BACKGROUND: Protein oxidation during food processing causes changes in the balance of protein–molecular interactions and protein–water interactions, ultimately leading to protein denaturation, which results in the loss of a range of functional properties. Therefore, how to control the oxidative modification of proteins during processing has been the focus of research. RESULTS: In the present study, the intrinsic fluorescence value of the myofibrillar proteins (MP) decreased and the surface hydrophobicity value increased, indicating that the heat treatment caused a significant change in the conformation of the MP. With an increase in heating temperature, protein carbonyl content increased, total sulfhydryl content decreased, and protein secondary structure changed from α‐helix to β‐sheet, indicating that protein oxidation and aggregation occurred. Sodium dodecyl sulfate‐polyacrylamide gel electrophoresis revealed that heat treatment can lead to the degradation of proteins, especially myosin heavy chain, although actin had a certain thermal stability. In total, 733 proteins were identified by proteomics, and the protein oxidation caused by low temperature vacuum heating (LTVH) was determined to be mild oxidation dominated by malondialdehyde and 4‐hydroxynonenal by oxidation site division. CONCLUSION: The present study has revealed the effect of LTVH treatment on the protein oxidation modification behavior of sturgeon meat, and explored the effect mechanism of LTVH treatment on the processing quality of sturgeon meat from the perspective of protein oxidation. Background: Diabetes mellitus (DM) is considered a major risk factor for myocardial infarction (MI), and MI patients with DM have a poor prognosis. Accordingly, we aimed to investigate the additive effects of DM on LV deformation in patients after acute MI. Materials and methods: One hundred thirteen MI patients without DM [MI (DM−)], 95 with DM [MI (DM+)] and 71 control subjects who underwent CMRscanning were included. LV function, infarct size and LV global peak strains in the radial, circumferential and longitudinal directions were measured. MI (DM+) patients were divided into two subgroups based on the HbA1c level (< 7.0% and ≥ 7.0%). The determinants of reduced LV global myocardial strain for all MI patients and MI (DM+) patients were assessed using multivariable linear regression analyses. Results: Compared with control subjects, both MI (DM−) and MI (DM+) patients presented higher LV end-diastolic and end-systolic volume index and lower LV ejection fraction. LV global peak strains progressively declined from the control group to the MI(DM−) group to the MI(DM+) group (all p < 0.05). Subgroup analysis showed that LV global radial PS and longitudinal PS were worse in MI(MD+) patients with poor glycemic control than in those with good glycemic control (all p < 0.05). DM was an independent determinant of impaired LV global peak strain in radial, circumferential and longitudinal directions in patients after acute MI (β = − 0.166, 0.164 and 0.262, both p < 0.05). The HbA1c level was independently associated with a decreased LV global radial PS (β = − 0.209, p = 0.025) and longitudinal PS (β = 0.221, p = 0.010) in MI (DM+) patients. Conclusions: DM has an additive deleterious effect on LV function and deformation in patients after acute MI, and HbA1c was independently associated with impaired LV myocardial strain. Purpose: The aim of our study was to investigate a comprehensive profile of streptozotocin (STZ)-induced early diabetic retinopathy (DR) mice to identify a risk scoring signature based on micorRNAs (miRNAs) for early DR diagnosis. Methods: RNA sequencing was performed to obtain the gene expression profile of retinal pigment epithelium (RPE) in early STZ-induced mice. Differentially expressed genes (DEGs) were determined with log2|fold change (FC)| > 1 and p value < 0.05. Functional analysis was carried out based on gene ontology (GO), Kyoto Encyclopedia of Genes and Genomes (KEGG) pathway enrichment analysis and the protein–protein interaction (PPI) network. We predicted the potential miRNAs via online tools and ROC curves were then conducted. Three potential miRNAs with AUC > 0.7 were explored via public datasets and a formula was further established to evaluate DR severity. Results: In total, 298 DEGs (200 up-regulating and 98 down-regulating) were obtained through RNA sequencing. Hsa-miR-26a-5p, hsa-miR-129-2-3p and hsa-miR-217 were three predicted miRNAs with AUC > 0.7, suggesting their potential to distinguish healthy controls from early DR. The formula of DR severity score = 19.257 − 0.004 × hsa-miR-217 + 5.09 × 10−5 × hsa-miR-26a-5p − 0.003 × hsa-miR-129-2-3p was established based on regression analysis. Conclusions: In the present study, we investigated the candidate genes and molecular mechanisms based on RPE sequencing in early DR mice models. Hsa-miR-26a-5p, hsa-miR-129-2-3p and hsa-miR-217 could work as biomarkers for early DR diagnosis and DR severity prediction, which was beneficial for DR early intervention and treatment. Background: Obesity is highly prevalent in patients with hypertrophic cardiomyopathy (HCM) and believed to influence its phenotype. Purpose: To explore the effects of obesity on left ventricular (LV) remodeling and long‐term clinical course in Chinese patients with HCM. Study Type: Longitudinal. Population: A total of 247 patients with HCM classified according to body mass index (BMI) (normal weight: BMI = 18.0–22.9 kg/m2 [N = 90]; overweight: BMI = 23.0–24.9 kg/m2 [N = 58]; and obese: BMI ≥ 25 kg/m2 [N = 99]). Field Strength/Sequence: 3.0 T/Balanced steady‐state free precession sequence and phase‐sensitive inversion recovery late gadolinium enhancement (LGE) sequence. Assessment: LV function and geometry were measured. LV peak strain analysis was performed. The presence and percentage of LGE in the LV were recorded. The endpoints including heart failure, sudden cardiac death, and overall composite outcome were assessed during a median follow‐up of 4.1 years (interquartile range, 3.0–6.2 years). Statistical Tests: One‐way analysis of variance, Kruskal–Wallis test, or chi‐square test; Pearson correlation coefficient (r); multivariable linear regression analysis; Kaplan–Meier survival analysis; and Cox proportional hazards model analysis were conducted. A two‐tailed P‐value < 0.05 was considered statistically significant. Results: Obese patients exhibited a significant progressive increase in LV mass compared with normal‐weight patients. The magnitude of all LV strain indices gradually and significantly decreased as BMI increased, whereas LV ejection fraction was not significantly different among BMI groups (P = 0.364). Multivariable linear regression analysis showed that obesity had a significant association with impaired strain indices as well as with indexed LV mass. Multivariable Cox model analysis retained obesity as an independent marker for future endpoints, and conveyed a > 3‐fold increase in risk compared with patients with normal weight (hazard ratio, 3.04; 95% confidence interval, 1.07–6.57). Data Conclusion: Obesity is an important environmental modifier that is associated with adverse LV remodeling and is independently associated with future clinical outcomes in Chinese patients with HCM. In this study, Buton rock asphalt (BRA), nano-aerogel (NA), and an antiaging agent (AA) were selected to modify base asphalt to improve its aging resistance. Based on the preliminary performance test results, the optimum dosage of the three modifiers was determined. The asphalt's rheological properties and microscopic properties were tested under different aging methods. Its high- and low-temperature rheological properties were characterized by dynamic shear rheological tests and bending beam rheological tests. Fourier transform infrared spectroscopy (FTIR) analyzed changes in the asphalt's functional groups before and after aging. The ultraviolet absorption capacity of the asphalt before and after aging was studied by an ultraviolet-visible spectrophotometer (UV-VIS). The results showed that the optimal dosing of the modifiers was 2% NA, 10% BRA, 0.3% antioxidant 1010, and 0.4% UV-327. Compared with the base asphalt, the composite modified asphalt with BRA, NA, and AA showed improvement in high- and low-temperature performance. The aging index of both the rheological test and the infrared spectroscopy test showed that the composite modified asphalt had the best resistance to aging. Its resistance to UV radiation was higher than that of the base asphalt, and the BRA/NA modified asphalt. Therefore, the addition of BRA and NA effectively improved the asphalt's high- and low-temperature performance. Addition of the antioxidants 1010 and UV-327 effectively improved its aging resistance. Artificial synapses and neurons are crucial milestones for neuromorphic computing hardware, and memristors with resistive and threshold switching characteristics are regarded as the most promising candidates for the construction of hardware neural networks. However, most of the memristors can only operate in one mode, that is, resistive switching or threshold switching, and distinct memristors are required to construct fully memristive neuromorphic computing hardware, making it more complex for the fabrication and integration of the hardware. Herein, we propose a flexible dual‐mode memristor array based on core–shell CsPbBr3@graphdiyne nanocrystals, which features a 100% transition yield, small cycle‐to‐cycle and device‐to‐device variability, excellent flexibility, and environmental stability. Based on this dual‐mode memristor, homo‐material‐based fully memristive neuromorphic computing hardware—a power‐free artificial nociceptive signal processing system and a spiking neural network—are constructed for the first time. Photodynamic antimicrobial chemotherapy (PACT), as a novel and effective therapeutic modality to eradicate drug resistant bacteria without provoking multidrug resistance, has attracted increasing attention. This study examined the antimicrobial efficacy of the novel cationic amino acid-porphyrin conjugate 4I with four lysine groups against two different clinical isolated strains (drug sensitive and multidrug resistant) of the Acinetobacter baumannii species and its toxicity on murine dermal fibroblasts in vitro, as well as the therapeutic effect of PACT on acute, potentially lethal multidrug resistant strain excisional wound infections in vivo. The PACT protocol exposed 4I to illumination, exhibiting high antimicrobial efficacy on two different strains due to a high yield of reactive oxygen species (ROS) and non-selectivity to microorganisms. The photoinactivation effects of 4I against two different strains were dose-dependent. At 3.9 μM and 7.8 μM, PACT induced 6 log units of inactivation of sensitive and multidrug resistant strains. In contrast, 4I alone and illumination alone treatments had no visibly antimicrobial effect. Moreover, cytotoxicity tests revealed the great safety of the photosensitizer 4I in mice. In the in vivo study, we found 4I-mediated PACT was not only able to kill bacteria but also accelerated wound recovery. Compared with non-treated mice, over 2.89 log reduction of multidrug resistant Acinetobacter baumannii strain was reached in PACT treat mice at 24 h post-treatment. These results imply that 4I-mediated PACT therapy is an effective and safe alternative to conventional antibiotic therapy and has clinical potential for superficial drug-resistant bacterial infections.

AIM: To study the effects of αB-crystallin on the αB-crystallin content in retina, growth associated protein-43(GAP-43)expression of retinal ganglion cells(RGCs), amplitude of electroretinogram(ERG)b-wave, and to investigate the promotive effect of αB-crystallin on axonal regeneration of RGCs after acute ocular hypertension(AOH). METHODS: One hundred and twenty male Sprague-Dawley(SD)rats(220±10g)were randomly divided into four groups(thirty in each group). αB group,(AOH following intravitreal injection of αB crystallin group, n=30). S group,(AOH following intravitreal injection of normal saline solution group, n=30). P group,(AOH following vitreous puncture group, n=30). H group,(AOH group, n=30). Western blot was assayed αB-crystallin content 7 and 14 days after model; Immunohistochemistry was observed GAP-43 expression 7, 14 and 21 days after model established; Amplitude of ERG b-wave was tested retinal function before and 1 month after model. RESULTS: The αB content in retina of αB group was higher 7 days after model established, fewer 14 days after model(P=0.000, P=0.000, P=0.001, P=0.000). But the αB content in retina of αB group was more serious compared with other groups at the same time points(P=0.019,P=0.016); The GAP-43 expression of RGCs in αB group was highest 7 days after model established, started to decrease 14 days after model established, which was fewest 21 days after model established(P=0.000, P=0.000, P=0.000, P=0.000). But the GAP-43 expression of RGCs in αB group was significantly higher than in other groups at each time point(P =0.002, P =0.011, P =0.009); The amplitude of ERG b-wave was lower 1 month after model(P=0.014, P=0.004, P=0.003, P=0.006). There were no statistical differences among groups on amplitude of ERG b-wave before model established(P=0.993). The amplitude of ERG b-wave in αB group was higher than others 1 month after model(P=0.002). CONCLUSION: Exogenous αB-crystallin could improve αB-crystallin expression of retinas; αB-crystallin contributed to the increase of GAP-43 expression of RGCs and RGCs axonal regeneration; αB-crystallin could promote the recovery of retinal function and was non-toxic on retinas of rat.

Background: Patients with type 2 diabetes mellitus (T2DM) are highly susceptible to cardiovascular disease, and coronary artery disease (CAD) is their leading cause of death. We aimed to assess whether computed tomography (CT) based imaging parameters and radiomic features of pericoronary adipose tissue (PCAT) can improve the diagnostic efficacy of whether patients with T2DM have developed CAD. Methods: We retrospectively recruited 229 patients with T2DM but no CAD history (146 were diagnosed with CAD at this visit and 83 were not). We collected clinical information and extracted imaging manifestations from CT images and 93 radiomic features of PCAT from all patients. All patients were randomly divided into training and test groups at a ratio of 7:3. Four models were constructed, encapsulating clinical factors (Model 1), clinical factors and imaging indices (Model 2), clinical factors and Radscore (Model 3), and all together (Model 4), to identify patients with CAD. Receiver operating characteristic curves and decision curve analysis were plotted to evaluate the model performance and pairwise model comparisons were performed via the DeLong test to demonstrate the additive value of different factors. Results: In the test set, the areas under the curve (AUCs) of Model 2 and Model 4 were 0.930 and 0.929, respectively, with higher recognition effectiveness compared to the other two models (each p < 0.001). Of these models, Model 2 had higher diagnostic efficacy for CAD than Model 1 (p < 0.001, 95% CI [0.129–0.350]). However, Model 4 did not improve the effectiveness of the identification of CAD compared to Model 2 (p = 0.776); similarly, the AUC did not significantly differ between Model 3 (AUC = 0.693) and Model 1 (AUC = 0.691, p = 0.382). Overall, Model 2 was rated better for the diagnosis of CAD in patients with T2DM. Conclusions: A comprehensive diagnostic model combining patient clinical risk factors with CT-based imaging parameters has superior efficacy in diagnosing the occurrence of CAD in patients with T2DM. Most of studies on change-point at a regional or global scale have only examined a single hydrometeorological variable and have been unable to identify any underlying explanations. In this study, we identified change-points and long-term trends of six wildfire-related variables and attempted to explain the cause of change-point from atmospheric–oceanic indices. As a result, we discovered that the main change-point dates for the precipitation, temperature, and drought codes, as well as the duff moisture code, fine fuel moisture code, and fire weather index, were 1995–2000 and 2000–2005, respectively. Furthermore, the relationship between the change-point of six variables and atmospheric–oceanic indices was discussed through the correlation coefficient. For example, the Atlantic Multidecadal Oscillation was found to dominate the precipitation in West Africa. The aim of this study is to evaluate the benefits of dexmedetomidine in epidural labor analgesia compared with lipophilic opioids. The databases of PubMed, Embase, Cochrane Library, Web of Science, Wanfang, and SinoMed were searched from inception to Mar. 25, 2021 for randomized controlled trials (RCT) that assessed dexmedetomidine versus lipophilic opioids as adjuvants to local anesthetics in epidural labor analgesia. Meta-analyses were conducted with RevMan 5.3, and a random-effects model was adopted. A total of 11 RCTs involving 1099 parturients were enrolled. The results showed that, compared with the control group, dexmedetomidine significantly reduced Visual Analogue Scale (VAS) scores both at 30 minutes after induction [weighted mean difference (WMD) = -0.40, 95% CI: -0.61 to -0.20] and on delivery (WMD = -0.83, 95% CI: -1.15 to -0.50), reduced analgesic consumption (WMD = -6.29 mL, 95% CI: -10.49 to -2.10), shortened the duration of the first (WMD = -9.58 minutes, 95% CI: -18.12 to -1.04) and second (WMD = -1.66 minutes, 95%CI: -3.20 to -0.12) stage of labor, increased maternal bradycardia [risk ratio (RR) = 2.44, 95% CI: 1.31 to 4.53] and motor blockade (RR = 5.30, 95% CI: 2.21 to 12.73), reduced nausea/vomiting (RR = 0.34, 95% CI: 0.20 to 0.57), pruritis (RR= 0.19, 95%CI: 0.06 to 0.58) and shivering (RR = 0.37, 95% CI: 0.18 to 0.77). There was no significant difference between groups in the rate of instrumental delivery (p = 0.68), and cesarean delivery (p = 0.40), Apgar scores at 1 minute (p = 0.24), at 5 minutes (p = 0.36), and the umbilical arterial PH (p = 0.16). Excessive intake of high-caffeine tea will induce health-related risk. Therefore, breeding and cultivating tea cultivars with less caffeine is a feasible way to control daily caffeine intake. Cocoa tea (Camellia ptilophylla Chang) is a wild tea plant which grows leaves with little or no caffeine. However, the vegetative propagation of cocoa tea plants is difficult due to challenges with rooting. Whether natural seeds collected from wild cocoa tea plants can be used to produce less-caffeinated tea remains unknown, because research on the separation of traits among the seed progeny population is lacking. The present study was set to investigate the variation of caffeine and other chemical compositions in seed-propagated plant individuals using colorimetric and HPLC methods. It shows that there were great differences in chemical composition among the seed-propagated population of wild cocoa tea plants, among which some individuals possessed caffeine contents as high as those of normal cultivated tea cultivars (C. sinensis), suggesting that the naturally seed-propagated cocoa tea seedlings are not suitable for directly cultivating leaf materials to produce low-caffeine tea. Therefore, the cocoa tea plants used for harvesting seeds for growing low-caffeine tea plants should be isolated in order to prevent their hybridization with normal cultivated C. sinensis plants. Interestingly, the leaves of cocoa tea seedlings contained high levels of gallocatechin gallate (GCG) and would be a good source of leaf materials for extracting more stable antioxidant, because GCG is a more stable antioxidant than epigallocatechin gallate (EGCG), the dominant component of catechins in normal cultivated tea cultivars. Some plant individuals which contained low levels of caffeine along with high levels of amino acids and medium levels of catechins, are considered to be promising for further screening of less-caffeinated green tea cultivars. Among the three DMTF1 splicing isoforms, DMTF1α acts as a tumour suppressor through promoting p14ARF expression, while DMTF1β exhibits an oncogenic role likely through antagonizing DMTF1α. However, the molecular mechanism underlying alternative splicing of DMTF1 pre-mRNA has not been delineated. In the current study, we discovered SRSF5 as a regulatory protein binding to a region located between DMTF1β and α acceptor splice sites to promote DMTF1β and γ splicing. We demonstrated that SRSF5 expression positively correlated with DMTF1β/α ratio in breast cancer samples, and ectopically expressed SRSF5 promoted the splicing of DMTF1β and γ, but not DMTF1α, when testing endogenous DMTF1 pre-mRNA and a reporter construct. Upon SRSF5 knockdown, we observed significantly decreased DMTF1β and γ ratios of endogenous transcripts. An RNA sequence just upstream of the α acceptor site contains two adjacent SRSF5 binding elements, one of which overlaps with an SF1 binding site. Our mechanistic studies revealed that SRSF5 binding to both elements in this region could consign SF1 to its distal-binding sites close to the β and γ acceptor sites, favouring splicing of their isoforms. Overall, our study revealed SRSF5 as a key regulator to promote DMTF1β and γ splicing, and consequently reduce DMTF1α splicing. Kunming city is located in the middle of Yunnan Province. Due to large-scale groundwater exploitation and urban development in recent years, this area has been affected by surface subsidence. In this paper, Interferometric Synthetic Aperture Radar (InSAR) and Global Navigation Satellite System (GNSS) data are used to monitor the surface subsidence in Kunming city area for better analysis and understanding. The study used data of Sentinel-1A from 2018 to 2020 with atmospheric correction based on GACOS to calculate the average annual subsidence rate in Kunming city area, and the results show that the maximum subsidence rate is 48 mm/year. The subsidence obtained by InSAR is compared with the vertical deformation information obtained by eight GNSS stations in continuous operation in the study area. The subsidence rate trend show by the two methods is consistent, which further verifies the validity of InSAR data to reflect the local deformation. Experimental results shown that the eastern and northeastern Dianchi lake areas were affected by underground resources mining, and the induced surface subsidence characteristics were obvious, with the surface subsidence rate reachde 48 mm/year and 37 mm/year respectively. The Kunyang Phosphate Mine also had different degrees of mining subsidence disaster, with the maximum subsidence rate reached 36 mm/year. The subsidence rate of InSAR and GNSS has the same trend on the whole. Purpose. To establish an effective and accurate prognostic nomogram for lung adenocarcinoma (LUAD). Patients and Methods. 62,355 LUAD patients from 1975 to 2016 enrolled in the Surveillance, Epidemiology, and End Results (SEER) database were randomly and equally divided into the training cohort (n = 31,179) and the validation cohort (n = 31,176). Univariate and multivariate Cox regression analyses screened the predictive effects of each variable on survival. The concordance index (C-index), calibration curves, receiver operating characteristic (ROC) curve, and area under the ROC curve (AUC) were used to examine and validate the predictive accuracy of the nomogram. Kaplan–Meier curves were used to estimate overall survival (OS). Results. 10 prognostic factors associated with OS were identified, including age, sex, race, marital status, American Joint Committee on Cancer (AJCC) TNM stage, tumor size, grade, and primary site. A nomogram was established based on these results. C-indexes of the nomogram model reached 0.777 (95% confidence interval (CI), 0.773 to 0.781) and 0.779 (95% CI, 0.775 to 0.783) in the training and validation cohorts, respectively. The calibration curves were well-fitted for both cohorts. The AUC for the 3- and 5-year OS presented great prognostic accuracy in the training cohort (AUC = 0.832 and 0.827, respectively) and validation cohort (AUC = 0.835 and 0.828, respectively). The Kaplan–Meier curves presented significant differences in OS among the groups. Conclusion. The nomogram allows accurate and comprehensive prognostic prediction for patients with LUAD. Nitrogen stripping plays a vital role during the sulfate-rich wastewater treatment process by sulfate-reducing bacteria (SRB). However, the interaction between nitrogen stripping, sulfate reduction capacity, S2– concentration, and microbial community structure is rarely studied. Here, two anaerobic systems were conducted to analyze these interactions simultaneously. Results showed that the toxic products (H2S) were stripped by nitrogen at first. Then, the increased relative abundance of fermentative bacteria accelerated the carbon sources conversion, and Desulfovibrio, Dethiosulfovibrio, and Sulfurospirillum facilitated the sulfate reduction after nitrogen stripping. Finally, the produced S2− concentration was increased by 12%, and the sulfate reduction rate was increased by 9.2% compared to that without nitrogen stripping. [ABSTRACT FROM AUTHOR]

Treatments are lacking for sarcopenia, which is an age‐related disease characterized by loss of skeletal muscle mass, strength, and/or physical performance. Icariin is a phytomolecule from herbal Epimedium, a traditional Chinese medicine widely used to treat musculoskeletal disorders for thousands of years. Here the effects of icariin against sarcopenia are investigated and the underlying mechanism is elucidated. A classic rat model of bilaterally orchiectomized (ORX) is used to induce sarcopenia. After administration for 8 weeks, compared to the control group, the forelimb grip strength, the specific tetanic forces of the soleus (SOL) and extensor digitorum longus muscle (EDL) are higher, and the fiber cross‐sectional areas (CSAs) of the gastrocnemius and tibialis anterior muscle are larger in the icariin group. In addition, icariin promotes mRNA and protein expressions of myosin heavy chain (MyHC) both in SOL and EDL. Mechanistically, icariin significantly suppresses the mRNA and protein expressions of FOXO3a, atrogin‐1, and MuRF‐1, which are related to the degradation of myosin heavy chain. Collectively, icariin protects from sarcopenia in ORX rats characterized by enhancing grip strength and skeletal muscle contraction, as well as increasing skeletal muscle CSA by inhibiting the ubiquitination degradation of the MyHC in skeletal muscle fibers. This paper reports on the flexural behavior of nine steel fiber reinforced-recycled aggregate concrete (SFRAC) beams through combined experimental and finite element analysis. The test parameters in this study include the steel fiber volume fraction, recycled aggregate replacement ratio, and concrete strength. The failure modes, crack development, load-deflection curves, and flexural bearing capacity of SFRAC beams are investigated in detail. The test results indicated that cracks and concrete crushing are formed in the compression zone of all specimens. The flexural bearing capacity of SFRAC beams increases with the increase of steel fiber volume fraction and concrete strength and the decrease of recycled aggregate replacement ratio. In addition, the results are compared with those of the finite element analysis. Based on the uniaxial compressive constitutive model of SFRAC, a new model for calculating the flexural bearing capacity of SFRAC beams is proposed. The prediction and test results are compared to evaluate the accuracy of the developed formula. Background: Previous studies reported that there was right ventricular (RV) systolic dysfunction in patients with hypertension. The aim of this study was to evaluate the impact of type 2 diabetes mellitus (T2DM) on RV systolic dysfunction and interventricular interactions using cardiac magnetic resonance feature tracking (CMR-FT) in patients with essential hypertension. Methods and methods: Eighty-five hypertensive patients without T2DM [HTN(T2DM −)], 58 patients with T2DM [HTN(T2DM +)] and 49 normal controls were included in this study. The biventricular global radial, circumferential and longitudinal peak strains (GRS, GCS, GLS, respectively) and RV regional strains at the basal-, mid- and apical-cavity, were calculated with CMR-FT and compared among controls and different patient groups. Backward stepwise multivariable linear regression analyses were used to determine the effects of T2DM and left ventricular (LV) strains on RV strains. Results: The biventricular GLS and RV apical longitudinal strain deteriorated significantly from controls, through HTN(T2DM-), to HTN(T2DM +) groups. RV middle longitudinal strain in patient groups were significantly reduced, and LV GRS and GCS and RV basal longitudinal strain were decreased in HTN(T2DM +) but preserved in HTN(T2DM-) group. Multivariable regression analyses adjusted for covariates demonstrated that T2DM was independently associated with LV strains (LV GRS: β = − 4.278, p = 0.004, model R2 = 0.285; GCS: β = 1.498, p = 0.006, model R2 = 0.363; GLS: β = 1.133, p = 0.007, model R2 = 0.372) and RV GLS (β = 1.454, p = 0.003, model R2 = 0.142) in hypertension. When T2DM and LV GLS were included in the multiple regression analysis, both T2DM and LV GLS (β = 0.977 and 0.362, p = 0.039 and < 0.001, model R2 = 0.224) were independently associated with RV GLS. Conclusions: T2DM exacerbates RV systolic dysfunction in patients with hypertension, which may be associated with superimposed LV dysfunction by coexisting T2DM and suggests adverse interventricular interactions. Esophageal squamous cell carcinoma (ESCC) is one of the most aggressive malignancies with complex tumor microenvironment (TME) which has been proven to be associated with therapeutic failure or resistance. A deeper understanding of the complex TME and cellular heterogeneity is urgently needed in ESCC. Here, we generated single‐cell RNA sequencing (scRNA‐seq) of 25 796 immune and 8197 non‐immune cells from three primary tumor and paired normal samples in ESCC patients. The results revealed intratumoral and intertumoral epithelium heterogeneity and tremendously differences in tumor and normal epithelium. The infiltration of myofibroblasts, one subtype of fibroblasts, might play important roles in the progression of ESCC. We also found that some differentially expressed genes and markers in epithelium and fibroblast subtypes showed prognostic values for ESCC. Diverse cell subtypes of T cells and myeloid cells were identified, including tumor‐enriched HAVCR2+ CD4+ T cells with significantly exhausted signature. The epithelium and myeloid cells had more frequent cell–cell communication compared with epithelium and T cells. Taken together, this study provided in‐depth insights into the cellular heterogeneity of TME in ESCC and highlighted potential therapeutic targets including for immunotherapy. [ABSTRACT FROM AUTHOR]

In a recent Comment, Chang et al. [Phys. Plasmas 29, 114703 (2022)] have studied the electron dynamics for an electron accelerating in a tightly focused Gaussian laser pulse with the higher order correction terms, and it is found that the initial position of the electron is the reason why the electron is pushed to the −z axis at the end. In this Reply, the electron dynamics and its nonlinear Thomson backscattering in a tightly focused Gaussian laser pulse with the higher order correction terms are presented, and it is found that the result is consistent with the lowest order approximation case in our paper [Hong et al., Phys. Plasmas 29, 043102 (2022)]. Meanwhile, it is also found that when the longitudinal deceleration effect of the ponderomotive force on the electron introduced by the falling part of the tightly focused laser pulse is greater than the longitudinal acceleration effect of the rising part, the electron that is initially stationary will be slightly pushed to the −z axis at the end, which can well explain the "return" phenomenon of the electron in the longitudinal direction both in our paper and in the Comment. Background: Type 2 diabetes mellitus (T2DM) often occurs conjunctly with the menopausal transition in female patients. In addition, epicardial adipose tissue (EAT) has an unfavorable impact on the myocardium and coronary arteries under the influence of metabolic disorders. Purpose To investigate the impact of T2DM on EAT and myocardial microvascular function in postmenopausal women. Study Type: Retrospective. Population: One‐hundred sixty‐one postmenopausal women divided into three groups: newly diagnosed (≤5 years) T2DM (n = 56, 58.6 ± 7.7 years), long‐term (>5 years) T2DM (n = 57, 61.9 ± 7.9 years), and healthy controls (n = 48, 59.4 ± 7.4 years). Field Strength/Sequence: 3.0 T; balanced steady‐state free precession and inversion recovery prepared echo‐planar sequences. Assessment EAT volume was quantified by delineating the epicardial border and the visceral layer of pericardium on the short‐axis cine stacks. Perfusion parameters including upslope, maximum signal intensity (MaxSI) and time to maximum signal intensity (TTM) were derived from the first‐pass perfusion signal intensity–time curves. Statistical Tests: One‐way analysis of variance, Pearson's and Spearman correlation, and multivariable linear regression. Two‐sided P < 0.05 was considered statistically significant. Results: EAT volume was significantly increased in diabetic postmenopausal women compared to the controls (48.4 ± 13.4 mL/m2 [newly diagnosed T2DM] vs. 58.4 ± 17.3 mL/m2 [long‐term T2DM] vs. 35.8 ± 12.3 mL/m2 [controls]). Regarding perfusion parameters, upslope and MaxSI were significantly reduced (2.6 ± 1.0 [newly diagnosed T2DM] vs. 2.1 ± 0.8 [long‐term T2DM] vs. 3.6 ± 1.3 [controls]; and 21.4 ± 6.9 [newly diagnosed T2DM] vs. 18.7 ± 6.4 [long‐term T2DM] vs. 28.4 ± 8.6 [controls]), whereas TTM was significantly increased in the T2DM groups compared to the control group (23.6 ± 8.7 [newly diagnosed T2DM] vs. 27.1 ± 9.4 [long‐term T2DM] vs. 21.4 ± 6.0 [controls]). Multivariable analysis (adjusted coefficient of determination [R2] = 0.489) showed that EAT volume (β = −0.610) and menopausal age (β = 0.433) were independently correlated with decreased perfusion upslope. Data Conclusion: Diabetic postmenopausal women had significantly higher EAT volume and more impaired microcirculation compared to the controls. Increased EAT volume and earlier menopausal age were independently associated with microvascular dysfunction in these patients. Two kinds of fluorescent hyaluronan (HA) analogs, one serving as normal imaging agent and the other used as a biosensitive contrast agent, were developed for the investigation of HA uptake and degradation. Our approach of developing HA imaging agents depends on labeling HA with varying molar percentages of a near-infrared (NIR) dye. At low labeling ratios, the hyaluronan uptake can be directly imaged while at high labeling ratios, the fluorescent signal is quenched and signal generation occurs only after degradation. It is found that the conjugate containing 1%–2% NIR dye can be used as a normal optical imaging agent, while bioactivable imaging agents are formed at 6% to 17% dye loading. It was determined that the conjugation of dye to HA with different loading percentages does not impact HA biodegradation by hyaluronidase (Hyal). The feasibility of using these two NIR fluorescent hyaluronan analogs for HA investigation was evaluated in vivo with optical imaging. The data demonstrates that the 1% dye loaded fluorescent HA can be used to monitor the behavior of HA and its fragments, whereas bioactivatable HA imaging agent (17% dye in HA) is more suitable for detecting HA fragments.
