1. Application of SCoT markers on genetic diversity analysis of Luffa Mil.

- Abstract
[Objective] The genetic diversity and relationship of Luffa Mill, germplasm resources were analyzed by using SCoT marker technology in order to provide references for resources utilization and variety breeding of loofah, [Method] The genetic diversity and cluster analysis of 81 loofah accessions were conducted by using 80 primers and SCoT-PCR, [Result]Based on the optimal reaction system of SCoT-PCR, 10 primers with abundant polymorphism and better repeatability were screened from 80 primers, and each primer could amplify out 10-17 bands with 100-2100 bp in length, A total of 140 bands were amplified, and 124 of which were polymorphic, the polymorphic rate was 87,57%, The genetic similar coefficient of 81 loofah accessions ranged from 0,5900 to 0, 9200, and all loofah accessions could be clustered into 2 groups at 0, 5900 of similar coefficients, The I group included 46 accessions of Luffa acutang;ula (L1 ) Roxb, which mainly contained most of loofah materials collected from Guangxi and Guangdong province, The "group included 35 accessions of Luffacy lindrica (L1) Roem, The loofah materials in I and " groups could be divided into 5 subgroups at 0, 7200 and 0, 7900 of similar coefficient, and each subgroup mainly contained the materials collected from the same region, [Conclusion]SCoT marker technology could be acted as effective supplement of ISSR and SRAP markers and provide a simple, reliable method on studying loofah germplasm resources.
