INTRODUCTION: Bumetanide, a loop diuretic, was identified as a candidate drug for repurposing for Alzheimer's disease (AD) based on its effects on transcriptomic apolipoprotein E signatures. Cross‐sectional analyses of electronic health records suggest that bumetanide is associated with decreased prevalence of AD; however, temporality between bumetanide exposure and AD development has not been established. METHODS: We evaluated Medicare claims data using Cox proportional hazards regression to evaluate the association between time‐dependent use of bumetanide and time to first AD diagnosis while controlling for patient characteristics. Multiple sensitivity analyses were conducted to test the robustness of the findings. RESULTS: We sampled 833,561 Medicare beneficiaries, 60.8% female, with mean (standard deviation) age of 70.4 (12). Bumetanide use was not significantly associated with AD risk (hazard ratio 1.05; 95% confidence interval, 0.99–1.10). DISCUSSION: Using a nationwide dataset and a retrospective cohort study design, we were not able to identify a time‐dependent effect of bumetanide lowering AD risk. Background: The Buffalo Concussion Treadmill Test (BCTT) is used to establish exercise tolerance for rehabilitation and identify injury subtypes for youth athletes after mild traumatic brain injury (mTBI). Its utility in adult community members is unknown. Objective: Primary: To describe how adults with and without mTBI tolerate the BCTT. Secondary: To explore relationships between baseline factors, mTBI‐related symptoms, and BCTT duration. Design: Prospective, observational, longitudinal. Setting: Academic medical center. Participants: Thirty‐seven adults treated in a level 1 trauma center emergency department with mTBI; 24 uninjured controls (UC). Interventions: N/A. Main Measures: Participants completed two visits 3 weeks apart (1 week and 1 month after mTBI) including a 15‐minute BCTT, the Rivermead Post Concussion Symptoms Questionnaire (RPQ), and preinjury International Physical Activity Questionnaire. Analyses characterized BCTT response and associations between baseline factors, RPQ scores, and BCTT duration. Results: Persons with mTBI discontinued earlier than UC at 1‐week postinjury using standard discontinuation criteria for exercise intolerance. The percentage of mTBI participants with signs of possible mTBI‐related intolerance was 55.6% at 1 week (36.1% for mTBI‐related symptom exacerbation, 19.4% for exertion/fatigue before reaching 85% of one's age‐predicted maximum heart rate [HR]) and 48.0% at 1 month (40.0% mTBI‐related symptom exacerbation, 8.0% exertion without reaching the target HR). Thirty percent of UCs completed the BCTT at both assessments. UCs met discontinuation criteria for increased nonspecific symptoms (eg, pain/general discomfort and increased Visual Analog Scale ratings; 39–61%) and physical exertion (9–26%). Shorter duration was associated with higher body mass index (r = −0.42 – −0.45), shorter height (r = 0.22–0.29), female gender (r = −0.26 – −0.27), and greater RPQ symptoms (r = −0.28 – ‐0.47). Conclusion: The BCTT exacerbates mTBI‐related symptoms in adult community members. Participant characteristics and noninjury factors influence performance. The findings imply the BCTT could be useful in clinical assessments of adults with mTBI. Interpretation should account for the unique characteristics of nonathletes. [ABSTRACT FROM AUTHOR]

Understanding the factors influencing species range limits is increasingly crucial in anticipating migrations due to human‐caused climate change. In the boreal biome, ongoing climate change and the associated increases in the rate, size, and severity of disturbances may alter the distributions of boreal tree species. Notably, Interior Alaska lacks native pine, a biogeographical anomaly that carries implications for ecosystem structure and function. The current range of lodgepole pine (Pinus contorta var. latifolia) in the adjacent Yukon Territory may expand into Interior Alaska, particularly with human assistance. Evaluating the potential for pine expansion in Alaska requires testing constraints on range limits such as dispersal limitations, environmental tolerance limits, and positive or negative biotic interactions. In this study, we used field experiments with pine seeds and transplanted seedlings, complemented by model simulations, to assess the abiotic and biotic factors influencing lodgepole pine seedling establishment and growth after fire in Interior Alaska. We found that pine could successfully recruit, survive, grow, and reproduce across our broadly distributed network of experimental sites. Our results show that both mammalian herbivory and competition from native tree species are unlikely to constrain pine growth and that environmental conditions commonly found in Interior Alaska fall well within the tolerance limits for pine. If dispersal constraints are released, lodgepole pine could have a geographically expansive range in Alaska, and once established, its growth is sufficient to support pine‐dominated stands. Given the impacts of lodgepole pine on ecosystem processes such as increases in timber production, carbon sequestration, landscape flammability, and reduced forage quality, natural or human‐assisted migration of this species is likely to substantially alter responses of Alaskan forest ecosystems to climate change. Rangelands are the dominant land use across a broad swath of central North America where they span a wide gradient, from <350 to >900 mm, in mean annual precipitation. Substantial efforts have examined temporal and spatial variation in aboveground net primary production (ANPP) to precipitation (PPT) across this gradient. In contrast, net secondary productivity (NSP, e.g., primary consumer production) has not been evaluated analogously. However, livestock production, which is a form of NSP or primary consumer production supported by primary production, is the dominant non‐cultivated land use and an integral economic driver in these regions. Here, we used long‐term (mean length = 19 years) ANPP and NSP data from six research sites across the Central Great Plains with a history of a conservative stocking to determine resource (i.e., PPT)–productivity relationships, NSP sensitivities to dry‐year precipitation, and regional trophic efficiencies (e.g., NSP:ANPP ratio). PPT–ANPP relationships were linear for both temporal (site‐based) and spatial (among site) gradients. The spatial PPT–NSP model revealed that PPT mediated a saturating relationship for NSP as sites became more mesic, a finding that contrasts with many plant‐based PPT–ANPP relationships. A saturating response to high growing‐season precipitation suggests biogeochemical rather than vegetation growth constraints may govern NSP (i.e., large herbivore production). Differential sensitivity in NSP to dry years demonstrated that the primary consumer production response heightened as sites became more xeric. Although sensitivity generally decreased with increasing precipitation as predicted from known PPT–ANPP relationships, evidence suggests that the dominant species' identity and traits influenced secondary production efficiency. Non‐native northern mixed‐grass prairie was outperformed by native Central Great Plains rangeland in sensitivity to dry years and efficiency in converting ANPP to NSP. A more comprehensive understanding of the mechanisms leading to differences in producer and consumer responses will require multisite experiments to assess biotic and abiotic determinants of multi‐trophic level efficiency and sensitivity. Including soil health and physical measurements with traditional soil fertility measurements has the potential to improve corn (Zea mays L.) P fertilizer recommendations. The objectives for this study were to (1) evaluate the accuracy of current South Dakota corn P recommendations in predicting yield increases and (2) determine if the predictability of yield response to P fertilization improves by including selected soil biological, physical, and chemical measurements. This project was conducted in central and eastern South Dakota from 2019 to 2022 at 117 experimental sites that varied in management, landform, and soil type. A treatment of 48.9 kg P2O5‐P ha−1 was compared to a control with no P fertilizer. Soil samples (0–15 cm) collected before fertilization were analyzed for selected soil physical, chemical, and biological parameters. Analysis suggested that the current critical P value of 16 mg Olsen‐P kg−1 was still accurate as it was still within the 68% confidence interval for the linear (12–19 mg kg−1) and quadratic plateau (16–26 mg kg−1) models. Using the critical value of 16 mg Olsen‐P kg−1, we correctly predicted P responsiveness 70% of the time. The accuracy of predicting P responsiveness was not further improved by considering selected enzymes, permanganate oxidizable C, soil respiration, total C, or total N, but was improved by considering pH, soil organic matter, and apparent cation exchange capacity. Therefore, the use of typical soil fertility measurements without soil physical and biological measurements is currently the best option for predicting the responsiveness of corn to P fertilization in South Dakota. Large‐scale tree planting on global rangelands is promoted as a natural climate solution (NCS), but there is little scientific evidence to support this narrative. The presumed benefits of rangeland afforestation originate from five major misconceptions: (1) conflation between reforestation and afforestation, (2) overestimation of carbon (C) sequestration potential, (3) insufficient recognition of rangeland ecosystem services, (4) potential for adverse ecological outcomes, and (5) neocolonial tendencies of afforestation programs. Rangeland afforestation possesses minimal potential for additional C storage, but it has high potential to reduce vital rangeland ecosystem services that benefit rangeland residents and non‐residents alike. Conservation of existing C—most of which is stored belowground, where it is less vulnerable to loss—may prove to be the most appropriate NCS for extensively managed rangelands. Stewardship strategies promoting rangeland multifunctionality will not only contribute to climate‐change mitigation but also support biodiversity conservation and sustainable production of high‐protein foods for marginalized populations. Introduction: The main mission of the Australian and New Zealand Children's Haematology and Oncology Group (ANZCHOG) is to develop and facilitate local access to the world's leading evidence‐based clinical trials for all paediatric cancers, including brain tumours, as soon as practically possible. Diffuse intrinsic pontine gliomas (DIPGs) — a subset of a larger group of tumours now termed diffuse midline glioma, H3K27‐altered (DMG) — are paediatric brain cancers with less than 10% survival at two years. In the absence of any proven curative therapies, significant recent advancements have been made in pre‐clinical and clinical research, leading many to seek integration of novel therapies early into standard practice. Despite these innovative therapeutic approaches, DIPG remains an incurable disease for which novel surgical, imaging, diagnostic, radiation and systemic therapy approaches are needed. Main recommendations: All patients with DIPG should be discussed in multidisciplinary neuro‐oncology meetings (including pathologists, neuroradiologists, radiation oncologists, neurosurgeons, medical oncologists) at diagnosis and at relapse or progression.Radiation therapy to the involved field remains the local and international standard of care treatment.Proton therapy does not yield a superior survival outcome compared with photon therapy and patients should undergo radiation therapy with the available modality (photon or proton) at their treatment centre.Patients may receive concurrent chemotherapy or radiation‐sensitising agents as part of a clinical trial.Biopsy should be offered to facilitate consideration of experimental therapies and eligibility for clinical trial participation.After radiation therapy, each patient should be managed individually with either observation or considered for enrolment on a clinical trial, if eligible, after full discussion with the family.Re‐irradiation can be considered for progressive disease. Changes in management as a result of the guideline: Every child diagnosed with DIPG should be offered enrolment on a clinical trial where available. Access to investigational drugs without biological rationale outside the clinical trial setting is not supported. In case of potentially actionable target identification with molecular profiling and absence of a suitable clinical trial, rational targeted therapies can be considered through compassionate access programs. Objective: We evaluated the effect of a 381‐mm minimum length limit (MLL) on saugeye (female Walleye Sander vitreus × male Sauger S. canadensis) population and fishery characteristics implemented in Ohio reservoirs in 2011. Methods: Using a before‐after–control‐impact study design (n = 6 control populations and n = 11 impact populations that received the MLL), we compared total CPUE, age‐2+ CPUE, proportional stock density‐harvestable (381‐mm), and length at age 2 (to index growth) determined from standardized gill‐net surveys and saugeye angler effort, catch rate, harvest rate, length of saugeye at harvest, yield, and satisfaction obtained from standardized creel surveys. Result: We found evidence that the 381‐mm MLL led to an increase in age‐2+ CPUE but had no effect on other saugeye population metrics. In the regulation reservoirs, harvest rates declined following MLL implementation. Total length of harvested saugeye did not increase significantly in response to the MLL, and combined with decreased harvest rates, yield was suppressed in the regulation reservoirs relative to the control reservoirs. However, over 90% of anglers reported that they were satisfied with the MLL during creel surveys. Conclusion: Despite marginal influence on saugeye populations and failure to increase yield, anglers supported the MLL. Our results suggest that the effects of harvest regulations may be marginal, and managers should consider the nuances of angler satisfaction in regulation decisions. Impact statementFisheries management agencies commonly use minimum length limits to attempt to improve recreational fisheries; however, robust evaluations of regulation success are often lacking. In saugeye fisheries, minimum length limits minimally affected population size structure and angler yield but still received strong angler support. Knowledge of how agricultural management interacts with weed seed banks and emergent weed communities is crucial for proactive weed management. Though studies have detailed how differences in disturbance and nutrient applications between organic and conventional herbicide‐based systems affect weed communities, few have focused on these same factors in contrasting organic systems. This study assessed the seed banks and emergent weed communities from the most recent crop rotation cycle (2017–2022) of a long‐term experiment, which compared four organic grain and forage cropping systems differing in nutrient inputs and soil disturbance. The high fertility (HF) system received high‐rate nutrient applications, low fertility (LF) received low‐rate applications, enhanced weed management (EWM) focused on weed control through frequent soil disturbance, and reduced tillage (RT) prioritized soil health with less intense or frequent soil disturbance. Soil samples for greenhouse germination assays were collected at the beginning (2017) and end (2022) of the rotation to explore how these four systems influenced seed bank dynamics over time. Weed community biomass was also sampled in each crop during this time. Treatment effects on weed abundance, taxonomic diversity, and community‐weighted means and functional dispersion of weed traits were analyzed with generalized mixed‐effect models. The RT system had the highest weed seed bank taxonomic diversity, and EWM had the lowest. RT and LF had higher functional dispersion of traits than HF in the seed bank. Weed seed bank communities in HF and RT were characterized by short, small‐seeded, and early germinating weed species. However, seed banks were also labile: Differences between systems in seed density and all other mean trait values were dependent on the crop, which preceded seed bank sampling. Likewise, differences among emergent weed communities in the four systems depended on an interaction between crop species and their planting year. Results suggest that resource availability and intensity of disturbance act as weed community assembly filters in organic cropping systems. Organic growers seeking to design systems that balance weed management and production goals can use relatively low soil disturbance and nutrient application to increase weed community taxonomic or functional diversity without necessarily increasing weed biomass or seed bank density. Objective: Flathead Catfish Pylodictis olivaris are a widespread aquatic invasive species within the United States and a recent invader in the Susquehanna River basin, Pennsylvania. Flathead Catfish are piscivores known to consume native and recreationally important fish species. In the mid‐Atlantic United States, it is unknown how this invader is impacting food webs and which species may be at greatest predation risk. To address this knowledge gap, we DNA barcoded stomach contents collected from Flathead Catfish to identify prey species and elucidate potential predatory impacts of Flathead Catfish in the Susquehanna River. Methods: We used a Bayesian hierarchical multivariate probit model to investigate if the probability of prey species occurrence in the diets of Flathead Catfish varied seasonally or was a function of Flathead Catfish length. Result: A total of 576 Flathead Catfish were collected during 2020–2021, with 241 individuals having recoverable stomach contents. In all, we identified 47 different prey species. The most common prey species were rusty crayfish Faxonius rusticus, Margined Madtom Noturus insignis, and shiners Notropis spp. While frequency of occurrence of prey species differed across Flathead Catfish length classes (<300 mm, 301–600 mm, 601–900 mm TL), rusty crayfish were commonly found (33.7–44.0% of diets) in stomachs of all size‐classes. Conclusion: We found that Flathead Catfish length and seasonality did influence occurrence probability differentially for several prey species. For example, Channel Catfish Ictalurus punctatus were more likely to appear in shorter Flathead Catfish while Smallmouth Bass Micropterus dolomieu appeared in larger Flathead Catfish. We demonstrate significant variation in Flathead Catfish predation, increasing our understanding of predator–prey dynamics, which is necessary to better manage and identify future impacts to aquatic communities in the Susquehanna River basin. Switchgrass (Panicum virgatum L.) is one of several grass species being bred for use as a biomass crop to support the biofuel industry. Increases in biomass yield are imperative to ensure that crops such as switchgrass can sustainably meet the needs of this industry. Genomic selection is one strategy that can accelerate breeding gains for complex traits such as biomass yield. The goal of this study was to conduct three cycles of genomic selection in a previously trained Liberty switchgrass population and compare that to one cycle of phenotypic selection, both of which required 3 years to complete. The advanced lines were tested across five locations and three hardiness zones in the Central United States using a randomized complete block design with four replicates. There were strong genotype × location interactions, but the first two generations of genomic selection were superior to Liberty at four of the five evaluation locations. Conversely, phenotypic selection failed to result in significant gains in biomass yield for any of the five evaluation locations. Ammonia (NH3) loss following manure application is an environmental concern and N loss for crop production. Manure injection typically reduces NH3 loss compared to surface application without incorporation but increases emissions of nitrous oxide (N2O), which is a potent greenhouse gas. Synchronizing manure in spring with cover crop (CC) growth may increase N recovery and reduce N2O emissions compared to applying manure later in the absence of growing crops. We compared the two following manure application methods: shallow-disk injection (IM) or surface banding without incorporation (BM) to annual ryegrass (Lolium multiflorum L.) and red clover (Trifolium pratense L.) CC at two times: early spring on growing CC (EARLY) and late spring on terminated CC (LATE). The randomized split-plot block experiment was conducted at Rock Springs, PA, during 2021–2022. After manure application, we measured NH3 for 72 h and N2O throughout the growing season. Aboveground CC biomass, N, and C:N ratio; pre-sidedress soil nitrate; corn (Zea mays L.) stalk nitrate; and silage yield were assessed. Averaged across application times, compared to BM, IM reduced cumulative NH3 loss, increased soil N, and resulted in 13% greater corn yield but increased yield-scaled N2O. Compared to BM LATE, BM EARLY reduced NH3 loss by 43%, increased CC N, reduced N2O emission by 50%, but decreased corn yield by 11%. When IM was EARLY compared to LATE, CC N increased 84%, cumulative N2O loss decreased 55%, and corn yield was similar. Injecting manure to growing CCs offers a strategy for reducing detrimental NH3 and N2O emissions and maintaining corn yield. [ABSTRACT FROM AUTHOR]

The current study was initiated to assess mungbean [Vigna radiata (L.) R. Wilczek] mutant lines for yield and their attributes in the mutation generation third (M3) generation using an augmented block design. Note that 1200 mutated mungbean lines were selected from the γ-irradiated (400–700 Gy) population and distributed into 21 blocks, along with three popular varieties (checks) replicated in each block. To evaluate mean performance, variance, and population genetics, the observations on days to flowering, days to maturity, plant height (cm), number of clusters per plant, number of pods per plant, pod length (cm), protein percentage, seed index, and seed yield (g) were measured and analyzed. All the lines under consideration showed highly significant variations for all the variables, except for days to flowering. Seventy-nine mutant lines were found to have significantly better yield attributes than checks and are currently being evaluated under station trial. Skewness and kurtosis analysis unveiled the presence of gene interactions, offering opportunities for targeted improvement and efficacy of γ rays as a mutagen, facilitating the release of variability within the population. Future mungbean breeding programs will benefit from the successful isolation of mutant plants with yield-enhancing traits, such as up to 20 clusters per plant, 71 pods per plant, 8.55 cm pod length, 26.69% protein content (5% higher than parent), and 29 gram seeds per plant. With an increase in the frequency of hot-dry-windy weather and reported significant increase in winter wheat (Triticum aestivum L.) in-season average temperatures, it is important to understand how soil management affects yield stability. This study quantified the combined effects of tillage and fertilizer-N on winter wheat yield stability in a wheat–sorghum [Sorghum bicolor (L.) Moench]–fallow crop rotation. The research was conducted from 1975 to 2022 and the experimental design was a randomized complete block with a split-split-plot arrangement. Crop phases were the main plots, tillage (conventional tillage [CT], reduced tillage [RT], and no-tillage [NT]) in the sub-plot, and N application rates (0, 22, 45, and 67 kg N ha−1 or 0, 45, 90, and 134 kg N ha−1) in the sub-sub-plot. Results showed winter wheat yield under NT increased by 0.8 Mg ha−1, compared with about 1.1 Mg ha−1 for CT and RT, when environmental average increased by 1 Mg ha−1 for N rates ≤90 kg ha−1. Yields with CT and RT increased by 0.8 Mg ha−1, compared with about 1.1 Mg ha−1 rate for NT, when environmental average increased by 1 Mg ha−1 for N >90 kg ha−1. In 30%–52% of the time, CT wheat yield was greater than NT, but yield advantage with CT decreased as fertilizer-N rate increases. Yield stability of winter wheat varied by tillage and fertilizer rate. We concluded that NT wheat required greater N rates to reach the same yield potential as CT and RT, and long-term CT or RT wheat at 45 kg N ha−1 was most stable [ABSTRACT FROM AUTHOR]

The maturation process of coffee (Coffea arabica) trees exhibits inherent variability, producing fruits at various physiological maturity stages. This variability affects the resistance between the fruit and its peduncle, posing a challenge in mechanized harvesting: non-selective harvesting. A precise classification of coffee fruit detachment force is essential to address this challenge, ensuring coffee’s quality and producer’s profitability. This study assesses the efficacy of machine learning (ML) models in determining the detachment force across various coffee cultivars under drip-irrigated and rainfed conditions. The dataset included detachment force measurements from 24 cultivars—13 drip-irrigated and 11 rainfed—yielding 1152 data points. Variance analysis compared irrigation methods and three maturity stages: green, cherry, and dry. Detachment force was categorized into four classes based on the dataset’s quartile distribution. The ML models utilized were random forest (RF), support vector machine (SVM), K-nearest neighbors, and artificial neural networks. The SVM model was notably effective in classifying detachment force for rainfed cultivars, with a Matthews correlation coefficient (MCC) of 0.78. In contrast, the RF model was particularly adept for drip-irrigated cultivars, with an MCC of 0.75. The highest classification accuracies were recorded for the extreme force classes I and IV, with precision values of 0.93 and 0.8, respectively, while classes II and III had lower precision at 0.57 and 0.69. The herbicide dicamba (3,6-dichloro-2-methoxybenzoic acid) is commonly used to control broadleaf weeds in soybeans. Dicamba, however, is susceptible to volatilization and drift, thereby causing significant plant damage to nontarget crops downwind. Dicamba was reformulated to reduce volatility and off-target movement. The effectiveness of the dicamba reformulation was assessed by quantifying dicamba emissions following spray application and investigated how meteorological factors influenced the off-target movement. The experiments were conducted at the University of Minnesota Agricultural Experiment Station (UMORE Park) during the growing season of 2018, 2019, 2021, and 2022. Multiple high-flow polyurethane foam air samplers were used to measure dicamba concentrations downwind from a 4-ha soybean field sprayed with dicamba. Dicamba emissions were estimated using backward Lagrangian modeling constrained by the air sample observations. The results indicate that dicamba emissions and downwind transport were significant for several days following application. Further, non-traited soybeans located within 15–45 m showed substantial dicamba-related damage. In warmer, drier seasons, increased dicamba emissions caused more severe damage to downwind soybeans, likely worsened by drought stress preventing recovery. Favorable atmospheric conditions that reduced potential drift can be difficult to achieve in terms of the typical weather experienced over agricultural sites in the Upper Midwest. Integrated forage–row cropping systems provide important agronomic and economic benefits to producers. However, little attention has been given to incorporating forages into row crop systems unique to the southeastern United States. This study assessed the viability of intercropping cotton (Gossypium hirsutum L.) on perennial, cool-season legumes during the summer months in the Southeast Coastal Plain over two production years. Treatments included a weedy fallow, annual ryegrass (ARG; Lolium multiflorum Lam.) monoculture, a red clover (RC; Trifolium pratense L.) and white clover (WC; Trifolium repens L.) mixture, and a three-species mixture of ARG, RC, and WC. Plots were established in fall 2020 with forage grown until May 2021 and 2022, when plots were strip-tilled and planted with cotton. Cotton was managed with minimal herbicide use to preserve perennial clovers. Results indicated WC was more persistent than RC (>40 plants m−2 vs. <40 plants m−2), and the presence of perennial clovers suppressed weeds at similar levels to the ARG monoculture (35 weeds m−2) during the second spring. Perennial clovers grew taller (4–5 cm) when mixed with ARG. The presence of clovers mixed with ARG during the second spring reduced acid detergent fiber (ADF) concentration and increased crude protein (CP) concentration (280 g ADF kg−1; 167 g CP kg−1) compared to the ARG monoculture (315 g ADF kg−1; 126 g CP kg−1). Understanding the relationship between a dam's size and its ecological effects is important for prioritization of river restoration efforts based on dam removal. Although much is known about the effects of large storage dams, this information may not be applicable to small dams, which represent the vast majority of dams being considered for removal. To better understand how dam effects vary with size, we conducted a multidisciplinary study of the downstream effect of dams on a range of ecological characteristics including geomorphology, water chemistry, periphyton, riparian vegetation, benthic macroinvertebrates, and fish. We related dam size variables to the downstream–upstream fractional difference in measured ecological characteristics for 16 dams in the mid‐Atlantic region ranging from 0.9 to 57 m high, with hydraulic residence times (HRTs) ranging from 30 min to 1.5 years. For a range of physical attributes, larger dams had larger effects. For example, the water surface width below dams was greater below large dams. By contrast, there was no effect of dam size on sediment grain size, though the fraction of fine‐grained bed material was lower below dams independently of dam size. Larger dams tended to reduce water quality more, with decreased downstream dissolved oxygen and increased temperature. Larger dams decreased inorganic nutrients (N, P, Si), but increased particulate nutrients (N, P) in downstream reaches. Aquatic organisms tended to have greater dissimilarity in species composition below larger dams (for fish and periphyton), lower taxonomic diversity (for macroinvertebrates), and greater pollution tolerance (for periphyton and macroinvertebrates). Plants responded differently below large and small dams, with fewer invasive species below large dams, but more below small dams. Overall, these results demonstrate that larger dams have much greater impact on the ecosystem components we measured, and hence their removal has the greatest potential for restoring river ecosystems. Wildfires and climate change increasingly are transforming vegetation composition and structure, and postfire management may have long‐lasting effects on ecosystem reorganization. Postfire aerial seeding treatments are commonly used to reduce runoff and soil erosion, but little is known about how seeding treatments affect native vegetation recovery over long periods of time, particularly in type‐converted forests that have been dramatically transformed by the effects of repeated, high‐severity fire. In this study, we analyze and report on a rare long‐term (23‐year) dataset that documents vegetation dynamics following a 1996 post‐fire aerial seeding treatment and a subsequent 2011 high‐severity reburn in a dry conifer landscape of northern New Mexico, USA. Repeated surveys between 1997 and 2019 of 49 permanent transects were analyzed for differences in vegetation cover, richness, and diversity between seeded and unseeded areas, and to characterize the development of seeded and unseeded vegetation communities through time and across gradients of burn severity, elevation, and soil‐available water capacity. Seeded plots showed no significant difference in bare ground cover during the initial years postfire relative to unseeded plots. Postfire seeding led to a clear and sustained divergence in herbaceous community composition. Seeded plots had a much higher cover of non‐native graminoids, primarily Bromus inermis, a likely contaminant in the seed mix. High‐severity reburning of all plots in 2011 reduced native graminoid cover by half at seeded plots compared with both prefire levels and with plots that were unseeded following the initial 1996 fire. In addition, higher fire severity was associated with increased non‐native graminoid cover and reduced native graminoid cover. This study documents fire‐driven ecosystem transformation from conifer forest into a shrub‐and‐grass‐dominated system, reinforced by aerial seeding of grasses and high‐severity reburning. This unique long‐term dataset illustrates that post‐fire seeding carries significant risks of unwanted non‐native species invasions that persist through subsequent fires—thus alternative postfire management actions merit consideration to better support native ecosystem resilience given emergent climate change and increasing disturbance. This study also highlights the importance of long‐term monitoring of postfire vegetation dynamics, as short‐term assessments miss key elements of complex ecosystem responses to fire and postfire management actions. There is a resurgent enthusiasm for Indigenous Knowledges (IK) across settler–colonial institutions of research, education, and conservation. But like fitting a square peg in a round hole, IK are being forced into colonial systems, and then only as marginal alternatives. To address this mismatch, the Traditional Ecological Knowledge Section of the Ecological Society of America (ESA) hosted a 2‐day workshop—entitled Elevating Indigenous Knowledges in Ecology—at the 2022 ESA Annual Meeting, which was held on Kanien'keháka (Mohawk) and Ho‐de‐no‐sau‐nee‐ga (Haudenosaunee) territories in Montreal, Canada. This gathering of 21 interdisciplinary Indigenous ecologists included scholars from across the career and professional spectrum. By consensus, workshop participants (including the authors of this article) identified four emergent themes and respective guiding questions as a pathway toward the transformation of settler–colonial institutions into IK‐led spaces. We highlight this pathway to support actions toward systemic change, inspire future directions for Indigenous and non‐Indigenous ecologists, and nurture stronger relationships between Indigenous communities and the Western sciences, toward actualized decoloniality. Objective: To evaluate an online training program for physician supervisors with the aim of promoting a mentally healthy workplace by improving their use of both responsive and preventive mental health support strategies. Study design: Cluster randomised, waitlist‐controlled trial. Setting, participants: Royal Australasian College of Physicians fellows who were supervising at least one resident physician in any of the 31 primary health networks in Australia and 20 district health boards in New Zealand (health network clusters). Intervention: A brief online skills‐based mental health training program, comprising twelve modules grouped into three topics: common mental illnesses; helping trainees you are concerned about (responsive strategies); and minimising mental health risks at work (preventive strategies). Main outcome measures: Change between baseline and the 3‐month assessment in self‐reported recommended supervisor behaviours; differences between intervention and control groups in recommended behaviour scores three weeks, three months, and six months after the program. Results: Ninety physicians from 20 health network clusters were allocated to the intervention group, 88 physicians from 22 clusters to the control group. Intervention group participants reported greater positive change in behaviour across the study period than those in the control group (mixed model repeated measures analysis, group × time interaction: P < 0.001). The mean change in self‐reported supervisory behaviour score was higher for the intervention than the control group at the 3‐week (mean difference in score, 1.6; 95% confidence interval [CI], 0.8–2.4), 3‐month (0.9; 95% CI, 0.2–1.6), and 6‐month assessments (1.9; 95% CI, 1.1–2.7). The mean change in self‐reported responsive behaviour score was also greater for the intervention group at the 3‐week (mean difference, 2.3; 95% CI, 1.5–3.1), 3‐month (1.0; 95% CI, 0.2–1.9), and 6‐month assessments (2.0; 95% CI, 1.1–2.9); differences in the mean change in preventive behaviour scores were statistically significant at the 3‐week (mean difference, 1.1; 95% CI, 0.1–2.2) and 6‐month assessments (1.8; 95% CI, 0.8–2.8), but not the 3‐month assessment (0.8; 95% CI, –0.1 to 1.7). Conclusions: Brief online mental health training for senior physicians can lead to changes in their self‐reported behaviour for supporting the mental health needs of resident physicians. Summary: The Australian health care system contributes 7% of the national greenhouse gas emission footprint and generates massive waste streams annually. Operating rooms are a particular hotspot, generating at least 20% of the total hospital waste.A systematic search of several global academic databases was conducted in mid‐2022 (articles from 1992 to 2022) for peer‐reviewed research relevant to waste management in the operating rooms. We then used thematic analysis to enumerate and characterise the strategies and barriers to sustainable waste management in the operating room.The waste reduction strategies focused on avoidance of high carbon products; correct waste segregation and reduced overage; reusing, reprocessing, and repurposing devices; and improved recycling. The first barrier identified was a constrained interpretation of the concept of "first do not harm", ingrained in surgeons' practices, in prioritising single‐use surgical products. The second barrier was ineffective or insufficient waste education. The third barrier was the immediate cost of implementing waste management compared with the long term realisation of environmental and economic benefits. The last barrier to implementing institutional practice change was the lack of policies and regulations at the local hospital, federal and international levels.We also evaluated the knowledge gaps in current surgical waste research, including lack of benchmarking data and standardised regulations concerning reusable or reprocessed devices, as well as the methods used to promote pro‐sustainability behavioural change. ABSTRACT Joint swarms can be important components of fractured reservoirs. They are often explained as damage around faults or related to mechanical differences between layers, although this does not explain the close spacing of the joints. Joint swarms around Bergen (Norway) are described, which are not related to exposed faults and are not influenced by layering or foliation in the Lower Palaeozoic gneisses. We suggest an evolution whereby: (1) a zone of microcracks develops; (2) one microcrack propagates and becomes connected to a source of mineralising fluid; (3) the fracture becomes a microvein, with a higher tensile strength than the microcracked host rock; (4) another microcrack propagates and the cycle is repeated, producing a zone of microveins; (5) the veins are partly weathered out, producing an apparent joint swarm, or the microveins crack at or near the ground‐surface. Background: Osimertinib is an irreversible epidermal growth factor receptor (EGFR) tyrosine‐kinase inhibitor (TKI) and an available therapy for patients with non‐small cell lung cancer (NSCLC) that have an EGFR or T790M mutation. It has become the preferred TKI in this patient group as it is superior to first‐generation TKIs; however, osimertinib may be discontinued due to various toxicities or reactions. Aim: We report two instances of successful osimertinib desensitisation in a 70‐year‐old woman requiring treatment for NSCLC following two hypersensitivity reactions presenting as angioedema and urticaria. Clinical details: Osimertinib desensitisation started at 5 mg/day and was gradually increased to 80 mg/day over a period of 30 days. Outcomes: The patient continued osimertinib 80 mg daily for over a year until treatment was withheld for 4 weeks due to thrombocytopenia and diverticulitis. She restarted osimertinib, completing a second desensitisation to a reduced dose of 40 mg daily without serious adverse effect. The patient continues reduced‐dose osimertinib with stable disease. Conclusion: This case report proposes an osimertinib desensitisation strategy useful for select patients experiencing osimertinib‐induced hypersensitivity reactions. It also demonstrates that if there is prolonged disruption to treatment, a second desensitisation can be completed successfully in the same patient so effective treatment in NSCLC may be continued. BACKGROUND: This study investigated the potential of phosphorylated plasma Tau217 ratio (pTau217R) and plasma amyloid beta (Aβ) 42/Aβ40 in predicting brain amyloid levels measured by positron emission tomography (PET) Centiloid (CL) for Alzheimer's disease (AD) staging and screening. METHODS: Quantification of plasma pTau217R and Aβ42/Aβ40 employed immunoprecipitation‐mass spectrometry. CL prediction models were developed on a cohort of 904 cognitively unimpaired, preclinical and early AD subjects and validated on two independent cohorts. RESULTS: Models integrating pTau217R outperformed Aβ42/Aβ40 alone, predicting amyloid levels up to 89.1 CL. High area under the receiver operating characteristic curve (AUROC) values (89.3% to 94.7%) were observed across a broad CL range (15 to 90). Utilizing pTau217R‐based models for low amyloid levels reduced PET scans by 70.5% to 78.6%. DISCUSSION: pTau217R effectively predicts brain amyloid levels, surpassing cerebrospinal fluid Aβ42/Aβ40's range. Combining it with plasma Aβ42/Aβ40 enhances sensitivity for low amyloid detection, reducing unnecessary PET scans and expanding clinical utility. INTRODUCTION: Mild cognitive impairment (MCI) is a prodromal stage of dementia. Understanding the mechanistic changes from healthy aging to MCI is critical for comprehending disease progression and enabling preventative intervention. METHODS: Patients with MCI and age‐matched controls (CN) were administered cognitive tasks during functional near‐infrared spectroscopy (fNIRS) recording, and changes in plasma levels of extracellular vesicles (EVs) were assessed using small‐particle flow cytometry. RESULTS: Neurovascular coupling (NVC) and functional connectivity (FC) were decreased in MCI compared to CN, prominently in the left‐dorsolateral prefrontal cortex (LDLPFC). We observed an increased ratio of cerebrovascular endothelial EVs (CEEVs) to total endothelial EVs in patients with MCI compared to CN, correlating with structural MRI small vessel ischemic damage in MCI. LDLPFC NVC, CEEV ratio, and LDLPFC FC had the highest feature importance in the random Forest group classification. DISCUSSION: NVC, CEEVs, and FC predict MCI diagnosis, indicating their potential as markers for MCI cerebrovascular pathology. INTRODUCTION: There is limited evidence about factors related to the timeliness of dementia diagnosis in healthcare settings. METHODS: In five prospective cohorts at Rush Alzheimer's Disease Center, we identified participants with incident dementia based on annual assessments and examined the timing of healthcare diagnoses in Medicare claims. We assessed sociodemographic, health, and psychosocial correlates of timely diagnosis. RESULTS: Of 710 participants, 385 (or 54%) received a timely claims diagnosis within 3 years prior to or 1 year following dementia onset. In logistic regressions accounting for demographics, we found Black participants (odds ratio [OR] = 2.15, 95% confidence interval [CI]: 1.21 to 3.82) and those with better cognition at dementia onset (OR = 1.48, 95% CI: 1.10 to 1.98) were at higher odds of experiencing a diagnostic delay, whereas participants with higher income (OR = 0.89, 95% CI: 0.81 to 0.97) and more comorbidities (OR = 0.94, 95% CI: 0.89 to 0.98) had lower odds. DISCUSSION: We identified characteristics of individuals who may miss the optimal window for dementia treatment and support. INTRODUCTION: Physical activity is associated with reduced risk of cognitive and functional decline but scalable, sustainable interventions for populations at risk for Alzheimer's disease (AD) and AD and related dementias (ADRD) are lacking. METHODS: A 12‐week randomized‐controlled trial was conducted with a 3‐week follow‐up using a national AD prevention registry (GeneMatch). The control group (n = 50) set step goals and received daily feedback. The intervention group (n = 44) also received a behaviorally designed game based on achieving step goals and reinforced by a support partner. RESULTS: Intervention participants (94 participants, mean age 70, 78% female) had greater change in mean daily step count than control of 1699 steps/day (95% confidence interval [CI], 1149–2249), P < 0.0001, which was sustained in the follow‐up period at 1219 steps/day (95% CI, 455–1983), P = 0.0018. Carriers of the apolipoprotein E ε4 gene (high risk) did not perform differently than non‐carriers; however, high self‐reported risk perception was associated with higher activity. DISCUSSION: A gamified intervention was effective in promoting and sustaining higher physical activity in older adults at genetic risk for AD/ADRD. INTRODUCTION: Spatial extent‐based measures of how far amyloid beta (Aβ) has spread throughout the neocortex may be more sensitive than traditional Aβ‐positron emission tomography (PET) measures of Aβ level for detecting early Aβ deposits in preclinical Alzheimer's disease (AD) and improve understanding of Aβ's association with tau proliferation and cognitive decline. METHODS: Pittsburgh Compound‐B (PIB)‐PET scans from 261 cognitively unimpaired older adults from the Harvard Aging Brain Study were used to measure Aβ level (LVL; neocortical PIB DVR) and spatial extent (EXT), calculated as the proportion of the neocortex that is PIB+. RESULTS: EXT enabled earlier detection of Aβ deposits longitudinally confirmed to reach a traditional LVL‐based threshold for Aβ+ within 5 years. EXT improved prediction of cognitive decline (Preclinical Alzheimer Cognitive Composite) and tau proliferation (flortaucipir‐PET) over LVL. DISCUSSION: These findings indicate EXT may be more sensitive to Aβ's role in preclinical AD than level and improve targeting of individuals for AD prevention trials. INTRODUCTION: Impaired brain protein synthesis, synaptic plasticity, and memory are major hallmarks of Alzheimer's disease (AD). The ketamine metabolite (2R,6R)‐hydroxynorketamine (HNK) has been shown to modulate protein synthesis, but its effects on memory in AD models remain elusive. METHODS: We investigated the effects of HNK on hippocampal protein synthesis, long‐term potentiation (LTP), and memory in AD mouse models. RESULTS: HNK activated extracellular signal‐regulated kinase 1/2 (ERK1/2), mechanistic target of rapamycin (mTOR), and p70S6 kinase 1 (S6K1)/ribosomal protein S6 signaling pathways. Treatment with HNK rescued hippocampal LTP and memory deficits in amyloid‐β oligomers (AβO)‐infused mice in an ERK1/2‐dependent manner. Treatment with HNK further corrected aberrant transcription, LTP and memory in aged APP/PS1 mice. DISCUSSION: Our findings demonstrate that HNK induces signaling and transcriptional responses that correct synaptic and memory deficits in AD mice. These results raise the prospect that HNK could serve as a therapeutic approach in AD. INTRODUCTION: Neighborhood socioeconomic status (SES) has been linked to dementia, but the distribution of SES within a neighborhood may also matter. METHODS: Data from 460 (47% Black, 46% White) older adults from the Michigan Cognitive Aging Project were linked to census tract–level data from the National Neighborhood Data Archive (NaNDA). Neighborhood SES included two composites reflecting disadvantage and affluence. Neighborhood racial income inequality was the ratio of median incomes for White versus Black residents. Generalized estimating equations examined associations between neighborhood factors and cognitive domains. RESULTS: Neighborhood racial income inequality was uniquely associated with worse cognitive health, and these associations did not differ by participant race. Neighborhood disadvantage was only associated with worse cognitive health among Black participants. DISCUSSION: Both the level and racial distribution of SES within a neighborhood may be relevant for dementia risk. Racial differences in the level and impact of neighborhood SES contribute to dementia inequalities. INTRODUCTION: This study derived composite scores for two novel cognitive measures, the No Practice Effect (NPE) battery and the Miami Computerized Functional Skills Assessment and Training system for use in early‐stage Alzheimer's disease (AD) clinical trials. Their psychometric properties and associations with AD risk markers were compared to those of well‐established measures. METHODS: For 291 older adults with healthy cognition or early mild cognitive impairment, Exploratory factor analyses were used to identify the factor structure of the NPE. Factor and total scores were examined for their psychometric properties and associations with AD risk biomarkers. RESULTS: Composite scores from the novel cognitive and functional measures demonstrated better psychometric properties (distribution and test‐retest reliability) and stronger associations with AD‐related demographic, genetic, and brain risk markers than well‐established measures, DISCUSSION: These novel measures have potential for use as primary cognitive and functional outcomes in early‐stage AD clinical trials. Objective: Saugeye (Sauger Sander canadensis × Walleye S. vitreus) have been introduced in reservoirs for several purposes, including as a top‐down control to combat stunting in crappie Pomoxis spp. populations. However, no comprehensive diet evaluation has been completed in southern reservoirs. Our objectives were to assess variability in saugeye diet compositions, explore factors influencing crappie presence in saugeye diets, and investigate trends in prey size to inform management strategies regarding predatory control of crappie populations. Methods: We collected 2638 saugeye diets from six Oklahoma reservoirs. We used permutational multivariate analysis of variance to test differences in diet compositions among saugeye lengths, study reservoirs, and seasons, and we used logistic regression models to correlate the presence of crappie in diets with saugeye size, season, crappie abundance, productivity, and turbidity. Finally, we used bivariate plots of diet indices and quantile regression to explore trends in prey importance and prey size relative to saugeye size. Result: Saugeye diet compositions were best explained by saugeye size, followed by study reservoir and season. Saugeye exhibited an ontogenetic shift from Inland Silverside Menidia beryllina to shad Dorosoma spp., and crappie were eaten infrequently in comparison. The presence of crappie prey in diets was most correlated with saugeye size (primarily large saugeye), followed by higher crappie abundance, lower reservoir productivity, and season (highest in fall). Saugeye consumed larger prey than most piscivores relative to predator size. Conclusion: Our results indicated that dominant prey of saugeye transitioned from smaller‐bodied fishes to larger shad as they grew when both prey types were available, which may have implications for growth and recruitment. Ultimately, the low and variable use of crappie prey may lead to inconsistent predatory effects on crappie populations, and large saugeye may exert greater top‐down pressure on crappie in lakes with lower productivity or higher crappie abundance. Objective: Biodiversity is declining due to invasive species and other factors that can affect individual species differently. Silver Chub Macrhybopsis storeriana are declining in their native range, and their conservation status in the Great Lakes ranges from secure to possibly extirpated. Lake Erie once supported a large Silver Chub population until it crashed in the 1950s. Additionally, the spawning behavior and reproductive guild of Silver Chub in Lake Erie is unknown. Our objective was to document Silver Chub spawning in the Maumee River, a Lake Erie tributary. Methods: Invasive Grass Carp Ctenopharyngodon idella are known to spawn in the Maumee River during high‐flow events from May to July, and the University of Toledo and U.S. Geological Survey regularly sample the lower 24 km for early life stages using paired bongo nets. Contents from paired bongo nets are returned to the laboratory for processing, and a subset of potential Grass Carp eggs are sent for genetic analysis. Result: On June 8, 2022, several potential Grass Carp eggs were captured at two sites on the Maumee River during a high‐flow event. Fifteen potential Grass Carp eggs were sent for genetic analysis, and DNA sequencing revealed that six of these eggs were Silver Chub. Discussion: This was the first known collection of Silver Chub eggs in a Lake Erie tributary, and our findings indicate that Silver Chub likely belong to the pelagophil reproductive guild. Although Grass Carp and Silver Chub spawn under similar conditions, management actions to control Grass Carp in the Maumee River may be unlikely to affect Silver Chub due to electrofishing settings used in the capture of Grass Carp. The verification of Silver Chub spawning in a Western Erie Basin tributary provides insights into their reproductive biology that could be useful in recovery planning in Lake Erie and throughout the Great Lakes. An immunosuppressive tumor microenvironment promotes tumor growth and is one of the main factors limiting the response to cancer immunotherapy. We have previously reported that inhibition of vacuolar protein sorting 34 (VPS34), a crucial lipid kinase in the autophagy/endosomal trafficking pathway, decreases tumor growth in several cancer models, increases infiltration of immune cells and sensitizes tumors to anti‐programmed cell death protein 1/programmed cell death 1 ligand 1 therapy by upregulation of C‐C motif chemokine 5 (CCL5) and C‐X‐C motif chemokine 10 (CXCL10) chemokines. The purpose of this study was to investigate the signaling mechanism leading to the VPS34‐dependent chemokine increase. NanoString gene expression analysis was applied to tumors from mice treated with the VPS34 inhibitor SB02024 to identify key pathways involved in the anti‐tumor response. We showed that VPS34 inhibitors increased the secretion of T‐cell‐recruitment chemokines in a cyclic GMP‐AMP synthase (cGAS)/stimulator of interferon genes protein (STING)‐dependent manner in cancer cells. Both pharmacological and small interfering RNA (siRNA)‐mediated VPS34 inhibition increased cGAS/STING‐mediated expression and secretion of CCL5 and CXCL10. The combination of VPS34 inhibitor and STING agonist further induced cytokine release in both human and murine cancer cells as well as monocytic or dendritic innate immune cells. Finally, the VPS34 inhibitor SB02024 sensitized B16‐F10 tumor‐bearing mice to STING agonist treatment and significantly improved mice survival. These results show that VPS34 inhibition augments the cGAS/STING pathway, leading to greater tumor control through immune‐mediated mechanisms. Objective: Both electronic tags (e.g., acoustic and radio transmitters) and conventional external tags are used to evaluate movement and population dynamics of fish. External tags are also sometimes used to facilitate the recovery of internal electronic tags or other instrumentation because healing can make it difficult to identify fish with internal tags based on appearance alone. With both tag types, tag shedding and failure of electronic tags can affect accuracy and precision of study results. Methods: We used a decade (2011-2021) of recapture data for Walleye Sander vitreus tagged in the Laurentian Great Lakes, where fish were double-or triple-tagged with external tags (T-bar, loop, or internal anchor tags) and internal acoustic transmitters, to quantify external tag and internal transmitter shedding and transmitter failure rates. Result: In total, 1125 (33%) Walleye were recovered that had retained at least one external tag or internal transmitter. No confirmed cases of transmitter shedding were observed; 15 of 899 transmitters (2%) that were checked for functionality failed prior to the expected battery expiration. The retention of external T-bar tags 1 year after release differed depending on whether the tag was placed anterior or posterior to the secondary dorsal fin (anterior, fish length = 420 mm: 73% retention; anterior, fish length = 700 mm: 73%, posterior: 63%) but was <26% after 4 years for both tag positions and fish sizes. Internal anchor tags had an 88% 1-year retention probability and 81% 4-year retention probability. Loop tags had the highest 1-year retention (89%) but after 4 years retention (28-34% depending on agency) was comparable to that of T-bar tags. Conclusion: Better understanding of tag retention characteristics through long-term tagging studies such as this can inform study design, be considered in model design, and ultimately improve inferences from mark-recapture studies. Objective: We investigated if large Paddlefish Polyodon spathula invest in testis weight disproportionately more than smaller males, as expressed by the gonadosomatic index (GSI). The approach was to assess the relation between testis weight F (i.e., the combined weight of both testes) and fish weight W in the expression F = aWb. The hypothesis was that sperm competition in Paddlefish would be expected (with b > 1) based on observations of several male fish attending a female spawner in a past study in the Osage River, Missouri. Methods: We used a large, three‐state data set from three fisheries (Montana, North Dakota, and Oklahoma) to evaluate isometry and allometry in the power function relationship between fish weight and testis weight. Result: All three data sets showed hyperallometric increases in testis weight with fish weight (i.e., b > 1). For Montana males, hyperallometry was greatest (b = 1.56; n = 8798), followed by North Dakota (b = 1.36; n = 8209) and Oklahoma (b = 1.22; n = 9571). Paddlefish from Montana and North Dakota had much lower testis weights through age 18 and much lower fish weights through age 25 than Oklahoma Paddlefish. By ages 17–25, mean male GSI in Oklahoma declined, whereas it remained level at about 1.5 for combined Montana and North Dakota Paddlefish. Throughout their lives, at each age, Montana and North Dakota Paddlefish allocated a higher percentage of their total weight to testis weight and reached a higher asymptotic GSI (GSI∞) than did Oklahoma Paddlefish; GSI∞ was 1.50 for Montana fish, 1.44 for North Dakota fish, and 1.26 for Oklahoma fish. Conclusion: Results provide evidence supporting the occurrence sperm competition (i.e., b > 1), as would occur when two or more males compete to fertilize the eggs of a female spawning Paddlefish. Comparisons among species and stocks are made but were limited by an acute shortage of data on male testis weights and other testis characteristics (e.g., sperm size, energy content), and reproductive investment in Paddlefish and most other fishes in other localities. Crop growth and development is affected by intraspecies competition. This study was conducted to determine whether cotton productivity and profitability could be manipulated through planting geometry. The effects of row spacing, planting pattern, and variety on cotton fruit distribution, yield, and net returns were investigated near Bella Mina, AL, on a Decatur silt loam (fine, kaolinitic, thermic Rhodic Paleudults); Jackson, TN, on a Calhoun silt loam (fine‐silty, mixed, active, thermic Typic Glossaqualfs); Starkville, MS, on a Leeper silty clay loam (fine, smectitic, nonacid, thermic Vertic Epiaquepts); and Stoneville, MS, on a Bosket very fine sandy loam (fine‐loamy, mixed, active, thermic Mollic Hapludalfs). For both the 76‐ and 97‐cm row spacing, transitioning from a solid to a 2 × 1 skip row pattern reduced cotton lint yield by 19%. Reduced lint yield in the 2 × 1 skip row pattern was attributed primarily to a 7% decrease in the number of Zone 1 and first‐position bolls. Consequently, planting on a solid rather than skip row pattern increased net returns by 21%. Planting cotton in either 76‐ or 97‐cm rows using a solid rather than skip row pattern likely increases crop productivity and profitability across the US cotton belt. Individuals with Down syndrome (DS) have a partial or complete trisomy of chromosome 21, resulting in an increased risk for early‐onset Alzheimer's disease (AD)‐type dementia by early midlife. Despite ongoing clinical trials to treat late‐onset AD, individuals with DS are often excluded. Furthermore, timely diagnosis or management is often not available. Of the genetic causes of AD, people with DS represent the largest cohort. Currently, there is a knowledge gap regarding the underlying neurobiological mechanisms of DS‐related AD (DS‐AD), partly due to limited access to well‐characterized brain tissue and biomaterials for research. To address this challenge, we created an international consortium of brain banks focused on collecting and disseminating brain tissue from persons with DS throughout their lifespan, named the Down Syndrome Biobank Consortium (DSBC) consisting of 11 biobanking sites located in Europe, India, and the USA. INTRODUCTION: The pace of innovation has accelerated in virtually every area of tau research in just the past few years. METHODS: In February 2022, leading international tau experts convened to share selected highlights of this work during Tau 2022, the second international tau conference co‐organized and co‐sponsored by the Alzheimer's Association, CurePSP, and the Rainwater Charitable Foundation. RESULTS: Representing academia, industry, and the philanthropic sector, presenters joined more than 1700 registered attendees from 59 countries, spanning six continents, to share recent advances and exciting new directions in tau research. DISCUSSION: The virtual meeting provided an opportunity to foster cross‐sector collaboration and partnerships as well as a forum for updating colleagues on research‐advancing tools and programs that are steadily moving the field forward. INTRODUCTION: The extent to which the Big Five personality traits and subjective well‐being (SWB) are discriminatory predictors of clinical manifestation of dementia versus dementia‐related neuropathology is unclear. METHODS: Using data from eight independent studies (Ntotal= 44,531; Ndementia= 1703; baseline Mage= 49 to 81 years, 26 to 61% female; Mfollow‐up range = 3.53 to 21.00 years), Bayesian multilevel models tested whether personality traits and SWB differentially predicted neuropsychological and neuropathological characteristics of dementia. RESULTS: Synthesized and individual study results indicate that high neuroticism and negative affect and low conscientiousness, extraversion, and positive affect were associated with increased risk of long‐term dementia diagnosis. There were no consistent associations with neuropathology. DISCUSSION: This multistudy project provides robust, conceptually replicated and extended evidence that psychosocial factors are strong predictors of dementia diagnosis but not consistently associated with neuropathology at autopsy. INTRODUCTION: White matter hyperintensities (WMH) may promote clinical Alzheimer's disease (AD) disparities between Black American (BA) and non‐Hispanic White (nHW) populations. Using a novel measurement, unhealthy white matter connectivity (UWMC), we interrogated racialized group differences in associations between WMH in AD pathology‐affected regions and cognition. METHODS: UWMC is the proportion of white matter fibers that pass through WMH for every pair of brain regions. Individual regression models tested associations of UWMC in beta‐amyloid (Aβ) or tau pathology‐affected regions with cognition overall, stratified by racialized group, and with a racialized group interaction. RESULTS: In 201 older adults ranging from cognitively unimpaired to AD, BA participants exhibited greater UWMC and worse cognition than nHW participants. UWMC was negatively associated with cognition in 17 and 5 Aβ‐ and tau‐affected regions, respectively. Racialization did not modify these relationships. DISCUSSION: Differential UWMC burden, not differential UWMC‐and‐cognition associations, may drive clinical AD disparities between racialized groups. Exposure to time‐varying electromagnetic fields (EMF) has the capacity to influence biological systems. Our results demonstrate that exposure to time‐varying EMF modeled after the physiological firing frequency of intercellular calcium waves can inhibit proliferation and induce apoptosis in malignant cells. Single exposure of B16‐BL6 cells to a Ca2+ EMF for 40 min reduced the number of viable cells by 50.3%. Cell imaging with acridine orange and ethidium bromide dye revealed substantial cellular apoptosis, preapoptotic cells, nuclear fragmentation, and large spacing between cells in the Ca2+ EMF condition when compared to the control condition. The ability of Ca2+ EMF to influence the proliferation and survival of malignant cells suggests that exposure to specific EMF may function as a potential anticancer therapy. [ABSTRACT FROM AUTHOR]

Background: The authors developed a pain monitoring app offering educational information, and real‐time health care professional feedback on clinically significant pain (>4 numeric rating scale [NRS]‐11) for children with cancer to reduce pain at home. Methods: This monocenter, nonblinded randomized controlled trial enrolled Dutch children (0–18 years old) receiving cancer treatment (≥3 months after diagnosis, ≥2 months treatment remaining). Children were randomly assigned to use the app or receive usual care (two parallel groups). We assessed whether use of the app yielded less clinically significant pain (aim 1) and whether it affected pain severity, duration, interference, pain management strategies, and parental emotional well‐being (aim 2). The app was also evaluated by families (aim 3). Results: A total of 94 children were randomized to use the app (15 drop‐outs), and 90 were to receive care as usual (11 drop‐outs). The app group (n = 79, mean age: 7.5 [5.1] years, 48% girls, 63% hemato‐oncology diagnosis) reported significantly less clinically significant pain compared to usual care (n = 79, mean age: 7.5 [5.4] years, 52% girls, 65% hemato‐oncology diagnosis) (odds ratio [OR], 0.38; 95% confidence interval [CI], 0.198–0.734]) (aim 1), as well as significantly lower pain severity (β = –0.27; 95% CI, –0.407 to –0.142). No differences were found for duration, interference, or management strategies. Parents in the app group reported significantly less distress compared to usual care (β = –0.84; 95% CI, –1.61 to –0.03]) (aim 2). Families generally evaluated the app positively (aim 3). Conclusions: Use of the app resulted in less clinically significant pain at home. The exact working mechanisms of the app should be further elucidated. Use of a pain monitoring app with educational information on pain (management) and real‐time health care professional feedback reduced clinically significant pain in children with cancer at home. A substantial body of empirical evidence suggests that anthropogenic disturbance can affect the structure and function of grassland ecosystems. Despite this, few studies have elucidated the mechanisms through which grazing and mowing, the two most widespread land management practices, affect the stability of natural grassland communities. In this study, we draw upon 9 years of field data from natural grasslands in northern China to investigate the effects of gazing and mowing on community stability, specifically focusing on community aboveground net primary productivity (ANPP) and dominance, which are two major biodiversity mechanisms known to characterize community fluctuations. We found that both grazing and mowing reduced ANPP in comparison to areas enclosed by fencing. Grazing reduced community stability by increasing the likelihood of single‐species dominance and decreasing the relative proportion of nondominant species. In contrast, mowing reduced the productivity of the dominant species but increased the productivity of nondominant species. As a consequence, mowing improved the overall community stability by increasing the stability of nondominant species. Our study provides novel insight into understanding of the relationship between community species fluctuation‐stability, with implications for ecological research and ecosystem management in natural grasslands. Background: Physiatry is a specialty with high rates of burnout. Although organizational strategies to combat burnout are key, it is also important to understand strategies that individual physiatrists can use to address burnout. Objective: To identify changes that resulted in improvement of occupational well‐being of physiatrists over a 6‐ to 9‐month period. Design: We employed two quantitative surveys spaced 6 to 9 months apart to identify physiatrists who experienced meaningful improvement in occupational burnout and/or professional fulfillment between the two survey timepoints. These physiatrists were subsequently recruited to participate in a qualitative study using semi‐structured interviews to identify changes that respondents felt contributed to improvements in burnout and professional fulfillment. Setting: Online surveys and interviews. Participants: Physiatrists in the American Academy of Physical Medicine and Rehabilitation (AAPM&R) Membership Masterfile. Main Outcome Measure: Burnout and professional fulfillment were assessed using the Stanford Professional Fulfillment Index. Results: One hundred twelve physiatrists responded to the baseline and follow‐up surveys. Of these, 35 were eligible for interviews based on improvements in the Stanford Professional Fulfillment Index, and 23 (64%) agreed to participate. Themes from the qualitative interviews highlighted the importance of personal lifestyle choices, approaches to improve professional satisfaction, and strategies to foster work‐life harmony. Personal lifestyle strategies included investing in wellness and mental health. Efforts to improve professional satisfaction included decreasing work intensity, prioritizing meaningful aspects of work, and building relationships with colleagues. Fostering work‐life harmony also included making trade‐offs in both domains, setting boundaries at work, setting expectations at home, and overcoming personal challenges. Conclusion: Our findings illustrate that, in addition to organizational strategies demonstrated to be effective, there are actions that individual physiatrists can take to recover from burnout and foster professional fulfillment. Objective: To quantify the benefits versus harms of amantadine in the treatment of irritability and aggression following traumatic brain injury. Methods: Secondary outcome data from a randomized controlled multisite trial of amantadine 100 mg twice daily were used to calculate number‐needed‐to‐treat (NNT). Given prior findings of positive clinician‐perceived effects and low incidence of adverse events, we hypothesized low number‐needed‐to‐treat for benefit (NNTB; high benefit) and high number‐needed‐to‐treat for harm (NNTH; low risk) based on the clinician ratings, supporting the use of amantadine in clinical practice. Specifically, NNTB values were calculated using number of individuals with improvement on the Clinician Global Impressions–Global Improvement scale (GI). NNTH values were computed using number of individuals with worsening on the GI and experiencing serious and any adverse events. Results: Based on clinician ratings, on average for every six patients treated with amantadine rather than placebo, one extra patient would be expected to improve (NNTB = 6.4; 95% confidence interval [CI]: [3.3–76.8]). More participants in the placebo group worsened than in the amantadine group, but the result was not statistically significant (NNTH = −92.4; 95% CI: [NNTB −32.9 to infinity to NNTH −19.2]). The amantadine and placebo groups did not differ on the numbers of adverse events experienced during the trial. Conclusion: Clinician ratings suggest modest benefit of amantadine 100 mg twice daily with low risk to appropriately selected patients with adequate renal clearance. Thus, amantadine should be considered a treatment option for the experienced brain injury clinician. Although there has been a resurgence in interest among farmers in producing narrow row hybrid maize (Zea mays L.) in the Eastern UnitedStates, agronomic recommendations for modern hybrids are limited. To address this issue, field trials were carried out from 2016 to 2018 in Ohio and Pennsylvania (10 unique site‐year combinations), aiming to answer the following questions: (i) Do modern hybrids differ in their response to narrow (38 cm) and conventional (76 cm) row spacing? (ii) Is there a yield increase for narrow row production? The study was planted in a split‐plot randomized complete block design with four replications. The whole plot factor was row spacing, and the subplot factor was hybrid. Two hybrids (P0506AM and P0604AM) were in an "early"‐season group, and two hybrids (P1197AM and P1443AM) were "full"‐season hybrids. Both P0506AM and P1443AM were identified as AQUAmax hybrids and had superior seed company drought tolerance ratings over their maturity pairing counterpart. Grain and forage yield were 4% and 8% greater, respectively, for the narrow row spacing across hybrids. Hybrid differences in drought tolerance designation did not result in grain or forage yield differences under varied row spacings. There was no hybrid × row spacing interaction. In most site‐years, P1197AM hybrid produced the greatest grain and forage yields, though P1443AM and P0506AM were similar in most cases. Results suggest a minor advantage in yield (≤8%) from narrow rows in Ohio and Pennsylvania, though other considerations that include cost to conversion should be considered prior to adoption.
