Lyman-alpha (Ly α) emission from galaxies is currently our most promising probe for constraining when and how reionization began, and thus when the first galaxies formed. At z > 7, the majority of galaxies detected with Ly α are in candidate overdensities. Here, we quantify the probability of these galaxies residing in large ionized bubbles. We create (1.6 Gpc)3 intergalactic medium (IGM) simulations: sufficient volume to robustly measure bubble size distributions around UV-bright galaxies and rare overdensities. We find |${M_{\small UV}}\lesssim -16$| galaxies and overdensities are ≳10–1000 × more likely to trace ionized bubbles compared to randomly selected positions. The brightest galaxies and strongest overdensities have bubble size distributions with highest characteristic size and least scatter. We compare two models: gradual reionization driven by numerous UV-faint galaxies versus rapid reionization by rarer brighter galaxies, producing larger bubbles at fixed neutral fraction. We demonstrate that recently observed z ∼ 7 overdensities are highly likely to trace large ionized bubbles, corroborated by their high Ly α detection rates. However, Ly α detections at z ≈ 8.7 in EGS and z  = 10.6 in GN-z11 are unlikely to trace large bubbles in our fiducial model – 11 and 7 per cent probability of >1 proper Mpc bubbles, respectively. Ly α detections at such high redshifts could be explained by: a less neutral IGM than previously expected; larger ionized regions at fixed neutral fraction; or if intrinsic Ly α flux is unusually strong in these galaxies. Disturbances in the circadian rhythm have been reported in patients following traumatic brain injury (TBI). However, the rhythmic expression of circadian genes in peripheral blood leukocytes (PBL) following TBI has not yet been studied. The messenger ribonucleic acid (mRNA) expression of period 1 (Per1), Per2, Per3, cryptochrome 1 (Cry1), Cry2, brain and muscle aryl hydrocarbon receptor nuclear translocator-like 1 (Bmal1), and circadian locomotor output cycles kaput (Clock) was quantified in PBLs from sham-operated rats and rats with acute subdural hematoma (ASDH) over a 48-h period. The rectal temperature of the animals was measured every 4 h over 2 days. The mesor, rhythm, amplitude, and acrophase were estimated using cosinor analysis. Cosinor analysis revealed that Per2, Cry1, and Bmal1 mRNAs were rhythmically expressed in the PBLs of sham-operated rats. In contrast, fluctuations in rhythmic expression were not observed following ASDH. The rectal temperature of sham-operated rats also exhibited rhythmicity. ASDH rats had a disrupted rectal temperature rhythm, a diminished amplitude, and an acrophase shift. TBI with ASDH results in dysregulated expression of some circadian genes and changes in body temperature rhythm. Further research is required to understand the pathophysiology of altered circadian networks following TBI. Rare and vulnerable endemic plants represent different evolutionary units that occur at different times, and protecting these species is a key issue in biological protection. Understanding the impact of the history of endangered plant populations on their genetic diversity helps to reveal evolutionary history and is crucial for guiding conservation efforts. Saussurea involucrata, a perennial alpine species mainly distributed in the Tianshan Mountains, is famous for its medicinal value but has become endangered due to over-exploitation. In the present study, we employed both nuclear and chloroplast DNA sequences to investigate the genetic distribution pattern and evolutionary history of S. involucrata. A total of 270 individuals covering nine S. involucrata populations were sampled for the amplification and sequencing of nrDNA Internal Transcribed Spacer (ITS) and chloroplast trnL-trnF, matK and ndhF-rpl32 sequences. Via calculation, we identified 7 nuclear and 12 plastid haplotypes. Among the nine populations, GL and BA were characterized by high haplotype diversity, whereas BG revealed the lowest haplotype diversity. Molecular dating estimations suggest that divergence among S. involucrata populations occurred around 0.75 Ma, coinciding with the uplift of Tianshan Mountains. Our results reveal that both isolation-by-distance (IBD) and isolation-by-resistance (IBR) have promoted genetic differentiation among populations of S. involucrata. The results from the ecological niche modeling analyses show a more suitable habitat for S. involucrata in the past than at present, indicating a historical distribution contraction of the species. In arid and semi-arid regions, rainfall takes on a critical significance to both agricultural and engineering construction activities, and the transport process and driving mechanism of soil water under rainfall conditions are in need of further investigation. To clarify the variations in soil moisture, temperature, and liquid and vapor flux under various rainfall scenarios, the Mu Us Sandy Land was selected as the study region, and a water–vapor–heat transport model was established using the Hydrus-1D software with in situ observed soil and meteorological data. The simulated results were in good agreement with the measured data during both the calibration and validation periods, suggesting that the model was accurate and applicable to the study region. The variations in the selected dry and rainy periods proved the significant effect of rainfall events on soil matric potential, temperature, and driving forces. When rainfall occurred, the hydraulic conductivity for liquid water rose by three to five orders of magnitude, driving the liquid water flow downward. In contrast, the vapor flux played a vital role in soil water movement, accounting for about 15% of the total water flux in the shallow layer when the soil was dry, while it became non-significant during rainy periods due to the reduction in hydraulic conductivity for vapor and the temperature gradient. These results clarified the mechanisms of soil liquid water and vapor movement in arid areas, which could provide scientific support for future studies on vegetation restoration and ecosystem sustainability in ecologically fragile areas. Pupillary light reflex (PLR) assessment is a crucial examination for evaluating brainstem function, particularly in patients with acute brain injury and neurosurgical conditions. The PLR is controlled by neural pathways modulated by both the sympathetic and parasympathetic nervous systems. Altered PLR is a strong predictor of adverse outcomes after traumatic and ischemic brain injuries. However, the assessment of PLR needs to take many factors into account since it can be modulated by various medications, alcohol consumption, and neurodegenerative diseases. The development of devices capable of measuring pupil size and assessing PLR quantitatively has revolutionized the non-invasive neurological examination. Automated pupillometry, which is more accurate and precise, is widely used in diverse clinical situations. This review presents our current understanding of the anatomical and physiological basis of the PLR and the application of automated pupillometry in managing neurocritical patients. Chemoradiation-induced hearing loss (CRIHL) is one of the most devasting side effects for nasopharyngeal carcinoma (NPC) patients, which seriously affects survivors' long-term quality of life. However, few studies have comprehensively characterized the risk factors for CRIHL. In this study, we found that age at diagnosis, tumor stage, and concurrent cisplatin dose were positively associated with chemoradiation-induced hearing loss. We performed a genome-wide association study (GWAS) in 777 NPC patients and identified rs1050851 (within the exon 2 of NFKBIA), a variant with a high deleteriousness score, to be significantly associated with hearing loss risk (HR = 5.46, 95% CI 2.93–10.18, P = 9.51 × 10–08). The risk genotype of rs1050851 was associated with higher NFKBIA expression, which was correlated with lower cellular tolerance to cisplatin. According to permutation-based enrichment analysis, the variants mapping to 149 hereditary deafness genes were significantly enriched among GWAS top signals, which indicated the genetic similarity between hereditary deafness and CRIHL. Pathway analysis suggested that synaptic signaling was involved in the development of CRIHL. Additionally, the risk score integrating genetic and clinical factors can predict the risk of hearing loss with a relatively good performance in the test set. Collectively, this study shed new light on the etiology of chemoradiation-induced hearing loss, which facilitates high-risk individuals' identification for personalized prevention and treatment. Background: Patients suffered from severe traumatic brain injury (TBI) have twice the risk of developing into neurodegenerative diseases later in their life. Thus, early intervention is needed not only to treat TBI but also to reduce neurodegenerative diseases in the future. Physiological functions of neurons highly depend on mitochondria. Thus, when mitochondrial integrity is compromised by injury, neurons would initiate a cascade of events to maintain homeostasis of mitochondria. However, what protein senses mitochondrial dysfunction and how mitochondrial homeostasis is maintained during regeneration remains unclear. Results: We found that TBI-increased transcription of a mitochondrial protein, phosphoglycerate mutase 5 (PGAM5), during acute phase was via topological remodeling of a novel enhancer-promoter interaction. This up-regulated PGAM5 correlated with mitophagy, whereas presenilins-associated rhomboid-like protein (PARL)-dependent PGAM5 cleavage at a later stage of TBI enhanced mitochondrial transcription factor A (TFAM) expression and mitochondrial mass. To test whether PGAM5 cleavage and TFAM expression were sufficient for functional recovery, mitochondrial oxidative phosphorylation uncoupler carbonyl cyanide 4-(trifluoromethoxy) phenylhydrazone (FCCP) was used to uncouple electron transport chain and reduce mitochondrial function. As a result, FCCP triggered PGAM5 cleavage, TFAM expression and recovery of motor function deficits of CCI mice. Conclusions: Findings from this study implicate that PGAM5 may act as a mitochondrial sensor for brain injury to activate its own transcription at acute phase, serving to remove damaged mitochondria through mitophagy. Subsequently, PGAM5 is cleaved by PARL, and TFAM expression is increased for mitochondrial biogenesis at a later stage after TBI. Hyperspectral image (HSI) provides rich spectral–spatial information and the light detection and ranging (LiDAR) data reflect the elevation information, which can be jointly exploited for better land-cover classification. However, due to different imaging mechanisms, HSI and LiDAR data always present significant image difference, current pixel-wise feature fusion classification methods relying on concatenation or weighted fusion are not effective. To achieve accurate classification result, it is important to extract and fuse similar high-order semantic information and complementary discriminative information contained in multimodal data. In this paper, we propose a novel coupled adversarial learning based classification (CALC) method for fusion classification of HSI and LiDAR data. In specific, a coupled adversarial feature learning (CAFL) sub-network is first trained, to effectively learn the high-order semantic features from HSI and LiDAR data in an unsupervised manner. On one hand, the proposed CAFL sub-network establishes an adversarial game between dual generators and discriminators, so that the learnt features can preserve detail information in HSI and LiDAR data, respectively. On the other hand, by designing weight-sharing and linear fusion structure in the dual generators, we can simultaneously extract similar high-order semantic information and modal-specific complementary information. Meanwhile, a supervised multi-level feature fusion classification (MFFC) sub-network is trained, to further improve the classification performance via adaptive probability fusion strategy. In brief, the low-level, mid-level and high-level features learnt by the CAFL sub-network lead to multiple class estimation probabilities, which are then adaptively combined to generate a final accurate classification result. Both the CAFL and MFFC sub-networks are collaboratively trained by optimizing a designed joint loss function, which consists of unsupervised adversarial loss and supervised classification loss. Overall, by optimizing the joint loss function, the proposed CALC network is pushed to learn highly discriminative fusion features from multimodal data, leading to higher classification accuracies. Extensive experiments on three well-known HSI and LiDAR data sets demonstrate the superior classification performance by the proposed CALC method than several state-of-the-art methods. Simple Summary: Mood disorders including depression and bipolar were associated with an increased hazard ratio of cervical cancer. The cumulative incidence of cervical cancer was significantly higher in participants with mood disorders than in the non-mood-disorder cohort. Thus, mood disorders could potentially increase the risk of developing subsequent cervical cancer. Objective: Psychological distress is considered a factor for cancer development. However, the impact of mood disorders (depression and bipolar) on the development of cervical cancer remains uncertain. We conducted a nationwide population-based retrospective cohort study to investigate the association between mood disorders and the subsequent risk of developing cervical cancer. Methods: A total of 138,130 participants' profiles between 2000 and 2012 were extracted from the National Health Insurance Research Database and subdivided into a mood-disorder cohort (27,626 participants) and a non-mood-disorder cohort (110,504 participants). Cohorts were propensity-matched for a 1:4 ratio according to age and index year. The Cox proportional hazards regression model was utilized for assessing cervical cancer risk between cohorts. Results: Kaplan–Meier analysis revealed that the mood-disorder cohort had a higher cumulative incidence of cervical cancer. The mood-disorder cohort was also associated with an increased risk of cervical cancer after adjustments for potential confounders. Subgroup analysis revealed a negative impact of mood disorders on cervical cancer, especially in the 30–50 years and white-collar groups. Conclusions: Our findings demonstrated that mood disorders were associated with an increased risk of cervical cancer development, which provide helpful information for clinical strategies to reduce the incidence of cervical cancer in this vulnerable population. Revealing the cosmic hydrogen reionization history is one of the main goals of the modern cosmology. z > 5 quasars (QSOs) have been used as back-lights to investigate the evolution of the intervening intergalactic medium (IGM) during the cosmic reionization since their first discovery. However, due to the small population of luminous QSOs (∼130 QSOs known to date), a tight constraint on the reionization history has not yet been placed. In this work, we aim to tighten the constraint using the 93 QSOs (5.5 < z < 7.1) recently discovered in the Subaru High-z Exploration of Low-Luminosity Quasars (SHELLQS) project. This is the largest QSO sample used to constrain the epoch of reionization. We measure the mean IGM Ly α transmission and the QSO near-zone size using the UV spectra of these QSOs. The mean IGM Ly α transmission rises above zero at z ≲ 6, indicating the end of the reionization. The near-zone sizes of the SHELLQs QSOs are consistent with sizes spanned by QSOs of lifetime t q ∼ 1–100 Myr in simulations. Due to the scatter created by the low signal-to-noise spectra and large Ly α redshift uncertainty, we cannot conclude whether the redshift evolution of the near-zone size is affected by the reionization effect. Backgroud: Postoperative nausea and vomiting (PONV) is one of the most common complications after total thyroidectomy under general anesthesia. Total intravenous anesthesia (TIVA) has been documented to prevent PONV in patients undergoing total thyroidectomy. Penehyclidine, an anticholinergic agent with an elimination half-life of over 10 h, is widely used as premedication to reduce glandular secretion. This study aimed to explore the preventative effects of penehyclidine with propofol-remifentanil-TIVA to single-TIVA on PONV in patients undergoing total thyroidectomy. Methods: A total of 100 patients scheduled for total thyroidectomy were randomly assigned to either the penehyclidine group (n = 50) or TIVA group (n = 50). Propofol and remifentanil were was used for TIVA in all patients. No patients who received premedication. Patients were administrated with either 5 ml of normal saline or 0.5 mg of penehyclidine soon after anesthesia induction. The incidence of nausea and vomiting, the severity of nausea, the requirement of rescue antiemetics, and adverse effects were investigated during the first 24 h in two time periods (0–2 h and 2–24 h). Results: The overall PONV incidence during the 24 h after surgery was significantly lower in the penehyclidine group compared with the TIVA group (12% vs 36%, P < 0.005). Besides, the incidence of nausea and the incidence of vomiting were significantly lower in the penehyclidine group compared with the TIVA group at 2–24 h after surgery. However, there was no significant difference between the two groups at 0–2 h after surgery. Conclusions: Administration of penehyclidine under TIVA with propofol-remifentanil is more effective for prevention of PONV than TIVA alone, especially 2–24 h after total thyroidectomy. By adopting atomic layer deposition (ALD) sidewall passivation, the electroluminescence (EL) and communication performances of mini-light emitting diodes (mini-LEDs) of different sizes were improved. In particular, the most significant improvement in the electroluminescence properties of the external quantum efficiency (EQE) (a 7.2% increase), leakage current, and the communication properties of the modulation bandwidth (a 20% increase) transmission rate and bit error rate (BER) was found in the smallest mini-LEDs (80 $\mu \text{m}\,\,\times $ 120 $\mu \text{m}$). According to the results of time-resolved photoluminescence (TRPL) measurements, the carrier lifetime of the samples can be affected by both the size and ALD sidewall passivation. A novel BiOBr/Ni-MOF composite was fabricated by in-situ growth of BiOBr nanosheets on Ni-MOF through a convenient precipitation process. The optimal photocatalytic performance was achieved with a Ni-MOF mass content of 45 wt% in the BiOBr/Ni-MOF composite. This composite exhibited remarkable efficiency, degrading 98.33 % of Rhodamine B in 90 min, surpassing the degradation rate constants of pristine BiOBr and Ni-MOF by 4.2 and 160 times, respectively. The superior performance was primarily ascribed to the efficient separation of photogenerated carriers facilitated by the formation of heterojunctions. Additionally, the increased specific surface area and pore structure provided numerous adsorption and active sites. Radical scavenger experiments and electron spin resonance experiments further revealed that O 2 − and h+ played crucial roles in the degradation process. Furthermore, the structural properties of the prepared photocatalysts and the effects of different photocatalytic conditions on the degradation of RhB were investigated. —Machine learning technologies have gained significant popularity in rechargeable battery research in recent years, and have been extensively adopted to construct data-driven solutions to tackle multiple challenges for energy storage in embedded computing systems. An important application in this area is the machine learning-based battery lifetime prediction, which formulates regression models to estimate the remaining lifetimes of batteries given the measurement data collected from the testing process. Due to the non-idealities in practical operations, these measurements are usually impacted by various types of interference, thereby involving noise on both input variables and regression labels. Therefore, existing works that focus solely on minimizing the regression error on the labels cannot adequately adapt to the practical scenarios with noisy variables. To address this issue, this study adopts total least squares (TLS) to construct a regression model that achieves superior regression accuracy by simultaneously optimizing the estimation of both variables and labels. Furthermore, due to the expensive cost for collecting battery cycling data, the number of labeled data samples used for predictive modeling is often limited. It, in turn, can easily lead to overfitting, especially for TLS, which has a relatively larger set of problem unknowns to solve. To tackle this difficulty, the TLS method is investigated conjoined with stepwise feature selection in this work. Stroke is an acute cerebrovascular disease caused by the rapid rupture or blockage of intracranial blood vessels for a variety of reasons, preventing blood from flowing into the brain and causing damage to brain tissue. The global burden of stroke disease is quickly increasing, and ischemic stroke (IS) accounts for 60 percent to 70 percent of all strokes, owing to the prevalence of people's bad lifestyles and the intensity of global ageing. Although most IS patients have received effective treatment, many patients still have certain dysfunction or death after treatment, and the recurrence rate is about 18%, which brings a heavy economic burden to society and families. Therefore, it is urgent to build a postoperative prediction model for IS, so as to take targeted clinical intervention measures, which has extremely important practical significance for improving the prognosis of IS. The following work has been done in this paper: (1) the theoretical background for the BP prediction model and logistic regression prediction model suggested in this work is offered, as well as the research progress and related technologies of IS recurrence prediction by domestic and foreign academics. (2) The basic principles of BPNN and logistic regression are introduced, and the logistic multifactor predictor is constructed. (3) The experimental results show that the consistency rate, sensitivity, and specificity of the prediction results of BPNN are higher than those of logistic regression, indicating that for diseases such as IS, which have many pathogenic factors and complex relationships between factors, the fitting effect of BPNN model is better than that of the logistic regression model. With the continuous improvement of medical technology and the aging of the population, the death rate of stroke is gradually decreasing, but the recurrence rate is still high, and the number of recurrences is increasing, resulting in disability and other symptoms, which brings great burden and distress to patients and their families. As the number of strokes increases, neurological impairment becomes more and more severe, affecting patients' ability to live, socialize, and work, and seriously reducing their quality of life. Clustered care is a combination of evidence-based linked interventions and a multidisciplinary team providing the best possible care through evidence-based research and highly operational practice, and it can improve outcomes for ischemic stroke patients more than implementation alone. This paper presents a Cox proportional risk regression-based model, using it to build the most used semi-parametric model for multifactorial survival analysis, due to its advantages of both parametric and nonparametric models, and to analyze the factors influencing survival time in study subjects with incomplete data. Microwave radiation is a relatively new energy source that is being considered for several applications in mineral processing and extractive metallurgy. In the present research, various gibbsite-type bauxite ores were subjected to microwave radiation. The main objective was to assess the effect of microwave dewatering on the compressive strength indices of the ores and to compare the results obtained to those for conventional heating. Firstly, the fundamental interactions of the microwaves with the ores were evaluated by determining both the real and the imaginary permittivities as a function of temperature, and these were related to the water content. Secondly, the microwave heating behavior was modeled using a 24 factorial statistical analysis. Thirdly, the effect of dewatering by microwave heating on the compressive strength indices of roughly spherical bauxite ore pisoids was studied, and these results were compared to those obtained using conventional heating. Fourthly, the effect of particle size on the compressive strengths of irregular-shaped single particles of bauxite ore was investigated using both heating techniques. Finally, the energy requirements for dewatering of the ores, and hence reducing their compressive strengths, were compared for both processes. On the laboratory scale, the results showed that in comparison to conventional dewatering, microwave dewatering resulted in lower strength indices at both lower moisture removals and energy inputs. In the rapid promotion of China's electricity spot market, a large number of electricity retailers and large consumers participate in power trading, of which medium- and long-term power trading accounts for a large proportion. In the electricity spot market, the previous medium- and long-term transactions need to be closely combined with the current spot market transaction settlement rules. This paper analyzes the trading strategy of large retailers in the power market. In order to effectively reduce the total electricity cost, it is necessary to optimize the medium- and long-term transactions based on three aspects: electricity quantity and benchmark price decisions of medium- and long-term contracts, the daily electricity decomposition method in the day-ahead (DA) market, and the daily load curve decomposition strategy. According to load history characteristics that are extracted by the X12 method, daily electricity is decomposed from the medium- and long-term electricity quantity in the DA market. This paper introduces three methods of decomposing the daily load curve and proves that the particle swarm algorithm is the best method for effectively minimizing the cost in the DA market. Through analyzing the total electricity cost change pattern, we prove that the basic component of decision making is the relative relationship between the electricity price of medium- and long-term contracts and the equivalent kWh price of medium- and long-term electricity in the DA market, which is determined by the decomposition daily curve method. If the equivalent kilowatt-hour price obtained by the decomposition method in the DA market is greater than the electricity price of medium- and long-term contracts, the larger the electrical energy of medium- and long-term contracts, the lower the costs. Store-operated channels (SOCs) are highly selective Ca2+ channels that mediate Ca2+ influx in non-excitable and excitable (i.e., skeletal and cardiac muscle) cells. These channels are triggered by Ca2+ depletion of the endoplasmic reticulum and sarcoplasmic reticulum, independently of inositol 1,4,5-trisphosphate (InsP3), which is involved in cell growth, differentiation, and gene transcription. Exploring effective spectral–spatial feature extraction methods is one of the most focused problems in current hyperspectral image (HSI) classification research. However, complex spectral–spatial structure characteristics in HSIs, e.g., shape-variable spatial structure and nonlinear spectral structure, are difficult to be effectively extracted and jointly represented. To overcome this issue, a novel superpixel-based Brownian descriptor (SBD) method for HSI classification is proposed in this article. In specific, superpixel segmentation is first used to extract shape-adaptive spatial structure information from dimension-reduced HSI, leading to generate nonoverlapping homogeneous 3-D image blocks. Then, similar pixels within the 3-D image block are jointly represented by a new local spectral–spatial feature based on the Brownian descriptor (BD). This is the first time that the BD is introduced to measure both linear and nonlinear correlations among different spectral bands in HSI. On one hand, the integration of superpixel and BD helps to provide much richer and more valuable information for better discrimination between different categories. On the other hand, the SBD can effectively represent the internal structure characters within each 3-D image block of different spatial shapes by a symmetric positive definite matrix of a united form. Finally, considering that the SBD lies on the Riemannian manifold space, a log-Euclidean kernel sparse representation (LKSR) classifier is introduced to obtain the classification results. Background. Mutations of the Ectodysplasin-A (EDA) gene are generally associated with syndrome hypohidrotic ectodermal dysplasia or nonsyndromic tooth agenesis. The influence of EDA mutations on dentinogenesis and odontoblast differentiation has not been reported. The aim of this study was to identify genetic clues for the causes of familial nonsyndromic oligodontia and explore the underlying mechanisms involved, while focusing on the role of human dental pulp stem cells (hDPSCs). Materials and Methods. Candidate gene sequences were obtained by PCR amplification and Sanger sequencing. Functional analysis was conducted, and the pathogenesis associated with EDA mutations in hDPSCs was investigated to explore the impact of the identified mutation on the phenotype. Capillary electrophoresis (CE) was used to detect X-chromosome inactivation (XCI) in the blood of female carriers. Results. In this study, we identified an EDA mutation in a Chinese family: the missense mutation c.1013C>T (Thr338Met). Transfection of hDPSCs with a mutant EDA lentivirus decreased the expression of EDA and dentin sialophosphoprotein (DSPP) compared with transfection of control EDA lentivirus. Mechanistically, mutant EDA inhibited the activation of the NF-κB pathway. The CE results showed that symptomatic female carriers had a skewed XCI with a preferential inactivation of the X chromosome that carried the normal allele. Conclusions. In summary, we demonstrated that EDA mutations result in nonsyndromic tooth agenesis in heterozygous females and that, mechanistically, EDA regulates odontogenesis through the NF-κB signalling pathway in hDPSCs. Background: Autoimmune polyendocrine syndrome type 1 (APS-1) is a rare monogenic inherited disease caused by mutations of the autoimmune regulator gene (AIRE). The three major components of this syndrome are chronic mucocutaneous candidiasis, hypoparathyroidism and adrenocortical insufficiency. Case presentation: We report a 20-year-old male who was clinically diagnosed with APS-1 at the age of 15. He was admitted to our department this time for suffering from polyuria and polydipsia for 6 months and was finally diagnosed with diabetes insipidus. Whole-exome sequencing (WES) revealed a novel compound heterozygous mutation of the AIRE gene —the c.239 T > G (p.Val80Gly) variant on one allele and the copy number variant (CNV) of 21q22.3(chr21:45,670,150–45,706,528)*1 on the other. Here, we characterized the BA101 mutant under controlled environmental conditions in parallel with the parental strain C. beijerinckii NCIMB 8052. To investigate the correlation between phenotype and genotype, we carried out the genome sequencing of BA101. Through comparative genomic analysis, several mutations in the genes encoding transcriptional regulator, sensor kinase, and phosphatase were identified in the BA101 genome as well as other sibling mutants. Among them, the SNP in the Cbei_3078 gene encoding PAS/PAC sensor hybrid histidine kinase was unique to the BA101 strain. Active learning (AL) attempts to actively select the most representative or useful training samples in an iterative manner. The aim is to simultaneously improve the classification performance and reduce the manual labeling effort. In this article, a novel subpixel-pixel-superpixel-based multiview AL (MAL) (SPS-MAL) method is proposed for hyperspectral image (HSI) classification. Here, the multiple views are generated via extracting the subpixel-level, pixel-level, and superpixel-level information. The multiple views can reflect various characteristics of HSI, i.e., spectral mixture, spectral discrimination, and spectral–spatial structure. Therefore, the joint use of diverse and complementary information in multiple views will contribute to a better identification ability of different classes. In addition, a coarse-to-fine MAL algorithm is introduced to effectively select the most representative samples with the most uncertainty. Specifically, a disagreement analysis on multiple views and joint posterior probability estimation is used to query unlabeled samples. Along with the expansion of training samples, view-specific confidence scores are estimated to adaptively integrate the classification results of multiple views, according to their discrimination performance. In this way, the classification accuracy will be further boosted while the number of necessary training samples can be significantly reduced. Background: Studies assessing the efficacy of pioglitazone solely for primary stroke prevention in Asian patients with type 2 diabetes mellitus (DM) and present multiple cardiovascular (CV) risk factors are rare. Thus, we aimed to assess the effect of pioglitazone on primary stroke prevention in Asian patients with type 2 DM without established CV diseases but with risk factors for CV diseases. Methods: Between 2000 and 2012, we enrolled patients aged ≥ 18 years, who were newly diagnosed with type 2 diabetes and had at least one of the following CV risk factors: hypertension and hyperlipidemia. Patients with a history of stroke and those using insulin or glucagon-like peptide-1 agonist for more than 3 months were excluded. Patients were divided into the pioglitazone and non-pioglitazone groups based on their receipt of pioglitazone during the follow-up period. Propensity-score matching (1:1) was used to balance the distribution of the baseline characteristics and medications. Follow-up was terminated upon ischemic stroke development, withdrawal from the insurance system, or on December 31, 2013, whichever occurred first. The overall incidence of new-onset ischemic stroke in the two groups was subsequently compared. The subgroup analyses of ischemic stroke were conducted using different baseline features. Additionally, the effect of pioglitazone exposure dose on the occurrence of ischemic stroke was evaluated. Chi square test, Student's t-test, competing risk regression models, Kaplan–Meier method, and log-rank test were some of the statistical tests conducted. Results: A total of 13 078 patients were included in the pioglitazone and non-pioglitazone groups. Compared with patients who did not receive pioglitazone, those administered pioglitazone had a lower risk of developing ischemic stroke (adjusted hazard ratio: 0.78; 95% confidence interval: 0.62–0.95). The subgroup analyses defined by different baseline features did not reveal significant alterations in the observed effect of pioglitazone. Moreover, a significant decreasing trend in ischemic stroke risk with an increase in pioglitazone dose (p-value for trend = 0.04) was observed. Conclusion: Pioglitazone use decreased the risk of new-onset ischemic stroke in Asian patients with type 2 DM and CV risk factors. Traditional hyperspectral imaging technique obtains numerous hyperspectral images (HSIs) with hundreds of spectral bands, leading to high cost in data acquisition, transmission, and storage. Compressed sensing (CS) theory provides a new imaging mechanism, which relies on the assumption that signals can be sparsely represented over a dictionary. By the CS imaging technique, original HSIs can be approximately reconstructed from only a few sampled measurements. In this article, a novel context-aware CS (CACS) method for HSIs is proposed by incorporating contextual prior to the dictionary learning and the sparse reconstruction. First, a patch-based online dictionary learning (ODL) algorithm is developed by introducing a joint sparse constraint. On the one hand, the online dictionary learning mechanism enables a more adaptive representation of HSIs with different scenes than using fixed-basis-based dictionaries, e.g., the discrete cosine transform (DCT) and the discrete wavelet transform dictionaries. On the other hand, the introduced joint sparse constraint promotes the learned dictionary to more sparsely and structurally represent spectral pixels. Then, with the well-learned dictionary, a weighted smoothing regularization is introduced to develop a new sparse reconstruction model. Considering the high spectral–spatial similarity of pixels in a neighborhood, the new sparse reconstruction model will encourage a locally smoothing reconstruction result. In this way, the spectral–spatial structures of the HSI can be well preserved, while possible artifacts can be effectively reduced. Our previous studies have revealed that a dominant mutation in vacuolar protein sorting 4B (VPS4B), a member of the AAA ATPase family, causes dentin dysplasia type I. The purpose of the present study was to investigate the roles of VPS4B in human dental pulp stem cells (hDPSCs) and to elucidate the underlying molecular mechanisms. In this study, we found that VPS4B was highly expressed in the dental pulp cells of the mouse molar tooth germ, and the expression of VPS4B increased significantly during the odontoblastic differentiation of hDPSCs. VPS4B downregulation inhibited the proliferation, migration, and odontoblastic differentiation of hDPSCs. Moreover, treatment with lithium chloride, an agonist of the Wnt-β-catenin signalling pathway, partially reversed the VPS4B knockdown-driven suppression of proliferation and of odontoblastic differentiation of hDPSCs. Collectively, our findings indicate that VPS4B, via Wnt-β-catenin signalling, acts as a regulator of the proliferation and differentiation of hDPSCs. Electrolytic Manganese Residue (EMR) is a solid waste containing soluble sulfate, discharged by electrolytic manganese industries. The accumulation of EMR in ponds poses a significant hazard to both safety and the environment. This study utilized innovative geotechnical test techniques to conduct a series of tests, investigating the effect of soluble salts on the geotechnical characteristics of EMR. The results revealed that soluble sulfates had a significant impact on the geotechnical characteristics of the EMR. In particular, the infiltration of water leached away the soluble salts, causing a non-uniform particle size distribution and decreasing the shear strength, stiffness, and liquefaction resistance of the EMR. Nevertheless, an increase in the stacking density of EMR could improve its mechanical characteristics and inhibited the dissolution of soluble salts. aimed to investigate the potential role of fatty acid synthase (FASN) in the development and progression of colorectal cancer (CRC). Materials and methods: FASN levels were analyzed in human CRC tissues and adjacent normal tissues by Western blots and immunohistochemistry. Potential roles of FASN in regulating CRC cell proliferation and migration were examined by genetic manipulation in vitro. The molecular signaling was determined to understand the mechanisms of observed FASN effects. Results: FASN level was upregulated in CRC tissues and high expression of FASN was significantly associated with lymph node metastasis, TNM (Tumor, Node, Metastases) stage and poor prognosis in patients with CRC. Knockdown of FASN attenuated CRC cell proliferation and migration in vitro while FASN overexpression possessed the opposite effects. Background Yes-associated protein (YAP) is upregulated in many cancer types, and its overexpression is involved in tumor cell proliferation, metastasis and resistance to chemotherapy. In the present study, we aimed to investigate the potential role of YAP in the development and progression of gastric cancer. Methods YAP levels were analyzed in human gastric cancer and adjacent normal tissues by Western blotting and immunohistochemistry. Potential roles of YAP in regulating gastric cancer cell proliferation and sensitivity to cisplatin were examined by genetic manipulation in vitro . Alloy-type materials are regarded as prospective anode replacements for lithium-ion batteries (LIBs) owing to their attractive theoretical capacity. However, the drastic volume expansion leads to structural collapse and pulverization, resulting in rapid capacity decay during cycling. Here, a simple and scalable approach to prepare NiM (M: Sb, Sn)/nitrogen-doped hollow carbon tubes (NiM C) via template and substitution reactions is proposed. The nanosized NiM particles are uniformly anchored in the robust hollow N -doped carbon tubes via Ni N C coordination bonds, which not only provides a buffer for volume expansion but also avoids agglomerating of the reactive material and ensures the integrity of the conductive network and structural framework during lithiation/delithiation. As a result, NiSb C and NiSn C exhibit high reversible capacities (1259 and 1342 mAh/g after 100 cycles at 0.1 A/g) and fascinating rate performance (627 and 721 mAh/g at 2 A/g), respectively, when employed as anodes of LIBs. The electrochemical kinetic analysis reveals that the dominant lithium storage behavior of NiM C electrodes varies from capacitive contribution to diffusion contribution during the cycling corresponding to the activation of the electrode exposing more NiM sites. Meanwhile, M (Sb, Sn) is gradually transformed into stable NiM during the de-lithium process, making the NiM C structure more stable and reversible in the electrochemical reaction. Both European and Asian historical records report that Jesuits were at the origin of enamelling technology transfers from France (and Italy) to Asia at the 17th–18th century turn. A mobile Raman set-up equipped with a high magnification (×200) microscope objective with long working distance is used to identify the use of European (arsenic-rich) cobalt in 17th and 18th century porcelains: twenty soft-paste porcelains from Rouen (L. Poterat’ Factory), Saint-Cloud, Paris (Pavie’, Hébert’, Chicanneau’, and Bellevaux’ factories), Lille, Mennecy, and Vincennes factories, of which almost all were produced before 1750, belonging to the Cité de la Céramique Collection (formerly Musée national de Céramique, Sèvres), have been analysed in order to get a more representative view of the type of cobalt ore used. A large proportion of blue decors exhibit signs of lead arsenate, even for productions after ~ 1750, which proves the wilful interest given to arsenic phases in magnifying blue hues. Different lead arsenate signatures are observed, one assigned to the product of reaction of As-rich cobalt ore with lead-based glaze and another characteristic of added arsenic. At least two groups of glaze are identified: a lead-rich one and a mixed lead-alkali glaze. Porcelains made during the second half of the 18th century exhibit the very strong As-O modes characteristic of the voluntary addition of a lead-arsenate forming compound. Key points: Auditory brainstem neurons of all vertebrates fire phase‐locked action potentials (APs) at high rates with remarkable fidelity, a process controlled by specialized anatomical and biophysical properties. This is especially true in the avian nucleus magnocellularis (NM) – the analogue of the mammalian anteroventral cochlear nucleus. In addition to high voltage‐activated potassium (KHVA) channels, we report, using whole cell physiology and modelling, that resurgent sodium current (INaR) of sodium channels (NaV) is equally important and operates synergistically with KHVA channels to enable rapid AP firing in NM. Anatomically, we detected strong NaV1.6 expression near hearing maturation, which was less distinct during hearing development despite functional evidence of INaR, suggesting that multiple NaV channel subtypes may contribute to INaR. We conclude that INaR plays an important role in regulating rapid AP firing for NM neurons, a property that may be evolutionarily conserved for functions related to similar avian and mammalian hearing. Abstract: Auditory brainstem neurons are functionally primed to fire action potentials (APs) at markedly high rates in order to rapidly encode the acoustic information of sound. This specialization is critical for survival and the comprehension of behaviourally relevant communication functions, including sound localization and distinguishing speech from noise. Here, we investigated underlying ion channel mechanisms essential for high‐rate AP firing in neurons of the chicken nucleus magnocellularis (NM) – the avian analogue of bushy cells of the mammalian anteroventral cochlear nucleus. In addition to the established function of high voltage‐activated potassium channels, we found that resurgent sodium current (INaR) plays a role in regulating rapid firing activity of late‐developing (embryonic (E) days 19–21) NM neurons. INaR of late‐developing NM neurons showed similar properties to mammalian neurons in that its unique mechanism of an ‘open channel block state’ facilitated the recovery and increased the availability of sodium (NaV) channels after depolarization. Using a computational model of NM neurons, we demonstrated that removal of INaR reduced high‐rate AP firing. We found weak INaR during a prehearing period (E11–12), which transformed to resemble late‐developing INaR properties around hearing onset (E14–16). Anatomically, we detected strong NaV1.6 expression near maturation, which became increasingly less distinct at hearing onset and prehearing periods, suggesting that multiple NaV channel subtypes may contribute to INaR during development. Clostridium beijerinckii is a predominant solventogenic clostridia with great attraction for renewable liquid biofuel and biochemical production. Metabolic engineering and synthetic biology can be employed to engineer the strain toward desirable phenotypes. However, current limited information such as promoter strength and gene regulation may hinder the efficient engineering of the strain. To investigate genetic information and complex cellular bioprocesses of C. beijerinckii , an in vivo fluorescence reporter system can be employed. In general, green fluorescence protein (GFP) and relative analogs have been widely used as real-time reporters. However, GFP-family proteins require molecular oxygen for fluorescence maturation. Considering the strict anaerobic growth requirement of the clostridia, an oxygen-independent fluorescence reporter such as a flavin mononucleotide-based fluorescent protein (FbFP) can be used as an alternative fluorescence reporter. In this study, we synthesized and expressed the codon-optimized FbFP gene for C. beijerinckii (CbFbFP) based on the nucleotide sequence of Bacillus subtilis YtvA variant EcFbFP in C. beijerinckii NCIMB 8052 wild-type. Protein expression and in vivo fluorescence of CbFbFP in C. beijeirnckii were confirmed under anaerobic growth conditions. Through fluorescence-activated cell sorting (FACS), we isolated the bright cells from the heterogenous population of C. beijerinckii cells expressing CbFbFP. Several mutations were found in the isolated plasmid which may be responsible for the high-level expression of CbFbFP in C. beijerinckii . A gas chromatography-isotope dilution mass spectrometry (GC-IDMS) technique was developed for the quantification of two heavy polyaromatic hydrocarbons (PAHs), benz[ a]anthracene and benzo[ a]pyrene, in yerba maté tea (maté). The optimisation of two extraction methods, namely liquid-liquid extraction and accelerated solvent extraction, was carried out. Both optimised methods were validated using a certified reference material of fine dust and the results were within the expanded uncertainties at 95% confidence level. Recoveries of 99.2-106.7% with RSD of measurements of 1.1-2.3% were achieved for benz[ a]anthracene. Recoveries of 95.7-101.9% with RSD of measurements of 0.4-1.4% were achieved for benzo[ a]pyrene. The validated methods were applied for the extraction of benz[ a]anthracene and benzo[ a]pyrene in maté powder from NIST. Four inorganic-organic hybrid compounds composed of Strandberg-type organophosphomolybdate anion [(C 6 H 5 PO 3 ) 2 Mo 5 O 15 ] 4− (abbreviated as (PhP) 2 Mo 5 ) and transition metal (TM)−2,2'-bipy/H 2 O complex units, namely [(TM(H 2 O)(bipy)) 2 (C 6 H 5 PO 3 ) 2 Mo 5 O 15 ] n (TM = Co, ( 1 ); Ni, ( 2 )), [(Cu(bipy) 2 ) 2 (C 6 H 5 PO 3 ) 2 Mo 5 O 15 ]·2H 2 O ( 3 ) and [(Zn(bipy)(µ-OH)) 2 (Zn(bipy) 2 (C 6 H 5 PO 3 ) 2 Mo 5 O 15 ) 2 ]·3H 2 O ( 4 ) (bipy = 2,2'-bipyridine), were successfully constructed under hydrothermal conditions, and their structures were determined by single crystal X-ray diffraction analysis and spectroscopic methods. Supervised classification of hyperspectral images (HSI) is a very challenging task due to the existence of noisy and mixed spectral characteristics. Recently, the widely developed spectral unmixing techniques offer the possibility to extract spectral mixture information at a subpixel level, which can contribute to the categorization of seriously mixed spectral pixels. Besides, it has been demonstrated that the discrimination between different materials will be improved by integrating the geometry and structure information, which can be derived from the variance between neighboring pixels. Furthermore, by incorporating the spatial context, the superpixel-based spectral–spatial similarity information can be used to smooth classification results in homogeneous regions. Therefore, a novel fusion framework for HSI classification that combines subpixel, pixel, and superpixel-based complementary information is proposed in this paper. Here, both feature fusion and decision fusion schemes are introduced. For the feature fusion scheme, the first step is to extract subpixel-level, pixel-level, and superpixel-level features from HSI, respectively. Then, the multiple feature-induced kernels are fused to form one composite kernel, which is incorporated with a support vector machine (SVM) classifier for label assignment. For the decision fusion scheme, class probabilities based on three different features are estimated by the probabilistic SVM classifier first. Then, the class probabilities are adaptively fused to form a probabilistic decision rule for classification. Experimental results tested on different real HSI images can demonstrate the effectiveness of the proposed fusion schemes in improving discrimination capability, when compared with the classification results relied on each individual feature. Aim: Rheumatoid arthritis (RA) is a chronic inflammation mediated by an autoimmune response. Baculoviral IAP repeat‐containing 2 (BIRC2) and tumor necrosis factor receptor 1‐associated death domain protein (TRADD) have been reported to be highly expressed in RA, while their specific roles during RA progression remain unclear. This study aims to explore the specific regulation of BIRC2/TRADD during the progression of RA. Methods: C28/I2 cells were stimulated by lipopolysaccharide (LPS) to establish an in vitro RA cellular model. The expression level of BIRC2 and TRADD was examined by quantitative real‐time polymerase chain reaction and western blot. Cell Counting Kit‐8 and flow cytometry assays were performed to examine cell viability and necroptosis, respectively. The oxidative stress markers were detected using commercial kits, and the pro‐inflammatory cytokines were measured by ELISA assay. The interaction between BIRC2 and TRADD was verified by co‐immunoprecipitation assay. Results: BIRC2 and TRADD were discovered to be highly expressed in LPS‐mediated C28/I2 cells. BIRC2 knockdown was demonstrated to inhibit LPS‐induced cell viability loss, necroptosis, oxidative stress, and inflammation in C28/I2 cells. BIRC2 could interact with TRADD and positively regulate TRADD expression. In addition, the protective role of BIRC2 knockdown against LPS‐mediated injuries in C28/I2 cells was partly weakened by TRADD overexpression. Conclusion: In summary, BIRC2 knockdown alleviated necroptosis, oxidative stress, and inflammation in LPS‐mediated C28/I2 cells, which might correlate to the regulatory role of TRADD, indicating a novel target for the treatment of RA. A novel hyperspectral image (HSI) classification method by the probabilistic fusion of pixel-level and superpixel-level classifiers is proposed. Generally, pixel-level classifiers based on spectral information only may generate “salt and pepper” result in the classification map since spatial correlation is not considered. By incorporating spatial information in homogeneous regions, the superpixel-level classifiers can effectively eliminate the noisy appearance. However, the classification accuracy will be deteriorated if undersegmentation cannot be fully avoided in superpixel-based approaches. Therefore, it is proposed to adaptively combine both the pixel-level and superpixel-level classifiers, to improve the classification performance in both homogenous and structural areas. In the proposed method, a support vector machine classifier is first applied to estimate the pixel-level class probabilities. Then, superpixel-level class probabilities are estimated based on a joint sparse representation. Finally, the two levels of class probabilities are adaptively combined in a maximum a posteriori estimation model, and the classification map is obtained by solving the maximum optimization problem. Experimental results on real HSI images demonstrate the superiority of the proposed method over several well-known classification approaches in terms of classification accuracy. A novel set-to-set distance-based spectral–spatial classification method for hyperspectral images (HSIs) is proposed. In HSIs, the spatially connected and spectrally similar pixels within each homogeneous region can be considered as one set of test samples, i.e., a test set, which should belong to the same class. In addition, each class of labeled pixels can be regarded as one set of training samples, i.e., a training set. Therefore, it is a natural consideration in the proposed method to measure the similarity between test and training sets via specific set-based distance criteria and then decide the classification label for each test set, accordingly. Specifically, the superpixel algorithm-based oversegmentation technique jointly exploits both the spatial similarity and structural information to first divide the HSI into multiple perceptually uniform regions. As a result, each segmented region corresponds to one test set. Then, each test/training set is represented with an affine hull (AH) model, which utilizes both the similarity and variance of pixels within each set to adaptively characterize the set. Finally, the class label for each test set is determined based on the closest geometry distance between test and training AHs. Experimental results on real HSI data sets demonstrate the superiority of the proposed algorithm over several well-known classification approaches, in terms of classification accuracy and computational speed. Aims. To measure plasma levels of superoxide dismutases 1, 2, and 3 (SOD1, 2, 3) and determine whether SODs can function as biomarkers for coronary artery disease (CAD). Patients & Methods. Patient groups were as follows: patients with stable angina pectoris (SAP, n=33), patients with acute coronary syndrome (ACS, n=49), and controls (n=42). Protein quantification was done using ELISA. Results. The concentrations of plasma SOD1 and SOD2 were higher in CAD than in healthy controls. No difference in SOD3 levels between CAD and control groups was found. Limited correlations were found between SODs and gender, age, and severity of coronary artery stenosis. Conclusions. Aberrant methylation has been associated with transcriptional inactivation of tumor-related genes in a wide spectrum of human neoplasms. The influence of DNA methylation in oligodendroglial tumors is not fully understood. Genomic DNA was isolated from 61 oligodendroglial tumors for analysis of methylation using methylation-specific multiplex ligation-dependent probe amplification assay ( MS- MLPA). We correlated methylation status with clinicopathological findings and outcome. The genes found to be most frequently methylated in oligodendroglial tumors were RASSF1A (80.3%), CASP8 (70.5%), and CDKN2A (52.5%). Kaplan-Meier survival curve analysis demonstrated longer duration of progression-free survival in patients with 19q loss, aged less than 38 years, and with a proliferative index of less than 5%. Methylation of the ESR1 promoter is significantly associated with shorter duration of overall survival and progression-free survival, and that methylation of IGSF4 and RASSF1A is significantly associated with shorter duration of progression-free survival. However, none of the methylation status of ESR1, IGSF4, and RASSF1A was of prognostic value for survival in a multivariate Cox model. A number of novel and interesting epigenetic alterations were identified in this study. Aneurysmal subarachnoid hemorrhage (aSAH) is one of the most severe neurological disorders, with a high mortality rate and severe disabling functional sequelae. Systemic inflammation following hemorrhagic stroke may play an important role in mediating intracranial and extracranial tissue damage. Previous studies showed that various systemic inflammatory biomarkers might be useful in predicting clinical outcomes. Anti-inflammatory treatment might be a promising therapeutic approach for improving the prognosis of patients with aSAH. China has implemented a series of long-term measures to control the spread of COVID-19, however, the effects of these measures on other chronic and acute respiratory infectious diseases remain unclear. Tuberculosis (TB) and scarlet fever (SF) serve as representatives of chronic and acute respiratory infectious diseases, respectively. In China's Guizhou province, an area with a high prevalence of TB and SF, approximately 40,000 TB cases and hundreds of SF cases are reported annually. To assess the impact of COVID-19 prevention and control on TB and SF in Guizhou, the exponential smoothing method was employed to establish a prediction model for analyzing the influence of COVID-19 prevention and control on the number of TB and SF cases. Additionally, spatial aggregation analysis was utilized to describe spatial changes in TB and SF before and after the COVID-19 outbreak. The parameters of the TB and SF prediction models are R2 = 0.856, BIC = 10.972 and R2 = 0.714, BIC = 5.325, respectively. TB and SF cases declined rapidly at the onset of COVID-19 prevention and control measures, with the number of SF cases decreasing for about 3–6 months and the number of TB cases remaining in decline for 7 months after the 11th month. The spatial aggregation of TB and SF did not change significantly before and after the COVID-19 outbreak but exhibited a marked decrease. These findings suggest that China's COVID-19 prevention and control measures also reduced the prevalence of TB and SF in Guizhou. These measures may have a long-term positive impact on TB, but a short-term effect on SF. Areas with high TB prevalence may continue to experience a decline due to the implementation of COVID-19 preventive measures in the future. [ABSTRACT FROM AUTHOR]

Synthetic microbial consortia represent a frontier of synthetic biology that promises versatile engineering of cellular functions. They are primarily developed through the design and construction of cellular interactions that coordinate individual dynamics and generate collective behaviors. Here we review recent advances in the engineering of synthetic communities through cellular-interaction programming. We first examine fundamental building blocks for intercellular communication and unidirectional positive and negative interactions. We then recap the assembly of the building blocks for creating bidirectional interactions in two-species ecosystems, which is followed by the discussion of engineering toward complex communities with increasing species numbers, under spatial contexts, and via model-guided design. We conclude by summarizing major challenges and future opportunities of engineered microbial ecosystems. Background: MicroRNAs are small noncoding RNAs about 22 nt long that play key roles in almost all biological processes and diseases. The fluorescent labeling and lipofection are two common methods for changing the levels and locating the position of cellular miRNAs. Despite many studies about the mechanism of DNA/RNA lipofection, little is known about the characteristics, mechanisms and specificity of lipofection of fluorescent-labeled miRNAs. Methods and Results: Therefore, miRNAs labeled with different fluorescent dyes were transfected into adherent and suspension cells using lipofection reagent. Then, the non-specific binding and its mechanism were investigated by flow cytometer and laser confocal microscopy. The results showed that miRNAs labeled with Cy5 (cyanine fluorescent dye) could firmly bind to the surface of adherent cells (Hela) and suspended cells (K562) even without lipofection reagent. The binding of miRNAs labeled with FAM (carboxyl fluorescein) to K562 cells was obvious, but it was not significant in Hela cells. After lipofectamine reagent was added, most of the fluorescently labeled miRNAs binding to the surface of Hela cells were transfected into intra-cell because of the high transfection efficiency, however, most of them were still binding to the surface of K562 cells. Moreover, the high-salt buffer which could destroy the electrostatic interactions did not affect the above-mentioned non-specific binding, but the organic solvent which could destroy the hydrophobic interactions eliminated it. Conclusions: These results implied that the fluorescent-labeled miRNAs could non-specifically bind to the cell surface by hydrophobic interaction. It would lead to significant errors in the estimation of transfection efficiency only according to the cellular fluorescence intensity. Non-Hodgkin’s lymphomas (NHLs) are a heterogeneous group of neoplasm in which 90% are B-cell lymphomas and 10% T-cell lymphomas. Although T-helper 17 (Th17) cells have been implicated to be essential in the pathogenesis of autoimmune and inflammatory diseases, its role in B-cell non-Hodgkin’s lymphoma (B-NHL) remains unknown. In this study, we observed a significantly decreased frequency of Th17 cells in peripheral blood from B-NHL patients compared with healthy individuals, accompanied with increased Th1 cells. IL-17AF plasma levels were remarkably decreased in B-NHL patients, accompanied with undetectable IL-17FF and unchangeable IL-17AA. Moreover, Th17 and Th1 cells became normalized after one or two cycles of chemotherapy. Interestingly, in B-NHL, circulating Th17 cells frequencies were significantly higher in relapsed patients than those in untreated patients or normal individuals. Meanwhile, there was no statistical difference regarding the frequencies of Th1 cells between relapsed and untreated patients. In this paper, a novel spectral–spatial adaptive sparse representation (SSASR) method is proposed for hyperspectral image (HSI) denoising. The proposed SSASR method aims at improving noise-free estimation for noisy HSI by making full use of highly correlated spectral information and highly similar spatial information via sparse representation, which consists of the following three steps. First, according to spectral correlation across bands, the HSI is partitioned into several nonoverlapping band subsets. Each band subset contains multiple continuous bands with highly similar spectral characteristics. Then, within each band subset, shape-adaptive local regions consisting of spatially similar pixels are searched in spatial domain. This way, spectral–spatial similar pixels can be grouped. Finally, the highly correlated and similar spectral–spatial information in each group is effectively used via the joint sparse coding, in order to generate better noise-free estimation. The proposed SSASR method is evaluated by different objective metrics in both real and simulated experiments. The aberrations of TP53 gene and dysregulation of the TP53 pathway are important in the pathogenesis of many human cancers, including malignant lymphomas, especially for diffuse large B cell lymphoma (DLBCL). By regulating many downstream target genes or molecules, TP53 governs major defenses against tumor growth and promotes cellular DNA repair, apoptosis, autophagy, cell cycle arrest, signaling, transcription, immune or inflammatory responses and metabolism. Dysfunction of TP53, including microRNA regulations, copy number alterations of TP53 pathway and TP53 itself, dysregulation of TP53 regulators, and somatic mutations by abnormal TP53 function modes, play an important role in lymphoma generation, progression and invasion. The role of TP53 in DLBCL has been widely explored recently.
