The development of nanomedicine involves complex nanomaterial research involving magnetic nanomaterials and their use in magnetic hyperthermia. The selection of the optimal treatment strategies is time-consuming, expensive, unpredictable, and not consistently effective. Delivering personalized therapy that obtains maximal efficiency and minimal side effects is highly important. Thus, Artificial Intelligence (AI) based algorithms provide the opportunity to overcome these crucial issues. In this paper, we briefly overview the significance of the combination of AI-based methods, particularly the Machine Learning (ML) technique, with magnetic hyperthermia. We considered recent publications, reports, protocols, and review papers from Scopus and Web of Science Core Collection databases, considering the PRISMA-S review methodology on applying magnetic nanocarriers in magnetic hyperthermia. An algorithmic performance comparison in terms of their types and accuracy, data availability taking into account their amount, types, and quality was also carried out. Literature shows AI support of these studies from the physicochemical evaluation of nanocarriers, drug development and release, resistance prediction, dosing optimization, the combination of drug selection, pharmacokinetic profile characterization, and outcome prediction to the heat generation estimation. The papers reviewed here clearly illustrate that AI-based solutions can be considered as an effective supporting tool in drug delivery, including optimization and behavior of nanocarriers, both in vitro and in vivo, as well as the delivery process. Objective(s): One applications of nanotechnology is in the area of medicine which is called nanomedicine. Primary instruments in nanomedicine can help us to detect diseases and used for drug delivery to inaccessible areas of human tissues. An important issue in simulating the motion of nanoparticles is modeling blood flow as a Newtonian or non- Newtonian fluid. Sometimes blood flow is simulated as a Newtonian fluid but actually blood has nature of non- Newtonian fluid. Simulation of targeted drug delivery and motion of nanoparticles in the blood flow as Newtonian and non-Newtonian fluid flow is investigated in this paper. Materials and Methods: In this paper, the blood flow is modeled as both Newtonian and non-Newtonian fluid and the effects of each case on the motion of nanoparticles in blood flow and targeted drug delivery is investigated. The flow is modeled with finite volume method. The particle modeled with discrete phase model. Results: Cross, Herschel-Bulkley and Power-law models are used for simulating the non-Newtonian blood flow. Numerical simulations show that trajectory of nanoparticle's movement and the required time to pass the vessel by blood flow is variable for different models. According to obtained results, non-Newtonian Power-law and Herschel-Bulkley models have closely similar results but they have significant differences compared with Newtonian model. Conclusion: According to the results, it is preferred in the simulation to model blood flow as a non-Newtonian fluid and uses one of Herschel- Bulkley or Power-law models. The field of nanomedicine has recently emerged as a product of the expansion of a range of nanotechnologies into biomedical science, pharmacology and clinical practice. Due to the unique properties of nanoparticles and the related nanostructures, their applications to medical diagnostics, imaging, controlled drug and gene delivery, monitoring of therapeutic outcomes, and aiding in medical interventions, provide a new perspective for challenging problems in such demanding issues as those involved in the treatment of cancer or debilitating neurological diseases. In this review, we evaluate the role and contributions that the applications of magnetic nanoparticles (MNPs) have made to various aspects of nanomedicine, including the newest magnetic particle imaging (MPI) technology allowing for outstanding spatial and temporal resolution that enables targeted contrast enhancement and real-time assistance during medical interventions. We also evaluate the applications of MNPs to the development of targeted drug delivery systems with magnetic field guidance/focusing and controlled drug release that mitigate chemotherapeutic drugs' side effects and damage to healthy cells. These systems enable tackling of multiple drug resistance which develops in cancer cells during chemotherapeutic treatment. Furthermore, the progress in development of ROSand heat-generating magnetic nanocarriers and magneto-mechanical cancer cell destruction, induced by an external magnetic field, is also discussed. The crucial roles of MNPs in the development of biosensors and microfluidic paper array devices (PADs) for the detection of cancer biomarkers and circulating tumor cells (CTCs) are also assessed. Different chemical agents are used for the biocompatibility and/or functionality of the nanoparticles used in magnetic hyperthermia to reduce or even eliminate cellular toxicity and to limit the interaction between them (van der Waals and magnetic dipolar interactions), with highly beneficial effects on the efficiency of magnetic hyperthermia in cancer therapy. In this paper we propose an innovative strategy for the biocompatibility of these nanoparticles using gamma-cyclodextrins (γ-CDs) to decorate the surface of magnetite (Fe3O4) nanoparticles. The influence of the biocompatible organic layer of cyclodextrins, from the surface of Fe3O4 ferrimagnetic nanoparticles, on the maximum specific loss power in superparamagnetic hyperthermia, is presented and analyzed in detail in this paper. Furthermore, our study shows the optimum conditions in which the magnetic nanoparticles covered with gamma-cyclodextrin (Fe3O4–γ-CDs) can be utilized in superparamagnetic hyperthermia for an alternative cancer therapy with higher efficiency in destroying tumoral cells and eliminating cellular toxicity. Among central nervous system (CNS) disorders, Alzheimer's disease (AD) is the most prevalent neurodegenerative disorder and a major cause of dementia worldwide. The yet unclear etiology of AD and the high impenetrability of the blood–brain barrier (BBB) limit most therapeutic compounds from reaching the brain. Although many efforts have been made to effectively deliver drugs to the CNS, both invasive and noninvasive strategies employed often come with associated side effects. Nanotechnology-based approaches such as nanoparticles (NPs), which can act as multifunctional platforms in a single system, emerged as a potential solution for current AD theranostics. Among these, magnetic nanoparticles (MNPs) are an appealing strategy since they can act as contrast agents for magnetic resonance imaging (MRI) and as drug delivery systems. The nanocarrier functionalization with specific moieties, such as peptides, proteins, and antibodies, influences the particles' interaction with brain endothelial cell constituents, facilitating transport across the BBB and possibly increasing brain penetration. In this review, we introduce MNP-based systems, combining surface modifications with the particles' physical properties for molecular imaging, as a novel neuro-targeted strategy for AD theranostics. Cancer is a disease that causes millions of deaths per year worldwide because conventional treatments have disadvantages such as unspecific tumor selectivity and unwanted toxicity. Most human solid tumors present hypoxic microenvironments and this promotes multidrug resistance. In this study, we present "Magnetogene nanoparticle vector" which takes advantage of the hypoxic microenvironment of solid tumors to increase selective gene expression in tumor cells and reduce unwanted toxicity in healthy cells; this vector was guided by a magnet to the tumor tissue. Magnetic nanoparticles (MNPs), chitosan (CS), and the pHRE-Luc plasmid with a hypoxia-inducible promoter were used to synthesize the vector called "Magnetogene nanoparticles" by ionic gelation. The hypoxic functionality of Magnetogene vector nanoparticles was confirmed in the B16F10 cell line by measuring the expression of the luciferase reporter gene under hypoxic and normoxic conditions. Also, the efficiency of the Magnetogene vector was confirmed in vivo. Magnetogene was administered by intravenous injection (IV) in the tail vein and directed through an external magnetic field at the site of tumor growth in C57Bl/6 mice. A Magnetogene vector with a size of 50 to 70 nm was directed and retained at the tumor area and gene expression was higher at the tumor site than in the others tissues, confirming the selectivity of this vector towards hypoxic tumor areas. Magnetic nanoparticles (MNPs) have great potential for a wide use in various biomedical applications due to their unusual properties. It is critical for many applications that the biological effects of nanoparticles are studied in depth. To date, many disparate results can be found in the literature regarding nanoparticle-biological factors interactions. This review highlights recent developments in this field with particular focuses on in vitro MNPs-cell interactions. The effect of MNPs properties on cellular uptake and cytotoxicity evaluation of MNPs were discussed. Some employed methods are also included. Moreover, nanoparticle-cell interactions are mediated by the presence of proteins absorbed from biological fluids on the nanoparticle. The purpose of this paper was to detect and separate the cluster intensity provided by Iron oxide nanoparticles (IO-NPs), in the MRI images, to investigate the drug delivery effectiveness. IO-NPs were attached to the macrophages and inserted into the eye of the inflamed mouse's calf. The low resolution of MRI and the tiny dimension of the IO-NPs made the situation challenging. IO-NPs serve as a marker, due to their strong intensity in the MRI, enabling us to follow the track of the macrophages. An image processing procedure was developed to estimate the position and the amount of IO-NPs spreading inside the inflamed mouse leg. A fuzzy Clustering algorithm was adopted to select the region of interest (ROI). A 3D model of the femoral region was used for the detection and then the extraction IO-NPs in the MRI images. The results achieved prove the effectiveness of the proposed method to improve the control process of targeted drug delivered. In recent years, much effort has been invested into developing multifunctional drug delivery systems to overcome the drawbacks of conventional carriers. Magnetic nanoparticles are not generally used as carriers but can be functionalised with several different biomolecules and their size can be tailored to present a hyperthermia response, allowing for the design of multifunctional systems which can be active in therapies. In this work, we have designed a drug carrier nanosystem based on Fe3O4 nanoparticles with large heating power and 4-amino-2-pentylselenoquinazoline as an attached drug that exhibits oxidative properties and high selectivity against a variety of cancer malignant cells. For this propose, two samples composed of homogeneous Fe3O4 nanoparticles (NPs) with different sizes, shapes, and magnetic properties have been synthesised and characterised. The surface modification of the prepared Fe3O4 nanoparticles has been developed using copolymers composed of poly(ethylene-alt-maleic anhydride), dodecylamine, polyethylene glycol and the drug 4-amino-2-pentylselenoquinazoline. The obtained nanosystems were properly characterised. Their in vitro efficacy in colon cancer cells and as magnetic hyperthermia inductors was analysed, thereby leaving the door open for their potential application as multimodal agents. The incidence of gastrointestinal cancers has increased in recent years. Current treatments present numerous challenges, including drug resistance, non-specificity, and severe side effects, needing the exploration of new therapeutic strategies. One promising avenue is the use of magnetic nanoparticles, which have gained considerable interest due to their ability to generate heat in tumor regions upon the application of an external alternating magnetic field, a process known as hyperthermia. This review conducted a systematic search of in vitro and in vivo studies published in the last decade that employ hyperthermia therapy mediated by magnetic nanoparticles for treating gastrointestinal cancers. After applying various inclusion and exclusion criteria (studies in the last 10 years where hyperthermia using alternative magnetic field is applied), a total of 40 articles were analyzed. The results revealed that iron oxide is the preferred material for magnetism generation in the nanoparticles, and colorectal cancer is the most studied gastrointestinal cancer. Interestingly, novel therapies employing nanoparticles loaded with chemotherapeutic drugs in combination with magnetic hyperthermia demonstrated an excellent antitumor effect. In conclusion, hyperthermia treatments mediated by magnetic nanoparticles appear to be an effective approach for the treatment of gastrointestinal cancers, offering advantages over traditional therapies. In this article, we provide an overview of the progress of scientists working to improve the quality of life of cancer patients. Among the known methods, cancer treatment methods focusing on the synergistic action of nanoparticles and nanocomposites have been proposed and described. The application of composite systems will allow precise delivery of therapeutic agents to cancer cells without systemic toxicity. The nanosystems described could be used as a high-efficiency photothermal therapy system by exploiting the properties of the individual nanoparticle components, including their magnetic, photothermal, complex, and bioactive properties. By combining the advantages of the individual components, it is possible to obtain a product that would be effective in cancer treatment. The use of nanomaterials to produce both drug carriers and those active substances with a direct anti-cancer effect has been extensively discussed. In this section, attention is paid to metallic nanoparticles, metal oxides, magnetic nanoparticles, and others. The use of complex compounds in biomedicine is also described. Dendritic cells (DCs)‐based tumor vaccines have the advantages of high safety and rapid activation of T cells, and have been approved for clinical tumor treatment. However, the conventional DC vaccines have some severe problems, such as poor activation of DCs in vitro, low level of antigen presentation, reduced cell viability, and difficulty in targeting lymph nodes in vivo, resulting in poor clinical therapeutic effects. In this research, magnetic nanoparticles Fe3O4@Ca/MnCO3 were prepared and used to actively and efficiently deliver antigens to the cytoplasm of DCs, promote antigen cross‐presentation and DC activation, and finally enhance the cellular immune response of DC vaccines. The results show that the magnetic nanoparticles can actively and quickly deliver antigens to the cytoplasm of DCs by regulating the magnetic field, and achieve cross‐presentation of antigens. At the same time, the nanoparticles degradation product Mn2+ enhanced immune stimulation through the interferon gene stimulating protein (STING) pathway, and another degradation product Ca2+ ultimately promoted cellular immune response by increasing autophagy. The DC vaccine constructed with the magnetic nanoparticles can more effectively migrate to the lymph nodes, promote the proliferation of CD8+ T cells, prolong the time of immune memory, and produce higher antibody levels. We present an investigation of the effects on BxPC3 pancreatic cancer cells of proton therapy combined with hyperthermia, assisted by magnetic fluid hyperthermia performed with the use of magnetic nanoparticles. The cells' response to the combined treatment has been evaluated by means of the clonogenic survival assay and the estimation of DNA Double Strand Breaks (DSBs). The Reactive Oxygen Species (ROS) production, the tumor cell invasion and the cell cycle variations have also been studied. The experimental results have shown that the combination of proton therapy, MNPs administration and hyperthermia gives a clonogenic survival that is much smaller than the single irradiation treatment at all doses, thus suggesting a new effective combined therapy for the pancreatic tumor. Importantly, the effect of the therapies used here is synergistic. Moreover, after proton irradiation, the hyperthermia treatment was able to increase the number of DSBs, even though just at 6 h after the treatment. Noticeably, the magnetic nanoparticles' presence induces radiosensitization effects, and hyperthermia increases the production of ROS, which contributes to cytotoxic cellular effects and to a wide variety of lesions including DNA damage. The present study indicates a new way for clinical translation of combined therapies, also in the vision of an increasing number of hospitals that will use the proton therapy technique in the near future for different kinds of radio-resistant cancers. With 10 million deaths in 2020 cancer remains one of the most challenging diseases in contemporary medicine. Chemotherapy, surgery, and radiation therapy are commonly used to treat tumors. These treatments harm healthy tissues and succeed rarely in advance stages of cancer. Recent studies indicate that magnetic hyperthermia, which involves targeted delivery of magnetic nanoparticles to tumor cells followed by localized remote heating of cancer tissues could revolutionize clinical practice in the treatment of cancer, either as standalone intervention or adjunct to radiotherapy and chemotherapy. Water dispersible magnetic nanoparticles (MNPs) of ferrites (AFe2O4; A = Fe, Mn, Co) are the promising candidates for magnetic hyperthermia due to their high chemical stability, biocompatibility, moderate magnetization and high specific absorption rates (SAR). In this article, we have evaluated magnetic hyperthermia efficiency of water based magnetic fluids of AFe2O4. AFe2O4 NPs were synthesized by chemical co-precipitation method. Nanoparticles were coated with a bilayer of oleic acid and dispersed in water. Structural and magnetic properties of MNPs were investigated by X-Ray diffraction (XRD) and vibrating sample magnetometer (VSM). XRD study revealed that AFe2O4 NPs exhibits cubic inverse spinel structure. Fe3O4 (Ms = 48 emu/g, Mr = 2.60 emu/g, Hc = 49 Oe), MnFe2O4 (Ms = 40 emu/g, Mr = 2.60 emu/g, Hc = 35 Oe) and CoFe2O4 (44 emu/g, Mr = 10.10 emu/g, Hc =440 Oe) NPs exhibits soft ferromagnetic behaviour. Magnetic hyperthermia measurements were performed as a function of magnetic field strength (2–10 mT), field frequency (162–935.6 kHz) for 10 minutes. MNPs exhibits highest SAR values for 10 mT field strength at 935.6 kHz. Amongst the tested MNPs, Fe3O4 possess the highest SAR value (27.35 W/g), followed by MnFe2O4 (1.91 W/g) and CoFe2O4 (0.94 W/g). Considering this, it is concluded that amongst the inverse spinel ferrites AFe2O4, Fe3O4 NPs and most suitable for magnetic hyperthermia applications. [ABSTRACT FROM AUTHOR]

The cancer therapy with the lowest possible toxicity is today an issue that raises major difficulties in treating malignant tumors because chemo- and radiotherapy currently used in this field have a high degree of toxicity and in many cases are ineffective. Therefore, alternative solutions are rapidly being sought in cancer therapy, in order to increase efficacy and a reduce or even eliminate toxicity to the body. One of the alternative methods that researchers believe may be the method of the future in cancer therapy is superparamagnetic hyperthermia (SPMHT), because it can be effective in completely destroying tumors while maintaining low toxicity or even without toxicity on the healthy tissues. Superparamagnetic hyperthermia uses the natural thermal effect in the destruction of cancer cells, obtained as a result of the phenomenon of superparamagnetic relaxation of the magnetic nanoparticles (SPMNPs) introduced into the tumor; SPMNPs can heat the cancer cells to 42–43 °C under the action of an external alternating magnetic field with frequency in the range of hundreds of kHz. However, the effectiveness of this alternative method depends very much on finding the optimal conditions in which this method must be applied during the treatment of cancer. In addition to the type of magnetic nanoparticles and the biocompatibility with the biological tissue or nanoparticles biofunctionalization that must be appropriate for the intended purpose a key parameter is the size of the nanoparticles. Also, establishing the appropriate parameters for the external alternating magnetic field (AMF), respectively the amplitude and frequency of the magnetic field are very important in the efficiency and effectiveness of the magnetic hyperthermia method. This paper presents a 3D computational study on specific loss power ( P s ) and heating temperature ( Δ T ) which allows establishing the optimal conditions that lead to efficient heating of Fe3O4 nanoparticles, which were found to be the most suitable for use in superparamagnetic hyperthermia (SPMHT), as a non-invasive and alternative technique to chemo- and radiotherapy. The size (diameter) of the nanoparticles (D), the amplitude of the magnetic field (H) and the frequency (f) of AMF were established in order to obtain maximum efficiency in SPMHT and rapid heating of magnetic nanoparticles at the required temperature of 42–43 °C for irreversible destruction of tumors, without affecting healthy tissues. Also, an analysis on the amplitude of the AMF is presented, and how its amplitude influences the power loss and, implicitly, the heating temperature, observables necessary in SPMHT for the efficient destruction of tumor cells. Following our 3D study, we found for Fe3O4 nanoparticles the optimal diameter of ~16 nm, the optimal range for the amplitude of the magnetic field of 10–25 kA/m and the optimal frequency within the biologically permissible limit in the range of 200–500 kHz. Under the optimal conditions determined for the nanoparticle diameter of 16.3 nm, the magnetic field of 15 kA/m and the frequency of 334 kHz, the magnetite nanoparticles can be quickly heated to obtain the maximum hyperthermic effect on the tumor cells: in only 4.1–4.3 s the temperature reaches 42–43 °C, required in magnetic hyperthermia, with major benefits in practical application in vitro and in vivo, and later in clinical trials. There are significant challenges in developing drug carriers for therapeutic perspective. We have investigated a novel nanocarrier system, based on combining functionalized magnetic nanocomposite with Metal–Organic Frameworks (MOFs). Magnetic nanoparticles modified using biocompatible copolymers may be suitable for delivering hydrophobic drugs, such as cisplatin. Furthermore, compared to polymeric nanocarriers, nanocomposite constructed from zeolitic imidazolate framework-8 (ZIF-8) have demonstrated better drug loading capacity, as well as excellent pH-triggered drug release. Cisplatin-encapsulated Fe3O4@SiO2-ZIF-8@N-Chit-FA has been evaluated to determine the antitumor effects of free cisplatin enhancement in cervical cancer cells. In order to increase the stability of the proposed nanocarrier in aqueous solutions, in addition to the density of functional groups, a nano-chitosan layer was coated on top of the magnetic nanocomposite. It was then added with cisplatin onto the surface of Fe3O4@SiO2-ZIF-8@N-Chit-FA to deliver anticancer treatment that could be targeted using a magnetic field. A mouse isograft model of TC1 cells was used to evaluate the in vivo tumor growth inhibition. In tumor-bearing mice, Fe3O4@SiO2-ZIF-8@N-Chit-FA-cisplatin was injected intraperitoneally, and the targeted delivery was amplified by an external magnet (10 mm by 10 mm, surface field strength 0.4 T) fixed over the tumor site. Based on in vivo results, cisplatin-Loaded Mesoporous Magnetic Nanobiocomposite inhibited the growth of cervical tumors (P < 0.001) through the induction of tumor necrosis (P < 0.05) when compared to cisplatin alone. With the application of an external magnetic field, the drug was demonstrated to be able to induce its effects on specific target areas. Treatment of certain diseases requires the administration of drugs at specific areas of tissues and/or organs to increase therapy effectiveness and avoid side effects that may harm the rest of the body. Drug targeting is a research field that uses various techniques to administrate therapies at specific areas of the body, including magnetic systems able to drive nano "vehicles", as well as magnetically labeled molecules, in human body fluids and tissues. Most available actuation systems can only attract magnetic elements in a relatively small workspace, limiting drug target applications to superficial tissues, and leaving no alternative cases where deep targeting is necessary. In this paper, we propose an electromagnetic actuation system able to push and deflect magnetic particles at distance of ~10 cm, enabling the manipulation of magnetic nano- and microparticles, as well as administration of drugs in tissues, which are not eligible for localized drug targeting with state-of-the-art systems. Laboratory experiments and modeling were conducted to prove the effectiveness of the proposed system. By further implementing our device, areas of the human body that previously were impossible to treat with magnetically labeled materials such as drugs, cells, and small molecules can now be accessible using the described system. Cancer is one of the major health issues with increasing incidence worldwide. In spite of the existing conventional cancer treatment techniques, the cases of cancer diagnosis and death rates are rising year by year. Thus, new approaches are required to advance the traditional ways of cancer therapy. Currently, nanomedicine, employing nanoparticles and nanocomposites, offers great promise and new opportunities to increase the efficacy of cancer treatment in combination with thermal therapy. Nanomaterials can generate and specifically enhance the heating capacity at the tumor region due to optical and magnetic properties. The mentioned unique properties of nanomaterials allow inducing the heat and destroying the cancerous cells. Silk fibroin is an FDA approved biopolymer for clinical applications with great potential in nanomedicine. However, silk-based nanoformulations are still facing several challenges in processing and drug delivery efficiency (such as reproducibility and targetability), especially in cancer therapy. To address these challenges, robust and controllable production methods are required for generating nanocarriers with desired properties. This study aimed to develop a novel method for the production of peptide-functionalized magnetic silk nanoparticles with higher selectivity for cancer cells for targeted delivery of the hydrophobic anticancer agent ASC-J9. A new microfluidic device with a swirl mixer was designed to fabricate magnetic silk nanoparticles (MSNP) with desired size and narrow size distribution. The surface of MSNPs was functionalized with a cationic amphiphilic anticancer peptide, G(IIKK) 3 I-NH 2 (G3), to enhance their selectivity towards cancer cells. The G3-MSNPs increased the cellular uptake and anticancer activity of G3 in HCT 116 colorectal cancer cells compared to free G3. Moreover, the G3-MSNPs exhibited considerably higher cellular uptake and cytotoxicity in HCT 116 colorectal cancer cells compared to normal cells (HDFs). Encapsulating ASC-J9 in G3-MSNPs resulted in augmented anticancer activity compared to free ASC-J9 and non-functionalized ASC-J9 loaded MSNPs within its biological half-life. Hence, functionalizing MSNPs with G3 enabled targeted delivery of ASC-J9 to cancer cells and enhanced its anticancer effect. Functionalization of nanoparticles with anticancer peptides could be regarded as a new strategy for targeted delivery and enhanced efficiency of anticancer drugs. Magnetic Fe3O4 nanoparticles (MNPs) have great potential for nucleic acid separation, detection, and delivery. MNPs are considered a valuable tool in biomedicine due to their cost-effectiveness, stability, easy surface functionalization, and the possibility of the manipulations under a magnetic field. Herein, the synthesis of magnetic nylon 6 nanocomposites (MNPs@Ny6) was investigated. Transmission electron microscopy (TEM) was used for morphology and size analysis. A new method of UV-induced immobilization of oligonucleotides on MNPs@Ny6 for nucleic acid magnetic separation was proposed. MNPs@Ny6 shows a high oligonucleotide binding capacity of 2.2 nmol/mg with 73.3% loading efficiency. The proposed system has been applied to analyze model mixtures of target RNA on the total yeast RNA background. The RNA target isolation efficiency was 60% with high specificity. The bind RNA release was 88.8% in a quantity of 0.16 nmol/mg. The total RNA capture efficiency was 53%. General principles are developed using a finite element model regarding how time-dependent power dissipation of magnetic nanoparticles can be used to optimize hyperthermia selectivity. To make the simulation more realistic, the finite size and spatial location of each individual nanoparticle is taken into consideration. When energy input into the system and duration of treatment is held constant, increasing the maximum power dissipation of nanoparticles increases concentrations of energy in the tumor. Furthermore, when the power dissipation of magnetic nanoparticles rises linearly, the temperature gradient on the edge of the tumor increases exponentially. With energy input held constant, the location and duration of maximum power dissipation in the treatment time scheme will affect the final energy concentration inside the tumor. This review tries to summarize the purpose of steadily developing surface-functionalized nanoparticles for various bio-applications and represents a fascinating and rapidly growing field of research. Due to their unique properties—such as novel optical, biodegradable, low-toxicity, biocompatibility, size, and highly catalytic features—these materials are considered superior, and it is thus vital to study these systems in a realistic and meaningful way. However, rapid aggregation, oxidation, and other problems are encountered with functionalized nanoparticles, inhibiting their subsequent utilization. Adequate surface modification of nanoparticles with organic and inorganic compounds results in improved physicochemical properties which can overcome these barriers. This review investigates and discusses the iron oxide nanoparticles, gold nanoparticles, platinum nanoparticles, silver nanoparticles, and silica-coated nanoparticles and how their unique properties after fabrication allow for their potential use in a wide range of bio-applications such as nano-based imaging, gene delivery, drug loading, and immunoassays. Magnetic ferrite-based nanoparticles have recently emerged as a promising alternative choice for the treatment of different types of cancer. Magnetic ferrite-based nanoparticles have already been used in diagnosis of various diseases and as a drug delivery vehicle. In this study, spinel ferrite nanoparticles were prepared using a hydrothermal approach which is a simple and cost-effective technique. Spinel ferrite nanoparticles were prepared from a variety of divalent metals including nickel, zinc, copper and cobalt. The impact of these nanoparticles on the composition of human blood serum was evaluated in vitro. Fourier-transform infrared and ultraviolet-visible absorption spectroscopies were used to examine the blood serum. According to the findings, all of the magnetic ferrite-based nanoparticles studies showed protein corona formation. The development of corona protein was due to the affinity of magnetic ferrite-based nanoparticles to interact with protein and size independent. The magnetic ferrite-based nanoparticles had no effect on the amino acid makeup of the serum. The magnetic ferrite-based nanoparticles exerted their efficacy by releasing a small amount of metal oxides into the serum. Besides the study and the medical application of iron oxide nanoparticles, ferrites produced with zinc and manganese are of particular interest for their properties. The introduction of these elements into the crystalline structure of the magnetic particle generates some changes in the material properties, enhancing their potential use in theranostic applications. With the growing demands for personalized medicine and medical devices, nanomedicine is a modern scientific field, and research continues to apply nanomaterials for therapeutic and damaged tissue diagnosis. In this regard, substantial progress has been made in synthesizing magnetic nanoparticles with desired sizes, chemical composition, morphologies, and surface chemistry. Among these materials, nanomagnetic iron oxides have demonstrated promise as unique drug delivery carriers due to cancer treatment. This carrier could lead to responsive properties to a specific trigger, including heat, pH, alternative magnetic field, or even enzymes, through functionalization and coating of magnetic nanoparticles, along with biocompatibility, good chemical stability, easy functionalization, simple processing, and ability to localize to the tumor site with the assistance of external magnetic field. Current studies have focused on magnetic nanoparticles' utilities in cancer therapy, especially for colorectal cancer. Additionally, a bibliometric investigation was performed on the public trends in the field of the magnetic nanoparticle to drug delivery and anticancer, which represented progressing applications of these carriers in the multidisciplinary zones with a general view on future research and identified potential opportunities and challenges. For application of nanoparticles (NPs) as delivery vectors for biomedical applications the size, charge, surface chemistry and aggregation of the particles are one of key parameters. We focused on characterization of maghemite and cobalt ferrite magnetic NPs by measuring size distribution and zeta potential in different culture media and PBS. We show that our NPs functionalized with PAA are relatively very stable also in different culture media, where level of aggregation depends on medium composition. Effect of divalent ions and serum presence on stability is specifically analysed and we present possible destabilization mechanisms. We show that stability of electrostatically stabilized suspensions is affected by the molar concentration and valence of destabilizing counterions like Na+ or Mg2+ where effective surface charge of nanoparticles and thus repulsion force is screened by these counterions. In agreement with other papers we demonstrate significant effect of media composition (divalent ions, serum) and thus show the importance of NPs characterization under conditions that are representative of cell culture media or physiological conditions for understanding of NPs interaction with biological systems and for assessments of nanoparticle cytotoxicity. Abstract: Engineered inorganic nanoparticles are essential components in the development of nanotechnologies. For applications in nanomedicine, particles need to be functionalized to ensure a good dispersibility in biological fluids. In many cases however, functionalization is not sufficient: the particles become either coated by a corona of serum proteins or precipitate out of the solvent. In the present paper, we show that by changing the coating of iron oxide nanoparticles from a low-molecular weight ligand (citrate ions) to small carboxylated polymers (poly(acrylic acid)), the colloidal stability of the dispersion is improved and the adsorption/internalization of iron toward living mammalian cells is profoundly affected. Citrate-coated particles are shown to destabilize in all fetal-calf-serum based physiological conditions tested, whereas the polymer coated particles exhibit an outstanding dispersibility as well as a structure devoid of protein corona. The interactions between nanoparticles and human lymphoblastoid cells are investigated by transmission electron microscopy and flow cytometry. Two types of nanoparticle/cell interactions are underlined. Iron oxides are found either adsorbed on the cellular membranes, or internalized into membrane-bound endocytosis compartments. For the precipitating citrate-coated particles, the kinetics of interactions reveal a massive and rapid adsorption of iron oxide on the cell surfaces. The quantification of the partition between adsorbed and internalized iron was performed from the cytometry data. Magnetic nanoparticle's different applications in nanomedicine, due to their unique physical properties and biocompatibility, were intensively investigated. Recently, Fe3O4 nanoparticles, are confirmed to be the best sonosensitizers to enhance the performance of HIFU (high intensity focused ultrasound). They are also used as thermo-sensitizers in magnetic hyperthermia. A new idea of dual, magneto-ultrasound, coupled hyperthermia allows the ultrasound intensity to be reduced from the high to a moderate level. Our goal is to evaluate the enhancement of thermal effects of focused ultrasound of moderate intensity due to the presence of nanoparticles. We combine experimental results with numerical analysis. Experiments are performed on tissue-mimicking materials made of the 5% agar gel and gel samples containing Fe3O4 nanoparticles with φ = 100 nm with two fractions of 0.76 and 1.53% w/w. Thermocouples registered curves of temperature rising during heating by focused ultrasound transducer with acoustic powers of the range from 1 to 4 W. The theoretical model of ultrasound-thermal coupling is solved in COMSOL Multiphysics. We compared the changes between the specific absorption rates (SAR) coefficients determined from the experimental and numerical temperature rise curves depending on the nanoparticle fractions and applied acoustic powers. We confirmed that the significant role of nanoparticles in enhancing the thermal effect is qualitatively similarly estimated, based on experimental and numerical results. This paper describes a magnetic nanotechnology that locally enables hyperthermia treatment of hollow organ tumors by using polymer hybrid stents with incorporated magnetic nanoparticles (MNP). The hybrid stents are implanted and activated in an alternating magnetic field to generate therapeutically effective heat, thereby destroying the tumor. Here, we demonstrate the feasibility of nanomagnetic actuation of three prototype hybrid stents for hyperthermia treatment of hollow organ tumors. The results show that the heating efficiency of stent filaments increases with frequency from approximately 60 W/gFe (95 kHz) to approximately 250 W/gFe (270 kHz). The same trend is observed for the variation of magnetic field amplitude; however, heating efficiency saturates at approximately 30 kA/m. MNP immobilization strongly influences heating efficiency showing a relative difference in heating output of up to 60% compared to that of freely dispersed MNP. The stents showed uniformly distributed heat on their surface reaching therapeutically effective temperatures of 43 °C and were tested in an explanted pig bile duct for their biological safety. Objectives: The project is based on extensive studies on applied nanoparticles in biology and medicine. This study was primarily designed to investigate the role of magnetic nanoparticles by comparing the absorbance and stabilization of DNA-C, L by the electrophoretic detection. Methods: Fe3O4 magnetic nanoparticles (MNPs) with core/shell structure of silica coatings were synthesized by a chemical coprecipitation method. This work is done at 15 min times with multitimes, that is, 20 numbers and nanoparticles are provided regular with good morphology which was synthesized in 20 nm in size, and its structure was analyzed by tools such as EDX analysis. Next, 20 mg of the magnetic nanoparticles were coated with silica in a heterogeneous solution at 25 μg/ml of the solution of each of the mixed DNAs (ring, linear) in separate containers. Finally, 15 minutes later, DNA was adsorbed on the surface of the nanoparticles. The amount of this adsorption was injected by spectrophotometry (UV-Vis, ith 99% accuracy and optimized by the standard Tris.HCl buffer required to separate DNA from its pure solution [unabsorbed DNA]) and electrophoresis. Results: The results showed that absorption and diffusion of DNA-C or L at the surface of nanoparticles were 95% and 85%, respectively (i.e., absorbance of DNA-C>DNA-L is) with rate of removing of on MNPs was >99%. Hence, after review, we received that a linkage of electrostatic bonding between nanoparticles and biomolecules was obtained, and the results of the EDX analysis confirmed this study. Conclusion: In this project, nanocomposites containing magnetic nanoparticles were synthesized and their structure was identified by relevant analyzes. Nanotechnology-based therapeutic approaches have attracted attention of scientists, in particular due to the special features of nanomaterials, such as adequate biocompatibility, ability to improve therapeutic efficiency of incorporated drugs and to limit their adverse effects. Among a variety of reported nanomaterials for biomedical applications, metal and metal oxide-based nanoparticles offer unique physicochemical properties allowing their use in combination with conventional antimicrobials and as magnetic field-controlled drug delivery nanocarriers. An ever-growing number of studies demonstrate that by combining magnetic nanoparticles with membrane-active, natural human cathelicidin-derived LL-37 peptide, and its synthetic mimics such as ceragenins, innovative nanoagents might be developed. Between others, they demonstrate high clinical potential as antimicrobial, anti-cancer, immunomodulatory and regenerative agents. Due to continuous research, knowledge on pleiotropic character of natural antibacterial peptides and their mimics is growing, and it is justifying to stay that the therapeutic potential of nanosystems containing membrane active compounds has not been exhausted yet. Cancer is considered as one of the most challenging health care problems. Though there are many approved drugs that can be used for cancer therapy, drug resistance and delivery are among of the barriers of the treatment. In addition, pathological characteristics of tumors and their abnormal blood vessel architecture and function also reduce the efficiency of the conventional cancer treatment. Therefore, looking for techniques that can increase the efficacy of the therapy such as nanoparticles (NPs) is vital. NPs have many properties such as their small size, ability to load various drugs and large surface area, and ability to increase the absorption of conjugated. Therefore, the NPs have been considered as excellent tumor-targeting vehicles. The recent nanoscale vehicles include liposomes, polymeric nanoparticles, magnetic nanoparticles, dendrimers, and nanoshells; lipid-based NPs have been used as conjugates. There are few examples of approved conjugated anticancer NPs including AmBisome® (amphotericin B liposomal) and Doxil® (liposomal doxorubicin). There are many other conjugated anticancer drugs at different stages of clinical trials for treatment of various cancers. This review will discuss the properties of different NPs in cancer treatment and their benefits of overcoming multidrug resistance. In addition, recent advances of using nanomedicine in different approaches of cancer treatment such as chemotherapy, radiotherapy, and immunotherapy will be highlighted in this review. Objective(s): One applications of nanotechnology is in the area of medicine which is called nanomedicine. Primary instruments in nanomedicine can help us to detect diseases and used for drug delivery to inaccessible areas of human tissues. An important issue in simulating the motion of nanoparticles is modeling blood flow as a Newtonian or non-Newtonian fluid. Sometimes blood flow is simulated as a Newtonian fluid but actually blood has nature of non-Newtonian fluid. Simulation of targeted drug delivery and motion of nanoparticles in the blood flow as Newtonian and non-Newtonian fluid flow is investigated in this paperMaterials and Methods: In this paper, the blood flow is modeled as both Newtonian and non-Newtonian fluid and the effects of each case on the motion of nanoparticles in blood flow and targeted drug delivery is investigated. The flow is modeled with finite volume method. The particle modeled with discrete phase model.Results: Cross, Herschel-Bulkley and Power-law models are used for simulating the non-Newtonian blood flow. Numerical simulations show that trajectory of nanoparticle’s movement and the required time to pass the vessel by blood flow is variable for different models. According to obtained results, non-Newtonian Power-law and Herschel-Bulkley models have closely similar results but they have significant differences compared with Newtonian model. Conclusion: According to the results, it is preferred in the simulation to model blood flow as a non-Newtonian fluid and uses one of Herschel- Bulkley or Power-law models. Otherwise the simulation is far different from real phenomena.

Novel magnetite-ammonium bisphosphonate graft ionic copolymer nanocomplexes (MGICs) have been developed for potential drug delivery, magnetic resonance imaging, and hyperthermia applications. The complexes displayed relatively uniform sizes with narrow size distributions upon self-assembly in aqueous media, and their sizes were stable under simulated physiological conditions for at least 7 days. The anticancer drugs, cisplatin and carboplatin, were loaded into the complexes, and sustained release of both drugs was observed. The transverse NMR relaxivities (r2s) of the complexes were 244 s−1 (mM Fe)−1 which is fast compared to either the commercial T2-weighted MRI agent Feridex IV® or our previously reported magnetite-block ionomer complexes. Phantom MRI images of the complexes demonstrated excellent negative contrast effects of such complexes. Thus, the bisphosphonate-bearing MGICs could be promising candidates for dual drug delivery and magnetic resonance imaging. Moreover, the bisphosphonate MGICs generate heat under an alternating magnetic field of 30 kA·m−1 at 206 kHz. The temperature of the MGIC dispersion in deionized water increased from 37 to 41°C after exposure to the magnetic field for 10 minutes, corresponding to a specific absorption rate of 77.0 W·g−1. This suggests their potential as hyperthermia treatment agents as well as the possibility of temperature-dependent drug release, making MGICs more versatile in potential drug delivery applications. Polydopamine (PDA)-coated magnetic nanoparticles functionalized with mono-6-thio-β-cyclodextrin (SH-βCD) were obtained and characterized by transmission electron microscopy (TEM), Fourier transform infrared spectroscopy (FT-IR), X-ray photoelectron spectroscopy (XPS), Nuclear and Magnetic Resonance Imaging (NMR and MRI), and doxorubicin (DOXO)-loading experiments. The liver cancer cellular internalization of DOXO-loaded nanoparticles was investigated by confocal imaging microscopy. Synthesized nanomaterials bearing a chemotherapeutic drug and a layer of polydopamine capable of absorbing near-infrared light show high performance in the combined chemo- and photothermal therapy (CT-PTT) of liver cancer due to the synergistic effect of both modalities as demonstrated in vitro. Moreover, our material exhibits improved T2 contrast properties, which have been verified using Carr-Purcell-Meiboom-Gill pulse sequence and MRI Spin-Echo imaging of the nanoparticles dispersed in the agarose gel phantoms. Therefore, the presented results cast new light on the preparation of polydopamine-based magnetic theranostic nanomaterials, as well as on the proper methodology for investigation of magnetic nanoparticles in high field MRI experiments. Background: Vascular endothelial growth factor is well known for its angiogenesis potential. The study was performed to determine the possible pro-angiogenic role of magnetic nanoparticles coupled to VEGF in vitro and their capacity to cross an endothelial monolayer. This novel treatment technique for angiogenesis could be potentially useful for therapeutic purposes using magnetic nanoparticles.Methods: Magnetic nanoparticles (MN) were synthesized and were conjugated with the vascular endothelial growth factor. The particles were tested in vitro in a 2D to 3D culture system. MN was seeded in different positions in relation to an HUVEC spheroid to assess a preferential migration. To evaluate the MN capacity to cross the endothelial barrier, a confluent monolayer of HUVEC cells was seeded on top of a collagen gel. MN was placed in dissolution on the cell culture media, and the MN position was determined by confocal microscopy for 24 h.Results: HUVEC spheroids were able to generate a preferential sprouting depending on the MN position. Meanwhile, there was random migration when the MN's were placed all over the collagen gel and no sprouting when no MN was added. The trans-endothelial migration capacity of the MN was observed after 20 h in culture in the absence of external stimuli.Conclusion: Here we show in vitro angiogenesis following the distribution of the MN conjugated with growth factors. These nanoparticles could be controlled with a magnet to place them in the ischemic area of interest and speed up vascular recovery. A new application of mass-spectrometric methods to the identification, characterization, and analysis of nanomaterials developed or used in medicine for diagnostics, treatment, and (or) target-oriented drug delivery is considered. The main advantage of mass spectrometry (MS) in this exponentially developed area is the possibility of obtaining specific elemental and molecular information on the composition, structure, chemical state, and concentration of nanoparticles based on metals and their compounds. In combination with different separation techniques, MS can be used for the monitoring of formation and identification of particle conjugates with both target and transport biomolecules. Attention is focused on inductively coupled plasma MS and its versions suitable for the material analysis of metal particles, including multipurpose (or theranostic) particles. Realizing the full potential of magnetic nanoparticles (MNPs) in nanomedicine requires the optimization of their physical and chemical properties. Elucidation of the effects of these properties on clinical diagnostic or therapeutic properties, however, requires the synthesis or purification of homogenous samples, which has proved to be difficult. While initial simulations indicated that size-selective separation could be achieved by flowing magnetic nanoparticles through a magnetic field, subsequent in vitro experiments were unable to reproduce the predicted results. Magnetic field-flow fractionation, however, was found to be an effective method for the separation of polydisperse suspensions of iron oxide nanoparticles with diameters greater than 20 nm. While similar methods have been used to separate magnetic nanoparticles before, no previous work has been done with magnetic nanoparticles between 20 and 200 nm. Both transmission electron microscopy (TEM) and dynamic light scattering (DLS) analysis were used to confirm the size of the MNPs. Drug delivery to the brain has been one of the toughest challenges researchers have faced to develop effective treatments for brain diseases. Owing to the blood–brain barrier (BBB), only a small portion of administered drug can reach the brain. A consequence of that is the need to administer a higher dose of the drug, which, expectedly, leads to a variety of unwanted side effects. Research in a variety of different fields has been underway for the past couple of decades to address this very serious and frequently lethal problem. One area of research that has produced optimistic results in recent years is nanomedicine. Nanomedicine is the science birthed by fusing the fields of nanotechnology, chemistry and medicine into one. Many different types of nanomedicine-based drug-delivery systems are currently being studied for the sole purpose of improved drug delivery to the brain. This review puts together and briefly summarizes some of the major breakthroughs in this crusade. Inorganic nanoparticle-based drug-delivery systems, such as gold nanoparticles and magnetic nanoparticles, are discussed, as well as some organic nanoparticulate systems. Amongst the organic drug-delivery nanosystems, polymeric micelles and dendrimers are discussed briefly and solid polymeric nanoparticles are explored in detail. [ABSTRACT FROM AUTHOR]

Humans are exposed to a complex mix of man-made electric and magnetic fields (MFs) at many different frequencies, at home and at work. Epidemiological studies indicate that there is a positive relationship between residential/domestic and occupational exposure to extremely low frequency electromagnetic fields and some types of cancer, although some other studies indicate no relationship. In this review, after an introduction on the MF definition and a description of natural/anthropogenic sources, the epidemiology of residential/domestic and occupational exposure to MFs and cancer is reviewed, with reference to leukemia, brain, and breast cancer. The in vivo and in vitro effects of MFs on cancer are reviewed considering both human and animal cells, with particular reference to the involvement of reactive oxygen species (ROS). MF application on cancer diagnostic and therapy (theranostic) are also reviewed by describing the use of different magnetic resonance imaging (MRI) applications for the detection of several cancers. Finally, the use of magnetic nanoparticles is described in terms of treatment of cancer by nanomedical applications for the precise delivery of anticancer drugs, nanosurgery by magnetomechanic methods, and selective killing of cancer cells by magnetic hyperthermia. The supplementary tables provide quantitative data and methodologies in epidemiological and cell biology studies. Although scientists do not generally agree that there is a cause-effect relationship between exposure to MF and cancer, MFs might not be the direct cause of cancer but may contribute to produce ROS and generate oxidative stress, which could trigger or enhance the expression of oncogenes. The fate of nanoparticle (NP) formulations in the multifaceted biological environment is a key determinant of their biocompatibility and therapeutic performance. An understanding of the degradation patterns of different types of clinically used and experimental NP formulations is currently incomplete, posing an unmet need for novel analytical tools providing unbiased quantitative measurements of NP disassembly directly in the medium of interest and in conditions relevant to specific therapeutic/ diagnostic applications. In the present study, this challenge was addressed with an approach enabling real-time in situ monitoring of the integrity status of NPs in cells and biomimetic media using Förster resonance energy transfer (FRET). Disassembly of polylactidebased magnetic NPs (MNPs) was investigated in a range of model biomimetic media and in cultured vascular cells using an experimentally established quantitative correlation between particle integrity and FRET efficiency controlled through adjustments in the spectral overlap between two custom-synthesized polylactide–fluorophore (boron dipyrromethene) conjugates incorporated in MNPs. The results suggest particle disassembly governed by diffusion–reaction processes with kinetics strongly dependent on conditions promoting release of oligomeric fragments from the particle matrix. Thus, incubation in gels simulating the extracellular environment and in protein-rich serum resulted in notably lower and higher MNP decomposition rates, respectively, compared with nonviscous liquid buffers. The diffusion–reaction mechanism also is consistent with a significant cell growth-dependent acceleration of MNP processing in dividing vs. contact-inhibited vascular cells. The FRET-based analytical strategy and experimental results reported herein may facilitate the development and inform optimization of biodegradable nanocarriers for cell and drug delivery applications. For decades now, conventional sinusoidal signals have been exclusively used in magnetic hyperthermia as the only alternating magnetic field waveform to excite magnetic nanoparticles. However, there are no theoretical nor experimental reasons that prevent the use of different waveforms. The only justifiable motive behind using the sinusoidal signal is its availability and the facility to produce it. Following the development of a configurable alternating magnetic field generator, we aim to study the effect of various waveforms on the heat production effectiveness of magnetic nanoparticles, seeking to prove that signals with more significant slope values, such as the trapezoidal and almost-square signals, allow the nanoparticles to reach higher efficiency in heat generation. Furthermore, we seek to point out that the nanoparticle power dissipation is dependent on the waveform's slope and not only the frequency, magnetic field intensity and the nanoparticle size. The experimental results showed a remarkably higher heat production performance of the nanoparticles when exposed to trapezoidal and almost-square signals than conventional sinusoidal signals. We conclude that the nanoparticles respond better to the trapezoidal and almost-square signals. On the other hand, the experimental results were used to calculate the normalized power dissipation value and prove its dependency on the slope. The aim of the present work was to demonstrate the possibility of selective detection of nanoparticle contrast agents (NPCAs) on diagnostic echographic images by exploiting the second harmonic component they introduce in the spectra of corresponding ultrasound signals, as a consequence of nonlinear distortion during ultrasound propagation. We employed silica nanospheres (SiNSs) of variable diameter (160 nm, 330 nm, and 660 nm) dispersed in different volume concentrations (range 0.07-0.8%) in agarose gel samples that were automatically scanned through a digital ecograph using narrow-band ultrasound pulses at 6.6 MHz and variable mechanical index (MI range 0.2-0.6). In the first part of the study, the intensity peaks of four different spectral components of the backscattered signal were considered: fundamental (detected in correspondence of the incident ultrasound frequency), subharmonic (detected at half of the fundamental frequency), ultra harmonic (detected at 1.5 times the fundamental frequency), and second harmonic (detected at twice the fundamental frequency). Subsequently, based on the experimental results of the first part of the study and on our recently reported findings, the focus was moved to a detailed comparison between subharmonic and second harmonic trend, which were determined as a function of nanoparticle composition, sample concentration, and MI. The experiments were also repeated on different agarose samples, containing SiNSs covered by an outer shell of smaller magnetic nanoparticles, made of either iron oxide (IO) or FePt-IO nanocrystals. Obtained results show that this new ultrasound-based method for NPCA imaging has a detection sensitivity similar to that of our previously introduced subharmonic-based technique in the presence of 330-nm SiNSs, but performs significantly better in the detection of both the types of “dual mode” NPCAs. The fact that the reported detection method was optimized for identification of 330-nm SiNSs (a sort of “ideal” size for the development of novel tumor-targeting NPCAs) and that the magnetically coated particles are detectable also through magnetic resonance imaging makes the presented second harmonic ultrasound method a valuable solution for the introduction of new protocols for multimodal molecular diagnoses employing only nonionizing radiations.
