Determining bacterial identity at the strain level is critical for public health to enable proper medical treatments and reduce antibiotic resistance. Herein, we used liquid chromatography, ion mobility, and tandem MS (LC-IM-MS/MS) to distinguish Escherichia coli (E. coli) strains. Numerical multivariate statistics (principal component analysis, followed by linear discriminant analysis) showed the capability of this method to perform strain-level discrimination with prediction rates of 96.1% and 100% utilizing the negative and positive ion information, respectively. The tandem MS and LC separation proved effective in discriminating diagnostic lipid isomers in the negative mode, while IM separation was more effective in resolving lipid conformational biomarkers in the positive ion mode. Because of the clinical importance of early detection for rapid medical intervention, a faster technique, paper spray (PS)-IM-MS/MS, was used to discriminate the E. coli strains. The achieved prediction rates of the analysis of E. coli strains by PS-IM-MS/MS were 62.5% and 73.5% in the negative and positive ion modes, respectively. The strategy of numerical data fusion of negative and positive ion data increased the classification rates of PS-IM-MS/MS to 80.5%. Lipid isomers and conformers were detected, which served as strain-indicating biomarkers. The two complementary multidimensional techniques revealed biochemical differences between the E. coli strains confirming the results obtained from comparative genomic analysis. Suzuki–Miyaura cross-coupling is one of the most powerful strategies for constructing biaryl compounds. However, classic Suzuki–Miyaura coupling suffers from hour-scale reaction time and competitive protodeboronation. To address these problems, a mild nonaqueous potassium trimethylsilanolate (TMSOK)-assisted Suzuki–Miyaura coupling strategy was designed for the microsynthesis of biaryls in paper spray ionization (PSI). Due to the acceleration power facilitated by microdroplet chemistry in reactive PSI, the microsynthesis of biaryls by reactive PSI was accomplished within minutes with comparable yields to the bulk, showing good substrate applicability from 32 Suzuki–Miyaura reactions of aryl bromides and aryl boronic acid/borates bearing different substituents. Based on the above TMSOK-assisted Suzuki–Miyaura coupling strategy, we further developed a high-sensitivity and selective PSI mass spectrometry (MS) method for quantitative analysis of aryl bromides, a class of environmentally persistent organic pollutants that cannot be directly detected by ambient mass spectrometry due to their low ionization efficiency. In situ derivatization of aryl bromides was achieved with aryl borates bearing quaternary ammonium groups in PSI. The proposed PSI-MS method shows good linearity over the 0.01–10 μmol L–1 range with low detection limits of 1.8–4.8 nmol L–1 as well as good applicability to the rapid determination of six aryl bromides in three environmental water samples. The proposed PSI-MS method also shows good applicability to brominated flame retardants (polybrominated diphenyls/diphenyl esters). Overall, this study provides a simple, rapid, low-cost, high-sensitivity, and high-selectivity strategy for trace aryl bromides and other brominated pollutants in real samples with minimal/no sample pretreatment. This work describes the development of a system that combines a derivatization protocol based on the Katritzky reaction with paper spray ionization mass spectrometry (PSI-MS) for the analysis of amino acid neurotransmitters in mouse brain tissues. The system is relatively simple, consisting of spraying the derivatization solution onto a mouse brain section mounted on a glass slide, applying a small volume of solvent to moisten the sample, pressing a triangular paper onto the sample surface to transfer the sample constituents to the paper surface, and using the paper as a substrate for PSI-MS analysis. The Katritzky reaction facilitated the ionization of the amino acids by reacting a pyrylium salt with the amino group of the analytes, forming very stable pyridinium cations, which greatly increased the sensitivity of the PSI-MS analysis. Most of the intensities of the amino acids modified by the Katritzky reaction were more than 10 times greater than the nonderivatized ones. The system was applied for the analysis of brain sections obtained from mice with Parkinson's disease, and the amino acids gamma-aminobutyric acid (GABA) and glycine (Gly), two compounds very well-known in studies of Parkinson's disease, were readily detected. The results suggest that the Katritzky reaction combined with PSI-MS might offer a significant advance in the knowledge on protocols that improve the sensitivity of detection of crucial biological compounds. Microfluidic paper-based analytical devices (μPADs) have garnered significant interest as a promising analytical platform in the past decade. Compared with traditional microfluidics, μPADs present unique advantages, such as easy fabrication using established patterning methods, economical cost, ability to drive and manipulate flow without equipment, and capability of storing reagents for various applications. This Review aims to provide a comprehensive review of the field, highlighting fabrication methods available to date with their respective advantages and drawbacks, device designs and modifications to accommodate different assay needs, detection strategies, and the growing applications of μPADs. Porous media made of nitrocellulose and glass fiber are common “paper” substrates for lateral flow assays, microfluidic paper analytical devices and other point-of-care diagnostic assays. Such assays commonly use optical labels such as gold nano-particles, latex beads, or fluorescent nanoparticles to visualize the presence of analytes. Fluorescent labels are commonly used in bioassays to enhance sensitivity, but autoluminescence of the paper substrate worsens signal-to-noise ratios of fluorescence-based assays. To date, there exists no systematic investigation of autoluminescence wavelengths or lifetimes of porous membranes used in lateral flow assays. In response, we quantified the autoluminescence of commonly used porous materials across the visible spectrum via excitation—emission spectroscopy and timeresolved fluorescence spectroscopy, and demonstrate that autoluminescence is solely due to autofluorescence with lifetimes of about 5 ns in the visible spectrum. Counterintuitively, we found that spectroscopy alone does not provide sufficient information to select candidate paper substrates for fluorophore-labeled assays. Therefore, we developed a simple quantitative framework to select a low-fluorescence substrate that minimizes both the overlap of paper and fluorophore emission spectra and the fluorescence intensity on an imaging system of interest (such as a gel imager). Use of this framework was shown to lower the limit of detection of an influenza A nucleoprotein immunoassay by over 50%. The tools developed in this manuscript enable assay developers to screen appropriate, low-fluorescence porous substrates and enhance the sensitivity of membrane-based fluorescence assays. A new approach using paper spray ionization mass spectrometry (PSI–MS) for the analysis of steroid hormones in wastewater samples has been demonstrated. Triangular papers containing paraffin barriers as microfluidic channels were used to direct the sample solution to the paper tip, preventing the sample from spreading over the corners of the paper. The method was used to analyze the hormones levonorgestrel and algestone acetophenide in industrial wastewaters. Analytical curves presented a correlation coefficient (R2) above 0.99. Limits of quantification were below 2.3 ppm and limits of detection below 0.7 ppm. Values of precision (coefficient of variation) and accuracy (relative error) were less than 15% for all analyses. Recovery results ranged from 82% to 102%. Levonorgestrel was also analyzed by high-performance liquid chromatography coupled to mass spectrometry in order to compare the analytical performance with PSI–MS. No statistically significant differences were found between both methods. This study demonstrates the usefulness of PSI–MS for rapid analysis of hormones in industrial wastewater samples and also indicates its potential to be employed as a simple and reliable analytical method in environmental sciences. In this study, we report the use of pyrolysis-GCxGC/MS profiles for an optimized treatment of data issued from pyrolysis-GC/MS combined with the automatic deconvolution software Automated Mass Spectral Deconvolution and Identification System (AMDIS). The method was illustrated by the characterization of marker compounds of East Asian handmade papers through the examination of pyrolysis-GCxGC/MS data to get information which was used for manually identifying low concentrated and co-eluting compounds in 1D GC/MS data. The results showed that the merits of a higher separation power for co-eluting compounds and a better sensitivity for low concentration compounds offered by a GCxGC system can be used effectively for AMDIS 1D GC/MS data treatment: (i) the compound distribution in pyrolysis-GCxGC/MS profiles can be used as “peak finder” for manual check of low concentration and co-eluting compound identification in 1D GC/MS data, and (ii) pyrolysis-GCxGC/MS profiles can provide better quality mass spectra with observed higher match factors in the AMDIS automatic match process. This paper aims to develop a construction and operation cost model of wastewater treatment for the paper industry in China and explores the main factors that determine these costs. Previous models mainly involved factors relating to the treatment scale and efficiency of treatment facilities for deriving the cost function. We considered the factors more comprehensively by adding a regional variable to represent the economic development level, a corporate ownership factor to represent the plant characteristics, a subsector variable to capture pollutant characteristics, and a detailed-classification technology variable. We applied a unique data set from a national pollution source census for the model simulation. The major findings include the following: (l) Wastewater treatment costs in the paper industry are determined by scale, technology, degree of treatment, ownership, and regional factors; (2) Wastewater treatment costs show a large decreasing scale effect; (3) The current level of pollutant discharge fees is far lower than the marginal treatment costs for meeting the wastewater discharge standard. Key implications are as follows: (l) Cost characteristics and impact factors should be fully recognized when planning or making policies relating to wastewater treatment projects or technology development; (2) There is potential to reduce treatment costs by centralizing wastewater treatment via industrial parks; (3) Wastewater discharge fee rates should be increased; (4) Energy efficient technology should become the future focus of wastewater treatment. This paper describes the electrospray of resorcinol-formaldehyde (RF) sol on a carbon paper for making carbon electrodes. The sol layer undergoes curing, solvent removal, and pyrolysis under an inert atmosphere. In a separate study, the powdered carbon samples were derived from bulk RF-gel samples, prepared in bottles. The gel composition and the processing method were systematically varied. The EDX and BET analyses of the carbon samples helped in arriving at an optimized composition of the precursor sol, and the processing method for the electrospray derived carbon film. The electrodes showed a specific capacitance of 200 F g-1 at 0.1 A g-1 of current density with 2 M potassium chloride solution as electrolyte. The high value of specific capacitance was possible by the splitting of precursor sol prior to deposition on the current collector, and also by removal of solvent by lyophilization that retained the pore structure of the gel. Proteinaceous toxins are harmful proteins derived from plants, bacteria, and other natural sources. They pose a risk to human health due to infection and also as possible biological warfare agents. Paper spray mass spectrometry (PS-MS) with wipe sampling was used to detect proteins from surfaces as a potential tool for identifying the presence of these toxins. Proteins ranging in mass between 12.4 and 66.5 kDa were tested, including a biological toxin simulant/vaccine for Staphylococcal enterotoxin B (SEBv). Various substrates were tested for these representative proteins, including a laboratory bench, a notebook cover, steel, glass, plant leaf and vinyl flooring. Carbon sputtered porous polyethylene (CSPP) was found to outperform typical chromatography paper used for paper spray, as well as carbon nanotube (CNT)-coated paper and polyethylene (PE), which have been previously shown to be well-suited for protein analysis. Low microgram quantities of the protein toxin simulant and other test proteins were successfully detected with good signal-to-noise from surfaces using a porous wipe. These applications demonstrate that PS-MS can potentially be used for rapid, sample preparation-free detection of proteins and biological warfare agents, which would be beneficial to first responders and warfighters. This study provides a systematic approach for assessment of contaminants in materials for recycling. Paper recycling is used as an illustrative example. Three selected chemicals, bisphenol A (BPA), diethylhexyl phthalate (DEHP) and mineral oil hydrocarbons (MOHs), are evaluated within the paper cycle. The approach combines static material flow analysis (MFA) with dynamic material and substance flow modeling. The results indicate that phasing out of chemicals is the most effective measure for reducing chemical contamination. However, this scenario was also associated with a considerable lag phase (between approximately one and three decades) before the presence of chemicals in paper products could be considered insignificant. While improved decontamination may appear to be an effective way of minimizing chemicals in products, this may also result in lower production yields. Optimized waste material source-segregation and collection was the least effective strategy for reducing chemical contamination, if the overall recycling rates should be maintained at the current level (approximately 70% for Europe). The study provides a consistent approach for evaluating contaminant levels in material cycles. Currently, all assays measuring acetylcholinesterase (AChE) activity following a suspected nerve agent exposure leverage methodologies that fail to identify the agent. This limits the overall effectiveness and ability to administer proper countermeasures. As such, there is an urgent need to identify novel, rapid, and more comprehensive approaches to establish AChE activity, including identification of the toxicant. Paper spray mass spectrometry was used to monitor the activity of acetylcholinesterase, both in-solution and on modified hydrophobic paper surface. Hydrophobic paper surfaces were prepared using vaporized trichloro(3,3,3-trifluoropropyl)silane. In both approaches, mixtures of diluted human whole blood with and without VX were mixed with a non-endogenous AChE specific substrate, 1,1-dimethyl-4-acetylthiomethylpiperidinium (MATP+). Formation of the cleaved MATP+ product was monitored over time and compared to MATP+ to determine relative AChE activity. This on-substrate assay was effective at determining AChE activity and identifying the toxicant; however, determination of AChE activity in-solution proceeded at a slower rate. The on-substrate assay serves as a pioneering example of an enzymatic reaction occurring on the surface of a paper spray ionization ticket. This work describes a novel rapid method to fabricate high-resolution paper-based microfluidic devices using wax-ink-based printing. This study demonstrates that both temperature and pressure are important knobs in controlling the device resolution. High-resolution lines and patterns were obtained by heating the paper asymmetrically from one side up to 110 °C while applying pressure up to 49 kPa. Starting with wax lines with an initial width of 130 μm, we achieve a thorough penetration through a 190 μm-thick paper with lateral spreading on the front as narrow as 90 μm. The role of temperature and pressure are systematically studied and compared with the prediction of the Lucas-Washburn equation. We found that the temperature dependence of spreading can be explained by the viscosity change of the wax, according to the Lucas-Washburn equation. The pressure dependence deviates from Lucas-Washburn behavior because of compression of the paper. An optimal condition for achieving full depth penetration of the wax yet minimizing lateral spreading is suggested after exploring various parameters including temperature, pressure, and paper type. Inthis study, polyaniline-coated filter papers PANI (ES)-FP andPANI (EB)-FP have been synthesized by chemical oxidation polymerization.This is a cost-effective and simple method of preparing PANI filmon an inexpensive filter paper. The PANI coated filter papers werecharacterized by UV–vis, IR, thermogravimetric analysis andscanning electron microscopy. These composite materials have beeneffectively used for adsorption of seven different dyes from aqueousmedium. It has been observed that these are good materials for anionicand cationic dye adsorption. Detailed equilibrium and kinetic studieshave been reported here by taking Eosin yellow and Methylene blueas anionic and cationic model dyes. The experimental data for Eosinyellow and Methylene blue adsorption were best fitted in Freundlichand Langmuir adsorption models, respectively. The Langmuir adsorptioncapacity of Eosin yellow and Methylene blue on PANI (ES)-FP and PANI(EB)-FP were 4.3 and 1.3 mg g–1, respectively, atneutral pH. We have developed a new method for characterizing paper coating structure using FIB milling technique and FE-SEM. A new image analysis method combining smart blur and locally adaptive thresholding technique was applied. Compared to conventional method, this method preserved the edge of target and gave better automatic image segmentation of pores. We applied this method for the measurement of the pore structure in pigment coating layers. Staining with OsO4and resin-embedding treatments were used for the sample preparation of coated papers for image analysis. A control sample without any treatment was also used in analysis. BSE images of osmium stained and resin-embedded coated paper gave the clearest image segmentation of coating components. The results showed that three sample preparation methods gave similar z-directional porosities. Aptamer-based paper strip sensor for detecting Vibrio fischeri was developed. Our method was based on the aptamer sandwich assay between whole live cells, V. fischeri and DNA aptamer probes. Following 9 rounds of Cell-SELEX and one of the negative-SELEX, V. fischeri Cell Aptamer (VFCA)-02 and -03 were isolated, with the former showing approximately 10-fold greater avidity (in the subnanomolar range) for the target cells when arrayed on a surface. The colorimetric response of a paper sensor based on VFCA-02 was linear in the range of 4 × 101 to 4 × 105 CFU/mL of target cell by using scanning reader. The linear regression correlation coefficient (R2) was 0.9809. Paper spray ionization has arisen relatively recently as a complement and alternative to electro- and nanospray ionization with silica capillaries. A majority of the work in the present literature focuses on the chemical aspect of paper spray. In order to study the physical and phenomenological facet of its implementation, we measured current and voltage distributions of Taylor cones. To study transport phenomena on filter paper, we addressed the behavior of large, sparingly soluble tetraalkylammonium ions, which are usually used as mobility standards, in paper spray. The variation of intensity with time of monomers and dimers of these ions was measured with a differential mobility analyzer and compared with that produced by contamination in the paper. At the same time, we evaluated the proficiency of different paper spray techniques for protein analysis using nano spray as a reference. Experiments suggest that Taylor cones in paper spray are subject to hysteresis, whereas transport phenomena in the porous substrate notably affects the ionization of the sample. Significant amounts of paper deinking residue (DIR) has been and is still being generated from paper deinking processes, representing both an economic and environmental burden for recycled paper mills. Our research on low-temperature (<200 °C) microwave-assisted (MW-assisted) pyrolysis of DIR allows for simultaneously efficient fast separation and recovery of the organic and inorganic content of DIR at relatively low temperature and within 15 min. Our study is the first highly detailed account of the use low-temperature MW-assisted pyrolysis to effect this change. The obtained liquid and solid products were characterized by a variety of analytical techniques (e.g., attenuated total reflection infrared, gas chromatography-mass spectrometry, liquid-state nuclear magnetic resonance (NMR), X-ray diffraction, solid-state cross-polarization/magic-angle spinning 13C NMR, and Bloch-decay 13C NMR). The results reveal that the process efficiently separates the inorganic minerals as microwave residue (mainly calcite and kaolinite) from organic matter, and hence the microwave residue could be reused to produce new paper/cardboard products. The organic fraction bio-oil generated is energy-densified and rich in carbohydrates and is a potential source for valuable aromatic compounds. [ABSTRACT FROM AUTHOR]

A simple, reliable, and low-cost fabrication method is proposed here for assembling paper-based electrochemical devices (PEDs) using a commercial desktop digitally controlled plotter/cutter, together with ordinary writing tools. Permanent markers (tips of 1 mm) were used to create effective hydrophobic barriers on paper, while micromechanical pencils (mounting 4B graphite leads, 0.5 mm in diameter) were adopted for automatically drawn precise reference, counter, and working carbon electrodes. Fabrication parameters, such as writing pressure and speed, were first optimized, and the electrochemical performance of these devices was then evaluated by using potassium hexacyanoferrate(II) as redox probe. The good interdevice reproducibility (4.8%) displayed by the relevant voltammetric responses confirmed that this strategy can be profitably adopted to easily assemble paper-based electrochemical devices in a highly flexible manner. The simplicity of the instrumentation used and the low cost of each single device (about $0.04), together with the speed of fabrication (about 2 min), are other important features of the proposed strategy. Finally, to confirm the effectiveness of this prototyping method for the analysis of real samples and rapid controls, PEDs assembled by this simple approach were successfully exploited for the analysis of vitamin B6 in food supplements. Three main fabrication steps for microfluidic paper-based analytical devices (μPADs) were fully integrated with accurate geometrical alignment between the individual steps in a simple and rapid manner. A wax printer for creating hydrophobic barriers was integrated with an inexpensive (ca. $300) electronic craft plotter/cutter for paper cutting, followed by colorimetric reagent deposition using technical pens. The principal shortcoming in the lack of accurate and precise alignment of the features created by these three individual fabrication steps was addressed in this work by developing appropriate alignment procedures during the multistep fabrication process. The wax printing step was geometrically aligned with the following cutting and plotting (deposition) steps in a highly accurate and precise manner using optical scanning function of the plotter/cutter based on registration marks printed on the paper using the wax printer and scanned by the plotter/cutter. The accuracy and precision of alignment in a two-dimensional plane were quantified by cutting and plotting cross-shaped features and measuring their center coordinates relative to wax printed reference lines. The average accuracy along the X- and Y-axis was 0.12 and 0.16 mm for cutting and 0.19 and 0.17 mm for plotting, respectively. The potential of this approach was demonstrated by fabricating μPADs for instrument-free determination of cobalt in waters using distance-based readout, with excellent precision (96RSD = 5.7) and detection limit (LOD) of 2.5 ng and 0.5 mg/L (mass and concentration LODs, respectively). As a cheap and abundant porous material, cellulose filter paper was used to immobilize nano-TiO2 and denoted as TiO2/cellulose paper (TCP). With high adsorption capacity for Cu(II) (more than 1.65 mg), TCP was used as an adsorbent, photocatalyst, and colorimetric sensor at the same time. Under the optimum adsorption conditions, i.e., pH 6.5 and 25 °C, the adsorption ratio of Cu(II) was higher than 96.1%. Humic substances from the matrix could be enriched onto TCP but the interference of their colors on colorimetric detection could be eliminated by the photodegradation. In the presence of hydroxylamine, neocuproine, as a selective indicator, was added onto TCP, and a visual color change from white to orange was generated. The concentration of Cu(ll) was quantified by the color intensity images using image processing software. This fully integrated visual analysis system was successfully applied for the detection of Cu(n) in 10.0 L of drinking water and seawater with a preconcentration factor of 104. The log-linear calibration curve for Cu(II) was in the range of 0.5-50.0 μg L-1 with a determination coefficient (R2) of 0.985 and its detection limit was 0.073 p% L-1. [ABSTRACT FROM AUTHOR]

We have demonstrated a multistep 2-dimensional paper network immunoassay based on controlled rehydration of patterned, dried reagents. Previous work has shown that signal enhancement improves the limit of detection in 2-dimensional paper network assays, but until now, reagents have only been included as wet or dried in separate conjugate pads placed at the upstream end of the assay device. Wet reagents are not ideal for point-of-care because they must be refrigerated and typically limit automation and require more user steps. Conjugate pads allow drying but do n o t offer any control of the reagent distribution upon rehydration and can be a source of error when pads do not contact the assay membrane uniformly. Furthermore, each reagent is dried on a separate pad, increasing the fabrication complexity when implementing multistep assays that require several different reagents. Conversely, our novel method allows for consistent, controlled rehydration from patterned reagent storage depots directly within the paper membrane. In this assay demonstration, four separate reagents were patterned in different regions of the assay device: a gold-antibody conjugate used for antigen detection and three different signal enhancement components that must not be mixed until immediately before use. To show the viability of patterning and drying reagents directly onto a paper device for dry reagent storage and subsequent controlled release, we tested this device with the malaria antigen Plasmodium falciparum histidine-rich protein 2 (PfHRP2) as an example of target analyte. Pulp and paper production consumesa large amount of water andsteam. In particular, water is used in brown stock washing system(BSWS) to separate spent white liquor from the pulp stream. The wastewaterstream generated from BSWS, known as black liquor, is a potentialbiomass. In the current practice, black liquor is concentrated andburned in a recovery boiler to produce steam and electricity. However,recent development in an integrated biorefinery offers increased marketabilityin pulp and paper mills through the generation of both bioproductsand bioenergy. It is common practice for steam generated from thecombustion of black liquor to be integrated with pulp and paper millto satisfy its heating demands. Note that the water consumption inBSWS directly affects the quantity and quality of black liquor. Thiswill later affect the potential for steam generation and integrationin pulp and paper mill. Thus, in this work, water and heat integrationis performed for a pulp and paper mill with biorefinery. An integratedpulp and paper biorefinery (IPPB) is proposed which accounts for differentconversion pathways of pulp and paper mill biomass. Apart from combustion,several gasification pathways are considered. A nonlinear program(NLP) is formulated to synthesize an optimum IPPB with maximum resourcesconservation. This article describes the use of paper spray mass spectrometry (PS-MS) for the direct analysis of black ink writings made with ballpoint pens. The novel approach was developed in a forensic context by first performing the classification of commercially available ballpoint pens according to their brands. Six of the most commonly worldwide utilized brands (Bic, Paper Mate, Faber Castell, Pentel, Compactor, and Pilot) were differentiated according to their characteristic chemical patterns obtained by PS-MS. MS on the negative ion mode at a mass range of m/ z 100-1000 allowed prompt discrimination just by visual inspection. On the other hand, the concept of relative ion intensity (RII) and the analysis at other mass ranges were necessary for the differentiation using the positive ion mode. PS-MS combined with partial least squares (PLS) was utilized to monitor changes on the ink chemical composition after light exposure (artificial aging studies). The PLS model was optimized by variable selection, which allowed the identification of the most influencing ions on the degradation process. The feasibility of the method on forensic investigations was also demonstrated in three different applications: (1) analysis of overlapped fresh ink lines, (2) analysis of old inks from archived documents, and (3) detection of alterations (simulated forgeries) performed on archived documents. A field-based metabolomic study was conducted during a shutdown of a pulp and paper mill (PPM) to assess the impacts of treated PPM effluent on endogenous polar metabolites in fathead minnow (FHM; Pimephales promelas) livers. Caged male and female FHMs were deployed at a Great Lakes area of concern during multiple periods (pre-, during, and post-shutdown) near the outflow for a wastewater treatment plant. Influent to this plant is typically 40% PPM effluent by volume. Additional FHMs were exposed to reference lake water under laboratory conditions. A bioassay using T47D-KBluc cells showed that estrogenic activity of receiving water near the outflow declined by 46% during the shutdown. We then used 1H NMR spectroscopy and principal component analysis to profile abundances of hepatic endogenous metabolites for FHMs. Profiles for males deployed pre-shutdown in receiving water were significantly different from those for laboratory-control males. Profiles were not significantly different for males deployed during the shutdown, but they were significant again for those deployed post-shutdown. Impacts of treated effluent from this PPM were sex-specific, as differences among profiles of females were largely nonsignificant. Thus, we demonstrate the potential utility of field-based metabolomics for performing biologically based exposure monitoring and evaluating remediation efforts occurring throughout the Great Lakes and other ecosystems. We present a microfluidic paper analytical device (μPAD) that relies on flow in hollow channels, rather than through a cellulose network, to transport fluids. The flow rate in hollow channels is 7 times higher than in regular paper channels and can be conveniently controlled from 0 to several mm/s by balancing capillary and pressure forces. More importantly, the pressure of a single drop of liquid (0.2 mbar) is sufficient to induce fast pressure-driven flow, making hollow channels suitable for point of care diagnostics. We demonstrate their utility for simple colorimetric glucose and BSA assays in which the time for liquid transport is reduced by a factor of 4 compared to normal cellulose channels. To determine the benthic transformation pathways and fate of carbon associated with secondary-treated pulp and paper mill (PPM) effluent, 13C-labeled activated sludge biomass (ASB) and phytoplankton (PHY) were added, separately, to estuarine intertidal sediments. Over 28 days, 13C was traced into sediment organic carbon, fauna, seagrass, bacteria, and microphytobenthos and into fluxes of dissolved organic carbon (DOC) and dissolved inorganic carbon (DIC) from inundated sediments, and carbon dioxide (CO2(g)) from exposed sediments. There was greater removal of PHY carbon from sediments (∼85% over 28 days) compared to ASB (∼75%). Although there was similar 13C loss from PHY and ASB plots via DIC (58% and 56%, respectively) and CO2(g) fluxes (<1%), DOC fluxes were more important for PHY (41%) than ASB (12%). Faster downward transport and loss suggest that tauna prefer PHY, due to its lability and/or toxins associated with ASB; this may account for different carbon pathways. Secondary-treated PPM effluent has lower oxygen demand than primary-treated effluent, but ASB accumulation may contribute to sediment anoxia, and respiration of ASB and PHY-derived DOC may make the water column more heterotrophic. Paper spray tandem mass spectrometry is used to identify and quantify eight individual amphetamines in whole blood in 1.3 min. The method has been optimized and fully validated according to forensic toxicology guidelines, for the quantification of amphetamine, methamphetamine, 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxy- N-methylamphetamine (MDMA), 3,4-methylenedioxy- N-ethylamphetamine (MDEA), para-methoxyamphetamine (PMA), para-methoxymethamphetamine (PMMA), and 4-fluoroamphetamine (4-FA). Additionally, a new concept of intrinsic and application-based selectivity is discussed, featuring increased confidence in the power to discriminate the amphetamines from other chemically similar compounds when applying an ambient mass spectrometric method without chromatographic separation. Accuracy was within ±15% and average precision was better than 15%, and better than 20% at the LLOQ. Detection limits between 15 and 50 ng/mL were obtained using only 12 μL of whole blood. Bisphenol A (BPA) is used in a variety of consumer products, including some paper products, particularly thermal receipt papers, for which it is used as a color developer. Nevertheless, little is known about the magnitude of BPA contamination or human exposure to BPA as a result of contact with paper and paper products. In this study, concentrations of BPA were determined in 15 types of paper products (n = 202), including thermal receipts, flyers, magazines, tickets, mailing envelopes, newspapers, food contact papers, food cartons, airplane boarding passes, luggage tags, printing papers, business cards, napkins, paper towels, and toilet paper, collected from several cities in the USA. Thermal receipt papers also were collected from Japan, Korea, and Vietnam. BPA was found in 94% of thermal receipt papers (n = 103) at concentrations ranging from below the limit of quantitation (LOQ, 1 ng/g) to 13.9 mg/g (geometric mean: 0.211 mg/g). The majority (81%) of other paper products (n = 99) contained BPA at concentrations ranging from below the LOQ to 14.4 μg/g (geometric mean: 0.016 μg/g). Whereas thermal receipt papers contained the highest concentrations of BPA (milligram-per-gram), some paper products, including napkins and toilet paper, made from recycled papers contained microgram-per-gram concentrations of BPA. Contamination during the paper recycling process is a source of BPA in paper products. Daily intake (DI) of BPA through dermal absorption was estimated based on the measured BPA concentrations and handling frequency of paper products. The daily intake of BPA (calculated from median concentrations) through dermal absorption from handling of papers was 17.5 and 1300 ng/day for the general population and occupationally exposed individuals, respectively; these values are minor compared with exposure through diet. The article examines the chemistry of paper preservation. Permanence in paper refers to a degree of chemical stability that allows only a very slow deterioration. Durability refers to a degree of physical strength and flexibility of paper required for extensive handling. Both permanence and durability are important qualities that must be considered in determining standards for permanent paper. Since lignin-containing papers tend to yellow on exposure to light, it was concluded that the presence of lignin contributed to strength loss in paper.

In this technical note, we describe a facile method for one-step fabrication of paper-based microfluidic devices, by simply using commercially available permanent markers and metal templates with specific patterns. The fabrication process involves only a single step of plotting pattern in paper; it can be typically finished within 1 min. The ink marks formed in the patterned paper will act as the hydrophobic barriers to define the hydrophilic flow paths or separate test zones. Various paper devices can be created by using different templates with corresponding patterns. Transparent adhesive tape-sandwiched devices could protect their assay surfaces from potential contamination. In the proof-of-concept experiments, circular paper test zones (~3 mm diameter) were fabricated for colorimetric and quantification detection of prostate-specific antigen (PSA) as a model target, based on dot-immunogold staining assays coupled with gold enhancement amplification. Several serum specimens were additionally evaluated with this new approach and the results were compared with the commercial chemiluminescence immunoassay, validating its feasibility of practical applications. Such a one-step plotting method for paper patterning does not require any specialized equipments and skills, is quite inexpensive and rapid, and thus holds great potential to find wide applications especially in remote regions and resource-limited environments such as small laboratories and private clinics. It has been reported that about 10−15% of the fresh water intake in a paper mill is used for feeding and diluting retention aids, so significant savings could be achieved by replacing fresh water with process water. Water from different sources and qualitiesfresh water, the outflow from an internal ultrafiltration placed in the machine circuit of a paper mill, and water from a membrane bioreactor used to treat the final effluent of this paper millwere used to prepare a dual retention system consisting of a cationic polyacrylamide and bentonite. While the behavior of bentonite was not significantly affected by the quality of the water used in its preparation, the efficiency of the cationic polyacrylamide was reduced to about 12% when it was prepared with water with high anionic trash content and conductivity as a result of a partial neutralization of the charged groups. Viral pathogens are a serious health threat around the world, particularly in resource limited settings, where current sensing approaches are often insufficient and slow, compounding the spread and burden of these pathogens. Here, we describe a label-free, point-of-care approach toward detection of virus particles, based on a microfluidic paper-based analytical device with integrated microwire Au electrodes. The device is initially characterized through capturing of streptavidin modified nanoparticles by biotin-modified microwires. An order of magnitude improvement in detection limits is achieved through use of a microfluidic device over a classical static paper-based device, due to enhanced mass transport and capturing of particles on the modified electrodes. Electrochemical impedance spectroscopy detection of West Nile virus particles was carried out using antibody functionalized Au microwires, achieving a detection limit of 10.2 particles in 50 μL of cell culture media. No increase in signal is found on addition of an excess of a nonspecific target (Sindbis). This detection motif is significantly cheaper (~$1 per test) and faster (~30 min) than current methods, while achieving the desired selectivity and sensitivity. Highly conductive, robust, and multifunctional integrated paper-supported electrodes are requisite to fulfill the promise of paper-based analytical application. Herein, an all-in-one Au electrode comprising of detection zone, waterproof electronic bridge, and signal output contactor was engineered via combining the double-sided growth method with the secondary wax-printing. Benefiting from the strongly omnidirectional conductivity and desirably mechanical robustness of the as-prepared electrode, a stackable lab-on-paper cyto-device integrated with high-efficiency photoelectrochemical strategy was developed for the MCF-7 cells assay. Specifically, the detection zone of the electrode, serving as the signal generator, was functionalized with a low-toxic cosensitized structure composed of corn-like ZnO nanorods, graphene quantum dots (GQDs), and Ag2Se QDs. With the proximity control of DNA hairpin-based aptamer probe (DHAP), a strong photocurrent could be promoted by the activated cosensitization effect and collected on the signal output contactor via the electron transport of waterproof electronic bridge. Upon the MCF-7 cells recognition, the DHAP switched from closed to open state with the formation of DNA-cell bioconjugates and the spatial separation of Ag2Se QDs linked on the terminal of DHAP from the electrode surface. The photocurrent was noticeably decreased due to the double inhibition of steric hindrance effect and vanished cosensitization effect. Based on the target-triggered photocurrent attenuation, the sensitive detection of target cells was achieved. This work not only provided a unique method for paper-based electrode preparation but also offered a powerful platform for the highly sensitive photoelectrochemical bioanalysis. This study demonstrates a novel strategy for colorimetric/surface-enhanced Raman scattering (SERS) dual-mode sensing of sulfur dioxide (SO2) by coupling headspace sampling (HS) with paper-based analytical device (PAD). The smart and multifunctional PAD is fabricated with a vacuum filtration method in which 4-mercaptopyridine (Mpy)-modified gold nanorods (GNRs)-reduced graphene oxide (rGO) hybrids (rGO/MPy-GNRs), anhydrous methanol, and starch-iodine complex are immobilized into cellulose-based filter papers. The resultant PAD exhibits a deep-blue color with a strong absorption peak at 600 nm due to the formation of an intermolecular charge-transfer complex between starch and iodine. However, the addition of SO2 induces the Karl Fischer reaction, resulting in the decrease of color and increase of SERS signals. Therefore, the PAD can be used not only as a naked-eye indicator of SO2 changed from blue to colorless but also as a highly sensitive SERS substrates because of the SO2-triggered conversion of Mpy to pyridine methyl sulfate on the GNRs. A distinguishable change in the color was observed at a SO2 concentration of 5 µM by the naked eye, and a detection limit as low as 1.45 µM was obtained by virtue of UV-vis spectroscopy. The PAD-based SERS method is effective over a wide range of concentrations (1 µM to 2000 µM) for SO2, and the detection limit for SO2 is found to be 1 µM. The HS-PAD based colorimetric/SERS method is applied for the determination of SO2 in wine, and the detection results match well with those obtained from the traditional Monier-Williams method. This study not only offers a new method for on-site monitoring of SO2 but also provides a new strategy for designing of paper-based sensing platform for a wide range of field-test applications. In this study, paper-based microfluidic devices (μPADs) capable of K+ quantification in aqueous samples, as well as in human serum, using both colorimetric and distance-based methods are described. A lipophilic phase containing potassium ionophore I (valinomycin) was utilized to achieve highly selective quantification of K+ in the presence of Na+, Li+, and Mg2+ ions. Successful addition of a suspended lipophilic phase to a wax printed paper-based device is described and offers a solution to current approaches that rely on organic solvents, which damage wax barriers. The approach provides an avenue for future alkali/ alkaline quantification utilizing μPADs. Colorimetric spot tests allowed for K+ quantification from 0.1-5.0 mM using only 3.00 μL of sample solution. Selective distance-based quantification required small sample volumes (6.00 μL) and gave responses sensitive enough to distinguish between 1.0 and 2.5 mM of sample K+. μPADs using distance-based methods were also capable of differentiating between 4.3 and 6.9 mM K+ in human serum samples. Here we introduce a new ambient ionization technique named laser assisted paper spray ionization mass spectrometry (LAPSI MS). In it, a 532 ± 10 nm, ≤10 mW laser pointer was shone on a triangularly cut paper along with high voltage, to effect ionization. The analyte solution was continuously pushed through a fused silica capillary, using a syringe pump, at a preferred infusion rate. LAPSI MS promises enhanced ionization with high signal intensity of polycyclic aromatic hydrocarbons (PAHs), which are normally not ionizable with similar ionization methods involving solvent sprays. LAPSI MS works both in positive and negative modes of ionization. A clear enhancement of signal intensity was visualized in the total ion chronogram for most analytes in the presence of the laser. We speculate that the mechanism of ionization is field assisted photoionization. The field-induced distortion of the potential well can be large in paper spray as the fibers comprising the paper are separated at tens of nanometers apart, and consequently, the analyte molecules are subjected to very large electric fields of the order of 107 Vcm-1. Ionization occurs from their distorted electronic states of reduced ionization energy, using the laser. Negative ion detection is also demonstrated, occurring due to the capture of produced photoelectrons. LAPSI MS can be used for monitoring in situ photoassisted reactions like the decarboxylation of mercaptobenzoic acid in the presence of gold and silver nanoparticles and the dehydrogenation reaction of 2,3-dihydro-1H-isoindole, which were chosen as examples. As an application, we have shown that paraffin oil, which is usually nonionizable by paper spray or by electrospray ionization can be efficiently detected using this technique. We report a novel and innovative electrochemical paper-based immunocapture assay (EPIA) to address the need for ultrasensitive detection of emerging pollutants without regulatory status and whose effects on environment and human health are not completely yet understood. In particular, we present the application of this system toward highly sensitive detection of the emerging pollutant ethinyl estradiol (EE2). The EPIA approach is based on the use of paper microzones modified with silica nanoparticles (SNs) and anti-EE2 specific antibodies for capture and preconcentration of EE2 from river water samples. After the preconcentration procedure, the paper microzones are placed onto a screen-printed carbon electrode modified with electrochemically reduced graphene (RG). The bound EE2 is subsequently desorbed adding a diluted solution of sulfuric acid on the paper microzones. Finally, recovered EE2 is electrochemically detected by OSWV. The proposed novel methodology showed an appropriate LOD and linear range for the quantification of EE2 for water samples with different origins. The nonsophisticated equipment required, the adequate recovery values obtained (from 97% to 104%, with a RSD less than 4.9%), and the appropriate LOD and linear range value (0.1 ng L-1 and 0.5-120 ng L-1, respectively) achieved by our immunocapture sensor present significant analytical figures of merit, particularly when the routine quantification of EE2 is considered. In addition, our system was based on electrochemical paper-based technology, which allows obtainment of portable, easy-to-use, inexpensive, and disposable devices. Rapid and selective sensing of sulfur dioxide (SO2) gas has attracted more and more attention because SO2 not only causes environmental pollution but also severely affects the health of human beings. Here we report an amino-functionalized luminescent metal-organic framework (MOF) material (i.e., MOF-5-NH2) and further investigate its sensing property for SO2 gas and its derivatives as a luminescent probe. The results indicate that the MOF-5-NH2 probe can selectively and sensitively sense SO2 derivatives (i.e., SO3 2-) in real time by a luminescence turn-on effect with a lower detection limit of 0.168 ppm and a response time of less than 15 s. Importantly, the luminescence turn-on phenomenon can be observed by the naked eye. We also assembled MOF-5-NH2 into a test paper to achieve the aim of portable detection, and the lower-limit concentration of the test paper for sensing SO2 in real time was found to be about 0.05 ppm. Moreover, MOF-5-NH2 also shows good anti-interference ability, strong luminescence stability, and reusability, which means that this material is an excellent sensing candidate. As paper-based diagnostics has become predominantly driven by more advanced microfluidic technology, many of the research efforts are still focused on developing reliable and versatile fluidic control devices, apart from improving sensitivity and reproducibility. In this work, we introduce a novel and robust paper fluidic control system enabling versatile fluidic control. The system comprises a linear push-pull solenoid and an Arduino Uno microcontroller. The precisely controlled pressure exerted on the paper stops the flow. We first determined the stroke distance of the solenoid to obtain a constant pressure while examining the fluidic time delay as a function of the pressure. Results showed that strips of grade 1 chromatography paper had superior reproducibility in fluid transport. Next, we characterized the reproducibility of the fluidic velocity which depends on the type and grade of paper used. As such, we were able to control the flow velocity on the paper and also achieve a complete stop of flow with a pressure over 2.0 MPa. Notably, after the actuation of the pressure driven valve (PDV), the previously pressed area regained its original flow properties. This means that, even on a previously pressed area, multiple valve operations can be successfully conducted. To the best of our knowledge, this is the first demonstration of an active and repetitive valve operation in paper microfluidics. Paper-based cultures are an emerging platform for preparing 3D tissue-like structures. Chemical gradients can be imposed upon these cultures, generating microenvironments similar to those found in poorly vascularized tumors. There is increasing evidence that the tumor microenvironment is responsible for promoting drug resistance and increased invasiveness. Acidosis, or the acidification of the extracellular space, is particularly important in promoting these aggressive cancer phenotypes. To better understand how cells respond to acidosis there is a need for 3D culture platforms that not only model relevant disease states but also contain sensors capable of quantifying small molecules in the extracellular environment. In this work, we describe pH-sensing optodes that are capable of generating high spatial and temporal resolution maps of pH gradients in paper-based cultures. This sensor was fabricated by suspending microparticles containing pH-sensitive (fluorescein) and pH-insensitive (diphenylanthracene) dyes in a polyurethane hydrogel, which was then coated onto a transparent film. The pH-sensing films have a fast response time, are reversible, stable in long-term culture environments, have minimal photobleaching, and are not cytotoxic. These films have a pK[sub a] of 7.61 ± 0.04 and are sensitive in the pH range corresponding to normal and tumorigenic tissues. In this work, we demonstrate a novel design that allows rapid online identification and quantitation of unsaturated fatty acid C=C location isomers via epoxidation and ambient mass spectrometry (MS). Unsaturated fatty acid solution was loaded on a paper strip placed between a low-temperature plasma probe and the inlet of a mass spectrometer. Reactive oxygen species in the plasma promoted epoxidation at the C=C, and the product was simultaneously ionized. Upon collision-induced dissociation (CID), the epoxidation product was fragmented to release diagnostic ions specific to the C=C location. The whole analytical workflow can be completed within 5 s and is particularly promising for point-of-care (POC) clinical diagnosis, considering its fast, high-throughput nature, and coupling with paper-based analytical devices. We report a quantitative analytical method using the naked eye. The method is based on the coffee-ring effect on paper, which converts a colored solution to stains on the paper. It is found that the width of the colored stains is correlated to the concentration of analyte so that quantitative analysis can be accomplished using a ruler. The influence of the shape and lamination of the paper reservoir to the test results is studied to find the optimal one for unambiguous analysis. It is also found that the sample color does not cause interference to the test. The applicability of the method to quantitative chemical analysis is demonstrated by detection of glucose and lactate. Owing to the multiple potential health risks caused by exposure to BPA and the strengthened regulation of BPA-containing thermal papers, much concern has been raised recently regarding a BPA-type color developer in thermal papers. However, few efforts have so far been focused on the development of safer alternative materials to BPA. In this study, we examined the use of polymeric materials as a safer replacement for BPA in thermal papers. For this objective, a series of phenolic polymers, phenol-formaldehyde novolac and resole (PF-N and PF-R) and BPA-formaldehyde novolac and resole (BPAF-N and BPAF-R), were prepared. Among these polymers, a BPAF-N-type polymer was determined to be the most appropriate candidate for a developer material in thermal papers. Through printing tests of the prepared thermal papers, it was found that BPAF-N_8 polymer has a high static sensitivity and dark contrast of thermal papers similar to those of BPA, whereas the BPAF-N_16 and BPAF-N_30 polymers have lower static sensitivities than BPFA-N_8 and BPA. This work reports a microfabricated electrochemical paper-based analytical device (ePAD) for the voltammetric determination of DNA. The device is patterned by wax-printing on paper and features a circular assay zone connected to an inlet zone and a sink via grooved microfluidic channels for accelerated flow rate. An electrochemical cell with integrated electrodes is formed on the reverse side of the paper by sputtering of thin metal films (Sn, Pt and Ag as the working, counter and reference electrode, respectively). Proof-of-principle of the ePAD for biosensing is demonstrated for a DNA assay involving attachment of capture DNA, hybridization with biotinylated target oligonucleotide and labeling with streptavidin-conjugated CdSe/ZnS quantum dots (QDs). After the acidic dissolution of the QDs, the released Cd(II) is quantified by anodic stripping voltammetry (ASV) at the Sn-film working electrode. Thanks to the synergistic effects of QDs amplification, the inherent sensitivity of ASV and the excellent detection capabilities of the Sn-film working electrode for Cd(II), the target DNA can be detected at levels as low as 0.11 pmol L-1 usingsample volumes as low as 1 μL. The developed microfluidic ePAD costs only 0.11$ and presents favorable fabrication and operational features that make it an excellent candidate biosensor for simple and ultrasensitive point-of-need testing. Salmonella causes over a million foodborne illnesses per year in the United States resulting in more hospitalizations and deaths than any other foodborne bacterial pathogen. To help prevent outbreaks, a rapid, portable, sensitive, and reliable method for onsite detection of bacteria that can be used in different sample matrices would be beneficial. Herein, we present a colorimetric paper-based analytical device (PAD) combined with immunomagnetic separation (IMS) for detecting Salmonella typhimurium. IMS anti-Salmonella coated magnetic beads were applied to capture and separate bacteria from the sample matrix and preconcentrate it into small volumes before testing on paper. To directly detect S. typhimurium after IMS, a sandwich immunoassay was implemented into the procedure with β-galactosidase (β-gal) as the detection enzyme. Using the antibody/enzyme complex, we performed a colorimetric assay with chlorophenol red-β-D-galactopyranoside (CPRG) for bacteria quantification. The method was confirmed to be highly specific to S. typhimurium without interference from other pathogenic bacteria like Escherichia coli. Using this system, the limit of detection of S. typhimurium was found to be 102 CFU mL-1 in culturing solution without any pre-enrichment. In addition, distance-based detection where the concentration is read as the length of colored band formed on the reaction was also demonstrated. This assay had a detection limit of 102 CFU mL-1 for S. typhimurium, providing an instrument-free quantitative analysis alternative to spot tests, which require image analysis. Finally, the proposed platform was applied for detection of S. typhimurium in inoculated Starling bird fecal samples and whole milk with detection limits of 105 CFU g-1 and 103 CFU mL-1, respectively, and this is the first published paper-based detection method for S. typhimurium in bird feces and whole milk. In this work, a disposable paper -- plastic hybrid microfluidic lab-on-a-chip (LOC) has been developed and successfully applied for the colorimetric measurement of urine by the smartphone-based optical platform using a "Urine-Analysis" Android app. The developed device was cost-effectively implemented as a stand-alone hybrid LOC by incorporating the paper-based conventional reagent test strip inside the plastic-based LOC microchannel. The LOC device quantitatively investigated the small volume (40 μL) of urine analytes for the colorimetric reaction of glucose, protein, pH, and red blood cell (RBC) in integration with the finger-actuating micropump. On the basis of our experiments, the conventional urine strip showed large deviation as the reaction time goes by, because dipping the strip sensor in a bottle of urine could not control the reaction volume. By integrating the strip sensor in the LOC device for urine analysis, our device significantly improves the time-dependent inconstancy of the conventional dipstick-based urine strip, and the smartphone app used for image analysis enhances the visual assessment of the test strip, which is a major user concern for the colorimetric analysis in point-of-care (POC) applications. As a result, the user-friendly LOC, which is successfully implemented in a disposable format with the smartphone-based optical platform, may be applicable as an effective tool for rapid and qualitative POC urinalysis.
