Objective To explore the value of dynamic contrast-enhanced (DCE)-MRI perfusion technique in distinguishing non-mass enhanced (NME) breast pathologies. Methods The clinical records and DCE-MRI of 56 women with NME breast lesions between January 2021 and December 2021 were retrospectively analyzed. Using pathological results as the reference standard, all lesions were divided into benign and malignant groups. The perfusion parameters and hemodynamic features between the two groups were compared using independent samples t-test and Mann-Whitney U test. The receiver operating characteristic (ROC) curve was used to evaluate the diagnostic performance of MRI parameters. Results The 56 NME lesions included 33 benign and 23 malignant lesions. The early enhancement ratio (EER) was significantly higher (P<0.05) in malignancy (235.615±49.946) than that of benign lesions (181.243±69.163) whereas the late enhancement ratio (LER) was not significantly higher (P=0.247) in malignancy (197.409±35.261) than benign lesions (184.071±40.842). The DCE-MRI parameters in malignancy including the rate constant (Kep, 1.606±0.671), volume transfer constant(Ktrans, 1.380 ±0.645), initial area under the gadolinium curve (0.651 ±0.263), contrast enhancement ratio (3.026 ±0.872), maximum slope increase (MSI, 0.045) were all significantly higher ( P<0.05) than those of benign lesions (0.732 ±0.326, 0.626±0.387, 0. 356 ±0. 317, 2.270±0.498, 0.023). ROC curve analysis showed that the semi-quantitative EER had better diagnostic performance (74.4%) than that of LER (63.5%). The quantitative parameters also had better diagnostic performance than semi-quantitative parameters with accuracy of 90.3% for Kep, 84.7% for Ktrans, and 86.1% for MSI. Conclusion Quantitative perfusion parameters of DCE-MRI have excellent diagnostic performance and can help differentiating benign from malignant NME breast lesions. [ABSTRACT FROM AUTHOR]

Objective: To explore the diagnostic efficacy of prenatal transabdominal gray scale combined with color flow ultrasound for placental implantation diseases. Methods: A total of 62 pregnant women with placenta previa diagnosed by prenatal ultrasound in our hospital from November 2019 -- December 2021, including 44 pregnant women with placenta implantation. All pregnant women underwent transabdominal gray-scale examination, transabdominal color ultrasound and their combination to detect placental implantation diseases. The screening efficiency of transabdominal gray-scale ultrasound combined with color blood flow ultrasound on placental implantation diseases was evaluated by analyzing the screening results of placental implantation diseases. Results: (1) The sensitivity and specificity of detecting placental implantation by gray-scale ultrasound were 73.42 % and 86.54 %; (2) the sensitivity and specificity of detecting placental implantation by color ultrasound were 76.89% and 89.07 %; (3) the sensitivity and specificity of detecting placental implantation by abdominal gray-scale combined with color blood flow ultrasound were 87.79 % and 90.36 %; (4) The positive rates of transabdominal gray scale examination, transabdominal color ultrasound examination and their combined examination in the screening of placental implantation diseases were 56.45 %, 62.90 % and 67.74 % respectively. The positive rates of the two combined examinations in the screening of prenatal placental implantation diseases were significantly higher than those of transabdominal gray scale examination and transabdominal color ultrasound examination (P<0.05). (5) The sensitivity was 72.26 %, the specificity was 90.54 %, the positive ratio was 95.55 %, and the diagnostic odds ratio was 78.89 %. Conclusion: Transabdominal gray scale combined with multi-parameter color flow ultrasound has high sensitivity and specificity in the diagnosis of prenatal placenta implantation diseases, which is worthy of clinical application. Objective　To analyze the application value of ultrasound scoring combined with enhanced flow in dangerous placenta previa（ PPP） combined with placenta accreta， in order to improve the ultrasonic diagnosis accuracy of PPP combined with placenta accreta. Methods　From January 2017 to June 2019，60 cases of pregnant and lying-in women with suspected PPP combined placental implantation in the Second People's Hospital of Yunnan Province were studied. Two-dimensional， ultrasound scoring and e-flow examination methods were routinely used in all pregnant women for the classification of PPP combined with placental implantation. Clinical surgery and pathological results were used as the gold standard for comparison， and the diagnostic efficacy of ultrasound scoring， e-flow and ultrasound scoring combined with e-flow for PPP combined with placental implantation was compared. Results　Among 60 pregnant women with suspected PPP combined with placental implantation， the sensitivity of ultrasound scoring combined with e-flow was higher than that of ultrasound scoring （P < 0.05） . The accuracy of ultrasonic scoring combined with E-flow was higher than that of ultrasonic scoring combined with e-flow （P < 0.05）. The diagnostic efficiency of ultrasound scoring combined with e-flow in PPP with placental implantation was higher than that of single ultrasound scoring or e-flow. Conclusion　Ultrasound scoring combined with E-flow has a higher diagnostic efficiency in the diagnosis of PPP combined with placental implantation. Objective To evaluate the diagnostic performance of SYSMEXUF-5000 automatic urinary sediment analyzer and explore its application value in the diagnosis and treatment of renal and urinary tract diseases. Methods 450 fresh urine samples were tested on UF-5000 and UF-1000i respectively, Compared with the results of manual microscope, the false positive rate and false negative rate of UF 5000 parameters were calculated. Then the sensitivity and specificity of UF 5000 and UF 1000I were calculated respectively. SPSS19.0 statistical software was used for statistical analysis, and X2 test was used for comparison. P < 0.05 was considered statistically significant. Results The false positive rate and false negative rate of 15 parameters, including UF 5000 red blood cells, white blood cells, and epithelial cells, were lower than 5%. Among those, white blood cells had the highest false positive rate of 4.01%, and the five commonly used parameters all had high sensitivity and specificity. However, there were no statistically significant differencs being found in the comparisons between the sensitivity of the five parameters and UF-1000i (P > 0.05). The specificity of the white blood cells and epithelial cells showed no differences, while the specificity of red blood cells, tube type, and pathological specificity of tube showed obvious differences (P<0.05). Conclusion Compared with UF 1000I, UF 5000 has similar or better diagnostic performance in most parameters and is suitable for the screening of renal and urinary tract diseases [ABSTRACT FROM AUTHOR]

Objective: To establish a nucleic acid assay for detection of Echinococcus granulosus based on recombinase-aided isothermal amplification (RAA) assay., Methods: The 12S rRNA gene of E. granulosus was selected as the target gene, and the specific primers and fluorescent probes for RAA assay were designed, screened and synthesized to establish a fluorescent RAA assay for detection of E. granulosus . The sensitivity of the fluorescent RAA assay was evaluated using different copy numbers of target gene sequence-contained recombinant plasmids and various concentrations of E. granulosus genomic DNA as templates, and the specificity of the fluorescent RAA assay was evaluated using the genomic DNA from E. granulosus , E. multilocularis , Schistosoma japonicum , S. mansoni , Ancylostoma duodenale , Clonorchis sinensis , Taenia saginata , Spirometra mansoni and Taenia solium as templates., Results: A fluorescent RAA assay was successfully established for detection of E. granulosus , which achieved specific amplification of E. granulosus genomic DNA within 20 min at 39 ℃. The lowest detection limit of the fluorescent RAA assay was 10 copies/μL of recombinant plasmids and 0.1 ng/μL E. granulosus genomic DNA, which exhibited a high sensitivity, and the fluorescent RAA assay was all negative for the genomic DNA from E. multilocularis , S. japonicum , S. mansoni , A. duodenale , C. sinensis , T. saginata , Spirometra mansoni and T. solium , which exhibited a high specificity. In addition, this fluorescent RAA assay successfully detected genomic DNA from E. granulosus cysts., Conclusions: A rapid, sensitive and specific fluorescent RAA assay is successfully established for nucleic acid detection of E. granulosus .

Objective: To establish a rapid nucleic acid detection technique for identification of Echinococcus multilocularis based on the recombinase aided isothermal amplification assay (RAA) and assess its diagnostic efficiency., Methods: The mitochondrial gene sequence of E. multilocularis (GenBank accession number: AB018440) was used as a target sequence. The primers were designed according to the RAA reaction principle and synthesized, and RAA was performed using the generated primers. E. multilocularis genomic DNA at various concentrations and the pMD19-T (Simple) vector containing various copies of the target gene fragment were amplified using RAA to evaluate its sensitivity for detection of E. multilocularis , and RAA was em- ployed to detect the genomic DNA of E. granulosus G1 genotype, Taenia saginata , T. asiatica , T. multiceps , Dipylidium caninum , Toxocara canis , Trichuris trichiura , Giardia lamblia , Fasciola hepatica , Paragonimus westermani , Fasciola gigantica and Clonorchis sinensis to evaluate its specificity. In addition, the optimized RAA was employed to detect nine tissue specimens of E. granulosus -infected animals, 3 fecal samples from E. granulosus -infected dogs and 2 fecal samples from field infected dogs to examine its reliability and feasibility., Results: The established RAA was able to detect the specific target gene fragment of E. multilocularis within 40 min. The lowest detect limit of RAA was 10 pg if E. multilocularis genomic DNA served as a template. If the re- combinant plasmid was used as a template, the minimally detectable copy number of RAA was 104. In addition, RAA was nega- tive for the genomic DNA of E. granulosus G1 genotype, T. saginata , T. asiatica , T. multiceps , D. caninum , T. canis , T. trichiura , G. lamblia , F. hepatica , P. westermani , F. gigantica and C. sinensis . The established RAA was positive for detection of the tissue specimens of infected animals, and simulated and field dog stool samples., Conclusions: A rapid, sensitive and specific RAA is established, which shows promising values in identification of E. multilocularis and gene diagnosis of alveolar echinococcosis.
