This research aimed to establish an analytical method for the quantification of 10-deacetylbaccatin Ⅲ (10-DAB Ⅲ) and Taxol in Taxus yunnanensis. The method of matrix solid phase dispersion coupled with high performance liquid chromatography (MSPD-HPLC) was used for the respective quantification of 10-DAB Ⅲ and Taxol of T. yunnanensis. The influences of various parameters were investigated, including 12 types of solid-phase dispersants (silica gel, florisil, acid alumina, neutral alumina, alkaline alumina, C18, C18-ME, C18-G1, C18-HC, Diol, Xamide, Xion), the mass of the dispersants, and the type, concentration, and volume of the eluent on the analysis of the two components. After optimization of these conditions, the methodology was validated. Additionally, a comparative analysis was conducted with the conventional ultrasonic extraction and hot reflux extraction pretreatment methods to ascertain their effectiveness of our novel method. The results were as follows: (1) Among the 12 solid-phase dispersants examined, alkaline alumina yielded a superior extraction detection rate for 10-DAB Ⅲ and Taxol when used in a 3∶1 ratio with the sample mass and 6 mL of methanol as the eluent. (2) The developed 10-DAB Ⅲ and Taxol analysis method in T. yunnanensis demonstrated that excellent linearity (r≥0.999 9) with limit of detection (LOD) and limit of quantification (LOQ) for 10-DAB Ⅲ and Taxol ranging from 0.023 9 to 0.030 1 μg·mL-1 and from 0.142 to 0.178 μg·mL-1, respectively. The average recoveries of the target analytes varied between 93.6% and 109.0%. (3) A comparative analysis revealed that negligible differences in extraction detection of the two taxanes between the three methods. However, the MSPD method stood out due to its lower solvent consumption, simple operation, short analysis time and high purification efficiency, making it more suitable for the rapid analysis of T. yunnanensis raw materials. Therefore, this study presents a rapid and efficient analytical method for the extraction and analysis of 10-DAB Ⅲ and Taxol in T. yunnanensis, using alkaline alumina as a dispersant, providing a significant contribution to the quantitative analysis of taxanes in this species.

Objective To assess the role of the ferroptosis-associated gene GLS2 in the prognosis of pan-cancer and immunity using bioinformatics methods. Methods GLS2 expression levels in pan-cancer were profiled using publicly available databases: The Cancer Genome Atlas, GTEx, Cancer Cell Line Encyclopedia and the International Cancer Genome Consortium. The aim was to explore the role of GLS2 gene expression, gene variation survival analysis, immune infiltration, immune checkpoint related genes, TMB, and MSI in different tumors. Results Difference in GLS2 expression levels between cancer and paraneoplastic tissues was statistically significant in most cancer types, and the expression level was correlated with survival in these cancer types. A positive correlation was found between GLS2 expression and immune cell infiltration in multiple cancer types, and GLS2 expression level was positively correlated with TMB, MSI, and methylation, and its expression is an indicator for potential therapeutic response. Conclusion Pan-cancer analysis shows that the ferroptosis-related gene GLS2 can be used as a diagnostic and prognostic marker of clear cell carcinoma of kidney, adrenocortical carcinoma, lung adenocarcinoma, and pancreatic cancer.

To solve the problem of icing on steel surface, BiOBr was synthesized by hydrothermal method using bismuth nitrate and potassium bromide as raw materials, and the BiOBr modified by silane coupling agent was coated on the surface of stearic acid modified iron sheet using the dip-coating method. The BiOBr and coating were characterized by XRD, FTIR and SEM, and the anti-icing and de-icing properties of the coating were studied. The experimental results demonstrate that the contact angle of water droplets on the coating surface is 154.23°, thus delaying the freezing time of water droplets on the surface of the iron sheet from 10 s to 60 s. Under the irradiation of ultraviolet light, the surface temperature of superhydrophobic iron sheet rises from 27.41 ℃ to 57.87 ℃. The photothermal conversion effect provided by BiOBr in the coating makes the ice on its surface melt rapidly. At the same time, the coating has good self-cleaning performance.

Ferroptosis is a novel regulatory cell death characterized by iron dependence and mainly caused by the accumulation of lipid peroxides and reactive oxygen species in the cell. This process plays an important role in the development of many malignancies, and has been extensively studied in lung cancer, especially in antitumor therapy. In recent years, the role of ferroptosis in tumor immunotherapy has been gradually explored. Studies showed that targeting ferroptosis can improve the therapeutic efficacy of antitumor immunotherapy. In addition, immunotherapy and ferroptosis can work synergistically to enhance the effectiveness of antitumor therapy, suggesting a potential relationship between ferroptosis and immunotherapy and the possible reversal of immune drug resistance. This study aims to elucidate the characteristics of ferroptosis, and the role and potential clinical applications of ferroptosis in the antitumor immunotherapy of advanced non-small cell lung cancer. We also explore the role of some nanomaterials that target the onset of tumor ferroptosis in facilitating immunotherapy.

To explore the clinical diagnosis and treatment experience of isolated fibrotic tumor （SFT） occurring in the larynx, hypopharynx and esophageal inlet with a wide range.The patient, admitted to the Department of Otolaryngology-Head and Neck Surgery of Tangdu Hospital of Air Force Medical University was a female aged at 78 years, who was diagnosed with SFT primarily occured at laryngeal, hypopharynx and esophageal entrance. The clinical data, surgical methods, histopathology characteristics of the patient were analyzed respectively. It's proved that a tumor sized about 3.8 cm×2.8 cm×2.0 cm with slippy surface was found at the entrance of the laryngeal, hypopharynx and esophageal entrance, covering the laryngeal vestibule, glottis and right piriform fossa, which was completely resected by transoral robotic surgery. The postoperative pathological diagnosis was SFT. The patient recovered well after surgery and showed no recurrence within 16-month follow-up. Objectives: The efficacy of immunotherapy for lung cancer is closely related to immune cell infiltration. Arachidonic acid 5-lipoxygenase (ALOX5) can activate inflammatory responses and trigger various cell death patterns; however, the relevance of ALOX5 to immune cell infiltration in lung cancer is unclear. The expression of ALOX5 in non-small cell lung cancer (NSCLC) is analyzed using an online database to explore the correlation between ALOX5 and immune cell infiltration in NSCLC and its relationship with prognosis., Methods: Differences in ALOX5 expression in NSCLC and normal lung tissues were analyzed by online databases such as TIMER, GEPIA and HPA; the UALCAN database was used to reveal the relationship between ALOX5 and clinical features; Kaplan-Meier database was applied to explore the prognostic value of ALOX5; GeneMANIA and String Website was used to explore genes and proteins associated with ALOX5 expression, respectively; the Gene Ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis were used to analyze ALOX5 differential genes which were picked up through the TCGA database; GSEA software was applied to predict the signal pathways that ALOX5 may be involved in; and the TIMER database was used to analyze the effect of ALOX5 expression on the level of immune cell infiltration., Results: Compared with the normal lung tissues, the ALOX5 expression was low in NSCLC tissues ( P <0.05), and which affected the prognosis of lung cancer patients. The expression level of ALOX5 was related to clinical features such as sex, age, metastasis, and pathological staging in NSCLC patients (all P <0.05). The gene interaction network analysis found that the genes interacting with ALOX5 mainly included the genes related to lipid oxidation and pro-inflammatory mediators such as coactosin like protein 1 ( COTL1 ), leukotriene C4 synthase ( LTC4S ), and prostaglandin endoperoxide synthase 2 ( PTGS2 ), and the protein-protein interaction analysis results were consistent. GO and KEGG analysis found that ALOX5 was involved in the biological process of activation of immune cell function and was involved in immune response function pathways. The GSEA analysis showed that ALOX5 may activate immune responses and mediate immune-related prognosis by affecting the cytokine-cytokine receptor interactions, natural killer-mediated cytotoxicity, and T cell receptor signaling pathways. The ALOX5 mRNA expressions in lung adenocarcinoma and lung squamous cell carcinoma were positively correlated with the tumor infiltration immune cells (B cells, CD8 + T cells, CD4 + T cells, etc.) (all P <0.05), and the ALOX5 mRNA expression was positively correlated with the expression of classic T cell immune checkpoint inhibitor genes ( P <0.001)., Conclusions: The ALOX5 gene expression in NSCLC is significantly downregulated, and which can affect NSCLC prognosis and immune cell infiltration levels. ALOX5 gene may be a potential biomarker of NSCLC prognosis associated with immune cell infiltration.

Objective: To investigate the clinical characteristics, management and prognosis of laryngotracheal stenosis induced by relapsing polychondritis. Method: A retrospective analysis was performed of 11 patients with laryngotracheal stenosis induced by relapsing polychondritis. Stenosis was classified as Myer and Cotton grade Ⅱ in 4 patients, grade Ⅲ in 3 and grade Ⅳin 4. Result: Long-term T-tube implantation of thoracic tracheal stenosis was performed in 1 patient, and one patient who had underwent 2 dilations was decannulated, and the tube was successfully extubated in 9 cases who had underwent laryngotracheal reconstruction with sternohyoid myocutaneous flap or costal cartilage. Conclusion: Treatment of laryngotracheal stenosis induced by relapsing polychondritis is challenging. Objective: The aim of this study is to o explore the diagnosis, treatment and prognosis of idiopathic subglottic stenosis （ISS） Method: The clinical data of 15 patients with idiopathic subglottic stenosis treated in our department were analyzed retrospectively. The degree of stenosis was classified by the Cotton Airway grading system of Myer, with 8 cases of gradeⅡ, 4 cases of grade Ⅲ and 3 cases of grade Ⅳ. Result: The time of follow-up of HTSS was 0.5-10 years. All 15 patients were successfully extubated without asphyxia, decannulation and wound nonunion. Conclusion: For patients with idiopathic subglottic stenosis in the non-progressive stage, active surgical treatment strategy should be adopted and treated individually. Objective: To investigate whether cannabinoid receptor 1 (CB1R) is involved in nerve growth in endometriosis-associated ectopic cyst., Methods: The effect of CB1R agonist and antagonist on the expression of pan-neuronal marker protein gene product (PGP) 9.5 in ectopic cyst was examined by immunofluorescence and Western blot in endometriosis model of 18 rats., Results: Immunofluorescence revealed that PGP 9.5 was expressed in the nerve fibers and was mainly distributed in the cyst hilum. Western blot revealed that the protein density of either PGP 9.5 (2 week: 0.38 ± 0.05; 4 week: 0.63 ± 0.03; 8 week: 0.80 ± 0.07, P < 0.01) or CB1R (2 week: 0.48 ± 0.04; 4 week: 0.68 ± 0.01; 8 week: 0.80 ± 0.03, P < 0.01) in the ectopic cyst increased with cyst size. In addition, compared to control group (0.75 ± 0.01), PGP 9.5 expression in the ectopic cyst was promoted by CB1R agonist ACPA (0.81 ± 0.01, P < 0.05), and inhibited by CB1R antagonist AM251 (0.67 ± 0.03, P < 0.01)., Conclusions: CB1R was involved in the nerve growth of ectopic cyst associated with endometriosis.

Objective: To detect the expression of pan-neuronal marker protein gene product (PGP)9.5 and its clinicopathologic significance in breast cancer., Methods: The expression of PGP9.5 was examined by immunohistochemistry EnVision method in 196 cases during 2007 to 2011, including 20 normal tissues, 14 cases of fibroadenoma, 18 cases of ductal carcinoma in situ (DCIS) and 144 cases of invasive ductal carcinoma (IDC) of the breast. The relationship between PGP9.5 expression and clinicopathologic characteristics of IDC was assessed., Results: PGP9.5 expression was localized in the stroma of all normal breast tissues, but there was no expression observed in all fibroadenomas and DCIS. Overall, the expression rate of PGP9.5 in IDC was 61.8% (89/144). PGP9.5 expression increased from grade 1 tumors (29.4%, 10/34) to grade 2-3 tumors (71.8%, 79/110; P = 0.000). In addition, patients with less than 3 years disease-free survival tended to show higher PGP9.5 expression (64.8%, 35/54), compared to patients with equal to and/or more than 3 years disease-free survival (46.7%, 42/90; P = 0.035). However, there was no correlation between PGP9.5 expression and tumor size, tumor stage, lymph metastasis, hormone receptor expression., Conclusion: PGP9.5 expression is correlated with tumor grade and prognosis in IDC of the breast.
