Objective To identify and analyze the phospholipase A (PLA) gene family in Talaromyces marneffei (T. marneffei), providing the basis for further research on the potential pathogenic effect and mechanism of T. marneffei PLA. Methods The members of the PLA gene family in T. marneffei were genome-wide identified by using bioinformatics, and their gene structure, chromosome distribution, protein physicochemical property and structure, conserved motif, as well as phyletic evolution, were analyzed. The expression levels of PLA genes in T. marneffei under macrophage treatment were analyzed using the qRT-PCR method. Results A total of eight PLA genes were identified from the genome of T. marneffei, unevenly located on 5 chromosomes without any tandem duplications or fragment duplications. The amino acid residues of PLA proteins ranged from 577 to 1546, molecular weights from 65 120 to 170 690, and the isoelectric point from 4.47 to 9.28. All PLA proteins were hydrophilic, with all but PLA3 classified as unstable hydrophilic proteins. Phylogenetic analysis revealed that PLA was divided into two subfamilies: PLA2 and patatin-like phospholipase, consisting of 3 and 5 members respectively. Within the same subfamily, gene structures, protein-conserved motifs, and protein structures were relatively similar, whereas significant differences existed between different subfamilies. qRT PCR results showed that the expression levels of PLA3, PLA6, and PLA8 were upregulated in T. marneffei infected with mouse macrophages. [Objectives] The aim of this study was to investigate the differences of endophytic fungal community characteristics between healthy and root rot-diseased Panax notoginseng, as well as the distribution pattern in different plant tissues and their changes with growth years. [Methods] 1-year-old and 2-year-old of healthy and root rot-diseased P. notoginseng were randomly collected and the endophytic fungi in their roots and stems were isolated by tissue isolation method. Based on morphological characteristics and ITS sequence analysis, the isolated strains were classified and identified, and the community composition and diversity of endophytic fungal community were analyzed. [Results] A total of 966 endophytic fungi strains were isolated and identified, which belonged to 3 phyla, 5 classes, 14 orders, 25 families and 31 genera. The number and isolation rate of endophytic isolates were as follows: diseased plant> healthy plant, 2-year-old P. notoginseng >1-year-old P. notoginseng, root>stem. The composition of the culturable endophytic fungi in healthy and root rot-diseased P. notoginseng was quite different, of which the genus Aspergillus, Irpex, Chaetomium, and Penicillium were the dominant endophytic fungi in the healthy P. notoginseng, while the predominant culturable endophytic fungi of root rot-diseased P. notoginseng were Fusarium, Aspergillus, Talaromyces, Chaetomium, and Trichoderma. The composition and diversity of endophytic fungi of P. notoginseng was significantly affected by plant tissues. Specially, the isolation frequency of genus Fusarium from roots of root rot-diseased P. notoginseng was 52%, which was significantly higher than that from the stems of 11.2%. Beneficial fungi such as Trichoderma and Talaromyces were also frequently isolated from diseased roots, with the isolation frequencies of 11.0% and 14.5%, respectively. Moreover, the diversity, richness, and evenness of endophytic fungal community in stems of P. notoginseng were higher than those in roots. With the increase of growth years, the composition and diversity of endophytic fungal community of healthy and root rot- diseased P. notoginseng also changed, and the isolation frequency of Fusarium oxysporum and F. solani in diseased P. notoginseng increased greatly, reaching 14.5% and 32.9%, respectively. Furthermore, the diversity, richness, and evenness of endophytic fungal community in healthy P. notoginseng increased gradually with growth year, while the diversity and evenness of endophytic fungal community in root rot-diseased P. notoginseng were gradually declined. [Conclusions] P. notoginseng harbored a rich diversity of endophytic fungi and its colonization quantity and community composition were significantly distinct between healthy and root rot- diseased plants. 【Objective】 The present paper aimed to know the below-ground ectomycorrhizal status of Castanea mollisima in the major producing area （Luotian county） of Chinese chestnut. The EMF community composition, morphological and molecular identification of C. mollissima were investigated in representative ≥80-year-old chestnut orchards in Luotian. 【Method】300 root samples of C. mollisima were collected and a total of 55 ECM morphotypes were obtained based on morphological characters.【Result】The top50 OTUs were assigned to 34 genera, 15 unclassified species and other taxa. The top 20 taxa in abundance accounted for 95.99% of the top50 OTUs, which were belonged to Basidiomycota （account for 60%, contained five dominant genera: Lactarius, Guehomyces, Inocybe, Russula and unclassified＿ Thelephoraceae）, Ascomycota（30%）, Mortierellomycota（5%）, and other（5%） were belonged to unclassified fungi. Furthermore, among the 22 EMF identified, genus Trichoderma had the largest variety of strains（6）, followed by Fusarium（4）, Penicillium（3）, Talaromyces（3）, Pestalotiopsis（2）, and other three genera: Ilyonectria, Castanediella, and Leptodontidium each possessed 1 specie. Of these fungi, 11 strains sharing high homology with strains which belonged to Fusarium, Trichoderma, Talaromyces and Penicillium were reported with biocontrol function previously, which indicated the 11 strains isolated might also have the potential of biological control. 【Conclusion】 This was the first study to characterize the ECM communities in major producing area of Chinese chestnut, which would lay a foundation for improving chestnut resistance by ectomycorrhizal fungi. Talaromycosis marneffei has been increasing in recent years. Our understanding of this disease has gradually deepened through extensive basic and clinical research, but there are still many limitations. In this article, by incorporating the latest research advancements, we discuss important issues in managing Talaromycosis marneffei trends, aiming to guide effective prevention and control of the disease, improving public health, and reducing the healthcare burden.

Objective: To investigate the clinical manifestations, treatments, and prognosis of pediatric patients with Talaromyces marneffei infection. Methods: In this retrospective study, 7 children diagnosed with Talaromyces marneffei infection in Shenzhen Children's Hospital from July 2017 to October 2021 were recruited. The clinical features, radiology, pathogen detection, immunological evaluation, treatments, and prognosis were analyzed. Results: In 7 cases, 5 were male, 2 were females. The age was from 0.75 to 8.75 years. The main clinical manifestations were fever in 7 cases, cough in 6 cases, malnutrition in 4 cases, papules in 2 cases and medical history of recurrent infection in 3 cases. Physical examination showed that all 7 patients had hepatosplenomegaly, 4 had superficial lymphadenopathy. Laboratory examination showed that 6 cases had decreased hemoglobin and 3 cases had decreased platelet. Chest CT showed that 4 cases had patchy shadows, pleural effusion, mediastinal or axillary lymph node enlargement, 3 had nodular shadows and 2 had cavities. The positive ratio of Talaromyces marneffei culture was 2/2 with tissue samples, 4/5 with bone marrow. The positive ratio was 3/4 by metagenomic next generation sequencing. The fungus was detected in 3 cases by smear microscopy of bone marrow and (or) peripheral blood. All patients were negative for human immunodeficiency virus by the immune function assay. However, 5 cases were confirmed as primary immunodeficiency disease, including 2 cases with high IgM syndrome, 2 with STAT1 gene variation, and the last with severe combined immunodeficiency (IL2RG gene variation). Exclude 1 case which gave up treatment due to acute intracranial infection, and the other patients received effective treatments along with amphotericin B, voriconazole, and itraconazole alone or in combination. Two cases relapsed after medication withdrawal, but 1 case got complete rehabilitation after hematopoietic stem cell transplantation. Conclusions: The clinical manifestations involve multisystem, the common charateristics are fever and cough. The chest CT imaging manifestations are diverse, it should be considered in differentiating tuberculosis. The amphotericin B, voriconazole and itraconazole are effective, but it will easily relapse when withdrawing those antifungal agents.

Objective: To improve the awareness of hyper-IgE syndrome (HIES) characterized by disseminated infection. Methods: We retrospectively analyzed a patient with HIES characterized by Talaromyces marneffei and Staphylococcus aureus mixed disseminated infection in Shenzhen People's Hospital. The clinical manifestations, results of laboratory tests/genetic examinations, therapeutic strategies and prognosis were summarized. The keywords "hyper-lgE syndrome" were used to search and review the literature in Wanfang databases and Pubmed database. Results: In February 2021, an 18-year-old male patient was admitted to our hospital with backache for over 3 weeks and fever for 4 days. Physical examination revealed deciduous teeth in the oral cavity, bilateral renal pain on percussion, and interphalangeal joint hyperextension. Laboratory studies demonstrated increased blood eosinophils and serum level of total IgE. Bacterial culture from bronchoscopic secretions, bronchial mucosa, and necrotic tissue from the left upper arm showed Talaromyces marneffei . Bacterial culture from alveolar lavage fluid, left upper arm necrotic tissue, puncture fluid of right retroauricular abscess and renal drainage fluid suggested methicillin-sensitive Staphylococcus aureus . The chest and abdominal CT revealed diffuse patchy and nodular lesions in bilateral lungs, cavitary lesions in the upper lobe of the left lung, multiple enlarged lymph nodes in the mediastinum, and infectious lesions within both kidneys and perirenal space. Furthermore, the patients was identified with STAT3 mutations by whole exome sequencing, which confirmed the diagnosis of HIES. Nineteen literature articles were retrieved, involving 27 adult patients with a median age of diagnosis of 23 years. The most common manifestations included: skin infection (16/27), eczema (15/27), elevated IgE (26/27) and eosinophils (17/27), as well as positive STAT3 mutation (11/27). Conclusion: Clinicians should be alert to the possibility of hyper-IgE syndrome in patients with severe or disseminated intracellular bacterial infections.

Objective To analyze the clinicopathological characteristics of lymphadenitis caused by Talaromyces marneffei (TM).Method s The clinical data,pathological features,pathogen examination,and treatment of 15 cases of TM-caused lymphadenitis were analyzed retrospectively.Results The 15 cases included 14 males and 1 females,who were aged 26-67 years,with an average age of (49.1±11.87) years.The 15 cases,including 13 cases of acquired immunodeficiency syndrome and 2 cases of diabetes mellitus,were accompanied by superficial lymph node enlargement in the neck and supraclavicular,axillary,and inguinal regions.The structure of cord-like lymph node tissue punctured by thick needle was completely or partially replaced by inflammatory lesions. Under microscope,8 cases showed mainly diffuse infiltration of phagocytes with pathogens;5 cases presented mainly extensive coagulation necrosis with a small amount of pathogens and nuclear debris;2 cases were characterized by small nodular hyperplasia of fibroblasts,formation of granulomatous structure,and scattered distribution of a few multinucleated giant cells.The pathogens were relatively consistent in size and shape,which were round,oval or sausage-shaped and clustered like mulberry.Diastase periodic acid-Schiff staining and hexamine silver staining highlighted the bacterial structure with transverse septum.TM growth was detected in the blood,alveolar lavage fluid,sputum or lymph node extract fungal culture of the 15 patients.Owing to the adequate antifungal treatment in time,these 15 patients were discharged after their conditions were improved.Conclusion Lymphadenitis is one of the major manifestations of the systemic invasion of TM at the late stage,which is tended to be misdiagnosed.Through core needle biopsy of lymph node,it can be diagnosed as soon as possible to avoid delayed treatment and improve the cure rate.

Talaromyces Marneffei infection is rarely reported in patients with chronic active Epstein-Barr virus (EBV) infection. We reported an old man with chronic fever, pleomorphic rash, cough, EBV viraemia, and secondary hemophagocytic syndrome. Repeated histological biopsy and culture of skin lesions revealed Talaromyces Marneffei . This patient was diagnosed as chronic active EBV infection, and Talaromyces Marneffei infection. After treated with glucocorticoid steroids and anti-fungal therapy, the patient finally recovered. EBV infection is usually seen in immune compromised patients, who are susceptible to opportunistic pathogens rarely as Talaromyces Marneffei in this case.

Talaromyces purpurogenus strain Q2 is a kind of beneficial microbe originally separated from the rhizosphere of healthy cucumber. In this study, we evaluated the biocontrol potential of strain Q2 against four soil-borne diseases by plate confrontation culture, potting in the greenhouse. We further estimated the control efficacy of strain Q2 combined metam-sodium fumigation against Fusarium wilt of bitter gourd in the field. The mechanism of strain Q2 controlling bitter gourd wilt and regulating soil microbial community was examined by plate dilution culture, high throughput sequencing and quantitative PCR. The results showed that strain Q2 could efficiently reduce disease incidence of Fusarium wilt of bitter gourd, potato stem canker, black shank of tobacco and black root rot of tobacco in the green house. Its biocontrol efficacy on black shank of tobacco and bitter gourd wilt was 75.3% and 63.4%, respectively. Biocontrol efficacy of strain Q2 on bitter gourd wilt was 51.0% in the artificial disease nursery inoculated pathogen of bitter gourd wilt, while the control efficacy of strain Q2 combined with soil fumigation technology was more than 80% in the same experiment condition. Strain Q2 application and soil fumigation altered soil microbial community composition and recovery trend. Metam-sodium fumigation significantly reduced the abundances of Fusarium oxysporum and causal agent of bitter gourd wilt. Strain Q2 further suppressed the efficient recovery trend of the pathogen. After application of strain Q2, Penicillium was enriched in soil, as well as the beneficial microbes involved in the suppression of F. oxysporum, such as Bacillus and Gaiella . Overall, after soil fumigation, biocontrol efficacy of strain Q2 on soil-borne diseases such as Fusarium wilt could attribute to the formation of beneficial microbial communities in soil and inhibition of strain Q2 on growth and development of F. oxysporum .

Glucoamylase is a critical ingredient for saccharification in the starch decomposition, and widely used in food, pharmaceutical and fermentation industries. Glucoamylases are usually thermostable and have peak activities at high temperature, as required for the industrial process of glucose production. In this study, a glucoamylase gene belonging to the glycoside hydrolase (GH) family 15, Tlga15A, was cloned from Talaromyces leycettanus JCM12802, and successfully expressed in Pichia pastoris GS115. Recombinant glucoamylase TlGA showed optimal activities at pH 4.5 and 75 °C. The result of thermostability analysis showed that TlGA retained above 70% activity after incubating for 1 h at 65 °C, and 43% residual activity after 30 min at 70 °C. Moreover, TlGA had high resistance to most metal ions and chemical reagents tested. Various starch substrates could be hydrolyzed by TlGA, including soluble starch (255.6±15.3) U/mg, amylopectin (342.3±24.7) U/mg, glycogen (185.4±12.5) U/mg, dextrin (423.3±29.3) U/mg and pullulan (65.7±8.1) U/mg. The primary, secondary and tertiary structures of glucoamylase were further analyzed. The low ratio of Gly in the primary structure and low exposed nonpolarity solvent accessible surface in the tertiary structure may be the main reasons for TlGA's thermostability. These results show that TlGA is great promising for potential use in the commercial production of glucose syrups. Moreover, this research will provide knowledge and innovating ideas for the improvement of glucoamylase thermostability.

Objective: To study the chemical constituents and monoamine oxidase inhibitory activity of Talaromyces wortmannii,an endophytic fungus was isolated from Tripterygium wilfordii., Methods: Chemical constituents were isolated from Talaromyces wortmannii by various column chromatography. The structures were elucidated by various spectral data. The activity was tested by using 96-well micro-plated method., Results: Ten compounds were identified as wortmannilactone A( 1),( +) brefeldin A( 2),( 2E,4R*)-4-hydroxy-4-{( 1R*,2S*)-4-oxo-2-[( 1E)-6-oxohept-1-en-1-yl]cyclopentyl} but-2-enoic acid( 3),phaseic acid( 4),deacetylisowortmin( 5),talaperoxide C( 6),brevianamide F( 7),cyclo( D-N-methyl-Leu-L-Trp)( 8),physcione( 9) and chrysophanol( 10). Compound 7 showed moderate anti-monoamine oxidase activity with the IC50 value of 55 μmol / L., Conclusion: Compounds 2 ~ 4,7 ~ 10 are isolated from Talaromyces genus for the first time,and the anti-monoamine oxidase activity of compound 7 is reported for the first time
