The Mediterranean will be one of the focal points of climate change. The predicted dry and hot summers will lead to water scarcity in agriculture, which may limit crop production and growth. The olive tree serves as a model woody plant for studying drought stress and improving water resource management; thus, it is critical to identify genotypes that are more drought tolerant and perform better under low irrigation or even rainfed conditions. In this study, the metabolomic approach was used to highlight variations in metabolites in stems and leaves of three Italian olive cultivars (previously characterized physiologically) under two and four weeks of drought stress. Phenolic and lipophilic profiles were obtained by gas chromatography-mass spectrometry and ultra-high performance liquid chromatography-mass spectrometry, respectively. The findings identified the leaf as the primary organ in which phenolic variations occurred. The Maurino cultivar exhibited a strong stress response in the form of phenolic compound accumulation, most likely to counteract oxidative stress. The phenolic compound content of ‘Giarraffa’ and ‘Leccino’ plants remained relatively stable whether they were exposed to drought or not. Variations in the lipid profile occurred in leaves and stems of all the cultivars. A high accumulation of compounds related to epicuticular wax components was observed in the leaf of ‘Giarraffa’, while a strong reduction of lipids and long-chain alkanes occurred in ‘Maurino’ when exposed to drought stress conditions. Since the beginning of the XXI century, Leishmaniasis has been integrated into the World Health Organization's list of the 20 neglected tropical diseases, being considered a public health issue in more than 88 countries, especially in the tropics, subtropics, and the Mediterranean area. Statistically, this disease presents a world prevalence of 12 million cases worldwide, with this number being expected to increase shortly due to the 350 million people considered at risk and the 2–2.5 million new cases appearing every year. The lack of an appropriate and effective treatment against this disease has intensified the interest of many research groups to pursue the discovery and development of novel treatments in close collaboration with the WHO, which hopes to eradicate it shortly. This paper intends to highlight the quinoline scaffold's potential for developing novel antileishmanial agents and provide a set of structural guidelines to help the research groups in the medicinal chemistry field perform more direct drug discovery and development programs. Thus, this review paper presents a thorough compilation of the most recent advances in the development of new quinoline-based antileishmanial agents, with a particular focus on structure–activity relationship studies that should be considerably useful for the future of the field. Simple Summary: The production of halophyte plants, like Salicornia ramosissima, is increasing as they are highly valued for human consumption and due to their ability to grow in unutilized cultivation areas, like saline soils. However, only the green tips are commercialized, while the remaining plant is considered a residue. This study aimed to explore the potential of these salicornia by-products to be included in aquafeeds for juvenile European seabass, partially replacing wheat meal. The results obtained indicate that this substitution is possible with no apparent adverse effects on the fish and on the economic viability of the feeds. Additionally, the inclusion of salicornia in the diets was related to the decrease in compounds in fish muscle usually used as stress biomarkers and with the increase in DHA levels, one of the most relevant Omega-3 fatty acids for human nutrition. Therefore, data from this study suggest that salicornia by-products are a viable alternative to partially replace wheat meal in diets for juvenile European seabass, allowing salicornia farmers to valorize a residue and to implement the principles of circular economy in halophyte farming and the aquaculture industry. A standard diet and three experimental diets including salicornia in 2.5%, 5% and 10% inclusion levels were tested in triplicate. After 62 days of feeding, no significant differences between treatments were observed in fish growth performances, feeding efficiency and economic conversation ratio. Nutrient digestibility of the experimental diets was unaffected by the inclusion of salicornia when compared to a standard diet. Additionally, salicornia had significant modulatory effects on the fish muscle biochemical profiles, namely by significantly decreasing lactic acid and increasing succinic acid levels, which can potentially signal health-promoting effects for the fish. Increases in DHA levels in fish fed a diet containing 10% salicornia were also shown. Therefore, the results suggest that salicornia by-products are a viable alternative to partially replace wheat meal in diets for juvenile European seabass, contributing to the valorization of a residue and the implementation of a circular economy paradigm in halophyte farming and aquaculture. Solenostemma oleifolium is a tropical plant growing in the Algerian desert that is traditionally used to treat several illnesses, including infection. We investigate Essential Oil components from leaves of S. oleifolium (EOSO) and its antibacterial activity. Using Gas Chromatography and Mass Spectrometry (GCMS), twenty compounds were identified in EOSO, including linalool (57.10%), terpineol (12.95%), trans-geraniol (12.65%), and nerol (4.67%). Nuclear magnetic resonance (NMR) analysis allowed us to confirm linalool as the main component of EOSO. Antibacterial activity was tested by agar diffusion and microdilution methods for minimal inhibitory concentration (MIC) and minimal bactericidal concentration (MBC). For EOSO, the inhibition diameters ranged from 16.79 to 39.84 mm, the MIC ranged from 1.066 to 8.54 mg mL-1 and the MBC ranged from 20 to 100 mg mL-1. For linanool, the inhibition diameters ranged from 11.1 to 31.87 mm, the MIC ranged from 2.68 to 14.3 mg mL-1 and, the MBC ranged from 40 to 100 mg mL-1. EOSO and linalool exhibited significant antibacterial activity against all the tested bacteria. Chestnut shells (CS), the principal by-product of the chestnut processing industry, contain high concentrations of flavonoids and other polyphenols with huge interest for the nutraceuticals field. Nonetheless, the bioaccessibility and bioactivity of phytochemicals can be influenced by their digestibility, making it imperative to evaluate these activities prior to application of CS as a nutraceutical ingredient. This work aims to appraise the effects of in vitro simulated gastrointestinal digestion on the bioaccessibility, bioactivity, and metabolic profiling of CS. An increase in the total phenolic and flavonoid contents, antioxidant/antiradical properties, radical scavenging capacity, and inhibition on acetylcholinesterase activity was evidenced during in vitro simulated digestion. Metabolomic profiling by LC-ESI-LTQ-Orbitrap-MS revealed changes during the simulated digestion, particularly in phenolic compounds (46% of total compounds annotated), lipids (22%), phenylpropanoids (9%), organic acids (7%), carbohydrates (5%), nucleosides (5%), amino acids (4%), and alcohols (1%). Phenolic acids (gallic acid, syringic acid, and hydroxyphenylacetic acid) and flavonoids (epicatechin) were the major polyphenolic classes identified. The heatmap-positive correlations highlighted that the bioactivity of CS is closely related to the phenolic compounds and their bioaccessibility. These findings suggest the reuse of CS as a potential nutraceutical ingredient with antioxidant and neuroprotective effects, encouraging the use of appropriate extraction and/or encapsulation techniques to enhance the bioaccessibility of phenolic compounds. (1) Background: In recent years, numerous studies have highlighted the beneficial effects of extra virgin olive oil (EVOO) as an active ingredient against chronic diseases. The properties of EVOO are due to its peculiar composition, mainly to its rich content of polyphenols. In fact, polyphenols may contribute to counteract oxidative stress, which often accompanies chronic diseases. In this work, the antioxidant effects of high-value polyphenol oleocanthal (OC) and its main metabolites, tyrosol (Tyr) and oleocanthalic acid (OA), respectively, have been investigated along with their impact on cell viability. (2) Methods: OC, Tyr, and OA have been evaluated regarding antiradical properties in term of scavenging capacity towards biologically relevant reactive species, including O2●−, HOCl, and ROO●, as well as their antioxidant/antiradical capacity (FRAP, DPPH●, ABTS●+). Moreover, the ability to permeate the intestinal membrane was assessed by an intestinal co-culture model composed by Caco-2 and HT29-MTX cell lines. (3) Results: The capacity of OC and Tyr as radical oxygen species (ROS) scavengers, particularly regarding HOCl and O2●−, was clearly demonstrated. Furthermore, the ability to permeate the intestinal co-culture model was plainly proved by the good permeations (>50%) achieved by all compounds. Chestnut shells (CSs) are an appealing source of bioactive molecules, and constitute a popular research topic. This study explores the effects of in vitro gastrointestinal digestion and intestinal permeability on the bioaccessibility and bioactivity of polyphenols from CS extract prepared by subcritical water extraction (SWE). The results unveiled higher phenolic concentrations retained after gastric and intestinal digestion. The bioaccessibility and antioxidant/antiradical properties were enhanced in the following order: oral < gastric ≤ intestinal digests, attaining 40% of the maximum bioaccessibility. Ellagic acid was the main polyphenol in the digested and undigested extract, while pyrogallol–protocatechuic acid derivative was only quantified in the digests. The CS extract revealed potential mild hypoglycemic (<25%) and neuroprotective (<75%) properties before and after in vitro digestion, along with upmodulating the antioxidant enzymes' activities and downregulating the lipid peroxidation. The intestinal permeation of ellagic acid achieved 22.89% after 240 min. The present study assessed two different plant parts (leaves and tubers) of Arum italicum species growing in Northeast Algeria for their phytochemical composition and pharmacological effects. The phytochemical content was determined using ultra-high-performance liquid chromatography coupled with a diode array detector and an electrospray mass spectrometer (UHPLC-DADESI/MS). The results revealed that the tuber extract was rich in lignans with a fraxiresinol glycoside as the major compound. In contrast, the leaf extract was rich in flavonoid glycosides, described for the first time in the aerial part of this species. The extract’s inhibitory activity against key enzymes was linked to hyperglycemia, α-glucosidase, and α-amylase, and their ability to inhibit the growth of human gastric carcinoma (AGS) and lung carcinoma (A549) cancer cell lines was also assessed. A cell line morphology study was also conducted with the most effective extract. The chromatin status of the cells was evaluated using DAPI, while the cytoplasmic morphology was evaluated using phalloidin. The tuber extract generally inhibited α-glucosidase and α-amylase enzymes more efficiently than the leaf extract. Its inhibition effect against the α-glucosidase was significantly higher when compared to the standard acarbose. The tuber extract also caused more viability loss of AGS and A549 cancer cells than the leaf extract in the cytotoxicity assay. In conclusion, our findings show that, compared to the leaf extract, the tuber extract exhibited more pronounced biological effects. The strong inhibitory potential of the tuber extract against the α-glucosidase enzyme should also be highlighted, which suggests it is a good candidate for discovering new antidiabetic agents. This study demonstrates an investigation into nanomineralogical and geochemical evolution for the detection of hazardous elements from old, abandoned coal mining deposits capable of causing negative environmental impacts. The general objective of this study is to evaluate the number of nanoparticulate chemical elements in sediments collected during the years 2017 and 2022 from deactivated coal mining areas in the La Guajíra and Cesar regions of Colombia. Sediments were collected and analyzed from areas that experienced spontaneous coal combustion (SCC). The analysis consisted of traditional mineralogical analysis by X-ray diffraction and Raman spectroscopy, nanomineralogy by field emission scanning electron microscope-FE-SEM, and high-resolution transmission electron microscope-HR-TEM (energy dispersive X-ray microanalysis system-EDS). The analyzed sediment samples contained high proportions of amorphous materials containing the chemical elements As, Cl, Hg, Mo, Pb, Sb, and Se. This study emphasizes the need to implement environmental recovery projects at former, now abandoned coal extraction areas located in the investigated region, as they have negative effects on the environment and human health across large regions. Chestnut (Castanea sativa) shells (CSs), an undervalued agro-industrial biowaste, have arisen as a source of bioactive compounds with promising health-promoting effects. This study attempted, for the first time, to develop a functional food, namely cookies, using a CS extract obtained by an eco-friendly technology (subcritical water extraction). The cookies were characterized regarding their nutritional composition, total phenolic and flavonoid contents (TPC and TFC, respectively), antioxidant/antiradical activities, phenolic profile, and sensory evaluation. The results demonstrated that the CS-extract-enriched cookies were mainly composed of carbohydrates (53.92% on dry weight (dw)), fat (32.62% dw), and fiber (5.15% dw). The phenolic profile outlined by HPLC-PDA revealed the presence of phenolic acids, flavonoids, and hydrolysable tannins, attesting to the high TPC and TFC. The in vitro antioxidant/antiradical effects proved the bioactivity of the functional cookies, while the sensory evaluation unveiled excellent scores on all attributes (≥6.25). The heatmap diagram corroborated strong correlations between the TPC and antioxidant/antiradical properties, predicting that the appreciated sensory attributes were closely correlated with high carbohydrates and phenolic compounds. Metabolomics is a powerful tool in diverse research areas, enabling an understanding of the response of organisms, such as plants, to external factors, their resistance and tolerance mechanisms against stressors, the biochemical changes and signals during plant development, and the role of specialized metabolites. Despite its advantages, metabolomics is still underused in areas such as nano-plant interactions. Nanoparticles (NPs) are all around us and have a great potential to improve and revolutionize the agri-food sector and modernize agriculture. They can drive precision and sustainability in agriculture as they can act as fertilizers, improve plant performance, protect or defend, mitigate environmental stresses, and/or remediate soil contaminants. Given their high applicability, an in-depth understanding of NPs' impact on plants and their mechanistic action is crucial. Being aware that, in nano-plant interaction work, metabolomics is much less addressed than physiology, and that it is lacking a comprehensive review focusing on metabolomics, this review gathers the information available concerning the metabolomic tools used in studies focused on NP-plant interactions, highlighting the impact of metal-based NPs on plant metabolome, metabolite reconfiguration, and the reprogramming of metabolic pathways. Leishmaniasis is one of the most neglected diseases in modern times, mainly affecting people from developing countries of the tropics, subtropics and the Mediterranean basin, with approximately 350 million people considered at risk of developing this disease. The incidence of human leishmaniasis has increased over the past decades due to failing prevention and therapeutic measures—there are no vaccines and chemotherapy, which is problematic. Acridine derivatives constitute an interesting group of nitrogen-containing heterocyclic compounds associated with numerous bioactivities, with emphasis to their antileishmanial potential. The present work builds on computational studies focusing on a specific enzyme of the parasite, S-adenosylmethionine decarboxylase (AdoMet DC), with several 1,2,3,4-tetrahydro-acridines emerging as potential inhibitors, evidencing this scaffold as a promising building block for novel antileishmanial pharmaceuticals. Thus, several 1,2,3,4-tetrahydroacridine derivatives have been synthesized, their activity against Leishmania (Leishmania) infantum promastigotes evaluated and a structure–activity relationship (SAR) study was developed based on the results obtained. Even though the majority of the 1,2,3,4-tetrahydroacridines evaluated presented high levels of toxicity, the structural information gathered in this work allowed its application with another scaffold (quinoline), leading to the obtention of N1,N12-bis(7-chloroquinolin-4-yl)dodecane-1,12-diamine (12) as a promising novel antileishmanial agent (IC50 = 0.60 ± 0.11 μM, EC50 = 11.69 ± 3.96 μM and TI = 19.48). Salicornia perennis is a halophyte belonging to the botanical subfamily Salicornioideae that forms extensive perennial salt marsh patches. This subfamily has excellent potential, still unexplored, as a source of food, medicine, and phytoremediation. This study aimed to evaluate the lipophilic composition of the Salicornia perennis different organs inhabiting salt marshes of Ria de Aveiro under different stress regimes. For this purpose, the lipophilic content was extracted with hexane and subsequent GC-MS analysis of the extracts for each plant organ, which was collected in three different salt marshes of the Ria de Aveiro. High sugar content was detected in the stems, whereas in fruiting articles, the higher content was in fatty acids. Shorter-chain organic acids were concentrated in the stems and vegetative articles; waxes were detected in greater quantity in photosynthetic organs. More or less stressful environments induce changes in the ratio and composition of molecules, such as acclimatization and oxidative stress reduction strategies; for example, fatty acid content was higher in plants subjected to a higher stress regime. These data contribute to understand the metabolic pathways of the species under study, suggesting new research approaches to its potential as food, medicine, and phytoremediator. The objective of this study is to obtain a more detailed assessment of particles that contain rare-earth elements (REEs) in abandoned deposits of Brazilian fine coal tailings (BFCTs), so as to aid current coal mining industries in the identification of methodologies for extracting such elements (Santa Catarina State, Brazil). The BFCT areas were sampled for traditional mineralogical analysis by X-ray Diffraction, Raman Spectroscopy and nanomineralogy by a dual beam focused ion beam (FIB) coupled with field emission scanning electron microscopy (FE-SEM) and high-resolution transmission electron microscopy (HR-TEM) coupled with an energy dispersive X-ray microanalysis system (EDS). The results show that the smaller the sampled coal fines were, the higher the proportion of rare-earth elements they contained. Although the concentration of REEs is below what would normally be considered an economic grade, the fact that these deposits are already ground and close to the surface negate the need for mining (only uncovering). This makes it significantly easier for REEs to be extracted. In addition, owing to their proximity to road and rail transport in the regions under study, the opportunity exists for such resources (BFCTs) to be utilized as a secondary market as opposed to simply being discarded as has been done in the past. Cystoseira abies-marina (reclassified as Gongolaria abies-marina) is a brown seaweed species rich in meroterpenoids, presenting interesting antioxidant, antitumor, and anti-inflammatory activities. However, there is still a lot to uncover regarding the bioactive potential of this species, as evidenced by the lack of records of antiaging activities from Cystoseira abies-marina, making this macroalga an excellent candidate for studies of its cosmeceutical potential. Ultrasound-(UAE) and microwave-assisted extraction (MAE) are advanced sustainable technologies that are very efficient in enhancing bioactive compound extraction. Applying these extraction techniques to a new biological matrix often calls for optimizing the parameters toward the best extraction yield. Since Cystoseira abies-marina is a new matrix for both UAE and MAE techniques, the present work proposes the optimization of the extraction process, using a novel approach: instead of only focusing on increasing the yield, the goal of this work is to determine the parameters for UAE and MAE that lead to extracts with better antiaging activities. For this bio-guided approach, several Cystoseira abies-marina extracts were prepared by UAE and MAE under varying conditions of solvent, time, and algae/solvent ratios. Their antiaging activities were then determined, and all the results combined to unveil the conditions yielding extracts with higher cosmeceutical potential. Using statistical tools, it was found that, for UAE, the best conditions were ethyl acetate, 15 min, and a ratio of 1:4, which led to an extract with high yield, and causing the strong inhibition of tyrosinase and elastase. In turn, ethanol, 10 min, and a ratio of 1:4 were the best conditions for MAE, leading to the extract with the best antioxidant activity. The results show that the proposed bio-guided approach was effective in obtaining extracts with high cosmeceutical potential, unveiling the possibility of modulating an extract's activity by changing the extraction method. This work focused on developing an active bilayer film based on natural extract. Thus, the jaboticaba peel extract (JPE) was produced and characterized and showed promising application as a natural additive in biopolymeric materials. The zein fiber and bilayer films were produced using a chitosan film (casting) and zein fiber (electrospinning), with and without JPE. All samples were evaluated according to thickness, solubility in water, water vapor permeability, and main diameter, and for these, zein fiber, chitosan/zein fiber, and chitosan/zein fiber + 3% JPE showed values of 0.19, 0.51, and 0.50 mm, 36.50, 12.96, and 27.38%, 4.48 × 10−9, 1.6 × 10−10, and 1.58 × 10−10 (g m−1 Pa−1 s−1), and 6.094, 4.685, and 3.620 μm, respectively. These results showed that the addition of a second layer improved the barrier properties of the material when compared to the monolayer zein fiber. The thermal stability analysis proved that the addition of JPE also improved this parameter and the interactions between the components of the zein fiber and bilayer films; additionally, the effective presence of JPE was shown through FTIR spectra. In the end, the active potential of the material was confirmed by antimicrobial analysis since the bilayer film with JPE showed inhibition halos against E. coli and S. aureus. [ABSTRACT FROM AUTHOR]

In the present study, two extracts from the aerial parts of the endemic species Satureja hispidula were analyzed for the first time by ultra-high-performance liquid chromatography coupled with a diode array detector and an electrospray mass spectrometer (UHPLC-DAD-ESI/MS) method in order to identify and quantify their phenolic compounds. These extracts' antioxidant, α-glucosidase and α-amylase inhibitory activities were also evaluated. UHPLC-DAD-ESI/MS allowed the identification of 28 and 20 compounds in the ethanolic and aqueous extracts, respectively; among them, 5-O-caffeoylquinic acid was the most abundant in both extracts. The biological assay results indicate that the species S. hispidula, besides its high antioxidant power, is also potentially useful for inhibiting the α-glucosidase enzyme. In both antioxidant and α-glucosidase inhibitory assays, the aqueous extract exhibited the most promising results, significantly better than the standards used as positive controls. The use of images from the Sentinel-3B SYN satellite (surface reflectance and aerosol parameters over land) is currently one of the most advanced technologies utilized to identify atmospheric aerosol concentrations on a global scale. The general aim of this study is to analyze the evolution of aerosols in the atmosphere of the Kiev region in northern Ukraine during 2019, 2020, 2021 and 2022. Due to this study's timing, both prior to and during the current military incursion into Ukraine, this study also evaluates the consequences of the invasion of the Russian army on the territory of Ukraine, in relation to the quantitative levels of aerosols present in the atmosphere. Satellite image data were modelled in SNAP software (Sentinel Application Platform). Using the JASP software (version 0.14.1.0), clusters with variations of T550 (Aerosol Optical Thickness) were generated. The Sentinel-3B SYN satellite images were made available by the European Space Agency (ESA), with moderate spatial resolution (>300 m), calibrated and normalized to an average standard of 0.83 µg/mg, with a maximum error of 6.62% in the 30 sampled points. Satellite image data were modelled in SNAP software. Using the JASP software (version 0.14.1.0), clusters with variations of T550 (Aerosol Optical Thickness) were generated. The results show variations in the concentration of T550 in different periods, revealing that the military conflict between Russia and Ukraine directly influenced the dynamics of aerosol concentration, attributed to factors incompatible with environmental sustainability.
