Despite breakthroughs in modern medical care, the incidence of cardiovascular disease (CVD) is even more prevalent globally. Increasing epidemiologic evidence indicates that emerging cardiovascular risk factors arising from the modern lifestyle, including psychosocial stress, sleep problems, unhealthy diet patterns, physical inactivity/sedentary behavior, alcohol consumption, and tobacco smoking, contribute significantly to this worldwide epidemic, while its underpinning mechanisms are enigmatic. Hematological and immune systems were recently demonstrated to play integrative roles in linking lifestyle to cardiovascular health. In particular, alterations in hematopoietic stem cell (HSC) homeostasis, which is usually characterized by proliferation, expansion, mobilization, megakaryocyte/myeloid-biased differentiation, and/or the pro-inflammatory priming of HSCs, have been shown to be involved in the persistent overproduction of pro-inflammatory myeloid leukocytes and platelets, the cellular protagonists of cardiovascular inflammation and thrombosis, respectively. Furthermore, certain lifestyle factors, such as a healthy diet pattern and physical exercise, have been documented to exert cardiovascular protective effects through promoting quiescence, bone marrow retention, balanced differentiation, and/or the anti-inflammatory priming of HSCs. Here, we review the current understanding of and progression in research on the mechanistic interrelationships among lifestyle, HSC homeostasis, and cardiovascular health. Given that adhering to a healthy lifestyle has become a mainstream primary preventative approach to lowering the cardiovascular burden, unmasking the causal links between lifestyle and cardiovascular health from the perspective of hematopoiesis would open new opportunities to prevent and treat CVD in the present age. Carbon trading risk management and policy making require accurate forecasting of carbon trading prices. Based on the sample of China's carbon emission trading pilot market, this paper firstly uses the Augmented Dickey–Fuller test and Autoregressive conditional heteroscedasticity model to test the stationarity and autocorrelation of carbon trading price returns, uses the Generalized Autoregressive Conditional Heteroscedasticity family model to analyze the persistence, risk and asymmetry of carbon trading price return fluctuations, and then proposes a hybrid prediction model neural network (generalized autoregressive conditional heteroscedasticity–long short-term memory network) due to the shortcomings of GARCH models in carbon price fluctuation analysis and prediction. The model is used to predict the carbon trading price. The results show that the carbon trading pilots have different degrees of volatility aggregation characteristics and the volatility persistence is long, among which only the Shanghai and Beijing carbon trading markets have risk premiums. The other pilot returns have no correlation with risks, and the fluctuations of carbon trading prices and returns are asymmetrical. The prediction results of different models show that the root mean square error (RMSE) of Hubei, Shenzhen and Shanghai carbon trading pilots based on the GARCH-LSTM model is significantly lower than that of the single GARCH model, and the RMSE values are reduced by 0.0006, 0.2993 and 0.0151, respectively. The RMSE in the three pilot markets improved by 0.0007, 0.3011 and 0.0157, respectively, compared to the standalone LSTM model. At the same time, compared with the single model, the GARCH-LSTM model significantly increased the R^2 value in Hubei (0.2000), Shenzhen (0.7607), Shanghai (0.0542) and Beijing (0.0595). Therefore, compared with other models, the GARCH-LSTM model can significantly improve the prediction accuracy of carbon price and provide a new idea for scientifically predicting the fluctuation of financial time series such as carbon price. Meat product is the main food and major source of daily protein intake. Polyphenols are always introduced into many meat products during processing. Some complex interactions may occur between polyphenol and meat protein during the processing, especially thermal processing, which may affect the digestion of protein. In this experiment, chicken protein and tea polyphenol were interacted in simulated systems to explore the effects of the interaction between meat protein and polyphenols on the digestion of meat protein. The mechanism of tea polyphenol inhibiting chicken protein digestion was studied by analyzing the changes of chicken protein in intrinsic fluorescence, surface plasmon resonance (SPR), reactive sulfhydryl group, and solubility in different solvents. The results showed that the chicken protein digestion had a negative correlation with tea polyphenol concentration and interaction temperature, and the meat protein has a higher affinity to EGCG than protease. The mechanism of tea polyphenol inhibiting chicken protein digestion was related to the changing spatial structure of chicken protein and the decreasing activity of proteases. In the simulation system, at low-concentration tea polyphenol, the inhibition of the tea polyphenol on the digestibility of chicken protein might be mainly caused by the changes in chicken protein structure, while at high concentration, the changes in protein structure and the inhibition of proteases activity played a role together. This experiment revealed the effect and the mechanism of polyphenols on the digestion performance of meat protein and provide more references for the further application of polyphenols in meat processing. The Mahu conglomerate reservoir is characterized by strong heterogeneity and the uneven stimulation of the horizontal lateral during hydraulic fracturing. The optimization of the perforation number per cluster is of great value for horizontal well multi-stage fracturing (HWMF) because the suitable perforation number not only promotes the uniform propagation of multiple fractures but also prevents excessive perforation erosion. In this work, a typical well in the Mahu conglomerate reservoir was selected, and a field test of optimizing the perforation number was carried out. The perforation schemes of three, five, and eight perforations per cluster were designed in nine fracturing stages, respectively. The wellhead pressure under different perforation schemes was compared and analyzed with the step-down flow rate test, and the optimal perforation number per cluster in the Mahu conglomerate reservoir was selected as eight. The theoretical calculation results show that eight perforations per cluster can generate the perforation friction of 5 MPa, sufficient to overcome the mechanical property differences among multiple clusters within one stage. The downhole video technology shows that the perforation erosion area is the most uniform with the case of eight perforations per cluster. Moreover, the optical fiber monitoring results show that the perforation number of eight per cluster can realize the simultaneous initiation and uniform propagation of six fractures or five fractures within one stage. Additives and antibiotic abuse during food production and processing are among the key factors affecting food safety. The efficient and rapid detection of hazardous substances in food is of crucial relevance to ensure food safety. In this study, a water-soluble quantum dot with glutathione as a ligand was synthesized as a fluorescent probe by hydrothermal method to achieve the detection and analysis of H2O2. The detection limits were 0.61 μM in water and 68 μM in milk. Meanwhile, it was used as a fluorescent donor probe and manganese dioxide nanosheets were used as a fluorescent acceptor probe in combination with an immunoassay platform to achieve the rapid detection and analysis of enrofloxacin (ENR) in a variety of foods with detection limits of 0.05–0.25 ng/mL in foods. Myelosuppression is a common and intractable side effect of cancer therapies including radiotherapy and chemotherapy, while the underlying mechanism remains incompletely understood. Here, using a mouse model of radiotherapy-induced myelosuppression, we show that inorganic phosphate (Pi) metabolism is acutely inhibited in hematopoietic stem cells (HSCs) during irradiation-induced myelosuppression, and closely correlated with the severity and prognosis of myelosuppression. Mechanistically, the acute Pi metabolic inhibition in HSCs results from extrinsic Pi loss in the bone marrow niche and the intrinsic transcriptional suppression of soluble carrier family 20 member 1 (SLC20A1)-mediated Pi uptake by p53. Meanwhile, Pi metabolic inhibition blunts irradiation-induced Akt hyperactivation in HSCs, thereby weakening its ability to counteract p53-mediated Pi metabolic inhibition and the apoptosis of HSCs and consequently contributing to myelosuppression progression. Conversely, the modulation of the Pi metabolism in HSCs via a high Pi diet or renal Klotho deficiency protects against irradiation-induced myelosuppression. These findings reveal that Pi metabolism and HSC survival are causally linked by the Akt/p53–SLC20A1 axis during myelosuppression and provide valuable insights into the pathogenesis and management of myelosuppression. Red mud is a strong alkaline solid waste pollutant produced in the process of aluminum smelting, which causes great pollution to the regional groundwater environment due to its high content of fluorine and aluminum and high concentration of strong alkali. In this study, fluoride ion was selected as the model contaminant, and a numerical model of the groundwater flow field and solute transport was developed using GMS software to simulate and analyze the migration patterns of fluoride contaminants caused by the red mud pit for the fractured karst geohydrological conditions. The results demonstrated that the groundwater model and flow pattern were mainly controlled by atmospheric precipitation recharge, given flow boundary conditions and leakage of rivers and drains. When the concentration of fluorine pollutants in the red mud yard was 60.0 mg/L, the maximum migration distance of F− in the groundwater of the ordovician limestone aquifer was 473, 1160, 1595 and 1750 m after 1, 5, 10 and 15 years of bottom leakage, and the additional transport distances were 687, 435 and 155 m every 5 years, respectively. The range of F− pollution plume was 0.37 km2, 1.15 km2, 1.95 km2 and 2.14 km2, respectively and the range of newly added pollution plume was 0.78 km2, 0.80 km2 and 0.19 km2, respectively, every five years. Both indicated that with the extension of time, the migration and diffusion rate of pollutants slow down, and the diffusion volume increased first and then decreased. The F− pollution plume spread from the red mud pit to the northeast, which was consistent with the flow of groundwater. The high-concentration pollution plume was mainly distributed in the Ordovician limestone fractured aquifer in the northeast. This study revealed the migration law of red mud pollutants, and provided a scientific decision-making basis for the prevention and control of red mud groundwater pollution in the future. We report on a sequential control scheme to realize a steady, quasi-continuous output of squeezed vacuum states, which eliminates the influence of the seed beam on the squeezing strength. The scheme, originating from time-division multiplexing, separates the generation process from the locking process. We confirm that the sequential control scheme does not reduce the squeezing strength and that the setup operates stably for a 3-h running test, with a duty ratio of 80% and cycle time of 5 s. The western flower thrips, Frankliniella occidentalis, is an aggressive agricultural insect pest causing significant damage to a wide range of fruit, vegetable, and ornamental crops. Beauveria bassiana is a broad-host-range entomopathogenic fungus capable of infecting and killing F. occidentalis. Infection of thrips by B. bassiana strain BbYT12 using a concentration of 1 × 108 conidia/mL resulted in 81.48% mortality in adults in 6 d (LT50 = 90 ± 15.1 h). Scanning electron microscopy of the infection process revealed preferential adhesion and germination of fungal spores to inter-segmental folds or grooves on the insect body surface with penetrating germlings and extended hyphae visualized during the initial stages of infection (6–24 h). Histological analyses showed the appearance of in vivo hyphal bodies in sagittal sections and the fat body as early as 24 h post-infection. Within 72 h, hyphal bodies and hyphae could be found throughout the infected organism including in the midgut, Malphigian tubules, alimentary canal, ovarioles (in females), and an extended hyphal network could be seen on insect cadavers (>72 h post-infection). Real-time RT-PCR analyses of the expression of select genes implicated in virulence including the Pr1 protease, beauvericin synthase, involved in the production of the secondary metabolite beauvericin, two cytochrome P450 monooxygenases implicated in cuticular hydrocarbon degradation, two multidrug efflux proteins, a perilipin involved in lipid storage, and the Hog1 MAP kinase and protein kinase A signaling factors revealed discrete patterns of infection-time dependent expression. Protein is one of the three major macronutrients and is essential for health. The reaction of α-dicarbonyl compounds (α-DCs) with glutenin during heat processing can modify its structure, thereby reducing its digestibility. Furthermore, advanced glycation end products (AGEs) formed by the Maillard reaction are associated with long-term diabetes-related complications. In this study, we established a heat processing reaction system for α-DCs and glutenin by simulating common food processing conditions. An in vitro digestion model was used to study the digestibility of glycated glutenin; whereupon the effects of the digestion products on macrophage inflammatory response were further investigated. It was found that reaction conditions, including temperature, treatment duration, pH, and reactant mass ratio, can significantly affect the digestibility of glycation glutenin, in which the mass ratio of reactants has the most significant influence. We demonstrated that when the mass ratio of glutenin to methylglyoxal (MGO) was 1:3, the level of inflammation induced by glycated glutenin was the highest. The mass ratio of reactants significantly affects the digestibility of glycation glutenin and the level of macrophage-induced inflammatory response. Hematopoietic stem cells (HSCs) regularly produce various blood cells throughout life via their self-renewal, proliferation, and differentiation abilities. Most HSCs remain quiescent in the bone marrow (BM) and respond in a timely manner to either physiological or pathological cues, but the underlying mechanisms remain to be further elucidated. In the past few years, accumulating evidence has highlighted an intermediate role of inflammasome activation in hematopoietic maintenance, post-hematopoietic transplantation complications, and senescence. As a cytosolic protein complex, the inflammasome participates in immune responses by generating a caspase cascade and inducing cytokine secretion. This process is generally triggered by signals from purinergic receptors that integrate extracellular stimuli such as the metabolic factor ATP via P2 receptors. Furthermore, targeted modulation/inhibition of specific inflammasomes may help to maintain/restore adequate hematopoietic homeostasis. In this review, we will first summarize the possible relationships between inflammasome activation and homeostasis based on certain interesting phenomena. The cellular and molecular mechanism by which purinergic receptors integrate extracellular cues to activate inflammasomes inside HSCs will then be described. We will also discuss the therapeutic potential of targeting inflammasomes and their components in some diseases through pharmacological or genetic strategies. [ABSTRACT FROM AUTHOR]

Background: It is widely believed that Maillard reactions could affect the sensitization of allergens. However, the mechanism of action of methylglyoxal (MGO) production in Maillard reactions in the sensitization variation of glutenin (a predominant allergen in wheat) during heat processing is still unclear. Methods: This research evaluated the effect of MGO on the immune response against glutenin in a mouse model. The resulting variations in conformation and corresponding digestibility of glutenin were determined. The immune response and gut microflora variation in mice were analyzed following administering of glutenin and MGO-glutenin. Results: The results of the study showed that MGO-glutenin induced a lower immune response than native glutenin. Cytokine analysis showed that MGO-glutenin regulated mouse immune response by inducing Treg differentiation. MGO decoration changed the structure and digestibility of glutenin. In addition, MGO-glutenin contributes to the maintenance of the beneficial gut microflora. Conclusion: MGO decoration of glutenin during heat processing could alleviate the resulting allergic reaction in mice. Decoration with MGO appears to contribute to the aggregation of glutenin, potentially masking surface epitopes and abating sensitization. Furthermore, Bacteroides induced regulatory T-cell (Treg) differentiation, which may contribute to inhibition of the Th2 immune response and stimulation of immune tolerance. [ABSTRACT FROM AUTHOR]

We report the use of biochar and Fe3O4 nanoparticles as co-stabilizers for oil-in-water (o/w) Pickering emulsion. The emulsion is subsequently used to prepare magnetic tetracycline-imprinted biochar composite microspheres (MMIPMs) with good uniformity and high selectivity. The MMIPMs were characterized by scanning electron microscopy (SEM), Brunner-Emmet-Teller (BET) measurements, Fourier transform infrared spectrometry (FT-IR), X-ray diffraction (XRD), vibrating sample magnetometer (VSM) and thermogravimetry analysis (TGA). The adsorption properties of tetracycline to the MMIPMs were investigated using different adsorption experiments including adsorption kinetic experiment, equilibrium binding experiment, selectivity evaluation and competitive adsorption tests. The theoretical maximum adsorption capacity of the MMIPMs (15.45 mg g−1) was greater than that of the raw biochar (2.10 mg g−1) and non-imprinted biochar composite microspheres (3.39 mg g−1) for tetracycline. Further, the MMIPMs were used as adsorbent for magnetic solid phase extraction (SPE) for the extraction of tetracycline present in drinking water, milk, fish and chicken samples. Under optimal conditions, the results showed good recovery yield ranging from 88.41% to 106.29% with a relative standard deviation (RSD) ranging from 0.35% to 6.83%, respectively. Sensor-based phenotyping technologies may offer a non-destructive, high-throughput and efficient assessment of herbage yield (HY) to replace current inefficient phenotyping methods. This paper assesses the feasibility of combining normalised difference vegetative index (NDVI) from multispectral imaging and ultrasonic sonar estimates of plant height to estimate HY of single plants in a large perennial ryegrass breeding program. For sensor calibration, fresh HY (FHY) and dry HY (DHY) were acquired destructively, and plant height was measured at four dates each in 2017 and 2018 from a selected subset of 480 plants. Global multiple linear regression models based on K-fold and random split cross-validation methods were used to evaluate the relationship between observed vs. predicted HY. The coefficient of determination (R2) = 0.67–0.68 and a root mean square error (RMSE) between 5.43–7.60 g was obtained for the validation of predicted vs. observed DHY. The mean absolute error (MAE) and mean percentage error (MPE) ranged between 3.59–5.44 g and 22–28%, respectively. For the FHY, R2 values ranged from 0.63 to 0.70, with an RMSE between 23.50 and 33 g, MAE between 15.11 and 24.34 g and MPE between ~22% and 31%. Combining NDVI and plant height is a robust method to enable high-throughput phenotyping of herbage yield in perennial ryegrass breeding programs. [ABSTRACT FROM AUTHOR]

Perennial ryegrass (Lolium perenne L.) is one of the most important forage grass species in temperate regions of the world, but it is prone to having poor persistence due to the incidence of abiotic and biotic stresses. This creates a challenge for livestock producers to use their agricultural lands more productively and intensively within sustainable limits. Breeding perennial ryegrass cultivars that are both productive and persistent is a target of forage breeding programs and will allow farmers to select appropriate cultivars to deliver the highest profitability over the lifetime of a sward. Conventional methods for the estimation of pasture persistence depend on manual ground cover estimation or counting the number of surviving plants or tillers in a given area. Those methods are subjective, time-consuming and/or labour intensive. This study aimed to develop a phenomic method to evaluate the persistence of perennial ryegrass cultivars in field plots. Data acquisition was conducted three years after sowing to estimate the persistence of perennial ryegrass using high-resolution aerial-based multispectral and ground-based red, green and blue(RGB) sensors, and subsequent image analysis. There was a strong positive relationship between manual ground cover and sensor-based ground cover estimates (p < 0.001). Although the manual plant count was positively correlated with sensor-based ground cover (p < 0.001) intra-plot plant size variation influenced the strength of this relationship. We conclude that object-based ground cover estimation is most suitable for use in large-scale breeding programs due to its higher accuracy, efficiency and repeatability. In this work, a novel molecularly imprinted polymer (MIP) with water-soluble CdTe quantum dots (QDs) was synthesized by oil-in-water Pickering emulsion polymerization using whole Listeria monocytogenes as the template. Listeria monocytogenes was first treated by acryloyl-functionalized chitosan with QDs to form a bacteria–chitosan network as the water phase. This was then stabilized in an oil-in-water emulsion comprising a cross-linker, monomer, and initiator, causing recognition sites on the surface of microspheres embedded with CdTe QDs. The resulting MIP microspheres enabled selective capture of the target bacteria via recognition cavities. The target bacteria Listeria monocytogenes was detected. Scanning electron microscopy (SEM) characterization showed that the MIPs had a rough spherical shape. There was visual fluorescence detection via quenching in the presence of the target molecule, which offered qualitative detection of Listeria monocytogenes in milk and pork samples. The developed method simplified the analysis process and did not require any sample pretreatment. In addition, the fluorescence sensor provided an effective, fast, and convenient method for Listeria monocytogenes detection in food samples. [ABSTRACT FROM AUTHOR]

Increasing the yield of perennial forage crops remains a crucial factor underpinning the profitability of grazing industries, and therefore is a priority for breeding programs. Breeding for high dry matter yield (DMY) in forage crops is likely to be enhanced with the development of genomic selection (GS) strategies. However, realising the full potential of GS will require an increase in the amount of phenotypic data and the rate at which it is collected. Therefore, phenotyping remains a critical bottleneck in the implementation of GS in forage species. Assessments of DMY in forage crop breeding include visual scores, sample clipping and mowing of plots, which are often costly and time-consuming. New ground- and aerial-based platforms equipped with advanced sensors offer opportunities for fast, nondestructive and low-cost, high-throughput phenotyping (HTP) of plant growth, development and yield in a field environment. The workflow of image acquisition, processing and analysis are reviewed. The "big data" challenges, proposed storage and management techniques, development of advanced statistical tools and methods for incorporating the HTP into forage breeding systems are also reviewed. Initial results where these techniques have been applied to forages have been promising but further research and development is required to adapt them to forage breeding situations, particularly with respect to the management of large data sets and the integration of information from spaced plants to sward plots. (Tb0.6Y0.4)3Al5O12 transparent ceramics were successfully fabricated by solid-state reactive sintering using Tb4O7, Y2O3, and α-Al2O3 powders as raw materials. The effect of (Tb+Y)/Al ratio on microstructure evolution and densification process was investigated in detailed. The results showed that the grain growth kinetics were significantly affected by (Tb+Y)/Al ratio. Al-rich and Tb-rich phases appeared in part of the samples of different ratios. Particularly, excess aluminum increased the diffusing process, leading to a higher densification rate, while samples with excess terbium ratios displayed a smaller grain size and lower relative density. The optical quality was highly related to the amount of the secondary phase produced by different (Tb+Y)/Al ratios. Finally, (Tb0.6Y0.4)3Al5O12 transparent ceramics have been fabricated through pre-sintering in vacuum, followed by hot isostatic sintering (HIP), and the best transmittance of sample with a 4 mm thickness was approximately 78% at 1064 nm. An accurate calculation of the indicator diagram of a pumping unit is the key factor in analyzing the performance of an oilfield production and operation and in preparing and optimizing an oilfield development plan. Aiming at the problems of the poor stability of the conventional load-displacement sensor method and the wave equation method, owing to the influence of an alternating load on the force sensor and the difficulty in measuring the crank angle using the electrical parameter method, a new soft sensing method employing the input electrical parameters of the motor and the beam inclination has been proposed to obtain the indicator diagram. At first, this method is established based on the beam angle of the pumping unit, which is easily measured using the suspension point displacement mathematics calculation model and the torque factor. Subsequently, the electric motor input parameters, the parameters of the four-bar linkage, and the relationship between the polished rod load have been established. Finally, the motor and the beam angle of the measured electrical parameters have been substituted into the calculation of the suspension point displacement and load value and pull in accordance with the guidelines to eliminate the singularity mutation values. After processing the measured data through a Butterworth filter, the indicator diagram is obtained. The results of the engineering experiment and application show that the average relative error of the method is less than 3.95%, and the maximum relative error remains within 2% for 6 months, which verifies the stability of the soft sensing method.

This study describes the development of a reproducible and label-free surface plasmon resonance (SPR) immunosensor and its application in the detection of harmful enrofloxacin (ENRO) in animal-derived foods. The experimental parameters for the immunosensor construction and regeneration, including the pH value (4.5), concentration for coating ENRO-ovalbumin conjugate (ENRO-OVA) (100 μg·mL -1 ), concentration of anti-ENRO antibody (80 nM) and regeneration solution (0.1 mol·L -1 HCl) were evaluated in detail. With the optimized parameters, the proposed SPR immunosensor obtained a good linear response to ENRO with high sensitivity (IC 50 : 3.8 ng·mL -1 ) and low detection limit (IC 15 : 1.2 ng·mL -1 ). The proposed SPR immunosensor was further validated to have favorable performances for ENRO residue detection in typical animal-derived foods after a simple matrix pretreatment procedure, as well as acceptable accuracy (recovery: 84.3-96.6%), precision (relative standard deviation (n = 3): 1.8-4.6%), and sensitivity (IC 15 ≤ 8.4 ng·mL -1 ). Each SPR chip for analysis can be reused at least 100 times with good stability and the analysis cycle containing the steps of sample uploading/chip regeneration/baseline recovery can be completed within 6 min (one cycle) and auto-operated by a predetermined program. These results demonstrated that the proposed SPR immunosensor provided an effective strategy for accurate, sensitive, and rapid detection for ENRO residue, which has great potential for routine analysis of large numbers of samples for measuring different types of compounds., Competing Interests: The authors declare no conflict of interest.

Programmed cell death 4 (PDCD4) is one multi-functional tumor suppressor inhibiting neoplastic transformation and tumor invasion. The role of PDCD4 in tumorigenesis has attracted more attention and has been systematically elucidated in cutaneous tumors. However, the normal biological function of PDCD4 in skin is still unclear. In this study, for the first time, we find that tumor suppressor PDCD4 is uniquely induced in a cell density-dependent manner in keratinocytes. To determine the potential role of PDCD4 in keratinocyte cell biology, we show that knockdown of PDCD4 by siRNAs can promote cell proliferation in lower cell density and partially impair contact inhibition in confluent HaCaT cells, indicating that PDCD4 serves as an important regulator of keratinocytes proliferation and contact inhibition in vitro. Further, knockdown of PDCD4 can induce upregulation of cyclin D1, one key regulator of the cell cycle. Furthermore, the expression patterns of PDCD4 in normal skin, different hair cycles and the process of wound healing are described in detail in vivo, which suggest a steady-state regulatory role of PDCD4 in epidermal homeostasis and wound healing. These findings provide a novel molecular mechanism for keratinocytes' biology and indicate that PDCD4 plays a role in epidermal homeostasis.
