In this work, 193 nm ultraviolet photodissociation-mass spectrometry (UVPD-MS) was applied to investigate supercharging reagent sulfolane in protein sequence and structure characterization. All datasets were collected by using an Orbitrap MS system equipped with an ArF 193 nm EX50 excimer Laser. The 193 nm UV laser has the advantages of high brightness, good stability, and propagate in air, which can directly excite and dissociate the protein backbone within 5 ns (single pulse). Previous reports demonstrated that the partial protein high-order structures and non-covalent interactions could be retained in the UVPD fragments and the site-specific fragmentation yields (FYs) revealed the dynamic protein structure alterations under different conditions. Therefore, the combination of UVPD with nMS provides a new research paradigm for the characterization of protein structure changes. In this work, we further demonstrated 193 nm UVPD was superior to high-energy collision induced dissociation (HCD) in fragmenting and sequencing the non-denatured carbonic anhydrase II (CA) generated in native electrospray ionization (ESI). UVPD could efficiently activate and fragmentate the non-denatured CA with different charge states in just 5 ns, achieving cleavage sequence coverage 79.8% for CA Z10. Then, 0.25% supercharging reagent sulfolane was added into the protein solution and the CA charge distribution was significantly moved to higher charge states under identical native ESI conditions. However, higher-order structures (HOS) and non-covalent interactions could be obtained as the CA charge distribution was still concentrated and significantly different from the spectra features of denatured CA. The UVPD cleavage sequence coverage of the CA was increased to 87.2%, including the regions that were difficult to be dissociated and characterized without sulfolane addition. In addition, the non-covalent of CA and Zn2+ remained stable, indicating that the addition of sulfoxide had little influence on the Zn2+ binding region. The UVPD sequence coverage of CA Z11 was increased by 7.4% after the addition of sulfoxide, but the site FYs were almost not changed, demonstrating the corresponding local structures were nearly not changed. However, the higher charge states introduced by sulfolane also exhibited the risk of inducing specific structure alteration of part of protein regions. In conclusion, it was infered that supercharging reagent is efficient in modulating the charge state distribution of proteins in native ESI-MS characterization and can expand the MS detection mass range. The introduction of supercharging reagent can further improve the UVPD sequence coverage, including the structure-compact regions hard to be fragmented. Although part of the protein HOS and non-covalent interactions seems retained, the influence of higher charge states on protein structures should be considered in dynamic protein structure investigation.

Abstract Background Tea trees originated in southwest China 60 million or 70 million years ago. Written records show that Chinese ancestors had begun drinking tea over 3000 years ago. Nowadays, with the aging of populations worldwide and more people suffering from non-communicable diseases or poor health, tea beverages have become an inexpensive and fine complementary and alternative medicine (CAM) therapy. At present, there are 3 billion people who like to drink tea in the world, but few of them actually understand tea, especially on its development process and the spiritual and cultural connotations. Methods We searched PubMed, Google Scholar, Web of Science, CNKI, and other relevant platforms with the key word “tea”, and reviewed and analyzed tea-related literatures and pictures in the past 40 years about tea’s history, culture, customs, experimental studies, and markets. Results China is the hometown of tea, tea trees, tea drinking, and tea culture. China has the oldest wild and planted tea trees in the world, fossil of a tea leaf from 35,400,000 years ago, and abundant tea-related literatures and art works. Moreover, tea may be the first Chinese herbal medicine (CHM) used by Chinese people in ancient times. Tea drinking has many benefits to our physical health via its antioxidant, anti-inflammatory, immuno-regulatory, anticancer, cardiovascular-protective, anti-diabetic, and anti-obesity activities. At the moment, COVID-19 is wreaking havoc across the globe and causing severe damages to people’s health and lives. Tea has anti-COVID-19 functions via the enhancement of the innate immune response and inhibition of viral growth. Besides, drinking tea can allow people to acquire a peaceful, relaxed, refreshed and cheerful enjoyment, and even longevity. According to the meridian theory of traditional Chinese medicine, different kinds of tea can activate different meridian systems in the human body. At present, black tea (fermented tea) and green tea (non-fermented tea) are the most popular in the world. Black tea accounts for over 90% of all teas sold in western countries. The world’s top-grade black teas include Qi Men black in China, Darjeeling and Assam black tea in India, and Uva black tea in Sri Lanka. However, all top ten famous green teas in the world are produced in China, and Xi Hu Long Jing tea is the most famous among all green teas. More than 700 different kinds of components and 27 mineral elements can be found in tea. Tea polyphenols and theaflavin/thearubigins are considered to be the major bioactive components of black tea and green tea, respectively. Overly strong or overheated tea liquid should be avoided when drinking tea. Conclusions Today, CAM provides an array of treatment modalities for the health promotion in both developed and developing countries all over the world. Tea drinking, a simple herb-based CAM therapy, has become a popular man-made non-alcoholic beverage widely consumed worldwide, and it can improve the growth of economy as well. Tea can improve our physical and mental health and promote the harmonious development of society through its chemical and cultural elements.

Objective: To investigate hypertriglyceridemia and hepatomegaly caused by Schisandrae Sphenantherae Fructus (FSS) and Schisandra chinensis Fructus (FSC) oils in mice. Methods: Mice were orally administered a single dose of Schisandrae Fructus oils. Serum and hepatic triglyceride (TG), triglyceride transfer protein (TTP), apolipoprotein B48 (Apo B48), very-low-density lipoprotein (VLDL), hepatocyte growth factor (HGF), alanine aminotransfease (ALT) and liver index were measured at 6-120 h post-dosing. Results: FSS and FSC oil caused time and dose-dependent increases in serum and hepatic TG levels, with maximum increases in the liver (by 297% and 340%) at 12 h post-dosing and serum (244% and 439%) at 24-h post-dosing, respectively. Schisandrae Fructus oil treatments also elevated the levels of serum TTP by 51% and 63%, Apo B48 by 152% and 425%, and VLDL by 67% and 38% in mice, respectively. FSS and FSC oil treatments also increased liver mass by 53% and 55% and HGF by 106% and 174%, but lowered serum ALT activity by 38% and 22%, respectively. Fenofibrate pre/ co-treatment attenuated the FSS and FSC oil-induced elevation in serum TG levels by 41% and 49% at 48 h post-dosing, respectively, but increased hepatic TG contents (by 38% and 33%, respectively) at 12 h post-dosing. Conclusions: Our findings provide evidence to support the establishment of a novel mouse model of hypertriglyceridemia by oral administration of FSS oil (mainly increasing endogenous TG) and FSC oil (mainly elevating exogenous TG).

As large-scale, high-proportion, and efficient distribution transformers surge into the grids, anti-short circuit capability testing of transformer windings in efficient distribution seems necessary and prominent. To deeply explore the influence of progressively short-circuit shock impulses on the core winding deformation of efficient power transformers, a finite element theoretical model was built by referring to a three-phase three-winding 3D wound core transformer with a model of S20-MRL-400/10-NX2. The distributions of internal equivalent force and total deformation of the 3D wound core transformer along different paths under progressively short-circuit shock impulses varying from 60% to 120% were investigated. Results show that the equivalent stress and total deformation change rate reach their maximum as the short-circuit current increases from 60% to 80%, and the maximum and average variation rate for the equivalent stress reach 177.75% and 177.43%, while the maximum and average variation rate for the total deformation corresponds to 178.30% and 177.45%, respectively. Meanwhile, the maximum equivalent stress and maximum total deformation reach 29.81 MPa and 38.70 μm, respectively, as the applied short-circuit current increased to 120%. In light of the above observations, the optimization and deployment of wireless sensor nodes was suggested. Therefore, a distributed monitoring system was developed for acquiring the vibration status of the windings in a 3D wound core transformer, which is a beneficial supplement to the traditional short-circuit reactance detection methods for an efficient grid access spot-check of distribution transformers. In recent years, the high-frequency hybrid sky-surface wave radar (HSSWR) has been increasingly used for target detection applications. Nevertheless, the specific bistatic system layout and the phase path disturbances induced by the ionospheric propagation channel may severely spread the sea clutter spectrum, thereby deteriorating the detection ability of the HSSWR for slow-moving targets. In this work, a novel subspace method based on the hybrid use of the amplitude and phase estimator (APES) and the orthogonal projection (OP) in the spatial–temporal domain, denoted as the APES-OP method, is proposed to suppress the spreading first-order sea clutter of the HSSWR. The distribution characteristics of targets and first-order sea clutter in the spatial–temporal domain were investigated, and a time-domain subspace signal model was adopted to describe targets perturbed by ionospheric phase path modulation. An APES filter was adopted to filter out the potential targets with a traversal approach to avoid attenuating desired signals while suppressing sea clutter. After that, sampling data from multi-channels and slow-time domains at the cell under test were employed to construct a spatial–temporal matrix, which was then utilized to obtain the sea clutter subspace by singular value decomposition. Simulation results indicate that the proposed algorithm can suppress sea clutter while retaining the target, even if the target is buried by sea clutter. The processing results of measured data further demonstrate the efficiency of the proposed algorithm. After sea clutter suppression, the target obscured by clutter can be revealed, and the signal-to-clutter ratio of the target is greatly improved. [ABSTRACT FROM AUTHOR]

The beta T-cell receptor (TRB) expressed by beta T cells is essential for foreign antigen recognition. The TRB locus contains a TRBV family that encodes three complementarity determining regions (CDRs). CDR1 is associated with antigen recognition and interactions with MHC molecules. In contrast to domestic pigs, African suids lack a 284-bp segment spanning exons 1 and 2 of the TRBV27 gene that contains a sequence encoding CDR1. In this study, we used the African swine fever virus (ASFV) as an example to investigate the effect of deleting the TRBV27-encoded CDR1 on the resistance of domestic pigs to exotic pathogens. We first successfully generated TRBV27-edited fibroblasts with disruption of the CDR1 sequence using CRISPR/Cas9 technology and used them as donor cells to generate gene-edited pigs via somatic cell nuclear transfer. The TRBV-edited and wild-type pigs were selected for synchronous ASFV infection. White blood cells were significantly reduced in the genetically modified pigs before ASFV infection. The genetically modified and wild-type pigs were susceptible to ASFV and exhibited typical fevers (>40 °C). However, the TRBV27-edited pigs had a higher viral load than the wild-type pigs. Consistent with this, the gene-edited pigs showed more clinical signs than the wild-type pigs. In addition, both groups of pigs died within 10 days and showed similar severe lesions in organs and tissues. Future studies using lower virulence ASFV isolates are needed to determine the relationship between the TRBV27 gene and ASFV infection in pigs over a relatively long period. [ABSTRACT FROM AUTHOR]

In the present study, two new Conidiobolus s.s. species were described relying on the morphological studies and phylogenetic analysis utilizing nuclear large subunit of rDNA (nucLSU), mitochondrial small subunit of rDNA (mtSSU), and elongation-factor-like gene (EFL) sequences. Conidiobolus jiangxiensis sp. nov. is distinguished by its short primary conidiophores, a feature not commonly observed in other Conidiobolus s.s. species. Conversely, Conidiobolus marcoconidius sp. nov. is characterized by larger primary conidia and the emergence of 2–5 secondary conidia from each branched secondary conidiophores. Additionally, the taxonomic reassessment of C. polyspermus confirms its distinct status within the genus Conidiobolus s.s. Moreover, molecular analyses, incorporating the nucLSU , mtSSU , and EFL sequences, provide robust support for the phylogenetic placement of the two newly described species and the taxonomic identity of C. polyspermus. This investigation contributes valuable insights into the species diversity of Conidiobolaceae in China, enhancing our understanding of the taxonomy within this fungal family. [ABSTRACT FROM AUTHOR]

This study developed a mobile app called Ubiquitous-Physics (U-Physics), which helps students explore inclined plane phenomena in authentic contexts and consolidates their physics learning in everyday contexts. The study investigated inquiry behaviors such as interpreting graphs, applying formulas, drawing conclusions, and peer sharing, and how these influenced learning achievements. The app's effects on learning perceptions and motivation were also analyzed. The study was conducted at a vocational senior high school and two activities (indoors and outdoors/authentic contexts). The results show that the experimental group using U-Physics significantly outperformed the control group, who used traditional tools such as a stopwatch for the inclined plane experiment in terms of learning achievements. Analysis of the correlations among inquiry behaviors and learning achievement revealed that the number of posts in authentic contexts had the greatest influence on learning achievement, confirming the importance of applying physics concepts in authentic contexts. Additionally, most students reported positive perceptions of the U-Physics app and were highly motivated to use it for future learning. The findings indicate that using U-Physics in authentic contexts is very meaningful and can contribute to physics learning, particularly when using mobile devices with sensors to facilitate inquiry learning in authentic physics scenarios.
