It is crucial to comprehend how fertilization and cultivation management alter the composition of dissolved organic carbon (DOC) and microbial communities to regulate the biogeochemical cycling of soil nutrients and mitigate adverse impacts on soil–water quality. Based on 15 years of long-term field trials conducted in purple soil on farmland with a slope of 15° in southwestern China, the following five treatments were examined: CK (no fertilizer was applied), T1 (NPK plus organic manure and downslope cultivation), T2 (NPK and downslope cultivation), T3 (1.5-fold NPK and downslope cultivation), and T4 (NPK and contour cultivation). Soil samples were obtained from summer maize at two soil depths (0–10 and 10–20 cm) and from rhizospheric soil, and the changes in the DOC content, UV–visible (UV–Vis) absorptivity, and phospholipid fatty acids (PLFAs) were assessed. Our results revealed a significant change in the DOC content following fertilization, especially in T1, as it was 136.0%, 179.4%, and 132.2% higher, respectively, than that in CK at the 0–10 and 10–20 cm depths and rhizospheric soil. Fertilization decreased the UV–Vis absorptivity variables of DOC (i.e., SUVA254, SUVA260, SUVA400, SUVA465, SUVA665, and C:C ratio) and raised the E4:E6 ratio (fulvic acid to humic acid in DOC), regardless of T2 and T3 at the 10–20 cm depth and in the rhizospheric soil compared with those in CK, respectively. Fertilization significantly increased the total PLFA content and selected microbial groups relative to CK. Among the treatments, T1 significantly increased the total PLFA content by 50.6%, 59.0%, and 46.2%, respectively, relative to CK, at the 0–10 and 10–20 cm depths and in the rhizospheric soil. The microbial community structure in contour cultivation (T4) was significantly greater than in downslope cultivation (T2). Random forest analysis (RFA) revealed that SOC and DOC were likely the primary variables for regulating the total PLFAs in the examined soil. Partial least squares path modeling (PLS-PM) further indicated that the DOC content and the ratio of E4:E6 among DOC compositions had greater effects on the soil microbial community structure in the examined soil. These observations suggested that long-term fertilization and cultivation management are effective approaches to regulating the soil microbial community structure by altering the composition of DOC in sloping farmland. A two-year experiment was carried out at Shenyang Agricultural University's research field area in China to evaluate the impact of the combined application of straw mulch (0 and 8 t ha−1) and biochar (0, 4, 12, and 36 t ha−1) on the morphological traits and grain development of rainfed maize during 2018 and 2019. The results showed that straw mulch and different biochar application rates significantly impacted the maize growth index. Compared to non-biochar-treated soils, the introduction of straw mulch improved plant height, stem diameter, leaf area index (LAI), leaves, stem, root, and crop growth rate (CGR), and dry weight of rainfed maize crop. The highest plant height, stem diameter, LAI, leaves, stem, root growth rate, CGR, and dry weight of rainfed maize crop were reported when soil was treated with a higher rate of biochar (36 t ha−1). Biochar increased grain filling rate while decreasing grain filling duration in rainfed maize crops. Our results indicate that straw mulch and biochar-based soil management strategies can improve the rainfed maize growth with the environmental benefits of global warming mitigation. However, due to the wide range of biochar properties, the interactions between straw mulch and biochar should be given special consideration in the maize cropping system. [ABSTRACT FROM AUTHOR]

Gloeostereum incarnatum has edible and medicinal value and was first cultivated and domesticated in China. We sequenced the G. incarnatum monokaryotic strain GiC-126 on an Illumina HiSeq X Ten system and obtained a 34.52-Mb genome assembly sequence that encoded 16,895 predicted genes. We combined the GiC-126 genome with the published genome of G. incarnatum strain CCMJ2665 to construct a genetic linkage map (GiC-126 genome) that had 10 linkage groups (LGs), and the 15 assembly sequences of CCMJ2665 were integrated into 8 LGs. We identified 1912 simple sequence repeat (SSR) loci and detected 700 genes containing 768 SSRs in the genome; 65 and 100 of them were annotated with gene ontology (GO) terms and KEGG pathways, respectively. Carbohydrate-active enzymes (CAZymes) were identified in 20 fungal genomes and annotated; among them, 144 CAZymes were annotated in the GiC-126 genome. The A mating-type locus (MAT-A) of G. incarnatum was located on scaffold885 at 38.9 cM of LG1 and was flanked by two homeodomain (HD1) genes, mip and beta-fg. Fourteen segregation distortion markers were detected in the genetic linkage map, all of which were skewed toward the parent GiC-126. They formed three segregation distortion regions (SDR1-SDR3), and 22 predictive genes were found in scaffold1920 where three segregation distortion markers were located in SDR1. In this study, we corrected and updated the genomic information of G. incarnatum. Our results will provide a theoretical basis for fine gene mapping, functional gene cloning, and genetic breeding the follow-up of G. incarnatum. [ABSTRACT FROM AUTHOR]

Color is an important quality attribute of fungi, and a useful marker for classification, genetic, and molecular research. However, there is much debate over which enzymes play key regulatory roles in pigment synthesis pathways among different fungi and even within the same species. Auricularia cornea is the most widely cultivated mushroom in the genus Auricularia ; 1.834 million tons of this mushroom were produced in 2016 in China. Thus, systematic studies on its color inheritance and the genes encoding key enzymes for pigment synthesis have high scientific and economic value. In this study, the white strain ACW001 and the purple strain ACP004 of A. cornea were used as dikaryotic parents. Selfing populations of ACW001 and ACP004 were constructed with their monokaryotic strains. The fruiting body color of the two populations was consistent with that of their parents, confirming that the two parents were color homozygotes. All strains in the hybrid population of the two parents produced purple fruiting bodies. A robust hybrid strain (ACW001-33×ACP004-33) was selected from the hybrid population, and 87 monokaryotic strains of ACW001-33×ACP004-33 were obtained as a mapping population. Finally, a testcross population was constructed by crossing the mapping population with the test strain ACW001-9. The color genotype of each monokaryotic strain in the mapping population was identified by a fruiting test. The genomes of the two monokaryotic strains ACW001-33 and ACP004-33 were sequenced, and then simple sequence repeat (SSR) and sequence-related amplified polymorphism (SRAP) molecular marker primers were developed. Then, 88 pairs of primers that could distinguish the genotypes of the mapping population were used to construct a genetic linkage map. The genetic linkage map consisted of 12 linkage groups (LGs) spanning 1315.2 cM. The color control locus was preliminarily located at 24.5 cM of the 11th LG. Fine-mapping primers were designed based on sequence differences between ACW001-33 and ACP004-33 in the primary location region. Four color control candidate genes were located in an 8.2-kb region of ACW001-33_contig733 and a 9.2-kb region of ACP004-33_contig802. Homologous alignment and prediction of conserved domain analyses indicated that two of the color control candidate genes encoded proteins with unknown function, and the other two, ACP004_g11815 and ACP004_g11816, encoded glutamyl aminotransferases. These two genes were consecutively arranged on ACP004-33_contig802, and were likely to encode key enzymes in the γ-glutamine-4-hydroxy-benzoate (GHB) pigment synthesis pathway. Primers were designed from the flanking sequences of the two genes and used to analyze the testcross population. Products were amplified only from the 30 testcross strains with purple fruiting bodies, confirming the accuracy of the localization results. We discuss the deficiencies and advantages of map-based cloning in fungi vs. plants, and summarize the steps and requirements of the map-based cloning method for fungi. Affected by climate variability and human activities, the changes of basin environment have altered runoff, posing new challenges to water resources and land use management strategies, especially in areas with limited water resources. The purpose of this study is to quantify factors that drive the changes in runoff components. Annual runoff data from six hydrological stations in the Weihe River Basin, China and sixteen meteorological stations data from 1961 to 2015 were used. The Mann-Kendall test and Pettitt method were used to diagnose the runoff sequence and detect its abrupt-point. The period before the abrupt-point of annual runoff was taken as the base period (Period I), while the period after the abrupt-point was taken as the calculation period (Period II). The elasticity coefficient of runoff (Q) was deduced based on the Budyko hypothesis, including climate variabilities such as precipitation (P) and potential evapotranspiration (Ep), and the catchment characteristics representing land surface changes (n). The elasticity coefficient was the smallest for Ep ranging from −1.210 ~ −2.847, intermediate for n ranging from −1.602 ~ −2.964, and greatest for P ranging from 2.210–3.847. Runoff exhibited a significant downward trend throughout the Weihe River Basin, where the land surface change is the main cause of runoff reduction, accounting for 75% at the largest Linjiacun hydrological station, with the smallest being 40.36% at the Zhuangtou hydrological station. Adaptive water resource management measures are necessary to cope with future environmental change and water resource scarcity. • The analytical method based on coupling water balance and the Budyko hypothesis on the elasticity coefficient is applied. • Runoff showed a total reduction at six hydrological stations in the Weihe River Basin. • Overall, the change of land surface is the main factor of runoff decrease in the Weihe River Basin, followed by precipitation, and the potential evapotranspiration was less affected.
