With the progression of big data and cloud computing technologies, there has been an escalating trend in cross-institutional data sharing and interaction within the financial sector. However, the frequent occurrence of privacy breaches poses significant challenges to ensuring data security protection. This necessitates the elevation of requirements for identity authentication across data centers and other technologies related to data security management. The traditional centralized identity authentication schemes are inherently flawed in security and scalability, which hampers their ability to meet the demands of high-frequency cross-center data sharing applications. The blockchain technology has emerged as a potential solution, offering exceptional characteristics such as a distributed nature, tamper-proof properties, traceability, and the ability to achieve multi-party consensus. In practical application scenarios, it is essential to not only protect the privacy of user identities but also to effectively regulate anonymous identities. An anonymous and supervisable cross-data center identity authentication scheme based on blockchain was proposed. This scheme did not necessitate alterations to the original architecture of each data center. Instead, a distributed multi-center consortium blockchain was formed by leveraging more than one server from each data center to establish a data center union. Blockchain technology was utilized to ensure the transparency and reliability of the identity authentication process. Additionally, attribute signatures were employed to safeguard user identity privacy while simultaneously creating an identity mapping table that facilitated the tracking of real user identities. Consequently, the scheme was designed to protect user privacy during cross-data center identity authentication and to enable effective supervision over anonymous identities. A thorough security analysis and the results of experiments demonstrate that the proposed scheme possesses the following attributes: it does not require users to re-register, it exhibits characteristics of unforgeability and traceability, and it is capable of withstanding various network attacks, including replay attacks, denial of service attacks, tampering attacks, and man-in-the-middle attacks.

Objective To observe the effect of Bushen Tiaojing ( Kidney-tonifying Menstruation-regulating) Formula (BSTJF) on quality and apoptosis of blastocyst in mice induced by repeated controlled ovarian stimulation (COS). Methods 160 female ICR mice were randomly divided into normal group, model group, low-dose BSTJF group and high-dose BSTJF group (25.6,51.2 g/mL). Except for the normal group, in the other three groups repeated COS model was established (COS three times). The mice in the normal group and model group were gavaged with distilled water, and the mice in the low-dose and high-dose BSTJF groups were gavaged with low- and high-dose BSTJF at 9 a. m. every day for 10 consecutive days. The model group, low- and high-dose BSTJF groups were intraperitoneally injected with pregnant mare serum gonadotropin (PMSG) 10 IU at 4 p. m. on the 11th day, and human chorionic gonadotropin (hCG) 10 IU 48 hours later. The normal group was intraperitoneally injected with saline 0. 1 mL at 4 p. m. on the 11th day. There were 3 consecutive cycles with an interval of 4 days between every two cycles. Mice in all groups were caged with male mice of the same strain immediately after the third intraperitoneal injection of hCG (for the model group, low- and high-dose BSTJF groups) or normal saline (for the normal group). It was recorded as pregnancy if a vaginal plug was identified or sperm was observed on the vaginal smear in the following morning. Pregnant mice were killed 96 hours after the last intraperitoneal injection to obtain blastocysts. The quality of blastocysts was observed and number of blastocysts were counted under inverted microscope, and the rate of high quality blastocysts was calculated. The apoptosis rate of blastocysts was detected by TUNEL, and the levels of mRNA expression of Bcl-2 and Bax were detected by real-time fluorescence quantitative PCR. The levels of expression of apoptosis-related factors (Bcl-2, Bax and cleaved caspase- 3) were detected with Simple Western, and the expression of cytochrome c was detected with immunofluorescence. Results Compared with the normal group, in the model group the number of blastocysts obtained was increased (P <0. 05), while the rate of high-quality blastocysts was decreased (P<0. 05); the total number of cells in blastocysts decreased, and the apoptosis rate increased (P < 0. 05); the RNA and protein ratio of Bcl-2 and Bcl-2/Baxm decreased (P<0. 05), while the mRNA and protein expression levels of Bax increased(P<0. 05), and the protein expression level of cleaved caspase- 3 increased(P<0. 05); cytochrome C was released from mitochondria and co-localization coefficients M1 and M2 decreased(P<0. 05). Compared with the model group, there was no significant difference in the number of blastocysts obtained in the low- and high-dose BSTJF groups (P>0. 05), but the rate of high-quality blastocysts increased significantly (P <0. 05); the total number of cells in blastocysts increased and the apoptosis rate decreased ( P < 0. 05); the mRNA and protein ratio of Bcl2 and Bcl-2/Bax increased (P<0. 05), while the mRNA protein expression level of Bax decreased(P<0. 05), and protein expression level of cleaved caspase-3 decreased (P<0. 05); there was no significant release of cytochrome C, but the co-localization coefficients M1 and M2 increased (P<0. 05). Conclusion Bushen Tiaojing Formula has the potential to inhibit blastocyst cell apoptosis induced by repeated COS and improve the quality of early embryos, and its mechanism may be related to mitochondrial apoptosis pathway. Yarrowia lipolytica is a non-conventional yeast with unique physiological and metabolic characteristics. It is suitable for production of various products due to its natural ability to utilize a variety of inexpensive carbon sources, excellent tolerance to low pH, and strong ability to secrete metabolites. Currently, Y . lipolytica has been demonstrated to produce a wide range of carboxylic acids with high efficiency. This article summarized the progress in engineering Y . lipolytica to produce various carboxylic acids by using metabolic engineering and synthetic biology approaches. The current bottlenecks and solutions for high-level production of carboxylic acids by engineered Y . lipolytica were also discussed, with the aim to provide useful information for relevant studies in this field.

Objective To investigate the effects of quercetin on cell viability,apoptosis,autophagy,and phosphatidylinositol 3-kinase(PI3K)/protein kinase B(Akt)/mammalian target of rapamycin(mTOR)signaling pathway in human prostate cell carcinoma PC-3 cells.Methods PC-3 cells were cultured in vitro ,and cell viability was detected by CCK-8.Apoptosis was detected by TUNEL staining.Autophagy vesicle was observed by acridine orange staining.Autophagosomes was observed by GFP-LC3 plasmid transfection analysis.Expressions of autophagy-related protein microtubule associated protein 1 light chain 3 fusion protein(LC3)and Beclin-1 and PI3K/Akt/mTOR signaling pathway protein were detected by Western blot analysis.Results Quercetin inhibited cell viability in a dose-time dependent manner and induced apoptosis.Quercetin increased the number of autophagy vesicles and autophagosomes in PC-3 cells.Quercetin increased the expressions of LC3-Ⅱ/LC3-Ⅰ and Beclin-1 in PC-3 cells and decreased the expression of phosphorylated-PI3K,phosphorylated-Akt and phosphorylated-mTOR.Conclusion Quercetin may induce autophagy by inactivating PI3K/Akt/mTOR signaling pathway in PC-3 cells.

Objective: To investigate the risk factors for post-stroke pneumonia and assess the value of A(2)DS(2) score in predicting post-stroke pneumonia in elderly stroke patients., Methods: The clinical data were retrospectively collected from elderly stroke patients from January, 2007 to December, 2012. A(2)DS(2) score was then assigned using the clinical information from the medical record. The ability of the score to discriminate between patients with post-stroke pneumonia and those without was quantified using ROC analysis. The calibration of the score was analyzed using Hosmer-Lemeshow goodness-of-fit test., Results: A total of 131 elderly male stroke patients were enrolled in this study, among whom the incidence of post-stroke pneumonia was 29.01%. The independent risk factors for post-stroke pneumonia identified included moderate (P=0.0081, OR: 5.6089; 95%CI: 1.5663-20.0854) and severe (P=0.0048, OR: 44.4827; 95%CI: 3.1847-621.3126) neurological impairment, dysphagia (P=0.0005, OR: 7.5265; 95%CI: 2.4282-23.3292), and atrial fibrillation (P=0.0226, OR: 4.1778; 95%CI: 1.2221-14.2825). The incidence of post-stroke pneumonia ranged from 2.2% in patients with a A(2)DS(2) score less than 3 to 75% in those with a score higher than 8. The C-statistic of A(2)DS(2) score for predicting post-stroke pneumonia was 0.86 (95%CI: 0.784-0.911) by the ROC analysis. The A(2)DS(2) score was well calibrated to predict post-stroke pneumonia in elderly patients by Hosmer-Lemeshow test (7.083, P=0.528)., Conclusion: The A(2)DS(2) score can be useful for predicting post-stroke pneumonia and for routine monitoring of high-risk elderly stroke patients in the clinical setting.

Objective: To study the correlation of CD44 with epithelial-mesenchymal transition(EMT) and metastasis in nasopharyngeal cancer cells, and explore the possible mechanism of CD44 regulates EMT and metastasis in nasopharyngeal cancer cells., Method: The CD44 and EMT-associated proteins in 5-8F and 6-10B nasopharyngeal cancer cell lines were assayed by Western blotting. The erasion trace test was performed to observe the migratory ability of 5-8F and 6-10B nasopharyngeal cancer cells. Using lipid-mediated DNA transfection technique, the low metastatic nasopharyngeal cancer cells 6-10B were transfected in vitro with plasmid which contained CD44 gene, and then new nasopharyngeal cancer cells were obtained. The CD44 and EMT-associated proteins in 6-10B, empty vector transfected and CD44-transfected cells were assayed by Western blotting. The erasion trace test was performed to observe the alteration of migratory ability of nasopharyngeal cancer cells before and after CD44 transfection., Result: The expression of CD44 and EMT-associated protein MMP-9 in 5-8F was higher than that in 6-10B, but EMT-associated protein E-Cadherin in 5-8F was lower than that in 6-10B. The migratory ability of 5-8F was higher than that of 6-10B. The expression of CD44 and MMP-9 were significantly higher in the CD44-transfected nasopharyngeal cancer cells than in the control groups. Compared with control groups, the migratory ability of CD44-transfected nasopharyngeal cancer cells was significantly increased., Conclusion: CD44 positively regulates the metastatic ability of nasopharyngeal cancer cells, which is relevant to the process of EMT.

Objective: To analyze deafness gene mutations by genechip., Method: The peripheral blood samples were obtained and DNA templates were extracted by extraction kits. The deafness gene mutations were distinguished by genechip., Result: Among 42 patients with non-syndromic hearing loss, GJB2 235delC was found in 11 cases (7 cases were homozygosis, 4 cases were heterozygosis); 4 cases were shown to carry the PDS IVS7-2A>G mutation., Conclusion: The incidence of GJB2 gene and PDS IVS7-2A>G mutations among the deaf- mute children in Guiyang city is 38.10%. Molecular genetic screening for these mutations and genetic counseling are effective methods to prevent the occurrence of hereditary hearing loss.

Objective: To identify the genetic characteristics in patients with nonsydromic hearing loss (NSHL) in Hunan province, to determine the prevalence and spectrum of mutations in GJB2 gene, and to explore the pathogenic mechanism., Methods: A total of 140 sporadic patients with NSHL were enrolled after clinical examination. Molecular studies were performed by amplifing the coding region of GJB2 gene, purifying the PCR products, and sequencing directly. Sequences were analysed by DNAStar software to determine GJB2 mutations in the patients. Special method was designed to confirm the unreported mutation., Results: We detected GJB2 mutation in 56 out of the 140 patients (40%, 56/140). Both of the 2 alleles were mutated in 29 patients and 1 allele in the other 27 patients, and the rate of allele mutation was 30.4%(85/280). Ten variations were detected, including 7 mutations and 3 polymorphisms. The deaf-causing mutations were nonsense mutation c.139G>T; frameshift mutation c.235delC and c.176-191del16; and missense mutation c.109G>A, c.344T>G, c.550C>T and c.571T>C. The unreported missense mutation was c.344T>G. The c.235delC mutation was the most prevalent mutation found in the 27 patients (19.3%, 27/140). The frequency of c.109G>A mutation was next to c.235delC found in 25 patients (17.9%, 25/140)., Conclusion: GJB2 mutation is a major cause for NSHL. The most common-spot in Chinese patients with NSHL is c.235delC. The unreported missense mutation is c.344T>G.

Objective: To summarize the clinical features and the experiences of diagnosis and treatment of extramedullary plasmacytomas (EMPs), and to enhance the cure rates of EMPs., Method: Clinical data of 8 patients with EMPs in head and neck, who treated in our hospital from Jan. 1990 to Dec. 2004, were reviewed. Of 8 cases, 3 cases with the tumors occurred in nasal cavities, 1 in maxillary sinus, 2 in nasopharynx, 1 in posterior wall of oropharynx., Result: Eight patients who were all pathologic confirmed EMP accepted surgical resections of the tumors, and 4 of 8 cases were boosted radiation therapy post-operation. Four cases were still alive disease-free for more than 1, 5, 10 and 12 years after treatments, respectively. One died of local recurrence in 2 years, 1 died of multiple myeloma in 3 year, and 1 died of heart attack in 2 years after treatments, respectively. One lost follow-up., Conclusion: EMPs in head and neck are low potential malignancy tumors. The diagnosis of EMPs mainly depends on clinical manifestations and pathological results. Surgery and radiation therapy are the main treatments for EMPs in head and neck.

Objective: To report the experience of surgical treatment of inflammatory labyrinthine fistula (LF)., Method: Twenty-three patients with cholesteatomatous otitis media complicated with LF underwent mastoidectomy, surgical repair and obliteration of LF. After removal of cholesteatoma matrix and inflammatory lesion around fistula, the fistulae were treated as following procedures: fistulae less than 2 mm in diameter were overlaid with fascia (7 cases), those 2 to 3 mm were obliterated with bone chips and fascia (13 cases), and for those more than 3 mm, the bony semicircular canal lumen were packed with fascia and bone chips, then overlaid with fascia., Result: In 23 patients with LF, which included 22 lateral semicircular canal fistulae (SCF) and one lateral combined with posterior SCF, two patients lost follow-up and 21 cases were observed with an average follow-up period of 2.5 years. In this series of cases, vertigo disappeared in 17 and diminished in 2. With the exception of total deafness in two cases, the average auditory thresholds in speech frequencies remained unchanged, showing that the thresholds of air conduction (AC) and bone conduction (BC) were 67.1 dB HL and 37.1 dB HL prior operation versus 69.1 dB HL and 30.6 dB HL after operation, respectively., Conclusion: Because of structural barrier between superior and inferior labyrinth and inflammatory adhesions, surgical treatment of inflammatory LF is feasible and effective. Successful operation depends on excellent microsurgery skill, complete removal of inflammatory lesion, precise obliteration of fistula, post-operative administration of antibiotics and steroids and close follow-up. In addition to fistula test, axial petrous bone CT scan is a good complemental method in preoperative diagnosis of LF.

Objective: To evaluate the clinical features of chronic stenosed otitis externa (CSOE) and its treatment., Methods: Fourteen patients with CSOE from 1992 to 2000 were analyzed retrospectively., Results: In etiologic study, otitis externa and eczema were found in nine cases, chronic suppurative otitis media in three, acetic acid burn in one, and no apparent precipitating causes in one. Ten cases were cured through single operation, 3 experienced 2 operations and one had had 3 operations. The restenosis of external auditory canal occurred in four ears., Conclusions: CSOE is a rare disease. Surgical treatment is the main effective method for CSOE. The important items for treatment are: removing all stenotic fibrous tissue, widening bony canal, covering all bare bone with skin, preventing wound infection and close postoperative follow-up.
