Motivation mRNA location corresponds to the location of protein translation and contributes to precise spatial and temporal management of the protein function. However, current assignment of subcellular localization of eukaryotic mRNA reveals important limitations: (1) turning multiple classifications into multiple dichotomies makes the training process tedious; (2) the majority of the models trained by classical algorithm are based on the extraction of single sequence information; (3) the existing state-of-the-art models have not reached an ideal level in terms of prediction and generalization ability. To achieve better assignment of subcellular localization of eukaryotic mRNA, a better and more comprehensive model must be developed. Results In this paper, SubLocEP is proposed as a two-layer integrated prediction model for accurate prediction of the location of sequence samples. Unlike the existing models based on limited features, SubLocEP comprehensively considers additional feature attributes and is combined with LightGBM to generated single feature classifiers. The initial integration model (single-layer model) is generated according to the categories of a feature. Subsequently, two single-layer integration models are weighted (sequence-based: physicochemical properties = 3:2) to produce the final two-layer model. The performance of SubLocEP on independent datasets is sufficient to indicate that SubLocEP is an accurate and stable prediction model with strong generalization ability. Additionally, an online tool has been developed that contains experimental data and can maximize the user convenience for estimation of subcellular localization of eukaryotic mRNA. Glutarylation is a post-translational modification which plays an irreplaceable role in various functions of the cell. Therefore, it is very important to accurately identify the glutarylation substrates and its corresponding glutarylation sites. In recent years, many computational methods of glutarylation sites have emerged one after another, but there are still many limitations, among which noisy data and the class imbalance problem caused by the uncertainty of non-glutarylation sites are great challenges. In this study, we propose a new semi-supervised learning algorithm, named FCCCSR, to identify reliable non-glutarylation lysine sites from unlabeled samples as negative samples. FCCCSR first finds core objects from positive samples according to reverse nearest neighbor information, and then clusters core objects based on natural neighbor structure. Finally, reliable negative samples are selected according to clustering result. With FCCCSR algorithm, we propose a new method named FCCCSR_Glu for glutarylation sites identification. In this study, multi-view features are extracted and fused to describe peptides, including amino acid composition, BLOSUM62, amino acid factors and composition of k-spaced amino acid pairs. Then, reliable negative samples selected by FCCCSR and positive samples are combined to establish models and XGBoost optimized by differential evolution algorithm is used as the classifier. On the independent testing dataset, FCCCSR_Glu achieves 85.18%, 98.36%, 94.31% and 0.8651 in sensitivity, specificity, accuracy and Matthew's Correlation Coefficient, respectively, which is superior to state-of-the-art methods in predicting glutarylation sites. Therefore, FCCCSR_Glu can be a useful tool for glutarylation sites prediction and FCCCSR algorithm can effectively select reliable negative samples from unlabeled samples. Roll-to-roll systems that include rotary components such as driven rolls and idle rollers have significant potential for application in fabrication of flexible functional devices. They are inexpensive, mass producible, and environmentally friendly; however, even minor defects in their component bearings can render them susceptible to severe damage, which necessitates accurate diagnoses of bearing quality. The main steps in machine learning for fault diagnosis include feature extraction and selection. In the case of high-dimensional feature data, critical study is required to identify the best feature combination for proper diagnosis. Thus, this study aims to develop a method that extracts fault characteristics of a bearing from the measured signal and qualify the bearing according to the Mahalanobis distances and differences in density between normal and faulty data groups. Features extracted from vibration data collected from industry-scale roll-to-roll systems and CWRU data were trained with principal component analysis, other modern feature selection techniques, and the proposed algorithm-based eight classifiers. Compared with the existing algorithm, the accuracy increased by up to 9.24%, the training time decreased by up to 34.46%, and the number of features to obtain the maximum accuracy decreased by up to 59.92%.
