Objective To explore the feasibility of tirofiban, a platelet surface glycoprotein (GP)Ⅱb/Ⅲa receptor antagonist intervene in transfusion-related acute lung injury (TRALI), by inhibiting platelet activation and by preventing platelet and neutrophil binding to form aggregates. Methods 1) Fifty wild-type male Balb/c mice, aged 8 to 10 weeks, were randomly divided into TRALI, normal, tirofiban TRALI intervention, isotype control and tirofiban normal intervention groups. In the TRALI model, tirofiban TRALI intervention and isotype control groups, each mouse was injected intraperitoneally with lipopolysaccharide (LPS) 0.1 mg/kg, and after 18 h with 4.5 mg/kg anti-MHC-I or IgG2a isotype control antibody, in which 0.5 μg/g tirofiban was injected 30 min before anti-MHC-I injection, and was labeled as tirofiban TRALI intervention. The group without any treatment was set as normal group. The tirofiban normal intervention group was injected with only 0.5 μg/g tirofiban into the tail vein, 30 min before the injection of anti-MHC-I. 2) After antibody injection, the mice were observed for 2 h, then executed with their lungs removed, and the extent of lung injury and the intervention effect of tirofiban were analyzed by comparing the differences in lung dry to wet ratio, total protein, myeloperoxidase (MPO), inflammatory factors and quantitative results of HE staining. The platelet activation level in whole blood and immunofluorescence (IF) quantification of platelet and neutrophil fluorescence were detected by flow cytometry to analyze the mechanism of tirofiban on TRALI. The Tc (collapse temperature) and thermal history of the solution were studied using freeze-drying microscopy and differential scanning calorimetry (DSC), the Tg´ (glass transition temperature of the maximally freeze-concentrated state) was determined according to the two results obtained. The effects of different heating and freezing rated on Tg´ and Tc of isomaltose (IMO) solution were analyzed. The effect of D-Mannitol solution, according to its characteristics reverse glass transition and enhancement of ice crystal structure, on Tg´ and Tc of IMO solution was investigated. It could be concluded that the low and high temperature transitions presented in the thermal history of sample solutions. The measured Tc existed between the end of low temperature transition and the beginning of high temperature transition, and the low temperature transition was identified to be glass transition according to the freeze-concentrated state of solutions. The freezing rates had no significant effect on Tc and Tg´, and Tc was 3~4 ℃ higher than Tg´. However, the higher the freezing rate, the more obvious the collapse phenomenon, with the increasing of heating rate , Tc and Tg´ increased and collapse phenomenon aggravates, Tc increased more obviously, the drying effect was better at 1 ℃/min. With the reduction of IMO/D-Mannitol ratio, Tg´ decreased and the devitrification phenomenon became more obvious, the most was 5:5, the drying effect of 9:1 and 5:5 was better than that of IMO solution, the collapse was worst at 8:2, Tc was 6~7 ℃ higher than Tg´. When a small amount of D-Mannitol was added, the dried structure was better preserved, Tc was higher than that of IMO solution at 9:1, and the drying layer maintained better.

Objective To evaluate the reliability of CardioChek PA portable blood analyzer in the detection of blood lipid and to explore the feasibility and accuracy of its application in lipid screening in high-risk stroke group. Methods According to the profile of the National Committee for Clinical Laboratory Standards of Method Comparison and Bias Estimation Using Patient Samples (NCCLS-EP9-A), two different biochemical analysis systems, CardioChek PA blood analyzer (testing system) and Siemens ADVIA2400 automatic analyzer (reference system), were used respectively to test lipids including total cholesterol (TC), triglyceride (TG), high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) to obtain the correlation coefficient and the linear regression equation as well as predicted bias. Based on the comparison, CardioChek PA blood analyzer was used to detect blood lipid of 1263 persons (more than 40 years old) at high-risk of stroke. Results The outcomes of TC, TG, HDL-C and LDL-C between CardioChek PA blood analyzer and Siemens ADVIA2400 automatic analyzer had significant correlations (r > 0.975, P < 0.05) and error allowance at different medical decision levels were all between 95% confidence interval (CI) of predicted bias, indicating that there were comparability of TC, TG, HDL-C and LDL-C between two detecting systems and the outcomes were acceptable. Moreover, there were347 cases (27.47% ) with abnormal TC, 348 cases (27.55% ) with abnormal TG, 62 cases (4.91% ) with abnormal HDL-C and 441 cases (34.92%) with abnormal LDL-C among 1263 persons. Conclusion There is comparability of TC, TG, HDL-C and LDL-C between CardioChek PA blood analyzer and ADVIA2400 biochemical automatic analyzer and the outcomes of CardioChek PA are accurate and confident, thus CardioChek PA can be used in epidemiological study.
