目的 探讨前循环串联病变患者颈动脉支架置入后支架内再闭塞的可能影响因素及预后情况。 方法 回顾性连续收集2020年6月—2022年9月于临沂市人民医院就诊的前循环串联病变且术中接受颈动脉支架置入治疗患者的临床资料，并分析上述患者手术操作过程、血管再通、术后并发症以及预后情况。同时依据90 d功能预后，将患者分为预后良好组与预后不良组，采用多元logistic回归分析影响患者预后的独立危险因素。 结果 本研究共纳入111例前循环串联病变且接受急诊颈动脉支架置入治疗的患者，术后有16例（14.4%）患者发生支架内再闭塞。分析发现支架内再闭塞患者术后残余狭窄程度较高且多数患者术前未进行充分的抗栓治疗。同时依据90 d功能预后，将111例患者分为预后良好组（66例）与预后不良组（45例），多元logistic回归分析显示术后支架内再闭塞（OR 0.315，95%CI 0.106～0.932，P=0.037）及术前梗死核心区体积（OR 0.978，95%CI 0.959～0.997，P=0.023）是影响患者预后的独立危险因素。 结论 前循环串联病变患者颈动脉支架置入后再闭塞可能与术后残余狭窄程度高、抗栓治疗不充分等因素相关,而术前梗死核心区体积以及支架内再闭塞是影响患者预后的独立危险因素。 Abstract: Objective To explore the possible influencing factors and prognosis of in-stent reocclusion of patients with anterior circulation tandem lesions after carotid artery stenting. Methods Retrospective continuous collection of the clinical data of patients with anterior circulation tandem lesions who underwent carotid artery stenting at Linyi People’s Hospital from June 2020 to September 2022, and analysis of surgical procedures, vascular recanalization, postoperative complications, and prognosis of these patients were performed in this study. Based on the 90-day functional outcome, patients were categorized into favorable and unfavorable prognosis groups, and multivariate logistic regression analysis was employed to identify independent risk factors influencing patient prognosis. Results This study included 111 patients with anterior circulation tandem lesions who received carotid artery stenting during emergency treatment. Postoperatively, 16 (14.4%) patients experienced in-stent reocclusion. Analysis displayed that patients with in-stent reocclusion had a higher degree of residual stenosis after surgery, and most patients did not receive sufficient antithrombotic therapy before surgery. Based on the 90-day functional prognosis, 111 patients were divided into a good prognosis group (66 cases) and a poor prognosis group (45 cases). Multivariate logistic regression analysis showed that postoperative in-stent reocclusion (OR 0.315, 95%CI 0.106-0.932, P=0.037) and preoperative infarct core volume (OR 0.978, 95%CI 0.959-0.997, P=0.023) were independent risk factors influencing the prognosis of patients. Conclusions Reocclusion after carotid artery stenting in patients with anterior circulation tandem lesions may be related to factors such as a high degree of postoperative residual stenosis and insufficient preoperative antithrombotic therapy. Preoperative infarct core volume and in-stent reocclusion are independent risk factors influencing the prognosis of patients.

Freeze-thaw cycle damage is one of the main diseases of buildings in cold regions which seriously affects the normal performance of buildings and the safety of long-term use. Under the condition of freeze-thaw cycle for concrete bridge cracks different freeze-thaw cycles were set up and several different crack repair methods were provided to test the freeze-thaw cycle and mechanical properties of concrete blocks and study the crack reinforcement effect then the relationship between the number of freeze-thaw cycles and the crack repair method is obtained. The experimental results show that with the increase of freeze-thaw cycles the carbon fiber cloth and epoxy resin have good effect on repairing cracks the five repair methods have obvious advantages over the control group in terms of mechanical properties and show good resistance to freeze-thaw damage. The Ordos Basin went through six main stages of geological evolution, developed three sets of soure rocks, two unconformities, several sets of reservoirs and caprocks, and had three typical petroleum systems, i.e. Lower Paleozoic Ordovician gas reservoirs, Upper Paleozoic Carboniferous-Permian gas reservoirs, and Mesozoic Triassic-Jurassic oil reservoirs. Three well-tie profiles of oil and gas reservoirs in the basin are analyzed, which are the Wuzhong-Mizhi profile, Guyuan-Yichuan profile, and Baotou-Chengcheng profile. The size and distribution of the oil and gas reservoirs are controlled by the spatial distribution of soure rocks. Mesozoic oil reservoirs are located inside or near hydrocarbon depressions, where oil and gas had a short secondary migration distance. Paleozoic gas in the basin, after having a large-scale short migration in the range of source rocks, eventually gathered in the effective traps of the Ordovician weathering crust, and in the low permeability effective sand traps inside or near Carboniferous-Permian source rocks. The future risk exploration targets are: (1) for Mesozoic oil exploration, it will extend northward and explore unconventional reservoirs in source rocks; (2) for Upper Paleozoic gas exploration, it will extend southward and explore marine clastic gas reserviors; (3) for Lower Paleozoic gas exploration, it will search for lithologic-structural traps of carbonate weathering crust in the north-central and southeastern Yishan slope and those of carbonate reef flat. Objective: In order to obtain piericidin intermediates of low toxicity by metabolic engineering, we studied the function of methyltransferase gene pieB2 in the biosynthetic cluster of piericidin A1., Methods: The methyltransferase pieB2 gene disrupted Streptomyces piomogeues var. Hangzhouwanensis was constructedby double crossover recombination. The methyltransferase gene pieB2 was also PCR amplified and cloned into the plasmid pET28a for overexpressing N-(His)6-tag PieB2 in E. coli BL21(DE3)/pLysE. The recombinant PieB2 was purified by affinity chromatography via AKTA FPLC system. The PieB2 catalyzed reactions were performed using SAM and demethyl-piericidinas substrates., Results: The disruption mutant LQ-9 produced demethyl-piericidininstead of piereicidin A1, which was restored by in trans complementation of the pieB2 gene. The N-(His)6-tag PieB2 was expressed in E. coli in soluble form and was successfully purified via Ni2+ mediated affinity chromatography. In vitro biochemical experiments showed that PieB2 could convert demethyl-piericidininto piereicidin A1 in the presence of SAM. The demethyl-piericidin intermediat showed an attractive biological activities as well as piericidin A1., Conclusion: We confirmed that PieB2 is function as a SAM dependent methyltransferase in the biosynthetic gene cluster of piericidin A1.

Objective: To observe the biological characteristics of the human olfactory mucosa mesenchymal stem cells (hOM-MSCs)., Methods: The hOM-MSCs were isolated, cultured and identified in vitro. Scanning electron microscope and transmission electron microscope were used to observe the ultrastructure of hOMMSCs. Th e cells were induced towards adipocyte, osteocyte, neural stem cells, neural-like-cells in vitro., Results: The hOM-MSCs were mainly in spindle shape, arranged with radial colony. The hOMMSCs expressed CD73 and CD90 but no CD34 and CD45. Th e short and thick microvilli processes were seen at the surface of hOM-MSCs by scanning electron microscope, and 2 different cellular morphology of hOM-MSCs were seen under transmission electron microscope. Moreover, the hOMMSCs could be differentiated into adipocyte, osteocyte, neural stem cells and neural cells., Conclusion: The hOM-MSCs possess general biological characteristics of MSCs and display multiple differentiation functions. They can be served as ideal seed cells in tissue-engineering for injury repair.

Objective: To evaluate the change of plasma level of retinol binding protein 4 (RBP4) in patients with coronary heart disease, and to explore the effect of hyperinsulinemia., Methods: This study was carried out at Xiangya Hospital of Central South University, China, from September 2009 to May 2010. Thirty patients with coronary artery disease (the CAD group) were confirmed by coronary angiography, 29 patients with CAD plus hyperinsulinemia (the CAD+HIns group), and 30 healthy subjects were enrolled as controls (the control group). The peripheral blood sample from the anticubital vein was collected aseptically in all the subjects to measure the RBP4 by enzyme linked immunosorbent-assay (ELISA). The height, weight, body mass index (BMI) the waist-to-hip ratio (WHR), the blood pressure, the fasting plasma glucose (FPG), the fasting insulin (Fins), the 2-hour postprandial inslulin (2hPIns), and the homeostasis model assessment-insulin resistance index (HOMA-IR) was measured. The lipids, high sensitivity C reactive protein (hsCRP), uric acid(UA), free fatty acids (FFA) were all examined., Results: The level of plasma RBP4 in the CAD+HIns group was higher than that in the CAD group and the control group (both P<0.01), with no significant difference of plasma RBP4 between the CAD group and the control group (P>0.05). Correlation analysis showed that the plasma RBP4 level was significantly correlated with BMI, FPG, FIns, 2hPIns, HOMA-IR, TG, HDL-C, UA, and hsCRP (r=0.259, 0.331, 0.582, 0.452, 0.600, 0.236, -0.290, 0.243, 0.231, respectively; all P>0.05). Multiple regression analysis showed that BMI, 2hPIns, and HOMA-IR were the independent factors related to RBP4., Conclusion: The plasma level of RBP4 does not increase in the CAD group, but it is high in the CAD +HIns group. RBP4 level is related to BMI, lipids, UA, and other cardiovascular risk factors. BMI, 2hPIns, and HOMA-IR are the independent factors associated with RBP4.

Objective: To examine the plasma adiponectin concentration in coronary heart disease (CHD) patients combined with abnormal glucose metabolism, and to explore the clinical significance of adiponectin., Methods: Eighty-seven hospitalized CHD patients confirmed by coronary angiography from August 2009 to April 2010 at Xiangya Hospital were enrolled and divided into 3 groups according to their glucose metabolic state: 31 patients were selected as a simple CHD group, 28 were selected as a CHD combined with impaired glucose tolerance group (CHD+IGT group), and the other 28 as a CHD combined with diabetes mellitus group (CHD+DM group). The 31 healthy subjects who got health checkup at the same time were enrolled as a normal control group (NC group). Plasma adiponectin was measured by enzyme linked immunosorbent assay. The height, weight,waistline and blood pressure of all the subjects were checked, and the fasting blood glucose (FBG), insulin, lipids, high-sensitivity C-reactive protein (hs-CRP), free fatty acids (FFA), the liver function and the renal function were checked as well. The body mass index and the homeostasis model were assessed for insulin resistance., Results: 1) Plasma adiponectin in the CHD group, the CHD+IGT group, and the CHD+DM group was all lower than that in the NC group (P<0.05); 2) Compared with the CHD group, the plasma adiponectin in the CHD+DM group was the lowest, followed by the CHD+IGT group, and there was significant difference in the 3 groups (P<0.05); 3) Plasma adiponectin level was positively related with the high density lipoprotein cholesterol-C (HDL-C) (r=0.483, P<0.01), while it was negatively related with the hs-CRP and Gensini score (r=-0.489, P<0.05;r=-0.252, P<0.05)., Conclusion: Plasma adiponectin concentration is reduced in the CHD patients, and significantly reduced in CHD patients combined with abnormal glucose metabolism. Plasma adiponectin concentration decreases significantly with the severity of abnormal glucose metabolism. CHD and the abnormal glucose metabolism are important influence factors for plasma adiponectin. That plasma adiponectin level significantly decreases may be the superimposed results of CHD and abnormal glucose metabolism. Plasma adiponectin combined with HDL-C, hs-CRP and Gensini score may provide the reference in the judgement of the severity of CHD patients with abnormal glucose metabolism.

Background and Objective: Radiation sensitivity is closely related to tissue oxygen, and rh-endostatin can induce the high level of oxygen content in tumor by "normalizing" tumor angiogenesis which is associated with radiotherapy sensitivity. The aim of this study is to observe the effect of combination of radiotherapy with rh-endostatin in the rats with lung cancer., Methods: Immediate lewis cancerous ascetic injection method was used to make rats tumors bearing model, then the rats was divided into four groups randomly: group A was treated with saline; group B was treated with rh-endostatin; group C was treated with irradiation and group D was treated with rh-endostatin and irradiation. After all rats were treated, inhibition rates and the tumor growth curve were calculated. Immunohistochemisty was adopted to check the expressions of vascular endothelial growth factor (VEGF) and microvessel density (MVD)., Results: Compared with group A, the growth rates of the tumors in the other group were obviously slower, and the tumor weights were significantly different form group A (P < 0.05). Compared with the other groups, the tumor weights of group D were obviously reduced (P < 0.05). Compared with group A, VEGF and MVD of other three groups were reduced (P < 0.05), and group D were significantly cut down., Conclusion: Combination with radiotherapy and rh-endostatin could inhibit the lung cancer significantly in rats. The possible mechanisms are to decrease the expression ofVEGF and inhibit the production of angiogenesis.

Background: It has been proven that radiotherapy can induce tumor cells over expressing HIF-1. The aim of this work is to investigate the effect of endostatin and radiotherapy on human lung cancer cell line A549 and the impact of HIF-1 expression after therapy., Methods: Analysed the cell proliferation periods of the A549 cells after treated by endostatin. The A549 cells were treated with endostatin (ES) and/or radiotherpy (RT), and then the cell growth inhibiting rate was detected by methyl thiazolyl tetrazolium (MTT), the expression of HIF-1 was determined by enzyme linked immunosorbent assay (Elisa)., Results: Every treatments can inhibit the growth of A549 cells, but the cells treated with endostatin and radiotherapy were inhibited more obviously. Endostatin had the function of block cell periods on A549, it stoped the A549 cells at G2/M and S periods. Treatment of endostatin and radiotherapy can reduce the expression of HIF-1 induced by radiotherapy., Conclusions: Endostatin was synergistical with the effect of radiotherapy on inhibiting the growth rate of the A549 cells. This synergism may come from: endostatin can make A549 cells accumulated in G2-M and S transition, thereby inhibiting the proliferation of the A549 cells. HIF-1 is a important factor in resistant of cemotherapy and radiotherapy. Endostatin can inhibite the producing of HIF-1 by cells itself or induced by radiation, so increasing the treatment effect of radiotherapy.
