In the early 1900s, mutation breeding to select varieties with desirable traits using spontaneous mutation was actively conducted around the world, including Japan. In rice, the number of fixed mutations per generation was estimated to be 1.38-2.25. Although this low mutation rate was a major problem for breeding in those days, in the modern era with the development of next-generation sequencing (NGS) technology, it was conversely considered to be an advantage for efficient gene identification. In this paper, we proposed an in silico approach using NGS to compare the whole genome sequence of a spontaneous mutant with that of a closely related strain with a nearly identical genome, to find polymorphisms that differ between them, and to identify the causal gene by predicting the functional variation of the gene caused by the polymorphism. Using this approach, we found four causal genes for the dwarf mutation, the round shape grain mutation and the awnless mutation. Three of these genes were the same as those previously reported, but one was a novel gene involved in awn formation. The novel gene was isolated from Bozu-Aikoku, a mutant of Aikoku with the awnless trait, in which nine polymorphisms were predicted to alter gene function by their whole-genome comparison. Based on the information on gene function and tissue-specific expression patterns of these candidate genes, Os03g0115700/LOC_Os03g02460, annotated as a shortchain dehydrogenase/reductase SDR family protein, is most likely to be involved in the awnless mutation. Indeed, complementation tests by transformation showed that it is involved in awn formation. Thus, this method is an effective way to accelerate genome breeding of various crop species by enabling the identification of useful genes that can be used for crop breeding with minimal effort for NGS analysis. Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disorder that causes the progressive loss of motoneurons and, unfortunately, there is no effective treatment for this disease. Interconnecting multiple pathological mechanisms are involved in the neuropathology of this disease, including abnormal aggregation of proteins, neuroinflammation and dysregulation of the ubiquitin proteasome system. Such complex mechanisms, together with the lack of reliable animal models of the disease have hampered the development of drugs for this disease. Protein kinases, a key pharmacological target in several diseases, have been linked to ALS as they play a central role in the pathology of many diseases. Therefore several inhibitors are being currently trailed for clinical proof of concept in ALS patients. In this review, we examine the recent literature on protein kinase inhibitors currently in pharmaceutical development for this diseaseas future therapy for AS together with their involvement in the pathobiology of ALS. Following the discovery of the involvement of the ribonucleoprotein TDP‐43 in amyotrophic lateral sclerosis (ALS) and frontotemporal lobar degeneration (FTLD), a major research focus has been to develop treatments that can prevent or alleviate these disease conditions. One pharmacological approach has been to use TDP‐43‐based disease models to test small molecules and drugs already known to have some therapeutic effect in a variety of neurodegenerative conditions. In parallel, various disease models have been used to perform high‐throughput screens of drugs and small compound libraries. The aim of this review will be to provide a general overview of the compounds that have been described to alter pathological characteristics of TDP‐43. These include expression levels, cytoplasmic mis‐localization, post‐translational modifications, cleavage, stress granule recruitment and aggregation. In parallel, this review will also address the use of compounds that modify the autophagic/proteasome systems that are known to target TDP‐43 misfolding and aggregation. We investigated the pollution levels of 6 heavy metals and 29 dioxins (polychlorinated dibenzo-p-dioxins (PCDDs), polychlorinated dibenzofurans (PCDFs), and dioxin-like polychlorinated biphenyls (DL-PCBs)) in intertidal and supratidal zones by using wharf roaches (Ligia spp.) collected from 12 sampling sites on the coast of Northeast Japan from November 2011 to June 2012. The total concentrations of heavy metals ranged from 177 to 377 µg/g-dry weight (dw), and the predominant metals were copper, zinc, and aluminum. The order of the detected level of heavy metals was zinc > aluminum > copper > cadmium > lead > chromium, and this trend was similar to a previous report. We reviewed the long-term trends and inter-annual variations in the surface shortwave irradiance in China and Japan. Pyranometer observations revealed decreases followed by increases in the shortwave irradiance in China and Japan between the 1960s and 2000s, while obvious long-term trends were not evident in the satellite observations after 1983. In China, surface shortwave irradiance decreased from 1961 until around 1990, but then began to increase. In Japan, on the contrary, the decreasing trend stopped in the 1960s, with little inter-annual variation during the 1970s and 1980s, and an increase began around 1990. The causes of the differences between the shortwave irradiance trends in China and Japan were ascribed to an increase in light-absorbing aerosols in China that began in the 1960s and a decrease in absorbing aerosols in Japan that began in the late 1970s. Absorbing aerosols decrease both direct and diffuse radiation, while non-absorbing aerosols decrease direct radiation but increase diffuse radiation. Although these aerosol influences are generally found under clear sky conditions, absorbing aerosols could have direct effects even under cloudy sky conditions. The trends of surface shortwave irradiance in China and Japan are in line with the so-called global dimming and brightening dimming processes, although the phases of the minimum periods in the two regions slightly differed. An increase in anthropogenic aerosol was responsible for the variation in the shortwave irradiance through the direct radiative effect of aerosol in the polluted area, while indirect radiative effects, i.e., changes in cloud cover due to an increase in cloud condensation nuclei, dominated in pristine areas. The effects of other factors, such as variations in water vapor and natural aerosol levels, appear to be small compared to the effects of cloud and anthropogenic aerosols. [ABSTRACT FROM AUTHOR]
