Background: Obstructive sleep apnea (OSA) is a common sleep disorder shown to be significantly correlated with metabolic syndrome (MS). Healthcare professionals affected by both MS and OSA may suffer from poor sleep quality, raising potential concerns about patient safety. Currently, there remains a gap in research specifically addressing the relationship between MS and OSA in healthcare professionals. Purpose: This study was conducted to examine the prevalence of employees with MS concomitant with OSA and to identify the factors of influence affecting MS employees with OSA in the workplace. Methods: A cross-sectional study design and convenient sampling were used, and participants were recruited from a district hospital in northern Taiwan. Data were collected using a structured questionnaire and a home sleep apnea test. Multiple logistic regression analysis was used to determine the significant factors influencing OSA in employees with MS. Results: A total of 89 participants completed the assessment, with 65.2% of those with MS also found to have OSA. The main factors found to be associated with these employees with MS having OSA as well include gender, job position, body mass index, neck circumference, waist circumference, oxygen desaturation index, and snoring f requency. Conclusions/ Implications for Practice: In light of the high percentage (65.2% in this study) of MS employees affected by OSA, regular health screenings should include a home sleep apnea test for high-risk employees to assess their risk of developing OSA. Public hospitals constitute the main body of China′s healthcare system and are a crucial force in comprehensively promoting the construction of a "Healthy China". Guiding the medical-prevention integration in public hospitals with the concept of integrated care is key to fully implementing the “Healthy China” strategy, a means of shifting disease control and management upstream, and the optimal strategy for achieving whole-life-cycle health management. This article reviews the background and necessity of implementing medical-prevention integration in public hospitals, summarizes the current status and problems of such integration in China, and, guided by the concept of integrated care, explores implementation path for medical-prevention integration in Chinese public hospitals from five perspectives: functional integration, financing integration, personnel integration, model integration, and information integration. The aim is to promote the development of medical-prevention integration in public hospitals in China.

During the 14th Five-Year Plan period, public hospitals in China have entered a phase of high-quality development, with health insurance payment system reforms becoming a crucial means to promote this advancement. However, from the current situation, although the diagnosis related group (DRG) payment reform has made great achievements in shortening the average length of stay, improving the case-mix index, and reducing the mortality rate of low-risk groups, there are also many problems and challenges such as incomplete supporting policies, the classification system needs to be updated, the quality of medical record home pages varies, the quality of coding urgently needs to be improved, and how to deal with the use of new technologies. Thus, revisiting the impact of the DRG payment method on the high-quality development of public hospitals will not only help to understand the current challenges more clearly, but also offer valuable insights for further improving and optimizing the payment method of health insurance in the future.

Objective To analyze the training content needs and preferences of administrative personnel in a tertiary public hospital under the backdrop of high-quality hospital development, aiming to enhance their overall performance and effectiveness in supporting hospital growth. Methods This study utilized a questionnaire survey and employed chi-square goodness-of-fit test for statistical analysis. Results A total of 70 questionnaires were collected. 84.29% of the respondents believed that the relevance of training content, 74.29% thought time conflicts, and 70.00% considered leadership attention, training participation awareness, and the competence of training instructors as the main factors affecting training quality. 65.71% of the respondents identified holistic thinking ability, 62.86% noted innovation capability, and 44.29% pointed out decision-making skills as the most needed training content. Conclusion The main factors affecting the quality of management ability training for hospital administrative staff are the relevance of training content and employees' willingness to participate in training. The ability that needs the most training is communication skills, while the greatest demand is for knowledge about the hospital's development history and culture. Objective To understand the current status of work stress among full -time research staff in public hospitals, analyze the problems in research management, and propose the targeted countermeasures and suggestions. Methods In August 2022, the full -time research staff in six public hospitals in Shanghai, chosen by a purposive sampling method, were surveyed with a self-designed Work Stress Source Scale for Full-time Research Staff in Public Hospitals. The work stress status and differences among the above staff with different characteristics were analyzed, and the correlation between the scores in various dimensions was examined. Results 231 questionnaires were distributed, and 212 valid questionnaires were obtained, with an effective recovery rate of 91.8%. The scores in various dimensions of the scale were as follows: the highest score was (4.02 ± 0.81) for workload pressure, followed by (4.01 ± 0.70) for work difficulty, (3.70 ± 0.81) for assessment pressure, (3.46 ± 0.86) for role ambiguity, and (3.42 ± 0.94) for job recognition. The overall work stress score was (3.72 ± 0.62), and on the overall score of work stress. Staff who had undertaken provincial or ministerial -level research projects had higher stress scores (P < 0.01); the higher the education level, the higher the scores in workload and work difficulty dimensions (both P < 0.01). There were statistically significant differences in the scores of workload and work difficulty dimensions among full-time research staff in different positions (P < 0.05 or P < 0.01), with the highest stress levels among research staff. The higher the professional title, the higher the workload dimension score (P < 0.05). There were statistically significant differences in the role ambiguity dimension scores among full -time research staff in different departments (P < 0.05), with clinical department staff scoring the highest, followed by staff in central laboratories, both higher than those in research institutes or research parks. Staff who had undertaken provincial or ministerial -level research projects had higher scores in workload, work difficulty, and job recognition dimensions (all P < 0.01). There were positive correlations between scores in various dimensions of the scale (r = 0.348 to 0.613, all P < 0.01). Conclusions Full-time research staff in public hospitals experienced significant work stress. Relevant hospital departments should actively take measures to reduce the workload of research staff, reasonably allocate work difficulty, improve job recognition, and alleviate the pressure of role ambiguity. Objective To understand the current status and existing problems of health science popularization work conducted by medical staff in municipal grade A tertiary public hospitals in a certain region, and to propose suggestions for improving their health science popularization services. Methods A questionnaire survey was conducted among 411 medical staff from four municipal grade A tertiary hospitals in a city. Relevant results were obtained through statistical analysis, and interviews were used to discuss the findings. Results Among the 411 medical staff, over 60.00% believed that health science popularization work was very important; 80.54% conducted health science popularization work in their daily routine. However, only 14.84% and 20.44% “often” (10-20 times per month) and “always” (more than 20 times per month) engaged in health science popularization work, respectively. There were statistically significant differences in the frequency of health science popularization work among medical staff of different ages and medical titles (P＜0.05). In terms of the types of health science popularization, the most common was health education or promotion during clinical diagnosis and treatment (86.1%),while only 34.14% of the work was conducted through internet platforms. In terms of overall evaluation of health science popularization work, over 80.00% of the medical staff considered it to be “quite good” or “very good”. The main factors constraining medical staff from conducting health science popularization work were insufficient time due to heavy workloads, lack of health science popularization capabilities, lack of incentive mechanisms, and scarcity of relevant materials and platforms for health science popularization. Conclusion Currently, the health science popularization capabilities of medical staff lag behind the development of the times, and their participation in health science popularization work needs to be improved. Objective To investigate the satisfaction of inpatients with telemedicine services, further improve their medical experience, and promote the quality of telemedicine services in Xinjiang region. Methods A total of 1 446 inpatients were randomly selected from 92 public hospitals in 14 prefectures and autonomous prefectures of Xinjiang. A self-designed electronic questionnaire was used to conduct a satisfaction survey among patients who received telemedicine services. Results The overall satisfaction rate of inpatients receiving telemedicine services in Xinjiang region was relatively high (94.16%), and the audience tended to be younger (patients under 50 years old accounted for 79.19%). The main reason for 72. 68% of patients choosing telemedicine services was to reduce medical costs, rather than the level of expertise provided by remote service specialists. After receiving telemedicine services, 93.50% of patients had increased confidence in subsequent treatments, and 85. 96% of patients believed that the treatment effect had been significantly improved. Conclusion Currently, the satisfaction with telemedicine services among inpatients in Xinjiang region is relatively high, but there exists a mismatch between telemedicine service capabilities and patient satisfaction. Objective To provide a reference for the health department and to improve the medical quality by analyzing and evaluating the medical quality of public traditional Chinese medicine hospitals in Shandong Province from 2019 to 2022 comprehensively. Methods Based on the survey data of public TCM hospitals participating in performance appraisal in Shandong Province, the medical quality of public TCM hospitals in Shandong Province was comprehensively evaluated by using the entropy-weighted TOPSIS method, RSR method and the fuzzy combination method of the two. Results According to the entropy-weighted TOPSIS method, the evaluation results of medical quality were the best in 2019, followed by 2022, with C values of 0.594 4 and 0.548 6, respectively, the evaluation results of RSR method were the best in 2022, followed by 2021, with RSR values of 0.750 0 and 0.688 0, respectively, and the best evaluation results of the two fuzzy joint methods were in 2022 and followed by 2021, and the three evaluation methods showed that the evaluation results of medical quality in 2020 were the worst. Conclusion Although the evaluation results of the entropy-weighted TOPSIS and RSR methods and the fuzzy joint method are slightly different, the trend is basically the same, and the medical quality of public TCM hospitals in Shandong Province has been improving year by year from 2019 to 2022. The fuzzy combination method of the two can reflect the evaluation results more scientifically and reasonably, and has certain reference significance for the improvement of medical quality level and management standards. Objective To understand the status quo of employee satisfaction in a public hospital under the background of high-quality development. Methods From March 23,2022 to April 22, 2022, a cluster sampling survey was conducted on the employees of a tertiary general hospital in a prefecture-level city in Anhui Province by using the National Health Commission Employee Satisfaction Questionnaire to analyze their job satisfaction. Results The total score of employee satisfaction was (72.14±10.59) score. The scores of five dimensions of salary and welfare, development and promotion, work content and environment, supervisor-subordinate relationship and peer relationship were(16.14±4.18),(13.10±3.45),(17.29±2.38),(10.40±1.50) and(15.20±1.83) score, respectively. There were statistically significant differences in the satisfaction scores of employees with different working years, jobs and professional titles (P<0.05). There were significant differences in the scores of salary and welfare, work content and environment dimensions between different genders(P<0.05). There were statistically significant differences in the scores of each dimension of employee satisfaction survey among different working years(P<0.05). There were statistically significant differences in the scores of each dimension of employee satisfaction in different jobs (P<0.05). There were statistically significant differences in the scores of salary and welfare, development and promotion, superior-subordinate relationship and peer relationship among employees with different professional titles (P<0.05). Conclusion The overall satisfaction of the hospital 's employees is good. OBJECTIVE: To probe into the effects of national centralized drug volume-based procurement policy, drug negotiation and key monitoring on application of proton pump inhibitors (PPI) in hospitals, so as to provide data support for the rational application of PPI and evaluation of implementation effect on related policies. METHODS: Application data of PPI from 2018 to 2022 were extracted from the drug use database of sample hospitals of the National Pharmaceutical Information Network (including 892 hospitals, with 645 tertiary hospitals and 247 secondary hospitals). Consumption amount and proportion, consumption sum and proportion, daily defined dose system (DDDs) and daily defined dose (DDC) and other indicators were analyzed and compared. RESULTS: From 2018 to 2022, due to the influence of relevant policies, the consumption sum and DDC of PPI in sample hospitals decreased year by year. The reduction of PPI injection was greater than that of PPI oral preparation. The consumption amount structure and consumption sum structure of PPI changed significantly. The consumption amount of PPI injection and DDDs of most injection varieties decreased significantly. CONCLUSIONS: From 2018 to 2022, Consumption sum and consumption amount of PPI injection in sample hospitals decrease significantly, yet the alternative application among different clinical varieties still deserves further evaluation. Background and objective Isoliquiritigenin (ISL) is an important pharmacological constituent of Glycyrrhiza glabra, which possesses a range of physiological and pharmacological activities, as well as significant antitumor activity, and can be used as a potential drug for targeted cancer therapy. LINC01503 is an oncogene, which has been closely associated with the malignant biological processes of many cancers. The aim of this study was to investigate the effects of ISL on the proliferation, apoptosis, invasion and migration of lung squamous carcinoma cells by regulating LINC01503. Methods Plasma was collected from lung squamous carcinoma patients and healthy individuals treated at Tangshan People's Hospital from January 2021 to December 2022. The expression of LINC01503 in lung squamous carcinoma plasma, tissues and cells was detected by real-time quantitative fluorescence polymerase chain reaction (qRT-PCR). Lung squamous carcinoma cells were treated with different concentrations of ISL for 24 h, and LINC01503 expression was detected by qRT-PCR. The cells were treated in groups: si-NC group, si-LINC01503 group, DMSO (0.1% dimethyl sulfone) group, ISL group, pc DNA3.1(+)-NC group, pc DNA3.1(+)-LINC01503 group, ISL+pc DNA3.1(+)-NC group and ISL+pc DNA3.1(+)- LINC01503 groups. CCK-8 assay, clone formation assay, flow cytometry, Transwell assay and scratch assay were used to explore the effect of LINC01503 on the functional phenotype of lung squamous carcinoma cells. Results Fluorescence in situ hybridization results showed that the average fluorescence intensity of LINC01503 in tissue microarrays of lung squamous carcinoma patients was higher than that in paracancerous tissues (P<0.05). The expression of LINC01503 in the plasma of patients with lung squamous carcinoma was higher than that in the plasma of healthy individuals (P<0.05). Knockdown of LINC01503 inhibited the proliferation, invasion and migration of lung squamous carcinoma cells and promoted apoptosis (P<0.05). ISL inhibited the proliferation, invasion, migration and promoted apoptosis of lung squamous carcinoma cells (P<0.05). Overexpression of LINC01503 followed by intervention with ISL reversed the promotional effect of overexpression of LINC01503 on the proliferation, invasion and migration of lung squamous carcinoma cells as well as the inhibitory effect on apoptosis (P<0.05). Conclusion LINC01503 was highly expressed in lung squamous carcinoma, and LINC01503 could promote the proliferation, invasion and migration of lung squamous carcinoma cells and inhibit the apoptosis, ISL could inhibit the proliferation, invasion and migration of lung squamous carcinoma cells and promote apoptosis of lung squamous carcinoma cells by regulating the expression of LINC01503. Objective To analyze the clinical prognosis of high - grade aneurysmal subarachnoid hemorrhage (aSAH) patients with different amounts of hematoma. Methods A total of 211 patients with high - grade aSAH hospitalized in Norhern Jiangsu People's Hospital from September 2013 to December 2020 were included. According to the intracerebral hematoma volume, they were divided into non - hematoma group (n = 105), hematoma volume < 50 ml group (n = 69) and hematoma volume ≥ 50 ml group (n = 37). All of them underwent aneurysm clipping or embolization. The modified Rankin Scale (mRS) was used to evaluate the clinical prognosis 6 months after surgery. Results Among 211 cases, 139 (65.87%) underwent aneurysm clipping and 72 (34.12%) underwent aneurysm embolization. Except for 3 patients who failed to successfully clamp and underwent decompression with bone flap due to intraoperative malignant encephaloceles, the remaining 208 patients were successfully clipped, and no aneurysms ruptured and bled again after surgery. There were 58 patients (27.49%) with good prognosis, 59 patients (27.96%) with poor prognosis, and 94 patients (44.55%) with death. The difference in neurological function prognosis among the 3 groups was statistically significant (χ² = 7.424, P = 0.024). Among them, the mortality rate in incidence of complication was 95.73% (202/211) in all patients, including 95.24% (100/105) in non - hematoma group, 95.65% (66/69) in hematoma volume < 50 ml group, and 97.30% (36/37) in hematoma volume ≥ 50 ml group. The difference in complication among the 3 groups was not statistically significant (χ² = 0.284, P = 0.867). Conclusions The prognosis of high - grade aSAH patients with intracerebral hematoma is poorer, especially in high-grade aSAH patients with hematoma volume ≥ 50 ml, and have a higher mortality rate. Objective To offer insights for enhancing the hierarchical medical system and guiding patient healthcare-seeking behaviors systematically, this study analyzes referral willingness and influencing factors of tertiary public hospitals and primary healthcare institutionsamong chronic disease patients in Sichuan Province. Methods A questionnaire survey was conducted on 297 chronic disease patients. Descriptive statistical analysis of the data was performed using SPSS 26.0 software. Chi-square tests were employed to analyze the downward referral willingness of chronic disease patients from tertiary public hospitals and the upward referral willingness to primary healthcare institutions. Results Patients in large public hospitals showed low downward referral willingness (44.2%). Rural residence, prior medical consortium services, and greater distance from tertiary hospitals increased willingness. Primary healthcare patients exhibited a 28.1% upward referral willingness, influenced by occupation, age, and use of outpatient special disease policy services. Conclusion Referral willingness is generally weak in large public hospitals and primary healthcare institutions. Enhancing the comprehensive capacity of primary healthcare institutions, strengthening collaborative mechanisms and the level of medical services within medical consortia, along with expanding reimbursement policies for chronic diseases, contributes to promoting bidirectional patient referrals and advancing the formation of a tiered diagnosis and treatment system. Objective To evaluate the impact of DRG medical insurance payment reform on the quality development of a tertiary comprehensive hospital in Guangxi. Methods This study selected data on all inpatient settlement cases in the hospital from 2020 to 2021, and calculated the DRG grouping of all cases according to the national CHS-DRG version 1.0 grouping plan. The interruption time series analysis method was further used to analyze the level and trend changes of indicators after the DRG policy reform in hospitals in December 2020.These indicators included total weight, case group index, cost consumption index, time consumption index, average length of stay, average hospitalization cost per visit, etc. They were used to evaluate the hospital' s medical service capacity, medical service efficiency, and medical safety, in order to understand the quality of the hospital. Results A total of 72 089 cases were included in this study. Indicators such as cost consumption index (β = -0.242, P = 0.001), time consumption index (β = -0.119, P = 0.002), average length of stay (β = -1.543, P = 0.011), proportion of high incidence cases (β = -0.028, P = 0.026), and average cost of hospitalization (β = -3 669.366, P = 0.026) showed an immediate decrease during the implementation of DRG policy. Conclusion After implementing the DRG policy reform, the hospital has significantly improved the efficiency of medical services, but still faces the challenge of adjusting structure and increasing quality. Objective To analyze the coupling and coordination relationship of high -quality development of tertiary public general hospitals in Guangxi, and to explore the path of high-quality development of tertiary public general hospitals, so as to provide a basis for related policy formulation. Methods The evaluation index system of high-quality development of tertiary public general hospitals in Guangxi was constructed from the four dimensions of medical quality, operational efficiency, sustainable development, and satisfaction evaluation. The coefficient of variation method and rank sum ratio method were used to comprehensively evaluate and classify the high-quality development level of the tertiary public general hospitals in 14 prefectures and cities in Guangxi in 2019, and the coupling coordination degree model was used to calculate the coupling and coordinated development level. Results Most of the sample hospitals were in the primary coordination stage, and the proportion of other coupling coordination types from high to low were intermediate coordination (22.73%), reluctant coordination (18.18%), on the verge of disorder (13.64%), and good coordination (4.55%). According to the principle of optimal classification, the tertiary public hospitals in Guangxi were divided into high, medium, and low grades. Most of the hospitals were in the middle level and there was much room for development and progress. Conclusion The coupling coordination degree of the tertiary public general hospitals in Guangxi is not high, and there are still some problems, such as uneven distribution of medical resources, large differences in medical quality, and lack of motivation for personnel training. Ambulatory medical care has undergone more than 20 years of development in China, gradually improving into a medical model that provides diagnosis and treatment services throughout the 24-hour hospitalization process. In the past decade, under the guidance of relevant national policies, the development scale and speed of ambulatory medical care have further expanded, providing more patients with efficient and low-cost diagnostic and treatment services, and playing a leverage role in the high-quality development of public hospitals. However, the development of ambulatory medical care is also facing challenges brought about by various factors such as the support of medical insurance policies, performance evaluation, and homogeneous development. Under the new situation and concepts of promoting the high-quality development of public hospitals and improving the medical service experience, the ambulatory medical service model has played a greater role in medical value. Objective To understand the development status and related reasons of smoking cessation clinics in Shenzhen public hospitals. Methods Using the census method, a survey was conducted on the current situation of smoking cessation clinics in 59 and 72 public hospitals identified by the Shenzhen Municipal Health Commission in 2015 and 2021, respectively, and 56 and 66 valid questionnaires were collected, respectively. SPSS 23. 0 was used for statistical analysis of the data, the composition ratio was mainly used for statistical description, and the chi-square test was used for statistical inference. Results As of 2021, the setting rate of smoking cessation clinics in Shenzhen public hospitals was 71. 21%, which was lower than the 78.57% (44/56) in 2015 (P >0. 05), still at the leading level in China. The application rate of brief smoking cessation intervention technology, the proportion of part-time staff, the ownership rate of equipment, the level of publicity, drug treatment and follow-up rates increased from 72. 73%, 47. 73%, 86. 36%, 36. 36%, 9. 09%, and 25. 58% in 2015 to 91. 49%, 76. 59%, 97. 87%, 59. 57%, 29. 79%, 46. 81% in 2021, the differences were all statistically significant. Among them, the weekly service time of each smoking cessation clinic in 2021 was [0. 5, 49] hours, the average service time was 15 hours/week, the number of visits per month was [ 0, 40 ], the average monthly service was 7 people, and the follow-up times were distributed between 1 and 3 times/month, mainly concentrated in 1 time/month (81. 82% ). Objective To investigate the allocation and utilization efficiency of bed resources in tertiary public hospitals in Henan Province from 2016 to 2020, to provide reference for improving the uneven utilization of bed resources and promoting the efficient utilization of bed resources. Methods Descriptive statistics, bed eficiency index, and bed utilization model were used to analyze the bed allocation and utilization efficiency of all kinds of tertiary public hospitals in Henan Province. Results From 2016 to 2020, the number of beds in tertiary public hospitals in Henan Province increased by 26%, the number of beds per 1 000 population increased by 0.27, the bed turnover accelerated, and the average length of stay decreased. The bed utilization in the tertiary public hospitals were generally in a state of high efficiency. The bed operation efficiency of general hospitals and specialist hospitals were higher, while that of traditional Chinese medicine (TCM) hospitals was relatively lower, and the bed utilization efficiency varied among different cities. Conclusion We should coordinate the allocation of beds in inter-regional tertiary public hospitals, improve the fairness of the allocation of bed resources, and reduce the uneven utilization of beds. Objective To formulate a high-quality development evaluation system of public TCM hospitals. Methods According to the relevant national policy documents and referring to the Quality Development Evaluation Indicators of Public Hospitals (Trial) and the Quality Development Evaluation Indicators of Public Traditional Chinese Medicine Hospitals (Trial) attached to the High-quality Development Evaluation Indicators of Public Hospitals (Trial), the method of combining subjective and objective weights was adopted, and the Delphi method and analytic hierarchy process were combined. Delphi method is used to sort, summarize and screen the three-level indicators to determine the indicators. Then analytic hierarchy process was used to calculate the weights of each index in the overall model, and the data of public TCM hospitals in Guangdong Province from 2019 to 2021 was collected for empirical study. Results An evaluation system for the high-quality development of public TCM hospitals has been formed, including 5 first-level indicators, 25 second-level indicators and 36 third-level indicators, and a weight table of indicators has been formed. Conclusion By collecting the data of public TCM hospitals in Guangdong Province from 2019 to 2021, the empirical study proves that the high-quality development evaluation index system of public TCM hospitals can be used to evaluate the high-quality development of public TCM hospitals. Objective To analyze the surgical treatment patterns and different surgical methods for benign prostatic hyperplasia in county-level hospitals in Gansu Province, and provide theoretical basis for appropriate perioperative management, surgical methods and health economics. Methods A public hospital was randomly selected from each county in Gansu Province, The perioperative management of urologists and the basic information of 1224 patients from 2019 to 2021 were collected by network questionnaire and grouped according to different years and prostate volume. Results The main operation for benign prostatic hyperplasia in county hospitals were M-TURP (16. 16%) and B-TURP (74. 27%). There were differences in postoperative catheter time, postoperative hospitalization time and hospitalization expenses among different prostate volumes (P < 0. 05). Conclusion The baseline characteristics of patients treated by BPH surgery in county hospitals in Gansu Province are similar to those in grade 3A hospitals of Gansu Province, and the preoperative examination and surgical indications are the same as those in developed countries, but they are significantly different from those in Gansu Province and Western countries in terms of postoperative bladder irrigation, postoperative catheter time and post-operative hospitalization, and lag behind the grade 3A hospitals of Gansu Province and Western developed countries in the promotion and use of new techniques. Objective To screen the risk factors for short-term poor clinical prognosis in patients with low-grade aneurysmal subarachnoid hemorrhage (aSAH), and to construct the Nomogram model. Methods A total of 293 aSAH patients with World Federation of Neurosurgical Societies (WNFS) grade I-II treated by aneurysm clipping or aneurysm embolization in The People's Hospital of Kaizhou District of Chongqing from January 2016 to December 2021 were enrolled. The modified Rankin Scale (mRS) was used to evaluate the clinical prognosis at 3 months after surgery. Univariate and multivariate stepwise Logistic regression analysis was used to screen the risk factors for short-term poor clinical prognosis in patients with low-grade aSAH, and Nomogram model was constructed. The receiver operating characteristic (ROC) curve was drawn and the area under the curve (AUC) was calculated to evaluate the prediction efficiency of the model. The Bootstrap internal verification method, Hosmer-Lemeshow goodness of fit test and calibration curve were used to verify the model. Results Total 293 patients were divided into good prognosis group (0-2 score, n = 238) and poor prognosis group (3-6 score, n = 55) according to mRS score. The proportion of poor prognosis group was age ≥ 65 years old (χ² = 18.516, P = 0.000) and WFNS grade II (χ² = 9.491, P = 0.002), the incidences of postoperative delayed cerebral ischemia (χ² = 28.355, P = 0.000), shunt dependent hydrocephalus (χ² = 33.497, P = 0.000) and intracranial hemorrhage (χ² = 17.744, P = 0.000) were higher than those of good prognosis group. Logistic regression analysis showed age ≥ 65 years old (OR = 1.241, 95%CI: 1.021-1.772; P = 0.000), postoperative delayed cerebral ischemia (OR = 9.462, 95%CI: 1.302-23.823; P = 0.010) and shunt dependent hydrocephalus (OR = 6.092, 95%CI: 2.730-16.201; P = 0.000) were the risk factors for short-term poor clinical prognosis in patients with low-grade aSAH. The Nomogram model was constructed based on the above 3 risk factors. ROC curve showed the AUC was 0.833 (95%CI: 0.772-0.897, P = 0.000), when Youden index was 0.520, the corresponding sensitivity was 79.28%, specificity was 72.69%, and the cut-off value was 128 points. The prediction efficiency of the model was better than that of the above 3 factors alone (P = 0.000, for all). Furthermore, the Bootstrap internal verification method, Hosmer-Lemeshow goodness of fit test and calibration curve confirmed the good discrimination (CI = 0.908), stability (χ² = 1.078, P = 0.693) and calibration. Conclusions Age ≥ 65 years old, postoperative delayed cerebral ischemia and shunt dependent hydrocephalus are the risk factors for short-term poor clinical prognosis in patients with low-grade aSAH. Objective: To explore the mechanism and clinical significance of carboxypeptidase A4 (CPA4) promoting the proliferation, migration, invasion and epithelial-mesenchymal transformation (EMT) progression of non-small cell lung cancer (NSCLC) cells. Methods: IHC and WB were employed to detect the expression of CPA4 in NSCLC tissues and adjacent tissues of 105 patients with advanced or metastatic NSCLC in Jinan Central Hospital from February 2012 to December 2015 and the expression of CPA4 in NSCLC cells. Chi-square test was used to analyze the association between CPA4 expression and the clinicopathological characteristics of the patients. The relationship between CPA4 expression and the overall survival (OS) of the patients was analyzed by Kaplan-Meier method. H1299 and A549 NSCLC cells with stable overexpression and downexpression of CPA4 were constructed by cell transfection. Changes in the cell proliferation, invasion and migration after overexpression and knockdown of CPA4 were detected by CCK-8 assay, cell colony formation assay, scratch healing assay and Transwell assay. Xenograft tumor models and tail vein-lung metastasis tumor models of nude mice were established to detect the role of CPA4 in tumor tumorigenesis and metastasis in vivo. Changes in the markers of EMT in the tissues of xenografts were detected by WB and IHC. Results: Compared with paracancer tissues and normal lung epithelial cells, CPA4 was highly expressed in cancer tissues and cancer cells (all P<0.05). The OS of NSCLC patients with high CPA4 expression was significantly shorter than that of patients with low CPA4 expression (P<0.05). High expression of CPA4 was more common in poor differentiation, N2-3 lymph node involvement and TNM stage IV (P<0.05 or P<0.01). Overexpression of CPA4 promoted H1299 cells, while knockdown of CPA4 inhibited the proliferation, migration and invasion of A549 cells. Overexpression of CPA4 promoted the growth of H1299 cell xenograft tumors and lung metastatic tumors in nude mice. Overexpression of CPA4 promoted the changes of EMT process-related molecules in H1299 cells. Conclusion: CPA4 is highly expressed in NSCLC tissues and cells. Overexpression of CPA4 can induce EMT, promote the proliferation, migration and invasion of NSCLC cells and is related to tumor progression and prognosis.
