Abstract

Dietary fish oil supplementation has long been shown to have significant health benefits, largely stemming from the anti-inflammatory activity of the ω-3 and ω-6 polyunsaturated fatty acids (PUFAs) present in fish oils. The anti-inflammatory properties of these fatty acids has been linked to beneficial health effects, such as protecting the heart, in individuals consuming diets rich in fish, or supplemented with fish oils. These effects are highly notable in the Māori people native to coastal regions of New Zealand; the significantly lower rates of heart problems compared to the inland populous has been attributed to the consumption of the green lipped mussel Perna Canaliculus. Commercially available health supplements based on the New Zealand green lipped mussel include a freeze-dried powder and a lipid extract (Lyprinol®), the latter of which has shown anti-inflammatory properties comparable to classical non-steroidal anti-inflammatory drugs (NSAIDs) such as Naproxen. GCMS analysis of Lyprinol by Murphy et al. showed the presence of a class of ω-4 and ω-6 PUFAs bearing a highly electron rich tri- or tetra-alkyl furan ring, which were designated furan fatty acids (F-acids). Due to their instability, isolation of F-acids from natural sources cannot be carried out and a general synthetic route toward this class of natural products was required. To accomplish this, the biosynthesis of F-acids was mimicked by utilising an oxidation of 1,3-dienes, followed by a dehydration/aromatisation to generate the heterocyclic furan ring. Singlet oxygen was chosen as the means of oxidising the conjugated dienes giving endoperoxides. To mimic the biological aromatisation of the peroxide intermediates the Appel reagent was chosen and, in a novel application of the reagent, was exploited as a mild, metal free method of dehydrating the cyclic peroxides to their corresponding furans. The biomimetic furan synthesis was applied toward a selection of 1,3-diene substrates bearing a range of pre-installed functionalities and substitution patterns including alkyl, aryl, alkenes, cyclopropyl rings, silyl ethers, and esters, alongside being applied to the total synthesis of the furan fatty acid F5. A brief exploration of the possibility of performing the aromatisation reaction under catalytic conditions was carried out, to determine whether endoperoxides could be converted to furans without needing a stoichiometric quantity of Appel reagent, by harnessing a catalytic quantity of triphenylphosphine oxide and regenerating the active P(V) species via reaction with oxalyl chloride. Furthermore, an optimisation study was carried out using a simple design of experiments procedure to ascertain the ideal conditions for carrying out the Appel-type dehydration of endoperoxides. Finally, the scope of the reaction sequence was expanded to be performed in a continuous flow reactor, with telescoping of the singlet oxygen diene oxidation and Appel-type aromatisation to increase oxidation yields and to omit the requirement for isolation of peroxide intermediates, and was applied to the synthesis of a selection of 2,5-diaryl furan motifs.

10. Aerosolized Particulate Matter and Blunting of Ciliary Dynamic Responses: Implications for Veterans and Active Duty Military in Southwest Asia.

Abstract

Introduction Respiratory diseases such as chronic rhinosinusitis and asthma are observed at increased rates in active duty and veteran military members, and they are especially prevalent in individuals who have been deployed in Southwest Asia during Operation Iraqi Freedom and Operation Enduring Freedom. Particulate matter, specifically the fine-grain desert sand found in the Middle East, may be a key source of this pathology because of deleterious effects on mucociliary clearance. Materials and Methods With IRB approval, human sinonasal tissue was grown at an air-liquid interface and cultures were exposed to different types and sizes of particulate matter, including sand from Afghanistan and Kuwait. Ciliary dynamic responses to mechanical stimulation and ATP application were assessed following particulate exposure. Results Particle size of the commercial sand was substantially larger than that of the sand of Afghan or Kuwaiti origin. Following exposure to particulate matter, normal dynamic ciliary responses to mechanical stimulation and ATP application were significantly decreased (P  < .01), with corresponding decreases in ATP-induced calcium flux (P  < .05). These changes were partially reversible with apical washing after a 16-h period of exposure. After 36 h of exposure to Middle Eastern sand, ciliary responses to purinergic stimulation were completely abolished. Conclusions There is a neutralization of the dynamic ciliary response following chronic particulate matter exposure, similar to ciliary pathologies observed in patients with chronic rhinosinusitis. Aerosolized particulate matter endured by military personnel in the Southwest Asia may cause dysfunctional mucociliary clearance; these data help to explain the increased prevalence of respiratory pathology in individuals who are or have been deployed in this region. Abstract

Background: The role of Akt in nasal immunity is unstudied. Akt phosphorylates and activates endothelial nitric oxide synthase (eNOS) expressed in epithelial ciliated cells. Nitric oxide (NO) production by ciliated cells can have antibacterial and antiviral effects. Increasing nasal NO may be a useful antipathogen strategy in chronic rhinosinusitis (CRS). We previously showed that small‐molecule Akt activator SC79 induces nasal cell NO production and suppresses IL‐8 via the transcription factor Nrf‐2. We hypothesized that SC79 NO production may additionally have antibacterial effects. Methods: NO production was measured using fluorescent dye DAF‐FM. We tested effects of SC79 during co‐culture of Pseudomonas aeruginosa with primary nasal epithelial cells, using CFU counting and live–dead staining to quantify bacterial killing. Pharmacology determined the mechanism of SC79‐induced NO production and tested dependence on Akt. Results: SC79 induced dose‐dependent, Akt‐dependent NO production in nasal epithelial cells. The NO production required eNOS and Akt. The NO released into the airway surface liquid killed P. aeruginosa. No toxicity (LDH release) or inflammatory effects (IL8 transcription) were observed over 24 h. Conclusions: Together, these data suggest multiple immune pathways are stimulated by SC79, with antipathogen effects. This in vitro pilot study suggests that a small‐molecule Akt activator may have clinical utility in CRS or respiratory other infection settings, warranting future in vivo studies. Abstract

Akt is an important kinase in metabolism. Akt also phosphorylates and activates endothelial and neuronal nitric oxide (NO) synthases (eNOS and nNOS, respectively) expressed in M0 (unpolarized) macrophages. We showed that e/nNOS NO production downstream of bitter taste receptors enhances macrophage phagocytosis. In airway epithelial cells, we also showed that the activation of Akt by a small molecule (SC79) enhances NO production and increases levels of nuclear Nrf2, which reduces IL-8 transcription during concomitant stimulation with Toll-like receptor (TLR) 5 agonist flagellin. We hypothesized that SC79's production of NO in macrophages might likewise enhance phagocytosis and reduce the transcription of some pro-inflammatory cytokines. Using live cell imaging of fluorescent biosensors and indicator dyes, we found that SC79 induces Akt activation, NO production, and downstream cGMP production in primary human M0 macrophages. This was accompanied by a reduction in IL-6, IL-8, and IL-12 production during concomitant stimulation with bacterial lipopolysaccharide, an agonist of pattern recognition receptors including TLR4. Pharmacological inhibitors suggested that this effect was dependent on Akt and Nrf2. Together, these data suggest that several macrophage immune pathways are regulated by SC79 via Akt. Abstract

This study aimed to investigate the impact of physical solid dispersions of spray-dried glibenclamide (SG) on the surface of microcrystalline cellulose (MC) and mannitol (M) surfaces, as well as their combination with phosphatidylcholine (P), on enhancing the dissolution rate of glibenclamide (G). Solid dispersions were prepared using varying proportions of 1:1, 1:4, and 1:10 for SG on the surface of MC (SGA) and M (SGM), and then combined with P, in a proportion of 1:4:0.02 using spray drying. The particle size, specific surface area, scanning electron microscopy (SEM), X-ray diffraction (XRD), and dissolution rate of SGA and SGM were characterized. SEM analysis revealed successful adhesion of SG onto the surface of the carrier surfaces. XRD showed reduced crystalline characteristic peaks for SGA, while SGM exhibited a sharp peaks pattern. Both SGA and SGM demonstrated higher dissolution rates compared to SG and G alone. Furthermore, the dissolution rates of the solid dispersions of SG, MC and P (SGAP), and SG, M, and P (SGMP) were sequentially higher than that of SGA and SGM. The study suggests that physical solid dispersions of SG on MC and M, along with their combination with P, can effectively enhance the dissolution rate of G. These findings may be valuable in developing of oral solid drug dosage forms utilizing SGA, SGM, SGAP, and SGMP. [ABSTRACT FROM AUTHOR]

Abstract

Bortezomib (BTZ), a boronic acid-derived proteasome inhibitor, is commonly employed in treating multiple myeloma (MM). However, the applications of BTZ are limited due to its poor stability and low bioavailability. Herein, we develop an optimized liposomal formulation of BTZ (L-BTZ) by employing a remote-loading strategy. This formulation uses Tiron, a divalent anionic catechol derivative, as the internal complexing agent. Compared to earlier BTZ-related formulations, this alternative formulation showed significantly greater stability due to the Tiron–BTZ complex's higher pH stability and negative charges, compared to the meglumine–BTZ complex. Significantly, the plasma AUC of L-BTZ was found to be 30 times greater than that of free BTZ, suggesting an extended blood circulation duration. In subsequent therapeutic evaluations using two murine xenograft tumor models of MM, the NCI-H929 and OPM2 models showed tumor growth inhibition (TGI) values of 37% and 57%, respectively. In contrast, free BTZ demonstrated TGI values of 17% and 11% in these models. Further, L-BTZ presented enhanced antitumor efficacy in the Hepa1-6 HCC syngeneic model, indicating its potential broader applicability as an antineoplastic agent. These findings suggest that the optimized L-BTZ formulation offers a significant advancement in BTZ delivery, holding substantial promise for clinical investigation in not merely MM, but other cancer types. Abstract

Importance: Recurrent coronary events in patients with recent myocardial infarction remain a major clinical problem. Non-invasive measures of coronary atherosclerotic disease activity have the potential to identify those at greatest risk.Objective: To determine whether non-invasive assessment of coronary atherosclerotic plaque activity could be associated with recurrent coronary events. Design: Prospective observational longitudinal cohort study recruiting participants between September 2015 and February 2020 with a minimum 2 years follow up.Setting: International multicenter study.Participants: Patients with multivessel coronary artery disease and recent myocardial infarction were eligible for inclusion. From 2,684 patients screened, 995 were eligible, 712 attended for imaging, and 704 had completed an interpretable scan and comprised the study population.Intervention: Coronary 18F-sodium fluoride positron emission tomography and coronary computed tomography angiography. Main Outcomes and Measures: Total coronary atherosclerotic plaque activity was assessed by 18F-sodium fluoride uptake. The primary endpoint was cardiac death or non-fatal myocardial infarction but was expanded during study conduct to include unscheduled coronary revascularization due to lower than anticipated primary event rates.Results: Participants were middle-aged (63.8±8.2 years) and predominantly male (85%). Total coronary atherosclerotic plaque activity was identified in 421 (60%) participants. After a median of 4 years follow-up, 141 participants experienced the primary endpoint: 9 had cardiac death, 49 non-fatal myocardial infarction and 83 unscheduled coronary revascularizations. Increased coronary plaque activity had no demonstrable association with the primary endpoint (hazard ratio (HR) 1.25 [95% confidence interval (CI) 0.89 to 1.76], P=0.20) or unscheduled revascularization (HR 0.98 [95% CI 0.64 to 1.49], P=0.91) but was associated with the secondary endpoints of cardiac death or non-fatal myocardial infarction (47 versus 19; HR 1.82 [95% CI 1.07 to 3.10], P=0.03) and all-cause mortality (30 versus 9; HR 2.43 [95% CI 1.15 to 5.12], P=0.02). These associations were similar after adjustment for differences in baseline clinical, coronary angiographic, and GRACE score characteristics (HR 1.76 [95% CI 1.00 to 3.10] (P=0.05) and HR 2.01 [95% CI 0.90 to 4.49], (P=0.09) respectively).Conclusions and Relevance: In patients with recent myocardial infarction, coronary atherosclerotic plaque activity was not associated with the primary composite endpoint. The findings suggesting risk of cardiovascular death or myocardial infarction in patients with elevated plaque activity warrants further research to explore its incremental prognostic implications.

Abstract

Literary value – in the sense of the worth, usefulness or importance of the literary – has been a topic of debate from no later than Plato’s impugning of poetry. But from the so-called canon wars of the last century to the present, literary value has also become a perplexing source of distress. With its complicities thoroughly unmasked, it no longer axiomatically serves as literary study’s central justification. Yet no consensus alternative has taken its place. This book, unlike other approaches to the topic, neither pursues an apologetic thesis about the most defining values of literature nor conversely provides a demystifying account of the ideological uses of specific ascribed values. Instead, arguing that the category of literary value is ultimately inescapable, it focuses pragmatically on everyday scholarly and pedagogical activities, proposing how we may reconcile that category’s inevitability with our understandable wariness of its intractable uncertainties and complicities. Toward these ends, it offers a preliminary theory of literary valuing and explores the problem of literary value and possible responses in respect to the literary edition, canonicity and interpretation. Much of this exploration occurs within Chaucer studies, which, because of Chaucer’s simultaneous canonicity and marginality, provides fertile ground for thinking through the problem’s challenges. The book thereby also supplies an extended reflection on the state of Chaucer studies. In using this subfield as a kind of synecdoche for the field as a whole, the book seeks to forge a viable rationale for literary studies within and without the academy.
