This paper constructs an immersive multimedia virtual interactive experience technology with the Great Wall World Heritage Site as the experimental object. By analyzing the VR vision system and algorithm, the process of stereo vision perception and the general algorithm of somatic image production are explained. The distance equation and depth calculation in stereoscopic projection are studied, and the virtual interactive experience technology is constructed and applied to the World Heritage Site of the Great Wall. The result data show that the immersive multimedia virtual interactive experience technology highlights the real feeling of users more, in which the model has 50% of the influence on the real feeling, the mapping has 15%, the lighting has 15%, and the rendering has 10%. The youth population has a higher percentage of interest in the virtual field, accounting for 50%, 65%, and 60%, respectively, while the middle-aged stage has a more average degree of interest in the virtual field, with 50% of the average interest, 30% of happy to experience, 25% of not interested and 50% of interested. This thesis is conducive to enriching the project of the Great Wall World Heritage Site and providing an effective reference for technological innovation of immersive multimedia virtual interactive experience.

Double-layer composite water-lubricated bearing is a new type of water-lubricated bearing which can integrate the good damping performance of low elastic under-layer bush and good tribological performance of plastic layer bush. This paper analyzes its elastohydrodynamic lubrication performance by fluid–structure interaction (FSI) method, and studies the effects of eccentricity ratio, rotational speed, elastic modulus distribution and thickness distribution of bearing bush on its lubrication performance. Results show that the lubrication performance of double-layer bearing is more like that of plastic bearing. As rotational speed and eccentricity ratio increase, the maximum water film pressure, the load carrying capacity and the maximum bush deformation increase significantly. As the elastic modulus of the low elastic under-layer bush decreases, the total bush deformation increases significantly, but the load carrying capacity decreases slightly. The bush thickness distribution influences the deformation distribution of both low elastic under-layer bush and plastic layer bush, but have little impact on the total bush deformation and bearing lubrication performance.

In this paper, we are concerned with the nonlinear elliptic systems in divergence form under controllable growth condition. We prove that the weak solution u is locally Hölder continuous besides a singular set by using the direct method and classical Morrey-type estimates. Here the Hausdorff dimension of the singular set is less than n-p{n-p}. This result not only holds in the interior, but also holds up to the boundary.

This paper is an instance of howa Chinese language teacher trainer cooperated with a game-based learning (GBL) researcher to design and develop a mobile applicationfor learning daily communication in Mandarin. The teacher trainer intendsto assist beginning learners, specifically those who speak Malay as their first language, to establish confidence in speaking Mandarin in Malaysian‘sdaily living contexts; while the researcher has set out to testthe ecologicalvalidity of a GBL cooperation model. The interdisciplinary cooperation between theseindividuals has given rise tothe production ofaMandarin communication learning application. The key challenge encountered in the development has been to determinethechoice of contents and communication contexts for beginning learners. This paper illustrates how the challenges have been resolvedand discusses lessons learnt from the cooperation. The outcomes of the study would benefit academics, GBL researchersor developers who plan to initiate cross-disciplinary cooperation towards developinglanguage learning applications.

Strengthening bilateral energy security cooperation is crucial in the process of fostering the sustainable growth of China and neighboring Asian nations. Using data chosen from China and 25 adjacent nations between 2010 and 2019, this article first assesses the level of energy security using the TOPSIS (Technique of Ranking Preference with Similarity to Ideal Solution) and GRA (Grey Relational Analysis) methodologies. Additionally, based on the level of energy security, this article analyzes the coupling degree of bilateral capacity structure and, in the end, suggests a theoretical model to assess the stability of bilateral energy security cooperation. The findings demonstrate that China and its neighboring countries share the energy trilemma, which is the inability of these nations to simultaneously achieve the needs of energy supply security, energy justice, and environmental sustainability. The report also makes the case that Thailand, Kazakhstan, and the Philippines all have theoretically stronger sustainability of bilateral energy security cooperation with China. Employing Chinese customs data and the Peking University Digital Financial Inclusion Index of China, this paper studies the impact of China's digital finance development on the upgrading of export at the city level and further explores the heterogeneity across cities and the mechanisms through which digital finance influences export upgrading. Benchmark results suggest that digital inclusive finance can significantly promote the upgrading of export. The heterogeneity analysis shows that cities with a smaller size, lower wage, higher human capital level, and better location advantage experience greater facilitating effects of digital inclusive finance on promoting export upgrading. It suggests that, compared with 'icing on the cake', the digital inclusive finance plays a better role in 'offering fuel in snowy weather', whereas full exertion of the inclusiveness of digital finance requires higher human capital and location advantage. Further mechanism analysis shows that innovation effect and market effect are the main channels where digital inclusive finance promotes the upgrading of a city's export. [ABSTRACT FROM AUTHOR]

In the face of increasingly severe climate change and its destructive effects, how to effectively reduce carbon dioxide emissions has become a challenging task. Developing a digital economy provides opportunities for countries to reduce pollution and carbon emissions and reach a goal of carbon neutrality. As an emerging trade form, digitizable product trade is of great significance to promoting economic growth and carbon emission reduction. This paper selects panel data for 94 countries from 2001 to 2019 and adopts the STIRPAT model to analyze the impact effect and impact mechanism of digitizable product trade on carbon emissions. Research results show that developing digitizable product trade will help countries reduce carbon emissions. The conclusion is robust by replacing the explained variable and core explanatory variable. The carbon emission reduction effect has heterogeneity due to differentiated national income levels and product categories. Mechanism analysis shows that digitizable product trade reduces carbon emissions through the technology effect. Our analysis indicates that countries developing digital trade and digital technology and actively responding to environmental issues have a greater chance of reduced carbon emissions. This paper used a new transient computational fluid dynamics and fluid–structure interaction method to investigate the journal bearing performance with the effect of thermal and cavitation, to reveal the performance of journal bearing in different lubricants and to provide substitution references for bearings in different lubricants. Considering thermal effect, elastic deformation and cavitation, a detailed discussion was conducted to show the performance of plain journal bearings lubricated by water, seawater, and lubricating oil by computational fluid dynamics (CFD) and fluid structure interaction (FSI) method. And the results in this work are compared with the published results. The variation of dimensionless load carrying capacity, maximum film pressure and temperature with eccentricity ratio are presented, which can provide reference for the design of bearings. Furthermore, a diagram is presented for journal bearings with different diameter, length-diameter ratio and lubricants, which can be used as a reference for the equivalent substitutions of bearings. The present research provides references as to the design of bearings and the substitutions of bearings by different lubricants.

Wheat (Triticum aestivum) is a commercially important crop and its production is seriously threatened by the fungal pathogen Puccinia striiformis f. sp. tritici West (Pst). Resistance (R) genes are critical factors that facilitate plant immune responses. Here, we report a wheat R gene NB-ARC-LRR ortholog, TaYRG1 , that is associated with distinct alternative splicing events in wheat infected by Pst. The native splice variant, TaYRG1.6 , encodes internal-motif-deleted polypeptides with the same N- and C-termini as TaYRG1.1, resulting in gain of function. Transient expression of protein variants in Nicotiana benthamiana showed that the NB and ARC domains, and TaYRG1.6 (half LRR domain), stimulate robust elicitor-independent cell death based on a signal peptide, although the activity was negatively modulated by the CC and complete LRR domains. Furthermore, molecular genetic analyses indicated that TaYRG1.6 enhanced resistance to Pst in wheat. Moreover, we provide multiple lines of evidence that TaYRG1.6 interacts with a dynamin-related protein, TaDrp1. Proteome profiling suggested that the TaYRG1.6-TaDrp1-DNM complex in the membrane trafficking systems may trigger cell death by mobilizing lipid and kinase signaling in the endocytosis pathway. By constructing a technology spillover connectedness index and connectedness matrix, this paper studies the technology spillover connectedness among innovation agents in China from the perspective of network topology. An empirical study based on invention patent data finds that there is noticeable technology spillover connectedness among the six innovation agents: central enterprises, other domestic enterprises, universities and scientific research institutes, troops, individuals and other organizations, and foreign-funded enterprises. Other domestic enterprises are the hub of technology spillover connectedness among these agents, while foreign-funded enterprises play a limited role. The study of further subdividing foreign-funded enterprises according to their sources finds that the United States, South Korea, and Japan are the centers of technology spillover connectedness in China. The dynamic evolution of technology spillover connectedness shows that the total technology spillover connectedness among innovation agents presents an obvious downward trend, which is closely related to the complex and changeable international economic situation and the uncertainty of China's economic policy in recent years. This paper concludes with insights into how China uses the spillover connectedness among innovation agents to spur agents' innovation performance and promote the nation's economic growth and competitiveness. Thinopyrum ponticum (2 n = 10 x = 70) is a wild relative of wheat with high tolerance to both biotic and abiotic stresses; it has been wildly used in wheat genetic improvement. A disomic substitution line named SN19647 was derived from a cross between Triticum aestivum and the wheat– Th. ponticum partial amphiploid SNTE20 (2 n = 8 x = 56). It was evaluated for disease resistance and characterized via sequential fluorescence in situ hybridization (FISH)-genomic in situ hybridization (GISH) and molecular markers. The results showed that SN19647 carried resistance to both powdery mildew and leaf rust. It contained 42 chromosomes with a pair of wheat chromosome 1B replaced by a pair of JS chromosomes from Th. ponticum. In addition to chromosomal substitution events, structural variation also occurred on wheat chromosomes 2A, 5A, 6B, and 7B. Based on marker analysis, 19 markers specific to the JS chromosome were obtained, of which seventeen markers belonged to homoeologous group one. These results indicated that SN19647 was a 1JS (1B) substitution line. Compared with the known 1JS (1D) substitution line CH10A5, it was found that 17 markers generated different specific bands to Th. ponticum , confirming the novelty of the 1JS chromosome in SN19647. Therefore, SN19647, resistant to powdery mildew and leaf rust, was a novel 1JS (1B) substitution line that can be used in wheat genetic improvement. [ABSTRACT FROM AUTHOR]

The stiffness mutation of shield tunnel-shaft junction makes the tunnel structure affected by the differential displacement and forms a complex spatial effect. Taking the subsea shield tunnel crossing under the Shantou Gulf, China, as a case study, a three-dimensional finite element global model and a refined local spatial end submodel are established. The nonlinear dynamic behaviors of the seabed soil and concrete, the simulation of the bolt joints between ring segments by using cohesive models and the SMA shape memory alloy flexible joints, and the input ground motions produced by scaling from the high-level earthquake records are considered in detail. The results show that the shield tunnel spatial end structure increases nonlinearly in response to the increase of seismic motion intensity. The opening width and the deformation between ring segments at the vault and the outside spandrel are larger, and serious seismic damage and stress concentration exist at the conjugate 45° directions of shaft. The seismic responses of the tunnel-shaft junction subjected to the seismic motions with rich low-frequency components are much stronger than those of seismic motions with rich high-frequency components. Adding SMA flexible joints, the structural deformation caused by seismic motion propagation can be induced to the preset flexible joint, and the structural damage and stress concentration can be effectively reduced. The seismic response characteristics of shield tunnel spatial end structure calculated by the global model are consistent with those calculated by the submodel, while the seismic response of the submodel is greater than that of the global model. Alcoholic liver disease, caused by abuse and consumption of alcohol, exhibits high morbidity and mortality. Boletus aereus Bull. (Boletaceae) (BA) shows antioxidant, anti-inflammatory and antimicrobial effects. To investigate the hepatoprotective effects of BA using an acute alcohol-induced hepatotoxicity mice model. The composition of BA fruit body was first systematically analyzed. Subsequently, a C57BL/6 mice model of acute alcohol-induced liver injury was established by intragastrically administration of alcohol, which was intragastrically received with BA powder at 200 mg/kg and 800 mg/kg for 2 weeks, 60 mg/kg silybin treatment was used as positive control group. By employing the pathological examination, ELISA, RT-PCR and western blot, the regulation of BA on oxidative stress signals was investigated. The LD50 of BA was much higher than 4 g/kg/p.o. In acute alcohol-damaged mice, BA reduced the levels of alanine aminotransferase (>18.3%) and aspartate aminotransferase (>27.6%) in liver, increased the activity of liver alcohol dehydrogenase (>35.0%) and serum acetaldehyde dehydrogenase (>18.9%). BA increased the activity of superoxide dismutase (>13.4%), glutathione peroxidase (>11.0%) and 800 mg/kg BA strongly reduced chemokine (C-X-C motif) ligand 13 (14.9%) and chitinase-3 like-1 protein (13.4%) in serum. BA reversed mRNA over-expression (>70%) and phosphor-stimulated expression (>45.0%) of an inhibitor of nuclear factor κ-B kinase (NF-κB, an inhibitor of nuclear factor κ-B α and nuclear factor κ-B in the liver. BA is effective in ameliorating alcohol-induced liver injury through regulating oxidative stress-mediated NF-κB signalling, which provides a scientific basis for further research on its clinical applications. Thinopyrum ponticum (2 n = 10 x = 70), a member of the tertiary gene pool of wheat (Triticum aestivum L.), harbors many biotic and abiotic stress resistance genes. CH10A5, a novel disomic substitution line from a cross of T. aestivum cv. 7182 and Th. ponticum , was characterized by cytogenetic identification, in situ hybridization, molecular marker analysis, and morphological investigation of agronomic traits and disease resistance. Cytological observations showed that CH10A5 contained 42 chromosomes and formed 21 bivalents at meiotic metaphase I. Genome in situ hybridization (GISH) analysis indicated that two of its chromosomes came from the Js genome of Th. ponticum , and wheat 15K array mapping and fluorescence in situ hybridization (FISH) revealed that chromosome 1D was absent from CH10A5. Polymorphic analysis of molecular markers indicated that the pair of alien chromosomes belonged to homoeologous group one, designated as 1Js. Thus, CH10A5 was a wheat– Th. ponticum 1Js (1D) disomic substitution line. Field disease resistance trials demonstrated that the introduced Th. ponticum chromosome 1Js was probably responsible for resistance to both stripe rust and powdery mildew at the adult stage. Based on specific-locus amplified fragment sequencing (SLAF-seq), 507 STS molecular markers were developed to distinguish chromosome 1Js genetic material from that of wheat. Of these, 49 STS markers could be used to specifically identify the genetic material of Th. ponticum. CH10A5 will increase the resistance gene diversity of wheat breeding materials, and the markers developed here will permit further tracing of heterosomal chromosome fragments in the future. The present study intended to explore the anti-fatigue and anti-hypoxia efficacy of Tremella extract in mouse model. Kunming mice were randomized into 5 groups randomly (n = 12/group; half male and female), and given 0.9%Nacl (10 mL/Kg Serving as blank control group)orally, 0.6 g/kg rhodiola capsule (Serving as positive group) and Tremella extract at doses of high (3.0 g/kg), middle (1.5 g/kg) and low (0.5 g/kg) once a day for 14 days. At the end of drug intragastric gavage, following endurance running test, rotating rod test, normobarie hypoxia test and sodium nitrite poisoning test were performed. 14-day Tremella extract administration fails to influence mouse horizontal and vertical movement indicating little neurotoxicity at chosen doses. Through rotating rod, forced running test, the anti-fatigue activity of Tremella extract was demonstrated. Via normobarie hypoxia test, sodium nitrite poisoning test, Tremella extract was confirmed to possess anti-hypoxia effect. Tremella extract treatment significantly enhanced the liver levels of superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px), increased the expressions of ATP and hepatic glycogen, muscle glycogen in tissue. We conclude that Tremella extract efficiently enhances mouse fatigue endurance and anti-hypoxia capability partly through energy reserves and antioxidant enzyme activity. [ABSTRACT FROM AUTHOR]

Multiobjective subset selection problems widely exist in many real‐world applications. Multiobjective ant colony optimisation (MOACO) is a strong and kind instrument for settling those issues. However, it still has two shortages which the authors must resolve. One, its solution construction process is inconsistent with the disorder characteristics of solutions, which prevent it from getting better solutions. Two, conventional MOACOs which most deal with biobjective optimisation problems are difficult to figure out high‐dimensional objectives optimisation problems. In this study, the authors propose a new MOACO to resolve those two disadvantages. They give its detailed descriptions and an exhaust experiment. And the results on C measure, spacing measure and inverted generational distance (IGD) show that the proposed algorithm has a powerful convergence ability and gets a better balance between convergence and diversity compared with other state of art approaches. In this paper, CMOS wafer level ring oscillator frequency variability improvement >40% is demonstrated by either spatial source/drain activation or ion implantation super scan in FinFET technology. Yield improvement (up to 17%) is verified with better within wafer uniformity and with no intrinsic device performance degradation. Furthermore, the combination of super scan and spatial source/drain activation is suggested for optimized variability improvement benefits from individual device type uniformity tuning (super scan) and all device type uniformity tuning (from spatial S/D activation). Phosphoribosyl pyrophosphate synthetase 1 (PRPS1) is closely associated with a number of diseases; however, its influence in B-cell acute lymphoblastic leukemia (B-ALL) and the potential molecular mechanisms involved remain unclear. The present study aimed to evaluate the expression of PRPS1 in Chinese children with B-ALL and to investigate the mechanism of action of PRPS1 in this disease. A Cell Counting Kit-8 (CCK-8) assay was performed to examine the proliferation of B-ALL Sup-B15 and Raji cells, and flow cytometric analysis was conducted to determine the cell cycle distribution and rate of apoptosis. The mRNA and protein expression levels of PRPS1, MYC proto-oncogene, bHLH transcription factor, cyclin E1, B-cell lymphoma-2 (Bcl-2), cyclin dependent kinase 2 and caspase-3 were detected by reverse transcription-quantitative polymerase chain reaction and western blot analysis, respectively. Elevated PRPS1 expression was associated with a high-risk stratification and poor prognosis in patients with B-ALL. Furthermore, overexpression of PRPS1 accelerated the growth of and inhibited apoptosis in Sup-B15 and Raji cells as well as increasing the expression of Bcl-2 to induce an anti-apoptotic effect in B-ALL cell lines. Tricholoma matsutake, one of widely accepted functional mushrooms, possesses various pharmacological activities, and its antitumor effect has become an important research point. Our study aims to evaluate the cytotoxicity activities of T. matsutake aqueous extract (TM) in HepG2 and SMMC-7721 cells. In in vitro experiments, TM strikingly reduced cell viability, promoted cell apoptosis, inhibited cell migration ability, induced excessive generation of ROS, and caused caspases cascade and mitochondrial membrane potential dissipation in hepatocellular carcinoma cells. In in vivo experiments, 14-day TM treatment strongly suppressed tumor growth in HepG2 and SMMC-7721-xenografted nude mice without influence on their body weights and liver function. Furthermore, TM increased the levels of cleaved poly-ADP-ribose polymerase (PARP), Bad, and Bax and reduced the expressions of B-cell lymphoma 2 (Bcl-2) in treated cells and tumor tissues. Tricholoma matsutake has been popular as food and biopharmaceutical materials in Asian countries for its various pharmacological activities. The present study aims to analyze the antifatigue effects on enhancing exercise performance of Tricholoma matsutake fruit body (ABM) and liquid cultured mycelia (TM) in mouse model. Two-week Tricholoma matsutake treatment significantly enhances the exercise performance in weight-loaded swimming, rotating rod, and forced running test. In TM- and ABM-treated mice, some factors were observed at 60 min after swimming compared with nontreated mice, such as the increased levels of adenosine triphosphate (ATP), antioxidative enzymes, and glycogen and the reduced levels of malondialdehyde and reactive oxygen species in muscle, liver, and/or serum. Further data obtained from western blot show that CM and ABM have strongly enhanced the activation of 5′-AMP-activated protein kinase (AMPK), and the expressions of peroxisome proliferator have activated receptor γ coactivator-1α (PGC-1α) and phosphofructokinase-1 (PFK-1) in liver. Our data suggest that both Tricholoma matsutake fruit body and liquid cultured mycelia possess antifatigue effects related to AMPK-linked antioxidative pathway. The information uncovered in our study may serve as a valuable resource for further identification and provide experimental evidence for clinical trials of Tricholoma matsutake as an effective agent against fatigue related diseases. Nonplanar In0.53Ga0.47As gate wrap around field-effect transistors (GWAFETs) with atomic-layer deposited high-k dielectric and metal gate have been demonstrated in this paper. By applying novel device structure and optimizing fabrication process, In0.53Ga0.47As GWAFETs exhibit significant performance improvements over planar MOSFETs on both current drive capability and electrostatics control. In0.53Ga0.47As GWAFETs with fin width (W \(_{\mathrm {{fin}}}\) ) 40–200 nm have been fabricated. Devices with narrower W \(_{\mathrm {{fin}}}\) exhibit higher drive current, transconductance, and better short channel effect control, which demonstrates the scalability of nonplanar In0.53Ga0.47As GWAFETs. Notable improvements in the \HfO2/\In0.53 \Ga0.47\As gate stack have been achieved by a post-\HfO2 fluorine plasma treatment, including excellent interface quality of low equivalent oxide thickness \HfO2 (1.4 nm) directly on \In0.53\Ga0.47\As without using an interface passivation layer, \sim\!\!\5\times reduction in interface trap density from \2.8 \times \10^12 to \4.9 \times \10^11\ \cm^-2\eV^-1, \sim\!\!\10\times reduced border traps from \1.6 \times \10^19 to \1.6 \times \10^18\ \cm^-3, and \sim40% less charge-trapping centers. As a result, improved electrical performances have been obtained. Frequency dispersion in capacitance–voltage characteristics has been reduced. In0.53Ga0.47As and In0.7Ga0.3As surface channel metal-oxide-semiconductor field-effect transistors (MOSFETs) with atomic layer deposition of Al2O3 and ZrO2 have been fabricated and investigated. The device performance of InGaAs channel MOSFETs with various indium compositions (53% and 70%) has been studied, including drain current, transconductance, subthreshold swing, and interface trap density (Dit). It has been found that In0.53Ga0.47As MOSFETs exhibit higher transconductance and lower subthreshold swing than In0.7Ga0.3As MOSFETs for both Al2O3 and ZrO2 gate dielectric (e.g., 125 mV/decade versus 160 mV/decade for Al2O3). The interface trap density measured using conductance method (1.1×1012 eV-1 cm-2 versus 3.2×1012 eV-1 cm-2 for Al2O3) suggests a better interface at high-k dielectric/In0.53Ga0.47As channel. Capacitance-voltage measurement at 77 K also correlates with the above results, showing smaller frequency dispersion for In0.53Ga0.47As devices. [ABSTRACT FROM AUTHOR]

MicroRNAs (miRNAs/miRs) are known to be involved in various human cancer types. Ultrasound-targeted microbubble destruction (UTMD) may improve the transfection efficiency of exogenous genes into target tissues and organs, thereby improving cancer treatment. In the present study, the role of miR-4284 in non-small cell lung cancer (NSCLC) was investigated and the effect of UTMD-mediated inhibition of miR-4284 on tumor progression was further analyzed. The expression of miR-4284 in NSCLC cells and tissues was detected by reverse transcription-quantitative PCR. UTMD-mediated inhibition of miR-4284 was achieved by co-transfection of microvesicles and miR-4284 inhibitors into NSCLC cells. A Cell Counting Kit-8 assay was used to determine NSCLC cell proliferation, and the migration and invasion of NSCLC cells were examined by Transwell assays. Compared with that in the control group, the expression of miR-4284 was increased in NSCLC tissues and cells. Knockdown of miR-4284 in NSCLC cells inhibited cell proliferation, migration and invasion. UTMD improved the transfection efficiency of miR-4284 inhibitors in NSCLC cells, resulting in more significant inhibition of tumor cell proliferation, migration and invasion. In conclusion, the results indicated that the expression of miR-4284 was increased in clinical samples and cell lines of NSCLC and that knockdown of miR-4284 inhibited the proliferation, migration and invasion of tumor cells. Aegilops geniculata Roth has been used as a donor of disease-resistance genes, to enrich the gene pool for wheat (Triticum aestivum) improvement through distant hybridization. In this study, the wheat–Ae. geniculata alien disomic substitution line W16998 was obtained from the BC1F8 progeny of a cross between the common wheat 'Chinese Spring' (CS) and Ae. geniculata Roth (serial number: SY159//CS). This line was identified using cytogenetic techniques, analysis of genomic in situ hybridization (GISH), functional molecular markers (Expressed sequence tag-sequence-tagged site (EST–STS) and PCR-based landmark unique gene (PLUG), fluorescence in situ hybridization (FISH), sequential fluorescence in situ hybridization–genomic in situ hybridization (sequential FISH–GISH), and assessment of agronomic traits and powdery mildew resistance. During the anaphase of meiosis, these were evenly distributed on both sides of the equatorial plate, and they exhibited high cytological stability during the meiotic metaphase and anaphase. GISH analysis indicated that W16998 contained a pair of Ae. geniculata alien chromosomes and 40 common wheat chromosomes. One EST–STS marker and seven PLUG marker results showed that the introduced chromosomes of Ae. geniculata belonged to homoeologous group 7. Nullisomic–tetrasomic analyses suggested that the common wheat chromosome, 7A, was absent in W16998. FISH and sequential FISH–GISH analyses confirmed that the introduced Ae. geniculata chromosome was 7Mg. Therefore, W16998 was a wheat–Ae. geniculata 7Mg (7A) alien disomic substitution line. Inoculation of isolate E09 (Blumeria graminis f. sp. tritici) in the seedling stage showed that SY159 and W16998 were resistant to powdery mildew, indeed nearly immune, whereas CS was highly susceptible. Compared to CS, W16998 exhibited increased grain weight and more spikelets, and a greater number of superior agronomic traits. Consequently, W16998 was potentially useful. Using desirability function, four indexes including mycelium dry weight, intracellular polysaccharide, adenosine and mannitol yield were uniformed into one expected value ( Da ) which further served as the assessment criteria. In our present study, Plackett–Burman design was applied to evaluate the effects of eight variables including initial pH, rotating speed, culture temperature, inoculum size, ventilation volume, culture time, inoculum age and loading volume on Da value during Marasmius androsaceus submerged fermentation via a five-liter fermentor. Culture time, initial pH and rotating speed were found to influence Da value significantly and were further optimized by Box–Behnken design. Results obtained from Box–Behnken design were analyzed by both response surface regression (Design-Expert.V8.0.6.1 software) and artificial neural network combining the genetic algorithm method (Matlab2012a software). After comparison, the optimum M. androsaceus submerged fermentation conditions via a five-liter fermentor were obtained as follows: initial pH of 6.14, rotating speed of 289.3 rpm, culture time of 6.285 days, culture temperature of 26 °C, inoculum size of 5%, ventilation volume of 200 L/h, inoculum age of 4 days, and loading volume of 3.5 L/5 L. The predicted Da value of the optimum model was 0.4884 and the average experimental Da value was 0.4760. The model possesses well fitness and predictive ability. [ABSTRACT FROM AUTHOR]

Gastrointestinal dysfunction (GD) is a common complication after endotoxemia, which can further aggravate the progress of infection. Acupuncture uses metal needles of different shapes and techniques to stimulate specific points on the human body, which are effective in treating various diseases, including gastrointestinal diseases. We aimed at exploring the clinical effect of acupuncture on the recovery of visceral sensation, proximal gastric compliance, and autonomic nervous function in patients with septic GD. A total of 73 sepsis with GD patients were selected using modified single section ultrasonography combined with clinical symptoms in the First People's Hospital of Lanzhou City during 2019. The participants were randomly allocated to routine-treatment (control group) and study group receiving acupuncture. The indexes before and after treatment included gastric residue, gastric dilatation, pressure and volume, gastric compliance, autonomic nerve function, APACHE II score, and infection index were measured and compared. Before treatment, there was no significant difference in the basic information of the two groups, including gastric volume and pressure, gastric residue, gastric compliance, autonomic nerve function, and APACHE II score. After treatment, the maximum gastric volume and pressure, gastric residue, and APACHE II score of the two groups were significantly improved (P < 0.05). In addition, the maximum gastric volume and pressure of the study group were significantly higher, while gastric residual, autonomic nerve function, and APACHE II were significantly lower than those of the control group (P < 0.05). However, our results showed that acupuncture did not further reduce inflammatory markers, including white blood cells, C-reactive protein, and procalcitonin. To sum up, on the basis of basic treatment, the application of acupuncture can further improve the clinical symptoms of GD in patients with sepsis, enhance gastrointestinal motility, and improve autonomic nervous function, which is worthy of clinical application and promotion. Background: Splicing factors are frequently mutated in patients with myelodysplastic syndromes and acute myeloid leukaemia. Recent studies have revealed convergent molecular defects caused by splicing factor mutations, among which R-loop dysregulation and resultant genome instability are suggested as contributing factors to disease progression. On the other hand, understanding how mutant cells survive upon aberrant R-loop formation and genome instability is essential for developing novel therapeutics., Methods: The immunoprecipitation was performed to identify R-loops in association with PARP1/poly-ADP-ribosylation. The western blot, immunofluorescence, and flow cytometry assays were used to test the cell viability, cell cycle arrest, apoptosis, and ATM activation in mutant cells following the treatment of the PARP inhibitor. The Srsf2(P95H) knock-in murine hematopoietic cells and MLL-AF9 transformed leukaemia model were generated to investigate the potential of the PARP inhibitor as a therapy for haematological malignancies., Results: The disease-causing mutations in SRSF2 activate PARP and elevate the overall poly-ADP-ribosylation levels of proteins in response to R-loop dysregulation. In accordance, mutant cells are more vulnerable to the PARP inhibitors in comparison to the wild-type counterpart. Objective: Given high risks of major bleeding during retroperitoneal sarcoma(RPS) surgeries, severe complications and deaths are common to see perioperatively. Thus, effective anesthetic management is the key point to ensuring the safety of patients. This study aimed to introduce anesthesia management and mortalities in RPS patients receiving massive blood transfusions during surgeries., Methods: Records of RPS surgeries under general anesthesia from January 2016 through December 2021 were retrospectively retrieved from our database. Patients who received massive blood transfusions (MBT) exceeding 20 units in 24h duration of operations were finally included in this study. Demographics, modalities of anesthesia management, blood loss, transfusion, peri-anesthesia biochemical tests as well as morbidities and mortalities were collected. Risk factors of postoperative 60d mortality were determined through logistic regression in uni-and multi-variety analysis using the statistics software STATA 17.0., Results: A total of 70 patients (male 31) were included. The mean age was 50.1 ± 15.8 years. All patients received combined resections of sarcoma with involved organs under general anesthesia. Mean operation time and anesthesia time were 491.7 ± 131.1mins and 553.9 ± 132.6mins, respectively. The median intraoperative blood loss was 7000ml (IQR 5500,10000ml). Median red blood cells (RBC) and fresh frozen plasma (FFP) transfusion were 25.3u (IQR 20,28u), and 2400ml (IQR 2000,3000ml), respectively. Other blood products infusions included prothrombin complex concentrate (PCCs), fibrinogen concentrate (FC), platelet(plt) and albumin(alb) in 82.9% (58/70), 88.6% (62/70), 81.4% (57/70) and 12.9% (9/70) of patients. The postoperative severe complication rate(Clavien-Dindo grade≥3a) was 35.7%(25/70). A total of 7 patients (10%) died during the postoperative 60-day period. BMI, volumes of crystalloid infusion in anesthesia, and hemoglobin and lactate levels at the termination of operation were found significantly associated with postoperative occurrence of death in univariate analysis. In logistic multivariate analysis, extended anesthesia duration was found associated with postoperative venous thrombosis embolism (VTE) and severe complication. The lactate level at the immediate termination of the operation was the only risk factor related to perioperative death (p<0.05)., Conclusion: RPS patients who endure MBT in surgeries face higher risks of death postoperatively, which needs precise and effective anesthesia management in high-volume RPS centers. The AlGaN-based deep ultraviolet light-emitting diode (DUV LED) has advantages of environmentally friendly materials, tunable emission wavelength, and easy miniaturization. However, an increase in Al composition leads to a decline in the lattice quality, thereby reducing the internal quantum efficiency (IQE). In addition, the light extraction efficiency (LEE) is limited due to the strong transverse magnetization polarization emission from the multiple quantum wells. Here, we designed the topological corner structure in AlGaN-MQWs, and the high electric field intensity in a tiny space at the corner results in an extremely high local density of optical states (LDOS), which could shorten the luminescence decay time of the emitter and increase the radiative rate by 26 times. Meanwhile, because the excited topological corner state resonance mode is a transverse-electric mode, enhancing only the transverse-electric luminescence without any gain for transverse-magnetic luminescence, thereby significantly improving the light extraction efficiency. Finally, according to theoretical calculations, the IQE could reach 68.75% at room temperature.

Accumulating evidence suggests that in various systems, not all bidirectional microbiota-host interactions involve direct cell contact. Bacterial extracellular vesicles (BEVs) may be key participants in this interkingdom crosstalk. BEVs mediate microbiota functions by delivering effector molecules that modulate host signaling pathways, thereby facilitating host-microbe interactions. BEV production during infections by both pathogens and probiotics has been observed in various host tissues. Therefore, these vesicles released by microbiota may have the ability to drive or inhibit disease pathogenesis in different systems within the host. In this paper, Lepista sordida polysaccharides (LSP) were separated from Lepista sordida ( L. sordida ) mainly using the Ultrasonic-Micro Wave Synergy Extraction (UMSE) method and purified by graded alcohol precipitation. Three polysaccharide components: 40%-LSP-UMSE, 60%-LSP-UMSE, and 80%-LSP-UMSE were obtained and further analyzed the physicochemical properties, structural characteristics, and antioxidant activity. And the effects on the proliferation of Lactobacillus casei of three polysaccharide components were studied. The characteristic absorption peaks and the β-glycosidic bond of three polysaccharide components were the direct expression at UV 200 nm using UV and FT-IR spectroscopy. The three polysaccharide components were mainly composed of glucose, mannose, galactose, and ribose using high-performance liquid chromatography (HPLC) analysis. The antioxidant activity study revealed that the polysaccharides obtained by the UMSE method had better antioxidant activity compared to the traditional "Hot Water Extraction (HWE)" method. In addition, the polysaccharide components promoted the proliferation of L. casei to some extent. 40%-LSP-UMSE, 80%-LSP-UMSE as the carbon source had better acid production than the control inulin. Three LSP-UMSE used as a carbon source compared with glucose for culturing L. casei could significantly improve its tolerance to bile salts. Wheat (Triticum aestivum) is a commercially important crop and its production is seriously threatened by the fungal pathogen Puccinia striiformis f. sp. tritici West (Pst). Resistance (R) genes are critical factors that facilitate plant immune responses. Here, we report a wheat R gene NB-ARC-LRR ortholog, TaYRG1, that is associated with distinct alternative splicing events in wheat infected by Pst. The native splice variant, TaYRG1.6, encodes internal-motif-deleted polypeptides with the same N- and C-termini as TaYRG1.1, resulting in gain of function. Transient expression of protein variants in Nicotiana benthamiana showed that the NB and ARC domains, and TaYRG1.6 (half LRR domain), stimulate robust elicitor-independent cell death based on a signal peptide, although the activity was negatively modulated by the CC and complete LRR domains. Furthermore, molecular genetic analyses indicated that TaYRG1.6 enhanced resistance to Pst in wheat. Moreover, we provide multiple lines of evidence that TaYRG1.6 interacts with a dynamin-related protein, TaDrp1. Proteome profiling suggested that the TaYRG1.6-TaDrp1-DNM complex in the membrane trafficking systems may trigger cell death by mobilizing lipid and kinase signaling in the endocytosis pathway. Zhenqi Fuzheng formula (ZQFZ), of which the main ingredients are Astragalus membranaceus and Ligustrum lucidum , has immune system regulatory functions and potential anti-tumor bioactivity. The inhibition of colorectal tumor growth by ZQFZ was analyzed in inflammatory cells and B6/JGpt- Apc em1Cin(MinC) /Gpt ( Apc Min/+ ) mice. ZQFZ exhibited anti-inflammatory activity by decreasing the phosphorylation of nuclear factor-kappa B (NF-κB) pathway-related proteins in lipopolysaccharide-induced RAW264.7 cells. After 56 days of treatment, ZQFZ alleviated the progression of colorectal cancer (CRC) and increased the body weight and thymic index values of the Apc Min/+ mice. An analysis of the intestinal microflora showed that ZQFZ affected the abundance of certain immune-related bacteria, which may explain its immunomodulatory effects. Moreover, the percentages of T cells and NK cells in peripheral blood were significantly increased and 15 immune-related cytokines were regulated in serum or the colon or both. ZQFZ upregulated the levels of CD4 and CD8 in the spleen and colorectal tumors and decreased the expression levels of cytotoxic T-lymphocyte-associated protein 4 and programmed death-ligand 1 in colorectal tumors. ZQFZ promoted an anti-tumor immune response and inhibited the occurrence and development of CRC by regulating the immune system. This study provides the experimental basis for the application of ZQFZ as a therapeutic agent for CRC.

Thinopyrum ponticum (2 n = 10 x = 70) is a wild relative of wheat with high tolerance to both biotic and abiotic stresses; it has been wildly used in wheat genetic improvement. A disomic substitution line named SN19647 was derived from a cross between Triticum aestivum and the wheat- Th. ponticum partial amphiploid SNTE20 (2 n = 8 x = 56). It was evaluated for disease resistance and characterized via sequential fluorescence in situ hybridization (FISH)-genomic in situ hybridization (GISH) and molecular markers. The results showed that SN19647 carried resistance to both powdery mildew and leaf rust. It contained 42 chromosomes with a pair of wheat chromosome 1B replaced by a pair of J S chromosomes from Th. ponticum . In addition to chromosomal substitution events, structural variation also occurred on wheat chromosomes 2A, 5A, 6B, and 7B. Based on marker analysis, 19 markers specific to the J S chromosome were obtained, of which seventeen markers belonged to homoeologous group one. These results indicated that SN19647 was a 1J S (1B) substitution line. Compared with the known 1J S (1D) substitution line CH10A5, it was found that 17 markers generated different specific bands to Th. ponticum , confirming the novelty of the 1J S chromosome in SN19647. Background: Long noncoding RNAs have been demonstrated to play important roles in different kinds of human malignancy. The purpose of this study was to evaluate the diagnostic and prognostic value of long intergenic non-protein coding RNA 1232 (LINC01232) in patients with pancreatic adenocarcinoma (PAAD) and further explore the clinical significance of the potential miRNAs that might be sponged by LINC01232., Methods: The potential target miRNAs that might be sponged by LINC01232 were analyzed using bioinformatics analysis. The Real-Time quantitative PCR was adopted to measure the relative expression of LINC01232 and target miRNAs in PAAD serum and tissue samples. The diagnostic and prognostic value of LINC01232 was evaluated using the receiver operating characteristic analysis and Kaplan-Meier survival analysis, respectively., Results: LINC01232 expression was upregulated in PAAD serum and tissues and associated with patients' TNM stage. Serum LINC01232 expression had diagnostic value, and the high levels of LINC01232 could predict unfavorable prognosis in PAAD patients. miR-204-5p, miR-370-5p and miR-654-3p were proposed as 3 targets of LINC01232 in PAAD, and their decreased expression levels in PAAD patients showed certain clinical significance in diagnosis and prognosis., Conclusion: The data of this study revealed that LINC01232 expression is upregulated in PAAD serum and tissue samples with considerable diagnostic and prognostic significance. In addition, miR-204-5p, miR-370-5p and miR-654-3p may be sponged by LINC01232 in PAAD, which also show potencies in PAAD diagnosis and prognosis.

Thinopyrum ponticum (2 n = 10 x = 70), a member of the tertiary gene pool of wheat ( Triticum aestivum L.), harbors many biotic and abiotic stress resistance genes. CH10A5, a novel disomic substitution line from a cross of T. aestivum cv. 7182 and Th. ponticum , was characterized by cytogenetic identification, in situ hybridization, molecular marker analysis, and morphological investigation of agronomic traits and disease resistance. Cytological observations showed that CH10A5 contained 42 chromosomes and formed 21 bivalents at meiotic metaphase I. Genome in situ hybridization (GISH) analysis indicated that two of its chromosomes came from the J s genome of Th. ponticum , and wheat 15K array mapping and fluorescence in situ hybridization (FISH) revealed that chromosome 1D was absent from CH10A5. Polymorphic analysis of molecular markers indicated that the pair of alien chromosomes belonged to homoeologous group one, designated as 1J s . Thus, CH10A5 was a wheat- Th. ponticum 1J s (1D) disomic substitution line. Field disease resistance trials demonstrated that the introduced Th. ponticum chromosome 1J s was probably responsible for resistance to both stripe rust and powdery mildew at the adult stage. Based on specific-locus amplified fragment sequencing (SLAF-seq), 507 STS molecular markers were developed to distinguish chromosome 1J s genetic material from that of wheat. Of these, 49 STS markers could be used to specifically identify the genetic material of Th. ponticum .
