Objective To investigate the expression and clinical significance of silent information regulator 2 (SIRT2) form tissue of urothelial bladder cancer(UBC) and the effects of down-regulation of SIRT2 on the proliferation, invasion and migration of bladder cancer cells. Methods The expression of SIRT2 protein was detected in 95 samples of urothelial bladder cancer tissues and 39 paracancerous tissues using the immunohistochemical EnVision method. Its relationship with clinicopathological parameters was analyzed. The expression of SIRT2 mRNA in bladder cancer cell lines was detected by RT-qPCR, and the expression levels of SIRT2 protein in bladder cancer cell lines, 12 pairs of fresh bladder cancer tissues were detected by Western blot. Lentiviral infection was used to stably downregulate the SIRT2 gene in bladder cancer cell line T24. CCK8 and cell colony assays were performed to detect changes in cell proliferation ability and Transwell assay to detect changes in cell invasion and migration activity. Results The positive expression rate of SIRT2 in urothelial bladder cancer tissues was 84%(80/95), which was higher than that in normal uroepithelial tissues (5%, 2/39) (PConclusions SIRT2 is highly expressed in bladder uroepithelial carcinoma, and interfers with SIRT2 significantly and so inhibites bladder cancer cell proliferation, invasion and migration.

The amplitude of AC line current at the inverter side is affected by DC commutation failure and fluctuates for a short time after single-phase trip.The fluctuation range increases with the increase of the number of DC commutation failures,resulting in the misjudgment of the traditional single-phase fault nature criterion based on the amplitude of the recovery voltage. To solve this problem,a transmission line fault equivalence model is built firstly to evaluate the impact of DC commutation failure on the sound phase voltage. Then the time domain expressions are derived for the disconnected phase voltage during the recovery voltage stage for different fault nature. After that,a single-phase fault nature identification criterion based on time domain voltage integration is proposed,and its effectiveness is verified by PSCAD/EMTDC simulation. The results show that the proposed method has good resistance to transition resistance and DC interference,and the fault nature identification criterion is suitable for DC commutation failure.

Objective To study the interventional effect of Yangyin Yiqi Heji (Yin-nourishing Qi-replenishing Mixture) to alveolus epithelial-mesenchymal transition in rats with bleomycin-induced pulmonary fibrosis. Methods Wistar rats in SPF class (n= 120) were randomly divided into normal group, model group, prednisone group and low-dose, mid-dose and high-dose Yangyin Yiqi Heji groups. The rat model of pulmonary fibrosis was copied by using intratracheal instillation of bleomycin, and all groups were orally given corresponding medicinals for 28 d after modeling for 1 d. The rats were executed respectively on the 14th d and 28th d to collect lung samples for detection of left lung pathological changes by using HE and Masson staining. The mRNA expressions of α-smooth muscle actin (α-SMA), E-cadherin, collagen I (Col I) and laminin (LN) were detected in right lung by using real time fluorescence quantitative polymerase chain reaction (RT PCR). Results The mRNA expressions of α-SMA, Col I and LN increased, and E cadherin expressions decreased in model group compared with normal group (P<0.05). After treatment, the mRNA expressions of α-SMA, Col I and LN decreased, and mRNA expression of E cadherin increased in all treatment groups (P <0.05). On the 28th d, mRNA expression of E cadherin increased in high-dose Yangyin Yiqi Heji group compared with prednisone groups (P < 0. 05), and mRNA expression of LN decreased in high dose, mid dose and low-dose Yangyin Yiqi Heji groups (P < 0.05). Conclusion Yangyin Yiqi Heji can inhibit the epithelial-mesenchymal transition in rats with bleomycin-induced pulmonary fibrosis, and the mechanism may be related to down-regulation of genes of α-SMA, Col I and LN and up-regulation of E-cadherin gene. Ulcerative colitis (UC) is an inflammatory bowel disease induced by multiple factors, which causes abnormal activation of intestinal immune cells and excessive release of antibodies and inflammatory factors, repeatedly damaging the intestinal mucosa. Macrophages, as innate intestinal immune cells, often maintain the balance of M1/M2 macrophages polarization to normalize the regression inflammation, and the imbalance of their polarization will cause repeated damage of intestinal mucosa and persistent inflammation, which is a main cause of UC. Nuclear factor E2-related factor 2 (Nrf2), as an important regulator of antioxidant and anti-inflammatory, is often used as a target for the treatment of autoimmune diseases.Nrf2 alleviates intestinal high oxidative stress and inflammatory factors by balancing macrophage polarization, which may be of great significance for the prevention and treatment of UC. Summarizing the mechanism of macrophage polarization imbalance on the course of UC and the possible regulatory mechanism of Nrf2 may provide basis for the development of UC targeted therapeutic drugs.

Objective: To investigate hemodynamic parameters in 2 anatomical segments (S1 and S2) of anterior cerebral artery (ACA) in normal pregnancy during the second and third trimester of gestation.
 Methods: The peak systolic velocity (PSV), end diastolic velocity (EDV), time-average maximum velocity (TAMAXV), peak systolic velocity/end diastolic velocity (S/D), resistance index (RI), and pulsation index (PI) in S1 and S2 of fetal anterior cerebral artery (ACA) in 288 normal pregnant women were detected by power Doppler and pulsed Doppler. Multiple regression models were fitted to estimate the relation between Doppler variables and gestational age. The differences of hemodynamic parameters between ACAS1 and ACAS2 were compared.
 Results: The PSV, EDV, and TAMAXV of ACAS1 and ACAS2 were positively correlated with the weeks of pregnancy (P<0.001), all fitted with the cubic curve. The S/D, PI, and RI values of ACAS1 and ACAS2 were not correlated with gestational ages (P>0.05). The PSV, TAMAXV, S/D, PI, and RI of ACAS1 were significantly higher than those of ACAS2, while EDV in ACAS1 was lower than that in ACAS2 (P<0.05).
 Conclusion: The velocity parameters (PSV, EDV, TAMAXV) of the 2 anatomical segments (ACAS1 and ACAS2) are increased with the increase of gestational age in normal pregnant fetus during the second and third trimester of gestation, and the resistance parameters (S/D, PI, RI) are not significantly correlated with gestational age. Distribution of blood flow is different in the blood supply territory between ACAS1 and ACAS2.

Objective: To study the blood flow parameters for 2 anatomical segments of posterior cerebral artery (PCA) in normal singleton fetal.
 Methods: The blood flow velocity parameters peak systolic velocity (PSV), end diastolic velocity (EDV), time-average maximum velocity (TAMAXV), velocity time integral (VTI) and resistance parameters systolic peak velocity and end diastolic velocity ratio (S/D), pulsatility index (PI), resistance index (RI) for 2 anatomical segments in PCA were recorded. The first segment of the PCA (PCAS1) was recorded between the origin of PCA and the proximal part of the posterior communicating artery. The second segment of the PCA (PCAS2) was recorded at the distal part of the posterior communicating artery. The blood parameters in both PCAS1 and PCAS2 were analyzed by using Pearson correlation and multiple curves fitting with gestational age (GA). Paired student's t test was performed to compare the difference in blood parameter between PCAS1 and PCAS2.
 Results: The blood flow velocity parameters in both PCAS1 and PCAS2 were increased with the GA (P<0.0001), with the best fitted curves of Quadratic curve (P<0.0001). There were no correlations between resistance parameters in PCA and GA (P>0.05). Resistance parameters in PCAS1 were higher than those in PCAS2 (P<0.05).
 Conclusion: The blood flow velocity parameters in both PCAS1 and PCAS2 are increased with GA. The resistance parameters in both PCAS1 and PCAS2 do not change with GA. Study on the velocities and resistance in these 3 arterial branches provides a more comprehensive evaluation on the process of brain circulation.

Objective: To review kidney transplantation in the center and analyze the risk factors affecting long-term allograft survival., Methods: Thirty-two relative variables were analyzed with SAS statistical software. Using Log-rank method, we investigated influence of these variables on short-and long-term survival of grafts. Kaplan-Meier analysis was used to estimate the 1-, 3-, 5-, 10-years graft survival rates and half-life. Proportional hazards regression analysis (Cox model) was used to assess and rank the relative risk of potential variables., Results: The 1-, 3-, 5-, 10-years graft survival rates were 83%, 75%, 66% and 48%. After excluding the patients died with functioning grafts, the 1-, 3-, 5-, 10 years grafts survival rate increased to 89%, 82%, 75% and 69%, respectively. The mean half-life was 8.78 +/- 0.14 and 14.09 +/- 0.20 years, respectively. By Log-rank analysis, factors affecting short- and long-term graft survival were identified as: renal function, duration of graft function became normal, cold-ischemia time, presence of acute rejection, delayed graft function, immunosuppressive regimen, complication, infection, anti-rejection therapy. Cox model multivariate analysis showed that there were 18 factors affecting graft survival., Conclusions: New immunosuppressive agents not only significantly increase short-term graft survival, but also have the better long-term outcome tendency. Making assurance to get high quality donor organ and minimizing the death with graft function may be the most feasible way to prolong graft survival at present.
