BackgroundThe app-based salt reduction intervention program in school children and their families (AppSalt) is a multicomponent mobile health (mHealth) intervention program, which involves multiple stakeholders, including students, parents, teachers, school heads, and local health and education authorities. The complexity of the AppSalt program highlights the need for process evaluation to investigate how the implementation will be achieved at different sites. ObjectiveThis paper presents a process evaluation protocol of the AppSalt program, which aims to monitor the implementation of the program, explain its causal mechanisms, and provide evidence for scaling up the program nationwide. MethodsA mixed methods approach will be used to collect data relating to five process evaluation dimensions: fidelity, dose delivered, dose received, reach, and context. Quantitative data, including app use logs, activity logs, and routine monitoring data, will be collected alongside the intervention process to evaluate the quantity and quality of intervention activities. The quantitative data will be summarized as medians, means, and proportions as appropriate. Qualitative data will be collected through semistructured interviews of purposely selected intervention participants and key stakeholders from local health and education authorities. The thematic analysis technique will be used for analyzing the qualitative data with the support of NVivo 12. The qualitative data will be triangulated with the quantitative data during the interpretation phase to explain the 5 process evaluation dimensions. ResultsThe intervention activities of the AppSalt program were initiated at 27 primary schools in three cities since October 2018. We have completed the 1-year intervention of this program. The quantitative data for this study, including app use log, activity logs, and the routine monitoring data, were collected and organized during the intervention process. After completing the intervention, we conducted semistructured interviews with 32 students, 32 parents, 9 teachers, 9 school heads, and 8 stakeholders from local health and education departments. Data analysis is currently underway. ConclusionsUsing mHealth technology for salt reduction among primary school students is an innovation in China. The findings of this study will help researchers understand the implementation of the AppSalt program and similar mHealth interventions in real-world settings. Furthermore, this process evaluation will be informative for other researchers and policy makers interested in replicating the AppSalt program and designing their salt reduction intervention. BackgroundSalt intake in China is over twice the maximum recommendation of the World Health Organization. Unlike most developed countries where salt intake is mainly derived from prepackaged foods, around 80% of the salt consumed in China is added during cooking. ObjectiveAction on Salt China (ASC), initiated in 2017, aims to develop, implement, and evaluate a comprehensive and tailored salt reduction program for national scaling-up. MethodsASC consists of six programs working in synergy to increase salt awareness and to reduce the amount of salt used during cooking at home and in restaurants, as well as in processed foods. Since September 2018, two health campaigns on health education and processed foods have respectively started, in parallel with four open-label cluster randomized controlled trials (RCTs) in six provinces across China: (1) app-based intervention study (AIS), in which a mobile app is used to achieve and sustain salt reduction in school children and their families; (2) home cook-based intervention study (HIS), in which family cooks receive support in using less salt; (3) restaurant-based intervention study (RIS) targeting restaurant consumers, cooks, and managers; and (4) comprehensive intervention study (CIS), which is a real-world implementation and evaluation of all available interventions in the three other RCTs. To explore the barriers, facilitators, and effectiveness of delivering a comprehensive salt reduction intervention, these RCTs will last for 1 year (stage 1), followed by nationwide implementation (stage 2). In AIS, HIS, and CIS, the primary outcome of salt reduction will be evaluated by 24-hour urinary sodium excretion in 6030 participants, including 5436 adults and 594 school children around 8-9 years old. In RIS, the salt content of meals will be measured by laboratory food analysis of the 5 best-selling dishes from 192 restaurants. Secondary outcomes will include process evaluation; changes in knowledge, attitude, and practice on salt intake; and economic evaluation. ResultsAll RCTs have been approved by Queen Mary Research Ethics Committee and the Institutional Review Boards of leading institutes in China. The research started in June 2017 and is expected to be completed around March 2021. The baseline investigations of the four RCTs were completed in May 2019. ConclusionsThe ASC project is progressing smoothly. The intervention packages and tailored components will be promoted for salt reduction in China, and could be adopted by other countries. Abstract Background Maternal diabetes mellitus can influence the development of offspring. Gestational diabetes mellitus (GDM) creates a short-term intrauterine hyperglycaemic environment in offspring, leading to glucose intolerance in later life, but the long-term effects and specific mechanism involved in skeletal muscle dysfunction in offspring remain to be clarified. Methods Pregnant mice were divided into two groups: The GDM group was intraperitoneally injected with 100 mg/kg streptozotocin on gestational days (GDs) 6.5 and 12.5, while the control (CTR) group was treated with vehicle buffer. Only pregnant mice whose random blood glucose level was higher than 16.8 mmol/L beginning on GD13.5 were regarded as the GDM group. The growth of the offspring was monitored, and the glucose tolerance test was performed at different time points. Body composition analysis and immunohistochemical methods were used to evaluate the development of lean mass at 8 weeks. The exercise capacity and grip strength of the male mouse offspring were assessed at the same period. Transmission electron microscopy was used to observe the morphology inside skeletal muscle at 8 weeks and as a foetus. The genes and proteins associated with mitochondrial biogenesis and oxidative metabolism were investigated. We also coanalyzed RNA sequencing and proteomics data to explore the underlying mechanism. Chromatin immunoprecipitation and bisulfite-converted DNA methylation detection were performed to evaluate this phenomenon. Results Short-term intrauterine hyperglycaemia inhibited the growth and reduced the lean mass of male offspring, leading to decreased endurance exercise capacity. The myofiber composition of the tibialis anterior muscle of GDM male offspring became more glycolytic and less oxidative. The morphology and function of mitochondria in the skeletal muscle of GDM male offspring were destroyed, and coanalysis of RNA sequencing and proteomics of foetal skeletal muscle showed that mitochondrial elements and lipid oxidation were consistently impaired. In vivo and in vitro myoblast experiments also demonstrated that high glucose concentrations impeded mitochondrial organisation and function. Importantly, the transcription of genes associated with mitochondrial biogenesis and oxidative metabolism decreased at 8 weeks and during the foetal period. We predicted Ppargc1α as a key upstream regulator with the help of IPA software. The proteins and mRNA levels of Ppargc1α in the skeletal muscle of GDM male offspring were decreased as a foetus (CTR vs. GDM, 1.004 vs. 0.665, p = 0.002), at 6 weeks (1.018 vs. 0.511, p = 0.023) and 8 weeks (1.006 vs. 0.596, p = 0.018). In addition, CREB phosphorylation was inhibited in GDM group, with fewer activated pCREB proteins binding to the CRE element of Ppargc1α (1.042 vs. 0.681, p = 0.037), Pck1 (1.091 vs. 0.432, p = 0.014) and G6pc (1.118 vs. 0.472, p = 0.027), resulting in their decreased transcription. Interestingly, we found that sarcopenia and mitochondrial dysfunction could even be inherited by the next generation. Conclusions Short-term intrauterine hyperglycaemia significantly reduced lean mass in male offspring at 8 weeks, resulting in decreased exercise endurance and metabolic disorders. Disrupted organisation and function of the mitochondria in skeletal muscle were also observed among them. Foetal exposure to hyperglycaemia decreased the ratio of phosphorylated CREB and reduced the transcription of Ppargc1α, which inhibited the transcription of downstream genes involving in mitochondrial biogenesis and oxidative metabolism. Abnormal mitochondria, which might be transmitted through aberrant gametes, were also observed in the F2 generation.

Abstract Background In recent years, with benefits from the continuous improvement of clinical technology and the advantage of fertility preservation, the application of embryo cryopreservation has been growing rapidly worldwide. However, amidst this growth, concerns about its safety persist. Numerous studies have highlighted the elevated risk of perinatal complications linked to frozen embryo transfer (FET), such as large for gestational age (LGA) and hypertensive disorders during pregnancy. Thus, it is imperative to explore the potential risk of embryo cryopreservation and its related mechanisms. Methods Given the strict ethical constraints on clinical samples, we employed mouse models in this study. Three experimental groups were established: the naturally conceived (NC) group, the fresh embryo transfer (Fresh-ET) group, and the FET group. Blastocyst formation rates and implantation rates were calculated post-embryo cryopreservation. The impact of FET on fetal growth was evaluated upon fetal and placental weight. Placental RNA-seq was conducted, encompassing comprehensive analyses of various comparisons (Fresh-ET vs. NC, FET vs. NC, and FET vs. Fresh-ET). Results Reduced rates of blastocyst formation and implantation were observed post-embryo cryopreservation. Fresh-ET resulted in a significant decrease in fetal weight compared to NC group, whereas FET reversed this decline. RNA-seq analysis indicated that the majority of the expression changes in FET were inherited from Fresh-ET, and alterations solely attributed to embryo cryopreservation were moderate. Unexpectedly, certain genes that showed alterations in Fresh-ET tended to be restored in FET. Further analysis suggested that this regression may underlie the improvement of fetal growth restriction in FET. The expression of imprinted genes was disrupted in both FET and Fresh-ET groups. Conclusion Based on our experimental data on mouse models, the impact of embryo cryopreservation is less pronounced than other in vitro manipulations in Fresh-ET. However, the impairment of the embryonic developmental potential and the gene alterations in placenta still suggested it to be a risky operation.

ObjectiveThe identification of agricultural activities plays a crucial role for greenhouse vegetables production, particularly in the precise management of cucumber cultivation. By monitoring and analyzing the timing and procedures of agricultural operations, effective guidance can be provided for agricultural production, leading to increased crop yield and quality. However, in practical applications, the recognition of agricultural activities in cucumber cultivation faces significant challenges. The complex and ever-changing growing environment of cucumbers, including dense foliage and internal facility structures that may obstruct visibility, poses difficulties in recognizing agricultural activities. Additionally, agricultural tasks involve various stages such as planting, irrigation, fertilization, and pruning, each with specific operational intricacies and skill requirements. This requires the recognition system to accurately capture the characteristics of various complex movements to ensure the accuracy and reliability of the entire recognition process. To address the complex challenges, an innovative algorithm: SlowFast-SMC-ECA (SlowFast-Spatio-Temporal Excitation, Channel Excitation, Motion Excitation-Efficient Channel Attention) was proposed for the recognition of agricultural activity behaviors in cucumber cultivation within facilities.MethodsThis algorithm represents a significant enhancement to the traditional SlowFast model, with the goal of more accurately capturing hand motion features and crucial dynamic information in agricultural activities. The fundamental concept of the SlowFast model involved processing video streams through two distinct pathways: the Slow Pathway concentrated on capturing spatial detail information, while the Fast Pathway emphasized capturing temporal changes in rapid movements. To further improve information exchange between the Slow and Fast pathways, lateral connections were incorporated at each stage. Building upon this foundation, the study introduced innovative enhancements to both pathways, improving the overall performance of the model. In the Fast Pathway, a multi-path residual network (SMC) concept was introduced, incorporating convolutional layers between different channels to strengthen temporal interconnectivity. This design enabled the algorithm to sensitively detect subtle temporal variations in rapid movements, thereby enhancing the recognition capability for swift agricultural actions. Meanwhile, in the Slow Pathway, the traditional residual block was replaced with the ECA-Res structure, integrating an effective channel attention mechanism (ECA) to improve the model's capacity to capture channel information. The adaptive adjustment of channel weights by the ECA-Res structure enriched feature expression and differentiation, enhancing the model's understanding and grasp of key spatial information in agricultural activities. Furthermore, to address the challenge of class imbalance in practical scenarios, a balanced loss function (Smoothing Loss) was developed. By introducing regularization coefficients, this loss function could automatically adjust the weights of different categories during training, effectively mitigating the impact of class imbalance and ensuring improved recognition performance across all categories.Results and DiscussionsThe experimental results significantly demonstrated the outstanding performance of the improved SlowFast-SMC-ECA model on a specially constructed agricultural activity dataset. Specifically, the model achieved an average recognition accuracy of 80.47%, representing an improvement of approximately 3.5% compared to the original SlowFast model. This achievement highlighted the effectiveness of the proposed improvements. Further ablation studies revealed that replacing traditional residual blocks with the multi-path residual network (SMC) and ECA-Res structures in the second and third stages of the SlowFast model leads to superior results. This highlighted that the improvements made to the Fast Pathway and Slow Pathway played a crucial role in enhancing the model's ability to capture details of agricultural activities. Additional ablation studies also confirmed the significant impact of these two improvements on improving the accuracy of agricultural activity recognition. Compared to existing algorithms, the improved SlowFast-SMC-ECA model exhibited a clear advantage in prediction accuracy. This not only validated the potential application of the proposed model in agricultural activity recognition but also provided strong technical support for the advancement of precision agriculture technology. In conclusion, through careful refinement and optimization of the SlowFast model, it was successfully enhanced the model's recognition capabilities in complex agricultural scenarios, contributing valuable technological advancements to precision management in greenhouse cucumber cultivation.ConclusionsBy introducing advanced recognition technologies and intelligent algorithms, this study enhances the accuracy and efficiency of monitoring agricultural activities, assists farmers and agricultural experts in managing and guiding the operational processes within planting facilities more efficiently. Moreover, the research outcomes are of immense value in improving the traceability system for agricultural product quality and safety, ensuring the reliability and transparency of agricultural product quality.

To improve the plasticization quality and process safety of Modified Double-Base (MDB) propellants during the calendering process. This study, using actual physical parameters of MDB propellant, a three-dimensional thermo-fluid coupled model was developed to investigate the effects of process parameters on the heat and mass transfer behavior of MDB materials during the calendering process. By comparing the effects of different roller speed ratios, roller gaps, and roller speeds on the material's velocity, temperature, and viscosity distribution during the calendering process, it was found that the material forms an asymmetric vortex reflux movement during the calendering process. This unique flow characteristic is the primary reason for the material's asymmetric mass and heat transfer properties. Changes in the rotational speed ratio have a minor impact on the reflux movement but significantly affect process safety. A rotational speed ratio of 1.25 can effectively reduce the temperature of the material at the vortex center, thereby decreasing the risk of combustion. The smaller the roll gap, the higher the starting position of the material reflux. By selecting a roll gap of 0.8 mm, it's possible to effectively reduce the viscosity gradient while simultaneously decreasing the material's residence time. Increasing the rotation speed enhances the intensity of reflux. A speed setting of 20:15 not only reduces heat generated from shearing but also controls the temperature distribution range of the material, thereby enhancing the efficiency of the calendering production process. These adjustments in process parameters enhanced the safety and production efficiency of the MDB propellant calendering process, providing valuable insights for the safe production of MDB propellant calendering technology.

The wettability of nanofluid is a key factor affecting oil recovery and fluid heat transfer. However, the traditional wettability measurement method consisting in determining the contact angle enables to assess the wettability at a solid–liquid contact point only and not within the continuous detection range. Therefore, in this work, a lateral friction detection system is used to continuously measure the wettability of hydroxylated multi-walled carbon nanotubes/deionized water (MWCNT-OH/DIW) nanofluids on polydimethylsiloxane surfaces. Specifically, MWCNT-OH/DIW nanofluids with various mass fractions are configured, and different surfactants are introduced to regulate the dispersion in deionized water. The results reveal that the surface tension and contact angle decrease simultaneously with the increase in MWCNT-OH/DIW mass fraction. Meanwhile, this change also causes an increase in the lateral friction at the nanofluid/PDMS interface. Furthermore, both sodium dodecyl sulfate and cetyltrimethylammonium bromide (CTAB) as surfactants can effectively reduce its surface tension. Notably, when CTAB is added, the maximum static friction (132.62 μN) exceeds that of SDS (113.13 μN). Therefore, the proposed method opens up new prospects in wettability detection of nanofluids.

ABSTRACT Background Marfan syndrome (MFS) is a complex genetic systemic connective tissue disorder. It is well known that genetic factors play a critical role in the progression of MFS, with nearly all cases attributed to variants in the FBN1 gene. Methods We investigated a Chinese family with MFS spanning two generations. Whole exome sequencing, in silico analysis, minigene constructs, transfection, RT‐PCR, and protein secondary structure analysis were used to analyze the genotype of the proband and his father. Results The main clinical manifestations of the proband and his father were subluxation of the left lens and high myopia with pectus deformity. Whole exome sequencing identified a novel single nucleotide variant (SNV) in the FBN1 gene at a non‐canonical splice site, c.443‐3C>G. This variant resulted in two abnormal mRNA transcripts, leading to a frameshift and an in‐frame insertion. Further in vitro experiments indicated that the c.443‐3C>G variant in FBN1 was pathogenic and functionally harmful. Conclusion This research identified a novel intronic pathogenic FBN1: c.443‐3C>G gene variant, which led to two different aberrant splicing effects. Further functional analysis expands the variant spectrum and provides a strong indication and sufficient basis for preimplantation genetic testing for monogenic disease (PGT‐M).

In this study, headspace solid phase microextraction-gas chromatography-mass spectrometry (HS-SPME-GC-MS) and gas chromatography-olfactory (GC-O) were applied to identify and analyze the volatile aroma compounds of the ethanol extract of figs. Its key characteristic flavor compounds were analyzed by odor activity (OAV) and aroma recombination and omission tests. The results showed that a total of 40 volatile aroma components were identified, of which 18 compounds, such as isobutyrate, γ-butyl lactone, aromatic camphor, nonaldehyde, vanillin and furfural, were important aroma components in the fig extract (OAV > 1). The aroma recombination experiments showed that the sensory properties of the recombined samples, which had typical aroma characteristics such as fruity, sweet, baked and burnt sweet with slight sour, ointment-like and milky, were similar to those of the fig extract. Furthermore, the aroma omission experiments identified gamma-hexanoate, ethyl palmitate, phenmethanol, aromatic camphor, vanillin, benzaldehyde, 4-hydroxy-2,5-dimethyl-3 (2H)-furan, 5-hydroxymyfuran, and methyl cyclopentanolone as key characteristic flavor compounds of the fig extract. The findings of this study provide a theoretical basis for the development and quality control of fig characteristic flavorings.

BackgroundDietary sodium and potassium are involved in the pathogenesis of cardiovascular disease. Data exploring the cardiovascular outcomes associated with these electrolytes within Australian children is sparse. Furthermore, an objective measure of sodium and potassium intake within this group is lacking. ObjectiveThe primary aim of the Salt and Other Nutrient Intakes in Children (“SONIC”) study was to measure sodium and potassium intakes in a sample of primary schoolchildren located in Victoria, Australia, using 24-hour urine collections. Secondary aims were to identify the dietary sources of sodium and potassium, examine the association between these electrolytes and cardiovascular risk factors, and assess children’s taste preferences and saltiness perception of manufactured foods. MethodsA cross-sectional study was conducted in a convenience sample of schoolchildren attending primary schools in Victoria, Australia. Participants completed one 24-hour urine collection, which was analyzed for sodium, potassium, and creatinine. Completeness of collections was assessed using collection time, total volume, and urinary creatinine. One 24-hour dietary recall was completed to assess dietary intake. Other data collected included blood pressure, body weight, height, waist and hip circumference. Children were also presented with high and low sodium variants of food products and asked to discriminate salt level and choose their preferred variant. Parents provided demographic information and information on use of discretionary salt. Descriptive statistics will be used to describe sodium and potassium intakes. Linear and logistic regression models with clustered robust standard errors will be used to assess the association between electrolyte intake and health outcomes (blood pressure and body mass index/BMI z-score and waist circumference) and to assess differences in taste preference and discrimination between high and low sodium foods, and correlations between preference, sodium intake, and covariates. ResultsA total of 780 children across 43 schools participated. The results from this study are expected at the end of 2015. ConclusionsThis study will provide the first objective measure of sodium and potassium intake in Australian schoolchildren and improve our understanding of the relationship of these electrolytes to cardiovascular risk factors. Furthermore, this study will provide insight into child taste preferences and explore related factors. Given the cardiovascular implications of consuming too much sodium and too little potassium, monitoring of these nutrients during childhood is an important public health initiative.
