Aiming at the problem of community discovery in relational networks, considering the strength of interaction between nodes and information seepage mechanism, an edge weight and connected component (EWCC) community discovery algorithm based on edge weight and connected branches is innovatively proposed.In order to verify effectiveness of the algorithm, firstly, five kinds of interactive two-layer network models are constructed.By analyzing influence of interaction degree of nodes between layers on the network topology, 30 data sets generated under five kinds of two-layer network models are determined.Secondly, the real data set is selected to compare with GN algorithm and KL algorithm in the evaluation criteria of modularity, algorithm complexity and community division number.Experimental results show that EWCC algorithm has high accuracy.Then, the numerical simulation shows that with the weakening of interaction relationship between layers, the module degree is inversely proportional to number of communities, and the community division effect is better when node relationship between layers is weaker.Finally, as an application of the algorithm, the “user-APP” two-layer network is constructed based on empirical data, and the community is divided.

Ubiquitin is a conserved signaling protein. It is able to interact with thousands of target proteins specifically and perform its biological functions via non-covalent interactions. Ubiquilin-1 (Ubql-1) and Rad23A are two transport factors in the protein-degradation process, and both contain ubiquitin-associated domains (UBAs). S65 of the ubiquitin can be phosphorylated specifically by the kinase PINK1, and the phosphorylated ubiquitin has two interchangeable conformations in solution, the relaxed conformation and the retracted conformation. In this work, nuclear magnetic resonance (NMR) methods were used to characterize the regulation of interactions between the ubiquitin and the UBAs of Rad23A/Ubql-1 by phosphorylation. The results indicated that the UBAs selectively interacted with the relaxed conformation of the phosphorylated ubiquitin without affecting the affinity. When interacting with the UBA of Ubql-1, inter-protein binding appeared to be able to promote the transferring from the retracted conformation to the relaxed conformation.

With the development of technology and the improvement of security requirements, a single biometric is hard to meet the needs of highly confidential identity authentication, so multimodal biometrics is required. This paper proposed a hand shape verification method, which could be used as auxiliary authentication. The algorithm did not limit the background of image acquisition, and adopted the white background that was difficult to deal with in the experiments. First, the hand sample area could be extracted by a rough segmentation, and then gained the hand shape by segmenting the rough hand shape contour meticulously according to the samples' characters. This algorithm could extract the hand shape contour from the shadows accurately and it had good adaptability and portability. Simultaneously, it proposed a novel hand shape recognition method named as local shape context, which extracted the shape context characters in local area by local uniform sampling, then matching the hand shape by above characters. The equal error rate for verification of the proposed method is 3.15%, which is decreased about 1.5% ~5% in the same time cost compared with the other shape context algorithms. In cooperative communication systems, in order to overcome the low spectral efficiency problem of one-way relay network, the relay selection problem of two -way relay channel was investigated. The single relay selection strategy was proposed based on amplify forwarding and two-way relay selection strategy was proposed based on network coding. Moreover, bit error rate (BER) and diversity expressions were derived, and the performance of two ways was compared by the simulation. The results indicated that two strategies can achieve full diversity in multi-relay wireless networks. In the high SNR region, the bit error rate of TWRS-NC is half of TWRS-AF, and the gain of TWRS-NC is two times of the TWRS-AF; when bit error rate is fixed at 10-4, the double maximum standard is better than MIN-W selection criteria 1 dB. This paper proposed a new variant of designated confirmer signatures model, called deniable attribute-based designated confirmer signature, in which both the signer and the designated confirmer could run the same protocols to confirm a valid designated confirmer signatures or disavow an invalid signature. And it provided the proof of correctness and security in the random oracle (RO) model . The moving progress o{ bubbles is continuously traced by CCD (Charge Coupled Device) camera, the digital photographs are captured when the experiment is dealt with using image processing technology, then the shape and scale of bubbles along the height can be obtained. In terms of those continuous photographs, and comparing with the calibration value, the real situation of bubbles in the bed can be determined, then the trajectories of bubbles can be obtained. Objective To investigate the expression of X-ray-repaired cross complementary protein 6 (Ku70/XRCC6) in epithelial ovarian cancer(EOC) and its effects on the proliferation, apoptosis and migration of ovarian cancer cells. Methods Western blot was used to detect the expression of Ku70 in 9 ovarian cancer tissues of different grades and 3 normal ovarian tissues. Immunohistochemistry was used to detect the expression of Ku70 in 119 ovarian cancer tissues, and the relationship between Ku70 expression and clinicopathological parameters was analyzed. Kaplan-meier survival curve was used to analyze the relationship between Ku70 expression and 5-year survival. Colony formation and CCK-8 assay were used to analyze the effect of Ku70 expression on proliferation of ovarian cancer cells, and flow cytometry was used to detect the effect of Ku70 expression on apoptosis of ovarian cancer cells. The effect of Ku70 expression on ovarian cancer cell migration was analyzed by scratch test and Transwell chamber method. Results The expression of Ku70 in ovarian cancer tissues was higher than that in normal ovarian tissues. The expression of Ku70 was closely correlated with pathological features (P＜0.05). The survival rate of ovarian cancer patients with high Ku70 expression was lower than that of patients with low Ku70 expression(P＜0.05). Inhibiting the expression of Ku70 decreased poliferation and migration, and increased apoptosis of ovarian cancer cells (P＜0.05). Conclusions The abnormal expression of Ku70 is closely related to the occurrence of ovarian cancer and so Ku70 is a potential therapeutic target for ovarian cancer treatment.

BACKGROUND As an important mineralization element, mercury is widely distributed in different geological bodies and participates in diagenesis and mineralization. With the rapid development of mass spectrometry technology, the field of mercury isotope geochemistry has made remarkable progress. Mercury isotopes have been widely used to trace the biogeochemical processes of the earth's surface and mercury pollution. In recent years, mercury isotopes have been applied to reveal the evolution of planets, identify large igneous provinces in geological history, and trace the sources of mineral deposits. OBJECTIVES To summarize the mercury isotope compositions of different geological reservoirs (meteorites, terrestrial rocks, coal, sediments, volcanic emissions, epithermal deposits) and investigate the factors controlling the Hg isotope fractionation during ore-forming processes in epithermal deposits. METHODS Literature reviewed that included published data from this research group and others. RESULTS Based on previous studies, the isotope composition of mercury in different geological reservoirs was systematically studied. The mercury isotopic composition of geological reservoirs such as meteorites, magmatic rocks, metamorphic rocks, sedimentary rocks, and volcanic gases varied greatly, and some samples also contained non-mass fractionation information. The occurrence and isotopic composition characteristics of low-temperature hydrothermal deposits (modern hot springs, mercury deposits, lead-zinc deposits, antimony deposits, gold deposits) was the focus of this review, and the basic framework of the mercury isotope system construction. Combined with the latest research results, a comprehensive summary of the mercury isotope fractionation mechanism that may have occurred in the mineralization process of the deposit was carried out. The mass fractionation of mercury isotopes in hydrothermal deposits may be caused by fluid volatilization or boiling, condensation, redox reactions, and sulfide precipitation. The non-mass fractionation of mercury isotopes in rocks and ores may be the product of mercury photochemistry during the geological history, or the inheritance of a specific source rock information. CONCLUSIONS In the future, mercury isotope has great application potential in tracing the ore-forming source of low-temperature hydrothermal deposits and characterizing the evolution of ore-forming fluids.

As one of the famous karst springs in Shanxi Province, the Gudui spring is the only medium-low temperature hot spring, with a long history of development and a rich cultural accumulation. The karst groundwater in the Gudui spring catchment was taken as the research object. Through systematic sample collection and isotope analysis, hydrochemistry (Durov map, ion ratio, Gibbs map, and hydrogen and oxygen isotope) methods were comprehensively used to analyze groundwater hydrochemistry and groundwater system runoff characteristics. The 87 Sr/ 86 Sr value of karst groundwater in the Gudui spring catchment was 0.709 to 0.717, and the Mg/(Mg+Ca) value was 0.27 to 0.74. By analyzing the Sr isotope composition and Mg/(Mg+Ca) and 1/Sr variation characteristics, it was concluded that the karst groundwater in the Gudui spring catchment was a mixture of deep hot water and shallow cold water. The karst water subsystem of Nanliang spring presented the characteristics of carbonate stratum runoff. The karst water subsystem of Fuling Mountain Gaoxian Haitou spring and the deep circulation subsystem of Houma Basin exhibited the runoff characteristics of carbonate rock and igneous rock strata. The karst water subsystem of Taiershan Jiuyuanshan Gudui spring presented the runoff characteristics of carbonate rock and ancient silicoaluminate strata. The δ 18 O value in karst groundwater of Guodui spring area ranged from -11.46‰ to -7.81‰, and the average value was -10.08‰. The range of the δ D value was -83.7‰ to -60.8‰, and the average value was -73.6‰. This showed that karst groundwater in the spring area was the result of mixing of various types of water. Through comparative analysis of hydrogen and oxygen isotopes of 2014 and 2021 sampling points at the same location, it was concluded that the change in water samples at the Guduiquan resulted from the gradual accumulation of water supplied by Sanquan Reservoir. The change in Sanquan Reservoir was due to the influence of Yellow River diversion. The karst groundwater in the spring area were characterized by large calcium ion, magnesium ion, and sodium ion values; a small potassium ion value; a large sulfate value; and a small chloride value. The hydrochemical types of karst groundwater in Gudui spring catchment could be divided into SO 4 -Na, SO 4 -Ca, HCO 3 -Na, HCO 3 -Mg, HCO 3 -Ca, and Cl-Na. The hydrochemical types of karst groundwater showed evident hydrochemical composition zoning from HCO 3 -Ca·Mg→HCO 3 ·SO 4 -Ca·Mg→SO 4 ·HCO 3 -Na·Ca→SO 4 ·Cl-Na·Ca. According to the comprehensive analysis of hydrochemical isotope and hydrogeological conditions, the karst water subsystem of Nanliang spring was primarily recharged by rainfall infiltration in the exposed limestone area and river infiltration, and its karst groundwater was recharged by runoff from south to north to the karst water subsystem of Fuling Mountain Gaoxian Haitou spring and the deep circulation subsystem of Houma Basin. The karst water subsystem of Taier Jiuyuan Mountain Gudui spring received rainfall infiltration supplement and upstream runoff supplement from the exposed limestone area. Its karst groundwater flowed from north to south and received the supply of Sanquan Reservoir from Yellow River water in the natural discharge area of Gudui spring.

Acupuncture-moxibustion has affirmative curative effect in the prevention and treatment of senile dementia. Starting from the literature research, a visualization and application method of acupuncture-moxibustion knowledge of senile dementia in ancient books based on partial order structure is proposed. This method could extract and integrate the acupuncture-moxibustion knowledge of senile dementia contained in ancient books of traditional Chinese medicine, and establish a standardized, structured and visual knowledge graph. Applying this method to knowledge visual analysis and clinical auxiliary guidance could provide reference for combing the knowledge of ancient books of traditional Chinese medicine and transforming the knowledge of ancient books into clinical application.

The Fenhe River Basin is the mother river of Shanxi Province. Due to the over-exploitation of water resources and the impact of social and economic development, the ecological environment has deteriorated. After a series of treatment and protection measures, the water quality has since been improved. Mathematical statistics, Piper diagrams, Gibbs model, hydrogen and oxygen isotopes, and other methods were used to analyze the characteristics and sources of hydrochemistry in the surface water of the Fenhe River basin, which revealed the evolution process of surface water quality of the Fenhe River basin. The results showed that the content of the main hydrochemical components in the main stream surface water of Fenhe River basin increased gradually along the runoff path. The hydrochemical types of surface water of Fenhe River basin were mainly HCO 3 ·SO 4 ·Cl-Ca·Na·Mg and SO 4 ·HCO 3 ·Cl-Ca·Na·Mg. There were great differences in hydrochemical components of tributaries and karst water in the basin. There were also great differences in hydrochemical components of tributaries in the basin. The hydrochemical types of surface water of karst water were mainly SO 4 ·HCO 3 -Ca·Mg. The hydrochemical composition of surface water in Fenhe River basin was mainly affected by rock weathering and evaporation crystallization, whereas rainfall had little effect. Na + and K + mainly came from the dissolution of evaporated salt rocks with Na in the surrounding loess. Ca 2+ , Mg 2+ , and HCO 3 - mainly came from the dissolution of carbonate rocks. SO 4 2- may have also come from the dissolution of sulfide minerals in the loess layer around Fenhe River in addition to the dissolution of gypsum. The values of δ D and δ 18 O of Fenhe River surface water were gradually enriched from upstream to downstream. The characteristics of hydrogen and oxygen isotopes further showed that the surface water was mainly affected by evaporation. The results of this study can provide evidence for ecological restoration and protection and ecological civilization construction in the Fenhe River basin.

Objective: To investigate the protective effect of electroacupuncture (EA) at "Zusanli"(ST36)and "Weiwanxiashu"(EX-B3) on capillary function around the renal tubule and renal tubule structure in diabetic mice based on two-photon microscopy (TPM) imaging, so as to providing visualizable evidence for the regulatory effect of EA on diabetic renal vascular microcirculation., Methods: Spontaneous type Ⅱ diabetes mellitus mice (db/db) were employed for this study. Twenty db/db mice were randomly divided into model group ( n =10) and EA group ( n =10), and 10 db/m mice used as the control group. EA was applied to bilateral ST36 and EX-B3 for 20 min/time, 6 times a week for 6 weeks. The body weight was recorded and the fasting blood glucose measured before and after the intervention. The urine production and water consumption of mice in each cage were recorded after EA. The renal in vivo imaging method based on TPM was established to display the morphological structure of renal tubules, and the mouse renal blood flow velocity was detected by injecting 500 kDa dextran-fluorescein into femoral vein after the intervention., Results: Compared with the control group, the proportion of mice with decreased body mass in the model group was increased, accounting for 40%, while that in the control group was 0%; and fasting blood glucose, urine production and water consumption were significantly increased in the model group ( P <0.001, P <0.000 1). A renal in vivo imaging method based on TPM was successfully established, which can be applied to quantitatively analyze the renal blood flow and renal tubular diameter of mice. Based on this method, the results showed that compared with the control group, the blood flow velocity of peritubular capillary in the model group was significantly decreased ( P <0.000 1, P <0.001), renal tubular cells were slightly exfoliated and the diameter of renal tubular was significantly increased ( P <0.000 1). Compared with the model group, EA reduced the body weight loss ratio from 40% to 0%, and significantly decreased the fasting blood glucose, urine production and water consumption ( P <0.01, P <0.000 1, P <0.001), and the blood flow velocity of peritubular capillary in the EA group was significantly increased ( P <0.001, P <0.05) and tubule dilatation significantly alleviated ( P <0.0 1)., Conclusion: EA at ST36 and EX-B3 can ameliorate renal vascular microcirculation disorder to relieve the renal structure damage and improve renal function in diabetes mice.

To explore the spatial and temporal response of water quality to external load reduction in Lake Taihu, Jiangsu Province, China, and clarify the exogenous load reduction under different water inflow and pollution conditions, a water quality model was constructed and the inflow boundaries were divided into seven groups based on the EFDC model. Taking COD and ammonia nitrogen as output targets, the sensitivities of Taihu Lake water quality boundaries were analyzed using a local sensitivity analysis. The results showed that COD and ammonia nitrogen concentrations of each lake area were more sensitive to the boundary load of the lake area than the rest of the lake area, and the sensitivity index was the highest in the Northwest Lake area. Furthermore, the improvement rates of mean COD concentrations in the whole lake decreased by 28.40%-34.71% in the dry season relative to the wet season, and the ranked sensitivity order of the boundaries was as follows:Northwest Lake boundary > Zhushan Lake boundary > Gonghu Lake boundary > Meiliang Bay boundary > Southwest Lake area boundary > Eastern Lake area boundary > East Lake Taihu boundary. The average improvement rates of ammonia nitrogen concentrations in the whole lake were 41.59%-42.34% higher in the dry season relative to the wet season, and the ranked boundary sensitivity order was as follows:Northwest Lake boundary > Meiliang Bay boundary > Zhushan Lake boundary > Gonghu Lake boundary > Southwest Lake boundary > East Lake Taihu boundary > Eastern Lake area boundary. This difference was affected by algal growth and metabolism, and artificial water diversion and drainage. Therefore, it is necessary to consider the reduction period and inflow location according to different water-quality indicators when planning external prevention and control measures in large lakes.

Objectives: This study aims to evaluate the short-term clinical outcomes and patient satisfaction of anterior and pterygoid implants in the rehabilitation of edentulous maxilla with posterior atrophy., Methods: Given a minimum follow-up of 1 year, 25 patients with fixed maxillary rehabilitation over anterior and pterygoid implants were enrolled in this retrospective study. The implant survival rates, peri-implant soft tissue status (including probing depth, modified sulcus bleeding index, and plaque index), marginal bone loss, and patient satisfaction were measured., Results: The survival rates for anterior and pterygoid implants at 1-year follow-up were 96.5% and 97.8%, respectively ( P >0.05). No statistically significant difference in probing depth, modified sulcus bleeding index, and plaque index was observed between the two types of implants ( P >0.05). The marginal bone losses of anterior implants were 0.62 mm± 0.44 mm (mesial) and 0.61 mm± 0.40 mm (distal), and those of pterygoid implants were 0.64 mm± 0.46 mm (mesial) and 0.68 mm± 0.41 mm (distal) mm. These results showed no statistical difference in mesial and distal sites ( P >0.05). Patients indicated a high degree of satisfaction with the full-arch prostheses supported by anterior and pterygoid implants., Conclusions: For the edentulous maxilla with posterior atrophy, full-arch fixed prostheses supported by anterior and pterygoid implants has an acceptable short-term clinical outcome and excellent patient satisfaction. It may be considered as a predictable and feasible method for maxillary rehabilitation.

Discharge in Niangziguan Spring is 7.19 m 3 ·s -1 , which is the main water supply source for Yangquan City and Pingding County. Every year, Yangquan municipal government take water from the mouth of Niangziguan Spring to supply drinking water to urban residents at a rate of approximately 1.7 m 3 ·s -1 . It is of great significance to determine the characteristics and causes of variations in spring water flow conditions for the appropriate utilization of water resources and pollution prevention. Here, sample collection and hydrochemical isotope analyses were undertaken for the Niangziguan Spring area to chemically characterize the water environment and genesis. The pH of the karst spring is 7.2-7.5 with an average of 7.36; the calcium content of the water is 112.1-135.2 mg·L -1 with a mean value of 131.4 mg·L -1 ; the concentration of magnesium ions is 34.8-42.3 mg·L -1 with an average of 40.8 mg·L -1 ; the concentration of K + +Na - ions is 41.6-46.7 mg·L -1 with an average of 45.2 mg·L -1 ; and the sulfate ion concentration is 185.6-271.8 mg·L -1 with a mean value of 255.4 mg·L -1 . The hydrochemical type of the aquifer is classified as HCO 3 ·SO 4 -Ca·Mg. The spring water is characterized by high Ca 2+ , Mg 2+ , and SO 4 2- concentrations, and low Na + , K + , and Cl - concentrations. The supply path of Chengxi Spring is shortest followed by Wulong Spring. The supply paths of the remaining five spring are much longer. The Niangziguan Spring water environment is characterized by increasing pollution from coal mine acid drainage alongside decreasing inputs from domestic sewage. Environmental isotope tracing shows that sulfate in Chengxi Spring mainly derives from precipitation and gypsum dissolution, and the concentrations of sulfate in Wulong Spring are increasing. These changes are mainly driven by the amount of coal mine acid water pollution in the area.

Objective: To detect the expression of miR-146a in patients with AML, and to evaluate the relationship between miR-146a expression level and clinical characteristics, treatment response, EFS and OS., Methods: 154 patients with newly diagnosed AML were enrolled in AML group, 50 controls (patients with thrombocytopenic purpura or voluntary donor of bone marrow) were enrolled in control group. The miR-146a expression levels in bone marrow mononuclear cells was detected by RT-PCR between 2 group. AML patients were treated with chemotherapeutic drugs, and their clinical response and survivals were assessed., Results: The expression level of MiR-146a in AML group was significantly lower than that in control group. The ROC showed that miR-146a could distinguish the patients in AML and control group better (area under curve 0.819 (95%CI: 0.761-0.877). Meanwhile, the proportion of good and moderate good prognosis (P＜0.001), proportion of WBC count ≤15.2×10 9 /L (P＜0.05), CR rate (P＜0.05), EFS (P＜0.01) and OS (P＜0.01) in patients with high miR-146a expression were higher than those in patients with low miR-146a expression. Cox's model showed that miR-146a expression level positively realated with incressed EFS and OS., Conclusion: MiR-146a is downregulated in AML patients, which might be served as a biomarker for predicting risk and prognosis of AML patients.

Objective: To observe the effect of acupoint catgut embedding on histopathological changes of gastric mucosa and expression of mucosal hypoxia inducible factor (HIF)-1α and vascular endothelial growth factor (VEGF) proteins in chronic atrophic gastritis (CAG) rats, so as to explore its mechanisms underlying improvement of CAG., Methods: Male SD rats were divided into blank group ( n =10) and model group ( n =9) and catgut embedment group ( n =10). The CAG model was established by free drinking of N-methyl-N'-nitro-N-nitroso-guanidine solution (100 μg/mL) and irregular diet. Catgut embedment was applied to "Zhongwan" (CV12) and bilateral "Zusanli" (ST36) and "Pishu" (BL20), once every 10 days, 6 times in total. Histopathological changes of gastric mucosal tissue were observed under microscope after H.E. staining. The expression of HIF-1α and VEGF proteins in the antrum of stomach was detected by Western blot., Results: H.E. staining showed that compared with the blank group, the number of rats with glandular necrosis, atrophy and mucosal stasis in the model group were increased significantly ( P <0.001, P <0.05).Compared with the model group, the number of rats with gland necrosis, atrophy and mucosal stasis in the catgut embedment group were significantly reduced ( P <0.01, P <0.05). Western blot displayed that the expression levels of HIF-1α and VEGF proteins were significantly higher in rats with blood stasis and model group than in those without blood stasis and the blank group ( P <0.01), and were considerably down-regulated in the catgut embedment group than in the model group ( P <0.05, P <0.01)., Conclusion: Acupoint catgut embedment can improve the injury of gastric mucosa in CAG rats, which may be associated with its function in down-regulating the expression of HIF-1α and VEGF proteins in the gastric mucosa.

The water of the Longzici Spring catchment is the source of industrial, agricultural, and urban domestic water in Linfen City, Shanxi Province. It is of great significance to identify the sources and influencing factors of karst water resources for their development and utilization, and the protection of the ecological environment. Taking the karst groundwater in the Longzici Spring Area as the research object, systematic sample collection and isotope analysis were carried out. Hydrochemistry methods (Durov diagram, ion ratio system, Gibbs diagram, hydrogen and oxygen isotopes, and sulfur isotopes) were used to analyze the hydrochemical characteristics of groundwater. The hydrogeochemical and environmental isotopic characteristics of Longzici Spring water are revealed. The hydrochemical field of karst groundwater in the Longzici Spring domain has obvious zonation, from the deep buried detention area to the supply area to the runoff and discharge area. The hydrochemical types of karst groundwater detention area are SO 4 -Ca and SO 4 -Ca·Mg, with total dissolved solids (TDS) of 960.5-2340 mg·L -1 ; the supply area is HCO 3 -Ca·Mg and HCO 3 ·SO 4 -Ca·Mg, with TDS of 340.5-562 mg·L -1 ; and the runoff and discharge areas are SO 4 ·HCO 3 -Ca·Mg·Na, SO 4 ·HCO 3 -Ca, and SO 4 ·HCO 3 -Ca·Mg, with TDS of 494.0-1168.5 mg·L -1 . The karst groundwater is mainly SO 4 ·HCO 3 -Ca and SO 4 ·HCO 3 -Ca·Mg type water. The content range of sulfate ions is 61.6-1503 mg·L -1 , with an average of 481.4 mg·L -1 . A total of 70.3% of the karst groundwater SO 4 2- samples exceed the standard. The sulfate ions in karst groundwater mainly come from the dissolution of gypsum in the Ordovician aquifer and the oxidation of pyrite in a coal aquifer. The ratio of sulfate ions in the Longzici Spring catchment that come from pyrite oxidation in water is 20.2%. According to the analysis of hydrogen, oxygen and sulfur isotopes, 70% of karst groundwater is polluted by the acid water of coal mines to different degrees. Using the software PHREEQC, it is concluded that the hydrochemical characteristics of the study area are mainly affected by dissolution and dedolomitization, rock salt, and gypsum.

The research aims to propose the approaches to the clinical trial of acupuncture and moxibustion and provide the methodological guide for the research in this field in terms of the inadequate systematic collection, lack of the specific characteristics of acupuncture-moxibustion treatment and insufficient persuasion in clinical evidences of acupuncture-moxibustion research. Through a systematic review of the registration of acupuncture-moxibustion clinical trials at home and abroad, the distribution of diseases with the recommendation of acupuncture and moxibustion in Cochrane Library database, combined with the evidence grade of the evidence-based medicine and the individual characteristics of clinical practice with acupuncture and moxibustion, some research ideas and framework are proposed for the clinical trial of acupuncture and moxibustion. To take the curative effect as the forerumer, the dominant disease and therapeutic methods of acupuncture and moxibustion should be initially generalized by the case experiences and clinical observation at the first. In view of the potential function of acupuncture and moxibustion in treatment, the rigorous and standardized clinical trial should be carried out to verify their therapeutic effect. Finally, the experimental research is adopted to explore the mechanism of acupuncture and moxibustion. Through the combination of quantitative and qualitative methods, all of the evidences are integrated to form an integrative evidence chain. It is anticipated that the construction of the integrative evidence chain in clinical trial of acupuncture and moxibustion may enhance the promotion of clinical practice with acupuncture and moxibustion and increase the influence of acupuncture-moxibusiton discipline.

Purpose: To investigate the effect of LncTUG1 on NK cell killing sensitivity in oral squamous cell carcinoma cells., Methods: Oral squamous cell carcinoma cells were used as experimental objects in vitro. TUG1 siRNA was transfected,the expression of TUG1, cell proliferation, cell apoptosis and NK cell killing rate were detected by qRT-PCR, MTT, flow cytometry and LDH. Bioinformatics software was used to predict that TUG1 and miR-212-3p will target and complement each other, so luciferase reporter vector was constructed and the targeting relationship was identified. TUG1 siRNA and miR-212-3p inhibitor were co-transfected into oral squamous cell carcinoma cells, the effects of miR-212-3p inhibitor on TUG1 siRNA on proliferation, apoptosis and NK cell killing sensitivity of oral squamous cell carcinoma cells were evaluated. The data were analyzed with SPSS 21.0 software package., Results: TUG1 siRNA could significantly reduce the expression of TUG1 in oral squamous cell carcinoma cells (P=0.000), decrease cell proliferation (P=0.001), promote cell apoptosis (P=0.000), increase the killing rate of NK cells (P<0.01). TUG1 siRNA targeted to increase the expression of miR-212-3p. miR-212-3p inhibitor could reverse the effects of TUG1 siRNA on proliferation, apoptosis and NK cell killing rate of oral squamous cell carcinoma cells., Conclusions: Down-regulation of TUG1 targeting and negative regulation of miR-212-3p inhibits proliferation, induces apoptosis and improves NK cell killing sensitivity of oral squamous cell carcinoma cells.

Purpose: To observe the effect of individualized healing abutment on patients with posterior dental implant prosthesis and its influence on implant stability and food impaction., Methods: Eighty patients who received posterior dental implants in Jiangsu Stomatological Hospital from June 2013 to June 2018 were selected as the study subjects. According to the different healing abutment, they were divided into traditional operation group (40 cases) and experimental group (40 cases). Patients in the experimental group used personalized healing abutment on the basis of traditional dental implants. Differences in dental implant stability, food impaction score, masticatory efficiency and incidence of complications between the two groups after implant restoration were compared. SPSS 13.0 software package was used to analyze the data., Results: At 1, 3 and 6 months after operation, the stability of implants in the experimental group was significantly higher than that in the control group (P<0.01); One, 3 and 6 months after operation, the food impaction score in the experimental group was significantly lower than that in the traditional operation group(P<0.01); There was no significant difference in masticatory ability between the two groups before operation; After treatment, the occlusive force and masticatory efficiency in the experimental group were significantly higher than those in the traditional operation group (P<0.05). The incidence of complications such as tooth loosening and periodontitis in the experimental group was significantly lower than in the traditional operation group (P<0.05)., Conclusions: Individualized healing abutment has a good therapeutic effect on patients with posterior implant prosthesis, and can significantly improve the incidence of food impaction and complications with a good application value.

The purpose of this study was to investigate the inhibitory effect of the main 9,10-dihydrophenanthrene orchinol isolated from Spiranthes sinensis Radix et Herba on the invasion and migration of human gastric cancer SGC-7901 cells and its preliminary molecular mechanism. SGC-7901 cells were cultured in vitro, after the cells were treated with different final concentrations(5, 10, 20, 40, 80 μmol·L⁻¹) of orchinol for 24, 48 or 72 hours, the effect of orchinol on cell viability was measured by MTT assay. Wound healing and Transwell assays were performed to determine the effects of different final concentrations(5, 10, 20, 40 μmol·L⁻¹) of orchinol for 48 hour on invasion and migration abilities of SGC-7901 cells, respectively. The protein expression levels of β-catenin, Wnt-3α, DvL2, cyclinD1 and GSK-3β were detected by Western blot. The results showed that 5-80 μmol·L⁻¹ orchinol inhibited the viability of SGC-7901 cells in a dose-dependent and time-dependent manner, and the IC₅₈ values of 24, 48 and 72 hours were 77.79, 42.96 and 7.85 μmol·L⁻¹, respectively. Compared with the control group, the ability of invasion and migration of SGC-7901 cells was significantly inhibited after treated with 5, 10 and 20 μmol·L⁻¹ orchinol for 48 hours ( P <0.05, P <0.01), and the dose-effect relationship was observed. The results of Western blot showed that orchinol could significantly down-regulate the protein expression levels of β-catenin, Wnt3a, DvL2 and cyclinD1, and up-regulate the protein expression level of GSK-3β( P <0.05, P <0.01, P <0.001). Objective: To observe the impact of catgut embedding on histopathological changes and expression of phosphorylated-Janus kinase-2 (p-JAK 2), phosphorylated-signal transducer and activator of transcription 3 (p-STAT 3), Cyclin D 1, Bcl-2, and suppressor of cytokine signaling (SOCS) of the gastric mucosa in chronic atrophic gastritis (CAG) rats, so as to explore its mechanisms underlying improving CAG., Methods: Forty-one SD rats were randomly divided into normal ( n ＝12), model ( n ＝9), natural recovery ( n ＝10) and catgut embedding ( n ＝10) groups. The CAG model was established by free drinking of N-methyl-N'-nitro-N-nitroso-guanidine (MNNG) solution (100µg/mL) and irregular diet for 24 weeks. Catgut embedding was applied at bilateral "Zusanli" (ST 36), "Pishu" (BL 20) and "Zhongwan" (CV 12), once every 10 days, 6 times altogether. The histopathological changes of the gastric mucosa were observed by naked eye and light microscope after H．E. staining. Wes-tern blot was used to detect the expression of p-JAK 2, p-STAT 3, CyclinD 1, Bcl-2 and SOCS 3 proteins in the gastric antrum tissue., Results: After modeling, the regional gastric tissue showed pale thinner gastric wall, and poorer elasticity with fewer plica, congestion of blood vessels, reduction of regional glands with necrosis and disordered arrangement as well as evident hyperplasia of the connective tissue, which was evidently milder in the catgut embedding group. Compared with the normal group, the expression levels of p-JAK 2, p-STAT 3, CyclinD 1, Bcl-2, SOCS 3 proteins were significantly up-regulated in both model group and natural recovery group ( P <0.01), without significant differences between the latter two groups ( P >0.05). After catgut embedding intervention, the expression levels of p-JAK 2, p-STAT 3, Cyclin D 1, Bcl-2 proteins were considerably down-regulated ( P <0.01), and that of SOCS 3 protein was notably further increased relevant to the natural recovery group ( P <0.01)．., Conclusion: Catgut embedding may improve pathological changes of gastric mucosa in CAG rats, which possibly associated with its effects in up-regulating gastric SOCS 3 protein expression and in inhibiting the activation of JAK 2-STAT 3 signaling pathway to reduce the expression of CyclinD 1 and Bcl-2 proteins.

Objective: To analyze the factors influencing the therapeutic effect of "Shoulder Tri-needles therapy" in the treatment of shoulder-hand syndrome of stroke patients by using machine learning approach, so as to provide a feasibility for improving clinical efficacy., Methods: A total of 586 stroke patients with shoulder-hand syndrome eligible for this study were involved in our machine learning experiments for classification of the influential factors. Their data including the age, gender, pulse condition, complexion, tongue quality, tongue coating, disease stage, body mass index (BMI), blood pressure, blood glucose, blood triglyceride, blood total cholesterol, smoking history, drinking history, and final outcomes were extracted from the medical record system (from Oct. of 2014 to Jan. of 2017 in the First Affiliated Hospital and Shenzhen Futian Hospital of Guangzhou University of Chinese Medicine). The single rule algorithm (1 R) was adopted to learn, followed by optimization with Repeated Incremental Pruning to Produce Error Reduction (RIPPER) algorithm, and C 5.0 decision tree algorithm., Results: The accurate classification rates of 1 R, RIPPER and decision tree model were 87.37%(512/586), 95.90% (562/586), and 97.10% (569/586), respectively. The final outcomes of machine learning of this study showed that the disease stage (acute or recovery stage), complexion difference, tongue coating difference, blood pressure level, consumption of alcohol, BMI, and smoking habit were the most important factors influencing the therapeutic effect of "Shoulder Tri-needles" in the treatment of shoulder-hand syndrome of stroke patients., Conclusion: The disease stage, complexion and tongue identification, blood pressure level, alcohol drinking and smoking habits, and BMI are the principal factors affecting the therapeutic effect of "Shoulder Tri-needles therapy" in the treatment of shoulder-hand syndrome of stroke patients.

Objective: To evaluate the effectiveness and superiority of acupoint catgut-embedding in the treatment of insomnia., Methods: Randomized controlled trials (RCTs) for insomnia treated by acupoint implantation of catgut were included in the present study. We searched the related articles from 6 databases (PubMed, Google Scholar, CBM, CNKI, VIP and Wanfang) from the date of inception to March 19, 2017 by using keywords "catgut implantation at acupuncture point" or "catgut implantation at acupoint" and "insomnia" or "agrypnia" and "randomised (or randomized) clinical trial". Quality assessments were performed by two researchers independently using GRADE profile 3.6, followed by crosscheck. Data were analyzed using software Revman 5.3 and STATA 14.0., Results: Twenty-two trials met the inclusion criteria were included. They recruited 1 714 cases of insomnia. GRADE grading suggests a low quality of these articles. Heterogeneous test (numerical test) showed χ 2 ＝31.11, P ＝0.07, I 2 ＝32%. Gabriz chart and sensitivity analysis ultimately determined that these trials have smaller or no heterogenity. The effective rate of acupoint catgut embedding for insomnia was higher than that of the control group ( OR ＝ 3.77; 95% CI : 2.63－5.41; P <0.00001)., Conclusion: Acupoint catgut embedding therapy has a positive role in clinical treatment of insomnia. But due to lower quality of current literature, more high-quality randomized controlled clinical trials are required.

Objective: To analyze the regularity of acupoint selection, main acupoints and theoretical basis in acupuncture treatment of urticaria., Methods: Papers collected from the time of establishment of each database to September of 2017 were retrieved from databases CNKI, CBM, VIP and WF by using keywords of "acupuncture" "moxibustion" "blood-letting therapy" "autohemotherapy" "cupping" "acupoint catgut embedding" "auricular points" "acupoint injection" "fire-needle" (or red-hot needle), "dermal needle" "needle-embedding" "urticaria" in both Chinese and English. The collected papers were brought into analysis according to the inclusion and exclusion criteria, from which the prescriptions for acupuncture treatment of urticaria were subjected into descriptive statistical analysis, association rule analysis, and cluster analysis by using Access 2010, Clementine 18.0 and Stata software., Results: Outcomes of analysis indicated that the treatment methods of urticaria with acupuncture and moxibustion, with different emphases, may be classified into eight categories. For treating the exterior syndrome of urticaria, acupoints of the Bladder Meridian, Governor Vessel and Conception Vessel were often employed to harmonize Ying and Wei and to dispel the pathogenic wind, while for treating the interior syndrome, acupoints of the Large Intestine Meridian, Spleen Meridian, and Stomach Meridian were usually used to invigorate the spleen to dispel dampness and to regulate blood circulation. The top five frequently used acupoints were Quchi (LI 11), Xuehai (SP 10), Zusanli (ST 36), Sanyinjiao (SP 6) and Geshu (BL 17). It was crucial to make use of the specific acupoints with adequate meridian- qi , such as He -Sea points, Back- Shu points, and Yuan -Primary points. There were some fixed forms in the combination of acupoints, including LI 11, SP 10, Dazhui (GV 14) and auricular Lung, Shenmen, Fengxi, Adrenal gland, which had the highest confidence coefficient for the meridian points and ear acu-points, respectively. The outcomes of cluster analysis about the acupoint prescriptions showed that 12 acupoint groups as the SP 6-Hegu (LI 4)-LI 11-SP 10-ST 36, etc. were frequently used., Conclusion: The regularity of acupuncture treatment of urticaria can be discovered using data mining technology, resulting in an in-depth understanding and having a solid theoretical basis.

Objective: To evaluate the efficacy and safety of acupuncture in the treatment of epilepsy., Methods: We first collected research data about randomized control trials (RCTs) of acupuncture treatment of epilepsy from databases CNKI, CBM, VIP and Wanfang using key words "(acupuncture)", "(epilepsy)"or "(epilepsy)"or "(epilepsy)", and from PubMed, Google Scholar, and Cochrane Library using key words "acupuncture" "needling" "prod" "epilepsy" "epilepsies" or "seizure disorder" or "simple seizure" from the date of database construction to May 5, 2017. Then, two researchers extracted the study outcomes and evaluated the evidence qualification of the research data independently using GRADE profile 3.6 software and analyzed the primary outcome indexes using RevMan 5.3 and STATA 14.0., Results: Eleven articles containing 827 cases were included in the Meta-analysis. We found that acupuncture or acupuncture in combination with other conventional drugs had a significantly better efficacy than conventional drugs in the treatment of epilepsy [ OR =3.94, 95%CI (2.49, 6.24), Z =5.85, P <0.000 01]. There was a statistical heterogeneity ( P =0.04, l 2 =60%), and the Meta-analysis was not feasible for the studies of acupuncture vs medication. Combination of acupuncture and Chinese traditional drugs was significantly better than simple Chinese traditional drugs [ OR = 4.61, 95%CI (2.18, 9.74), Z =4.01, P <0.000 1], and combination of acupuncture and wes-tern medicines was obviously better than simple western medicines [ OR =3.07, 95%CI(1.24, 7.65), Z =2.41, P =0.02] in the treatment of epilepsy., Conclusion: Acupuncture therapy may have a positive effect in the treatment of epilepsy but the conclusion needs further verification due to very fewer high-quality and well-designed RCTs found at the present.

Purpose: To investigate the effect of miR-21 on promoting vascular differentiation of umbilical cord blood derived mesenchymal stem cells (UCBMSCs) in vitro., Methods: UCBMSCs (from Stem Cell Bank of Tongji University) were cultured; Lenti-LacZ-Luciferase and Lenti-miR-21-Luciferase Lentiviral vector were reconstructed; Real-time fluorescence quantification PCR and Western blot were performed to detect the expression of key angiogenic factors at 0, 1, 4, 7 and 14 d after target gene was transduced into UCBMSCs; UCBMSCs of the third generation were transduced with Lenti-LacZ-Luciferase and Lenti-miR-21- Luciferase; matrigel tube assay was performed to observe the tube-like structure under microscope. The data were statistically analyzed using Student's t test and variance analysis with SPSS13.0 software package., Results: UCBMSCs were successfully cultured; The results of qPCR and Western blot showed the expression of HIF-1α and VEGF in miR-21 group significantly increased at 4 d and continued until 7 d and 14 d; Matrigel results showed the tubular structures in Lenti-miR-21-Luciferase group were more than other groups both in amount and in length., Conclusions: MiR-21 can promote vascular differentiation of UCBMSCs.

Purpose: To observe the clinical effects of screw-retained implant-supported casted abutment integrated crowns (IACs) in the molar region with limited interocclusal space., Methods: This study involved 507 implants in 376 patients with limited interocclusal space in the molar region. All implants were inserted to the jaw by standard one-stage protocol. Screw-retained IACs were used as the final prosthesis. With 6 to 24 months follow-up, the clinical effects of screw-retained IACs were recorded and analyzed using SPSS 17.0 software package., Results: During 6 to 24 months of follow-up, the implant survival rate was 99.61%. 37 patients had ceramic fracture which mainly happened in the group with 3-4 mm interocclusal space. In each group, porcelain fused to metal (PFM) prosthesis had ceramic fracture more easily than PFM prosthesis without porcelain on occlusal surface(P<0.05). As the interocclusal space became smaller, the probability of collapsing porcelain increased(P<0.05).13 patients had screw loosening. 13 patients suffered from gingival swelling and bleeding, they were given periodontal treatment and oral hygiene instruction. Patients were satisfied with the restoration results., Conclusions: Screw-retained IACs can be used to repair missing teeth in the molar region with limited interocclusal space.

Objective: To evaluate the feasibility of minimally invasive internal fixation of pelvic anterior and posterior ring for the treatment of type C pelvic fracture, and to explore its operative techniques and therapeutic efficacy., Methods: From December 2010 to December 2015, 18 patients with type C pelvic fracture were treated by reconstructive plates fixation through minimally invasive ilioinguinal approach for pelvic anterior ring injuries, and by invasive percutaneous sacroiliac joint screw fixation for pelvic posterior ring injuries. There were 11 males and 7 females ranging from 29 to 68 years old with an average age of 43.6 years old. According to Tile classification, there were 14 cases of type C1, 3 cases of type C2, 1 case of Type C3. To be specific, 12 cases with hemi-fracture of rami ossa pubis accompanied with fracture of the sacrum, 2 cases with hemi-fracture of rami ossa pubis accompanied with sacro-iliac joint dislocation, 3 cases with bilateral-fracture of rami ossa pubis combined with pubic symphysis separation accompanied with single-fracture of the sacrum, 1 case with bilateral-fracture of rami ossa pubis combined with bilateral-fracture of sacro-iliac joint were included. Operation time, intra-operative blood loss, injuries of lumbosacral nerves and iliac blood vessels, and fracture reduction were observed., Results: All wounds were primary healing. No complications such as infection, deep venous thrombosis, injuries of lumbosacral nerves and iliaca vessels or heterotopic ossification occurred. According to Matta criterion of fracture reduction, 14 cases got excellent results, 3 good and 1 fair. Sixteen patients were followed up in a period varying from 6 to 33 months with 16.7 months on average. And according to functional score of Majeed, 13 cases obtained excellent results, 2 good and 1 fair, with an average score of 92.13±5.44., Conclusions: Internal fixation with reconstructive plates through the ilioinguinal approach and with percutaneous iliosacral screw for type C pelvic facture on pelvic anterior ring and pelvic posterior ring respectively have advantages of shorter operation time, smaller invasive trauma, less blood loss and etc. Thus, this technique is safe and practicable, yielding satisfying results., Competing Interests: The authors of this article and the planning committee members and staff have no relevant financial relationships with commercial interests to disclose.

Crohn’s disease and ulcerative colitis are two forms of inflammatory bowel diseases. They are autoimmune disorders caused by excessive inflammatory response to antigens in the intestine. In addition to the hygiene hypothesis which suggests the potential application of helminthic infection in the treatment of inflammatory bowel diseases, helminthic infection has shown preventive and treatment effects in animal models of inflammatory bowel disease. Clinical trials have been initiated. For example, Trichuris suis ova infection at a certain dose has a promising efficacy in the treatment of inflammatory bowel diseases. Helminthic infection may also have adverse effects. Therefore, helminth-derived immunomodulatory molecules are needed to overcome these problems. It is traditionally considered that the Th1/Th2 axis is involved in the mechnisms of the efficacy of helminthic infection. More recent research has pointed out the much more participation of the Tregs/Th17 axis. The mechanisms may also involve other palyers such as mucosal barrier, Toll like receptor and macrophages. This paper reviews the effect and mechanism of helminthic infection on the prevention and treatment of inflammatory bowel disease.

Purpose: To evaluate the clinical effectiveness of one-piece implant-supported detachable telescopic fixed bridge in edentulous patients., Methods: Seventeen patients were treated with one-piece implant-supported detachable telescopic fixed bridge. A total of 18 prostheses were fabricated with 8 in the upper jaws and 10 in the lower jaws.Fixed bridges retained by telescopic crowns were used as final prostheses, with milling titanium or all-ceramic abutments as primary crowns, gold-electroforming crowns as secondary crowns. Surveys about clinical and radiographic examination, satisfaction and prosthetic complications were conducted after 3 months,1 year, 2 years, 3 years after final rehabilitation. Data analysis was performed using SPSS 17.0 software package., Results: Radiography showed stable bone levels for all implants except 2 implants, which were observed slight marginal bone resorption. The results of one-way ANOVA showed that no significant difference in modified plaque index or modified sulcus blooding index was found during the follow-up period (P>0.05). The probing attachment level deteriorated by 1.5 mm during the first 3 years (P<0.05). Eighteen restoration provided sufficient fixation and stability. Two porcelain fractures occured but had no influence on restoration. The patients were highly satisfied with the outcomes. The frequency of prosthetic maintenance per patient per year was 0.11., Conclusions: One-piece implant-supported detachable telescopic fixed bridge is an effective method with satisfactory long-term aesthetic and stable outcomes in edentulous patients.

Objective: To explore the expression of regulatory B cells (Bregs) in peripheral blood of patients with systemic lupus erythematosus (SLE) and elucidate the function of Bregs in the pathogenesis of SLE., Methods: A total of 38 active SLE patients, 22 inactive SLE subjects and 20 healthy controls were enrolled. The expressions of IL-10+ CD19+ Breg and CD19+ CD24hi CD38hi Breg in peripheral blood mononuclear cells (PBMC) were evaluated by flow cytometry. And IL-10 was measured in the culture supermatants by enzyme-linked immunosorbent assay (ELISA)., Results: The expression of IL-10+ CD19+ Breg in PBMCs of active SLE patients was (1.54±0.64)%. And it was lower than those in inactive SLE patients (2.42±0.75)% (P<0.01) and healthy controls (4.35±1.00)% (P<0.01). And the expression of CD19+ CD24hi CD38hi Breg was (1.26±0.45)%. Also it was lower than those in inactive SLE subjects (2.01±0.61)% (P<0.01) and healthy controls (3.14±0.87)% (P<0.01).The level of IL-10 significantly decreased in culture supermatants of active SLE patients. And it was lower than those in inactive SLE subjects (P<0.05) and healthy controls (P<0.01). The percentage of IL-10+ CD19+ Breg and CD19+ CD24hi CD38hi Breg in PBMCs of SLE patients was positively correlated with the level of IL-10 in culture supernatants (r=0.652, P<0.01 and r=0.574, P<0.01)., Conclusion: The expression of IL-10-related Bregs significantly decreases in PBMCs of active SLE patients. And Bregs may play some role in the pathogenesis of SLE.

Objective: To observe the effect of acupuncture therapy on 14-3-3, Bcl-2 and Bax expression levels in the cerebral cortex in neonatal rats with hypoxic-ischemic brain damage(HIBD)., Methods: Timed pregnant Sprague-Dawley rat dams were delivered either vaginally (normal group), or by C-section (sham-operation group) or by C-section with 5 min of global anoxia (anoxia group), with 8 rats in each group. The rat pups of the anoxia group were randomly divided into model group and acupuncture group (n =8). Acupuncture stimulation of "Naosanzhen" "Niesanzhen" and "Zhisanzhen" acupoints was given begin- ning from the 14th day after birth, once daily for 7 consecutive days. All rat pups were killed by decapitation on day 21 after birth, and then 14-3-3, Bcl-2 and Bax immunoactivity (expression) in the cerebral cortex were detected by immunohistochemistry., Results: In comparison with the normal group, the expression level of cerebral cortical 14-3-3 was significantly decreased, and that of Bax remarkably increased in the model group (P<0. 01, P<0. 05). Compared to the model group, cortical 14-3-3 and Bcl-2 expression levels were markedly up-regulated in the acupuncture group (P<0.01, P<0.05). Compared to the normal group, cortical 14-3-3 expression level was obviously lower, but Bax expression level significantly higher in the sham-operation group (P<0. 05, P<0. 01). No significant differences were found between the model and normal groups in the expression levels of Bcl-2, and between the acupuncture and model groups in the expression levels of Bax (P>0. 05)., Conclusion: Acupuncture intervention can increase the expression of 14-3-3 and Bcl-2 in the cerebral cortex in HIBD rats.

Objective: To establish a method for screening nicotinamide phosphoribosyl transferase (NAMPT) inhibitors based on endogenous fluorescence determination., Methods: The double mutants of NAMPT, G355C/D393C, was cross-linked by using 1, 4-Bismaleimidobutane (BMB) to block the entrance of enzymatic active site of NAMPT. The binding of compounds to NAMPT was evaluated according to the change of spontaneous fluorescence of NAMPT and BMB-NAMPT with 280 nm excitation and 333 nm emmision. The in vitro enzamatic activity of NAMPT was determined by nuclear magnetic resonance. The cell viability was determined by MTT assay., Results: FK866 significantly decreased the spontaneous fluorescence of NAMPT but not of BMB-NAMPT. Rosmaric, cynarine and 1, 3-dicaffeoylquinic acid also decreased the spontaneous fluorescence of both NAMPT and BMB-NAMPT. However, the inhibition on two proteins was equivalent. FK866 significantly inhibit the catalysis of NAMPT. Rosmarinic acid, cynarine and 1, 3-dicaffeoylquinic acid failed to inhibit the catalysis of NAMPT. FK866 inhibited the viability of A549 cells, but rosmarinic acid, cynarine and 1, 3-dicaffeoylquinic acid did not., Conclusion: Endogenous fluorescence spectrometry based on NAMPT and BMB-NAMPT protein can be used for screening compounds that bind with NAMPT, and distinguishing the binding site - either within the enzymatic active site or not. Rosmarinic acid, cynarine and 1, 3-dicoffeoylquinic acid can bind to NAMPT out its enzymatic active site.

Objective: To observe changes of brain glucose metabolism by needling at Waiguan (SJ5) in cerebral infraction (CI) patients using 18F-fluorodeoxyglucose (FDG) positron-emission computer tomography (PET/CT), thus exploring its effect and mechanisms., Methods: A total of 21 patients with CI were recruited in this study. The location of lesion was limited to the left basal ganglia by CT or MRI scan. All patients were randomly assigned to three groups. i.e., the acupoint group (Group A), the non-acupoint group (Group B), the blank control group (Group C), 7 in each group. Patients in Group A were needled at right Waiguan (SJ5). Those in Group B were needled at non-acupoint [10 mm beside Waiguan (SJ5)], whereas those in Group C did not receive any treatment. All patients underwent PET/CT head scan. All data were statistically analyzed using SPSS 13.0 Software and SPM8 Software., Results: Compared with Group C, glucose metabolism increased in bilateral superior temporal gyrus (BA38), right superior frontal gyrus (BA9), left cingulate gyrus (BA24), left culmen and pyramid of cerebellum, and right cerebellar tonsil of cerebellum in Group A. Compared with Group C, glucose metabolism increased in bilateral superior frontal gyrus (BA6, BA9, BA10), bilateral middle frontal gyrus (BA6, BA10), left middle frontal gyrus (BA4), bilateral uncus of limbic lobe (BA36, BA38), left cingulate gyrus (BA24, BA31), left posterior cingulate gyrus (BA30), left precuneus (BA7), left inferior parietal lobule (BA4), and left lingual gyrus of occipital lobe (BA18) in Group B. Compared with Group B, glucose metabolism increased in bilateral superior temporal gyrus (BA22, BA38), right inferior frontal gyrus (BA47), left culmen and cerebellar tonsil of cerebellum in Group A. Activated encephalic regions of needling at Waiguan (SJ5) were mainly dominated in the healthy side, bilateral superior temporal gyrus, and right inferior frontal gyrus. Activated encephalic regions of cerebellum were located at the left cerebellar hemisphere, left culmen of anterior cerebella lobe, and bilateral cerebellar tonsil of posterior cerebella lobe., Conclusions: Needling at Waiguan (SJ5) of CI patients induced increased glucose metabolism in local cerebral regions. Functional neuroimaging using PET/CT could directly reflect changes of brain glucose metabolism by acupuncture.

Objective: To explore the optimal treatment for non-tuberculous mycobacterial keratitis (NTMK)., Methods: Experimental study.Fifty-one New Zealand white rabbits were inoculated with non-tuberculous mycobacteria (NTM) to establish the animal model of NTMK. Eyes with NTMK were randomly divided into 7 groups and topically treated with amikacin (2.5%) (B), gatifloxacin (3%) (C), azithromycin (1%) (D), amikacin (2.5%) plus gatifloxacin(3%) (E) , amikacin (2.5%) plus gatifloxacin (3%) combined with debridement (F), early lamellar keratoplasty (G) and balanced salt water (as a control) (A). All the drugs were administered 8 times per day within initial 7 days followed by 4 times per day after 1 week. The clinical manifestations of each rabbit were observed and recorded every week. The score of clinical manifestations before and after treatment does not meet the normal distribution, so it was presented by the median ± quantile interval and compared with rank sum test and SNK test. Fisher exact test was used to compare differences in the distribution of presenting the healing rate of keratitis., Results: After 1 week treatment, six eyes were cured in group G, and none were cured in other groups, the scores of clinical manifestations of group B, C, D, E, F and G were 6 ± 4, 8 ± 2, 6 ± 4, 8 ± 3, 7 ± 4, 3 ± 1, they were higher than that of group A (q = 20.36, 11.43, 19.21, 16.93, 18.64, 37.50, P < 0.05). After 2 weeks, the scores of group B, C, E, F and G were higher than that of group A (q = 23.50, 16.71, 15.21, 12.43, 30.87, P < 0.05), and the healing rate of group G was higher than that of group A. After 3 weeks, the score of clinical manifestations of group B, D, F and G was higher than that of group A, and the healing rate of group F and G was higher than that of group A. After 4 weeks, the inflammation of group F and G were healed, and a little inflammation remained in the eye of group A. The score of clinical manifestations of group D, E, F and G was higher than that of group A. None was detected in the bacterial culture after 4 weeks. In histopathological examination, there were integrated and proliferated corneal epithelium, fibroblast cells proliferation in the eyes of experimental groups was found, while corneal cells edema, stroma degeneration and necrosis with lymphocytes and plasma cells in the ulcerated lesion in the corneal stromal of group A., Conclusions: Amikacin, gatifloxacin or azithromycin is effective in arresting NTM, while antibiotic combined with debridement is more effective and early lamellar keratoplasty is the most rapid and thorough treatment of NTMK.

Objective: To explore the significance and evaluate the early structural erosion through the expressions of Th17 cells in peripheral blood of patients with rheumatoid arthritis (RA) in clinical remission., Methods: A total of 41 active RA patients without structural erosion were selected. Intracelluar flow cytometric detection of Th17 cells in peripheral blood was performed. And the supernatant level of interleukin (IL)-17A was determined simultaneously in RA patients and control groups at baseline and endpoint of 24-month therapy. The correlations were analyzed between Th17 cells and RA disease activity index DAS28. They were classified into radiographic progression (P, n = 10) and radiographic non-progression groups (NP, n = 26) by the Sharp/van der Heijde score (SHS) at the endpoint. The differences of Th17 cells and IL-17A levels were analyzed between P (SHS > 0.5) and NP groups (SHS ≤ 0.5)., Results: The expression of Th17 cells in active RA patients was significantly higher than that of controls [(1.63 ± 0.45)% vs (0.91 ± 0.26)%, P < 0.01]. And the results of IL-17A level were similar [1510 ± 280) vs (320 ± 31) ng/L, P < 0.05]. The expression of Th17 cells was positively correlated with DAS28 score (r = 0.87, P < 0.01). Thirty-six RA patients were followed up at the endpoint and all of them stayed in clinical remission (DAS28 < 2.6). The peripheral blood expressions of Th17 cells of P group were significantly higher than those of NP group . At the same time, no differences of IL-17A levels existed between two groups., Conclusion: Structural erosion still progresses in some RA patients despite an apparent clinic remission. And a high-level peripheral expression of Th17 hints at structural erosion.

Objective: To investigate the association of rs2910164 G > C polymorphism and rs11614913 T > C polymorphism in pre-miR-146a and pre-miR-196a2 with genetic damage levels in coke oven workers., Methods: A total of 575 nonsmoking workers who have worked for more than one year in a coke-oven plant at Wuhan, Hubei Province were enrolled in this study in September to October, 2010. The general characteristics as well as blood and urine samples were collected. The genetic damage levels were detected by cytokinesis-block micronucleus cytom assay and represented as micronucleus (MN) frequencies of binucleate cells in peripheral blood lymphocytes. The rs2910164 G > C polymorphisms in pre-miR-146a and rs11614913 T > C polymorphisms in pre-miR-196a2 were genotyped by using TaqMan assay. The plasma concentrations of benzo[a]pyrene-diolepoxide (BPDE)-albumin adducts were determined by using ELISA. All data were analyzed, the frequency ratio (FR) and 95%CI were calculated., Results: Totally, 575 workers were taken into consideration. The rs2910164 C allele was associated with increased MN frequencies in the coke oven workers (P trend = 0.025), and the MN frequencies were higher in rs2910164 CC genotype carriers (4.38 ± 3.46) than in wild-type rs2910164 GG genotype carriers (4.02 ± 3.09) (FR = 1.18, 95%CI:1.04-1.34). The further stratified analyses by working years, gender, alcohol consumption, and the levels of BPDE-albumin adducts showed that the effects of rs2910164 C allele in increasing MN frequencies were robust in subjects who were males (FR = 1.11, 95%CI:1.02-1.20), nondrinkers (FR = 1.07, 95%CI:1.00-1.14), working years less than 20 (FR = 1.12, 95%CI:1.03-1.22), and workers with lower BPDE-albumin adducts levels (FR = 1.11, 95%CI:1.02-1.21) (P trend = 0.011, 0.044, 0.006 and 0.020, respectively). In addition, the MN frequencies were higher in workers with rs11614913 TC genotype (4.27 ± 2.91) than workers with rs11614913 TT genotype (3.90 ± 3.32) (FR = 1.12, 95%CI:1.02-1.23).Workers carried both rs2910164 GG and rs11614913 TT genotypes were set as a control, and the MN frequencies of workers with both rs2910164 CC and rs11614913 CC genotypes (5.32 ± 4.94) were 1.51 (1.21-1.89) times higher than the control (3.75 ± 3.01)., Conclusion: The rs2910164 C allele in pre-miR-146a and rs11614913 C allele in pre-miR-196a2 were associated with increased genetic damage levels in coke oven workers.

Objective: To discuss the expression and clinical significance of VEGF-C and nm23-H1 in stage II and III colorectal carcinomas., Methods: SP immunohistochemical staining was employed to determine the expression of vascular endothelial growth factor-C (VEGF-C) and nm23-H1 in the tumor tissues of 110 cases of stage II and III colorectal carcinomas and in the adjacent mucosal tissues of 53 cases as control, and analyze their correlation with cliniopathological features and prognosis., Results: The positive expression of VEGF-C in the carcinoma tissues was 71.8%, significantly higher than that in the adjacent mucosal tissues (22.6%, P < 0.001). The positive expression of nm23-H1 in the carcinoma tissues was 57.3%, significantly lower than that in the adjacent mucosal tissues (90.6%, P < 0.001). The expression of VEGF-C was significantly correlated with lymph node metastasis (P < 0.05), and the nm23-H1 expression was significantly correlated with lymph node metastasis and pathological type (P < 0.05). The expression of VEGF-C and nm23-H1 did not show a significant correlation with age, gender, primary tumor site, tumor size and depth of invasion (P > 0.05). The VEGF-C expression was negatively related with nm23-H1 expression in colorectal carcinoma (r = -0.361, P < 0.001). The median overall survival (MOS) and median disease free survival (MDFS) of 110 patients with colorectal carcinoma were 55 and 48 months, respectively. The colorectal patients with different VEGF-C and nm23-H1 expression showed significant differences in the 5-year OS rate and 5-year DFS rate (P < 0.001). The patients with negative VEGF-C expression and positive nm23-H1 expression had a better prognosis., Conclusions: The joint detection of VEGF-C and nm23-H1 expression is very promising in prediction of the prognosis of patients with stage II and III colorectal carcinoma. However, whether it can be used as a marker in prognosis judgment needs further investigation.

Objective: To observe the arousal effect of electroacupuncture (EA) stimulation of Baihui (GV 20), Shuigou (GV 26), etc. on severe craniocerebral injury patients., Methods: A total of 90 cases of severe craniocerebral injury were randomly allocated to routine medication, naloxone and EA groups, with 30 cases in each group. For patients of the routine medication group, mild hypothermia therapy, medicines for dehydration, hormonal therapy, vascular dilation, cerebral nutrition supporting, anti-inflammation, etc. were given. For patients of the naloxone group, intravenous drip of naloxone 0.4 mg/kg in the first 3 days, 0.2 mg/kg for 7 days and 0. 1 mg/kg afterwards. For patients of the EA group, EA (1 Hz/50 Hz) was given for 30 min once daily. All the treatments were conducted once a day for 14 days. The Glasgow Coma Scale (GCS) and Glasgow Outcome Scale (GOS) were used for assessing the therapeutic effect., Results: In comparison with pre-treatment in each one of the routine medication, naloxone and EA groups, GCS scores were all obviously increased in the 3 groups following the treatment, and one month's follow-up (P<0. 05). The GCS scores of both naloxone and EA groups were significantly higher than those of the routine medication group (P<0.05). No significant difference was found between the naloxone group and EA group in GCS scores (P>0. 05). According to the GOS, one month's follow-up showed that of the three 30 cases in the routine medication, naloxone and EA groups, 6, 12 and 14 were improved, 8, 10 and 10 moderate handicap, 8, 3 and 2 severe handicap, 5, 3 and 2 vegetative state, and 3, 2 and 2 dead, with the arousal rates being 46. 66% , 73. 33% and 80. 00%, respectively. The therapeutic effects of both naloxone and EA groups were significantly superior to those of the routine medication group (P<0.05)., Conclusion: EA intervention at early stage can promote the recovery of neurological function, accelerate the consciousness from coma and improve the outcomes of patients with severe traumatic brain injury.
