Objective To investigate the changes in serum thyrotropin receptor antibody (TRAb), thyroid stimulating antibody (TSAb), and thyroid-stimulating blocking antibody (TSBAb) and their significance in patients with Hashimoto’s thyroiditis with hypothyroidism (HT hypothyroidism). Methods A total of 133 patients with HT hypothyroidism (HT hypothyroidism group) and 125 healthy subjects (control group) were enrolled and their clinical data were collected. The serum levels of free triiodothyronine (FT3), free thyroxine (FT4), thyrotropin (TSH), and TRAb were measured and compared between the two groups. TSAb and TSBAb were also compared across the patients who were positive for TRAb (group B), who were negative for TRAb (group C), and the healthy subjects who were negative for TRAb (group A). A Pearson correlation analysis was used to examine the relationships between the serum levels of TSAb and TSBAb and the serum levels of FT3, FT4, TSH, and TRAb in patients with HT hypothyroidism. Objective To report the clinical and pathological features of 4 patients with oculopharyngodistal myopathy type 1 (OPDM1) caused by CGG repeat expansion in the 5'untranslated region (5'UTR) of LRP12 gene. Methods and Results This study included 4 patients with OPDM1 who were admitted to Peking University First Hospital from January 2008 to August 2023. All 4 patients presented with weakness of eyes closed, dysarthria, dysphagia, facial muscle weakness and muscle weakness mainly involving distal muscles. None of 4 patients had restricted eye movements. Two of the patients had ptosis (Case 1 and Case 2). Serum creatine kinase (CK) levels were mildly to moderately elevated (190- 1211 U/L) in 4 patients. Three patients (Case 1, Case 2 and Case 3) underwent lower extremity muscle MRI which showed varying degrees of fatty infiltration, with the soleus and tibialis anterior muscles being worst affected. Skeletal muscle biopsies in all 4 patients showed rimmed vacuoles with muscular dystrophy‑like pathological changes. Immunofluorescence staining revealed p62 ‑ positive and ubiquitin ‑ positive intranuclear inclusions in all 4 patients. Genetic testing revealed that all 4 patients carried CGG repeat expansion in the 5'UTR of the LRP12 gene. Conclusions Most OPDM1 patients start with weakness in the distal extremities of both limbs and dysarthria. The involvement of muscles in the lower extremities is asymmetrical, and the involvement of tissues and organs other than muscles is not obvious. The remaining clinical phenotype and pathological features are consistent with other subtypes of OPDM.

Short tandem repeat (STR) is a component of the human genome, the number of repeats of STR is prone to change during DNA replication, and when it exceeds a certain threshold, it will lead to the occurrence of diseases, most of which involve the nervous system. With the rapid development and wide application of the third generation long read long sequencing technology, more and more neuromuscular diseases have been confirmed to be related to STR variation. This article reviews the clinical and mechanism research progress on STR‑related neuromuscular diseases, so as to provide guidance for clinical research.

Objective To explore the effect of enolase1 (ENO1) expression on maintaining the self-renewal activity of human embryonic stem cells (hESCs) and reveals its mechanism. Methods RNAs and proteins bound to ENO1 were predicted and analyzed through Gene Oncology (GO) enrichment. Endogenous expression of ENO1 was inhibited by shRNA in hESCs. The colony forming of hESCs was examined by clonogenesis and alkaline phosphatase (AP) staining experiments. The pluripotency and differentiation marker gene expression in hESCs were detected by qPCR. RNA fluorescence in situ hybridization was used to detect the localization changes of poly (A) + RNAs. Results Inhibition of ENO1 endogenous expression significantly inhibited colony forming of hESCs (P＜0.05)and the expression of differentiation marker genes was increased (P＜0.001). Localization of poly(A)+ RNA changed from cytoplasm to nucleus (P＜0.01) with ENO1 inhibition. Conclusions ENO1 regulates the activity of hESCsself-renew by regulating mRNA localization.

Objective To delineate RNA binding profile of NAT10(N-acetyltransferase 10) in human chronic myelogenous leukemia cell line K562. Methods Enhanced UV crosslinking and immunoprecipitation (eCLIP) were used to capture the transcript collection bound by NAT10 in K562 cells. Data were obtained through high throughput sequencing and then analyzed with bioinformatics. The type and regions of NAT10 binding genes were identified via peak-calling and annotation. The function of the binding RNA was further explored by gene function enrichment analysis. Results NAT10 binding transcripts which were enriched in 3′UTR region, were mainly protein coding genes. Further analysis showed that they were functionally associated with DNA damage and repair. Conclusions NAT10 may regulate gene expression by binding to the 3′UTR region of mRNA associated with DNA damage repair.

Objective To investigate the effect of cytoplasmic poly(A) binding protein-1(PABPC1) on translation of human monocytic leukemia cells line THP-1. Methods Specific short hairpin RNA(shRNA) was transfected into THP-1 cells. Western blot was used to verify the interference efficiency. The polysome profiling was used to examine the translation ability of THP-1 cells. Results The expression of PABPC1 protein was decreased in shPABPC1 transfected THP-1(P＜0.05). Compared with control group, the translation ability of THP-1 cells in shPABPC1 group was reduced. Conclusions Knockdown of PABPC1 may inhibit the translation in THP-1 cells and PABPC1 is a potential target for the treatment of acute myeloid leukemia (AML).

Compared with the advanced and sophisticated industrial production, agriculture and food production appear backward and sloppy, and agricultural production management methods are lacking. The fourth industrial revolution, represented by information technologies such as intelligent perception, Internet of Things, block chain, big data and artificial intelligence, has empowered traditional industries, and agriculture has stepped into a new digital, intelligence and even wisdom, which could provide a new opportunity to improve agricultural production management. Existing agricultural production management research is mostly limited to the study of concept definition, applicable theory and other issues. The development of information technology has built up a low-level Internet of Things with certain coverage, which brings some convenience, but cannot effectively support agricultural management and decision-making due to data silos, data waste, data abuse, data falsification and other thorny issues. From the perspective of top-level design and system management, this paper addressed the problems in the development of smart agriculture, conceptualized the characteristics of the event and natural event chains in the agricultural industry chain. This paper also proposed for the first time an event-correlation management approach and its wisdom strategy. The implementation of the event-correlation method included five aspects, including goal alignment, index systematization, asset linkage, process linkage and effect traceability. The application of the event-correlation management approach and its wisdom in the 5T management standard designation for Jilin rice brand building and the “six-step fresh rice precision control technology” innovation system of Wilmar rice industry have shown that it can improve the culture of rice cultivation, and systematically reduce losses to improve quality and increase economic benefits.

The influence of science and technology on cognitive change has a long history, which lead to forming philosophy of science and philosophy of technology respectively. The development of grain storage technology in China has gone through three stages: the initial exploration before the reform and opening up, the construction of intellingence in the past 10 years modern granaries in 30 years after the reform and opening up, and the creation of digital. The scientific and technological progress of grain storage in China has been witnessed by all, including building a modern granary with "four in one" new grain storage technology, to forming the world's largest grain and food networking with information technology, original multi-field coupling and bio-field theory, and realizing the dynamic supervision of grain reserves with grain cloud image scanning technology. In the new era, the political and economic situations have not changed significantly in the past century, and the new requirements of China's second centenary goal for food security and the impact of the fourth industrial revolution have made the trend of food science and technology becoming more and more intelligent. Based on the accumulation of scientific and technological achievements in grain storage practice for more than 40 years in China, combined with the team's research on multi-field coupling and bio-field theory of grain storage, dynamic supervision of cloud image of grain reserves, 5T information management of natural block chain for grain storage, and intelligent EAT window measurement and control methods for grain drying, this paper attempted to take the basic idea of "one divides into two, two divides into three, and three generates changes" on the basis of deconstructing the micro and macro ternary microstructure of grain storage system. Taking the joint driving of mechanism and data as the intelligent strategy, the granary philosophy is introduced to improve the recognition of existing scientific and technological achievements. The granary philosophy also guided the exploration of grain storage technology in the new era, and provided support for building a high-quality intelligent food system. The granary philosophy is a deep reflection on the practical situation of grain storage, and has certain universality, which originates from grain storage practice, but is not limited to grain storage practice.

In the optimization design of twin-screw compressor line, the influence of geometric parameters on compressor performance is considered to be different. In order to judge the advantages and disadvantages of the designed profile more directly, proposed a comprehensive performance index to judge the advantages and disadvantages of the rotor profile, and designed a rotor with better profile based on the comprehensive performance index for fluid simulation and experimental verification. Compared with the original profile, the length of the contact line of the optimized profile increases slightly, the area of leakage triangle decreases, area utilization coefficient increased. But the single performance index which reflects the whole performance index is better, which also preliminarily proves the correctness of the proposed method.

Background and purpose: Anti-programmed death-1 (anti-PD-1) adjuvant therapy and targeted therapy have resulted in longer recurrence-free survival (RFS) for malignant melanoma patients in clinical trials. However, the efficacy of adjuvant anti-PD-1 therapy in Chinese melanoma patients has not been previously reported. The purpose of this study was to retrospectively analyze the RFS in Chinese resected stage Ⅲ malignant melanoma patients treated with adjuvant anti-PD-1 therapy and to preliminarily compare it with BRAFV600 inhibitor vemurafenib. Methods: This study retrospectively collected the information of 120 patients with resected stage Ⅲ malignant melanoma from three major cancer centers in China from August 30, 2018 to August 30, 2020. These patients received adjuvant pembrolizumab/toripalimab or vemurafenib. RFS rates were assessed by Kaplan-Meier curves and log-rank tests were used to examined differences. Results: Among the 90 patients with stage Ⅲ malignant melanoma receiving adjuvant anti-PD-1 therapy, the median-RFS (mRFS) was 16 months (95% CI: 14-18 months); the 12-month RFS rate was 69.5% (95% CI: 64.6%-74.4%) and the 24-month RFS rate was 45.8% (95% CI: 40.5%-51.1%). The mRFS for 30 patients who received vemurafenib was 18 months (95% CI: 10-21 months), the 12-month RFS rate was 62.8% (95% CI: 54.0%-71.6%), and the 24-month RFS rate was 32.9% (95% CI: 27.1%-44.7%). In the stratified analysis, firstly, there was no difference in RFS between anti-PD-1 therapy group and vemurafenib group. Secondly, the BRAFV600E mutation status may did not affect the RFS of anti-PD-1 therapy. In addition, in this study, for the patients with BRAFV600E mutation, the RFS showed no difference between these patients received anti-PD-1 therapy or vemurafenib. Finally, although the RFS was better in patients with cutaneous melanoma than in patients with acral and worst in patients of unknown origin, the data for adjuvant therapy in China were still poor. Conclusion: These are the real-world data from adjuvant anti-PD-1 therapy in patients with resected stage Ⅲ cutaneous and acral melanoma patients in China. In our study, adjuvant anti-PD-1 therapy is indistinguishable from vemurafenib indicates more combination therapies are needed to improve outcome in China.

Developing large-scale complex information systems, such as the Smart Court system-of-systems (SoSs) of China, is a worldwide engineering challenge. This paper, from a methodological perspective, aims to expound the theoretical construction and practical progress of Smart Court system-of-systems engineering (SoSE) of China. The concept and key task requirements of SoSE are explored, technical difficulties faced by the Smart Court SoSE are analyzed, and a “two-track parallel, six-ring linkage” pattern framework is proposed for the progressive collaboration SoSE of large-scale autonomous information systems. Based on the theories including a universal information model, information metric system, and dynamic configurations of information systems, a key evaluation indicator system for an information SoSs is proposed. To satisfy the SoSE design requirements, an overall design method based on information relationships and its enabling tool are proposed, and a reference model of the Smart Court SoSs is designed to provide a top-level reference for the system development and integration of the Smart Court. Moreover, the development and collaborative integration of the autonomous and backbone systems in the Smart Court SoSE are presented in a comprehensive manner. The nationwide application and promotion of the Smart Court SoSs support the upgrade and transformation of the conventional judicial operation pattern of people's courts in China. Through continuous analyses of the quality and effectiveness of the Smart Court based on the key evaluation indicators, targeted improvement can be conducted to further enhance the SoSs capabilities, thereby contributing to the progress of judicial civilization in the information age.

Objective To detect the pathological changes in the liver of mice with non-alcoholic fatty liver disease (NAFLD) and explore an experimental method to obtain a large number of primary hepatocytes with high activity and purity from the mice with NAFLD. Methods A total of 60 mice were randomly divided into control group (chow diet, CD) and high-fat group (high-fat diet, HFD) and had been fed separately for 20 weeks. The body weight of the mice was regularly measured and the pathological characteristics of the liver tissue were observed by hematoxylin-eosin (HE) staining and Sirius red staining. After 15 weeks of high-fat diet, 6-8 mL collagenase Ⅳ were added for digestion to obtain mixed hepatocytes, which were spread on 40% Percoll separation solution and subjected to density gradient centrifugation to obtain purified hepatocytes. The viability of the cells was determined by trypan blue staining, and the morphology was observed by microscope. Results With the prolongation of modeling time, the body weight of mice in the HFD group were gradually increased, which was significantly higher than that in the CD group. In the HFD group, the liver of mice gradually developed fatty degeneration, and liver fibrosis could be detected at week 20. At week 15, about 5×107-7×107 hepatocytes could be obtained from each NAFLD mouse with cell viability ≥85% and cell purity ≥95%. Cells were round or elliptical, with smooth edges and few clusters. The number and quality of cells obtained were significantly improved by optimizing conditions such as accurate temperature control, reduced the concentration of collagenase, and the use of low-speed centrifugation and low-speed density gradient centrifugation. Conclusions The NAFLD mouse model was successfully established and the pathological characteristics of liver in different disease stages were revealed. An efficient and stable method for obtaining primary hepatocytes of NAFLD mice was established. The number and activity of hepatocytes obtained met the requirements of most experiments.

Objective Single-cell RNA sequencing (scRNA-seq) was used to systematically delineate the cell atlas and molecular tracks of human early erythroid development at the single-cell level. Methods The dataset of scRNA-seq of human embryo yolk sac at CS15 stage was analyzed by principal component analysis (PCA), cluster analysis and pseudostime analysis to observe the expression of marker genes in different cell clusters, and to predict the occurrence time and development path of erythroid. Results In different cell clusters of yolk sac at CS15 stage, marker genes of erythrocytes and progenitor cells were differentially expressed. The occurrence time of various cells was different, and there was some heterogeneity in erythrocytes in the yolk sac. There were abundant types of cells in the yolk sac at CS15 stage, which could include vascular endothelial cells, hematopoietic progenitor cells, granulocytes, different status of red blood cells and monocytes. Conclusions It is speculated that early development path of human erythroid is as follows: vascular endothelial cells differentiate into hematopoietic progenitor cells, further differentiate into red myeloid progenitor cells, and finally produce megakaryocytes and erythrocytes.

Recent research progress in curing system of silicone rubber was reviewed from the optimization of traditional vulcanization system and the development of new vulcanization system with 53 references. The advantages and disadvantages of organic peroxide curing systems, the effect of the catalysts and inhibitors on hydrosilylation curing systems, and the effect of the crosslinking agents and catalysts on functional group condensation curing systems were analyzed, and the new curing systems including radiation-induced curing systems, photoinitiated curing systems, curing systems based on click chemistry, and reversible covalent (non-covalent) curing systems were briefly introduced. Finally, some suggestions on the future development of silicone rubber curing system were also put forward.

Objective To investigate the biological function of RNA binding protein polycytosine-binding protein 1 (PCBP1) in the maintenance of pluripotency of human embryonic stem cells (hESC) and to explore the new mechanism of regulating hESC pluripotency. Methods PCBP1 knockdown technology was used to detect the clonogenicity. The alkaline phosphatase staining profile and the expression of pluripotency/differentiation marker genes were detected by qPCR. Meanwhile, the nucleus and cytoplasm of hESC were isolated for RNA sequencing, and the nucleoplasmic changes of RNA generated by PCBP1 knockdown were analyzed by multi-level analysis. Results PCBP1 knockdown significantly inhibited clonogenesis of human embryonic stem cells (P＜0.01) and reduced the RNA level of pluripotency related genes (P＜0.01). The expression of differentiation related genes increased(P＜0.01)that significantly affected the overall nucleoplasmic distribution of RNA in hESC and the nucleoplasmic distribution of mRNA related to embryonic development and cell differentiation. Conclusions PCBP1 regulates hESC pluripotency by regulating the expression of pluripotency/differentiation marker genes and the nucleoplasmic localization of mRNA.

Countersunk bolt joints with sleeves have been used in composite connection structures, and their fatigue performances need to be studied. Based on the experimental investigation conducted on the single lap CFRP/titanium alloy bolted joint with 3-countersunk head bolt and sleeves, the fatigue models of composite laminates and metal structures were established to analyze the fatigue performances of the structure, and finite element analysis on joints without sleeves was also carried out to study the influences of the sleeves on fatigue performance of the joint. The results show that the use of sleeves can more effectively improve the fatigue performance of the joint compared to that only using interference fit. The fatigue life of laminate is increased by about 3.6 times, the titanium alloy plate increased by about 2.7 times, and the bolts are increased by about 14 times. Only the titanium alloy plate of the joint fails, but not the fastener. Combined with analysis on the experimental results, it is found that the fatigue failure mode of bolted joints structure will change with different load levels due to the different fatigue properties between composite materials and metal materials; and the metal structure is prone to be damaged under a certain load level.

Coagulation factor ⅩⅢ (FⅩⅢ) is a member of the plasma transglutaminase family.Plasma FⅩⅢ (pFⅩⅢ)consists of two catalytic A subunits and two non-catalytic B subunits(FⅩⅢ-A2B2)combined with non-covalent bonds to form a heterotetramer.The cellular FⅩⅢ(cFⅩⅢ) exists in a form of A subunit homodimer(FⅩⅢ-A2).Although it is currently believed that cellular FⅩⅢ can be internalized and externalized from the cell through an unknown mechanism and cross-linked extracellular substrates to function in a variety of pathophysiological events,its specific functions and mechanisms need to be clarified by further systematic research.

The intensive aquaculture-induced environmental pollution has impeded the sustainable and healthful progression of the aquaculture industry. In order to explore a more sustainable and efficient aquaculture model, we enhanced the circulating aquaculture system in freshwater ponds in this study. The physicochemical indexes and nitrogen and phosphorus budget of ponds were analyzed during the 5-month experiment period. The results showed fluctuating changes in the physicochemical indexes of pond water, with total nitrogen and total phosphorus concentrations measuring 4.85 and 1.04 mg/L, respectively, at the end of the experiment. Notably, the total nitrogen concentrations met the secondary standards for freshwater aquaculture tailwater discharge. Feed and sediment emerged as principal contributors to nitrogen and phosphorus input, accounting for 50.6% and 43.7% of nitrogen input and 49.4% and 46.9% of phosphorus input, respectively. Sediment accumulation emerged as the primary mechanism for nitrogen and phosphorus output in ponds, accounting for 53.3% and 78.7% of nitrogen and phosphorus output, respectively. The nitrogen and phosphorus utilization rates in experimental ponds were determined to be 65.2% and 16.6%, respectively. The utilization rate of nitrogen and phosphorus of carp (Cyprinus carpio) were significantly higher than those of other fish (P<0.05). However, there was no significant difference in the phosphorus utilization rate between silver carp (Hypophythalmichthys molitrix) and bighead carp (Aristichthys nobilis) (P>0.05). The research shows that the enhanced pond circulation aquaculture system can effectively mitigate nutrient load, enhance the nitrogen and phosphorus utilization rate of aquaculture organisms, diminish the impact of aquaculture tailwater on the surrounding environment, and concurrently consider ecological and economic benefits. Cyclophilin A (CypA) serves as a principal receptor of Cyclosporin A (CsA) within cells, exhibiting considerable biodiversity and playing an important role in immune system. In this study, the full-length cDNA sequence (GenBank ID: MN121742) of CypA of Hyriopsis cumingii was obtained using PCR and RACE technology. The cDNA sequence spans 1237 bp, comprising a 57 bp 5' noncoding region (UTR), a 688 bp 3' noncoding region, and a 492 bp open reading frame, encoding a total of 164 amino acids. The theoretical isoelectric point of the CypA cDNA sequence is 8.9, and the relative molecular weight is 17.29 kD. A search for homologous sequences in GenBank revealed high similarities between CypA amino acid sequence of Hyriopsis cumingii and those of other shellfish species, including Hyriopsis schlegelii (99.39%), Cyclina sinensis (77.44%), Ruditapes philippinarum (78.05%), Crassostrea ariakensis (78.66%), and Solen grandis (75.05%). Utilizing SWISS-MODEL, it was predicted that CypA of Hyriopsis cumingii contains 8 p-pleated sheets and 2 a helixes, potentially forming the active central region of CypA. A multiple sequence comparative analysis was carried out by using the MEGA 5.1 Software, and a phylogenetic tree of the genetic system was constructed employing the neighbor-joining method. The analysis revealed that CypA of Hyriopsis cumingii clustered with CypA of shellfish from the same genus, particularly exhibiting a close genetic relationship with CypA of Hyriopsis schlegelii. The expressions levels of CypA in different tissues of Hyriopsis cumingii were assessed through qRT-PCR, indicating the highest expression in the gonad, followed by the liver, kidney and gill. Upon exposure to Aeromonas hydrophila, a bacterial infection, expression levels of CypA in gonad, liver, gill, and intestines exhibited an initial increase followed by a subsequent decrease within the first 72h of infection. Specifically, peak expression levels were observed at 6h in gonad and gill, at 12h in liver, and at 24h in intestines. This proves that Aeromonas hydrophila infection significantly induce the expressions of CypA in Hyriopsis cumingii. Expression level of CypA in gonad reaches the peak earlier than that in liver and intestines. Moreover, the relative expression times (14.91) of CypA in gonad at peak is significantly higher than those in liver (9.89), intestines (9.78) and gill (7.53). It is speculated that CypA plays an important role in defending against bacterial infections in gonad, which may not only serve reproductive functions, but also contribute significantly to immune defense. Objective To explore the biological function of RNA binding protein heterogeneous nuclear ribonucleoprotein A3(hnRNPA3) in the maintenance of pluripotency of human embryonic stem cells(hESCs) and to reveal its new mechanism in RNA transport. Methods The interaction of hnRNPA3 with nuclear pore and localization were examined by immunoprecipitation of nuclear pore complex and cytoplasmic separation in hESCs. After inhibiting the endogenous expression of hnRNPA3, the expression of the pluripotency/differentiation marker genes was detected by qPCR, and the effect on the nucleoplasmic distribution of polyA+ RNA and OCT4 mRNA was tested by RNA fluorescence in situ hybridization to predict the binding of hnRNPA3 to known RNA export associated proteins. Results hnRNPA3 was localized in the nucleus and interacted with the nuclear pore. Inhibiting the endogenous expression of hnRNPA3 significantly inhibited the cloning-formation ability of hESC(P＜0.05) and resulted in the increase of the expression of cell differentiation markers(P＜0.001) and significantly inhibited the export of polyA+ RNA as well as OCT4 mRNA(P＜0.001). The protein interaction network predicted that hnRNPA3 may bind to about one-third of the known RNA export associated proteins. Conclusions hnRNPA3 regulates hESC pluripotency by impacting on mRNA export.

Objective To establish an inducible CRISPR library screening method for human embryonic stem cells (hESCs) to screen the cis-acting elements involed in the maintainance the pluripotency of hESCs. Methods The inducible Cas9 expression elements were inserted into the hESC genome to construct an inducible Cas9 stable expression strain (iCas9-hESC). Analyze the published chromatin interaction information in hESCs to identify the cis-acting elements that may be involved in the regulation of pluripotency genes, then to construct a double gRNA CRISPR knockout library. The CIRSPR library was used to infect iCas9-hESC at low multiplicity of infection followed by induction of Cas9 expression, and gene editing of the cells. Using the expression level of OCT4 as a standard to measure pluripotency, the cells after gene editing were stained with OCT4 and analyzed by flow cytometry to determine whether the knockout of cis-elements would affect the pluripotency of hESCs. Results The iCas9-hESC was successfully constructed, and the cells still maintained a pluripotent state. Construct a CRISPR knockout library of ciselements and successfully infect the target cells. Flow cytometric analysis of the expression level of OCT4 was performed on the cells after the library infection, and it was found that there were indeed some populations with decreased pluripotency in the cells after gene editing. Conclusions A feasible scheme for screening cis-acting elements involved in maintaining pluripotency in hESCs through CRISPR library is established.

Objective To investigate the translational regulation of human embryonic kidney (HEK)293 cell line. Methods Polysome profiling coupled with Western blot was used to verify the translational activity of HEK293 cells. RPL10A and RPS17 RNA immunoprecipitation coupled with sequencing (RIP-seq) and bioinformatic analysis were used to investigate the actively translating mRNAs. Results A total of 129 genes were found in the RNA immunoprecipitation experiments RPL10A and RPS17, among which 93.8% were protein-coding genes and enriched in RNA splicing, metabolism, MAPK signal pathway, chromatin localization and RNA transport. Conclusions A variety of molecules involved in HEK293 cells translation are obtained by RPL10A and RPS17 RIP-seq analysis.

Objective To investigate the expression of Met-tRNAiMetcarrier proteins eIF2A, eIF2D and MCTS1 in normal hematopoietic stem cells (HSCs) and acute myeloid leukemia (AML) cells, and to study the effect of carrier proteins on the functions of AML cell lines. Methods The expression of carrier proteins among multiple stages of mouse HSCs differentiation was analyzed by public datasets. The expression of carrier proteins among multiple stages of human HSCs differentiation was analyzed in single cell RNA-sequencing datasets. The expression of carrier pro- teins among AML patients and normal was analyzed in public datasets. eIF2A or eIF2D expression was inhibited by lentivirus-mediated gene transduction in AML cell line MOLM13, cell proliferation and cell cycle was analyzed.AML cell line was treated with salubrinal (10 μmol/L) and serum starvation, which make cells in stress, and the expression of eIF2A and eIF2D were detected. Results The expression of eIF2A and eIF2D in hematopoietic stem and progenitor cells (HSPCs) was higher than mature blood cells, and in AML patients was higher than normal. On the contrary, the expression of MCTS1 had no difference in normal hematopoietic differentiation process, as well as AML vs normal. eIF2A or eIF2D knock down significantly suppressed cell proliferation (P＜0.0001), as well as caused cell cycle arrest in G2/M (P＜0.001). After MOLM13 was treated with salubrinal and serum starvation, the expression of eIF2A and eIF2D was upregulated. Conclusions eIF2A and eIF2D may regulate AML cell growth through breaking cell cycle arrest.

China is a country with a high incidence of esophageal cancer. The pathological type is mainly squamous cell carcinoma. Squamous intraepithelial neoplasia is the most recognized precancerous lesion of esophageal squamous cell carcinoma, and its monitoring and intervention is an effective method to reduce the incidence of esophageal squamous cell carcinoma and improve the quality of life of patients. Understanding the etiology, clinical features, diagnosis and treatment of esophageal squamous cell carcinoma plays a crucial role in the prevention and early diagnosis and treatment of esophageal squamous cell carcinoma. At present, the clinical research related to esophageal squamous intraepithelial neoplasia is still insufficient, and there are some differences in clinical treatment. This review summarizes the risk factors, clinical features, diagnosis, prognosis and treatment of esophageal squamous intraepithelial neoplasia, hoping to provide ideas for the clinical management of esophageal squamous intraepithelial neoplasia. Chinese liquor was made from sorghum, corn, wheat, rice, and glutinous rice with high starch content as raw materials, and was prepared through multiple processes. The amount of starch in the raw material was directly related to the liquor yield. In order to effectively shorten the time for detecting starch content in grain and reduce human error during operation, a fast method for detecting starch content was established. In the sample pre-treatment stage, fast microwave digestion instead of reflux acid hydrolysis is used to speed up starch hydrolysis and improve the completeness of hydrolysis. In the sample detection stage,instrumental detection instead of artificial micro-boiling titration is used which improves the ease of experimental operation. In this paper, the starch content of a variety of brewing grains was tested using the rapid starch detection method, and compared with the results of the national standard method GB 5009.9—2016. There were no significant differences between the results of the two detection methods. The rapid starch detection method can be applied to the detection of total starch content in a variety of brewing grains,greatly shorten the detection time, reduce the artificial operation error, and have good repeatability and reproducibility. Objective To investigate the localization of RNA in cytoplasm and nucleus of human embryonic stem cells (hESCs). Methods After separating cytoplasm and nucleus of hESCs, RNAs were extracted for sequencing, and the localization characteristics of RNAs were analyzed by bioinformatics. RT-qPCR was used to verify the localization of these RNAs. Results More RNA types were found in the cytoplasm than nucleus, as well as specifically locational RNAs (2 952 in the cytoplasm and 634 in the nucleus, respectively). Conclusions The localization map of RNA in cytoplasm and nucleus of hESCs is described,and the localization of many RNAs is verified.

Objective To investigate the characteristics of serum sodium disorders in children with different QST types of craniopharyngioma (CP). Methods There were 48 children underwent resection of CP from January 2016 to June 2017 were analyzed retrospectively. Statistical methods were used to evaluate the differences in serum sodium disorders among QST classification following removal of CP. Results There were 27 cases (56.25% ) had postoperative hyponatremia, including 12 cases (25% ) of moderate or severe, and 32 cases (66.67%) of hypernatremia, of which 16 cases (33.33%) were moderate or severe. There were significant differences in incidence of hyponatremia ( χ 2 = 11.420, P = 0.003) and hypernatremia (χ2 = 15.702, P = 0.000) in children with different QST types of CP. Among them, incidence of moderate ⁃ severe hyponatremia ( Z = ⁃ 3.324, P = 0.001) and moderate ⁃ severe hypernatremia ( Z = ⁃ 3.874, P = 0.000) of type T was significantly higher than that of type Q. Logistic regression analysis showed that QST classification was an influential factor for moderate ⁃severe serum sodium disorders after operation, and type T was a risk factor ( OR = 315.422, 95% CI: 16.804 5920.786; P = 0.000). Conclusions Long ⁃ term monitoring of serum sodium levels is necessity in children after removal of CP. Grasping the QST classification is in favour of comparing and predicting the regularity and severity of postoperatively sodium imbalance. Sorghum is the main raw material for Chinese liquor brewing, and the brewing Strong or Sauce-flavor liquor has more special requirements for the original cooking properties. In order to shorten the time and reduce the human error effectively during the operation of amylopectin detection, a rapid detection method was established for the detection of amylopectin content in this experiment, the performance evaluation of the method and instrument was carried out, and the rapid detection method was compared with the national standard method, the results showed that there was no significant difference between the two methods.

The priority during speckle reduction in Polarimetric Synthetic Aperture Radar (Pol-SAR) data is to maintain the polarization information. For this reason, it is undesirable to separate each element of polarimetric SAR. Based on the multiplicative noise model and complex matrix, an effective distance similarity measure method is derived, which can process covariance matrix C or coherent matrix T directly. Experiments show that this method can effectively reduce speckle and maintain the polarization information, point targets, and texture structure. Moreover, it can handle large polarimetric SAR with simplicity and ease.

Rumen microorganisms determine the optimal utilization rate of nutrients, and Prevotella is one of the most representative bacteria in the rumen biological system. Prevotella is involved in food digestion and metabolism of complex forms of nutrients such as cellulose and protein, and can cooperate with other forms of bacteria in the digestive tract of body. It promotes the biodegradation of some forms of nutrients by enzymes. The paper summarizes the research results of Prevotella at home and abroad, summarizes its classification and growth conditions, and focuses on the role of Prevotella in digestion and metabolism in rumen of ruminants. The paper introduced a design scheme of motor protection device based on dual-CPU of DSP56F807 and W78E516B, and analyzed circuit designs of main control board and man-machine interaction board and protection algorithm of the device. The device uses DSP56F807 to realize functions of multi-channel AD conversion, electricity-quantity calculation, kinds of protection algorithm, real-time clock control, data storage, CAN communication and output control of switching value of relay, uses W78E516B to realize function of man-machine interaction, and uses Fourier algorithm that can compensate any decaying time constant and filtrate decaying DC component to improve precision and correctness of motor protection algorithm. The experimental result showed that over-current protection algorithm of the device is good in sensibility and reliability with better running effect.

Background & Problems: The abdominal drainage tube is an important device used in disease treatment and life maintenance. Drainage tube slippage leads to complications that increase both length of stay and costs of care. Four and seven cases of drainage tube slippage were reported, respectively, in 2018 and 2019 in our trauma wards. Among these, 9 cases were under the care of nurses in their post graduate year (PGY) training program. Purpose: To increase to 94% the abdominal drainage tube care-completeness rate of nurses in the PGY program. Methods: Methods used included: establishing a standardized care module for abdominal drainage tube in the trauma wards, using the "drainage tube model" and multimedia teaching material in education to enhance skill proficiency, flexibly adjusting education schedules, using a creative-thinking teaching model in education, employing direct observation to evaluate PGY nurses' abdominal drainage tube skills; and establishing a drainage tube skill proficiency audit mechanism. Results: After the intervention, the rate of care completeness for the abdominal drainage tube rose to 98.1%, participant awareness rose to 100%, and the rate of abdominal drainage tube slippage reduced to 0%. Conclusions: This project achieved good outcomes that may be expanded horizontally to other surgical wards. The use of the creative-thinking teaching model in training activities received good feedback from the nurse participants and will be incorporated into in-service education standards along with the computerized direct observation of procedural skills in the PGY e-learning passport to strengthen the completeness of learning processes. Meteorological disaster is one of the main factors restricting agriculture development in China. It is important to clarify the risk of summer maize agrometeorological disaster for disaster prevention and mitigation. Based on the natural disaster risk theory, we collected meteorological data and maize yield data from 1981 to 2018 in a typical area in the northern mountainous area of Sichuan Basin (Wangcang County). The main disaster-causing factors affecting summer maize production were determined. A comprehensive agro-meteorological disaster risk assessment model for summer maize was constructed in combination with the sensitivity of pregnant environment and vulnerability of disaster bearing to evaluate the agrometeorological disasters risk of summer maize production in the northern mountainous area of Sichuan Basin. The results showed that during the study period, high temperature in mature period, rainstorm in flowering period, rainstorm in mature period, continuous rain in filling period and drought in booting stage were the main agrometeorological disasters affecting the growth and development of summer maize. The agrometeorological disaster risk of maize generally distributed in a southwest-northeast pattern, with the distribution areas of high-risk and higher-risk areas accounting for half of the total area of Wangcang County. The high-risk areas were mainly located in the southwest of the county, which was basically consistent with the high-value areas of hazard-pregnant environment sensitivity. The low-risk areas were mostly concentrated in the western part of county territory, which were also low-risk areas of high temperature in mature period, rainstorm in mature period, and rainstorm in flowering period disasters. In order to study the influence of the linear shaped pier on the inflow into the curved forked river, this paper uses the Tongcheng Sluice as the research model. Firstly, the most complex flow pattern is selected from different working conditions, and then based on the worst flow pattern, a linear shaped pier is set at different positions in front of the gate and the bend. The flow field parameters are respectively measured in Tongcheng Sluice's curved model, and the angle index between the inlet velocity vector and the direction of the gate pier is established to analyze the state of the curved forked river. At the same time, the FLUENT software is used for numerical simulation verification. Tecplot software is used for post-processing analysis. Finally, the optimal rectification scheme is selected based on the physical model. The results show that the angle deflection coefficient between the gate inlet velocity vector and the gate pier direction can directly evaluate the pros and cons of the inlet flow state. A linear shaped pier is placed in front of the gate. As the linear shaped pier is closer to the gate, the flow pattern in front of the gate will be more chaotic. In addition, the vortex in the bend can be significantly improved by setting a linear shaped pier in the bend, and the linear shaped pier is arranged 20 m from the convex bank on the center line of the bend, and the flow state in front of the bend and gate are significantly improved. Under the combined influence of climate change and highly intensive anthropogenic activities, the natural hydrological regimes of rivers have changed to some extent, interfering the structural stability and the function of riverine ecosystems. Ecological water requirement assessment is very important to the maintenance of river ecosystem health and is one of the important research contents in water science. The improved monthly frequency calculation method based on the Copula function is proposed to calculate the ecological flow and applied in a case study of the Qujiang River. The joint distribution between the monthly average flow and the annual average flow is constructed by using the Copula function. Based on the correlation between the conditional probability of the monthly average flow and the joint probability, the conditional probability target of the monthly average flow in the dry year is set up, and the joint probability is calculated. The design frequency of the monthly average flow is calculated by the joint probability function, and the design value of the monthly average flow is analyzed according to the marginal distribution function and checked for reasonableness. The results show that the Clayton Copula function has a good fitting effect on the joint probability distribution between the monthly average flow and the annual average flow of the Qujiang River. The basic ecological water requirement of the Qujiang River is 2.12 billion m³, accounting for 36% of the multi-year mean annual runoff. The analysis results of the improved monthly frequency method based on the Copula function pass the rationality test of "Specification for the Calculation of Environmental Flow in Rivers and Lakes", and are relatively safe in riverine ecological security. This method considers the frequency difference between annual average flow and monthly average flow and is an effective method for ecological flow assessment. The research results can provide a scientific basis and technical support for the development and utilization of water resources and the protection of river ecological environment in the Qujiang River, and have an important reference value for the evaluation of ecological water demand and the construction of water ecological civilization in other river basins. Objective: Using Xinjiang melon as raw material, explore melon slices in weak microwave-assisted hot air drying in the process of drying characteristics. Methods: Using R², X² and RMSE as evaluation indexes, compare melon slices of wet basis moisture content under different conditions, the drying rate, moisture content than the distinction, and melon dry dynamics model was set up, and the drying characteristic curve of melon slices was gotten at different slice thickness, the cycle time of microwave, the hot air cycle time. Results: The thinner the slice, the longer the circulation drying time of microwave and dry hot air, the faster the moisture content of the dry base decreased. The faster the drying rate, and the more the moisture ratio changes. The drying process of melon slice can be divided into two stages, accelerated drying and reduced drying, and accelerated drying was the main drying stage. Different slice thickness, microwave circulation time and dry-hot ar circulation time, the Thompson equation can better reflect the drying characteristics and laws of melon slices. The R² ranged from 0.992 9~0.999 9, χ² ranged from 9.55 X 10-6~1.51X10-3, RMSE ranged from 0.002 68~0.027 47. The predicted values of Thompson equation were similar to the true ones, R² was 0.995 0, χ² was 3. 93 X 10-4, and RMSE was 0.016 58. Based on the political institutional environment of China, this paper analyzes the influence mechanism of the interaction between central and local governments on the evolution of the PPP (Public Private Partnership) model under the administrative contracting system. Taking the time dimension as the starting point, based on the reform of fiscal system, infrastructure investment, financing institution and the change of external environment. The evolution of PPP model in China can be divided into four stages. These four stages are early exploration, BT development, middle development and late development. By analyzing the application characteristics and main problems of the PPP model at each stage, this paper explores the behavioral logic of local government and the institutional design of central government under the fiscal restraint and horizontal promotion incentives of the administrative contracting system. Objective To assess the efficacy of occipital artery (OA)-posterior inferior cerebellar artery (PICA) bypass for the treatment of vertebral aneurysms involving PICA. Methods and Results A total of 6 patients with vertebral aneurysms involving PICA treated by OA-PICA bypass in Tongji Hospital, Tongji Medical College, Huazhong University of Science and Technology from January 2016 to December 2020 were retrospectively reviewed. Complications were recorded and outcomes were assessed by modified Rankin Scale (mRS). OA-PICA bypass was successfully performed in 6 patients and bypass patency was confirmed by intraoperative indocyanine green angiography (ICGA). OA-PICA bypass and surgical trapping were performed in the same session in one patient (Case 4). Elective endovascular trapping was applied after bypass in 3 cases (Case 1, Case 3, Case 5). Elective stent implantation was conducted in one patient (Case 2) after the bypass surgery. One patient (Case 6) died of aneurysm rehemorrhage 10 h after OA-PICA bypass. No complications were observed during the hospitalization in the remaining 5 patients. Bypass patency was achieved in a mean follow-up time of 37 months and outcome was favorable (mRS was 1, for all) one year after the final surgery. In this paper, Korla fragrant pear was used as the experimental material, and the basic physical and chemical indices of fermented mash of whole fruit pulp or juice fermentation with three yeasts, EC118, KI and KD, as well as the aroma components of fragrant pear fruit and the six resulting treated fruit wines were examined. There were no differences among the treatments in the overall changing trend of each index of the fermented mashes for the three yeasts in the whole fruit pulp or juice fermentation process, although certain differences were still detected in the change level of each index. The total phenolic contents of the six kinds of fruit wines were in the range of 244.62~296.91 mg/L, with the total phenolic content of the fruit wine fermented with whole fruit pulp being higher than that of the fruit wine fermented with fruit juice. The aroma types of the EC118-, KI- and KD-whole fruit pulp and fruit juice wine were 25, 30, 28, 22, 23 and 24, respectively, with 14 kinds of aroma in common (mainly including alcohols, esters and acids). Among the three yeasts, KI and KD yeast had better aroma production capacities, with their corresponding fruit wines having appropriate acidity, high alcohol content, clear and bright color, higher total phenolic contents and more aroma types in whole fruit-derived fermented wine than in fruit juice-derived wines. KI yeast led to the greatest number of aroma components, and the Kl-whole fruit pulp fermented wine had the highest total phenolic content, therefore, KI and whole fruit pulp were selected as the best strain and fermentation raw material for fragrant pear fruit wine. To explore the current status and historical change of the Lake Qiandao ecosystem, the Ecopath model of the Lake Qiandao ecosystem was constructed based on the survey data of fishery resources and water environment in Lake Qiandao in 2016. And the trophic level structure, energy flow and ecosystem characteristics were analyzed based on this model. The Ecopath pedigree index (P index) was 0.54, with high data reliability. Ecopath model was consisted of 18 functional groups, and the fractional trophic levels ranged from 1 to 3.14. Energy flow of Lake Qiandao ecosystem was dominated by grazing food chain, which accounted for 56% of the energy source. Parameters reflected the complexity of the ecosystem, For example, the system connectance index (CI), the omnivory index (SOI), the Finn cycling index and the Finn mean path length were 0.26, 0.13, 5.15% and 2.46, respectively. Compared with other reservoirs, it showed that the functional groups of Lake Qiandao ecosystem had a higher polymerization, a closer connection, a higher rate of material recycling and the ecosystem was more mature. However, the total system throughput of Lake Qiandao in 2016 was low with 24698.27 t/(km2·a), and the total primary production was 6.51 folds of the total respiration, which indicate that the scale of Lake Qiandao ecosystem was small and still in a developmental stage. According to the historical analyses of the Lake Qiandao ecosystem, the scale of this ecosystem has become larger, and the stability and complexity have been enhanced, but the nutrient interaction relationship was weakened, and the ability of the ecosystem to resist external interference was still weak. At the same time, the conversion efficiency of primary producers of Lake Qiandao ecosystem was low, and the food web was simple. Objective To study the clinical significance and cut-off value of white blood cell (WBC) count in the diagnosis of early-onset sepsis (EOS) in neonates. Methods A retrospective analysis was performed on 306 neonates with EOS who were admitted from January 2019 to March 2020. A total of 580 children without infection who were admitted during the same period of time were enrolled as the control group. General status and WBC count were compared between the two groups. The diagnostic value of WBC count was analyzed based on the diagnostic and therapeutic protocol of neonatal sepsis in 2003 (referred to as the 2003 diagnostic and therapeutic protocol) and the expert consensus on the diagnosis and treatment of neonatal sepsis (2019 edition) (referred to as the 2019 expert consensus). Results According to the two different diagnosis and treatment protocols, the statistical analysis showed that WBC count had a relatively positive rate (51.3% and 32.0% respectively) but a relatively high specificity (93.3% and 98.6% respectively). The receiver operating characteristic (ROC) curve analysis showed that the area under the ROC curve of WBC count in the 2003 diagnostic and therapeutic protocol was larger than that in the 2019 expert consensus (P<0.05). Conclusions The cut-off value of WBC ≥25x109/L in the 2003 diagnostic and therapeutic protocol is more reasonable in the diagnosis of EOS. [ABSTRACT FROM AUTHOR]

[Objectives] The greater waxmoth (Galleria mellonella) is a ubiquitous pest of honeybee colonies and causes significant economic losses to the global apicultural industry. The objective of this study is to establish the antennal transcriptome database of G. mellonella and explore olfaction-related gene data, which could provide information useful facilitating the prevention and control of G. mellonella using olfaction-related gene targeting. [Methods] The antennal transcriptome of G. mellonella adults was sequenced using an Illumina HiSeqTM 2500 platform, analyzed bioinformatically and successfully assembled. [Results] In total, 188 278 unigenes with a mean length of 781 bp and an N50 length of 1 161 bp were obtained. Of these unigenes, 108 047 unigenes were annotated using a BLAST search based on seven public databases. 78 746 unigenes (41.82%) were annotated to the NR database. G. mellonella unigenes had the highest similarity (18.4%) to those of Bombyx mori. 69 794 unigenes were divided into 55 branches and three categories (biological processes, cellular components and molecular function) using Gene Ontology (GO), and many were found to be related to binding, catalytic activity and metabolic processes. In the KEGG database, 14 699 unigenes were assigned to 231 known metabolic pathways. Among these, 2 401 unigenes (16.33%) are involved in the signal transduction pathway. By further analyzing transcriptome data, 226 olfaction-related genes of G. mellonella including 52 odorant binding protein (OBP) genes, 36 chemosensory protein (CSP) genes, 80 odorant receptor (OR) genes, 56 ionotropic receptor (IR) genes and 2 sensory neuron membrane protein (SNMP) genes, were identified. After comparison of the transcriptome of male and female antennae, 114 differentially expressed genes (DEGs) including 5 olfaction-related genes (OBP8, OBP17, CSP3, CSP34 and OR15) were screened. Of these DEGs, 66 genes were highly expressed in the antennae of females and 48 in the antennae of males. [Conclusion] This study acquired the antennal transcriptome data of G. mellonella and identified olfaction-related genes.
