8. Dose-response relationship between daily screen time and the risk of low back pain among children and adolescents: a meta-analysis of 57831 participants

Background: The risk of low back pain (LBP) increases steeply during adolescence, and adolescents with LBP are more likely to have low back pain in their adult years. This study aimed to investigate the dose-response relationship between daily screen time and the risk of low back pain among children and adolescents. Methods: PubMed, the Cochrane Library, Embase, and Web of Science were searched to collect relevant studies on daily screen time and the risk of low back pain from the establishment of the database up to December 2022. Two investigators independently screened the literature, extracted data, and evaluated the risk of bias in the included studies. Stata16.0 was used to perform a dose-response meta-analysis and the methodological quality evaluation of the included studies. Results: The results of the meta-analysis showed that there is a positive correlation between daily computer time (OR = 1.32, 1.05–1.60), daily mobile phone time (OR = 1.32, 1.00–1.64), daily TV watching (OR = 1.07, 1.04–1.09) and the risk of low back pain, separately. The dose–response meta-analysis showed that there is a linear relationship between daily computer use and low back pain. The risk of low back pain increased by 8.2% for each 1-hour of daily computer use. Conclusions: Screen time is related to the risk of low back pain, and there is a linear relationship between daily computer use and the risk of low back pain. A number of strategic measures should be taken to prevent adolescents from developing severe low back pain.

Density is an important parameter for both geological research and geophysical exploration. However, for model-driven seismic inversion methods, high-fidelity density inversion is challenging due to seismic wave traveltime insensitivity to density and crosstalk that density has with velocity. To circumvent the challenge of density inversion, some inversion methods treat density as a constant value or derive density from velocity through empirical equation. On the other hand, deep learning approaches are completely driven by data and have strong target-oriented characteristics, offering a new way to solve multiparameter coupling problems. Nevertheless, the accuracy of the inversion results of data-driven algorithms is directly related to the amount and diversity of the training data, and thus, they lack the universality of model-driven algorithms. To achieve accurate density inversion, we propose a simultaneous inversion algorithm for velocity and density that combines the advantages of data- and model- driven approaches: A neural network model (U-T), combining the U-net and Transformer architectures, is proposed to construct non-linear mappings between seismic data as inputs and the velocity and density as predictions. Next, the model-driven inversion algorithm uses the U-T prediction as the initial model to obtain the final accurate solution. In the model-driven module, envelope-based sparse constrained deconvolution is used to obtain full-band seismic data, while a variable dominant frequency full waveform inversion algorithm is used to perform multiscale inversion, ultimately leading to accurate inversion results of velocity and density. 10. Comparing the Aging Processes of PLA and PE: The Impact of UV Irradiation and Water.

The aging processes of microplastics (MPs) are prevalent in natural environments. Understanding the aging mechanisms of MPs is crucial for assessing their environmental behavior and potential risks. In this study, we selected polylactic acid (PLA) and polyethylene (PE) as representatives of biodegradable and conventional plastics, respectively, to examine changes in their physicochemical properties induced by water and UV light exposure. Laboratory aging resulted in significant fragmentation, characterized by cracks and pores on the surfaces, for both types of MPs, with PLA MPs exhibiting more severe changes, particularly under combined UV and water exposure. Notably, PLA MPs tended to become progressively smaller after aging, whereas PE MPs did not show significant size changes. Chemical analyses of aged MPs using micro-Fourier-transform infrared spectroscopy (FTIR) and X-ray photoelectron spectroscopy (XPS) revealed a substantial increase in the carbonyl index (CI) and oxygen content for PE, suggesting surface oxidation during photo-oxidation. Conversely, PLA MPs displayed a CI decrease, along with an oxygen content increase, indicating the breakdown of ester linkages in PLA and the formation of other oxidation products. Furthermore, we developed and optimized pyrolysis–gas chromatography–mass spectrometry (Py–GC–MS) methods to identify potential chemical degradation products of PE and PLA, considering their differing thermal stabilities. We observed a distinct trend regarding the peaks in the chromatogram of aged MPs and identified the typical oxidation and crosslinking products for PLA. Additionally, after the aging process, both PE and PLA exhibited a significant increase in organic carbon content, with the eluate containing submicron/nano-sized particles. This study provides a scientific foundation for a deeper understanding of the environmental aging mechanisms of various MPs, particularly in regards to the effects of UV irradiation and water exposure. A quasi-continuous-wave (QCW) laser diode (LD) driver is commonly used to drive diode bars and stacks designed specifically for QCW operations in solid-state lasers. Such drivers are optimized to deliver peak current and voltage pulses to LDs while maintaining low average power levels. As a result, they are widely used in laser processing devices and laser instruments. Traditional high-energy QCW LD drivers primarily use capacitors as energy storage components and pulsed constant-current sources with op-amps and power metal-oxide-semiconductor field-effect transistors (MOSFETs) as their core circuits for generating repeated constant-current pulses. The drawback of this type of driver is that the driver's output voltage needs to be manually adjusted according to the operating voltage of the load before use to maximize driver efficiency while providing a sufficient current. Another drawback is its inability to automatically adjust the output voltage to maintain high efficiency when the load changes during the driver operation. Drastic changes in the load can cause the driver to fail to function properly in extreme cases. Based on the above traditional circuit structure, this study designed a stability compensation circuit and realized a QCW LD driver for driving a GS20 diode stack with a maximum repetition rate of 100 Hz, a constant current of approximately 300 A, a load voltage of approximately 10 V, and a pulse width of approximately 300 μ s. In particular, a high-efficiency, load-adaptive driving method was used with the MOSFETs in the critical saturation region (i.e., between the linear and saturated regions), controlling its power loss effectively while achieving maximum output current of the driver. The experiments demonstrated that the driver efficiency could be maintained at more than 80% when the load current varied from 50 to 300 A. [ABSTRACT FROM AUTHOR]

The core body temperature serves as a pivotal physiological metric indicative of sow health, with rectal thermometry prevailing as a prevalent method for estimating core body temperature within sow farms. Nonetheless, employing contact thermometers for rectal temperature measurement proves to be time-intensive, labor-demanding, and hygienically suboptimal. Addressing the issues of minimal automation and temperature measurement accuracy in sow temperature monitoring, this study introduces an automatic temperature monitoring method for sows, utilizing a segmentation network amalgamating YOLOv5s and DeepLabv3+, complemented by an adaptive genetic algorithm-random forest (AGA-RF) regression algorithm. In developing the sow vulva segmenter, YOLOv5s was synergized with DeepLabv3+, and the CBAM attention mechanism and MobileNetv2 network were incorporated to ensure precise localization and expedited segmentation of the vulva region. Within the temperature prediction module, an optimized regression algorithm derived from the random forest algorithm facilitated the construction of a temperature inversion model, predicated upon environmental parameters and vulva temperature, for the rectal temperature prediction in sows. Testing revealed that vulvar segmentation IoU was 91.50%, while the predicted MSE, MAE, and R

Sow body condition scoring has been confirmed as a vital procedure in sow management. A timely and accurate assessment of the body condition of a sow is conducive to determining nutritional supply, and it takes on critical significance in enhancing sow reproductive performance. Manual sow body condition scoring methods have been extensively employed in large-scale sow farms, which are time-consuming and labor-intensive. To address the above-mentioned problem, a dual neural network-based automatic scoring method was developed in this study for sow body condition. The developed method aims to enhance the ability to capture local features and global information in sow images by combining CNN and transformer networks. Moreover, it introduces a CBAM module to help the network pay more attention to crucial feature channels while suppressing attention to irrelevant channels. To tackle the problem of imbalanced categories and mislabeling of body condition data, the original loss function was substituted with the optimized focal loss function. As indicated by the model test, the sow body condition classification achieved an average precision of 91.06%, the average recall rate was 91.58%, and the average F1 score reached 91.31%. The comprehensive comparative experimental results suggested that the proposed method yielded optimal performance on this dataset. 18. A Novel PWM Signal-Generation Strategy for Pockels Cell Drivers.

A Pockels cell driver (PCD) can be viewed as a high-voltage pulse width generator for controlling the bi-refringence effect of electro-optical crystals. The main features of a PCD include a high repetition rate, fast on and off switching, variable pulse duration, and a true square pulse shape. The most commonly used PCD has a narrow pulse width tuning range, typically within a few microseconds. In this paper, we propose a PCD based on a novel pulse width modulation (PWM) signal-generation strategy that can continuously adjust its pulse width with a minimum step size of 10 ns and no restriction on the maximum width. Therefore, it is easily compatible with both "On-type" and "Off-type" applications of the electro-optic crystal quarter-wave voltage. The experimental results show that the rising and falling times of the proposed PCD are approximately 7.3 ns and 7.8 ns, respectively, with a maximum repetition rate of 1 MHz and a maximum voltage of approximately 2.0 kV. The power generation of the photovoltaic plant is related to the cleanliness of the photovoltaic modules. The accumulation of natural dust is the main source of pollution, which is affected by human activities and meteorological factors such as temperature, humidity, wind speed, and rainfall concentration in the current region. On the basis of particle swarm optimization (PSO) and the least‐squares support vector machine (LSSVM), the density of dust on photovoltaic modules was estimated. The authors proposed that the inertia weight decreased as a concave function to improve the efficiency of optimization and let the two positive constants change dynamically to improve convergence performance. The dust accumulation prediction model was established considering natural rainfall and the authors obtained the attenuation rate of the photovoltaic power output. Finally, the experiments in Hangzhou showed that the model can predict the density of accumulated dust quickly, which provides a theory for predicting PV power generation and managing the cleaning frequency. Insect metabolites play vital roles in regulating the physiology, behavior, and numerous adaptations of insects, which has contributed to them becoming the largest class of Animalia. However, systematic metabolomics within the insects is still unclear. The present study performed a widely targeted metabolomics analysis based on the HPLC-MS/MS technology to construct a novel integrated metabolic database presenting comprehensive multimetabolite profiles from nine insect species across three metamorphosis types. A total of 1442 metabolites were identified, including amino acids and their metabolites, organic acids and their derivatives, fatty acids (FAs), glycerophospholipids (GPs), nucleotides and their metabolites, and benzene and its substituted derivatives. Among them, 622 metabolites were used to generate a 0 and 1 matrix based on their presence or absence, and these metabolites were enriched in arachidonic acid metabolism, tyrosine metabolism, phenylalanine metabolism, and insect hormone biosynthesis pathways. Our study revealed that there is a high coincidence between the evolutionary relationships of the species and the hierarchical cluster based on the types of metabolites, while the quantities of the metabolites show a high diversity among species. The metabolome of the nine representative insects provides an important platform for implementing the analysis of insect systemic metabolites and biological events at the metabolic level. The rapid rise of glacioeustatic change is the most extreme paleoenvironment alteration in the aftermath of Snowball Earth. Although geologists conducted a lot of multi-subdiscipline research on this issue previously, there still exists the potential room for further discussions of the process in detail. For decades, the practice proved that the Fischer plot is a simple and robust tool to illustrate the fluctuations of accommodation patterns v.s. cycle sets or strata depth which could be interpreted as relative sea-level changes. This research simulates the Fischer plot to unravel the sea-level change in the aftermath of Marinoan glaciation by measuring the lower Ediacaran Doushantuo Formation in Shennongjia, South China. The result shows that 131 fifth-order cycles and nine third-order cycles help us propose a completed second-order cycle variation of ice melting-forced sea-level change, i.e., (1) early high-frequency and slow to rapid stepwise rising, and (2) followed by a stable decrease in the latter. In addition, the vertical sedimentary facies of the lower Doushantuo Formation display, in ascending order, (1) intertidal carbonate rock, (2) subtidal clastics with turbidite, and (3) intertidal lagoon fine clastics, indicating the process of the relative waxing and waning of sea-level. Such interpretation of the Fischer plot and the sedimentary facies' vertical evolution is beneficial for studying the high-frequency sea-level change and paleogeographic reconstruction of post-Marinoan deglaciation. 22. Association of the Systemic Immune-Inflammation Index with Outcomes in Acute Coronary Syndrome Patients with Chronic Kidney Disease.

Background: The systemic immune-inflammation index (SII; neutrophil × platelet/lymphocyte) is a novel marker for immune and inflammatory status and is associated with adverse prognosis in cardiovascular disease. Methods: In total, 744 patients diagnosed with acute coronary syndrome (ACS) and chronic kidney disease (CKD) were included in our study, received standard therapies, and were followed up. Patients were divided into high and low SII groups according to the baseline SII. The primary endpoint was major cardiovascular events (MACEs), defined as cardiovascular death, nonfatal myocardial infarction, and nonfatal stroke. Results: During a median follow-up of 2.5 years, a total of 185 (24.9%) MACEs were recorded. Analysis of the ROC curve revealed that the best cutoff value of SII was 1159.84× 10
9 /L for predicting MACEs. The Kaplan–Meier analysis showed that those patients in the low SII group had higher survival rates than those in the high SII group (p < 0.001). Compared to those in the low SII group, patients in the high SII group were at significantly higher risk of MACEs (134 (38.8%) vs 51 (12.8%), p < 0.001). Univariate and multivariable Cox regression analyses revealed that a high SII level was independently associated with MACEs in ACS patients with CKD (adjusted hazard ratio [HR]: 1.865, 95% confidence interval [CI]: 1.197– 2.907, p = 0.006). Conclusion: The present study showed that an elevated SII is associated with adverse cardiovascular outcomes in ACS with CKD patients, suggesting that SII may be a valuable predictor of poor prognosis in ACS with CKD patients. The accurate and effective early warning of sow behaviors in farrowing helps breeders determine whether it is necessary to intervene with the farrowing process in a timely manner and is thus essential for increasing the survival rate of piglets and the profits of pig farms. For large pig farms, human resources and costs are important considerations in farrowing supervision. The existing method, which uses cloud computing-based deep learning to supervise sow farrowing, has a high equipment cost and requires uploading all data to a cloud data center, requiring a large network bandwidth. Thus, this paper proposes an approach for the early warning and supervision of farrowing behaviors based on the embedded artificial-intelligence computing platform (NVIDIA Jetson Nano). This lightweight deep learning method allows the rapid processing of sow farrowing video data at edge nodes, reducing the bandwidth requirement and ensuring data security in the network transmission. Experiments indicated that after the model was migrated to the Jetson Nano, its precision of sow postures and newborn piglets detection was 93.5%, with a recall rate of 92.2%, and the detection speed was increased by a factor larger than 8. Background. Senescence is thought to be an imperative effect on the development of cancer. However, few studies pay an attention to the senescence-associated genes in pancreatic cancer (PC). The prognostic value of senescence-related genes (SRGs) and their involvement in tumor microenvironment (TME) in the PC remain obscure. The aim of this research was to investigate the prognostic role of senescence-associated genes and their affection in TME in PC. Methods. The transcriptome and clinical information of PC patients were obtained from The Cancer Genome Atlas (TCGA) and the Gene Expression Omnibus (GEO) databases. Two SRG-mediated molecular clusters were comprehensively identified. In total, data from the 285 PC patients were randomly used to develop a senescence-associated gene signature in the training set and verified in the validation set. Immune microenvironment analysis pertained to senescence-related genes was performed. Results. A SRG_score including five senescence-associated genes was established to separate PC patients into two risk groups. High-risk patients had worse overall survival than low-risk patients. The result of the multivariate Cox regression analysis identified the risk score and stage as independent prognostic factors for PC patients. Receiver operating characteristic curve (ROC) analysis confirmed the credible predictive ability of the nomogram. The area under time-dependent ROC curve (AUC) reached 0.746 at 1 year, 0.781 at 3 years, and 0.868 at 5 years in the training set and 0.653 at 1 year, 0.755 at 3 years, and 0.785 at 5 years in the validation set. Moreover, the SRG_score was associated with TME, tumor mutation burden (TMB), and chemotherapeutic drug sensitivity. Conclusions. This study found that the novel SRG_score could be an independent prognostic target for PC patients. Neuroradiological methods play important roles in neurology, especially in cerebrovascular diseases. Fluid-attenuated inversion recovery (FLAIR) vascular hyperintensity (FVH) is frequently encountered in patients with acute ischemic stroke and significant intracranial arterial stenosis or occlusion. The mechanisms underlying this phenomenon and the clinical implications of FVH have been a matter of debate. FVH is associated with large-vessel occlusion or severe stenosis, as well as impaired hemodynamics. Possible explanations suggested for its appearance include stationary blood and slow antegrade or retrograde filling of the leptomeningeal collateral circulation. However, the prognostic value of the presence of FVH has been controversial. FVH can also be observed in patients with transient ischemic attack (TIA), which may have different pathomechanisms. Its presence can help clinicians to identify patients who have a higher risk of stroke after TIA. In this review article, we aim to describe the mechanism and influencing factors of FVH, as well as its clinical significance in patients with cerebrovascular disease. [ABSTRACT FROM AUTHOR]

- Abstract
The occurrence of ovarian cancer (OC) is a major factor in women's mortality rates. Despite progress in medical treatments, like new drugs targeting homologous recombination deficiency, survival rates for OC patients are still not ideal. The tumor microenvironment (TME) includes cancer cells, fibroblasts linked to cancer (CAFs), immune-inflammatory cells, and the substances these cells secrete, along with non-cellular components in the extracellular matrix (ECM). First, the TME mainly plays a role in inhibiting tumor growth and protecting normal cell survival. As tumors progress, the TME gradually becomes a place to promote tumor cell progression. Immune cells in the TME have attracted much attention as targets for immunotherapy. Immune checkpoint inhibitor (ICI) therapy has the potential to regulate the TME, suppressing factors that facilitate tumor advancement, reactivating immune cells, managing tumor growth, and extending the survival of patients with advanced cancer. This review presents an outline of current studies on the distinct cellular elements within the OC TME, detailing their main functions and possible signaling pathways. Background: With the establishment of the cardiac-gut axis concept, increasing evidence has suggested the involvement and important regulatory role of the gut microbiota (GM) and short chain fatty acid (SCFA) in cardiovascular diseases. However, the relationship between GM and atrial fibrillation (AF) is still poorly understood., Objectives: The aim of this study was to investigate whether there were differences in GM and SCFA between AF patients and healthy controls., Methods: In this study, we enrolled 30 hospitalized patients with AF and 30 matched patients with sinus rhythm (SR). GM species in fecal samples were evaluated through amplicon sequencing targeting the 16Sribosomal RNA gene. The feces SCFAs were describe step by step the quantitative analysis using gas chromatography-mass spectrometry (GC-MS). GM species richness, diversity, differential abundance of individual taxa between AF and SR were analyzed., Results: AF patients showed decreased species richness and α-diversity compared to SR patients, but there was no statistical difference. The phylogenetic diversity was significant decreased in AF group. The β-diversity indexes revealed significant differences in GM community structure between the AF group and the SR group. After investigated the individual taxa, AF group showed altered relative abundance in several taxa compared to the SR group. linear discriminant analysis (LDA) effect size (LEfSe) analysis revealed, a significant decrease in Bifidobacterium and a greater abundance of Lactobacillus, Fusobacterium , Haemophilus in AF group compared with the SR group. The abundance of haemophilus was negative correlated with isovaleric acid and isobutyric acid., Conclusions: In AF patients, the GM phylogenetic diversity and β-diversity decreased, the relative abundance altered in several taxa and the bacterial community structure changed as well as the SCFA level. - Abstract
Importance: Candida auris is a recently emerged pathogenic fungus of grave concern globally due to its resistance to conventional antifungals. This study takes a whole-genome approach to explore how C. auris overcomes growth inhibition imposed by the common antifungal drug fluconazole. We focused on gene disruptions caused by a "jumping genetic element" called transposon, leading to fluconazole resistance. We identified mutations in two genes, each encoding a component of the Ubr2/Mub1 ubiquitin-ligase complex, which marks the transcription regulator Rpn4 for degradation. When either protein is absent, stable Rpn4 accumulates in the cell. We found that Rpn4 activates the expression of itself as well as the main drug efflux pump gene CDR1 by binding to a PACE element in the promoter. Furthermore, we identified an amino acid change in Ubr2 in many resistant clinical isolates, contributing to Rpn4 stabilization and increased fluconazole resistance.

31. Long Non-coding RNA LINC01969 Promotes Ovarian Cancer by Regulating the miR-144-5p/LARP1 Axis as a Competing Endogenous RNA.

- Abstract
Accumulating evidence has shown that long non-coding RNAs (lncRNAs) can be used as biological markers and treatment targets in cancer and play various roles in cancer-related biological processes. However, the lncRNA expression profiles and their roles and action mechanisms in ovarian cancer (OC) are largely unknown. Here, we assessed the lncRNA expression profiles in OC tissues from The Cancer Genome Atlas (TCGA) database, and one upregulated lncRNA, LINC01969 , was selected for further study. LINC01969 expression levels in 41 patients were verified using quantitative real-time polymerase chain reaction (qRT-PCR). The in vitro effects of LINC01969 on OC cell migration, invasion, and proliferation were determined by the CCK-8, ethynyl-2-deoxyuridine (EdU), wound healing, and Transwell assays. Epithelial-mesenchymal transition (EMT) was evaluated using qRT-PCR and Western blotting. The molecular mechanisms of LINC01969 in OC were assessed through bioinformatics analysis, RNA-binding protein immunoprecipitation (RIP), dual luciferase reporter gene assays, and a rescue experiment. Finally, in vivo experiments were conducted to evaluate the functions of LINC01969 . The results of the current study showed that LINC01969 was dramatically upregulated in OC, and patients with lower LINC01969 expression levels tended to have better overall survival. Further experiments demonstrated that LINC01969 promoted the migration, invasion, and proliferation of OC cells in vitro and sped up tumor growth in vivo . Additionally, LINC01969 , which primarily exists in the cytoplasm, boosted LARP1 expression by sponging miR-144-5p and promoted the malignant phenotypes of OC cells. 32. CircASH2L Promotes Ovarian Cancer Tumorigenesis, Angiogenesis, and Lymphangiogenesis by Regulating the miR-665/VEGFA Axis as a Competing Endogenous RNA.

- Abstract
Ovarian cancer is the leading cause of gynecologic cancer-related deaths. Emerging research has revealed a close relationship between circular RNAs (circRNAs) and ovarian cancer development, metastasis, and prognosis. The objective of our research was to further explore the relationship between circASH2L and ovarian cancer. Quantitative real-time polymerase chain reaction was used to detect the differential expression of circRNAs between normal ovaries and ovarian cancer tissues. The impact of circASH2L on the proliferation, invasion, and tumorigenicity of ovarian cancer cells was evaluated using gain- and loss-of-function experiments. The molecular mechanisms of circASH2L function were investigated using bioinformatics analysis, RNA fluorescence in situ hybridization, western blots, and dual-luciferase reporter assays. The results showed that circASH2L was remarkably upregulated in ovarian cancer. The invasion and growth of ovarian cancer cells were suppressed by circASH2L knockdown in vitro , and downregulation of circASH2L restrained both angiogenesis and lymphangiogenesis of tumor xenografts in vivo . Furthermore, circASH2L was mostly distributed in the cytoplasm, where it competes with vascular endothelial growth factor A (VEGFA) for binding to miR-665. These findings indicate that circASH2L has an oncogenic function in ovarian cancer. 34. Electroluminescence from light-emitting devices based on erbium-doped ZnO/n-Si heterostructures: Enhancement effect of fluorine co-doping.

We report on erbium (Er) related electroluminescence (EL) in the visible and near infrared (NIR) regions from the light-emitting device (LED) based on the Er-doped ZnO (ZnO:Er)/n-Si isotype heterostructure formed by sputtering ZnO:Er film on n-Si/n
+ -Si epitaxial wafer. Herein, the ZnO:Er film exhibits n-type in electrical conduction. The aforementioned LED is electroluminescent only under sufficiently high forward bias with the negative voltage connecting to n+ -Si substrate. Such forward bias enables the electrons from n-Si to enter into the ultra-thin SiOx (x ≤ 2) layer inherently existing between the ZnO:Er film and n-Si via Poole-Frenkel conduction mechanism and, subsequently, to drift into the ZnO:Er film thus becoming hot electrons, which impact-excite the Er3+ ions to emit characteristic visible and NIR light. Furthermore, the Er-related EL from the aforementioned LED can be significantly enhanced through adopting the strategy of co-doping F- ions into the ZnO host, which brings about twofold primary effects. Firstly, due to the atomic size compensation between F- and Er3+ ions, the ZnO crystal grains become larger to accommodate much more optically active Er3+ ions. Secondly, the partial substitution of F- ions for O2- ions around the Er3+ ion reduces the symmetry of pseudo-octahedral crystal field of Er3+ ion, thus increasing the probabilities of intra-4f transitions of Er3+ ions. We believe that this work sheds light on developing efficient silicon-based LEDs using the Er-doped oxide semiconductors.- Published

35. Physiological electric field works via the VEGF receptor to stimulate neovessel formation of vascular endothelial cells in a 3D environment.

- Abstract
Electrical stimulation induces significant neovessel formation in vivo We have shown that electrical stimulation of endothelial cells functions as an important contributor to angiogenesis in monolayer culture. Because angiogenesis occurs in a three-dimensional (3D) environment, in this study we investigated the effects of a direct current (DC) electrical field (EF) on endothelial neovessel formation in 3D culture. There was a significant increase in tube formation when endothelial cells were stimulated with EF for 4 h. The lengths of the tube-like structures were augmented further by the continued EF exposure. The lengths of the tubes also increased dose-dependently in the EF-treated cultures in the field strengths of 50 mV/mm∼200 mV/mm for 6 h. Electrical fields of small physiological magnitude enhanced VEGF expression by endothelial cells in 3D culture. EF treatment also resulted in activation of VEGFR2, Akt, extracellular regulated kinase 1,2 (Erk1/2), as well as the c-Jun NH2-terminal kinase (JNK). The tyrosine kinase inhibitor SU1498 that blocks VEGFR2 activity exhibited a potent inhibition of tube growth, and the Akt inhibitor MK-2206 2HCl, the Erk1/2 inhibitor U0126 and the JNK inhibitor SB203580 significantly reduced EF-stimulated tubulogenesis. These results suggest the importance of the VEGFR2 signaling pathway during EF-induced angiogenesis.
