ObjectiveTo analyze the mental health status and the influencing factors among the residents taking standardized residency training during the outbreak and stable period of COVID-19， so as to provide theoretical basis for their mental health education.MethodsOn February 8 to February 10 and April 11 to April 13， 2020， Symptom Checklist 90 （SCL-90）， Beck Srivastava Stress Inventory （BSSI） and Simplified Coping Style Questionnaire （SCSQ） were distributed in online chat groups involving all grades of residents taking standardized residency training in the Second Affiliated Hospital of Kunming Medical University. Through two rounds of questionnaire survey， non-probability sampling method was used to obtain survey samples， and their mental status were analyzed.ResultsA total of 159 valid questionnaires were collected in the first round， and 99 valid questionnaires in the second round. The first survey showed that the total score of SCL-90 was （117.69±37.74） and the detection rate of positive symptoms in SCL-90 was 25.8%， and mainly dominated by obsession， fear and interpersonal sensitivity. In the second survey， the results showed that the total score of SCL-90 was （127.19±51.44）， and the main positive symptoms included obsession， depression and interpersonal sensitivity， with a positive detection rate of 30.3 %. Objective To prepare the monoclonal antibodies (mAb) against hemagglutinin of H4 subtype avian influenza virus (AIV), and develop a sandwich ELISA for the detection of H4 subtype AIV. Methods The BALB/c mice were immunized with inactive H4 subtype AIV. A mAb against H4 subtype AIV, designated as 6G4, was obtained by cell fusion, hemagglutination inhibition (HI) screening and subcloing. Immuofluorescence cytochemistry and Western blotting were used to detect the reactivity of 6G4 with H4 subtype AIV, and the specificity, broad spectrum and stability of 6G4 were characterized by HI assay. Subclass of 6G4 was determined by kit. With chicken polyclonal antibody against H4 subtype AIV as coated antibody, 6G4 mAb as capture antibody and HRP-labeled goat anti-mouse IgG as the enzyme-labeled antibody, a sandwich ELISA for the detection of H4 subtype AIV was established by optimization of the reaction conditions and serial verification. Results 6G4 belonged to IgG1 subclass, and the light chain belonged to κ. It could secrete antibody stably and had good reactivity, specificity, broad spectrum and stability. ELISA based on 6G4 was specific, sensitive, accurate and suitable for the detection of a large number of samples. Conclusion We successfully achieved the anti-H4 subtype AIV mAb, and developed the sandwich ELISA for the detection of H4 subtype AIV.

Objective: To evaluate the application of various obstructive sleep apnea (OSA) screening scales in patients with acute ischemic stroke., Methods: One hundred and two patients with acute ischemic stroke were enrolled continuously during July 2016 and March 2017 from Sir Run Run Shaw Hospital, Zhejiang University School of Medicine. All patients were assessed by the same physician with various OSA screening scales, including Epworth scale, Berlin questionnaire, STOP-Bang questionnaire, SOS scale, four-variable scale and its modified version. Polysomnography was performed in Sleeping Disorder Center on each patient. According to the apnea-hypopnea index (AHI) acquired from polysomnography, patients were divided into moderate-severe OSA group (AHI ≥ 15, n =58) and normal-mild OSA group (AHI<15, n =44). The efficiencies of the scales in identification of patients with moderate or severe OSA were analyzed and compared., Results: The ROC curves showed that the four-variable scale and its modified version had higher area under curve (0.806 and 0.807, both P <0.01) for diagnosis of moderate-severe OSA, and the cut-off values for Epworth scale, Berlin questionnaire, STOP-Bang questionnaire, SOS scale, four-variable scale and its modified version were 9, 2, 4, 15, 11, 10, respectively. The sensitivities, specificities, positive and negative predictive values of four-variable scale and its modified version in diagnosis of moderate-severe OSA were higher than those of other scales., Conclusions: Compared with Epworth scale, Berlin questionnaire, STOP-Bang questionnaire and SOS scale, the four-variable scale and its modified version are more effective in screening of OSA for patients with acute ischemic stroke.

Objective: Establish an effective method for in vivo short-time standardized discriminating system of thyroid hormone disruptors., Methods: 4 experimental groups treated with PTU lavage for 3, 6, 9, 12 days respectively and one dissolvent group for control. 10 rats in each group were sacrificed in due time with the serum being extracted and detected for T3, T4 and TSH by chemoluminescence. Histopathological changes were visualized by HE staining and changes in colloid were visualized by PAS staining. PCNA was detected by IHC and western-blotting., Results: Comparing with control group, 3-day treated started to show a decrease in serum concentration of TT3 (P < 0.05), 6-day, 9-day and 12-day treated presented significant decreases in both concentration of TT3 and TT4, with a significant increase in TSH concentration (P < 0.05). Viscera coefficients of thyroid gland were significantly greater than the controls in 6-day, 9-day and 12-day treated groups (P < 0.05). Histopathological observation found thyroid follicle epithelium stratification in 3-day sections, diffused hyperplasia, vast hyperplasia plaque and solid bubbles in 6-day sections (P < 0.05), aggragated fibrilblasts and capillary endothelium hyperplasia and formation of large amount of solid sequential follicles in 9-day sections, mass hollow sequential follicles in 12-day sections. PAS staining showed weak positive in 3-day, 6-day, 9-day thyroid gland follicle colloid sections, and negative in 12-day sections. A coherent of changes with time was observed in PCNA labeling index and PCNA/beta-actin ratio, i.e. starting to increase in 3-day group (P < 0.05), reaching its peak at 6-day group (P < 0.05) and decreasing from 9-day group (P < 0.05), while no significant difference comparing with control in 12-day group., Conclusion: Hyperplastic patch, solid bubble and PCNA labeling index detected by IHC used as early index for thyroid gland hyperplasia are stable and relient, and 6 day is optimization experiment phase.
