Asymmetric tidal dynamics are of great significance for material transport and morphological evolution in estuaries. The tidal dynamics of the macro-tidal Hangzhou Bay (HZB) are characterized by flood-ebb asymmetries, spring-neap asymmetries, surface-bottom asymmetries, and up-downstream asymmetries. The mechanisms of spatio-temporal asymmetric tides and lateral flows in HZB were studied through a fully calibrated three-dimensional numerical model. The results show that tidal tides, tidal currents, and tidal asymmetry in HZB varied specially and temporally. In general, the bay was mostly flood-dominant. Temporally, tidal duration asymmetry in the bay fluctuated between spring and neap tides, with larger skewness during spring tides and smaller skewness during neap tides. The locally produced overtides are the primary sources of shallow-water tides in the bay, and the interaction between the lunar semi-diurnal tide M2 and the solar semi-diurnal tide S2 generates shallow-water overtides and deforms tidal asymmetries. The dissipated tidal energy may consumed by the bottom friction, with less passed to the generated shallow water overtides M4 and M6 tides (AM4 = 12.07 cm, AM6 = 3.91 cm) when comparing with the experiments that open boundary is purely forced by M2 tide (AM4 = 13.63 cm, AM6 = 6.31 cm). The increased depth reduces the bottom friction and the convergence of volume, resulting in skewness values close to zero (γTDA = 0.220, γM2-M4 = 0.141, γM2-M4-M6 = −0.002, γM2-S2-MS4 = 0.105). The changes of tidal duration asymmetry caused by the increased channel convergence, reduced bay width, and reclaimed intertidal zone spatially vary in different parts. The bottom friction contributed to the generation of the shallow-water tides and asymmetries in the bay (the RVRs for M4 and M6 are −73.5% and −92.5%), while the Coriolis force (the RVRs for M4 and M6 are 4.8% and 8.9%) and nonlinear advection (the RVRs for M4 and M6 are −17.3% and − 21.8%) are minor factors. Purpose: To describe the clinical and sonographic features of ovarian benign Brenner tumor (BBT) and malignant Brenner tumor (MBT), and to compare performance of four diagnostic models in differentiating them. Methods: Fifteen patients with BBTs and nine patients with MBTs were retrospectively identified in our institution from January 2003 and December 2021. One ultrasound examiner categorized each mass according to ovarian-adnexal reporting and data system (O-RADS), international ovarian tumor analysis (IOTA) Simple Rules Risk (SR-Risk) assessment and assessment of different neoplasias in the adnexa (ADNEX) models with/without CA125. Receiver operating characteristic curves were generated to compare diagnostic performance. Results: Patients with MBT had higher CA125 serum level (62.5% vs. 6.7%, P = 0.009) and larger maximum diameter of lesion (89 mm vs. 43 mm, P = 0.009) than did those with BBT. BBT tended to have higher prevalence of calcifications (100% vs. 55.6%, P = 0.012) and acoustic shadowing (93.3% vs. 33.3%, P = 0.004), and lower color scores manifesting none or minimal flow (100.0% vs. 22.2%, P < 0.001). Areas under curves of O-RADS, IOTA SR-Risk and ADNEX models with/without CA125 were 0.896, 0.913, 0.892 and 0.896, respectively. There were no significant differences between them. Conclusion: BBTs are often small solid tumors with sparse color Doppler signals, which contain calcifications with posterior acoustic shadowing. The most common pattern of MBT is a large multilocular-solid or solid mass with irregular tumor borders, and most were moderately or richly vascularized at color Doppler. Friction stir welded (FSWed) AZ31 Mg alloy joints were prepared at two different rotation speeds, namely, 150 rpm and 500 rpm, and are referred to as FSW-150 and FSW-500, respectively. The microstructure of the FSW joints was characterized and the local strain evolution in the tensile tests was monitored by a high-resolution digital image correlation method. The results show that the stir zone (SZ) of both FSW joints showed an equiaxed grain structure and the thermal-mechanically affected zone (TMAZ) had a gradually varied texture in FSW-150, compared with the relatively stable texture in FSW-500. A basin-shaped SZ was formed in FSW-150, while a cylindrical-shaped SZ was formed in FSW-500. Due to the different shape of SZ, the strain was gradually increased in TMAZ through the thickness of the FSW-500 specimen in tensile tests. However, the FSW-150 specimen showed an inhomogeneous strain distribution through the thickness, which resulted in the shear tensile stress to be localized at the TMAZ/SZ boundary and early fracture of the tensile specimen. Finally, the FSW-500 specimen showed an improved tensile strength and elongation of 250 MPa and 17%, compared with the 210 MPa and 6% for the FSW-150 counterpart. [ABSTRACT FROM AUTHOR]

Sauerkraut is a traditionally fermented cabbage, with a unique taste and beneficial properties, in northeast China. The taste and flavor of sauerkraut vary from region to region, owing to the differences in microorganisms. Illumina MiSeq sequencing was used to identify and quantify the microbial community composition of the broth and leaves of the naturally fermented Suan-cai collected from northeast China. The alpha and beta diversity of the samples from three areas in Heilongjiang province shown that the complexity of bacterial diversity of the three samples was C, A and B in turn. The Lactobacillus widely existed in fermented sauerkraut, of these, Latilactobacillus sakei, Loigolactobacillus coryniformis subsp. torquens, Lactiplantibacillus plantarum subsp. plantarum, and Secundilactobacillus malefermentans were more abundant in the sauerkraut leaves than in fermentation broth. Other genera of lactic acid bacteria Pediococcus and Leuconostoc, which have potential probiotic properties, were also present. A high-efficiency propeller can enable a long mission duration for autonomous underwater vehicles (AUVs). In this study, a new method with OpenProp coupled with computational fluid dynamics was developed to design a propeller for an Explorer100 AUV. The towed system simulation of the AUV was used to measure the nominal wake, and a self-propulsion simulation was used to measure the effective wake at the disc plane just in front of a propeller. Two propellers referring to the nominal wake (propeller 1) and effective wake (propeller 2) were designed with OpenProp and appended with the AUV for self-propulsion simulations, respectively. Through the numerical simulation of the AUV self-propulsion tests, the cruising velocity of AUV was obtained. The flow characteristics of the self-propulsion in pressure and velocity contours were also analyzed. The propeller designed with an effective wake improved the thrust, velocity, and efficiency by approximately 11.3%, 6.7%, and 2.5%, respectively, as compared with those with a nominal wake. The cruising velocity of the final designed propeller for the Explorer100 AUV improved by 21.8%, as compared to that of the original propeller from the AUV free-running tests. [ABSTRACT FROM AUTHOR]

It was suggested that the embryo pooling was an alternative for patients with insufficient number of embryos for preimplantation genetic testing (PGT) in a single ovarian stimulation cycle. However, limited study noticed whether it is an efficient strategy to pool cleavage-stage embryos by vitrification. This study included 71 cycles with vitrified-warmed and fresh embryos simultaneously for PGT between May 2016 and May 2021. The embryos from the same patients were split into two groups based on the origin: warming group and fresh group. Embryo development, sequencing results, clinical and neonatal outcomes were compared. The results showed that the rate of high-quality embryos in the warming group was significantly higher than that in the fresh group (64.53% versus 52.61%, P = 0.011); however, the available blastocyst rate in this group was significantly lower than that in the fresh group (47.29% versus 57.83%, P = 0.026). There were 96 and 144 blastocysts that underwent trophectoderm (TE) biopsy in warming and fresh groups, respectively. The high-quality blastocyst rate was significantly lower in the warming group compared to the fresh group (57.29% versus 70.14%, P = 0.041). The rates of genetic transferable blastocyst were comparable between the two groups (P = 0.956). There were no statistical differences in terms of embryo implantation, clinical pregnancy, miscarriage rates, and neonatal outcomes between the two groups. In conclusion, this study demonstrated that the cleavage-stage embryo pooling strategy might be unfavorable for the maintenance of embryonic development potential. In this study, a series of high purity Mg coating on Mg-2Zn-0.46Y-0.5Nd alloys were successfully prepared by friction stir processing technique at different tool travelling speeds. For all the samples, the high purity Mg coating were strongly bonded with the Mg-2Zn-0.46Y-0.5Nd alloy substrate, and no intermetallic compounds were found at the interface. Along the interface, a mixed structure consisting of coarse pure Mg and fine Mg-2Zn-0.46Y-0.5Nd grain structure were formed due to the upward flowing of the materials from the substrate. The corrosion current density of the high purity Mg coated Mg-2Zn-0.46Y-0.5Nd alloy in 3.5% NaCl solution reached 4.8063 × 10−6 A/cm2, which was much lower than that of the friction stir processed Mg-2Zn-0.46Y-0.5Nd alloy without coating (2.0139 × 10−5 A/cm2). In the high purity Mg coating, galvanic corrosion was not easy to form, which greatly improved the corrosion resistance of the substrate. [ABSTRACT FROM AUTHOR]

We investigated the copy number variation (CNV) of PDGFRA pathway across all common cancer types as well as its clinical relevance. This study included a total of 10,678 patients with pan-cancerous species involving 33 types of cancers and patient information was obtained from The Cancer Genome Atlas. According to the PDGFRA pathway CNV, all samples were divided into copy number gain (CN gain) group and No CN gain group. The analysis of loss of heterozygosity (LOH) fraction, CNV burden, tumor mutation burden (TMB), and the number of immunogenic mutations were performed, as well as the correlation analysis of PDGFRA pathway CN gain with tumor-related signaling pathways and tumor-infiltrating immune cell subpopulations. The results showed that CN gain of PDGFRA pathway in the cancer patients was associated with significantly shorter overall survival. The CN gain of PDGFRA pathway was identified as a prognostic risk factor for some tumors. CN gain was accompanied by an altered percentage of LOH, CNV burden, TMB, the number of immunogenic mutations were increased and tumor-infiltrating immune cell subpopulations were less. While certain tumor-related signaling pathways, such as hypoxia, cell cycle, DNA repair, and epithelial-mesenchymal transition were more enriched in the CN gain group, quiescence, and inflammation pathways were more enriched in the No CN gain group. Immunotherapy has emerged as an effective therapeutic strategy for various cancers, including colorectal cancer (CRC), but only a subset of MSI-H patients can benefit from such therapy. Patched1 (PTCH1) is a frequently altered gene in CRCs and its mutations contribute to unregulated Hedgehog (Hh) signaling. In the study, we evaluated the association of PTCH1 mutations with CRC immunity based on our single-center cohort and multiple cancer genomic datasets. Among 21 enrolled patients, six (28.6%) harbored a PTCH1 mutation based on WES analyses. In CRC patients, the PTCH1 mutation subgroup experienced a higher durable clinical benefit rate than the PTCH1 wild-type subgroup (100% vs. 40%, P = 0.017). In addition, patients with the PTCH1 mutation experienced greater progression-free survival (PFS, P = 0.037; HR, 0.208) and overall survival (OS, P = 0.045; HR, 0.185). A validation cohort from the MSKCC also confirmed the correlation between PTCH1 mutation and better prognosis (P = 0.022; HR, 0.290). Mechanically, diverse antitumor immune signatures were more highly enriched in PTCH1-mutated tumors than in PTCH1 wild-type tumors. Furthermore, PTCH1-mutated tumors had higher proportions of CD8 + T cells, activated NK cells, and M1 type macrophage infiltration, as well as elevated gene signatures of several steps in the cancer-immunity cycle. Notably, the PTCH1 mutation was correlated with tumor mutational burden (TMB), loss of heterozygosity score, and copy number variation burden. Immune checkpoint blockade (ICB) of the programmed cell death 1/programmed cell death ligand 1 (PD-1/PD-L1) immune checkpoint pathway has led to unprecedented advances in cancer therapy. However, the overall response rate of anti-PD-1/PD-L1 monotherapy is still unpromising, underscoring the need for predictive biomarkers. In this retrospective study, we collected pretreatment plasma samples from two independent cohorts of patients receiving ICB. To determine whether a signature of plasma cytokines could be associated with therapeutic efficacy, we systemically profiled cytokine clusters and functional groups in the discovery and validation datasets by using 59 multiplexed bead immunoassays and bioinformatics analysis. We first attempted to functionally classify the 59 immunological factors according to their biological classification or functional roles in the cancer-immunity cycle. Surprisingly, we observed that two signatures, the "checkpoint signature" and "trafficking of T-cell signature", were higher in the response subgroup than in the nonresponse subgroup in both the discovery and validation cohorts. Moreover, enrichment of the "checkpoint signature" was correlated with improved overall survival and progression-free survival in both datasets. In addition, we demonstrated that increased baseline levels of three checkpoint molecules (PD-L1, T-cell immunoglobulin mucin receptor 3 and T-cell-specific surface glycoprotein CD28) were common peripheral responsive correlates in both cohorts, thus rendering this "refined checkpoint signature" an ideal candidate for future verification. Purpose: This study aimed to analyze the impact of different biopsy protocols on the rate of mosaic blastocysts. Methods: This is a retrospective cohort study which included 115 cycles with pre-implantation genetic testing for aneuploidy (PGT-A). Two groups were allocated based on the biopsy protocols: method 1 group, the zona pellucida (ZP) was drilled on day 3 embryos followed by trophectoderm (TE) biopsy; and method 2 group, the ZP was opened on day 5 or 6 blastocysts followed by TE biopsy. All biopsy samples were assessed using next-generation sequencing (NGS) at a single reference laboratory. The euploid, aneuploid, and mosaic blastocyst rates and clinical outcomes were compared. Results: The mosaicism rate in the method 1 group was 19.58%, significantly higher than the method 2 group (8.12%; P < 0.05). No statistically significant difference was observed in euploid, aneuploid blastocyst rates, and clinical pregnancy rates between the two groups. Logistic regression analysis indicated that the biopsy protocols were independently associated with the mosaicism rates among all the variables. Conclusions: The present study showed that different biopsy protocols may have an impact on the mosaic blastocyst rate. The enrichment of innate immune cells and the enhanced inflammation represent the hallmark of non-alcoholic steatohepatitis (NASH), the advanced subtype with a significantly increased risk of progression to end-stage liver diseases within the spectrum of non-alcoholic fatty liver disease. Neutrophils are traditionally recognized as key components in the innate immune system to defend against pathogens. Recently, a growing body of evidence supports neutrophils as emerging key player in mediating the transition from steatosis to NASH, which is largely inspired by the histological findings in human liver biopsy indicating the enhanced infiltration of neutrophils as one of the key histological features of NASH. In this review, we discuss data regarding histological perspectives of hepatic infiltration of neutrophils in NASH. We also highlight the pathophysiological role of neutrophils in promoting metabolic inflammation in the liver through the release of a vast array of granule proteins, the interaction with other pro-inflammatory immune cells, and the formation of neutrophil extracellular traps. Neutrophil granule proteins possess pleiotropic effects on regulating neutrophil biology and functions. A variety of granule proteins (including lipocalin-2, myeloperoxidase, proteinase 3, neutrophil elastase, etc.) produced by neutrophils enhance liver metabolic inflammation, thereby promoting NASH progression by mediating neutrophil-macrophage interaction. Therapeutically, pharmacological inhibitors targeting neutrophil granule proteins hold promise to combat NASH. Micro/nanobubbles play an essential role in ultrasound-based biomedical applications. Here, a green and simple method to fabricate micro/nanobubbles was developed by the temperature-regulated self-assembly of lipids in the presence of free bubbles. The self-assembly mechanism of lipids interacting with gas-water interfaces was investigated, and the ultrasound imaging of the obtained lipid-encapsulated bubbles (LBs) was further confirmed. Above the phase transition temperature (Tm), fluid lipids transform from vesicles to micelles, and further assemble to the free bubbles interface to be a compressed monolayer, resulting in lipid shelled microbubbles. Cooling below Tm induces the lipid shell to glassy state and stables the LBs. Moreover, increasing the 1,2-distearoyl-sn-glycero-3-phosphoethanolamine-N-[methoxy(polyethylene glycol)-2000] (DSPE-PEG2K) content in lipids formulation can further manipulate the shell curvature and reduce the LBs size into nanobubbles. LBs with diameters of 1.68 ± 0.11 µm, 704 ± 7 nm and 208 ± 6 nm were successfully prepared. The in vitro and in vivo ultrasound imaging results showed that all of the LBs had excellent echogenicity. The nanosized LBs revealed elongated imaging duration time and greater microvascular details for the liver tissue. Avoiding the organic solvent and complicated multiple preparation process, this method has great potential in construction of various multifunctional micro/nanobubbles with size control for theranostic applications. Objectives: To establish methodological criteria for the clinical application of E-cervix technology in non-pregnant women, evaluate cervical differences between non-pregnant CI patients and normal women, and explore the predictive value of E-cervix for CI in non-pregnant women.Methods: In all, 46 non-pregnant women with CI and 46 non-pregnant women with full-term pregnancy experience were included. Elastic parameters (HR, IOS, EOS, IOS/EOS, CL) of the cervix were measured by transvaginal examination with a mid-sagittal section as the ROI, followed by examination of the whole cervix. The pressure of the probe on the cervix was divided into three levels. The elastic data of three consecutive images were separately obtained by two doctors for three pressure levels. Image stability and the effects of pressure were evaluated and the repeatability of the measurements was tested. In the case and control groups, cervical elasticity images were selected by standardized operation methods. Differences in the parameters between the two groups were determined and the diagnostic efficiency of each parameter was analyzed.Results: The HR, IOS, and EOS showed significant differences when measured on different sequential images obtained by continuous mapping (p < 0.05). The elastic parameters obtained from the first and last two images were significantly different (p < 0.05) and there were no significant differences between the elastic parameters obtained from the last two images (p > 0.05). The HR, IOS, EOS, and IOS/EOS showed significant differences among the pressure levels (p < 0.05), while the IOS showed a significant difference only between grades 0 and 2 (p < 0.05). E-cervix showed good repeatability within and among operators. The elastic parameters obtained by E-cervix in the CI group and control group were significantly different. ROC curve analysis showed that the HR was the most sensitive index for diagnosing CI.Conclusion: In E-cervix data acquisition, the first image should be excluded and the probe should not put pressure on the cervix. E-cervix has good retest and inter-observer reliability. Compared with normal women, CI patients showed a relatively softer and shorter cervix. This study explores change of precipitation and temperature using the Mann-Kendall test and the spatiotemporal variation of dryness/wetness using the standardized precipitation evapotranspiration index and empirical orthogonal function (EOF) analysis on 1-month time scales in Zhejiang Province, China, over 1971-2015. The results show that monthly precipitation had significant decreasing trends during April, May, September, and October, and significant increasing trends during November and December. Monthly temperatures had significant increasing trends in each month. Increasing temperature significantly increased drought events and intensity. There were consistent spatial patterns of dryness/wetness in Zhejiang. There were dryness trends in April, May, and September, a wetness trend in August, and no dryness/wetness pattern change in other months. The second EOF modes showed that dryness/wetness patterns were anti-phase between northern and southern Zhejiang during April-October. The proinflammatory factor high mobility group box protein 1 (HMGB1) has been implicated as an important mediator of many chronic inflammatory diseases, including asthma. Human bronchial epithelial cells (HBECs) play a central role in the pathogenesis of asthma. However, the effects of HMGB1 on HBECs and the underlying mechanisms remain unknown. Here, we investigated receptor expression and proinflammatory cytokine production by primary cultures of HBECs stimulated by HMGB1. We then examined the effects of specific receptor blockade and inhibition of p38 MAPK, ERK1/2, or PI3-K on HMGB1-induced expression of proinflammatory cytokines. HMGB1 increased the expression and secretion of TNF-α, TSLP, MMP-9, and VEGF in a dose- and time-dependent manner. HMGB1 also induced elevated expression of RAGE protein. Secretion of TNF-α, VEGF, MMP-9, and TSLP was significantly decreased by RAGE blockade and p38 MAPK pathway inhibition, while a less pronounced effect was mediated by ERK1/2 inhibition. These observations suggest that HMGB1 binds RAGE and promotes activities of p38 MAPK and ERK1/2 pathways in HBECs. This then enhances the expression of TNF-α, VEGF, MMP-9, and TSLP, which are the important inflammatory factors in asthma. Based on phenological and economic output models established and meteorological data from 1972 to 2013, changes in the phenology, frost risk, and economic output of the Longjing-43 tea tree in the Yuezhou Longjing tea production area of China were evaluated. As the local climate has changed, the beginning dates of tea bud and leaf plucking of this cultivar in all five counties studied has advanced significantly by −1.28 to −0.88 days/decade, with no significant change in the risk of frost. The main tea-producing stages in the tea production cycle include the plucking periods for superfine, grade 1, and grade 2 buds and leaves. Among the five bud and leaf grades, the economic output of the plucking periods for superfine and grade 1 decreased significantly, that for grade 2 showed no significant change, and those for grades 3 and 4 increased significantly. The economic output of large-area tea plantations employing an average of 45 workers per hectare and producing superfine to grade 2 buds and leaves were significantly reduced by 6,745-8,829 yuan/decade/ha, depending on the county. Those tea farmers who planted tea trees on their own small land holdings and produced superfine to grade 4 tea buds and leaves themselves experienced no significant decline in economic output. Chromosomal DNA replication is one of the central biological events occurring inside cells. Due to its large size, the replication of genomic DNA in eukaryotes initiates at hundreds to tens of thousands of sites called DNA origins so that the replication could be completed in a limited time. Further, eukaryotic DNA replication is sophisticatedly regulated, and this regulation guarantees that each origin fires once per S phase and each segment of DNA gets duplication also once per cell cycle. The first step of replication initiation is the assembly of pre-replication complex (pre-RC). Since 1973, four proteins, Cdc6/Cdc18, MCM, ORC and Cdt1, have been extensively studied and proved to be pre-RC components. Recently, a novel pre-RC component called Sap1/Girdin was identified. Sap1/Girdin is required for loading Cdc18/Cdc6 to origins for pre-RC assembly in the fission yeast and human cells, respectively. At the transition of G1 to S phase, pre-RC is activated by the two kinases, cyclindependent kinase (CDK) and Dbf4-dependent kinase (DDK), and subsequently, RPA, primase-polα, PCNA, topoisomerase, Cdc45, polδ, and polɛ are recruited to DNA origins for creating two bi-directional replication forks and initiating DNA replication. As replication forks move along chromatin DNA, they frequently stall due to the presence of a great number of replication barriers on chromatin DNA, such as secondary DNA structures, protein/DNA complexes, DNA lesions, gene transcription. Stalled forks must require checkpoint regulation for their stabilization. Otherwise, stalled forks will collapse, which results in incomplete DNA replication and genomic instability. Distributions of residual stresses and microstructure in the surface layers of shot-peened duplex stainless steel (DSS) S32205 were investigated. The results reveal that both compressive residual stresses (CRS) and microhardness increase with the enhancement of shot-peening (SP) intensity in the surface deformation layers. The maximum value of CRS of ferrite lies in the surface layer but that of austenite locates below the surface layer after SP. SP influence on the microstructure of DSS was studied using x-ray diffraction profiles, and the domain size and microstrain were calculated via Voigt method. After SP, the domain sizes are refined, and microstrain becomes severe at surface layers in both phases. On comparing the calculated results, it is found that the more evident domain size subdivision and the more serious microstrain increase in austenite than those in ferrite are due to the higher work hardening of austenite. This study adopts a loss assessment and indemnity approach for rice crops at risk of flooding in Yuhang County, Zhejiang Province. Employing a hydrological model for simulating floods and a reduction model for predicting rice yield, the relationship between the rate of reduction in the rice yield and precipitation is discussed. We argue that the yield reduction rate can be assessed according to the amount of precipitation and designed a weather-based indemnity index for agricultural insurance purposes in Zhejiang Province. With geographic information system technology, the yield reduction rate and weather-based indemnity index were refined and found to effectively reduce the shortcomings of traditional agricultural insurance, i.e., moral hazard, large error in assessing disaster loss and high basis risk. The validity of the method was verified by the amount of rice lost due to No. 16 typhoon Krosa in 2007, and the results show that the proposed method can well simulate the reduction rate of rice yield according to precipitation data.
