Objective With urban regional cultural inheritance as the background, the expression and application of Wu Region cultural elements in the public space of Suzhou metro are discussed, and the application value and strategies are summarized. Method Based on the connotation of Wu Region culture, such as inclusiveness, worship of literature and education, flexibility and talent, the extraction method of Wu Region cultural elements is put forward through three ways: typical extraction, abstraction rule and cultural symbol reconstruction. The case analysis is used to analyze the application of Wu Region cultural elements in Suzhou Metro entrances and exits, public art within station and inside carriage compartment from the aspects of design description, material technology, construction technology and technical requirements. The concrete ways of constructing the unique cultural landscape of Suzhou metro with architectural elements, garden elements, cultural character, catering culture, religious beliefs, language culture, opera culture and other aspects of Wu Region culture are introduced in detail. Result & Conclusion Based on the rules of Wu Region cultural elements extraction, to realize the translation and transmission of traditional culture in Suzhou Metro public space by way of establishing Wu Region cultural symbol design system, endowing Wu Region cultural symbol functions and cultural semantics, and applying modern design means will promote significance for inheriting the urban context, shaping the urban spirit in the new era, and enhancing the soft power of urban culture.

Dynamic disaster formation and triggering are closely related to the mechanical behavior of the regional stress field. The regional local stress field is continuously adjusted and changed under the effect of coal mining disturbance. In order to study the precursor response of regional stress field change to dynamic disasters, we used the analytical methods of variational modal decomposition and Hilbert transform to perform time-frequency analysis on the data collected from the Baodian coal mine and to identify and extract the intrinsic modal function (IMF) components reflecting the abnormal dynamic changes in the borehole strain data. The results show that the borehole strain observation data can effectively record the small changes inside the rock mass related to the mining disturbance. After decomposition and transformation, the abnormal fluctuation characteristics of IMF components appear two days before the dynamic pressure event, which is characterized by a “sudden jump” from the stage of stable deformation to the stage of rapid change, “shock” in the stage of rapid deformation change and “drop” in the stage of instability. According to the three-stage theory of seismic deformation, the normal starting time of deformation before the occurrence of dynamic disasters and the “sudden jump-shock-fall” are used as the precursor criteria for the occurrence of coal mine dynamic disasters. Based on the accurate observation of the regional stress field by borehole strainmeter, a criterion method applied to the warning of coal mine power hazards is constructed, which can provide a reference for the safe and efficient recovery of working faces under similar mining environments in coal mines.

Burkitt lymphoma is a highly aggressive subtype of non-Hodgkin lymphoma. And it is one of the fastest growing human tumors. The lymphoma can be divided into three subclasses: endemic type, sporadic type and immunodeficiency-associated type, according to epidemiology and different miRNA profiles. The disease is often found in children and teenagers, rarely in the adults. Outcome with short and intensive chemotherapy is excellent. But the prognosis is poor in adults who are in later period and resistant to therapy. PI3K/AKT/mTOR signaling pathway plays an important role in kinds of cellular functions, including growing, differentiation, metabolism, survival and proliferation. Studies have shown that the PI3K/AKT/mTOR signaling pathway is activated in Burkitt lymphoma, and the inhibitors of this pathway inhibit Burkitt lymphoma cells. We understand the pathogenesis of Burkitt lymphoma by exploring the effects and interaction of PTEN, c-Myc, autophagy and PI3K/AKT/mTOR signaling pathway. And we will manage the combination of PI3K/AKT/mTOR signaling pathway inhibitors with monoclonal antibodies or other pathway inhibitors to find accurate, effective and low-toxic targeted therapy for the adults with drug resistance or in later period.

Objective: To explore the association between egg consumption and the risk of non-alcoholic fatty liver disease(NAFLD), which will provide epidemiological evidence for NAFLD prevention., Methods: A total of 6734 non-NAFLD participants(5500 men and 1234 women, aged(45±14) years old)in the Kailuan cohort followed up in 2014-2015 were enrolled. Semi-quantitative food frequency questionnaires were used to collect the diet frequency, including the consumption of eggs, over the past years. Then the score of dietary approaches to stop hypertension(DASH) was calculated. Abdominal ultrasound was applied for fatty liver diagnosis. The Kaplan-Meier method was used to estimate the cumulative incidences of NAFLD for the three groups(0-4, 5-7 and >7 eggs/week). Furthermore, Cox regression models under various adjusted factors were used to compare the risks of NAFLD., Results: During the median follow-up of 45 months, 1484 NAFLDs were identified among 6734 non-NAFLD participants. The 4-year cumulative incidence rates of NAFLD for the individuals consuming 0-4, 5-7, and >7 eggs/week were 27.1%, 19.9% and 29.6%(P<0.05). The DASH score was significantly higher in those who consumed 5-7 eggs/week than in the other two groups(P<0.001). After adjusting confounders, including DASH score and dietary cholesterol, we found that compared with those consuming 5-7 eggs/week, the individuals with 0-4 and >7 eggs/week had a higher NAFLD hazard, with hazard ratios(95% confidence interval) of 1.20(1.03, 1.41) and 1.25(1.06, 1.47), respectively. The association was more significant in the population with a higher DASH score(P_(interaction)=0.02)., Conclusion: Eating 5-7 eggs/week may have the lowest risk of NAFLD, suggesting taking a moderate amount of eggs to prevent NAFLD, even if the overall diet quality is relatively healthy.

Objective: To detect pathogenic variant in a neonate suspected for Cornelia de Lange syndrome (CdLS)., Methods: Potential mutations of CdLS-related genes (NIPBL, SMC1A, SMC3, RAD21 and HDAC8) were detected by high-throughput target region capture and next-generation sequencing. Suspected variants was verified by Sanger sequencing., Results: The child was found to harbor a heterozygous splice site variant, c.6109-1G>A, of the NIPBL gene. Sanger sequencing suggested that neither parent has carried the same variant, suggesting that it was de novo. The variant was unreported by HGMD and ExAC database, and was predicted to alter an acceptor splicing site. No pathogenic variants of SMC1A, SMC3, RAD21 and HDAC8 genes were detected., Conclusion: The heterozygous c.6109-1G>A splicing variant of the NIPBL gene may underlie the disease in this child. Above finding has expanded the variant spectrum of the NIPBL gene.

Objective: To study the behavioral characteristics of memory maintenance and regression in a mouse model of combined learning and memory training with fluoxetine treatment and explore the neural basis for learning and memory in the barrel area of the brain., Methods: Twenty-six 16-day-old C57 mice were randomized into two equal groups and were given daily intraperitoneal injection of saline (control) or fluoxetine. The mice were subjected to stimulation of the right whiskers using a multi-sensory stimulation simulator and were given simultaneously olfactory stimulation with butyl acetate. In the initial 10 days of the experiment, the mice were given corresponding drug treatment followed by whisker and olfactory stimulations on a daily basis; from day 11 to day 17, only the drugs were administered without the stimulations; on day 18, both the drugs and stimulations were administered. The daily performance of the mice was recorded and analyzed. In the field potential experiment, the left barrel cortex of the mouse brain was selected to record the frequency of field potential signals in response to whisker stimulation., Results: In the behavioral test, the mice treated with fluoxetine showed greater increments of the frequency and angle of whisker deflection than the control mice ( P < 0.01). Compared with the peak levels that occurred on the 10th day, the swing angle and frequency of the whisker deflection decreased on the 17th day decreased in both groups, and the reduction was more obvious in the control group ( P < 0.05). During the training on the 18th day, the whisker movement of the mice increased rapidly to the peak level and showed significant differences between the two groups ( P < 0.05). In the field potential experiment on the 10th and 17th day, the frequencies of field potential signal in response to whisker stimulation was significantly higher in fluoxetine group than in the control group ( P < 0.05)., Conclusions: Combined training of the mice results in the formation of combined memory. Fluoxetine can enhance combined learning and memory abilities and prolong such memories in mice by promoting the function of the barrel cortex cells.

Objective: To explore clinicopathological features and prognostic factors of bone metastasis from primary hepatic carcinoma (PHC)., Methods: Eighty patients were enrolled with bone metastasis from PHC in Tianjin Medical University Cancer Institute and Hospital from January 2005 to June 2014 respectively. The clinicopathological features and prognostic factors of primary tumor were collected and bone metastases. Kaplan-Meier method was adopted for survival analysis. Log-rank method and Cox regression were used for univariate and multivariate analysis., Results: The median survival was 4.7 (95% CI range from 3.4 to 6.0) months. The 6-, 12-, and 24-month survival was 42.5%, 17.5%, 2.5%, respectively. Univariate analysis indicated that Child-Pugh grading (P<0.01), ECOG score (P=0.014), serum cholinesterase (P=0.002), total bilirubin (P=0.006), ascites (P=0.003), TACE (P<0.01) were significant prognostic factors of survival. Multivariate analysis revealed that Child-Pugh classification (P<0.01), TACE (P<0.01) were independent prognostic factors of survival., Conclusion: Bone metastasis from PHC patients had poor survival. Patients can benefit from TACE and liver protecting treatment.

Objective: To investigate the influence of pH on the properties of colloidal gold labeling monoclonal antibody (mAb)., Methods: The pH value of colloidal gold was adjusted by K(2)CO(3) solution. Then colloidal gold with a range of pH 5.0-9.0 labeled enterovirus 71 (EV71)-VP1 mAb respectively, and bovine serum albumin (BSA) was added to block the unreacted sites on the gold colloids for further experiments. The changes in the properties of colloidal gold in the progress of colloidal gold labeling mAb were monitored by UV/Vis spectroscopy. Finally, Mey's test was adopted to identify the stability of immunogold, and the sensitivity of the strip was evaluated by detecting gradual dilution of serum., Results: EV71-VP1 mAb could be conjugated with colloidal gold at pH7.0-8.5, and at this optimal pH, the test strip presented a good sensitivity., Conclusion: pH is an important factor to ensure the stability of immunogold and to determine the conjugation effect between colloidal gold and mAb. The study confirmed that UV/Vis spectroscopy can evaluate the influence of pH on the properties of colloidal gold labeling mAb and set up an optimal pH for colloidal gold labeling using UV/Vis spectroscopy.
