1. The effect of FGF and HMGB proteins on the survival and proliferation potential of neural stem cells in culture

Ovaj rad opisuje utjecaj FGF i HMGB1 molekula na živčane matične stanice u kulturi (NSC). Primjenom metode magnetofekcije živčane matične stanice transfecirane su plazmidima s FGFR1 receptorom. Osim NSC stanica u eksperimentima su korištene i embrionalne matične stanice bubrega (HEK293T). Pokazalo se da FGF utječe pozitivno na preživljenje HEK293T stanica s transfeciranim plazmidima u usporedbi sa HEK stanicama koje nisu transfecirane. HMGB1 je smanjio pozitivan učinak FGF-a u kulturi HEK293T stanica. NSC stanice u svojem mediju već imaju FGF te je postignuto bolje preživljenje NSC stanica transfeciranih FGFR1-EGFP plazmidom. Korištenjem dvostruko veće količine plazmida u odnosu na magnetne čestice postigla se dvostruko bolja uspješnost transfekcije metodom magnetofekcije u usporedbi sa prethodnim istraživanjima. HMGB1 je na njihovo preživljenje djelovao negativno kao i kod HEK293T stanica. The aim of this study was to describe the effects of FGF and HMGB1 molecules on cultured neural stem cells (NSCs). Using magnetofection, NSCs were transfected with plasmides containing the FGFR1 receptor. Along with NSCs, embryonal kidney stem cells (HEK293T) were also used. The results show that FGF positively affects the survival of HEK293T cells, which were transfected with plasmides, as opposed to non-transfected cells. The HMGB1 molecule reduced the positive effect of FGF in cultured HEK293T cells. NSCs were already grown in a medium containing FGF. The ones transfected with the FGFR1-EGFP plasmide showed increased survival. Using twice as many plasmides as magnet particles resulted in a twofold increase in magnetofection success rate, compared to previous studies. At the same time, HMGB1 affected their survival negatively, just as in HEK293T cells.
