We present the size–mass relation of the brightest cluster galaxies (BCGs) at 0.1 < z < 1.4 using the imaging data obtained by the Hyper Suprime-Cam Subaru Strategic Program. Our sample consists of 471 photometrically selected BCGs with stellar mass log M */M⊙ = 11–12. We measure the size of the BCGs using i -band imaging and model fits based on a single Sérsic light profile. Stellar masses are determined through spectral energy distribution fitting using Sérsic-modelled photometry data across five optical bands (grizy). The size–mass scaling relation of BCGs is |$r_\mathrm{ e}\propto M_*^{0.73-1.00}$| at z < 1, with a slope that does not evolve significantly. The slope of the size–mass relation for BCGs is steeper than other non-BCG galaxies, which implies that BCGs are a special galaxy population. The size of BCGs at a given stellar mass evolves rapidly as ∝ (1 + z)−1.58 ± 0.13 and increases with redshift by a factor of 2.5 from z ∼ 1.2 to z ∼ 0.2. The rapid size growth is in agreement with semi-analytical model results, indicating that the BCG growth is dominated by frequent minor mergers. Furthermore, we explore the size–mass relationship of BCGs with respect to the halo mass of the cluster and find there is no significant correlation, which might imply that a dependence on the environment predominantly affects the outer envelope of the BCGs. We propose that observations of supermassive galaxies contain cosmological statistical constraining power similar to conventional cluster cosmology, and we provide promising indications that the associated systematic errors are comparably easier to control. We consider a fiducial spectroscopic and stellar mass complete sample of galaxies drawn from the Dark Energy Spectroscopic Instrument (DESI) and forecast how constraints on Ωm–σ8 from this sample will compare with those from number counts of clusters based on richness λ. At fixed number density, we find that massive galaxies offer similar constraints to galaxy clusters. However, a mass-complete galaxy sample from DESI has the potential to probe lower halo masses than standard optical cluster samples (which are typically limited to λ ≳ 20 and M halo ≳ 1013.5 M⊙  h −1); additionally, it is straightforward to cleanly measure projected galaxy clustering w p for such a DESI sample, which we show can substantially improve the constraining power on Ωm. We also compare the constraining power of M *-limited samples to those from larger but mass-incomplete samples [e.g. the DESI Bright Galaxy Survey (BGS) sample]; relative to a lower number density M *-limited samples, we find that a BGS-like sample improves statistical constraints by 60 per cent for Ωm and 40 per cent for σ8, but this uses small-scale information that will be harder to model for BGS. Our initial assessment of the systematics associated with supermassive galaxy cosmology yields promising results. The proposed samples have a ∼10 per cent satellite fraction, but we show that cosmological constraints may be robust to the impact of satellites. These findings motivate future work to realize the potential of supermassive galaxies to probe lower halo masses than richness-based clusters and to potentially avoid persistent systematics associated with optical cluster finding. The Merian survey is mapping ∼ 850 deg2 of the Hyper Suprime-Cam Strategic Survey Program (HSC-SSP) wide layer with two medium-band filters on the 4-m Victor M. Blanco telescope at the Cerro Tololo Inter-American Observatory, with the goal of carrying the first high signal-to-noise (S/N) measurements of weak gravitational lensing around dwarf galaxies. This paper presents the design of the Merian filter set: N708 (λ c  = 7080 Å, Δλ = 275 Å) and N540 (λ c  = 5400 Å, Δλ = 210 Å). The central wavelengths and filter widths of N708 and N540 were designed to detect the |$\rm H\alpha$| and |$\rm [OIII]$| emission lines of galaxies in the mass range |$8\lt \rm \log M_*/M_\odot \lt 9$| by comparing Merian fluxes with HSC broad-band fluxes. Our filter design takes into account the weak lensing S/N and photometric redshift performance. Our simulations predict that Merian will yield a sample of ∼ 85 000 star-forming dwarf galaxies with a photometric redshift accuracy of σΔ z /(1 + z) ∼ 0.01 and an outlier fraction of |$\eta =2.8~{{\ \rm per\ cent}}$| over the redshift range 0.058 < z < 0.10. With 60 full nights on the Blanco/Dark Energy Camera (DECam), the Merian survey is predicted to measure the average weak lensing profile around dwarf galaxies with lensing S/N ∼32 within r < 0.5 Mpc and lensing S/N ∼90 within r < 1.0 Mpc. This unprecedented sample of star-forming dwarf galaxies will allow for studies of the interplay between dark matter and stellar feedback and their roles in the evolution of dwarf galaxies. Intestinal microbiota is a potential determinant of obesity, with probiotic bile salt hydrolase (BSH) as one of the key mechanisms in the anti-obesity effects. In this study, we present a Lactobacillus acidophilus GOLDGUT-LA100 (LA100) with high BSH activity, good gastric acid and bile salt tolerance, and a potential anti-obesity effect. LA100's anti-obesity effects were evaluated in a high-fat diet-induced, obese mouse model. LA100 administration alleviates high-fat diet-induced pathophysiological symptoms, such as body weight gain, high serum glucose and cholesterol level, hepatic lipid accumulation, and adipose inflammation. The administration of probiotics is an effective approach for treatment of Helicobacter pylori , which is associated with human gastrointestinal diseases and cancers. To explore more effective probiotics for H. pylori infection elimination, bacteria from infant feces were screened in this study. We successfully isolated the Bifidobacterium animalis subsp. lactis strains and evaluated its efficacy to inhibit H. pylori growth in vitro and in vivo. The results showed that a B. animalis strain (named BB18) sustained a high survival rate after incubation in gastric juice. The rapid urease test suggested that B. animalis BB18 reduced pathogen loads in H. pylori -infected Mongolian gerbils. Alleviation of H. pylori infection-induced gastric mucosa damage and decreased levels inflammatory cytokines were observed after the B. animalis BB18 administration. These findings demonstrated that B. animalis BB18 can inhibit H. pylori infection both in vitro and in vivo , suggesting its potential application for the prevention and eradication therapy of H. pylori infection. The Gonghe Basin in the northeast Tibetan Plateau presents significant potential for hot dry rock (HDR) geothermal resources. A 1990 M w 6.4 earthquake in the basin furthers the need for an improved understanding of its sedimentary structure. In this study, we utilize data from a dense seismic array of 88 short-period seismometers deployed at an interstation spacing of approximately 3 km to scrutinize the sedimentary structure of the Gonghe Basin. By analysing teleseismic P waveforms, we identify P -to- S converted waves (Ps wave) originating from the sedimentary basement. We then determine the delay time between the Ps waves and the direct P waves (P wave) through waveform cross-correlation. By integrating this delay time with empirical velocity structure models, HDR borehole data and results from teleseismic receiver function analysis, we derive a sediment thickness model of the Gonghe Basin for the Qabqa geothermal area. Our findings reveal a gradual increase in sediment thickness from around 500 m in the east to approximately 3000 m in the west, which is consistent with other geophysical surveys and borehole data. The thick sediments in the basin could potentially serve as an excellent thermal storage cover for HDR. The strong ground motion simulation using our sediment thickness model shows that thick sediments can amplify seismic waves, increasing the risk of seismic hazards. Moreover, our study indicates that the clear Ps waves can be effectively extracted to construct a dependable sediment thickness model using teleseismic P waves recorded by a short-period dense seismic array. We compare stellar mass surface density, metallicity, age, and line-of-sight velocity dispersion profiles in massive (⁠|$M_*\ge 10^{10.5}\, \mathrm{M_\odot }$|⁠) present-day early-type galaxies (ETGs) from the MaNGA survey with simulated galaxies from the TNG100 simulation of the IllustrisTNG suite. We find an excellent agreement between the stellar mass surface density profiles of MaNGA and TNG100 ETGs, both in shape and normalization. Moreover, TNG100 reproduces the shapes of the profiles of stellar metallicity and age, as well as the normalization of velocity dispersion distributions of MaNGA ETGs. We generally also find good agreement when comparing the stellar profiles of central and satellite galaxies between MaNGA and TNG100. An exception is the velocity dispersion profiles of very massive (⁠|$M_*\gtrsim 10^{11.5}\, \mathrm{M_\odot }$|⁠) central galaxies, which, on average, are significantly higher in TNG100 than in MaNGA (⁠|$\approx 50\, \mathrm{km\, s^{-1}}$|⁠). We study the radial profiles of in situ and ex situ stars in TNG100 and discuss the extent to which each population contributes to the observed MaNGA profiles. Our analysis lends significant support to the idea that high-mass (⁠|$M_*\gtrsim 10^{11}\, \mathrm{M_\odot }$|⁠) ETGs in the present-day Universe are the result of a merger-driven evolution marked by major mergers that tend to homogenize the stellar populations of the progenitors in the merger remnant. Plasticizer di(2-ethylhexyl) phthalate (DEHP) is employed to make polyethylene polymers. Some studies in epidemiology and toxicology have shown that DEHP exposure over an extended period may be hazardous to the body, including nephrotoxicity, and aggravate kidney damage in the context of underlying disease. However, studies on the toxicity of DEHP in diabetes-induced kidney injury have been rarely reported. Using a high-fat diet (HFD) and streptozotocin (STZ, 35 mg/kg)-induced kidney injury in mice exposed to various daily DEHP dosages, we explored the impacts of DEHP on diabetes-induced kidney injury. We discovered that DEHP exposure significantly promoted the renal inflammatory response and oxidative stress in mice, with increased P-p38 and P-p65 protein levels and exacerbated the loss of podocin. The same findings were observed in vitro after stimulation of podocytes with high glucose (30 mmol/L) and exposure to DEHP. Our results suggest that DEHP exacerbates diabetes-induced kidney injury by mediating oxidative stress and activating p38MAPK/NF-κB. [ABSTRACT FROM AUTHOR]

Due to the complexity and diverse causes, the pathological mechanism of diet-induced colonic injury and colitis remains unclear. In this study, we studied the effects of the combination of a high-fat diet (HFD) plus alcohol on colonic injury in mice. We found HFD plus alcohol treatment induced disturbance of the gut microbiota; increased the production of intestinal toxins lipopolysaccharide (LPS), indole, and skatole; destroyed the stability of the intestinal mucosa; and caused the colonic epithelial cells damage through the activation of nuclear factor (NF)-κB and aromatic hydrocarbon receptors (AhR) signaling pathways. To mimic the effect of HFD plus alcohol in vivo, NCM460 cells were stimulated with alcohol and oleic acid with/without intestinal toxins (LPS, indole, and skatole) in vitro. Combinative treatment of alcohol and oleic acid caused moderate damage on NCM460 cells, while combination with intestinal toxins induced serious cell apoptosis. Western blot data indicated that the activation of NF-κB and AhR pathways further augmented after intestinal toxins treatment in alcohol- and oleic acid-treated colonic cells. This study provided new evidence for the relationship between diet pattern and colonic inflammation, which might partly reveal the pathological development of diet-induced colon disease and the involvement of intestinal toxins. It is clear that within the class of ultra-diffuse galaxies (UDGs), there is an extreme range in the richness of their associated globular cluster (GC) systems. Here, we report the structural properties of five UDGs in the Perseus cluster based on deep Subaru/Hyper Suprime-Cam imaging. Three appear GC-poor and two appear GC-rich. One of our sample, PUDG_R24, appears to be undergoing quenching and is expected to fade into the UDG regime within the next ∼0.5 Gyr. We target this sample with Keck Cosmic Web Imager (KCWI) spectroscopy to investigate differences in their dark matter haloes, as expected from their differing GC content. Our spectroscopy measures both recessional velocities, confirming Perseus cluster membership, and stellar velocity dispersions, to measure dynamical masses within their half-light radius. We supplement our data with that from the literature to examine trends in galaxy parameters with GC system richness. We do not find the correlation between GC numbers and UDG phase space positioning expected if GC-rich UDGs environmentally quench at high redshift. We do find GC-rich UDGs to have higher velocity dispersions than GC-poor UDGs on average, resulting in greater dynamical mass within the half-light radius. The problem of anomaly detection in astronomical surveys is becoming increasingly important as data sets grow in size. We present the results of an unsupervised anomaly detection method using a Wasserstein generative adversarial network (WGAN) on nearly one million optical galaxy images in the Hyper Suprime-Cam (HSC) survey. The WGAN learns to generate realistic HSC-like galaxies that follow the distribution of the data set; anomalous images are defined based on a poor reconstruction by the generator and outlying features learned by the discriminator. We find that the discriminator is more attuned to potentially interesting anomalies compared to the generator, and compared to a simpler autoencoder-based anomaly detection approach, so we use the discriminator-selected images to construct a high-anomaly sample of ∼13 000 objects. We propose a new approach to further characterize these anomalous images: we use a convolutional autoencoder to reduce the dimensionality of the residual differences between the real and WGAN-reconstructed images and perform UMAP clustering on these. We report detected anomalies of interest including galaxy mergers, tidal features, and extreme star-forming galaxies. A follow-up spectroscopic analysis of one of these anomalies is detailed in the Appendix; we find that it is an unusual system most likely to be a metal-poor dwarf galaxy with an extremely blue, higher-metallicity H  ii region. We perform a consistent comparison of the mass and mass profiles of massive (M ⋆ > 1011.4 M⊙) central galaxies at z ∼ 0.4 from deep Hyper Suprime-Cam (HSC) observations and from the Illustris, TNG100, and Ponos simulations. Weak lensing measurements from HSC enable measurements at fixed halo mass and provide constraints on the strength and impact of feedback at different halo mass scales. We compare the stellar mass function (SMF) and the Stellar-to-Halo Mass Relation (SHMR) at various radii and show that the radius at which the comparison is performed is important. In general, Illustris and TNG100 display steeper values of α where |$M_{\star } \propto M_{\rm vir}^{\alpha }$|⁠. These differences are more pronounced for Illustris than for TNG100 and in the inner rather than outer regions of galaxies. Differences in the inner regions may suggest that TNG100 is too efficient at quenching in situ star formation at M vir ≃ 1013 M⊙ but not efficient enough at M vir ≃ 1014 M⊙. The outer stellar masses are in excellent agreement with our observations at M vir ≃ 1013 M⊙, but both Illustris and TNG100 display excess outer mass as M vir ≃ 1014 M⊙ (by ∼0.25 and ∼0.12 dex, respectively). We argue that reducing stellar growth at early times in |$M_\star \sim 10^{9-10} \, \mathrm{M}_{\odot }$| galaxies would help to prevent excess ex-situ growth at this mass scale. The Ponos simulations do not implement AGN feedback and display an excess mass of ∼0.5 dex at r < 30 kpc compared to HSC which is indicative of overcooling and excess star formation in the central regions. The comparison of the inner profiles of Ponos and HSC suggests that the physical scale over which the central AGN limits star formation is r ≲ 20 kpc. Joint comparisons between weak lensing and galaxy stellar profiles are a direct test of whether simulations build and deposit galaxy mass in the correct dark matter haloes and thereby provide powerful constraints on the physics of feedback and galaxy growth. The splashback radius (R sp) of dark matter haloes has recently been detected using weak gravitational lensing and cross-correlations with galaxies. However, different methods have been used to measure R sp and to assess the significance of its detection. In this paper, we use simulations to study the precision and accuracy to which we can detect the splashback radius with 3D density, 3D subhalo, and weak lensing profiles. We study how well various methods and tracers recover R sp by comparing it with the value measured directly from particle dynamics. We show that estimates of R sp from density and subhalo profiles correspond to different percentiles of the underlying R sp distribution of particle orbits. At low accretion rates, a second caustic appears and can bias results. Finally, we show that upcoming lensing surveys may be able to constrain the R sp–Γdyn relation directly. [ABSTRACT FROM AUTHOR]

We use the UniverseMachine to analyse the source of scatter between the central galaxy mass, the total stellar mass in the halo, and the dark matter halo mass, for massive (M vir > 1013 M⊙) haloes. We also propose a new halo mass estimator, the cen+N mass: the sum of the stellar mass of the central and the N most massive satellites. We show that, when real space positions are perfectly known, the cen+N mass has scatter competitive with that of richness-based estimators. However, in redshift space, using a simple cluster finder, the cen+N mass suffers less from projection effects in the UniverseMachine model. The cen+N mass is therefore a potential candidate to constrain cosmology with upcoming spectroscopic data from DESI. We analyse the scatter in stellar mass at fixed halo mass and show that the total stellar mass in a halo is uncorrelated with secondary halo properties, but that the central stellar mass is a function of both halo mass and halo age. This is because central galaxies in older haloes have had more time to grow via accretion. If the UniverseMachine model is correct, this implies that haloes selected using the centrals stellar mass will be biased old and that accurate galaxy-halo modelling of mass selected samples therefore needs to consider halo age in addition to mass. Using deep images from the Hyper Suprime-Cam (HSC) survey and taking advantage of its unprecedented weak lensing capabilities, we reveal a remarkably tight connection between the stellar mass distribution of massive central galaxies and their host dark matter halo mass. Massive galaxies with more extended stellar mass distributions tend to live in more massive dark matter haloes. We explain this connection with a phenomenological model that assumes, (1) a tight relation between the halo mass and the total stellar content in the halo, (2) that the fraction of in situ and ex situ mass at r <10 kpc depends on halo mass. This model provides an excellent description of the stellar mass functions (SMFs) of total stellar mass (⁠|$M_{\star }^{\mathrm{max}}$|⁠) and stellar mass within inner 10 kpc (⁠|$M_{\star }^{10}$|⁠) and also reproduces the HSC weak lensing signals of massive galaxies with different stellar mass distributions. The best-fitting model shows that halo mass varies significantly at fixed total stellar mass (as much as 0.4 dex) with a clear dependence on |$M_{\star }^{10}$|⁠. Our two-parameter |$M_{\star }^{\mathrm{max}}$| – |$M_{\star }^{10}$| description provides a more accurate picture of the galaxy–halo connection at the high-mass end than the simple stellar–halo mass relation (SHMR) and opens a new window to connect the assembly history of haloes with those of central galaxies. The model also predicts that the ex situ component dominates the mass profiles of galaxies at r < 10 kpc for log  M ⋆ ≥ 11.7. Although photometric redshifts (photo- z 's) are crucial ingredients for current and upcoming large-scale surveys, the high-quality spectroscopic redshifts currently available to train, validate, and test them are substantially non-representative in both magnitude and colour. We investigate the nature and structure of this bias by tracking how objects from a heterogeneous training sample contribute to photo- z predictions as a function of magnitude and colour, and illustrate that the underlying redshift distribution at fixed colour can evolve strongly as a function of magnitude. We then test the robustness of the galaxy–galaxy lensing signal in 120 deg2 of HSC–SSP DR1 data to spectroscopic completeness and photo- z biases, and find that their impacts are sub-dominant to current statistical uncertainties. Our methodology provides a framework to investigate how spectroscopic incompleteness can impact photo- z -based weak lensing predictions in future surveys such as LSST and WFIRST. We study the faint stellar halo of isolated central galaxies, by stacking galaxy images in the HSC survey and accounting for the residual sky background sampled with random points. The surface brightness profiles in HSC r band are measured for a wide range of galaxy stellar masses (9.2 < log10 M ∗/M⊙ < 11.4) and out to 120 kpc. Failing to account for the stellar halo below the noise level of individual images will lead to underestimates of the total luminosity by ≤15%. Splitting galaxies according to the concentration parameter of their light distributions, we find that the surface brightness profiles of low-concentration galaxies drop faster between 20 and 100 kpc than those of high-concentration galaxies. Albeit the large galaxy-to-galaxy scatter, we find a strong self-similarity of the stellar halo profiles. They show unified forms once the projected distance is scaled by the halo virial radius. The colour of galaxies is redder in the centre and bluer outside, with high-concentration galaxies having redder and more flattened colour profiles. There are indications of a colour minimum, beyond which the colour of the outer stellar halo turns red again. This colour minimum, however, is very sensitive to the completeness in masking satellite galaxies. We also examine the effect of the extended PSF in the measurement of the stellar halo, which is particularly important for low-mass or low-concentration galaxies. The PSF-corrected surface brightness profile can be measured down to ∼31  |$\mathrm{mag}\, \mathrm{arcsec}^{-2}$| at 3σ significance. PSF also slightly flattens the measured colour profiles. Narrow-line regions excited by active galactic nuclei (AGNs) are important for studying AGN photoionization and feedback. Their strong [O  iii ] lines can be detected with broad-band images, allowing morphological studies of these systems with large-area imaging surveys. We develop a new broad-band imaging technique to reconstruct the images of the [O  iii ] line, using the Subaru Hyper Suprime-Cam (HSC) Survey aided with spectra from the Sloan Digital Sky Survey (SDSS). The technique involves a careful subtraction of the galactic continuum to isolate emission from the [O  iii ]λ5007 and [O  iii ]λ4959 lines. Compared to traditional targeted observations, this technique is more efficient at covering larger samples without dedicated observational resources. We apply this technique to an SDSS spectroscopically selected sample of 300 obscured AGN at redshifts 0.1–0.7, uncovering extended emission-line region candidates with sizes up to tens of kpc. With the largest sample of uniformly derived narrow-line region sizes, we revisit the narrow-line region size–luminosity relation. The area and radii of the [O  iii ] emission-line regions are strongly correlated with the AGN luminosity inferred from the mid-infrared (15 $$\mu$$ m rest frame) with a power-law slope of $$0.62^{+0.05}_{-0.06}\pm 0.10$$ (statistical and systematic errors), consistent with previous spectroscopic findings. We discuss the implications for the physics of AGN emission-line regions and future applications of this technique, which should be useful for current and next-generation imaging surveys to study AGN photoionization and feedback with large statistical samples. We use ∼100 deg2 of deep (>28.5 mag arcsec−2 in i band), high-quality (median 0.6 arcsec seeing) imaging data from the Hyper Suprime-Cam (HSC) survey to reveal the halo mass dependence of the surface mass density profiles and outer stellar envelopes of massive galaxies. The i -band images from the HSC survey reach ∼4 mag deeper than Sloan Digital Sky Survey and enable us to directly trace stellar mass distributions to 100 kpc without requiring stacking. We conclusively show that, at fixed stellar mass, the stellar profiles of massive galaxies depend on the masses of their dark matter haloes. On average, massive central galaxies with log10(M ⋆, 100 kpc/M⊙)>11.6 in more massive haloes at 0.3 <  z  < 0.5 have shallower inner stellar mass density profiles (within ∼10–20 kpc) and more prominent outer envelopes. These differences translate into a halo mass dependence of the mass–size relation. Central galaxies in haloes with log10(M 200b/M⊙)>14.0 are ∼20 per cent larger in R 50 at fixed M ⋆, 100 kpc. Such dependence is also reflected in the relationship between the stellar mass within 10 and 100 kpc. Comparing to the mass–size relation, the M ⋆, 100 kpc– M ⋆, 10 kpc relation avoids the ambiguity in the definition of size, and can be straightforwardly compared with simulations. Our results demonstrate that, with deep images from HSC, we can quantify the connection between halo mass and the outer stellar halo, which may provide new constraints on the formation and assembly of massive central galaxies. Background Cervical cancer is the most common cancer among females in Swaziland, yet the screening rate remains low. The study intended to explore the knowledge, attitudes and practice on cervical cancer and screening. Methods A cross-sectional questionnaire survey was conducted among 202 men and 213 women aged 30–65 years. The participants were from the Manzini and Shiselweni regions of Swaziland, sampled from both rural (77.8%) and urban areas. Results About half (53.5%) of women and 22.8% of men correctly named at least one symptom of cervical cancer. Many (58.1%) participants had misconceptions on the risk factors of cervical cancer, such as witchcraft, abortion, or birth control. Only 5.2% of the women had been screened. Among women, 40.0% reported that they need to seek their spouses' permission to visit the clinic. Men with less misconceptions on cervical cancer heard of screening and with no preference of the gender of the health care worker were more likely to report that they would allow their partners to be screened. Conclusions Men have less knowledge on cervical cancer yet may make important health related decisions for women. Respiratory tract toxicity represents a significant cause of attrition of inhaled drug candidates targeting respiratory diseases. One of the key issues to allow early detection of respiratory toxicities is the lack of reliable and predictive in vitro systems. Here, the relevance and value of a physiologically relevant 3D human airway in vitro model (MucilAir) were explored by repeated administration of a set of compounds with (n=8) or without (n=7) respiratory toxicity following inhalation in vivo. Predictability for respiratory toxicity was evaluated by readout of cytotoxicity, barrier integrity, viability, morphology, ciliary beating frequency, mucociliary clearance and cytokine release. Interestingly, the data show that in vivo toxicity can be predicted in vitro by studying cell barrier integrity by transepithelial electrical resistance (TEER), and cell viability determined by the Resazurin method. Both read-outs had 88% sensitivity and 100% specificity, respectively, while the former was more accurate with receiver operating characteristic (ROC) AUC of 0.98 (p = .0018) compared with ROC AUC of 0.90 (p = .0092). The loss of cell barrier integrity could mainly, but not fully, be attributed to a loss of cell coverage in 6 out of 7 compounds with reduced TEER. Notably, these effects occurred only at 400 µM, at concentration levels significantly above primary target cell potency, suggesting that greater attention to high local lung concentrations should be taken into account in safety assessment of inhaled drugs. The prozone phenomenon is associated with a wide range of antibody titers as well as pregnancy, primary syphilis, and neurosyphilis.Background. The prozone phenomenon is known to be associated with high antibody titers; other associations, such as host factors, have not been elucidated.Methods. A retrospective analysis was conducted to evaluate the incidence of the prozone phenomenon of the syphilis rapid plasma reagin (RPR) test among 46 856 clinical samples, between June 2010 and June 2013. Logistic regression was used to analyze the risk factors of the prozone phenomenon.Results. Our results showed that the incidence of the prozone phenomenon was low (0.83%) and could occur during any clinical phase, particularly during primary and secondary syphilis. Pregnancy and neurosyphilis were associated with the prozone phenomenon; sex, age, and whether the patient had been treated were not. The results also revealed that the prozone phenomenon not only occurred in patients with a high titer but also could occur in patients with a moderate/low titer. In fact, almost 31% of the patients with the prozone phenomenon had titers ≤1:16.Conclusions. The prozone phenomenon in the RPR test was associated with the phase of syphilis, pregnancy, and neurosyphilis as well as a range of RPR titers between 1:8 and 1:512. Syphilis screening should begin with a treponemal immunoassay that is followed by a second, different treponemal assay as confirmation. A nontreponemal assay is unnecessary for diagnosis but can be used for determining serological activity and the effect of syphilis treatment.Background. Algorithms for the diagnosis of syphilis continue to be a source of great controversy, and numerous test interpretations have perplexed many clinicians.Methods. We conducted a cross-sectional study of 24 124 subjects to analyze 3 syphilis testing algorithms: traditional algorithm, reverse algorithm, and the European Centre for Disease Prevention and Control (ECDC) algorithm. Every serum sample was simultaneously evaluated using the rapid plasma reagin, Treponema pallidum particle agglutination, and chemiluminescence immunoassay tests. With the results of clinical diagnoses of syphilis as a gold standard, we evaluated the diagnostic accuracy of the 3 syphilis testing algorithms. The κ coefficient was used to compare the concordance between the reverse algorithm and the ECDC algorithm.Results. Overall, 2749 patients in our cohort were diagnosed with syphilis. The traditional algorithm had the highest negative likelihood ratio (0.24), a missed diagnosis rate of 24.2%, and only 75.81% sensitivity. However, both the reverse and ECDC algorithms had higher diagnostic efficacy than the traditional algorithm. Their sensitivity, specificity, and accuracy were 99.38%–99.85%, 99.98%–100.00%, and 99.93%–99.96%, respectively. Moreover, the overall percentage of agreement and κ value between the reverse and the ECDC algorithms were 99.9% and 0.996, respectively.Conclusions. Our research supported use of the ECDC algorithm, in which syphilis screening begins with a treponemal immunoassay that is followed by a second, different treponemal assay as a confirmatory test in high-prevalence populations. The heat resistance of lactic acid bacteria ( LAB) has been extensively investigated due to its highly practical significance. Reconstituted skim milk ( RSM) has been found to be one of the most effective protectant wall materials for microencapsulating microorganisms during convective drying, such as spray drying. In addition to proteins and carbohydrate, RSM is rich in calcium. It is not clear which component is critical in the RSM protection mechanism. This study investigated the independent effect of calcium. Ca2+ was added to lactose solution to examine its influence on the heat resistance of Lactobacillus rhamnosus ZY, Lactobacillus casei Zhang, Lactobacillus plantarum P8 and Streptococcus thermophilus ND03. The results showed that certain Ca2+ concentrations enhanced the heat resistance of the LAB strains to different extents, that is produced higher survival and shorter regrowth lag times of the bacterial cells. In some cases, the improvements were dramatic. More scientifically insightful and more intensive instrumental study of the Ca2+ behavior around and in the cells should be carried out in the near future. This paper presents the second data release of the Hyper Suprime-Cam Subaru Strategic Program, a wide-field optical imaging survey using the 8.2 m Subaru Telescope. The release includes data from 174 nights of observation through 2018 January. The Wide layer data cover about 300 deg |$^2$| in all five broad-band filters (⁠|$grizy$|⁠) to the nominal survey exposure (10 min in |$gr$| and 20 min in |$izy$|⁠). Partially observed areas are also included in the release; about 1100 deg |$^2$| is observed in at least one filter and one exposure. The median seeing in the i -band is |${0_{.}^{\prime \prime }6}$|⁠ , demonstrating the superb image quality of the survey. The Deep (26 deg |$^2$|⁠) and UltraDeep (4 deg |$^2$|⁠) data are jointly processed and the UltraDeep-COSMOS field reaches an unprecedented depth of |$i\sim 28$| at |$5 \, \sigma$| for point sources. In addition to the broad-band data, narrow-band data are also available in the Deep and UltraDeep fields. This release includes a major update to the processing pipeline, including improved sky subtraction, PSF modeling, object detection, and artifact rejection. The overall data quality has been improved, but this release is not without problems; there is a persistent deblender problem as well as new issues with masks around bright stars. The user is encouraged to review the issue list before utilizing the data for scientific explorations. All the image products as well as catalog products are available for download. The catalogs are also loaded into a database, which provides an easy interface for users to retrieve data for objects of interest. In addition to these main data products, detailed galaxy shape measurements withheld from Public Data Release 1 (PDR1) are now available to the community. The shape catalog is drawn from the S16A internal release, which has a larger area than PDR1 (160 deg |$^2$|⁠). Reactive oxygen species (ROS) have been implicated in tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) resistance of many cancers. We evaluated the role of peroxiredoxin (Prx) I in TRAIL resistance governed by coupling of nicotinamide adenosine dinucleotide phosphate oxidase (Nox)-derived ROS signaling with the p38 mitogen-activated protein kinase (MAPK)/caspase-signaling cascade in liver cancer cells. Upregulated Prx I expression was found in neoplastic regions of human patient liver, and Prx I knockdown resulted in accelerated TRAIL-induced cell death in SK-Hep-1 human hepatoma cells. The TRAIL cytotoxicity by Prx I knockdown was dependent on activation of caspase-8/3 cascades, which was ablated by addition of inhibitors for p38 MAPK, ROS or Nox, suggesting the association with Nox-driven redox signaling. Furthermore, we found that Nox4 was constitutively expressed in both SK-Hep-1 cells and tumor regions of patient livers, knockdown of Nox4 expression could alleviate ROS generation and TRAIL-mediated cytotoxicity. In accordance with previous findings, increased activation of both p38 MAPK and caspase cascades by Prx I knockdown was inhibited by either Nox4 knockdown or SB203580 addition. Collectively, these data suggest that Prx I functions to block propagation of Nox-derived ROS signaling to the p38 MAPK/caspase/cell death cascade during TRAIL treatment and also provides a molecular mechanism by which Prx I contributes to TRAIL resistance in liver cancers.
