Metacaspases (MCs) are cysteine proteases that are important for programmed cell death (PCD) in plants. In this study, we identified 89 MC genes in the genomes of four Gossypium species (Gossypium raimondii, Gossypium barbadense, Gossypium hirsutum, and Gossypium arboreum), and classified them as type-I or type-II genes. All of the type-I and type-II MC genes contain a sequence encoding the peptidase C14 domain. During developmentally regulated PCD, type-II MC genes may play an important role related to fiber elongation, while type-I genes may affect the thickening of the secondary wall. Additionally, 13 genes were observed to be differentially expressed between two cotton lines with differing fiber strengths, and four genes (GhMC02, GhMC04, GhMC07, and GhMC08) were predominantly expressed in cotton fibers at 5−30 days post-anthesis (DPA). During environmentally induced PCD, the expression levels of four genes were affected in the root, stem, and leaf tissues within 6 h of an abiotic stress treatment. In general, the MC gene family affects the development of cotton fibers, including fiber elongation and fiber thickening while four prominent fiber- expressed genes were identified. The effects of the abiotic stress and hormone treatments imply that the cotton MC gene family may be important for fiber development. The data presented herein may form the foundation for future investigations of the MC gene family in Gossypium species.

Microtubules (MTs) are of importance to fiber development. The Xklp2 (TPX2) proteins as a class of microtubule-associated proteins (MAPs) play a key role in plant growth and development by regulating the dynamic changes of microtubules (MTs). However, the mechanism underlying this is unknown. The interactions between TPX2 proteins and tubulin protein, which are the main structural components, have not been studied in fiber development of upland cotton. Therefore, a genome-wide analysis of the TPX2 family was firstly performed in Gossypium hirsutum L. This study identified 41 GhTPX2 sequences in the assembled G. hirsutum genome by a series of bioinformatic methods. Generally, this gene family is phylogenetically grouped into six subfamilies, and 41 G. hirsutum TPX2 genes (GhTPX2s) are distributed across 21 chromosomes. A heatmap of the TPX2 gene family showed that homologous GhTPX2 genes, GhWDLA2/7 and GhWDLA4/9, have large differences in expression levels between two upland cotton recombinant inbred lines (69307 and 69362) that are different in fiber quality at 15 and 20 days post anthesis. The relative data indicate that these four genes are down-regulated under oryzalin, which causes microtubule depolymerization, as determined via qRT-PCR. A subcellular localization experiment suggested that GhWDLA2 and GhWDLA7 are localized to the microtubule cytoskeleton, and GhWDLA4 and GhWDLA9 are only localized to the nucleus. However, only GhWDLA7 between GhWDLA2 and GhWDLA7 interacted with GhTUA2 in the yeast two-hybrid assay. These results lay the foundation for further function study of the TPX2 gene family.

In this study, we synthetized a series of 5-aryl-4,5-dihydrotetrazolo[1,5-a]thieno[2,3-e]pyridine derivatives containing tetrazole and other heterocycle substituents, i.e., triazole, methyltriazole, and triazolone. The forced swim test (FST) and tail suspension test (TST) were used to evaluate the antidepressant activity of the target compounds. The compound 5-[4-(trifluoromethyl)phenyl]-4,5-dihydrotetrazolo[1,5-a]thieno[2,3-e]pyridine (4i) showed the highest antidepressant activity, with a reduced immobility time of 55.33% when compared with the control group. Using an open-field test, compound 4i was shown to not affect spontaneous activity of mice. The determination of in vivo 5-hydroxytryptamine (5-HT) concentration showed that compound 4i may have an effect in the mouse brain. The biological activities of all synthetized compounds were verified by molecular docking studies. Compound 4i showed significant interactions with residues of the 5-HT1A receptor homology model.

Upland cotton (Gossypium hirsutum) is grown for its elite fiber. Understanding differential gene expression patterns during fiber development will help to identify genes associated with fiber quality. In this study, we used two recombinant inbred lines (RILs) differing in fiber quality derived from an intra-hirsutum population to explore expression profiling differences and identify genes associated with high-quality fiber or specific fiber-development stages using RNA sequencing. Overall, 72/27, 1137/1584, 437/393, 1019/184, and 2555/1479 differentially expressed genes were up-/down-regulated in an elite fiber line (L1) relative to a poor-quality fiber line (L2) at 10, 15, 20, 25, and 30 days post-anthesis, respectively. Three-hundred sixty-three differentially expressed genes (DEGs) between two lines were colocalized in fiber strength (FS) quantitative trait loci (QTL). Short Time-series Expression Miner (STEM) analysis discriminated seven expression profiles; gene ontology (GO) and Kyoto Encyclopedia of Genes and Genomes (KEGG) annotation were performed to identify difference in function between genes unique to L1 and L2. Co-expression network analysis detected five modules highly associated with specific fiber-development stages, especially for high-quality fiber tissues. The hub genes in each module were identified by weighted gene co-expression network analysis. Hub genes encoding actin 1, Rho GTPase-activating protein with PAK-box, TPX2 protein, bHLH transcription factor, and leucine-rich repeat receptor-like protein kinase were identified. Correlation networks revealed considerable interaction among the hub genes, transcription factors, and other genes.

Carbon dots (CDs) can be widely used in the field of sensing because of its good water solubility, low toxicity, high fluorescence stability and excellent biocompatibility. It has become a popular trend to prepare high-value, inexpensive, renewable and environmentally friendly CDs sensors from biomass resources. This article reviewed the research progress of biomass-derived CDs as chemical, physical and biological sensors in recent years and studied their preparation processes and sensing abilities. Furthermore, the prospects and challenges of biomass-CDs sensors were discussed. Handwritten Text Recognition has achieved an impressive performance in public benchmarks. However, due to the high inter- and intra-class variability between handwriting styles, such recognizers need to be trained using huge volumes of manually labeled training data. To alleviate this labor-consuming problem, synthetic data produced with TrueType fonts has been often used in the training loop to gain volume and augment the handwriting style variability. However, there is a significant style bias between synthetic and real data which hinders the improvement of recognition performance. To deal with such limitations, we propose a generative method for handwritten text-line images, which is conditioned on both visual appearance and textual content. Our method is able to produce long text-line samples with diverse handwriting styles. Once properly trained, our method can also be adapted to new target data by only accessing unlabeled text-line images to mimic handwritten styles and produce images with any textual content. Extensive experiments have been done on making use of the generated samples to boost Handwritten Text Recognition performance. Purpose: Pancreatic cancer is a malignant tumor with a high degree of malignancy, strong heterogeneity, and high lethality. Trop2 is a transmembrane glycoprotein associated with the occurrence, development, and poor prognosis of pancreatic cancer. This study aims to develop 64Cu/177Lu-labeled anti-Trop2 monoclonal antibody (hIMB1636) for positron emission tomography (PET) imaging and radioimmunotherapy (RIT) application in pancreatic cancer tumor models. Methods: The binding kinetics of hIMB1636 to Trop2 antigen was measured by Biolayer interferometry (BLI). Western blotting was used to screen the Trop2 expression of pancreatic cancer cell lines. Flow cytometry and cell immunofluorescence were used to evaluate the binding ability of hIMB1636 and Trop2 on the cell surface. hIMB1636 were conjugated with p-SCN-Bn-NOTA (NOTA) and DOTA-NHS-ester (DOTA) for 64Cu and 177Lu radiolabeling respectively. ImmunoPET imaging and RIT studies were performed using 64Cu-NOTA-hIMB1636 and 177Lu-DOTA-hIMB1636 in subcutaneous pancreatic cancer tumor models. Results: hIMB1636 had a strong binding affinity to Trop2 according to the results of BLI. The T3M-4 cell line showed the strongest expression of Trop2 and specific binding ability of hIMB1636 according to the results of Western blotting, flow cytometry, and cell immunofluorescence. The radiochemical purity of 64Cu-NOTA-hIMB1636 and 177Lu-DOTA-hIMB1636 exceeded 95%. PET imaging showed gradually an accumulation of 64Cu-NOTA-hIMB1636 in T3M-4 tumor models. The maximum tumor uptake was 8.95 ± 1.07%ID/g (n = 4) at 48 h post injection (p.i.), which had significant differences with T3M-4-blocked and PaTu8988-negative groups (P < 0.001). The high-177Lu-hIMB1636 group demonstrated the strongest tumor suppression with standardized tumor volume about 94.24 ± 14.62% (n = 5) at 14 days p.i., significantly smaller than other groups (P < 0.05). Ex vivo biodistribution and histological staining verified the in vivo PET imaging and RIT results. Conclusions: This study demonstrated that 64Cu/177Lu-labeled hIMB1636 could noninvasively evaluate the expression level of Trop2 and inhibit the Trop2-overexpressed tumor growth in pancreatic cancer tumor models. Aiming at the calculation problem of bearing pressure of typical overlying strata structure, in this paper, mechanical calculation, field monitoring and other methods are used to study the influence range of abutment pressure. Recently, many major rock burst accidents in working surface show that with the increase of mining depth and the change of overlying strata structure, the abutment pressure distribution has become one of the problems of safe mining. The influence range, distribution of high stress area and peak position of working surface support pressure need to be solved urgently; and through theoretical analysis, this paper reveals (whole rock single-layer structure, whole rock layered structure, whole rock single-layer structure—without arch in soil layer, whole rock layered structure—without arch in soil layer, whole rock single-layer structure—with soil arching, whole rock layered structure—with soil arching) the stress transfer mode, transfer size, and arching conditions of overlying strata in different areas. Mesoporous carbon nanotubes (mCNTs) hold great promise interests, owing to their superior nano-platform properties for biomedicine. To fully utilize this potential, the toxicity and biodistribution of pristine and surface-modified mCNTs (-OH/-COOH) should preferentially be addressed. The results of cell viability suggested that pristine mCNTs induced cell death in a concentration-dependent manner. As evidence of reactive oxygen species (ROS), malondialdehyde (MDA) and superoxide dismutase (SOD), pristine mCNTs induced noticeable redox imbalance. 99mTc tracing data suggested that the cellular uptake of pristine mCNTs posed a concentrate-dependent and energy-dependent manner via macropinocytotic and clathrin-dependent pathways, and the main accumulated organs were lung, liver and spleen. With OH modification, the ROS generation, MDA deposition and SOD consumption were evidently reduced compared with the pristine mCNTs at 24/48 h high-dose exposure. With COOH modification, the modified mCNTs only showed a significant difference in SOD consumption at 24/48 h exposure, but there was no significant difference in the measurement of ROS and MDA. The internalization mechanism and organ distribution of modified mCNTs were basically invariant. Together, our study provides evidence that mCNTs and the modified mCNTs all could induce oxidative damage and thereby impair cells. 99mTc-mCNTs can effectively trace the distribution of nanotubes in vivo. Brassica napus inap cytoplasmic male sterility (CMS) is a novel sterile line with potential application in rapeseed hybrid breeding. Sterile cytoplasm was obtained from Isatis indigotica through somatic fusion and then recurrent backcrossing with B. napus. Previous studies have shown that inap CMS abortion occurred before the stamen primordia (stage 4–5), but the genetic mechanism of sterility needs to be studied. RNA-seq analyses were performed on the floral buds at two stages (0–5 and 6–8), before and after the formation of stamen primordium. As a result, a total of 1769 and 594 differentially expressed genes (DEGs) were detected in the CMS line compared to its maintainer line at the two stages, respectively. In accordance with the CMS phenotype, the up- and downstream regulators of the stamen identity genes AP3 and PI were up- and downregulated in the CMS line, respectively. Furthermore, isobaric tags for relative and absolute quantitation (iTRAQ) analysis showed that a total of 760 differentially abundant proteins (DAPs) were identified in flower buds at stages 0–8, and most of the proteins related to the anther development, oxidative phosphorylation, and programmed cell death (PCD) were downregulated in inap CMS. In combined transcriptomic and proteomic analysis, a total of 32 DEGs/DAPs were identified, of which 7 common DEGs/DAPs had the same expression trend at stage 0–8 of flower development. The downregulation of genes related to the energy deficiency, hormone signal transduction, and the maintenance of mitochondrial metabolic homeostasis at stage 0–5 might disturb the normal differentiation of stamen primordium, resulting in carpelloid stamen of inap CMS. We report a photoconductor-based terahertz metadevice whose transmission characteristics can be comprehensively controlled by a combination of an optical excitation and electrical bias. A metasurface with interdigitated electrodes is proposed to simultaneously support a terahertz resonance and the photoconductive effect, allowing efficient local and global dual-tuning of the carriers in the semiconductor and thus the terahertz wave, based on the simultaneous external optical and electrical stimuli. Experimental results reveal the opposite tuning trend of optical and electrical excitations, leading to the comprehensive transmission modulation of terahertz wave. In this work, the influence of temperature on the corrosion behavior of AlCoCrFeNi2.1 eutectic high-entropy alloy in a chloride-containing sulfuric acid solution was investigated using electrochemical measurement, X-ray photoelectron spectroscopy, and scanning electron microscopy. Results show that the passive film of AlCoCrFeNi2.1 is stable in chloride-containing sulfuric acid solutions at low temperatures, while an unstable film forms on the alloy at high temperatures. Furthermore, temperature changes the proportion of hydroxide and oxide in Fe and Cr, but it has no noticeable effect on Al and Ni, which is a significant factor on the passive behavior. L12 phase exhibits good corrosion resistance at different temperatures. Pitting occurred on B2 phase in the chloride-containing sulfuric acid solution at a low temperature of 5 °C, while pitting and dissolution take place on AlCoCrFeNi2.1 in the acid solution at room temperature and above. The flexural ultrasonic transducer has traditionally been limited to proximity measurement applications, such as car-parking systems and industrial metrology. Principally, their classical form is unsuitable for environments above atmospheric 1 bar pressure, due to an internal air cavity which creates a pressure imbalance across the transducer’s vibrating membrane. This imbalance leads to physical deformation and degradation of the transducer’s structure, restricting the membrane’s capacity to vibrate at resonance to transmit and receive ultrasound. There is a requirement for ultrasonic sensors which can withstand environments of elevated pressure, for example in ultrasonic gas metering. Recent research demonstrated the dynamic performance of flexural ultrasonic transducers with vented structures, allowing the pressure to balance across the transducer membrane. However, a hermetically sealed transducer is a more practical and robust solution, where the internal components of the transducer, such as the piezoelectric ceramic disc, will be protected from harmful environmental fluids. In this research, the design and fabrication of a new form of flexural ultrasonic transducer for environments of elevated pressure is demonstrated, where the internal air cavity is filled with an incompressible fluid in the form of a non-volatile oil. Dynamic performance is measured through acoustic microphone measurements, electrical impedance analysis, and pulse-echo ultrasound measurement. Together with finite element analysis, stable ultrasound measurement is achieved above 200 bar in air, opening the possibility for reliable ultrasound measurement in hostile environments of elevated pressure. The resonant nature and geometric scalability make metamaterials an ideal platform for an enhanced light–matter interaction over a broad frequency range. The mid-infrared (IR) spectral range is of great importance for vibrational spectroscopy of molecules, while IR metamaterials created from lithography-based planar nanostructures have been used to demonstrate enhanced molecular detection. Compared with visible and near-infrared, the relative long wavelengths of IR light make it possible to achieve three-dimensional (3D) IR metamaterials via the state-of-the-art 3D fabrication techniques. Here, we design and fabricate a 3D printed plasmonic metamaterial absorber (MMA), and by performing Fourier-transform IR spectroscopy, we demonstrate that a series of molecular fingerprint vibrations of glycine can be significantly enhanced by the high absorption mode supported by the 3D meta-atoms of the MMA. The observed enhanced IR detection can also be partially attributed to the improved accessibility offered by the 3D architecture of the MMA. In particular, due to capillary forces during the drying process, the microscale 3D printed features lead to selective analyte deposition in high-field regions, which provides another degree of freedom in the design of the 3D printed structures for surface-enhanced IR detection. Our study shows the flexibility of metastructures based on advanced 3D printing technology in tailoring the interaction between IR light and materials on a subwavelength scale. Coal seam impact risk assessment is the premise of coal mine safety, which can reduce the occurrence of underground impact pressure accidents and directly affect the safety, coal production, economic and social benefits of coal mining enterprises. In order to evaluate the impact risk of coal seams more reasonably and comprehensively, and consider the weights of different influencing factors on the impact risk of coal seams, the neural network model is proposed to evaluate the impact risk of coal seams. Mining depth, impact tendency, geological structure and mining technology are selected as the influencing factors of coal seam impact risk. Each influencing factor contains different evaluation indices, a total of 18. The 18 evaluation indices and the impact risk level are normalized and quantified. The BP neural network model for evaluating coal seam impact risk level is established, and the impact risk of 2-1 coal seams in a mine in Inner Mongolia is comprehensively evaluated and analyzed in this study. The results show that the BP neural network model can represent coal seam impact risk level well. The application of the BP neural network model to evaluate coal seam impact risk level has the characteristics of high precision, fast calculation speed and less artificial calculation, which provides an efficient and convenient method for the evaluation of coal seam impact risk. MicroRNAs (miRNAs) have been shown to be dysregulated in virus-related cancers; however, miRNA regulation of virus-related cancer development and progression remains poorly understood. Here, we report that miR-148a is repressed by hepatitis B virus (HBV) X protein (HBx) to promote cancer growth and metastasis in a mouse model of hepatocellular carcinoma (HCC). Hematopoietic pre-B cell leukemia transcription factor-interacting protein (HPIP) is an important regulator of cancer cell growth. We used miRNA target prediction programs to identify miR-148a as a regulator of HPIP. Expression of miR-148a in hepatoma cells reduced HPIP expression, leading to repression of AKT and ERK and subsequent inhibition of mTOR through the AKT/ERK/FOXO4/ATF5 pathway. HBx has been shown to play a critical role in the molecular pathogenesis of HBV-related HCC. We found that HBx suppressed p53-mediated activation of miR-148a. Moreover, expression of miR-148a was downregulated in patients with HBV-related liver cancer and negatively correlated with HPIP, which was upregulated in patients with liver cancer. In cultured cells and a mouse xenograft model, miR-148a reduced the growth, epithelial-to-mesenchymal transition, invasion, and metastasis of HBx-expressing hepatocarcinoma cells through inhibition of HPIP-mediated mTOR signaling.
