Absent in melanoma 2 (AIM2) inflammasome is a multi-protein platform that recognizes aberrant cytoplasmic dsDNA and induces cytokine maturation, release and pyroptosis. It is composed of AIM2, apoptosis-associated speck-like protein containing a CARD (ASC), and caspase-1. Recent X-ray crystallographic and high resolution cryo-electron microscopic (cryo-EM) studies have revealed a series of structures in AIM2 inflammasome activation and regulation. One prominent feature common in multiple steps is the assembly of high-order structures, especially helical filaments nucleated by upstream molecules, rather than stoichiometric complexes. In this review, we track the AIM2 inflammasome activation process step by step, using high-resolution structures to illustrate the overall architecture of AIM2 inflammasome and its assembly and regulatory mechanisms. Abstract: Members of the tumor necrosis factor receptor superfamily play key roles in innate and adaptive immunity. Here, we review recent structural studies in the intracellular signal transduction of these receptors. A central theme revealed from these structural studies is that upon ligand binding, multiple intracellular proteins form higher-order signaling machines to transduce and amplify receptor activation information to different cellular fates, including NF-κB activation, apoptosis, and programmed necrosis. The coexistence of an evaporation potential and streaming potential in a NWEG has been proposed using ZSM-5 as the generation material. A streaming potential formed as water flowed inside the ZSM-5 nanochannels, driven by electrodynamic effects that increased from the bottom to the top of the generator. Conversely, an evaporation potential existed at the surface-gas interface of the ZSM-5-solution, which decreased from the bottom to the top as the evaporation height increased. The resulting open-circuit voltage (V oc) depended on the balance between evaporation and streaming potential, both of which were influenced by the evaporation enthalpy (E e) or vapor pressure. Generally, a higher E e or lower vapor pressure led to a lower evaporation potential and subsequently a lower V oc. A dual mechanism involving synergistic evaporation potential and streaming potential is proposed to explain the mechanism of NWEGs. [Display omitted] Electricity generation by natural water evaporation generators (NWEGs) using porous materials shows great potential for energy harvesting, but mechanistic investigations of NWEGs have mostly been limited to streaming potential studies. In this study, we propose the coexistence of an evaporation potential and streaming potential in a NWEG using ZSM-5 as the generation material. The iron probe method, salt concentration regulation, solution regulation, and side evaporation area regulation were used to analyze the NWEG mechanism. The fern genus Hymenasplenium (Aspleniaceae) is one of the two genera in the family. It is generally recognized among modern pteridologists. However, its infrageneric relationships and species diversity have been unclear and controversial. The molecular studies so far have had small taxon and character sampling. In the present study, DNA sequences of six plastid markers of 158 accessions representing ca. 40 out of ca. 50 known species of Hymenasplenium , and 16 species of Asplenium were used to infer a phylogeny with maximum likelihood, Bayesian inference, and maximum parsimony approaches. Our major results include: (1) Hymenasplenium as currently defined is strongly supported as monophyletic; (2) three major clades representing early splits in Hymenasplenium are identified, with the Old World species being strongly supported as monophyletic; it is ambiguous if the New World species are monophyletic; (3) extensive cryptic speciation in the Old World is discovered demonstrating the complexity of evolution of the genus; and (4) six strongly or moderately supported subclades in the Old World clade are revealed, differing from one another in molecular, morphological, and geographical features. Charge dynamics play essential role in many practical applications such as semiconductors, electrochemical devices and transmembrane ion channels. A Maxwell-Ampère Nernst-Planck (MANP) model that describes charge dynamics via concentrations and the electric displacement is able to take effects beyond mean-field approximations into account. To obtain physically faithful numerical solutions, we develop a structure-preserving numerical method for the MANP model whose solution has several physical properties of importance. By the Slotboom transform with entropic-mean approximations, a positivity preserving scheme with Scharfetter-Gummel fluxes is derived for the generalized Nernst-Planck equations. To deal with the curl-free constraint, the dielectric displacement from the Maxwell-Ampère equation is further updated with a local relaxation algorithm of linear computational complexity. We prove that the proposed numerical method unconditionally preserves the mass conservation and the solution positivity at the discrete level, and satisfies the discrete energy dissipation law with a time-step restriction. Numerical experiments verify that our numerical method has expected accuracy and structure-preserving properties. Applications to ion transport with large convection, arising from boundary-layer electric field and Born solvation interactions, further demonstrate that the MANP formulation with the proposed numerical scheme has attractive performance and can effectively describe charge dynamics with large convection of high numerical cell Péclet numbers. • A structure-preserving numerical scheme for MANP model with correlations. • Numerical scheme based on Slotboom transform with entropic mean approximation. • Numerical analysis on structure-preserving properties at discrete level. • A convergent, linear and local relaxation algorithm to realize curl-free. • Robust simulations of ion transport with large convection. Abstract: Peroxiredoxins (Prx), a family of small non-seleno peroxidases, are important regulators for cellular reactive oxygen species (ROS), which contribute to many signaling pathways and pathogenesis of diseases. Targeting redox homeostasis is being developed as a promising therapeutic strategy for many diseases such as cancers. This mini-review attempts to focus on our recent discoveries on adenanthin as the first natural molecule to specifically target the resolving cysteines of Prx I and Prx II and thus inhibit their peroxidase activities, and its role in differentiation induction in vitro and in vivo of acute myeloid leukemic cells. Emerging evidence has indicated that transforming growth factor-beta 1 (TGF-β1) induces the epithelial–mesenchymal transition (EMT) in cancer cells, thus promoting their motility and invasiveness. Quercetin, a member of the polyphenolic flavonoid family, has been reported to display anticancer activity against a broad range of cancer cell types. Indeed, numerous studies have shown the cancer preventive effects and molecular mechanisms of quercetin in vitro using diverse cell model systems. However, the potential effect of quercetin on EMT remains unclear. In this study, we identified a unique function of quercetin in inhibiting the EMT process induced by TGF-β1. In particular, quercetin rescued the morphological changes and EMT-like phenotypes in TGF-β1-activated SW480 cells, and this inhibition of TGF-β1-induced EMT was mediated via the suppression of Twist1 expression. In addition, quercetin strongly suppressed TGF-β1-induced invasion of SW480 cells. Thus, quercetin may be considered a novel therapeutic agent for the treatment of patients with refractory cancer and for the prevention of the metastatic cascade initiated by EMT. Abstract: Experiments have been carried out to confirm the cross-resistance between abamectin and tebufenozide in Plutella xylostella and demonstrate its mechanism. The results showed that the resistant strain of P. xylostella selected by tebufenozide (RF 99.38) really showed high cross-resistance to abamectin (RF 29.25). When this strain was subjected to resistance decaying treatment, breeding without contacting any insecticides, and abamectin resistance selection for 20 generations, the former resulted in decrease of its resistance to both tebufenozide and abamectin to about one third of the original (RF 35.03 and 11.67, respectively), and the later enhanced its resistance to abamectin dramatically (RF 303.77), but not to tebufenozide(RF 50.04). PBO showed high synergism to abamectin (SR 2.11–12.23), and the synergism ratio positively related to the resistance level among different strains. Enzyme analysis also proved that the activity of cytochrome P450 monooxygenase (MFO) was notable enhanced in the strains resistant to both tebufenozide and abamectin (1.71- to 3.01-fold). Based on discussion, it was concluded that tebufenozide selection could resulted in significant cross-resistance of P. xylostella to abamectin. The major mechanism for the cross-resistance should be the enhancement of MFO activity. For resistance management, tebufenozide and abamectin would not recommend for rotational use. Staphylococcus aureus is a common human pathogen that causes various diseases including infections on the skin, in the bloodstream and the lower respiratory tracts. The emergence of methicillin-resistant S. aureus (MRSA) made the treatment of the bacterial infection more difficult, calling for development of new therapeutics. Compared with conventional antibiotic therapy, phage therapy offers a promising alternative to combat infections caused by MRSA. Here we showed that phage VB_SauS_SH-St 15644 isolated from sewage inhibited MRSA isolates in vitro and in the murine skin infection model. Phage VB_SauS_SH-St 15644 belongs to Siphoviridae. The genome of the phage is a linear, 45,111 bp double-stranded DNA with GC content of 33.35%. Among the 37 clinical MRSA isolates tested, 12 (32%) were lysed by the phage in vitro. The phage was relatively stable at temperatures up to 40 °C or between pH 6 and 9. However, the phage was sensitive to UV light. 80% of the phage was approximately adsorbed to the host MRSA isolate in 4 min. The one-step growth curve showed that the latent period was about 12 min followed by the growth period (about 9 min). The burst size was estimated at 13 PFU per infected cell. Furthermore, in a murine skin infection model, the phage significantly inhibited MRSA infection. Our study suggested that phage VB_SauS_SH-St 15644 has a potential to inhibit MRSA skin infection. • The phage VB_SauS_SH-St 15644 was isolated from sewage and well characterized regarding physiological and genetic aspects. • It belongs to Siphoviridae and the genome of the phage is a linear, 45,111 bp double-stranded DNA with GC content of 33.35%. • It inhibited MRSA isolates in vitro and in the murine skin infection model.
