Wheat gluten protein (WG) is a kind of plant protein with high quality, low price and rich nutrition. But its low solubility limits its application in food products. Therefore, this work modified WG by pH cycling combined with heat treatment, to enhance its solubility and processing properties. Under the condition of pH cycling combined with heat treatment at 80 °C, the solubility of WG increased from 6.7% to 71.1%; the free sulfhydryl group and surface hydrophobicity were significantly improved (from 2.45 μmol/g and 340.86 to 15.27 μmol/g and 2080.73 respectively). The results of SDS-PAGE and the particle size showed that the WG molecules became smaller. The proteomic method was used and there were 3 subunits (A0A3B6LVB5, A0A3B5XV32 and A0A0A7AA82) observed having an impact on the improvement of solubility. Besides, the emulsifying activity index and emulsifying stability index of modified WG have been improved 8 times and 1.6 times, respectively. Furthermore, the powder oils made of the modified WG exhibited enhanced emulsifying and rehydrating characteristics than those of powder oils made of sodium caseinate and soybean protein isolates. Two New hole transport type exciton blocking (HTEB) materials, EB01 and EB02 for red Phosphorescent OLEDs (PhOLEDs), were designed and synthesized, both of which have an azaindolo[3,2,1- jk ]carbazole moiety chemically bonded with arylamine. As HTEB materials, their performances were investigated by comparing with device of HTEBL free. The results revealed that significant improvement in efficiencies can be realized by devices employing EB01 and EB02 compared to device without HTEBL. Moreover, in top-emitting devices using EB01 as HTEBL，very low driving voltages of 3 V, outstanding CE of more than 87 cd/A and PE of more than 91 Im/W at 1000 cd/m2, and remarkable long lifetime of LT95 with 84.03h at a constant current density of 50 mA/cm2 were simultaneously achieved, with deep-red emission of CIE x > 0.68, which is among the best performance in the reported deep-red PhOLEDs. Hepatotoxic microcystins (MCs) are produced and released by the harmful bloom-forming cyanobacteria, which severely threaten drinking water safety and human health due to their high toxicity, widespread distribution, and structural stability. The linearized microcystinase (MlrB) further hydrolyses the poisonous linearized MCs produced by the microcystinase-catalysed MCs to form tetrapeptides. Here, the purification and activity of MlrB were investigated. The results showed that the linearized products generated by 12.5 mg/L MC-LR and MC-RR were removed by purified recombinant MlrB at a protein concentration of 1 mg/L within 30 min. The high catalytic activity of MlrB can be obtained via heterologous expression and affinity purification, which lays the foundation for further studies on the properties and mechanism of MCs biodegradation enzymes. We previously reported that regenerating islet-derived protein 3 alpha (REG3A) exacerbates pancreatic malignancies. The mechanism of this effect has not been clearly elucidated. Here we first identified key differentially expressed genes (DEGs) and signal pathways in the pancreatic cancer cell line SW1990, compared to two control cell lines, by microarray analysis. We then identified key genes and pathways regulated by REG3A or the cytokine IL6 in SW1990 cells. Afterwards, these DEGs induced by REG3A or IL6 were subjected to KEGG pathway enrichment analysis and GO function analysis by the DAVID online tool. Ultimately, we constructed protein–protein interaction networks among the DEGs by Cytoscape. Among the three pancreatic cell lines, SW1990 exhibited highly deterioration with the activation of genes and pathways related to proliferation, survival, angiogenesis, and invasion. As a result, 50 DEGs enriched in 11 pathways were identified in REG3A-treated SW1990 cells, and 28 DEGs enriched in 9 pathways were detected in IL6-treated cells. Overall, results of microarray analysis followed by qRT-PCR and Western blotting suggest that REG3A regulates pancreatic cell growth by increasing the expression of at least 8 genes: JAK1 , STAT3 , IL10 , FOXM1 , KRAS , MYC , CyclinD1 , and c-fos ; and activation of at least 4 signal pathways: TGFβ, PDGF, angiogenesis and RAS. Similar results were obtained with IL6 treatment. In this work, by doping the lanthanide(iii)–hexafluoroacetylacetone complex into LAPONITE®, we obtained a lanthanide-based organic–inorganic hybrid material. The resulting hybrid materials were fully characterized with elementary analysis, scanning electron microscopy (SEM) and powder X-ray diffraction (PXRD) techniques. The Ln3+ and HFA loadings were experimentally determined to be roughly 0.3 per u.c. and 0.72 per u.c. by analyzing the supernatant (titration against EDTA) and elemental analysis, respectively. XRD patterns suggest that at least partial complexes are intercalated within the interlayers of the LAPONITE®. The in situ formation of luminescent Ln3+ complexes is confirmed by the luminescence data. Furthermore, the emission intensity ratio of the 5D4→7F5 transition (Tb3+) to the 5D0→7F2 transition (Eu3+) of the hybrid material containing both Eu3+ and Tb3+ can be linearly related to temperature in the range from 197 K to 287 K (temperature sensitivity: 1.107% per K), which will be an appealing alternative for in situ and real time detection of temperature in many special areas. Water-soluble hybrid composites with great potential for selective and sensitive sensing and that are obtained through simple, rapid, and environmentally friendly methods are highly desirable and remain a challenging task. Herein, we present luminescent hybrid composites that were realized by linking Na3[Ln(dpa)3] (dpa=2,6-pyridinedicarboxylic acid) to octa-amino functionalized polyhedral oligomeric silsesquioxane (POSS-NH2) through hydrogen-bonding interactions between the oxygen atoms of the carboxylate groups of dpa and the hydrogen-bond-donor amino groups. The resulting hybrid composites Ln(dpa)3@POSS-NH2 are highly soluble in aqueous solutions and the quantum yield of Eu(dpa)3@POSS-NH2 is as high as 56.5 % or 46.3 % in the solid state and in aqueous solution, respectively, as determined by using the integrating sphere method. The novel water-soluble luminescent hybrid composites exhibit high thermal and photostability, and the emitted colors of the resulting hybrid composite can be finely tuned by changing the Eu3+/Tb3+ ratio. The emergence of novel infectious bronchitis viruses (IBVs) has been reported worldwide. Between 2011 and 2014, eight IBV isolates were identified from disease outbreaks in northeast China. In the current study we analysed the S1 gene of these eight IBV isolates in addition to the complete genome of five of them. We confirmed that these isolates emerged through the recombination of LX4 and Taiwan group 1 (TW1) viruses at two switch sites, one was in the Nsp 16 region and the other in the spike protein gene. The S1 gene in these viruses exhibited high nucleotide similarity with TW1-like viruses; the TW1 genotype was found to be present in southern China from 2009. Pathogenicity experiments in chickens using three of the eight virus isolates revealed that they were nephropathogenic and had similar pathogenicity to the parental viruses. Through the combination of polyhedral oligosilsesquioxane (POSS) and a lanthanide complex, a hybrid material with enhanced luminescent properties was achieved. FTIR, XRD, SEM, and photoluminescence spectroscopy were employed to characterize the obtained material. Improved photo- and thermal-stability were observed in the hybrid material when compared to the mono-modified POSS derivative and individual lanthanide complex. Modification at each silicon vertex of the POSS core effectively increased the lanthanide concentration that was doped in the hybrid material and the 3D divergent configuration of octa-benzoic acid functionalized POSS (OBAPOSS) ensured the uniform distribution of lanthanide in the hybrid material to prevent aggregation of the lanthanide complex at a high concentration. The combination of POSS with lanthanide used in this study for designing and preparing luminescent hybrid materials is simple and applicable, and has provided new facilities for the design of optical devices and the substitution of pure lanthanide complexes in crystal engineering. The unique luminescence properties of the hybrid material, together with its good stability and processability, make it an ideal candidate for a wide range of applications such as materials for flat-panel displays, luminescent probes in bioassays, UV sensors and as laser materials and optoelectronic devices. An HPLC-MS method has been developed and validated for the quantitative determination of milbemycin oxime (MBO) in dog plasma. The developed method has been then applied in in vivo clinical studies to obtain pharmacokinetics of MBO in blood after its oral administration. Samples were extracted using solid-phase extraction (SPE). Sample proteins were precipitated with acetonitrile (ACN) and sodium chloride (NaCl) and then diluted with methanol and water. Calibration standards were prepared by using plasma matrix and following the same SPE procedure. Chromatographic separation was performed on a Waters C18 packed column (3.5 μm particles diameter; 3 × 100 mm) with a C18 guard column (3.5 μm particles diameter; 3 × 20 mm). The mobile phase was an 85:15 (v/v) mixed solution of ACN and 5 mM ammonium acetate. The calibration curve was linear over a concentration range of 2.0–500 ng/mL with a limit of quantitation of 2.0 ng/mL. The oral administration of a pellet of 2.5 mg MBO produced blood concentrations ranging from 6.10 ± 0.92 to 78.81 ± 4.38 ng/mL within 6 h, with a terminal half-time of 11.66 ± 0.93 h. The study was aimed to evaluate the morphology and immune function development of the jejunum and ileum in squab pigeons (Columba livia), by determining the villus ultrastructure, secretory IgA, and cytokines. Eight squabs were randomly selected and sampled on the day of hatch (DOH), d 7 (D 7), 14 (D 14), and 21 (D 21) post-hatch, respectively. The results showed that under transmission electron microscope, the enterocyte circumference in jejunum and ileum decreased with age. The tight junction involved in enterocyte circumference of jejunal villi plateaued from D 7, whereas that of ileal villi changed irregularly. The microvilli of jejunal and ileal villi was maximum at D 14. Under scanning electron microscope, the villus morphology of jejunum and ileum appeared finger-shaped at DOH. After D 7, the jejunal villi were still finger-shaped whereas the ileal villi were leaflike. The secretory IgA in jejunum was significantly increased at D 21. The TGF-β decreased linearly in jejunum and ileum. The anti-inflammatory cytokines increased linearly and proinflammatory cytokines decreased linearly in jejunum and ileum with age. In conclusion, the morphological changes of jejunal epithelium were concentrated at DOH–D 7 and ileal epithelium at DOH–D 14 mainly. Near infrared reflectance spectroscopy (NIRS) and reference data were used to determine the amylose contents of single maize seeds to enable rapid, effective selection of individual seeds with desired traits. To predict the amylose contents of a single seed, a total of 1069 (865 as calibration set, 204 as validation set) single seeds representing 120 maize varieties were analyzed using chemical methods and performed calibration and external validation of the 150 single seeds set in parallel. Compared to various spectral pretreatments, the regression of partial least squares (PLS) with mathematical treatment of Harmonization showed the final optimization. The single-seed amylose contents showed the root mean square error of calibration (RMSEC) of 2.899, coefficient of determination for calibration (R2) of 0.902, and root mean square error of validation (RMSEV) of 2.948. In external validations, the coefficient of determination in cross-validation (r2), root mean square error of the prediction (RMSEP) and ratio of the standard deviation to SEP (RPD) were 0.892, 2.975 and 3.086 in the range of 20–30%, respectively. The use of poly(vinylidene fluoride) (PVDF) microfiltration (MF) membranes to purify oily water has received much attention. However, it is challenging to obtain high-performance PVDF microfiltration membranes due to severe surface fouling and rapid decline of permeability. This study explored a new approach to fabricate high-performance PVDF/silica (SiO2) composite membrane via the use of a polymer solution featuring lower critical solution temperature (LCST) characteristics and the non-solvent thermally induced phase separation method (NTIPS). Coupling with morphological observations, the membrane formation kinetics were analyzed in depth to understand the synergistic effect between the LCST solution properties and fabrication conditions in NTIPS. Utilizing such a synergistic effect, the transition from finger-like macrovoid pores to bi-continuous highly connected pores could be flexibly tuned by increasing the PVDF concentration and the weight ratio of SiO2/PVDF in the dope solution and by raising the coagulation temperature to above the LCST of the solution. The filtration experiments with surfactant-stabilized oil-water emulsion showed that the permeation flux of the PVDF/SiO2 composite membranes was higher than 318 L·m−2·h−1·bar−1 and the rejection above 99.2%. It was also shown that the PVDF/SiO2 composite membranes, especially those fabricated above the LCST, demonstrated better hydrophilicity, which resulted in significant enhancement in the anti-fouling properties for oil/water emulsion separation. Compared to the benchmark pure PVDF membrane in oily water purification, the optimal composite membrane T70 was demonstrated via the 3-cycle filtration experiments with a significantly improved flux recovery ratio (Frr) and minimal reduced irreversible fouling (Rir). Overall, with the developed method in this work, facile procedure to tune the membrane morphology and pore structure was demonstrated, resulting in high performance composite membranes suitable for oil/water emulsion separation. • The multiplex dPCR system newly established here was the first reported quadruple dPCR assay for four common human Plasmodium species, which enables a four-in-one detection. • The analytical sensitivity of multiplex dPCR was as low as 0.557 copies/μL (95% CI 0.521 to 0.607), and it had a sensitivity of 98.0% and a specificity of 100% in clinical samples to make up for the false negative of microscopic examination. • The multiplex dPCR system has great potential in diagnosing mixed-infection and monitoring pharmacotherapy effectiveness. • The multiplex dPCR system presents a promising candidate as an absolute quantitative tool in clinical practice. Blood smear examination through traditional optical microscopy is the gold standard for malaria diagnosis. However, it imposes strict requirements for operational staff and its sensitivity cannot perfectly satisfy the needs of clinical requirements. More sensitive and accurate modern technologies should be applied to this field. Digital PCR (dPCR), as an absolute quantification detection method, can serve as an effective tool to facilitate the diagnosis and classification of different malaria species. We aimed to establish a new multiplex dPCR detection system for four main Plasmodium species: P. vivax , P. falciparum , P. ovale and P. malariae , which can distinguish exact species of malaria by one PCR reaction. A total of 39 patients were identified as malaria-positive by microscopic examination in Huashan Hospital from 2016 to 2021; seventy blood samples from these patients were collected. Additionally, 20 healthy individuals, 20 patients with fever and 6 patients with other types of blood parasites infection were also included in this study. Each blood sample was subjected to examination by both blood smears and dPCR. By optimizing four different fluorescence-labeled probes in one reaction system, dPCR permitted the performance of accurate quantitation and working out the exact number of copies of malaria DNA per microliter in whole blood. Rapid diagnostic tests were also conducted to verify part of the results obtained by dPCR. The dPCR system was able to make rapid diagnosis and quantification of malaria DNA samples. The analytical sensitivity of multiplex dPCR was as low as 0.557 copies/μL (95% CI 0.521 to 0.607), and it had a sensitivity of 98.0% and a specificity of 100% in clinical samples. Additionally, three multiple malaria co-infection samples have been detected by this dPCR system, including one triple malaria infection case. By testing consecutive daily blood samples of Patient 39, dPCR facilitated monitoring the efficacy of drug treatment. It showed that the DNA concentrations of P. falciparum ranged from 5474 copies/μL to 0 copies/μL, which can reflect the efficacy of antimalarials in real time. This study also found that haemocyte samples (plasma removed) rather than whole blood had higher malaria detection capability and an enhanced positive rate. The multiplex dPCR system newly established here made a substantial contribution in detecting malaria infection at low concentrations. Cholestasis is an important predisposing factor of liver diseases, such as hepatocyte necrosis, liver fibrosis and primary biliary cirrhosis. In this study, we aimed to investigate the effects of Kinsenoside (KD), a natural active ingredient of Anoectochilus roxburghii, on estrogen-induced cholestatic liver injury in Sprague-Dawley rats and the underlying mechanism. The rats were randomly divided into six groups: control group, model group, low-dose KD group (50 mg/kg body weight, KD-L), medium-dose KD group (100 mg/kg body weight, KD-M), high-dose KD group (200 mg/kg body weight, KD-H) and ursodeoxycholic acid group (40 mg/kg body weight, UDCA). 17α-Ethinylestradiol (EE) was used to establish an experimental animal model of estrogen-induced cholestasis (EIC). The results demonstrated that KD alleviated liver pathologic damage, serum biochemical status and inhibited hepatocellular microstructure disorder and bile duct hyperplasia in EE-induced cholestatic rats. Mechanically, KD alleviated EE-induced cholestatic liver injury by inhibiting inflammatory responses and regulating bile acid homeostasis. Concretely, KD reduced the expression of IL-1β and IL-6 by inhibiting NF-κB p65 to suppress EE-mediated inflammation in rat liver. KD enhanced the expression of FXR and inhibited EE-mediated reduction of FXR in vitro and in vivo. It was the potential mechanism that KD mitigates cholestasis by increasing efflux and inhibiting uptake of bile acids via FXR-mediated induction of bile salt export pump (BSEP) and reduction of Na+-dependent taurocholate cotransport peptide (NTCP) to maintain bile acid homeostasis. Moreover, KD repressed the bile acid synthesis through reducing the expression of synthetic enzyme (CYP7A1), thereby normalizing the expression of metabolic enzyme (SULT2A1) of bile acid. By using the saturation shake-flask method, the saturated solubility of bezafibrate in neat water, methanol, ethanol, n-propanol and isopropanol and aqueous co-solvent mixtures of methanol (1) + water (2), ethanol (1) + water (2), isopropanol (1) + water (2) and n-propanol (1) + water (2) was experimentally acquired at temperatures from 283.15 Kto 323.15 K and local atmospheric pressure. The maximum solubility magnitudes in mole fraction were recorded in the neat solvents of methanol/ethanol/ n -propanol/isopropanol for corresponding solutions. The mole fraction solubility was described mathematically by using the van't Hoff-Jouyban-Acree and Jouyban-Acree models. The largest value of relative average deviations was 6.23%; and, root-mean-square deviations, 8.92 × 10−5. The local mole fractions of water and co-solvent (methanol, ethanol, n -propanol or isopropanol) around the drug bezafibrate were quantitatively investigated by the use of Inverse Kirkwood–Buff integrals method. Results indicated that the bezafibrate was preferentially solvated by methanol (ethanol, n -propanol or isopropanol) in alcohol-rich and intermediate compositions in the alcohol mixtures; while in water-rich compositions, bezafibrate was preferentially solvated by water. The main factors relating to solvent effect were inspected through Kamlet and Taft linear solvation energy relationships, demonstrating that the cavity formation energy and dipolarity-polarizability in methanol solutions, cavity formation energy and basicity in ethanol solutions, and cavity formation energy in n -propanol (isopropanol) solutions played the principal role upon the bezafibrate solubility variation. Ubiquitous noise pollution has been associated with significant negative impacts on human health. However, current porous sound‐absorbing materials encounter considerable obstacles such as thick density, narrow absorbing band, and limited function. Here, a facile‐producing method for lightweight and efficiently sound‐absorbing aerogels made from bacterial cellulose (BC) and poly(vinyl alcohol) (PVA) is presented. The fabricated anisotropic aerogels with directional pores exhibit a minimum density as low as 11.3 mg cm−3. Meanwhile, the lamellar aerogels with low areal density (20.89 mg cm−2) exhibit remarkable noise attenuation performance with the noise reduction coefficient of 0.51.Furthermore, the BC‐PVA‐Ba0.85Ca0.15Zr0.9Ti0.1O3 (BCZT) aerogels show enhanced sound absorption performance, and these aerogels are self‐powered sensors to monitor vehicle collisions and human gestures. The algorithm yields high accuracy in human gesture recognition (100%) based on the deep‐learning model. Severe acute pancreatitis (SAP) begins with premature activation of enzymes, promoted by the immune system, triggering a potential systemic inflammatory response that leads to organ failure with increased mortality and a bleak prognosis. Interleukin-22 (IL-22) is a cytokine that may have a significant role in SAP. IL-22, a member of the IL-10 cytokine family, has garnered growing interest owing to its potential tissue-protective properties. Recently, emerging research has revealed its specific effects on pancreatic diseases, particularly SAP. This paper provides a review of the latest knowledge on the role of IL-22 and its viability as a therapeutic target in SAP. Highlights: 1. Transparent heat mirrors have been attracting a great deal of interest in the last few decades due to their broad applications, which range from solar thermal convection to energy-saving. Here, we present a flexible Polyethylene terephthalate/Ag-doped Indium tin oxide/Polydimethylsiloxane (PAIP) thin film that exhibits high transmittance in visible range and low emissivity in the thermal infrared region. Experimental results show that the temperature of the sample can be as high as 108 °C, which is ~23 °C higher than that of a blackbody control sample under the same solar radiation. Without solar radiation, the temperature of the PAIP thin film is ~6 °C higher than that of ordinary fabric. The versatility of the large-area, low-radiation-loss, highly-transparent and flexible hydrophobic PAIP thin film suggest great potential for practical applications in thermal energy harvesting and manipulation. • Three unusual alkaloids were isolated from Aspergillus versicolor F210. • 21- Epi -taichunamide D features a rare methylsulfonyl unit. • (±)- Versicalin A exist as two pairs of keto-enol tautomers. • (±)-Versicalin A exhibited moderate cytotoxicity against HL-60 cells. Three new alkaloids, 21- epi -taichunamide D (1) and (±)-versicalin A ((±)- 6), along with four known analogues (2 – 5) were isolated from the endophytic Aspergillus versicolor F210, which harbored in the bulbs of Lycoris radiata. Structurally, compound 1 is the third natural indole alkaloid with a rare methylsulfonyl unit, and (±)- 6 existing as two pairs of keto-enol tautomers ((+)- 6a/ (+)- 6b and (−)- 6a/ (−)- 6b , respectively), represent the first naturally occurring alkaloids with a rare 2,3-dihydro-1 H -isoindol-1-one unit fused to a tautomeric diisopentenyl cyclohexenone unit. Gas-liquid chromatography is an effective method to determine infinite dilution activity coefficients (γ ∞). Wall-coated open-tubular (WCOT) column which offers more advantages over packed column should be a preferable column type; however, the small carrier gas flow rate and stationary phase amount in WCOT columns limit its application in the determination of γ ∞. Mathematical strategy made some progress to avoid the quantification problem in the determination of γ ∞ by static-wall-coated open-tubular (SWCOT) columns. However, the previously reported strategy was based on the assumption that SWCOT column was geometrically an ideal hollow cylinder, which indeed deviates from the reality. In this study, without that assumption, we derived a new microgeometry-independent equation by using the relationship between the hold-up volume (V M) and the volume of stationary phase (V L), and used it to measure the γ ∞ of various organic solutes in two ionic liquids (ILs) 1‑butyl‑3-methylimidazolium dicyanamide and 1,3-dibutyronitrile-imidazolium bis((trifluoromethyl)sulfonyl)imide, both of which contain double cyano groups in the anion or cation. Phase loading study was adopted to eliminate the influence of interfacial adsorption to partition. The infinite dilution partial molar excess enthalpy, selectivity and capacity were directly calculated from the experimental γ ∞ values, and the linear solvation energy relationship (LSER) model was used to characterize the specific properties of both ILs. Simple Summary: Research on antioxidant status and lipid metabolism in pigeon squabs as meat type poultry is limited. The objective of this study was to explore the age-related changes in the antioxidant status and lipid metabolism of pigeon squabs (Columba livia). Ten squabs were randomly selected, out of 400 hatched squabs, on the following days: day of hatching (DOH) and days 7 (D7), 14 (D14), 21 (D21) post-hatch, respectively. BW, serum lipid levels, antioxidant capacity indices, lipid metabolism-related enzyme activities, lipid metabolism-related gene expression, and liver morphology were determined. Results indicated that the phase from DOH to D14 was a growth spurt, especially in the first seven days. The antioxidant capacity of squabs had a continuous decline from DOH to D14. Besides, changes regarding lipid metabolism mainly occurred in the phase from DOH to D14. The first seven days mainly showed less lipid breakdown, while the second seven days displayed more complicated lipid metabolism. The results obtained from this study suggested that, in the pigeon industry, it is better to take nutritional manipulation in squabs before D14. In this paper, low Ni-doped Co 3 O 4 (Ni: 2 wt%, 4 wt%, 6 wt% and 8 wt%) porous nanoplates are synthesized by hydrothermal method and annealing treatment. For the low Ni-doped Co 3 O 4 , X-ray diffraction (XRD) and Raman data show that the Ni ions are successfully incorporated into the Co 3 O 4 lattices. Electrochemical measurements indicate that Co 3 O 4 with low amount of Ni doping possesses remarkably enhanced activity for oxygen evolution reaction (OER) and hydrogen evolution reaction (HER) in alkaline media. And especially, 4 wt% Ni-doped Co 3 O 4 possesses the most efficient activity with an overpotential of 240 mV at the current density of 10 mA cm−2 for OER and 120 mV for HER in 1 M KOH than the pure Co 3 O 4 and other Ni-doped Co 3 O 4 samples. To investigate the structure and immunomodulatory activity of polysaccharide from Solanum muricatum, a novel acid polysaccharide named SMP-3a was purified from Solanum muricatum pulp through DEAE-52 cellulose column and Sephadex G-200 chromatography. Monosaccharide composition analysis showed that SMP-3a was mainly composed of rhamnose, arabinose, galactose, and galacturonic acid with the molar ratio of 1.09:2.64:1.54:1. The average molecular weight was found to be 227 kDa by high performance gel permeation chromatography (HPGPC). Thermal studies revealed the SMP-3a was a thermally stable polymer. Based on the results of methylation and NMR analysis, the backbone chain of SMP-3a was composed of →2)-α-l-Rhap-(1→, →4)-α-d-GalpA-(1→ and →4)-α-d-Galp-(1→. The side chain was consisted of α-l-Araf-(1→ and →5)-α-l-Araf-(1→. Microcystinase (MlrA) catalyzes the first and most important biodegradation step of hepatotoxic microcystin-LR (MC-LR) produced and released from cyanobacterial cells, and the underlying catalytic mechanism is not completely understood yet. MlrA was postulated previously to be a metalloprotease with an active site of H260AIH263NE265, a variant of the common metal-binding motif of HEXXH. Through comparison with representative modes in HEXXH-containing metalloproteases, molecular dynamics simulation, homology modeling, and docking, the active sites of MlrA involved in the MC-LR biodegradation by Sphingomonas sp. USTB-05 were predicted. Site-directed mutants of MlrA were constructed for verification then. The results show that MlrA is likely not a metalloprotease, but a glutamate protease belonging to type II CAAX prenyl endopeptidases. Biodegradation is efficient for removing cyanobacterial toxins, such as microcystins (MCs) and nodularin (NOD). However, not all the microbial strains with the microcystin-biodegrading enzymes MlrA and MlrC could biodegrade NOD. Studies on genes and enzymes for biodegrading NOD can reveal the function and the biodegradation pathway of NOD. Based on successful cloning and expression of the USTB-05-A and USTB-05-C genes from Sphingopyxis sp. USTB-05, which are responsible for the biodegradation of MCs, the pathway for biodegrading NOD by these two enzymes was investigated in this study. The findings showed that the enzyme USTB-05-A converted cyclic NOD (m/z 825.4516) into its linear type as the first product by hydrolyzing the arginine and Adda peptide bond, and that USTB-05-C cut off the Adda and glutamic acid peptide bond of linearized NOD (m/z 843.4616) and produced dimeric Adda (m/z 663.4377) as the second product. Further, based on the homology modeling of enzyme USTB-05-A, site-directed mutants of USTB-05-A were constructed and seven crucial sites for enzyme USTB-05-A activity were found. For the first time, waste-seashell-derived CaO catalysts were used as high-performance solid base catalysts for cyclopentanone self-condensation, which is an important reaction in bio-jet fuel or perfume precursor synthesis. Among the investigated seashell-derived catalysts, Scapharca Broughtonii-derived CaO catalyst (S-shell-750) exhibited the highest dimer yield (92.1%), which was comparable with commercial CaO (88.2%). The activity sequence of different catalysts was consistent with the CaO purity sequence and contact angle sequence. X-ray diffraction (XRD) results showed that CaCO3 in waste shell were completely converted to CaO after calcination at 750 °C or above for 4 h. CO2 temperature-programmed desorption (CO2-TPD) results indicate that both the amount and strength of base sites increase significantly when the calcination temperature climbs to 750 °C. Therefore, we can attribute the excellent performance of S-shell-750/850/950 catalysts to the higher CaO content, relatively low hydrophilicity, and stronger acidity and basicity of this catalyst. This study aimed to delineate correlation between stress hyperglycemia ratio (SHR) and clinical outcomes among patients in the cardiac intensive care unit (CICU). Participants were categorized based on their SHR threshold values. Key outcomes were short-term mortality and major adverse cardiovascular events (MACEs) at 1-year follow-up. The association between SHR and outcomes was estimated using inverse probability of treatment weighting (IPTW) and Kaplan-Meier analyses. The C-statistic was used to gauge the predictive capability of SHR. The study included 1,133 patients from the Medical Information Mart for Intensive Care IV and 412 from the Second Affiliated Hospital of Wenzhou Medical University. Kaplan-Meier curves revealed that individuals with elevated SHR exhibited higher 90-day mortality and MACEs. When considering SHR levels and diabetes status simultaneously, those with increased SHR but non-diabetes had the highest 90-day mortality and MACEs. SHR was associated with short-term mortality and MACEs (HR adjusted 1.63 95%CI 1.15–2.30; HR IPTW 1.47 95%CI 1.05–2.05). Upon integrating SHR into the foundational model, the C-statistic was 0.821, outperforming other hyperglycemia metrics. SHR is a valuable indicator for predicting poor outcomes in CICU patients. A promising bacterial strain for biodegrading dibutyl phthalate (DBP) was successfully isolated from activated sludge and characterized as a potential novel Microbacterium sp. USTB-Y based on 16S rRNA sequence analysis and whole genome average nucleotide identity (ANI). Initial DBP of 50 mg/L could be completely biodegraded by USTB-Y both in mineral salt medium and in DBP artificially contaminated soil within 12 h at the optimal culture conditions of pH 7.5 and 30 ℃, which indicates that USTB-Y has a strong ability in DBP biodegradation. Phthalic acid (PA) was identified as the end-product of DBP biodegraded by USTB-Y using GC/MS. The draft genome of USTB-Y was sequenced by Illumina NovaSeq and 29 and 188 genes encoding for putative esterase/carboxylesterase and hydrolase/alpha/beta hydrolase were annotated based on NR (non redundant protein sequence database) analysis, respectively. Gene3781 and gene3780 from strain USTB-Y showed 100% identity with dpeH and mpeH from Microbacterium sp. PAE-1. But no phthalate catabolic gene (pht) cluster was found in the genome of strain USTB-Y. Simple Summary: Clustered regularly interspaced short palindromic repeats (CRISPR) ribonucleoprotein (RNP)-mediated precise genome editing can modify a specific targeted gene to establish cell models and improve animal traits, thus playing a significant role in animal breeding. However, the efficiency of precise gene editing is low, which limits its widespread use. To overcome this problem in porcine cells, we used small molecules to improve the efficiency of CRISPR RNP-mediated precise genome editing in porcine fetal fibroblasts (PFFs). We demonstrated that a single small molecule including L-189, NU7441, SCR7, L755507, RS-1, and Brefeldin A increased the efficiency of homology-directed repair (HDR) during precise gene editing in PFFs, respectively. These results provided methodological support for the establishment of an efficient and precise gene editing system in pigs. The aim of this study was to verify whether small molecules can improve the efficiency of precision gene editing using clustered regularly interspaced short palindromic repeats (CRISPR) ribonucleoprotein (RNP) in porcine cells. CRISPR associated 9 (Cas9) protein, small guide RNA (sgRNA), phosphorothioate-modified single-stranded oligonucleotides (ssODN), and different small molecules were used to generate precise nucleotide substitutions at the insulin (INS) gene by homology-directed repair (HDR) in porcine fetal fibroblasts (PFFs). These components were introduced into PFFs via electroporation, followed by polymerase chain reaction (PCR) for the target site. All samples were sequenced and analyzed, and the efficiencies of different small molecules at the target site were compared. The results showed that the optimal concentrations of the small molecules, including L-189, NU7441, SCR7, L755507, RS-1, and Brefeldin A, for in vitro-cultured PFFs' viability were determined. Compared with the control group, the single small molecules including L-189, NU7441, SCR7, L755507, RS-1, and Brefeldin A increased the efficiency of HDR-mediated precise gene editing from 1.71-fold to 2.28-fold, respectively. There are no benefits in using the combination of two small molecules, since none of the combinations improved the precise gene editing efficiency compared to single small molecules. Background: Fluvoxamine is one of the selective serotonin reuptake inhibitors (SSRIs) that are regarded as the first-line drugs to manage mental disorders. It has been also recognized with the potential to treat inflammatory diseases and viral infection. However, the effect of fluvoxamine on autoimmune diseases, particularly type 1 diabetes (T1D) and the related cellular and molecular mechanisms, are yet to be addressed. Method: Herein in this report, we treated NOD mice with fluvoxamine for 2 weeks starting from 10-week of age to dissect the impact of fluvoxamine on the prevention of type 1 diabetes. We compared the differences of immune cells between 12-week-old control and fluvoxamine-treated mice by flow cytometry analysis. To study the mechanism involved, we extensively examined the characteristics of CD4+ T cells with fluvoxamine stimulation using RNA-seq analysis, real-time PCR, Western blot, and seahorse assay. Furthermore, we investigated the relevance of our data to human autoimmune diabetes. Result: Fluvoxamine not only delayed T1D onset, but also decreased T1D incidence. Moreover, fluvoxamine-treated NOD mice showed significantly attenuated insulitis coupled with well-preserved β cell function, and decreased Th1 and Th17 cells in the peripheral blood, pancreatic lymph nodes (PLNs), and spleen. Mechanistic studies revealed that fluvoxamine downregulated glycolytic process by inhibiting phosphatidylinositol 3-kinase (PI3K)-AKT signaling, by which it restrained effector T (Teff) cell differentiation and production of proinflammatory cytokines. Simple Summary: This study investigated the developmental differences between in vivo-developed (IVV) and in vitro embryos (in vitro fertilization (IVF) and parthenogenetic activation (PA)) in pigs using single-cell RNA sequencing data from the oocyte to the 8-cell stages. Findings show that in vitro embryos have more similar developmental trajectories and lower gene diversity, especially PA, compared to IVV. Significant variations were observed in maternal mRNA, influencing mRNA splicing, energy metabolism, and chromatin remodeling, where key genes like SMARCB1 and HDAC1 (in vivo) and SIRT1 and EZH2 (in vitro) might be playing crucial roles. Despite similar ZGA timings across embryo types, IVV embryos exhibited more upregulated genes, particularly during major ZGA, and global epigenetic modification differences were evident from the oocyte stage and continued to expand during minor ZGA. They also uniquely upregulated genes linked to mitochondrial function, ATP synthesis, and oxidative phosphorylation. A notable difference in mRNA degradation between in vivo and in vitro embryos was observed during major ZGA, and the timely decay of maternal mRna in IVV except in in vitro embryos was related to phosphatase activity and cell junctions. The study also highlighted the higher expression of mitochondrially encoded genes in in vitro embryos, though nucleosome occupancy and ATP8 expression were higher in IVV. In vitro-fertilized (IVF) and parthenogenetically activated (PA) embryos, key to genetic engineering, face more developmental challenges than in vivo-developed embryos (IVV). We analyzed single-cell RNA-seq data from the oocyte to eight-cell stages in IVV, IVF, and PA porcine embryos, focusing on developmental differences during early zygotic genome activation (ZGA), a vital stage for embryonic development. (1) Our findings reveal that in vitro embryos (IVF and PA) exhibit more similar developmental trajectories compared to IVV embryos, with PA embryos showing the least gene diversity at each stage. (2) Significant differences in maternal mRNA, particularly affecting mRNA splicing, energy metabolism, and chromatin remodeling, were observed. Key genes like SMARCB1 (in vivo) and SIRT1 (in vitro) played major roles, with HDAC1 (in vivo) and EZH2 (in vitro) likely central in their complexes. (3) Across different types of embryos, there was minimal overlap in gene upregulation during ZGA, with IVV embryos demonstrating more pronounced upregulation. During minor ZGA, global epigenetic modification patterns diverged and expanded further. Specifically, in IVV, genes, especially those linked to H4 acetylation and H2 ubiquitination, were more actively regulated compared to PA embryos, which showed an increase in H3 methylation. Additionally, both types displayed a distinction in DNA methylation. During major ZGA, IVV distinctively upregulated genes related to mitochondrial regulation, ATP synthesis, and oxidative phosphorylation. Coumarin is widely used in personal care products and pharmaceutical industry, which leads to the release of this compound into environment as an emerging contaminant. Here, a promising strain USTB-Z for biodegrading coumarin was successfully isolated from botanical soil and characterized as a potential novel Pseudomonas sp. based on 16S rDNA sequence analysis and orthologous average nucleotide identity tool. Initial coumarin up to 800 mg/L could be completely removed by USTB-Z within 48 h at the optimal culture conditions of pH 7.3 and 30 °C, which indicates that USTB-Z has a strong capacity in coumarin biodegradation. The biodegradation products of coumarin were further investigated using HPLC and Q-TOF LC/MS, and melilotic acid and 2,3-dihydroxyphenylpropionic acid were identified. The draft genome of strain USTB-Z was sequenced by Illumina NovaSeq, and 21 CDSs for NAD (P)-dependent oxidoreductase, 43 CDSs for hydrolase, 1 CDS for FAD-depend monooxygenase, 1 CDS for 3-hydroxycinnamic acid hydroxylase, 21 CDSs for dioxygenase, and 5 CDSs for fumarylacetoacetate (FAA) hydrolase were annotated and correlated to coumarin biodegradation. Background: Gastric cancer at different locations has distinct prognoses and biological behaviors, but the specific mechanism is unclear. Methods: Non-targeted metabolomics was performed to examine the differential metabolite phenotypes that may be associated with the effects of tumor location on the prognosis of gastric cancer. And silencing of the rate-limiting enzyme to evaluate the effect of abnormal changes in metabolic pathway on the functional biological assays of gastric cancer cells HGC-27 and MKN28. Results: In a retrospective study of 94 gastric cancer patients, the average survival time of patients with gastric cancer in the middle third of the stomach was significantly lower than that of patients with gastric cancer in other locations (p < 0.05). The middle third location was also found to be an independent risk factor for poor prognosis (HR = 2.723, 95%CI 1.334–5.520), which was closely associated with larger tumors in this location. Non-targeted metabolomic analysis showed that the differential metabolites affected 16 signaling pathways including arginine synthesis, retrograde endocannabinoid signaling, arginine biosynthesis, and alanine and aspartate and glutamate metabolism between gastric cancer and normal tissue, as well as between tumors located in the middle third of the stomach and other locations. Argininosuccinate synthetase 1 (ASS1), the rate-limiting enzyme of the arginine biosynthesis pathway, catalyzes the production of argininosuccinic acid. Here, knockdown of ASS1 significantly inhibited the proliferation, colony formation, and migration/invasion of gastric cancer cells, and promoted apoptosis. Conclusions: Our study suggests that abnormal arginine synthesis may lead to larger tumor size and worse prognosis in gastric cancer located in the middle third position of the stomach. Purpose: Typically, IgA nephropathy is a slowly progressive type of glomerulonephritis. High-grade proteinuria and hypertension are predictors of reduced kidney function. However, we found some normotensive patients with mild proteinuria could exhibit impaired renal function at the time of IgAN diagnosis. We therefore conduct a study to highlight the occurrence of these cases and to define their clinical characteristics and outcomes. Methods: The clinical, laboratory, pathological manifestations and outcomes of these IgAN patients were collected and were compared with normotensive IgA nephropathy patients with mild proteinuria and normal renal function. Patients were analyzed according to different pathological characteristics. Survival curves were constructed according to the Kaplan–Meier method. Results: Of all normotensive IgA nephropathy patients with mild proteinuria, 108 (10.1%) patients had impaired renal function. Ischemic sclerosis (79 patients) and fibrous crescent (25 patients) were the main pathological characteristics. Macroscopic hematuria (1.3%), prodromal infection (13.9%) and high serum IgA (11.4%) were significantly lower prevalences, but only proteinuria (26.6%) was more common in ischemic sclerosis group patients. Only hematuria were not found in ischemic sclerosis group and crescent group patients. The median follow-up were about 5 years. Patients in crescent group had a poor outcome compared with patients in ischemic sclerosis group. Conclusions: Some normotensive IgA nephropathy patients with mild proteinuria had impaired renal function at diagnosis. Ischemic sclerosis and fibrous crescent were the main pathological features in these patients. Patients in the crescent group had a worse outcome than patients in the ischemic sclerosis group. Both inosine and guanosine are precursors of uric acid that may cause the diseases of hyperuricemia and gout in humans. Here, a promising bacterial strain for efficiently biodegrading both inosine and guanosine was successfully isolated from a healthy human intestine and identified as Bacillus paranthracis YD01 with 16S rRNA analysis. An initial amount of 49.6 mg·L−1 of inosine or 49.9 mg·L−1 of guanosine was completely removed by YD01 within 12 h, which showed that YD01 had a strong ability to biodegrade inosine and guanosine. Furthermore, the initial amount of 49.2 mg·L−1 of inosine or 49.5 mg·L−1 of guanosine was totally catalyzed by the intracellular crude enzymes of YD01 within 6 h, and the initial inosine amount of 49.6 mg·L−1 or guanosine of 49.7 mg·L−1 was biodegraded by the extracellular crude enzymes of YD01 within 9 h. Illumina Hiseq sequencing and database gene annotation were used to elucidate the genomic characteristics of B. paranthracis YD01. Purine nucleoside phosphorylase, encoded by gene 1785, gene 3933, and gene 4403, was found in the KEEG database, which played a crucial role in the biodegradation of inosine and guanosine. Extensive use of phthalic acid esters (PAEs) as plasticizer causes diffusion into the environment, which posed a great threat to mankind. It was reported that Comamonas sp. was a potentially robust aromatic biodegrader. Although the biodegradation of several PAEs by Comamonas sp. was studies, the comprehensive genomic analysis of Comamonas sp. was few reported. In the present study, one promising bacterial strain for biodegrading diethyl phthalate (DEP) was successfully isolated from activated sludge and characterized as Comamonas sp. USTBZA1 based on the 16S rRNA sequence analysis. The results showed that pH 7.5, 30 °C and inoculum volume ratio of 6% were optimal for biodegradation. Initial DEP of 50 mg/L could be completely biodegrade by strain USTBZA1 within 24 h which conformed to the Gompertz model. Based on the Q-TOF LC/MS analysis, monoethyl phthalate (MEP) and phthalic acid (PA) were identified as the metabolic products of DEP biodegradation by USTBZA1. Furthermore, the whole genome of Comamonas sp. USTBZA1 was analyzed to clarify the molecular mechanism for PAEs biodegradation by USTBZA1. There were 3 and 41 genes encoding esterase/arylesterase and hydrolase, respectively, and two genes regions (pht34512 and pht4253) were responsible for the conversion of PA to protocatechuate (PCA), and two genes regions (ligCBAIKJ) were involved in PCA metabolism in USTBZA1. The resistance of sperm to freezing varies widely among boars. The semen ejaculate of different boars can be grouped into poor freezability ejaculate (PFE) and good freezability ejaculate (GFE). In this study, five Yorkshire boars each of the GFE and PFE were selected by comparing the changes in sperm motility before and after cryopreservation. Firstly, we found that the sperm plasma membrane of the PFE group showed weak integrity after PI and 6-CFDA staining. Then the electron microscopy results verified that the plasma membrane condition of all segments of GFE was better than that of PFE segments. Furthermore, the lipid composition of sperm plasma membranes in GPE and PFE sperm was analyzed by using mass spectrometry, and 15 lipids showed differences between the two groups. Among those lipids, only phosphatidylcholine (PC) (14:0/20:4) and phosphatidylethanolamine (PE) (14:0/20:4) were higher in PFE. The remaining lipid contents, including those of dihydroceramide (18:0/18:0), four hexosylceramides (18:1/20:1, 18:0/22:1, 18:1/16:0, 18:1/18:0), lactosylceramide (18:1/16:0), two hemolyzed phosphatidylethanolamines (18:2, 20:2), five phosphatidylcholines (16:1/18:2, 18:2/16:1, 14:0/20:4, 16:0/18:3, 18:1/20:2), and two phosphatidylethanolamines (14:0/20:4, 18:1/18:3), were all positively correlated with resistance to cryopreservation (p < 0.05, r > 0.6). Moreover, we analyzed the metabolic profile of sperm using untarget metabolomic. KEGG annotation analysis revealed that the altered metabolites were mainly involved in fatty acid biosynthesis. Finally, we determined that the contents of oleic acid, oleamideetc, N8-acetylspermidine etc., were different between GFE and PFE sperm. The present study assesses the rheological behaviors of TEMPO-oxidized bacterial cellulose (TOBCs) with diverse oxidation degrees to explore the potential relationship between the rheology and stability of the emulsions. The TOBCs with different oxidation degrees were prepared at the NaClO dosages of 2, 6, and 10 mmol/g, designated 2-TOBC, 6-TOBC, and 10-TOBC, respectively. The small amplitude dynamic rheology and diffusing wave spectroscopy results showed that 2-TOBC emulsion had higher storage modulus G' and loss modulus G" with lower tan δ than 6-TOBC emulsion and possessed the lowest solid-liquid balance among all of the TOBC Pickering emulsions, indicating the highest emulsion stability of 2-TOBC emulsion. Positive correlations existed between the viscoelasticity and stability of the TOBCs Pickering emulsions, with the coefficients of correlation over than 0.6. These results should improve understanding of the relationships between emulsions' stability and rheological characteristics, and lead to a potential application of TEMPO-oxidized bacterial cellulose as an emulsion stabilizer. This study investigated the effect of mercury (Hg) on progesterone secretion in ovarian granulosa cells of laying hens. The gene expressions of steroidogenic acute regulatory protein (StAR), cytochrome P450 cholesterol side‐chain cleavage (P450scc) and 3β‐hydroxysteroid dehydrogenase (3β‐HSD), cyclic adenosine monophosphate (cAMP)‐protein kinase A (PKA) pathway and intracellular calcium ion (Ca2+) were further investigated to uncover the molecular mechanism. Results revealed that the cell viability was gradually decreased after Hg exposure from 0 to 24 hr. Besides, progesterone secretion was significantly decreased (p < 0.05) as the concentration of Hg increased from 0 to 4 μM followed by a plateau in 6 μM Hg group at 12‐hr time point. Compared with 0 μM Hg group, 4 and 6 μM Hg for 48 hr had significantly decreased progesterone secretion (p < 0.05), while Hg exposure for 6 and 24 hr had no apparent effect on progesterone secretion. In addition, positive correlations occurred among intracellular progesterone, cAMP, PKA, mRNA expressions of StAR, P450scc and 3β‐HSD at 12‐h and 24‐h time points. On the contrary, intracellular Ca2+ level was negatively related to cAMP level at 6 time point and was negatively correlated with progesterone and PKA level at 48 time point. It could be concluded that Hg dose‐ and time‐dependently inhibited progesterone secretion by means of attenuating cAMP‐PKA signal pathway, gene expressions of StAR, P450scc and 3β‐HSD and enhancing intracellular Ca2+ in ovarian granulosa cells of laying hens. Whey is an important byproduct of the dairy industry containing mainly lactose and proteins. However, few studies have been conducted on co-utilization of lactose and proteins in whey for chemical production. To establish an efficient bioconversion method for the comprehensive utilization of cheese whey, a low-cost and labor-saving method for D-lactic acid production from cheese whey powder (CWP) was investigated and optimized in this study. CWP could be directly utilized by Lactobacillus bulgaricus CGMCC 1.6970 without the addition of extra nutrients. To fully utilize proteins in CWP, different proteases were selected to hydrolyze CWP, and the highest D-lactic acid concentration was obtained using neutral protease pretreated CPW as substrate. D-Lactic acid concentration could be further improved by adding a small amount of yeast extract. Based on the optimization, 70.70 g/L D-lactic acid with an average productivity of 1.47 g/L/h was obtained from batch fermentation. In fed-batch fermentation, a high D-lactic acid concentration (113.18 g/L) was obtained with an average productivity of 2.36 g/L/h. All the fermentations were carried out under an open condition without sterilization. This study presented a simple and economic strategy for CWP valorization and D-lactic acid production. Chinese spell checking is a task to detect and correct Chinese spelling errors, which is very important for natural language understanding. Generally, studies on Chinese spell checking are mainly based on n-gram language model and neural network models. However, the validity of the n-gram model needs to balance the value n and the storage resources, and most neural networks cannot efficiently handle the cases with severely uneven distribution of the correct and incorrect characters. This makes spell checking be limited in text application scenarios that contain many oral expressions. To solve the issues, a confusionset-guided decision network for spoken Chinese spell checking is proposed. By using confusionset to generate candidate set, the model can reasonably locate the wrong characters with decision network which ensures bidirectional long short-term memory pay more attention to the characteristics of the wrong characters. To verify the correctness and effectiveness of our model, extensive experiments were carried out on a logistics question and answer corpus and SIGHAN Bake-off dataset. Experimental results show that the model is efficient. It is much effective in spell checking for spoken Chinese, and it outperforms all competitor models. We investigated the genomic characteristics of 110 LX4 genotype strains of infectious bronchitis viruses (IBVs) isolated between 1995 and 2005 in China. The genome of these IBVs varies in size from 27596 bp to 27790 bp. Most IBV strains have the typical genomic organization of other gamacoronaviruses, however, two strains lacked 3a and 5b genes as a result of a nucleotide change within the start codon in the 3a or 5b genes. Analysis of our 110 viruses revealed that recombination events may be responsible for the emergence of the LX4 genotype with different topologies. Most of these viruses disappeared (before mid-2005) because they were not “fit” to adaptation in chickens. Finally, those of the “fit” viruses (after mid-2005) continued to evolve and have become widespread and predominant in commercial poultry. The main barriers to cost-effective lactic acid production from lignocellulose are the high cost of enzymes and the ineffective utilization of the xylose within the hydrolysate. In the present study, the thermophilic Bacillus coagulans strain CC17 was used for the simultaneous saccharification and fermentation (SSF) of bagasse sulfite pulp (BSP) to produce l -lactic acid. Unexpectedly, SSF by CC17 required approximately 33.33% less fungal cellulase than did separate hydrolysis and fermentation (SHF). More interestingly, CC17 can co-ferment cellobiose and xylose without any exogenous β-glucosidase in SSF. Moreover, adding xylanase could increase the concentration of lactic acid produced via SSF. Up to 110 g/L of l -lactic acid was obtained using fed-batch SSF, resulting in a lactic acid yield of 0.72 g/g cellulose. These results suggest that SSF using CC17 has a remarkable advantage over SHF and that a potentially low-cost and highly-efficient fermentation process can be established using this protocol. Crowdsourcing platforms are now extensively used for conducting subjective pairwise comparison studies. In this setting, a pairwise comparison dataset is typically gathered via random sampling, either with or without replacement. In this paper, we use tools from random graph theory to analyze these two random sampling methods for the HodgeRank estimator. Using the Fiedler value of the graph as a measurement for estimator stability (informativeness), we provide a new estimate of the Fiedler value for these two random graph models. In the asymptotic limit as the number of vertices tends to infinity, we prove the validity of the estimate. Based on our findings, for a small number of items to be compared, we recommend a two-stage sampling strategy where a greedy sampling method is used initially and random sampling without replacement is used in the second stage. When a large number of items is to be compared, we recommend random sampling with replacement as this is computationally inexpensive and trivially parallelizable. The initiation and progression of allergic asthma (AA) are associated with complex interactions between inflammation and immune response. Herein, we report the specific mechanisms underlying the molecular action of interferon (IFN)-γ in AA regulation. We speculated that IFN-γ inhibits Th9 differentiation by regulating the secretion of interleukin (IL)-27 from dendritic cells (DCs), thereby suppressing airway inflammation in asthma. We constructed a mouse model of ovalbumin-induced AA and overexpressed IFN-γ to evaluate the effect on the IL-27/Th9 axis via the in vitro effect of IFN-γ on IL-27 secretion by DCs and their influence on Th9 differentiation and asthmatic inflammation. IFN-γ overexpression reduced the proportion of Th9 cells and DCs and altered lung morphology and cytokine production in AA-induced mice, thus suppressing the AA phenotype. In addition, exogenous IFN-γ stimulation promoted the secretion of IL-27 and suppressed Th9 differentiation of CD4+ T cells via signal transducer and activator of transcription 1/3 (STAT1/3) signaling in a time-dependent manner. Simple Summary: Developing safe and effective antibiotic growth promoters (AGPs) substitutes is particularly important to improve animal health and production performance. As an essential plant oil, the oregano oil's main bioactive substance is carvacrol, which has been proven to have antioxidant, anti-inflammatory, antibacterial, and antiviral properties. The sanguinarine from macleaya cordata is the primary bioactive substance. Sanguinarine has anti-tumor, immune-enhancing, antibacterial, and anti-inflammatory effects. However, it has not been reported whether the compatibility of oregano oil and macleaya cordata extract could produce better results. This study is the first to report the effect of oregano oil combined with macleaya cordata oral solution on the growth of broilers. The oregano oil combined with macleaya cordata oral solution significantly improved the growth performance of broilers. At the same time, serum biochemical indices, serum antioxidant indices, serum immune indices, serum cytokines, and intestinal morphology were significantly improved. In summary, our results demonstrated that the mixed solution of oregano oil and macleaya cordata has substantial potential to be an alternative to AGPs for broilers to reduce costs and improve benefits. This study provides basic data and technical support for further research. In recent years, extracts of oregano oil and macleaya cordata have become a hot spot in the research and application of AGP substitutes for their safety and high efficiency. This study is the first to report the effect of oregano oil combined with macleaya cordata oral solution on broiler growth performance. A total of 960 one-day-old broiler chickens were randomly divided into four treatment groups (240 chickens per group). Each treatment group was divided into six replicate groups (40 birds per replicate group). There were four groups in this study: the solvent control group, the oregano essential oil combined with macleaya cordata extract oral solution group (OS group), the oregano essential oil oral solution group (OEO group), and the macleaya cordata extract oral solution group (MCE group). Two chickens from each replicate group were collected and mixed into a composite sample. Six composite samples were obtained for each treatment group. The results showed that the oregano oil combined with macleaya cordata oral solution significantly improved the growth performance of broiler chickens. At the same time, serum biochemical indices, serum antioxidant indices, serum immune indices, serum cytokines, and intestinal morphology were significantly improved by the OS group. Cyanobacterial hepatotoxins, including microcystins (MCs) and nodularins (NODs), are widely produced, distributed and extremely hazardous to human beings and the environment. However, the catalytic mechanism of microcystinase for biodegrading cyanobacterial hepatotoxins is not completely understood yet. The first microcystinase (MlrA) catalyzes the ring opening of cyclic hepatotoxins, while being further hydrolyzed by the third microcystinase (MlrC). Based on the homology modeling, we postulated that MlrC of Sphingopyxis sp. USTB-05 was a Zn2+-dependent metalloprotease including five active sites: Glu56, His150, Asp184, His186 and His208. Here, the active recombinant MlrC and five site-directed mutants were successfully obtained with heterologous expression and then purified for investigating the activity. The results indicated that the purified recombinant MlrC had high activity to catalyze linearized hepatotoxins.
