The Alternative splicing (AS) of Carnitine palmitoyltransferase 1a (CPT1a) and their expression profiles had never been illuminated in goats until now. Herein, a novel splice transcript in the CPT1a gene that is predicted to result in the skipping of exons 6–19 (CPT1a-sv1) has been isolated in addition to the full-length transcript in goats. The result of RT-PCR showed that CPT1a-sv1 is 606 bp in length and consists of 6 exons. A novel exon 6 was consisted of partial exon 5 and partial exon 19, compared to that in CPT1a. RT-qPCR analysis showed that the expression patterns of CPT1a and CPT1a-sv1 are spatially different. In both kid and adult goats, the CPT1a transcript is strongly expressed in the liver, spleen, lung, kidney, and brain tissues. However, CPT1a-sv1 has a strong tissue-specific expression pattern, with moderate RNA levels in the liver and brain of kids, while highly expressed in the liver and minimally expressed in the brain of adults. We observed two transcripts to be involved in brain development. Recent evidence suggested that long non-coding RNAs (lncRNAs) and microRNAs (miRNAs) play critical roles in the pathogenesis of vascular calcification (VC). In this study, we tried to explore the expression and role of a lncRNA, i.e., metastasis-associated lung adenocarcinoma transcript 1 (MALAT1), and a miRNA, i.e., miR-30c, in VC. In vitro VC model was induced in human vascular smooth muscle cells (VSMCs) after 10 days culture in calcifying medium containing 2 mM Na2HPO4. Alizarin red S staining, calcium assay and western blot analysis of runt-related transcription factor 2 (Runx2) and alpha smooth muscle actin (α-SMA) were performed to evaluate VC. Knockdown of MALAT1 and up-regulation of MALAT1, miR-30c and Runx2 was performed to determine the impact of these molecules on VSMCs calcification. Dual-luciferase report assay was performed to confirm the relationship between MALAT1 and miR-30c or miR-30c and Runx2. In addition, quantitative reverse transcription PCR and western blot were used to determine gene and protein expression. MALAT1 was increased, while miR-30c was decreased in calcified VSMCs. Knockdown of MALAT1 suppressed VSMCs calcification; on the contrary, up-regulation of MALAT1 promoted VSMCs calcification. The effect of MALAT1 over-expression on VSMCs calcification was reversed by upregulation of miR-30c, which was reversed again by upregulation of Runx2. Dual-luciferase report assay confirmed that there is a direct interaction between MALAT1 and miR-30c, and Runx2 is a direct target of miR-30c. Patients who suffer from dyspnea while dying from COVID-19 are treated with opioids and benzodiazepines. In some instances, patients may experience refractory dyspnea at the end of life. Palliative sedation can be prescribed to alleviate such patients' suffering. We describe two patients being treated for severe COVID-19 pneumonia in a tertiary hospital. Both developed intractable dyspneic crises despite high-dose opioids and benzodiazepines. This led to their requirement of palliative sedation in the general ward using subcutaneous phenobarbitone (phenobarbital). We outline clinical considerations for the use of palliative sedation in COVID-19 related dyspnea. In particular, we discuss the evidence for, benefits and limitations of using phenobarbitone for palliative sedation in COVID-19 patients. [ABSTRACT FROM AUTHOR]

B7 family members and their receptors have been confirmed to participate in T cell response signaling through the induction of immunosuppressive effects. The V-domain Ig suppressor of T-cell activation (VISTA) is expressed in myeloid cells as well as T lymphocytes and plays a crucial role in immune responses. In this study, the phage display technique was employed to isolate specific antibodies targeting VISTA in chickens. In in vitro experiments, VISTA could trigger the inhibition of T-cell activation through interaction during binding. However, isolated single-chain variable fragments (scFvs) effectively inhibited immune cells from producing exhaustion responses, freeing the T cells in peripheral blood mononuclear cells from inhibition and allowing them to remain activated. In addition, the isolated scFvs can block the interaction between VISTA and the VSIG3 ligand. Further analysis on one scFv VS10 revealed that a significant insertion mutation occurred at the CDR3 of the VH domain compared to the chicken germline sequence. Finally, the complex structures of scFv VS10 and VISTA were simulated using molecular docking, and the interaction produced was analyzed. The experimental results support the applicability of VISTA as a target for immune checkpoints and contribute to explaining geometric structures and properties in chicken antibodies. The molecular mechanism underlying modulation of metamorphosis of the bivalve Mytilus coruscus by bacteria remains unclear. Here, the functional role of the thioesterase gene tesA of the bacterium Pseudoalteromonas marina in larval metamorphosis was examined. The aim was to determine whether inactivation of the tesA gene altered the biofilm-inducing capacity, bacterial cell motility, biopolymers, or the intracellular c-di-GMP levels. Complete inactivation of tesA increased the c-di-GMP content in P. marina, accompanied by a reduced fatty acid content, weaker motility, upregulation of bacterial aggregation, and biofilm formation. The metamorphosis rate of mussel larvae on ΔtesA biofilms was reduced by ∼ 80% compared with those settling on wild-type P. marina. Exogenous addition of a mixture of extracted fatty acids from P. marina into the ΔtesA biofilms promoted the biofilm-inducing capacity. This study suggests that the bacterial thioesterase gene tesA altered the fatty acid composition of ΔtesA P. marina biofilms (BF) through regulation of its c-di-GMP, subsequently impacting mussel metamorphosis. Abstract Fibroblast growth factor 7 (FGF7) is involved in lipid metabolism, which is considered as a candidate gene with close relation with muscle development by eGWAs and RNA-Seq analyses. To date, limited research has been conducted on the relationship between FGF7 gene and growth traits. The main objective of this work was to further investigate the association between novel InDel within FGF7 gene and growth traits in goat. Herein, FGF7 mRNA expression levels were investigated in various Fuqing goat tissues. We found that FGF7 gene was expressed in six adult goat tissues with the highest mRNA levels in adipose tissue. This result suggested that FGF7 gene might play a critical role in fat deposition. We also detected potential polymorphisms in Fuqing, Nubian and Jianyang Daer breeds. A 22-bp InDel polymorphism in FGF7 gene was detected in 396 goats and the three genotypes were designated as II, ID, and DD. Correlation analysis revealed that InDel polymorphism was significantly associated with growth traits (P < 0.05). Goats with genotypes ID and/or II had superior growth traits compared to those with genotype DD. In summary, our findings suggested that the 22-bp InDel within FGF7 gene could act as a molecular marker to improve the growth traits of goats in breeding programs. [ABSTRACT FROM AUTHOR]

Abstract The oriental armyworm, Mythimna separata has led to great economic losses of corn in China. The main method to control this pest relies on chemical insecticides. Plenty of genes in armyworm can be activated in response to insecticides. In this study, the critical genes involved in detoxification of chlorantraniliprole in armyworm were investigated using RNA-seq (transcriptome sequencing), digital gene expression profiles, and functional identification by RNA interference (RNAi) technology. The de novo assembly of the transcriptomes was achieved based on Illumina short-read sequencing technology in M. separata. There were 70,845 unigenes produced and 35,327 (49.86%) annotated. 748 unigenes were significantly up-expressed and 741 were significantly down-expressed in chlorantraniliprole treatment group compared to the control group. Silencing of cytochrome B6-F (CYB6), and long chain fatty acid CoA ligase 4 (CoA) of the top 18 highly expressed genes selected in chlorantraniliprole treatment group, resulted in more significant sensibility of M. separata to chlorantraniliprole, growth retardation and antifeedant phenomenon, which indicated that both CYB6 and CoA were most possibly responsible for chlorantraniliprole metabolism/detoxication.
