Ruspolia differens (Serville) (Orthoptera: Tettigoniidae), also known as the 'edible grasshopper', 'African edible bush-cricket', and 'nsenene', is regarded as one of the most promising edible insect species that can be used for food, particularly in Sub-Saharan Africa. However, there is insufficient information on suitable diets and their effects on survival, adult weight, fecundity, and developmental time of this species, which are preconditions for large-scale production. In this study, we experimentally evaluated the effects of 12 diets (wheat bran, rice seed head, finger millet seed head, soya bran, maize bran, fresh maize comb, millet flour, chicken feed egg booster, simsim cake, sorghum seed head, powdered groundnut, and germinated finger millet), that are known to be accepted by R. differens , on their growth and reproductive parameters. The survival rate, developmental time, and adult weight varied considerably on the various diets. The highest nymphal survival rates, shortest development times, and highest adult weights were recorded for both sexes when fed fresh maize comb and germinated finger millet diet. Lifetime fecundity of females fed on germinated finger millet also was, on average, more than twice higher compared to other diets. The present study demonstrated that relatively inexpensive and locally available germinated finger millet, fresh maize seed (at the silking stage on the comb), sorghum seedhead, and finger millet seedhead could be successfully used to rear and sustain populations of R. differens. Cropping practice can affect pests and natural enemies. A three-year study of the strawberry tortricid, Acleris comariana (Lienig and Zeller) (Lepidoptera: Tortricidae), its parasitoid Copidosoma aretas Walker (Hymenoptera: Encyrtidae), and its entomopathogenic fungi was conducted in seven pairs of organic and conventional farms to test the hypothesis that farming practice (organic versus conventional) will affect the level of pest infestation and will affect the natural enemies. In addition, the number of years with strawberries on the farm, field age, and other factors that may affect pests and their natural enemies were considered. Farms were characterized by their cropping practices, cropping history, and other parameters. Field-collected larvae were laboratory reared to assess mortality from parasitoids and entomopathogenic fungi. In 2010, a survey of nematodes was made to assess the response of an unrelated taxonomic group to cropping practice. 2,743 larvae were collected. Of those, 2,584 were identified as A. comariana. 579 A. comariana were parasitized by C. aretas and 64 A. comariana were parasitized by other parasitoid species. Finally, 28% of the larvae and pupae of A. comariana died from unknown causes.Only two of the field-collected A. comariana larvae were infected by entomopathogenic fungi; one was infected by Isaria sp. and the other by Beauvaria sp. The density of A. comariana was on average four times lower in organic farms, which was significantly lower than in conventional farms. A. comariana was more dominant on conventional farms than on organic farms. The effect of crop age (One, two, or three years) on A. comariana infestation was significant, with higher infestations in older fields. Crop age had no effect on A. comariana infestation in a comparison of first- and second-year fields in 2010. Cropping practice did not lead to significant differences in the level of total parasitism or in C. aretas parasitism; however, C. aretas contributed to a higher proportion of the parasitized larvae on conventional farms than on organic farms. Plant parasitic nematodes were found in more samples from conventional farms than from organic farms. The low density of A. comariana in organic farms exposes the specialist C. aretas to a higher risk of local extinction. In organic farms, where the density of A. comariana is low, other parasitoids may play an important role in controlling A. comariana by supplementing C. aretas. Other tortricid species may serve as alternative hosts for these other parasitoids, contributing to conserving them in the habitat. The higher unknown mortality of larvae from organic fields may be the result of non-consumptive parasitoid or predator effects. This study reports an example of the effects of cropping practice on an insect pest, with similar effects on nematodes. Three DNA regions (ITS 1, LSU rRNA and GPD) of isolates from the insect-pathogenic fungus genus Entomophthora originating from different fly ( Diptera) and aphid ( Hemiptera) host taxa were sequenced. The results documented a large genetic diversity among the fly-pathogenic Entomophthora and only minor differences among aphid-pathogenic Entomophthora. The evolutionary time of divergence of the fly and the aphid host taxa included cannot account for this difference. The host-driven divergence of Entomophthora, therefore, has been much greater in flies than in aphids. Host-range differences or a recent host shift to aphid are possible explanations. Abstract: The toxicity of Bacillus thuringiensis subsp. israelensis to dipteran larvae (mosquitoes and black flies) depends on the presence of the pBtoxis plasmid. In this paper, two antibiotic resistance tagged pBtoxis were transferred by conjugation to other Bacillus cereus group strains. Among 15 potential recipients, only a lepidopteran active B. thuringiensis subspecies kurstaki and a B. cereus strain received the plasmid pBtoxis with a low transfer rate of about 10−8 transconjugants/recipient. The resulting B. thuringiensis subspecies kurstaki transconjugant was active to both lepidopteran and dipteran targets and the B. cereus transconjugant was active against dipteran insects. Phase contrast microscopy showed that the B. cereus transconjugants could produce only round crystalline inclusion bodies while B. thuringiensis subspecies kurstaki transconjugant could produce both round and bipyramidal crystals during sporulation. SDS–PAGE revealed that all the major mosquitocidal proteins from pBtoxis could express in the two transconjugants, including Cry4Aa, Cry4Ba, Cry10Aa, Cry11Aa and Cyt1Aa. However, none of the experiment showed any indications of mobilising abilities of pBtoxis. The limited number of strains, which could receive and maintain pBtoxis using a conjugational helper plasmid, indicates a very narrow host range of the B. thuringiensis subsp. israelensis pBtoxis plasmid. The plasmid pHT73 containing cry1Ac and tagged with an erythromycin resistance gene was transferred from Bacillus thuringiensis subspecies kurstaki KT0 to several Bacillus cereus group strains by conjugation. Sodium dodecyl sulfate–polyacrylamide gel electrophoresis (SDS–PAGE) and phase contrast microscopy showed that the transconjugants containing plasmid pHT73 could express Cry1Ac toxin and produce bipyramidal crystalline inclusion bodies during sporulation. The study demonstrated that pHT73 could be transferred to B. thuringiensis subsp. kurstaki, several B. cereus strains and Bacillus mycoides. Under non-selective conditions, the stability of the pHT73 plasmid in the transconjugants was found to be 58.2–100% after 100 generations and 4–96% after 200 generations. The fungus Metarhizium anisopliae is a facultative insect pathogen used as biological control agent of several agricultural pests worldwide. It is a dimorphic fungus that is able to display two growth morphologies, a filamentous phase with formation of hyphae and a yeast-like phase with formation of singlecelled blastospores. Blastospores play an important role for M. anisopliae pathogenicity during disease development. They are formed solely in the hemolymph of infected insects as a fungal strategy to quickly multiply and colonize the insect's body. Here, we use comparative genome-wide transcriptome analyses to determine changes in gene expression between the filamentous and blastospore growth phases in vitro to characterize physiological changes and metabolic signatures associated with M. anisopliae dimorphism. Our results show a clear molecular distinction between the blastospore and mycelial phases. In total 6.4% (n = 696) out of 10,981 predicted genes in M. anisopliae were differentially expressed between the two phases with a fold-change. 4. The main physiological processes associated with up-regulated gene content in the single-celled yeast-like blastospores during liquid fermentation were oxidative stress, amino acid metabolism (catabolism and anabolism), respiration processes, transmembrane transport and production of secondary metabolites. In contrast, the up-regulated gene content in hyphae were associated with increased growth, metabolism and cell wall re-organization, which underlines the specific functions and altered growth morphology of M. anisopliae blastospores and hyphae, respectively. Our study revealed significant transcriptomic differences between the metabolism of blastospores and hyphae. These findings illustrate important aspects of fungal morphogenesis in M. anisopliae and highlight the main metabolic activities of each propagule under in vitro growth conditions.
