponin Rh₂ in inducing apoptosis of human leukemia K562 cells, and explore its mechanism from the aspect of autophagy pathway. CCK-8 assay was used to examine the growth inhibition of human leukemia cell lines K562 treated with ginsenoside Rh₂; flow cytometry (FCM) was used to detect cell apoptosis; Hoechst staining was used to observe the changes of cell morphological apoptosis; Acridine and MDC staining were used to detect the effects of the Rh₂ on autophagy; Western blot and RT-PCR were used to detect the expression levels of the proteins closely associated with autophagy and apoptosis. In order to study the effect of autophagy in proliferation and apoptosis, we used the autophagy inhibitor (3-MA).CCK-8 indicated that Rh₂ at low concentration could effectively inhibit the proliferation of leukemia cellsin dose- and time-dependent manners in K562 cells; FCM indicated that Rh₂ induced apoptosis; Hoechest staining showed that K562 cells had typical apoptotic morphological changes by treated Rh₂; Acridine and MDC staining showed that Rh₂ enhanced the green fluorescence and a large number of acidic autophagy vesicles were present; Western blot and RT-PCR results showed that Rh₂ increased the expression levels of Beclin-1, LC3A, LC3B, activated Caspase-3 and p-p38 in K562 cells; application of autophagy inhibitors(3-MA) could weaken the inhibition effect of Rh₂ on proliferation and induction effect on apoptosis in K562 cells. Objective: To study the antalgic and antiphlogistic functions and mechanism of ronggudingtong (RGDT) plaster (traditional Chinese medicine)., Methods: The painful models were established with hot plate test or acetic acid writhing and the inflammatory models were established with daubing dimethylbenzene on auricle or injecting formaldehyde in toe or synovial envelope to study the antalgic and antiphlogistic functions of RGDT Plaster. The total protein and leukotriene B4(LTB4) in inflammatory exudate were detected to investigate the antalgic and antiphlogistic mechanism of RGDT plaster. The mice were randomly divided into different groups (n = 11), on the basis of drug using, the indexes of pain threshold, swelling degree were observed. Sixty-six mice were used to establish gasbag synovitis model and randomly divided into normal control group,model control group, positive control group (Voltaren gel 0.8 mg/d)and low/medium/high dosage RGDT plaster treating groups(30 mg/d, 60 mg/d, 120 mg/d)., Results: 30 mg/d, 60 mg/d,120 mg/d RGDT plaster could upgrade the pain thresholds, remit auricular and foot swelling (P < 0.05, P < 0.01), and degrade total protein and LTB4 in inflammatory exudates (P < 0.05, P < 0.01)., Conclusion: RGDT plaster has some antalgic and antiphlogistic functions, and one of the mechanisms is depressing synthesis of LTB4.

Objective: To evaluate the clinical efficacy of electroacupuncture(EA) for apoplectic urinary incontinence., Methods: Two hundred and four cases of apoplectic urinary incontinence were randomized into an EA group (136 cases) and an indwelling catheter group (68 cases). The EA was applied at Qugu (CV 2), Zhongji (CV 3), Shuidao (ST 28), Qihai (CV 6) and Guanyuan (CV 4), etc. in the EA group,5 times a week. Indwelling catheter was applied in the indwelling catheter group at intervals of 2-4 hours, and periodic bladder irrigation along with bladder rehabilitation training were also given. The efficacies were evaluated after 4 weeks of treatment. Before and after treatment, the urination diary (including the interval of urination, nocturia frequency, urination difficulty, urinary incontinence severity), bladder capacity, patients' satisfaction of the two groups were observed and the efficacy was evaluated., Results: The total effective rate was 96.2% (125/130) in the EA group, which was apparently superior to 87.5% (56/64) in the indwelling catheter group (P < 0.05); except for nocturia frequency in the indwelling catheter group, the total score and the subitem score in the urination diary were all improved significantly after treatment in both groups (all P < 0.001), which were more obvious in the EA group (P < 0.001, P < 0.05); the patients' satisfaction and bladder capacity were all improved significantly after treatment in both groups (all P < 0.001), which were more obvious in the EA group (both P < 0.001)., Conclusion: The EA has an obvious effect for apoplectic urinary incontinence in urinary incontinence alleviation and bladder capacity increase, which has better efficacy than indwelling catheter therapy.

Objective: To analyse the genetic diversity of wild Schisandra sphenanthera in Qinling Mountains., Methods: The genetic diversity of wild S. sphenanthera was investigated by SSR (Simple Sequences Repeats) markers., Results: Nine pair primers generated a total of 61 bands 53 of which were polymorphic, and 6.8 genotypes were detected by each SSR primer pair on an average ranging from 5 to 11. The percentage of polymorphic band was 86.88%. The coefficient of genetic differentiation (G(st) = 0.3524) showed great genetic variation occurred within populations. Gene flow (N(m)) was 1.1850, which showed that gene flow occurred among populations. The highest genetic diversity of five populations was Yangxian population. And cluster analysis was based on genetic similarity coefficient. The five populations of wild S. sphenanthera samples were classified into two large groups, group I included Pingli population, Shanyang population and Ningqiang population; Group II included Taibai population and Yangxian population., Conclusion: The S. sphenanthera in Qinling Mountains, especially the regions of Hanzhong,Baoji and Shangluo in Shaanxi province, reveals high genetic diversity, which benefits the breeding of new varieties. It is necessary to protect the resource and ensure the sustainable development and utilization of it.

Objective: To determine the content of four lignans in the fruits of Schisandra sphenanthera, Schisandra grandiflora, Schisandra rubriflora and Schisandra propinqua subsp. sinensis., Methods: The HPLC separation was performed on Diamonsil C18 (150 mm x 4.6 mm, 5 microm) columm using methanol-water (70 : 30) as the mobile phase at a flow rate of 0.8 mL/min, with the detection wavelength of 280 nm., Results: Schisandrin, deoxyschizandrin and gamma-schizandrin were detected in four species of Schisandra. The content of schisantherin A in S. sphenanthera was measured up to the set standards of the Chinese Pharmacopoeia (2010 Edition). The content of deoxyschizandrin was high in S. grandiflora and S. rubriflora, and that of gamma-schizandrin was high in S. propinqua subsp. sinensis., Conclusion: The content of lignans is different in four species of Schisandra fruits in Qinling Mountains, it provides scientific reference for further study.

Objective: Fabry disease is a rare lysosome storage disease featuring X-linked recessive inheritance. The study was to explore potential mutations of alpha-galactosidase A (GLA) gene and their correlation with clinic manifestations in three Chinese pedigrees with Fabry disease., Methods: All exons and flanking sequences of GLA gene were amplified with PCR. Potential mutations were detected with bidirectional DNA sequencing. Correlation between particular mutations and clinic features were analyzed., Results: A unreported missense mutation, c.797A>C (D266A) in GLA exon 5 was identified in pedigree 1. Also in exon 5, a missense mutation c.644A>G (N215S) was found in pedigree 2. In pedigree 3, a nonsense mutation c.355C>T (Q119X) was found in exon 2. The c.797A>C mutation was not detected in 200 unrelated male controls. The probands of pedigrees 1 and 3 had presented mainly with skin damage and chronic renal insufficiency, whilst the proband of pedigree 2 had presented with hypertrophic cardiomyopathy., Conclusion: The unreported c.797A>C (D266A) mutation is the sixth missense type mutation of the 266th codon of GLA gene, and all other 5 missense mutations reported previously had been confirmed to be responsible for Fabry disease. The c.797A>C mutation, not found in 200 unrelated male controls, may be the causative mutation in pedigree 1. The c.644A>G and c.355C>T mutations were first detected in Chinese patients. Variable phenotypes of Fabry disease may be in part attributed to the natures of particular mutations of GLA gene.

Objective: To analyze the clinical manifestation, clinicopathologic features and alpha-galactosidase A (GLA) gene mutations in a pedigree with Fabry disease., Methods: In this study, we retrospectively collected the clinical data of the members in the pedigree with Fabry disease, then the clinicopathologic features of the male proband were analyzed by renal biopsy, and GLA gene was detected by PCR and direct sequencing., Results: 1) The proband was characterized by pigmentation of bilateral lower extremities, episodes of neuropathic pain, and renal dysfunction. The renal biopsy showed secondary focal segmental glomerulosclerosis with massive foam-cell liked podocytes under light microscope and abundant inclusions in podocytes which were round, comprising concentric layers of dense material separated by clear spaces under electron microscope. 2) The proband was identified to present a missense mutation as CAG119TAG (Q119T). The mother and niece of the proband were the carriers of this missense mutation., Conclusion: We identified a family with Fabry disease resulting from a novel point mutation of GLA gene, which has not been reported before in Chinese population.

Objective: To establish a method to test citrinin (CIT) in monascus products by immuno-affinity chromatography (IAC)-high performance liquid chromatography (HPLC), and to detect the content of CIT in monascus products in Fujian province., Methods: IAC-HPLC was applied to detect the CIT content in monascus products. The conditions to use HPLC were as follows: C(18) reversed-phase chromatographic column, 150.0 mm×4.6mm×3 µm; mobile phase: the volume ratio of acetonitrile and 0.1% phosphoric acid solution at 65:35; isocratic elution; column temperature: 28°C; flow velocity: 0.8 ml/min; fluorescence detector, excitation wavelength (λ(ex)) was 331 nm and emission wavelength (λ(em)) was 500 nm. The standard curved was established by the linear regression of peak area (Y) to CIT content (X, ng/ml). The accuracy and precision of the method would then be verified. And 32 kinds of monascus products were determined and their color values were compared by this method., Result: The standard curve established in this study was Y = 4634.8X-136.42, r = 1.000; whose limits of detection was 20 µg/kg and the limits of qualification was 64 µg/kg. In the range between 200 and 800 µg/kg, the standard recovery rate was 98.9% - 110.0% (n = 3), and the relative standard deviation (RSD) was 0.51% - 1.76%. Out of the 32 samples, CIT was detected from 11 samples of monascus rice, 9 samples of monascus powder and 5 samples of monascus pigments, the content was around 0.212 - 14.500 mg/kg. 4 out of 7 functional monascus samples were detected out CIT, whose content at 0.142 - 0.275 mg/kg., Conclusion: The method to detect CIT in monascus products by IAC-HPLC has been established.

Objective: To observe the gene expression of herpes simplex virus type 1 thymidine kinase (HSVl-tk) in rat malignant ovarian tumor tissues and the therapeutic effect of ganciclovior (GCV) after intra-arterial infusion of HSVl-tk gene therapy mediated by GE7-delivery system., Methods: A GE7-polylysine/pCMV-HSV1-tk/polylysine-HA20 4-element complex was constructed. Eighteen rats with DMBA-induced ovarian tumor were divided into 3 groups as Atk, ANS and Vtk groups. The 4-element complex GE7-polylysine/pCMV-HSV1-tk/polylysine-HA20 was injected via the ovarian artery into the rats of Atk group, saline buffer was injected in the ANS groups, and the 4-element complex was injected via the tail vein into the rats of Vtk group. All rats received intraperitoneal injection of GCV in a dose of 50 mg/kg daily for 10 days. The rats were sacrificed 3 days after the final dose of GCV, and the tumor weight was measured and tumor growth inhibition rate was calculated. Flow cytometry was used to assess the cell cycle and apoptosis., Results: The tumor weight in the rats of Atk group was (4.77 ± 2.31) g, significantly lower than that of ANS group [(14.66 ± 6.26) g, P < 0.01] and Vtk group [(17.53 ± 7.19) g, P < 0.01]. The tumor growth inhibition rate of the Atk group was 67.5%, while that of Vtk group was -19.6%. The flow cytometry showed that S-phase tumor cells in the Atk group were (54.32 ± 9.65)%, significantly higher than that in the ANS (27.43 ± 9.22)% and (30.16 ± 11.57)% in the Vtk group (both P < 0.01). The tumor cell apoptosis rate in the Atk group was (39.15 ± 12.16)%, significantly higher than that in the ANS group [(11.86 ± 5.28)%, P < 0.01] and Vtk group [(14.32 ± 6.43)%, P < 0.01]., Conclusion: HSV1-tk/GCV gene therapy system mediated by GE7 non-viral delivery system via ovarian arterial infusion effectively causes cell cycle arrest at S phase and enhances cell apoptosis, therefore, exerts an inhibitory effect on tumor growth.

Specific primers were designed and synthesized based on the reported EgA31 gene of Echinococcus granulosus (GenBank Accession No. AF067807). Total RNA was extracted from E. granulosus and its EgA31 gene was amplified by reverse transcription-polymerase chain reaction (RT-PCR). The PCR product was purified and cloned into plasmid pUCM-T, then transformed into Escherichia coli DH5alpha. The recombinant plasmids were screened and identified by digestion with restriction enzyme and PCR amplification. The positive recombinant plasmid pUCM-T/EgA31 was confirmed by sequencing and homology comparison. Five parameters and methods were used to predict B-cell epitopes in amino acid sequence of EgA31. The amplified DNA fragment (636 bp) had an identity of 100% with the EgA31 gene sequence of E. granulosus. B-cell and T-cell epitopes of EgA31 were probably at or adjacent to 32-79, 79-95, 105-124 and 141-154 in its amino acid sequence.

Objective: This study was to identify the post-traumatic stress disorder (PTSD) changes and the relative risk factors within one year after Wenchuan earthquake among middle school students in the disaster area., Methods: A total of 1966 students from 3 schools in Wenchuan earthquake region were selected as the target population. For each student, personal basic information and standard psychological scale (PCL-C, PSSS) were investigated by a self-administrated questionnaire in the 3rd, the 6th, the 9th and the 12th month after the earthquake, respectively. PTSD trends over the time and the associated risk factors were analyzed through the establishment of multi-level random coefficient model., Results: There were 1677 middle school students fully participated in the PTSD follow-up study by turning in the valid questionnaires. The averaged scores of PTSD at the time of the 3rd, the 6th, the 9th and the 12th month after the earthquake were 35.14 ± 11.08, 32.90 ± 11.03, 30.67 ± 11.28 and 29.75 ± 11.22, respectively. Meanwhile, the general incidences of PTSD were 36.6% (613/1677), 30.7% (515/1677), 24.8% (416/1677)and 22.2% (373/1677), respectively. The median score of perceived social support system was 60.00 and the general incidences of PSS was 17.20% (289/1677). The PTSD scores for the students had a decreasing trend during the period of our observation (β(time) = -1.879, χ(2) = 47.03, P < 0.05). The averaged scores for boys for the 4 follow-up studies were 33.71, 31.61, 29.66, 28.83; for girls were 36.33, 33.98, 31.51, 30.52; for junior school students were 35.46, 33.28, 30.18, 29.22; for senior school students were 34.89, 32.62, 31.04, 30.15. Moreover, two factors, gender and grade, were related with the decreasing trend (the trend for girls and senior school students was sharper than that for boys and junior school students) (β(gender-time) = -0.354, χ(2) = 4.83, P < 0.05; β(grade-time) = 0.622, χ(2) = 11.30, P < 0.05)., Conclusions: The prevalence of PTSD was high. Meanwhile, there was a trend of self-recovery for adolescent's PTSD during the first year of post-earthquake, but boys and junior school students recovered more slowly.

Objective: To observe the gene and protein expression of herpes simplex virus type I-thymidine kinase (HSV(1)-tk) in the ovarian tumor tissues and other organs after arterial infusion of HSV(1)-tk gene mediated by GE7 delivery system., Methods: GE7-polylysine/pCMV-HSV(1)-tk/polylysine-HA20 complexes were constructed. Nine rats with induced ovarian tumor were divided into 3 groups, injecting the 4-element complexes or saline buffer through the ovarian artery and complexes through the tail vein, respectively. The ovarian tumors, hearts, livers, spleens, lungs and kidneys were obtained at 72 hours after injection. RT-PCR and Western Blot were preceeded to determine the expression of HSV(1)-tk gene and protein in the tumor tissues and other organs., Results: In the group of arterial injection with 4-element complexes, the HSV(1)-tk gene and protein were expressed strongly in the tumor tissues, while little or none was detected in other organs. In the group of arterial injection with saline buffer, no HSV(1)-tk gene and protein was detected in both tumor tissues and other organs. In the group of tail vein injection, none was detected in tumor tissues and only little was found in the livers, spleens, lungs and kidneys., Conclusion: High target and gene transfer rates can be obtained when HSV(1)-tk gene is transferred via the artery route mediated by GE7 delivery system. HSV(1)-tk protein can be expressed after the gene transfer. The results may provide a new strategy for target killing of HSV(1)-tk/GCV system in ovarian tumors.

Objective: To describe the personal experiences of treating cavernous sinus hemangiomas (CSH) with gamma knife or cyberknife and evaluate the medium and long-term outcomes., Methods: From June 1999 to December 2008, 20 CSH patients were treated by radiosurgery (gamma knife or cyberknife). Among them, 11 had residual tumor after surgery and 9 were diagnosed by neuroradiology. All cases on magnetic resonance imaging (MRI) showed well-defined and homogeneous hyperintensity on T2-weighted images. The mean maximum diameter of tumors was 38.7 ± 9.3 mm (range: 23.0 - 60.6). The volume of tumors ranged between 4.1 - 52.6 cm(3) with a mean of (18.7 ± 12.5) cm(3). The tumor margin dose irradiated by gamma knife ranged from 9 to 16 Gy (mean 12.3 ± 2.3 Gy). The tumor margin dose irradiated by cyberknife was 20 - 21 Gy in 3 fractions. The mean follow-up period was (60 ± 32) months (range: 12 - 120)., Results: Follow-up imaging showed tumor volume decreased over 90% (n = 5), tumor regressed over 50% in volume (n = 9) and no change in tumor volume (n = 1). One patient with large tumor (irradiated 10 Gy) was operated by open surgery at 5 months after gamma knife. Four giant tumors decreased over 50% in volume after cyberknife. Neurologically, none of these patients showed any deterioration. And 16 patients demonstrated an obvious improvement after radiosurgery. At the last follow-up, there were no complications related to radiosurgery., Conclusion: Radiosurgery (Gamma knife or cyberknife) is a safe and effective modality for small- and medium-sized CSH. Fractionated Cyberknife proves to be an effective treatment for large or giant CSH. Thus radiosurgery will be an alternative option to surgery.

Objective: To investigate mental health status among middle school students in Wenchuan earthquake region three months after the earthquake., Methods: 38 classes of 3 middle schools in the disaster region were selected and 1966 students were invited to participate the investigation. The post-traumatic stress disorder (PTSD) checklist-civilian version was used to screen positive PTSD, and the perceived social support scale (PSSS) was used to evaluate the level of perceived social support., Results: 1958 valid questionnaires were selected. The general positive rate of PTSD was 78.29% (1533/1958). PTSD prevalence of the girls was 82.30%(851/1034), higher than that of the boys (73.92% (683/924), chi(2) = 20.22, P < 0.01); the rate of the minority students was 80.27% (1375/1713), higher than that of the Han students (64.98% (154/237), chi(2) = 28.75, P < 0.01); the rate of the rural students was 83.52% (872/1044), higher than that of the urban students (72.43% (662/914), chi(2) = 35.37, P < 0.01); the rate of those who were injured in the quake was 90.38% (94/104), higher than that of the uninjured (77.66% (1439/1853), chi(2) = 9.40, P < 0.01). The level of perceived social support and family support of the subjects was high while outside family support was average. The medians of the score were 21, 60 and 39, respectively. The severity of PTSD was negatively correlated with perceived social support, and perceived support within and outside family (r values were -0.191, -0.130 and -0.190 respectively, all P values < 0.01)., Conclusion: The incidence rate of PTSD is high among the middle school students in disaster region. The symptoms and severity of PTSD are different in the middle school students with different characteristics. Moreover, the more perceived social support they obtain, the better health status they keep.

Objective: To explore the relationship between post-traumatic stress disorder (PTSD) and perceived social support (PSS) ability among middle school students in earthquake-stricken areas after the Wenchuan earthquake accident., Methods: A total of 1966 students from three secondary schools of Wenchuan earthquake-stricken areas were evaluated by PTSD Checklist-Civilian Version (PCL-C) and Perceived Social Support Scale (PSSS)., Results: The overall incidence rate of PTSD was 78.3%, with severe PTSD as 24.38%. Significant differences on the incidence rates of PTSD were found among the students who were in different PSS levels (P < 0.05) and significant negative correlation existed between the levels of severity on PTSD and PSS (gamma = -0.226, P < 0.05). Significant differences on PTSD incidence rates were found among those students who were from different families or out-family PSS levels (P = 0.009, P < 0.05). Significant negative correlation existed between the severity of PTSD and family or out-family PSS level (gamma = -0.176, P < 0.05, gamma = -0.214, P < 0.05)., Conclusion: Relationships between the incidence rate, severity of PTSD and PSS levels existed among the middle school students in Wenchuan earthquake-stricken areas, with higher PSS, lower incidence rate and lighter severity of PTSD. Psychological intervention for earthquake-stricken students should be carried out.

Objective: To investigate the geographic distribution pattern of patients with anorectal atresia/stenosis in China, in order to provide clue for research on its etiology., Methods: Data were collected from Chinese Birth Defects Monitoring Network (CBDMN), which was a hospital-based congenital malformations registry system. From 2001 to 2005, all fetuses with more than 28 weeks of gestation and neonates up to 7 days of age, were monitored. Two-dimensional graphic cluster method was used to divide monitoring stations into different classes with the incidence rates of anorectal atresia/stenosis., Results: The overall incidence of anorectal atresia/stenosis was 3.17 per 10,000 during 2001 to 2005. The incidence was higher in Eastern than that in Mid or Western parts of China and the difference was statistically significant (z = 2.50, 3.69; P = 0.012, < 0.001). The monitoring stations were grouped into 6 classes. Class I was with Heilongjiang, Jilin and Liaoling; Class II was with Fujian, Guangdong, Hainan, Guangxi, and South Hunan and Jiangxi; Class III was with Beijing, Tianjin, Hebei, Shandong, and North Jiangsu and Anhui; Class IV was with Zhejiang, Shanghai, South Anhui and Jiangsu, North Hunan and Jiangxi, Hubei, Henan, Shanxi and Inner Mongolia, Class V was with Ningxia, Gansu and Qinghai; and Class VI was with Shaanxi, Sichuan, Chongqing, Yunnan, Guizhou, Xinjiang and Tibet., Conclusion: Our findings discovered the geographic distribution patterns of patients with anorectal atresia/stenosis in China. It is important to further analyze the relevant environmental factors attached to it so a better regional monitoring system for anorectal atresia/stenosis can be operated.

Based on the investigation of arthropods biodiversity and insect pests controlling effect in the paddy fields of demonstration area of organic rice production in Guangzhou, a comprehensive assessment on the economic, social, and ecological profits and costs of organic rice production in study area was conducted by using analytic hierarchy process (AHP). The results indicated that among all insect pests controlling measures, organic rice planting had the best comprehensive benefits, with a weight value of 0.5355. Ecological cost played a dominant role in comprehensive costs, and that of traditional pesticide-based insect pests control was most cruel, with a weight value of comprehensive costs being 0.6252. From the viewpoint of the ratio of comprehensive profits to comprehensive costs (RPC), organic rice planting was better than any other insect pests controlling modes, whose RPC was 2.4776, being accorded with the requirement of sustainable development of agriculture.

Objective: To investigate the carcinogenic and mutagenic mechanism of bitumen fume., Methods: The experimental mice were forced to inhale the bitumen fume at different exposure level (55 mg/m(3), 165 mg/m(3)) and in different time (30 days, 60 days). The pathological changes of the lung tissue in mice were observed with H.E staining. The content of the DNA in the lung tissue of mice and the cell circles were determined with flow cytometry., Results: The lesion of the lung tissue in mice comprised the atypical hyperplasia of different levels and the carcinoma in situ with the increase of the containment time and dosage; the cycle index was changed: the number of the G 1 phase cells was decreased, the S phase was retarded, the cells entering the G 2/S phase were decreased, the cell proliferation index (P I) was increased and the heteroploid DNA index (DI) was increased (P < 0.05). The cell index in the 55 mg/m(3) group and the 165 mg/m(3) group was higher than that in the control group when the containment time was same. The heteroploid DNA index (DI) in the 55 mg/m(3) group was significantly higher than that in the 165 mg/m(3) group (P < 0.05 or P < 0.01). When the containment dosage was same, the DI in the 60 days treatment group (1.16 +/- 1.51 x 10(-2), 1.20 +/- 2.3 x 10(-2)) was all significantly higher than those in the 30 days treatment group (1.14 +/- 8.8 x 10(-2), 1.16 +/- 1.47 x 10(-2)) (P < 0.05)., Conclusion: The precancerous lesion in the lung tissue of the mice induced by the bitumen fume may be related with the changes of the cell cycle.

Objective: To observe the microstructure of human long bone with scanning electron and atomic force microscopes to understand the ultrastructural organization and the composition and the morphology of the bone collagen and minerals., Method: Fresh human cadaveric humeral bone were fixed and dehydrated to prepare the ground sections and thin slices, respectively, which were observed for bone microstructure using optical, scanning electron and atomic force microscopy, respectively., Results: Under optical microscope, the bone lacunae were in circular permutation around the Haversian canal, and the canaliculi communicated the lacunae and Haversian canal or between the lacunae. Atomic force microscopy represented distinct morphology of the large clusters of collagen, and the collagen fibers thickened in the mineralized zone and appeared in the shape of overlapping discs, whereas the calcium-phosphorus crystal displayed ellipse cylinders. The size, shape and relation of the canaliculi and lacunae were observed clearly. The bone matrix was observed in circular arrangement along the Haversian canal and the calcium-phosphorus crystal was in regular alignment under scanning electron microscope., Conclusion: Atomic force microscopy can help analyze the three-dimensional configuration of calcium-phosphorus crystal and bone collagen as well as the canaliculi and lacuna. The canaliculi serve as the channels mediating alimentation and molecular signals into the lacuna.

Objective: To study the in vitro biocompatibility of novel hydroxyapatite (HA) and AO artificial bone beta-tricalcium phosphate (beta-TCP) with rhesus bone marrow stromal cells (rBMSCs) ., Methods: The third passage of rBMSCs were cultured with HA and beta-TCP respectively, with the cells cultured without the materials as the control. The morphology and proliferation of cells were observed by inverted phase-contrast microscope and scanning electron microscope (SEM). MTT assay was used to semiquantitatively evaluate the cell proliferation., Results: The rBMSC cocultured with HA exhibited good growth as observed under inverted phase-contrast microscope, without significant difference from the cells in the control group. Some small particles were seen pealing off from beta-TCP, and some of the cells died. Under SEM, rBMSCs showed good adhesion to HA with obvious proliferation, but the ratio of adhesive cells was not as high as that in beta-TCP group. MTT assay showed no significant difference in the cell number between HA and the control groups, but the cell number in beta-TCP group was notably less than that of control group., Conclusion: Novel HA has good biocompatibility with rBMSCs for bone tissue engineering, and AO artificial bone still needs improvement to serve as scaffold material for BMSCs.

Objective: To investigate the efficiency and targeting of the GE7 system mediated gene transfer in the chemically induced ovarian tumor by intraarterial route., Methods: Animal models were chemically induced by 7,12-dimethylbenz[a]anthracene (DMBA). GE7-polylysine/beta-galactosidase (beta-gal)/polylysine-HA20 complexes were constructed. Fifteen rats with induced ovarian tumor were divided into three groups, the complexes were injected through ovarian artery and tail vein, respectively. The tumor, heart, liver, spleen, lung and kidney were obtained at 72 h. X-gal staining was used to check expression of beta-gal., Results: beta-gal was expressed strongly and well-distributed in tumor in the first group, and stained blue in the liver, spleen, lung and kidney, but much weaker than that of tumor. In the second group, beta-gal was expressed in the tumor, and weakly expressed in the liver, none was detected in the other organs. In the third group beta-gal was detected in the lung slightly, none was detected in the tumor and other organs., Conclusions: When GE7 complexes were administrated through ovarian artery, the introduced gene expressed preferentially in ovary. This result was the basis of intraarterial administration of therapeutic gene to treat ovarian cancer.
