The world has undergone a remarkable transformation from the era of famines to an age of global food production that caters to an exponentially growing population. This transformation has been made possible by significant agricultural revolutions, marked by the intensification of agriculture through the infusion of mechanical, industrial, and economic inputs. However, this rapid advancement in agriculture has also brought about the proliferation of agricultural inputs such as pesticides, fertilizers, and irrigation, which have given rise to long-term environmental crises. Over the past two decades, we have witnessed a concerning plateau in crop production, the loss of arable land, and dramatic shifts in climatic conditions. These challenges have underscored the urgent need to protect our global commons, particularly the environment, through a participatory approach that involves countries worldwide, regardless of their developmental status. To achieve the goal of sustainability in agriculture, it is imperative to adopt multidisciplinary approaches that integrate fields such as biology, engineering, chemistry, economics, and community development. One noteworthy initiative in this regard is Zero Budget Natural Farming, which highlights the significance of leveraging the synergistic effects of both plant and animal products to enhance crop establishment, build soil fertility, and promote the proliferation of beneficial microorganisms. The ultimate aim is to create self-sustainable agro-ecosystems. This review advocates for the incorporation of biotechnological tools in natural farming to expedite the dynamism of such systems in an eco-friendly manner. By harnessing the power of biotechnology, we can increase the productivity of agro-ecology and generate abundant supplies of food, feed, fiber, and nutraceuticals to meet the needs of our ever-expanding global population. Traditional breeding programmes have been limited to the selection and introduction of genotypes with promising agronomic characteristics, but studies focused on genetic diversity in the low hills of the North West(NW) Himalayas have not been conducted, which is very important for the identification of potential parents for breeding programmes in guava. In the present study, six best performing guava cultivars/hybrids were characterised using morphological descriptors and SCoT markers. Broad phenotypic variability among the guava cultivars/hybrids was detected using morphological descriptors. A set of 36 SCoT markers were used for polymorphism, out of which 31(86.1%) markers showed polymorphism, indicating high genetic variability in the guava cultivars/hybrids. During the analysis, 291 polymorphic amplicons were obtained, ranging from four to 19, with an average of 9.4 amplicons per primer and average polymorphic information content of 0.47.The UPGMA classified hybrids and cultivars into two groups. Based on morphological and molecular performance, L-49(Sardar) was ranked as the most promising cultivar for the low hills of NW Himalayan conditions. Morphological descriptors along with SCoT markers proved efficient in detecting the levels of genetic variability among the collections maintained in the field. Waste rice straw biomass and its burning in open fields have become a serious issue of greenhouse gases emission and air pollution, which has a negative impact on public health and the environment. However, the environmental impact of burning this agro-waste can be mitigated by diverting it towards green biorefinery through the sustainable production of energy, biofuels, organic chemicals, and building blocks for various polymers. This will not only help to reduce the reliance on limited fuels and various chemicals derived from petroleum, but also help in the restoration of the environment in a sustainable manner through its complete utilization. To maximize the inherent conversion potential of rice straw biomass into valuable products, this agriculture waste biomass requires a comprehensive analysis and a techno-economic review for its sustainable management. This review article focuses on the sustainable management of rice straw waste biomass via innovative valorization approaches, as well as the opportunities and challenges encountered in this sector for meeting the demand of current and future green biorefineries. Continuous urbanization and modernization have increased the burning of fossil fuels to meet energy needs across the globe, emanating environmental pollution and depleting fossil fuels. Therefore, a shift towards sustainable and renewable energy is necessary. Several techniques to exploit biomass to yield energy are trending, with pyrolysis one of them. Usually, a single feedstock is employed in pyrolysis for anoxygenic generation of biochar together with bio-oil at elevated temperatures (350–600 °C). Bio-oil produced through pyrolysis can be upgraded to crude oil after some modification. However, these modifications of bio-oil are one of the major drawbacks for its large-scale adoption, as upgradation increases the overall cost. Therefore, in recent years the scientific community has been researching co-pyrolysis technology that involves the pyrolysis of lignocellulosic biomass waste with non-biodegradable waste. Co-pyrolysis reduces the need for post-modification of bio-oil, unlike pyrolysis of a single feedstock. This review article discusses the recent advancements and technological challenges in waste biomass co-pyrolysis, the mechanism of co-pyrolysis, and factors that affect co-pyrolysis. The current study critically analyzes different recent research articles presented in databases such as PubMed, MDPI, ScienceDirect, Springer, etc. Hence, this review is one-of-a-kind in that it attempts to explain each and every aspect of the co-pyrolysis process and its current progress in the scientific field. Alkaline protease has tremendous applications in different industries, and to fulfil industrial and commercial enzyme requirements, overproduction and optimization of production medium are prerequisites. Protease production by Bacillus amyloliquefaciens SP1, isolated from apple tree rhizosphere was studied. Plackett Burmann design and response surface methodology was used to optimize medium for alkaline protease production. Higher levels of protease activity were observed in presence of disaccharides owing to diauxic growth and inorganic nitrogen sources proved to be less favourable. After screening through one-factor-at-a time experiments and Plackett Burmann design, casein, yeast extract, maltose and KH2PO4 were identified as the most significant variables. Statistical design based three-dimensional (3-D) and contour plots were generated to understand the relationship between enzyme activity and medium variables. The protease production was found to increase from 1730 to 3630 µg/ml/min in optimized medium i.e. approx. 2.1 fold increased compared with original medium. The study assumes significance in the ability of bacterium to survive in wide variety of salts and agricultural wastes and yield optimum levels of extracellular protease. [ABSTRACT FROM AUTHOR]

Alkaline protease from Bacillus amyloliquefaciens SP1 has been characterized in detail for its ecofriendly application of release of silver particles from gelatin layers of used X-ray films. It exhibited optimum activity at broad temperature range and maximum at 60°C under alkaline pH environment (8-12). Thermal inactivation of the crude enzyme followed first order kinetics. The half-life of the enzyme at 50, 60 and 65°C was 70, 15 and 12.6 min, respectively and the denaturation energy was 114.87 kJ/mol. Enzyme retained 53.83 and 108.33% of its initial activity after heating for 15 min at pH 8.0 and temperature 60°C, in presence and absence of 10 mM MnSO4, respectively. Enzymatic decomposition of gelatin layers was enhanced by increase of enzyme concentration from 38 to 3630 µg/ml/min, at 60°C and pH 8.0. This study reported the shortest time of 1.30 min at 3630 µg/ml/min and 4 : 30 min at 74 µg/ml/min of enzyme concentration for hydrolysis of gelatin layers. Keeping in mind that, nowadays recycling is needed and imperative, this is the first study to report that after addition of Mn2+ ions, thermal stability of enzyme increased and it could be effectively reused for 8 cycles as compared with enzyme without protective agents which also increase its yield of silver recovery i.e. 19.56 ± 0.78% by eight times. An alkaline protease gene was amplified from genomic DNA of Bacillus amyloliquefaciens SP1 which was involved in effective biocontrol of Fusarium oxysporum. We investigated the antagonistic capacity of protease of B. amyloliquifaciens SP1, under in vitro conditions. The 5.62 fold purified enzyme with specific activity of 607.69 U/mg reported 24.14% growth inhibition of F. oxysporum . However, no antagonistic activity was found after addition of protease inhibitor i.e. PMSF (15 mM) to purified enzyme. An 1149 bp nucleotide sequence of protease gene encoded 382 amino acids of 43 kDa and calculated isoelectric point of 9.29. Analysis of deduced amino acid sequence revealed high homology (86%) with subtilisin E of Bacillus subtilis . The B. amyloliquefaciens SP1 protease gene was expressed in Escherichiax coli BL21. The expressed protease was secreted into culture medium by E. coli and exhibited optimum activity at pH 8.0 and 60 °C. The most reliable three dimensional structure of alkaline protease was determined using Phyre 2 server which was validated on the basis of Ramachandran plot and ERRAT value. A thermostable extracellular alkaline protease producing Bacillus amyloliquefaciens SP1 was isolated from apple rhizosphere having multifarious plant growth promoting activities. Strain SP1 was purified to 6.48-fold using four-step purification protocol and characterized in detail for its robustness and ecofriendly application in leather and detergent industries. Structural analysis revealed that the protease was monomeric and had a molecular weight of 43 kDa. It exhibited optimum activity at 60°C in alkaline environment (pH 8.0) and stable in the presence of surfactants and oxidizing agents. Enzyme was thermostable at 50°C and retained more than 70% activity after 30 min incubation. It has shown stain removal property and dehairing of goat skin without chemical assistance and hydrolyzing fibrous proteins. This protease showed Km of 0.125 mg ml−1 and Vmax of 12820 μg ml−1 indicating its excellent affinity and catalytic role. Thermal inactivation of the pure enzyme followed first-order kinetics. The half life of the pure enzyme at 50, 60, and 65°C was 77, 19.80, and 13.33 min, respectively. The activation energy was 37.19 KJ mol−1. A total of 206 phosphate solubilizing bacteria (PSBs) were isolated from rhizosphere soil (RS) (116) and endorhizosphere (90) of apple trees of four locations: Chamba, Shimla, Kinnaur and Kullu of Himachal Pradesh, India. The population count of PSBs was invariably higher in rhizosphere than in root endosphere (ER) of apple. The greatest Shannon diversity indices for PSBs from RS (1.77) and ER (1.69) were obtained from Kinnaur. Polymorphism among 85 PSBs was detected using PCR-RAPD analysis. Alkalophilic Cellulosimicrobium cellulans CKMX1 isolated from mushroom compost is an actinomycete that produces industrially important and environmentally safer thermostable cellulase-free xylanase, which is used in the pulp and paper industry as an alternative to the use of toxic chlorinated compounds. Strain CKMX1 was previously characterized by metabolic fingerprinting, whole-cell fatty acids methyl ester analysis, and 16S rDNA and was found to be C. cellulans CKMX1. Crude enzyme (1027.65 U/g DBP) produced by C. cellulans CKMX1, having pH and temperature optima of 8.0 and 60 °C, respectively, in solid state fermentation of apple pomace, was used in the production of bleached wheat straw pulp. Pretreatment with xylanase at a dose of 5 U/g after pulping decreased pulp kappa points by 1.4 as compared with the control. Prebleaching with a xylanase dose of 5 U/g pulp reduced the chlorine charge by 12.5%, increased the final brightness points by approximately 1.42% ISO, and improved the pulp strength properties. Xylanase could be substituted for alkali extraction in C–Ep–D sequence and used for treating chemically bleached pulp, resulting in bleached pulp with higher strength properties. Modification of bleached pulp with 5 U of enzyme/g increased pulp whiteness and breaking length by 1.03% and 60 m, respectively; decreased tear factor of pulp by 7.29%; increased bulk weight by 3.99%, as compared with the original pulp. Reducing sugars and UV-absorbing lignin-derived compound values were considerably higher in xylanase-treated samples. P-solubilizing bacterial isolate CB7 isolated from apple rhizosphere soil of Himachal Pradesh, India was identified as Bacillus circulans on the basis of phenotypic characteristics, biochemical tests, fatty acid methyl esters analysis, and 16S rRNA gene sequence. The isolate exhibited plant growth-promoting traits of P-solubilization, auxin, 1-aminocyclopropane-1-carboxylate deaminase activity, siderophore, nitrogenase activity, and antagonistic activity against Dematophora necatrix. In vitro studies revealed that P-solubilization and other plant growth-promoting traits were dependent on the presence of glucose in PVK medium and removal of yeast extract had no significant effect on plant growth-promoting traits. Plant growth-promoting traits of isolate CB7 were repressed in the presence of KH2PO4. P-solubilization activity was associated with the release of organic acids and a drop in the pH of the Pikovskaya's medium. HPLC analysis detected gluconic and citric acid as major organic acids in the course of P-solubilization. Remarkable increase was observed in seed germination (22.32%), shoot length (15.91%), root length (25.10%), shoot dry weight (52.92%) and root dry weight (31.4%), nitrogen (18.75%), potassium (57.69%), and phosphorus (22.22%) content of shoot biomass over control. Vegetables are important component of balanced human diet. Advanced production technologies are being followed to measure productivity and quality of produce. Protected cultivation is an improved agro technique being used worldwide to register 3-4 times increase in production. Tomato is grown extensively in the plastic greenhouses for higher productivity. The growing conditions especially light and temperature are known to influence both composition and quality of tomato fruits. Hence, studies have been conducted to estimate the quality parameters of tomato grown both under open and in polyhouse with shade nets during cloudy weather in rainy season The results reveals that the fruits harvested from the field had higher TSS:TA ratio (11.73), acidity (0.49%), total sugar (2.5%), ascorbic acid (14.7 mg 100g-1), TSS (5.76°B) and the lycopene content (8.7mg 100 g-1) than the fruits grown under protected conditions. However, the environment in the polyhouse favour the growth and development of tomato plant through increased plant height (1.4m) and number of branches. The fruit yield obtained from the polyhouse was higher (2.6 kg plant-1) than the open field (1.5 kg plant-1). Hence, both types of environmental conditions are favourable in some aspects. The parameters of economic importance like fruit weight and yield are better under protected conditions but the nutritional parameters like vitamin C and lycopene content are higher in open conditions during rainy season. Due to rapid urbanization and industrialization, the population density of the world is intense in developing countries. This overgrowing population has resulted in the production of huge amounts of waste/refused water due to various anthropogenic activities. Household, municipal corporations (MC), urban local bodies (ULBs), and industries produce a huge amount of waste water, which is discharged into nearby water bodies and streams/rivers without proper treatment, resulting in water pollution. This mismanaged treatment of wastewater leads to various challenges like loss of energy to treat the wastewater and scarcity of fresh water, beside various water born infections. However, all these major issues can provide solutions to each other. Most of the wastewater generated by ULBs and industries is rich in various biopolymers like starch, lactose, glucose lignocellulose, protein, lipids, fats, and minerals, etc. These biopolymers can be converted into sustainable biofuels, i.e., ethanol, butanol, biodiesel, biogas, hydrogen, methane, biohythane, etc., through its bioremediation followed by dark fermentation (DF) and anaerobic digestion (AD). The key challenge is to plan strategies in such a way that they not only help in the treatment of wastewater, but also produce some valuable energy driven products from it. This review will deal with various strategies being used in the treatment of wastewater as well as for production of some valuable energy products from it to tackle the upcoming future demands and challenges of fresh water and energy crisis, along with sustainable development. Aneurinibacillus aneurinilyticus strain CKMV1 was isolated from rhizosphere of Valeriana jatamansi and possessed multiple plant growth promoting traits like production of phosphate solubilization (260mg/L), nitrogen fixation (202.91nmolethylenemL -1 h -1 ), indole-3-acetic acid (IAA) (8.1μg/mL), siderophores (61.60%), HCN (hydrogen cyanide) production and antifungal activity. We investigated the ability of isolate CKMV1 to solubilize insoluble P via mechanism of organic acid production. High-performance liquid chromatography (HPLC) study showed that isolate CKMV1 produced mainly gluconic (1.34%) and oxalic acids. However, genetic evidences for nitrogen fixation and phosphate solubilization by organic acid production have been reported first time for A. aneurinilyticus strain CKMV1. A unique combination of glucose dehydrogenase (gdh) gene and pyrroloquinoline quinone synthase (pqq) gene, a cofactor of gdh involved in phosphate solubilization has been elucidated. Nitrogenase (nif H) gene for nitrogen fixation was reported from A. aneurinilyticus. It was notable that isolate CKMV1 exhibited highest antifungal against Sclerotium rolfsii (93.58%) followed by Fusarium oxysporum (64.3%), Dematophora necatrix (52.71%), Rhizoctonia solani (91.58%), Alternaria sp. (71.08%) and Phytophthora sp. (71.37%). Remarkable increase was observed in seed germination (27.07%), shoot length (42.33%), root length (52.6%), shoot dry weight (62.01%) and root dry weight (45.7%) along with NPK (0.74, 0.36, 1.82%) content of tomato under net house condition.
