With the rapid development of the e-commerce industry, RMFS is prevalent in the modern logistic center. As for this advanced system, robots can apply two following strategies: all-time following and non-following in a task. Alltime following strategy means that robot follows the picking rack until the task is completed, and non-following strategy refers to that robot will leave the rack to perform other tasks when the rack is queued at the workstation, and the robot will be dispatched when the rack needs to be moved. For the robot scheduling problem under two kinds of following strategies, aiming at the shortest total time for the robot to complete the wave tasks, the robot scheduling models under two kinds of following strategies are designed. The simulation experimental platform under two kinds of following strategies are designed based on genetic algorithm, and the system performance of all-time following strategy and non- following strategy is compared through simulation experiments. The decision-making curve of the following strategy based on the shortest total time is obtained, and it is proved that selecting the appropriate following strategy, which is combined with the specific working conditions, can effectively improve the operation efficiency of the picking system. To explore the inhibitory effect and mechanism of theaflavin (TF1), human hepatoma carcinoma Bel-7402 cells were treated with different concentrations of TF1. Cell viability was detected by CCK8 and cell proliferation was detected by colony formation assay. Cell migration was detected by cell wound healing experiment and transwell chamber assay. Cell apoptosis was detected by flow cytometry. Apoptosis-related proteins (Bax, Bcl-2, PARP), migration related proteins (E-cad, N-cad, Vimentin, MMP9) and signaling pathway related proteins (TGF-β, Smad3, p-smad3) were detected by western blot. The results show that different concentrations of TF1 could inhibit the proliferation and migration of Bel-7402 cells and promote their apoptosis in a dose-dependent manner. The higher the dose, the stronger the inhibition effect (P<0.05). Compared with the control group, the expression levels of Bax and E-cad proteins in the experimental group were higher, while the expression levels of Bcl-2, PARP, N-cad, Vimentin, MMP9, TGF-β, Smad3 and p-smad3 were lower (P<0.05). In conclusion, TF1 could inhibit the proliferation and migration of Bel-7402 cells and promote their apoptosis through TGF-β signaling pathway. [ABSTRACT FROM AUTHOR]

DW871 is a Brassica napus dwarf line with unusual and excellent plant type. To study the dwarf-related genes and their expression patterns of DW871. we took B. napus HW871 as control and selected stem tissues at the bolting stage for transcriptome gene expression analysis. Results showed that there were 8665 significant differentially expressed genes between DW871 and HW881. of which 2582 were up-regulated and 6083 down-regulated. The enrichment analysis of GO could be annotated to 22 subcategories in 3 categories. the enrichment analysis of KEGG was annotated to 295 pathways in 31 subcategories in 5 categories. We screened genes closely related to dwarfing traits from significantly different genes, 43 in pectin degradation, 69 in auxin signal transduction, and annotated 22 in the monolignol biosynthesis. It was found that the dwarfing character of DW871 was related to cell elongation, abnormal cell wall formation and cell division, and obstruction of auxin signal transduction. In particular, in monolignol biosynthesis, it is possible to regulate a series of physiological and biochemical processes related to lignin by regulating the expression of enzymes such as PAL and CCR. The differential genes related to dwarfing traits were screened in this research. It could be hypothesized that the dwarfing mechanism of DW871 involving abnormal auxin response and lignin synthesis, which lead to abnormal cell elongation, growth and cell wall changes, and ultimately lead to the dwarfing of DW871. LSW2018, a Brassica napus germplasm, has typical "dwarf" character at seedling stage, and the plant is short with thick stem and strong lodging resistance. As an ideal dwarf germplasm resource, LSW2018 is especially suitable for mechanized operation and has important utilization value. In order to explore the breeding advantages, and study the inheritance of dwarf character of LSW2018, 119 normal high-stalk Brassica napus cultivars (lines), 12 Brassica rapta cultivars (lines) and 6 Brassica juncea varieties (lines) were selected to construct cross combinations with LSW2018 (P1), and the populations of F1, F2, F3, aBC1 and bBC1 were analyzed. The results indicated that dwarf trait was controlled by a pair of dominant genes, plant height has major gene-polygenic genetic effect, and there is no cytoplasmic effect. [ABSTRACT FROM AUTHOR]

In this study, a hybrid combination was contracted with F1 of Deyou 5, a Brassica napus variety, as female parent and a new line 273R as male parent. In the F2 generation, 6 mutant plants with obvious shorter plant types at seedling stage were selected. After 5 generations of purification, we bred a stable new germplasm, LSW2018. The germplasm shows dwarf marker characters at seedling stage, with shrunken, thicker and darker leaves, shorter petiole. The growth is lightly weak but stable, plant height is significantly shorter than that of the normal variety. From the budding stage to maturity, LSW2018 gradually turned into a normal shape, with shorter internodes and thicker stalks, stronger lodging resistance, and lower branching positions. The flowering habit, branching angle, inflorescence characteristics, primary and secondary branches, grains, and other characteristics of LSW2018 are not significantly different from those of the homologous normal high-stalk line HSW2018, but the plant height and branch height are extremely different. The average transplanting plant height of LSW2018 is 136.24 cm and the direct seeding is 110.4 cm. In short, LSW2018 is considered to be a new short-stem Brassica napus germplasm suitable for mechanized and light-simplified planting, which might have high utilization value and broad application prospects. Objective: To systematically evaluate the efficacy and safety of levetiracetam (LEV) versus phenytoin (PHT) as second-line drugs for the treatment of convulsive status epilepticus (CSE) in children., Methods: English and Chinese electronic databases were searched for the randomized controlled trials comparing the efficacy and safety of LEV and PHT as second-line drugs for the treatment of childhood CSE. RevMan 5.3 software was used for data analysis., Results: Seven studies with 1 434 children were included. The Meta analysis showed that compared with the PHT group, the LEV group achieved a significantly higher control rate of CSE ( RR =1.12, 95% CI :1.00-1.24, P =0.05), but there was no significant difference between the two groups in the recurrence rate of epilepsy within 24 hours ( RR =0.82, 95% CI :0.22-3.11, P =0.77) and the rate of further antiepileptic drug therapy ( RR =0.97, 95% CI :0.64-1.45, P =0.87). There was no significant difference in the incidence rate of adverse events between the two groups ( RR =0.77, 95% CI :0.55-1.09, P =0.15)., Conclusions: LEV has a better clinical effect than PHT in the treatment of children with CSE and does not increase the incidence rate of adverse events.

This study aims to construct a eukaryotic expression system for envelope gene of Jaagsiekte sheep retrovirus, observes its localization in 293T cells, and investigates the potential in inducing malignant transformation of NIH3T3 cells. By RT-PCR, the full-length cDNA of envelope gene of Jaagsiekte sheep retrovirus (exJSRV-env) was amplified from the extract of naturally infected sheep lung. The clone of target gene was sub-cloned into eukaryotic expression system pEGFP-C1, and validated by PCR, restriction endonuclease, and sequencing. Bioinformatic analysis concerning biological function and cellular localiza tion of exJSRV-env was also performed. The recombinant clone of exJSRV-env was transfected into 293T cells and NIH3T3 cells by Lipofectamine LTX. The expression and celluar localization in 293T cells were validated by confocal microscopy. Soft agar colony formation assay was employed to test the anchorage-independent growth of NIH3T3. DNA sequencing and restriction enzyme digestion with Kpn I and Hind III indicated the correct construction of the recombinant plasmid, which was named pEGFP-C1/exJSRV-env. Amino acid sequence alignment of exJSRV-env with reference sequences found 85%-100% homogeneity. A YRNM motif was discovered at the cytoplasmic tail of envelope gene, which is exclusively found in exogenous viruses. Phylogenetic tree analysis showed that our clone of exJSRV-env clustered closely with pathogenic exogenous Jaagsiekte sheep retroviruses. Fluorescence microscopy indicated typical membrane localization of exJSRV-env protein. NIH3T3 cells transfected with exJSRV-env lost contact inhibition, and acquired colony forming ability in soft agar. This study indicated that envelope protein of Jaagsiekte sheep retrovirus can induce malignant transformation of mouse fibroblast cell NIH3T3. Discoveries of this study provide a basis for further structural and functional research on Jaagsiekte sheep retrovirus envelope protein.

Aim: To determine the most important region in tartary buckwheat allergen., Methods: The gene of epitopes was amplified by PCR using the primers designed according to TBa cDNA. Four expression vectors containing the gene of epitopes were constructed, and then transformed into the E.coli BL21(DE3) host cells. The expression products were purified by Ni(2+);-NTA agarose affinity chromatography column and indirect ELISA, inhibition ELISA and Dot blot was performed using sera from allergenic patients., Results: The purified proteins were obtained and the immunological results showed that E1 exhibite stronger IgE binding to patient's serum than the other epitopes., Conclusion: E1 is probably the most important region in tartary buckwheat allergen binding to buckwheat allergic sera IgE.

Aim: To prepare the rat antibody against the recombinant buckwheat trypsin inhibitor., Methods: The recombinant buckwheat trypsin inhibitor was expressed in E.coli. BL21 (DE3) and used as an immunogen to immunize the rat. The titer and specificity of the anti-BTI antibody from the rat were analyzed by ELISA, Western blot and immunohistochemistry, respectively., Results: ELISA detection indicated the titer of the antiserum was about 1:128 000. Western blot analysis showed the antibody reacted specifically with rBTI. The immunohistochemistry analysis proved that rBTI was expressed in the cytoplast and nucleus of EC9706 cells., Conclusion: The rat antibody against rBTI has high titer and specificity, which is beneficial to further study on the molecular mechanisms in rBTI-induced apoptosis of tumor cells and also provide an important theory basis for further exploring the relationship between the structure and function of BTI.

The study was purposed to investigate the apoptosis of HL-60 cells induced by recombinant common buckwheat trypsin inhibitor (rBTI) and its mechanism. The inhibition rate of rBTI on HL-60 cells was detected by MTT (3-[4, 5-dimethylthiazol-2-yl]-2, 5-diphenyltetrazolium bromide); the morphology of HL-60 nuclei was observed by fluorescence microscopy; the apoptosis cells of HL-60 detected by agarose gel electrophoresis and the changes of apoptosis rate was assayed by flow cytometry (FCM), when the HL-60 cells were treated with different concentration of rBTI for 24 hours. The results showed that the growth of HL-60 cells was inhibited evidently after treatment with rBTI in a dose-dependent manner, but there were minimal effects on normal human peripheral blood mononuclear cells (PBMNCs). The nuclei of HL-60 cells showed the characteristics of apoptosis, the analysis by flow cytometry indicated that the apoptosis rate of HL-60 cells was 52% after treatment with rBTI (100 microg/ml), DNA analyzed by agarose gel electrophoresis showed "ladder" pattern. It is concluded that rBTI obviously inhibits growth of HL-60 and induces its apoptosis which provides a foundation for use of recombinant common buckwheat trypsin inhibitor to cure the acute myeloid leukemia.

In the present research, suppression subtractive hybridization (SSH) and high density membrane techniques were employed to analysis genes induced by water stress in wheat seedling at 2-leaf stage. The purpose was to comprehensively understand the genetic bases of drought resistance and to find the key genes related to drought resistance in wheat. A total of 181 positive clones were obtained by screening the SSH library including 1 530 individual recombinant clones. The result of the sequence homologous comparison and function querying showed that 83.2% differentially expressed genes were high homologous with those induced by abiotic or biotic stresses in plant and their functions were to protect the cell directly or indirectly from stress strain. Seventeen differentially expressed ESTs found first were registered in GenBank. After further identifying by reverse Northern, RT-PCR and Northern, the gene expression profile induced by water stress in wheat seedling was preliminarily revealed.
