Background Thyroid autoantibody is a marker for the diagnosis of Hashimoto's thyroiditis (HT), and B cells are essential in the pathogenesis of HT. Enhancer of Zeste homolog 2 (EZH2), which is an important epigenetic regulator, plays an important role in the regulation of lymphocytes development and function. Objective To investigate EZH2 expression in plasmoblasts and plasma cells in HT, and further explore the therapeutic effect of EZH2 inhibitors in experimental autoimmune thyroiditis (EAT) model. Methods The thyroid tissues from 6 patients who underwent thyroidectomy (3 HT patients with PTC, 3 patients with PTC alone) in Peking University First Hospital between 2010 and 2020 were obtained from the contralateral lobe with thyroid cancer, and screened for the expression of B-lymphocyte-related genes by RNA-seq; thyroid tissues from 16 HT patients and 8 normal thyroid tissues were collected and verified for the the expression of EZH2 in B cells in HT thyroid tissues by immunohistochemistry or immunofluorescence, respectively. Fine-needle aspiration (FNA) samples from patients with HT (n=25), and peripheral blood from patients with HT (n=19) or healthy donors (n=12) were analyzed by flow cytometry to define altered EZH2 expression in plasmablasts and plasma cells. Fifteen seven-week-old NOD.H-2h4 mice were randomly divided into the control (n=5), EAT without injection group (n=5), and EZH2 inhibitor+ GSK126 injection group (10 mg/kg, intraperitoneal injection 3 times /week, n=5). The degree of thyroid inflammation and changes in TgAb levels were observed after 8 weeks. Results RNA sequencing analysis showed that EZH2 and genes associated with the B-cell phenotype such as CD19, CD27, CD38, CD52 were higher expressed in HT hyroid tissues compared with normal thyroid tissues. Immunohistochemical results showed that immunohistochemical staining for EZH2 in 16 HT thyroid tissue specimens was strongly positive with positive cells observed in the GC region, and no positive cells were observed in the staining of 8 normal thyroid tissues. EZH2 staining in HT thyroid tissue was highly expressed in the GC region, and EZH2 was specifically expressed in CD19+ B cells. Objective: To establish the quality standard for the compatibility solution of Alpinia officinarum Pogostemon cablin. Methods: Thin layer chromatography was used for qualitative identification for the two indicator components of galangin and pogostone in the compatibility solution of Alpinia officinarum Pogostemon cablin: the mixture of anpetroleum ether ethyl acetate (5:3), (5:1)as the developing solvent system on silica gel G thin layer plate, high efficiency silica gel G thin layer plate; High performance liquid chromatography was used to determine the content of galangin and pogostone: C18 chromatographic column (4.6 mmx250 mm, 5 μm)was used, and the mobile phase was methanol (A) 0.2% phosphoric acid (B), with a detection wavelength of 310 nm and gradient elution. Results:For the two indicator components of galangin and pogostone in the compatibility solution of Alpinia officinarum Pogostemon cablin, the fluorescent spots were clear, without interference from negative controls; Mass concentration was between 10.00-1 000.00 μg/mL and 10.00-640.00 μg/mL, and the linear relationship coefficients with peak area within the range of were R²= 0.999 0, R²= 0.999 9, respectively; The precision RSD was 0.63% and 0.56%, the stability RSD was 1.82% and 0.24%, and the repeatability RSD was 1.30% and 0.89%; The average recovery rates (n=9) were 103.19% (RSD=0.93%) and 100.87% (RSD=0.4%), respectively; The content of these two indicator components should not be less than 233.71 μg/mL and 61.48 μg/mL. Conclusion: A research method has been established for the quality standard of Alpinia officinarum Pogostemon cablin compatibility solution. Water and fertilizer integration technology can effectively improve nutrient utilization efficiency. However, the existing water and fertilizer machines have some shortcomings, such as huge cost, single fertilizer injection, need for cleaning water and so on, which hinder the development of water and fertilizer integration technology. Aiming at the problems of precise and low-cost compounding of compound fertilizer at the local farm, the water and fertilizer integrated intelligent irrigation and fertilization system were taken as the research object. In this research, new concept of an intelligent sensing system was proposed， and accurate proportioning system of water and fertilizer concentration was constructed and implemented. Firstly, a fast on-line method of intelligent sensing model of water and fertilizer was established based on a series of concentration gradient compound fertilizer solutions. The conductivity values of these formulated solutions were tested by contactless conductivity detection electrodes. Subsequently, the data analysis algorithms were discussed and compared to fit regression model. Based on the intelligent sensing model of water and fertilizer , the framework structure of in-situ intelligent sensing and accurate proportioning system of water and fertilizer concentration was designed, and the working principle of the system was also explained. The system proposed includs a first-level water and fertilizer concentration intelligent perception model building subsystem and a second-level water and fertilizer accurate proportioning subsystem. The first-level subsystem was designed as a portable device, which mainly included a precise pump for quantitative dosing, a large-range online conductivity sensor, a plastic bucket and supporting control and model building software. The second-level subsystem was designed as a dynamic and precise fertilizer distribution device. The effectiveness of the system was verified by three types of water intelligent fertilizer application so as to guide the in-situ water and fertilizer concentration ratio. The testing results showed that the second-order polynomial fitting curve under regularization conditions was the best model to express the relationship between the conductivity and the concentration of water and fertilizer, and the correlation coefficients R2 was higher than 0.999. Combined with the proportion of each index of compound fertilizer, the concentration of each index of compound fertilizer that the user cares about can be obtained according to this model. The results of three types of water intelligent fertilizer application showed that the conductivity of natural water had an effect on the water and fertilizer system, and the relative deviation was more than 0.1. The online water and fertilizer perception and ratio system proposed in this research realized the elimination of the interference of the local water conductivity on the accuracy of the ratio of water and fertilizer, and the accurate calculation of compound fertilizer was achieved through model calculation. This system has a simple structure and accurate ratio, low cost， and can be easily combined with the existing water and fertilizer integrated machine or artificial fertilizer system. The system could be widely used in facility agriculture, orchard cultivation and field cash crop cultivation, et al.

Objective To investigate the glucose metabolic status in patients with previous gestational diabetes mellitus (GDM) in the 5-6 years after delivery, and to explore the changes in insulin resistance and islet β cell function from postpartum short-term (6-12 weeks) to the long-term (5-6 years) after delivery, as well as the relationship with glucose metabolic status. Methods A total of 72 patients with a history of GDM who delivered in Peking University First Hospital and had complete short-and long-term postpartum follow-up data were included. They were divided into normal glucose metabolism (NGT) and abnormal glucose metabolism (AGT) groups according to their glucose metabolism status in 5-6 years after delivery. Pre-pregnancy, pregnancy, perinatal, short-and long-term postpartum clinical data were collected and compared between the two groups, including demographic indicators, glucose and lipid metabolism indicators [oral glucose tolerance test (OGTT) fasting blood glucose (FPG), 1-hour blood glucose (1hPG), 2-hour blood glucose (2hPG), 3-hour blood glucose (3hPG), total cholesterol, triglycerides (TG), high-density lipoprotein cholesterol, low-density lipoprotein cholesterol], insulin use ratio, fasting insulin (FINS), OGTT 2-hour insulin (2hINS), insulin sensitivity index (ISI), homeostasis model assessment of insulin resistance index (HOMA-IR) and homeostasis model assessment of β cell function index (HOMA-β) were calculated. The comparison between the two groups was conducted using independent sample t-test, rank sum test χ² test or Fisher's exact probability test. Multivariate logistic regression analysis was used to analyze the influence factors of abnormal glucose metabolism. Results There were 38 (52.78%) cases in NGT group and 34 (47.22%) cases in AGT group. Compared with the NGT group, there are differences in related metabolic indicators in AGT group from pregnancy to long-term postpartum period: During pregnancy, 3hPG in 75 g OGTT and the proportion of insulin users were higher in AGT group. As for 6-12 weeks postpartum, OGTT 2hPG and the incidence of blood glucose abnormalities and the proportion of hypertriglyceridemia were higher (all P<0.05). However, there was no statistically significant difference in HOMA-IR and HOMA-β between the two groups(P>0.05). At 5-6 years postpartum, FPG, OGTT 2hPG, and FINS were all higher, with a higher degree of insulin resistance and lower insulin sensitivity (ISI) in AGT group (all P<0.05). Compared with short-term postpartum period (6-12 weeks), FPG, FINS, HOMA-IR, HOMA-β were all significantly increased regardless of glucose metabolic status at 5-6 years postpartum, while OGTT 2hPG and 2hINS increased further in the AGT group (all P<0.05). OGTT 2hPG and insulin use during pregnancy were risk factors for abnormal glucose metabolism 5-6 years postpartum according to multivariate logistic regression analysis, with OR values (95%CI) of 1.646 (1.015-2.671) and 3.570 (1.009-12.624), respectively. Conclusions The incidence of AGT was still high in patients with previous GDM 5-6 years after delivery. Insulin resistance progressed gradually after delivery regardless of glucose metabolism, especially in patients with abnormal glucose metabolism. OGTT 2hPG and insulin use in pregnancy were risk factors for abnormal glucose metabolism in postpartum 5-6 years, suggesting that there is continuity in time and similarity in pathogenesis between GDM and T2DM. In view of problems of large amount of maintenance and high failure rate existed in actual running process of mine-used electric locomotive used traditional DC speed regulation system, a speed regulation system of mine-used electric locomotive based on permanent magnet synchronous motor was designed. The system uses TMS320F28335 as control core and realizes high-precision control of the electric locomotive combining with strategy of direct torque control as well as peripheral circuits of current sampling, speed measurement. The experimental result shows that the system has good speed regulation performance, fast dynamic response and less maintenance.

Objective: To elaborate the clinical characteristics of adrenal non-Hodgkin's lymphoma and to expand the clinical thinking of adrenal tumors., Methods: Subjects with adrenal tumors and non-Hodgkin's lymphomas between January. 1994 and December.2012 in Peking University First Hospital retrospectively were included and these with adrenal lymphoma patients were analyzed in the present study., Results: Among 1100 adrenal tumors and 1 002 non-hodgkin's lymphomas, 21 patients (aged 35 to 80 years, mean 56 years) were diagnosed as having adrenal non-Hodgkin's lymphoma with 14 males and 7 females. Among the 21 patients, 15 were with pain on the waist and the back, 3 with fever, 1 had weight loss. Two patients were diagnosed by regular health examination. Only 2 subjects accompanied by superficial lymph node enlargement, while 10 by abdominal cavity lymph node enlargement.Eleven subjects were with extranodal involvement. Bilateral adrenal were involved in 8 patients with 9 on the left and 4 the right. The average diameter of the masses was 7.2 cm. There were no specific features in conventional imaging such as CT and MRI scan. All of the 21 cases were found normal adrenal function by endocrinological examinations. Pathology reviewed that 18 of 21 were diffuse large B cell non-Hodgkin's lymphoma, 2 were T cell lymphoma, 1 were anaplastic large cell lymphoma. Only 7 of 21 cases were diagnosed with adrenal lymphoma pre-operation. Seventeen patients were followed up by telephone in September 2014, in which 14 died and the average survival time was 5.5 months, 2 were tumor-free survival for 4 and 10 months respectively, 1 were currently undergoing chemotherapy., Conclusions: Adrenal lymphoma is rare with less superficial lymph node, more bilateral adrenal involved with high malignant degree. It progressed quickly with poor prognosis. Clinical and imaging features were not specific, so misdiagnosis was very common. Pathology is the diagnostic gold standard. The most common type was diffuse large B cell type.

Leather is one of the important exporting products to European Union (EU), and tris(2-chloroethyl) phosphate (TCEP) is a commonly used flame retardant in leather and leather products. Recently, TCEP has been classified as a kind of substance of very high concern (SVHC) by EU for its carcinogenicity and reproductive toxicity. But to date, there is not a recognized method for the determination of TCEP in leather and leather products due to the serious matrix interferences and relatively low recovery of TCEP. In this work, a home-made mixed-mode sorbent (Silica-WCX) with carboxyl and alkyl groups was tested as the sorbent of solid phase extraction (SPE) to extract TECP from leather. The results demonstrated that, making the carboxyl groups protonized under acidic condition, Silica-WCX exhibited better extraction performance towards TCEP over some frequently used commercial sorbents tested. After the optimization of the SPE conditions based on Silica-WCX, a method of gas chromatography/mass spectrometry (GC-MS) was established for the determination of TCEP in leather samples. The linear range for TCEP ranged from 0.10 to 100.0 μg/L and the limit of quantification (LOQ, S/N = 10) was 44.46 ng/kg. The recoveries of TCEP spiked in samples at varied levels were in the range of 91.45%-99.98% with the relative standard deviations (RSDs) of 4.33%-5.97%. The method is simple, sensitive and reliable for the analysis of TCEP in leather and leather products.

Objective: To investigate the metabolic changes in obese adolescents and the relationship between obese adolescents and metabolic syndrome., Methods: 198 obese adolescents [body mass index (BMI) >/= 25 kg/m(2)] and 78 normal adolescents (BMI 18.5 approximately 23.0 kg/m(2)) were chosen from 2 217 middle school students of 14 approximately 16 years. The levels of serum leptin, insulin, proinsulin, glucose and lipids of these adolescents were measured and compared., Results: (1) There was significant gender difference of serum leptin levels in adolescents of 14 approximately 16 years. The level of serum leptin was higher in female than that in male [(18.53 +/- 1.41) microgram/L vs (6.33 +/- 1.79) microgram/L]. (2) The levels of serum leptin, insulin and proinsulin of obese adolescents were significantly higher than those of normal adolescents [(19.94 +/- 1.91) microgram/L vs (11.27 +/- 2.04) microgram/L, (15.34 +/- 1.66) microIU/L vs (13.17 +/- 1.43) microIU/L and (16.19 +/- 1.64) pmol/L vs (11.79 +/- 1.70) pmol/L respectively]. (3) The levels of blood glucose and triglyceride of obese adolescents were significantly higher than those of normal adolescents [(4.63 +/- 0.50) mmol/L vs (4.13 +/- 0.33) mmol/L and (1.20 +/- 0.56) mmol/L vs (0.90 +/- 0.32) mmol/L respectively], but the levels of serum high-density lipoprotein cholesterol (HDL-C) of obese adolescents were significantly lower than those of normal adolescents [(1.14 +/- 0.24) mmol/L vs (1.38 +/- 0.26) mmol/L]., Conclusion: It is suggested that there are leptin resistance, insulin resistance and potential disorders of blood glucose and lipid metabolism in obese adolescents. Our study indicated that adolescent obesity would be related with early occurrence of type 2 diabetes.
