3. Contrasting responses of forest phenological guilds to complex floodplain change.

Understanding the seasonal dynamics of the forest herb layer and the factors that influence it is essential for predicting how the forest ecosystem, including mutualistic interactions, will respond to global change. However, to date, no research has investigated how understorey phenological guilds respond to major environmental threats and forest interventions.We showed the marked changes in the phenological structure of floodplain forest plant communities over half a century at a multi‐regional scale in Central Europe. Unlike previous studies focusing on individual species' responses to climate change, we examined shifts in species richness, frequency and abundance between phenological guilds within the plant community. We examined the effects of temporal variation in climate, hydrology, soil conditions and canopy structure, and assessed the effects of non‐intervention and intervention management on phenological guild responses.Our results show a significant loss of local species richness and a decline in species frequency in the forest understorey vegetation, associated with species exhibiting summer phenology. In contrast, an increase in abundance and richness of spring flowering species was observed, attributed to high nutrient loads and reduced flooding, respectively. The abundance of spring species increased only in stands where canopy cover increased over time, probably due to the suppression of summer flowering species, allowing spring flowering species to spread on open, bare ground. The community experienced a shift in maximum flowering plant richness from June to May. Despite management interventions such as clearcutting and tree planting, our data show similar temporal trajectories in intervention and non‐intervention forests, indicating that changes in phenological structure are largely independent of recent management activities.Synthesis. We propose that a complex interplay of environmental factors, rather than climate change alone, is shaping shifts towards earlier phenological guilds in floodplain forests. This study highlights the importance of considering a comprehensive view of phenological guild dynamics and the impact of environmental factors on forest plant community structure in the face of global change. Long noncoding RNAs (lncRNAs) are RNA molecules of 200 nucleotides or more in length that are not translated into proteins. Their expression is tissue-specific, with the vast majority involved in the regulation of cellular processes and functions. Many human diseases, including cancer, have been shown to be associated with deregulated lncRNAs, rendering them potential therapeutic targets and biomarkers for differential diagnosis. The expression of lncRNAs in the nervous system varies in different cell types, implicated in mechanisms of neurons and glia, with effects on the development and functioning of the brain. Reports have also shown a link between changes in lncRNA molecules and the etiopathogenesis of brain neoplasia, including glioblastoma multiforme (GBM). GBM is an aggressive variant of brain cancer with an unfavourable prognosis and a median survival of 14–16 months. It is considered a brain-specific disease with the highly invasive malignant cells spreading throughout the neural tissue, impeding the complete resection, and leading to post-surgery recurrences, which are the prime cause of mortality. The early diagnosis of GBM could improve the treatment and extend survival, with the lncRNA profiling of biological fluids promising the detection of neoplastic changes at their initial stages and more effective therapeutic interventions. This review presents a systematic overview of GBM-associated deregulation of lncRNAs with a focus on lncRNA fingerprints in patients' blood. [ABSTRACT FROM AUTHOR]

Dural arteriovenous fistula (DAVF) is a rare condition not constantly taken into consideration in the differential diagnosis of pulsatory tinnitus. For reliable detection of DAVF, digital subtraction angiography is necessary. Here we report one case of DAVF and comment on clinical aspects of that potentially very serious anomaly. Public goods and services administration represents an important tool supporting functioning of the state. Provision of public goods and services is closely related to the necessity to ensure relevant sources and setting the appropriate distribution channels, what in fact means, finding the suitable provider. Production of public goods can be provided by both sectors, private or public. If a public entity produces a public good, it is possible to talk about in-house production. In the opposite situation, when external private entity produces a public good, it is possible to talk about outhouse production. This contribution is focused on the issue of Public Private Partnership (PPP) projects as one of the outhouse production variants. The aim of the contribution is to compare the availability and complexity of information about the given projects for potential contractors in the Czech Republic and Slovak Republic (Slovakia). To be able to make this comparison, authors of the article conducted the qualitative research. Used method was document and text analysis. To identify the relevant literary sources, systematic literature review followed by the traditional literature review, were used. Authors expected that there will be solid database of information available to the potential conductors of PPP projects. Also the way of its presentation was expected to be thoughtful and complex. Provision of information and methodical support in specified area can influence the final willingness of both sides, contractors and contracting authorities, to enter this project focused on provision of public services, traditionally provided by the public sector, by the private sector. The contribution is finalized using the method of synthesis that enables the authors to describe the main differences in ways of PPP projects information presentation. It was found that currently provided information and methodical support in the area of PPP projects administered by the authorities on the central level in selected countries can be described as insufficient. Ochrana osobních údajů je tématem, které začalo být řešeno v souvislosti s náboženskými konflikty. Do popředí se dostalo po druhé světové válce, která zviditelnila možné negativní důsledky zneužití osobních údajů. V současnosti je ochrana osobních údajů zmiňována v souvislosti s implementací nového Nařízení Evropského parlamentu a Rady EU členskými státy EU. Cílem předkládaného článku je vyhodnotit postoje dále specifikovaného výzkumného vzorku podnikatelských subjektů, které vykonávají svou činnost v České republice, ke změnám stanoveným prostřednictvím nové právní úpravy ochrany osobních údajů. Příspěvek prezentuje zejména výstupy provedeného kvantitativního výzkumu založeného na sběru dat prostřednictvím dotazníkového šetření, identifikuje slabá místa procesu zavádění GDPR a navrhuje možná zlepšení vedoucí ke komfortnějšímu přechodu podnikatelských subjektů na současné legislativní podmínky v oblasti ochrany osobních údajů stanovené Evropským parlamentem a Radou EU.

The heterogeneity of the glioma subtype glioblastoma multiforme (GBM) challenges effective neuropathological treatment. The reliance on in vitro studies and xenografted animal models to simulate human GBM has proven ineffective. Currently, a dearth of knowledge exists regarding the applicability of cell line biomolecules to the realm of GBM pathogenesis. Our study's objectives were to address this preclinical issue and assess prominin-1, ICAM-1, PARTICLE and GAS5 as potential GBM diagnostic targets. The methodologies included haemoxylin and eosin staining, immunofluorescence, in situ hybridization and quantitative PCR. The findings identified that morphology correlates with malignancy in GBM patient pathology. Immunofluorescence confocal microscopy revealed prominin-1 in pseudo-palisades adjacent to necrotic foci in both animal and human GBM. Evidence is presented for an ICAM-1 association with degenerating vasculature. Significantly elevated nuclear PARTICLE expression from in situ hybridization and quantitative PCR reflected its role as a tumor activator. GAS5 identified within necrotic GBM validated this potential prognostic biomolecule with extended survival. Here we present evidence for the stem cell marker prominin-1 and the chemotherapeutic target ICAM-1 in a glioma animal model and GBM pathology sections from patients that elicited alternative responses to adjuvant chemotherapy. This foremost study introduces the long non-coding RNA PARTICLE into the context of human GBM pathogenesis while substantiating the role of GAS5 as a tumor suppressor. The validation of GBM biomarkers from cellular models contributes to the advancement towards superior detection, therapeutic responders and the ultimate attainment of promising prognoses for this currently incurable brain cancer. Ekonomické důsledky pandemie COVID-19 jsou, a samozřejmě v budoucnu budou, vážné. Tyto důsledky se velmi často projevují i na trhu práce a ve vzdělávání. Některé z nich nemusí být chápány pouze jako problémy, lze je chápat jako nové příležitosti: zlepšení pracovních podmínek, modernizace služeb zaměstnanosti, posílení systémů zaměstnávání a sociální ochrany, pracovní doba vyhovující rodinnému životu, podpora příležitostí k celoživotnímu vzdělávání, posílení digitálních dovedností, atd. Příspěvek se zaměřuje na různé přístupy a reakce na změny ve společnosti způsobené pandemií COVID-19. V současnosti jsou dostupné různé vědecké zdroje analyzující hrozby vyvolané pandemií a jejich dopady na různé oblasti lidské činnosti, včetně zaměstnávání a vzdělávání. Každá krize se však dá do určité míry pojmout také jako příležitost. Tento příspěvek se proto zabývá především příležitostmi, které současná krize přináší zejména v oblasti zaměstnanosti a vzdělávání. Práce je založena na vyhodnocení analýz (literární rešerše) zpracovávaných mezinárodními institucemi, jako je Mezinárodní úřad práce (ILO), a dalších výzkumných studií publikovaných Evropskou unií nebo OECD. The economic consequences of the COVID-19 pandemic are, and of course in the future will be, serious. These consequences are very often foundable also in labour market and education. Some of them are not only problems, but also new opportunities: improving of working conditions, modernising employment services and making them more flexible, strengthening employment and social protection systems, family-friendly working time, promoting lifelong learning opportunities, strengthening digital skills, etc. The paper focuses on different approaches and responses to changes in society caused by the COVID-19 pandemic. There are currently various scientific sources analyzing the threats initiated by the pandemic and their effects on various areas of human activity, including employment and education. However, every crisis can be taken also as an opportunity. This paper therefore primarily deals with the opportunities that the current crisis brings, especially in the field of employment and education. The paper is based on assessment of analyses (literary research) provided by international institutions such as the International Labour Office (ILO), and documents and other research studies prepared and published by the European Union or OECD.

Variation in genes involved in the absorption, distribution, metabolism, and excretion of drugs (ADME) can influence individual response to a therapeutic treatment. The study of ADME genetic diversity in human populations has led to evolutionary hypotheses of adaptation to distinct chemical environments. Population differentiation in measured drug metabolism phenotypes is, however, scarcely documented, often indirectly estimated via genotype-predicted phenotypes. We administered seven probe compounds devised to target six cytochrome P450 enzymes and the P-glycoprotein (P-gp) activity to assess phenotypic variation in four populations along a latitudinal transect spanning over Africa, the Middle East, and Europe (349 healthy Ethiopian, Omani, Greek, and Czech volunteers). We demonstrate significant population differentiation for all phenotypes except the one measuring CYP2D6 activity. Genome-wide association studies (GWAS) evidenced that the variability of phenotypes measuring CYP2B6, CYP2C9, CYP2C19, and CYP2D6 activity was associated with genetic variants linked to the corresponding encoding genes, and additional genes for the latter three. Instead, GWAS did not indicate any association between genetic diversity and the phenotypes measuring CYP1A2, CYP3A4, and P-gp activity. Genome scans of selection highlighted multiple candidate regions, a few of which included ADME genes, but none overlapped with the GWAS candidates. Our results suggest that different mechanisms have been shaping the evolution of these phenotypes, including phenotypic plasticity, and possibly some form of balancing selection. We discuss how these contrasting results highlight the diverse evolutionary trajectories of ADME genes and proteins, consistent with the wide spectrum of both endogenous and exogenous molecules that are their substrates. Introduction: While the role of physiotherapy as part of a comprehensive inpatient rehabilitation is indisputable, clear evidence concerning the effectiveness of different rehabilitation managements [interdisciplinary implementing the International Classification of Functioning, disability and health (ICF) vs. multidisciplinary model] and physiotherapy categories (neuroproprioceptive "facilitation, inhibition" vs. motor/skill acquisitions using technologies) are still lacking. In this study, four kinds of comprehensive inpatient rehabilitation with different management and content of physical therapy will be compared. Moreover, focus will be placed on the identification of novel biological molecules reflective of effective rehabilitation. Long non-coding RNAs (lncRNAs) are transcripts (>200 bps) of limited coding potential, which have recently been recognized as key factors in neuronal signaling pathways in ischemic stroke and as such, may provide a valuable readout of patient recovery and neuroprotection during therapeutic progression. Methods and analysis: Adults after the first ischemic stroke in an early sub-acute phase with motor disability will be randomly assigned to one of four groups and undergo a 3 weeks comprehensive inpatient rehabilitation of different types: interdisciplinary team work using ICF model as a guide; multidisciplinary teamwork implementing neuroproprioceptive "facilitation and inhibition" physiotherapy; multidisciplinary teamwork implementing technology-based physiotherapy; and standard multidisciplinary teamwork. Primary (the Goal Attainment Scale, the Patient-Reported Outcomes Measurement Information System, and the World Health Organization Disability Assessment Schedule) and secondary (motor, cognitive, psychological, speech and swallowing functions, functional independence) outcomes will bemeasured. A blood sample will be obtained upon consent (20 mls; representing pre-rehabilitation molecular) before and after the inpatient program. Primary outcomes will be followed up again 3 and 12 months after the end of the program. The overarching aim of this study is to determine the effectiveness of various rehabilitation managements and physiotherapeutic categories implemented by patients post ischemic stroke via analysis of primary, secondary and long non-coding RNA readouts. This clinical trial will offer an innovative approach not previously tested and will provide new complex analysis along with public assessable molecular biological evidence of various rehabilitation methodology for the alleviation of the effects of ischemic stroke. 24. Enhanced Extracellular Transfer of HLA-DQ Activates CD3 + Lymphocytes towards Compromised Treg Induction in Celiac Disease.

Celiac disease (CeD) manifests with autoimmune intestinal inflammation from gluten and genetic predisposition linked to human leukocyte antigen class-II (HLA-II) gene variants. Antigen-presenting cells facilitate gluten exposition through the interaction of their surface major histocompatibility complex (MHC) with the T cell receptor (TCR) on T lymphocytes. This fundamental mechanism of adaptive immunity has broadened upon recognition of extracellular exosomal MHC, raising awareness of an alternative means for antigen presentation. This study demonstrates that conditioned growth media (CGM) previously exposed to monocyte-derived dendritic cells from CeD significantly downregulates the CD3
+ lineage marker of control T cells. Such increased activation was reflected in their elevated IL-2 secretion. Exosome localization motif identification and quantification within HLA-DQA1 and HLA-DQB1 transcripts highlighted their significant prevalence within HLA-DQB1 alleles associated with CeD susceptibility. Flow cytometry revealed the strong correlation between HLA-DQ and the CD63 exosomal marker in T cells exposed to CGM from MoDCs sourced from CeD patients. This resulted in lower concentrations of CD25+ CD127− T cells, suggestive of their compromised induction to T-regulatory cells associated with CeD homeostasis. This foremost comparative study deciphered the genomic basis and extracellular exosomal effects of HLA transfer on T lymphocytes in the context of CeD, offering greater insight into this auto-immune disease. 30. Dopady elektronického evidence prodeje (EET) Úvod do malých a středních podniků v České republice

Hlavním důvodem zavedení elektronické evidence prodeje (EET) v České republice byl efektivnější výběr daní - zejména v oblasti daně z příjmů právnických osob, daně z příjmů fyzických osob a daně z přidané hodnoty. Cílem příspěvku je analyzovat a vyhodnotit dopady elektronických záznamů o uvedení do prodeje malých a středních podniků v České republice. Výzkum byl prováděn mezi malými a středními podniky, které mají s tímto novým legislativním nařízením něco společného. Autoři použili strukturovaný dotazník zaslaný 1 705 respondentům. Jako metoda hodnocení byla použita korelační analýza. Na základě výsledků výzkumu lze konstatovat, že více než polovina všech respondentů nesouhlasí se zaváděním elektronických záznamů o prodeji, a to zejména díky svým velkým administrativním nárokům. Výzkum také potvrdil, že největší část respondentů zaznamenala investice související se zavedením ERS do výše 5 000 Kč. Měsíčně zaznamenané náklady související s ERS byly nejčastěji v rozmezí 201 - 500 Kč. Kromě zjištění popsaných v článku byl také prokázán vliv elektronických záznamů o zavedení prodeje na trend růstu tržeb v hodnoceném segmentu vzhledem k hrubému domácímu produktu (HDP). Realizovaný výzkum bude sloužit jako výchozí bod pro další výzkum v popsané oblasti. The main reason for electronic records of sales (ERS) introduction in the Czech Republic was more effective tax collection - mainly in the area of corporate income tax, income tax on natural person and value added tax. Objective of this contribution is to analyze and evaluate impacts of electronic records of sales introduction to the small and medium-sized enterprises (SMEs) in the Czech Republic. The research was conducted among SMEs having something in common with this new legislative regulation. Authors used the structured questionnaire sent to 1 705 respondents. As the evaluation method was used the correlation analysis. On the basis of the research results can be stated that more than half of all respondents does not agree with the introduction of electronic records of sales, mainly due to its great administrative demands. The research also confirmed that the greatest part of respondents recorded investment related to ERS introduction in height up to 5 000 CZK. Monthly recorded costs related to ERS were most often in the range 201 - 500 CZK. In addition to findings described in the article was also proven effect of electronic records of sales introduction on sales growth trend in evaluated segment relative to gross domestic product (GDP). Conducted research will be used as a starting point for further research in described area.

Hlavním cílem příspěvku je identifikovat, analyzovat a vyhodnotit současný stav využití tohoto systému českými nekótovanými společnostmi s ohledem na budoucí možnosti rozšíření implementace IAS/IFRS na všechny účetní jednotky působící v České republice. Na základě zjištěných informací byly určeny hlavní výhody, nevýhody a bariéry aplikace IAS/IFRS a současný stav celkového povědomí nekótovaných společností o IAS/IFRS v České republice. S ohledem na získané informace autoři poukázali na nejčastěji se vyskytující problémy spojené s dalším rozšiřováním systému IAS / IFRS i mezi ostatními českými účetními jednotkami. Porovnání obdobně zaměřených výzkumů provedených výzkumnými pracovníky v různých zemích bylo provedeno tam, kde bylo možné zjistit, zda se trendy viditelné v České republice značně liší od trendů zaznamenaných v zahraničí či nikoliv. The main objective of the paper is to identify, analyze and evaluate current state of use of this system by the Czech non-listed companies with regard to future possibilities of extension of IAS/IFRS implementation to all accounting units operating in the Czech Republic. On the basis of the identified information were determined main advantages, disadvantages and barriers of the IAS/IFRS application and current state of the overall awareness of non-listed companies about IAS/IFRS in the Czech Republic. With regard to obtained information, the authors pointed out the most frequently met problems related to further expansion of the IAS/IFRS system also among other Czech accounting units. Comparison of similarly focused researches conducted by researchers in different countries were made where it was possible, to find out if the trends visible in the Czech Republic vary significantly from those recorded in foreign countries or not.

Controlling je disciplínou, která ovlivňuje efektivnost a konkurenceschopnost obchodních jednotek. Controlling nabízí nástroje, které manažerům pomáhají zajistit běžné obchodní aktivity (plánování, controlling, management). Cílem předkládaného příspěvku je demonstrovat vliv nástrojů controllingu na obchodní jednotkou standardně zajišťované procesy a navrhnout změny v controllingových aktivitách analyzované obchodní jednotky, které povedou ke zlepšení její pozice na trhu. Byla využita literární rešerše, analýza interních finančních dat sledované společnosti a polostrukturované rozhovory. K dosažení cíle bylo využito ERP systému specifikovaného v článku a nástrojů MS Excel a MS Project. Analyzovány byly controllingové a reportingové aktivity a jejich vhodnost pro danou společnost. Výsledkem analýzy je kompilace doporučení, která povedou ke zlepšení či alespoň udržení pozice sledované společnosti na trhu, ve kterém se pohybuje. Důraz byl kladen na využití informací, komunikačních technologií, finančních a lidských zdrojů. Navržená zlepšení (změny) může společnost využít v budoucnu. Controlling is taken as the discipline influencing effectiveness and competitiveness of business units. Controlling offers tools daily used by managers of the companies who ensure this way smooth running of all business activities, mainly planning, controlling and management. Objective of this contribution is to demonstrate the influence of controlling tools on processes standardly ensured by business units and propose available improvements in controlling activities of specific business unit leading to improvement of its market position. To reach the defined objective, literary research of relevant area, analysis of internal financial data of specific company and semi-structured interviews with responsible persons were used. During the analysis was used enterprise resource planning (ERP) system further specified in the contribution and other available tools suitable for conducted analysis such as MS Excel or MS Project. The main focus was given to cost controlling and reporting activities and suitability of their setting for analysed business unit. Results of conducted research were used to compile list of recommendations enabling the business unit keep or support its current business position. The emphasis was given on used information and communication technologies, financial and human resource planning and reporting. Selected proposals may be further used by analysed company and other companies providing their activities in the same or similar area.

Příspěvek reaguje na skutečnost, že nové průmyslové paradigma Průmysl 4.0 vyžaduje diskusi o sociálních a organizačních účincích tohoto paradigmatu a zaměřuje se na služby založené na znalostech. Cílem příspěvku je analyzovat a porovnat data zpracovaná Eurostatem, OECD a dalšími vědeckými pracemi z databáze Web of Science a nastavit (s využitím jejich srovnání) parametry pro typologii podniků znalostně náročných služeb. Autoři příspěvku nastavili 4 parametry: vysoká míra vysokoškolsky vzdělaných pracovních sil, začlenění do odvětví s vysokým podílem výdajů na výzkumnou činnost, existence pozitivního přístupu k inovaci a dostupnost základního procesu (jehož řízení je strategické), obsahově klíčových technologií, využívání systémů umožňujících řízení výroby, logistiky, marketingu a lze očekávat vysokou úroveň její bezpečnosti, resp. vysoké znalosti. Dalším cílem bylo pomocí údajů poskytnutých Českým statistickým úřadem identifikovat počet podniků, které patří k identifikovaným znalostně náročným službám a jeho vývoj v letech 2014 - 2017 se záměrem pokračovat ve výzkumu formou dotazníkového šetření. Tato analýza přinesla zjištění, že podniky, které splňují stanovené požadavky, se vyskytují především ve dvou sektorech národního hospodářství: Informační a komunikační aktivity s podílem 12,7% z celkového počtu výzkumných center v ČR a v sektoru: Profesionální, vědecké a technické činnosti s podílem 15,3%. Oba sektory vykazují nejvyšší míru malých podniků, ale ve sledovaném období se zvýšil počet malých, středních i velkých podniků. To může vést k navýšení disponibilního počtu podniků, které mohou být zařazeny do služeb založených na znalostech na 800. Tento příspěvek představuje částečný výsledek probíhajícího výzkumného projektu. Issue of the contribution responds to the fact that the new industrial paradigm Industry 4.0 requires the discussion about social and organizational effects of this paradigm and focuses on knowledge-intensive services. Objective of the contribution is to analyze and compare data processed by Eurostat, OECD and other scientific works from the Web of Science database and set (using their comparison) parameters for typology of enterprises of knowledge-intensive services. Authors of the contribution set 4 parameters: high rate of university-educated workforce, incorporation to sectors with high share of expenditures for research activities, existence of positive approach to innovation and availability of the core process, whose control is strategic, contents key technologies, uses systems enabling to manage production, logistics, marketing and it is possible to expect high level of its security, respectively high knowledge intensity. Further objective was to identify, using data provided by the Czech Statistical Office, the amount of enterprises belonging to identified knowledge-intensive services and its development in 2014 - 2017 with the intention to continue the research using the form of questionnaire survey. This analysis brought the finding that enterprises meeting set requirements occur mainly in two sectors of national economy: Information and communication activities with a share of 12,7 % of the whole amount of research centers in the Czech Republic and in the sector: Professional, scientific and technical activities with a share of 15,3 %. Both sectors show the highest rate of small businesses, but in monitored period increased the amount of both small and medium-sized and large enterprises. This may lead to increase of available amount of enterprises which can be included to the knowledge-intensive services to 800. This contribution represents partial result of the ongoing research project.

Předkládaný článek se zabývá hospodářskými aktivitami církevní organizace. Autorky se zaměřují zejména na účetní a daňová specifika vybrané organizace, na oblasti odlišné od podnikové praxe. Článek popisuje aktuální pozici církevních institucí v rámci národní ekonomiky a legislativní rámec limitující aktivity církevních právnických osob. V souvislosti se stanoveným cílem jsou vysvětleny účetní a daňové aspekty aktivit církevních organizací. Hlavním cílem článku je identifikace slabých míst ve dvou výše uvedených oblastech na příkladu zvolené církevní instituce (farnosti) a návrh změn vedoucích k efektivnějšímu zajištění činností sledované organizace v oblasti účetnictví a daní. K identifikaci slabých míst použily autorky metodu literární rešerše, kvalitativního výzkumu (polostrukturovaný rozhovor) a detailní analýzy interních dokumentů analyzované společnosti, navrhly způsob práce s informacemi v uvedených oblastech a doporučily řešení, které bude farností v budoucnu využito. Conducted article deals with the issue of ecclesiastical institutions. Authors are focused specifically on accounting and tax treatment of such institutions that differ in many areas from corporate practices. The article describes current position of ecclesiastical institutions in the national economy and legislative framework related to providing activities as the religious legal person. Then the accounting and tax aspects of these entities are explained in relation to the article objective. The main objective of conducted article is to identify the weak points in two mentioned areas using the example of selected ecclesiastical institution (parish) and recommend changes leading to more efficiency in activities currently provided by the organization in the accounting and tax area. Authors used methods of literary research, qualitative research (semistructured interviews) and deep analysis of internal financial documents of analyzed organization to be able to identify weak points, asses their threatment and propose final solution that will be further implemented and used by the parish.

Use of suitable IS, ERP, is one of the key elements supporting the optimal setting of processes in any selected area. Influence of accounting information system on accounting processes setting in the specified production company has become an object of interest analyzed by the article authors. Objective of this contribution is to compare the expected and actual changes of current processes and accounting procedures setting and propose the changes leading to their future higher efficiency and security. Selected type of conducted survey is qualitative research using the document analysis, participating observations and non-standardized conversation. Conducted research pointed out the most significant problems usually solved by the selected business unit in relation to their daily use of information system. Currently used information system was described at the level of selected modules relevant for the work of company accountants. The actual utilization rate of the used modules was analyzed and weak areas related to the use of substitution tools were identified. Weak areas were discussed with the accounting department representatives. Then the proposed changes and improvements were described and the related legislative framework was incorporated. Proposed changes are represented by the acquisition of additional modules dealing with document management, customer relationship management or production planning. It is also suggested to purchase improved barcode readers and improve the quality assurance system. The research results and proposed improvements were considered to be implementable and they are expected to be realized in future. There exist the reasonable requirement for cooperation of the accounting department with other, especially manufacturing, departments with respect to legislative framework in the area of information risk management. 39. Prenatal Exposure to Methamphetamine: Up-Regulation of Brain Receptor Genes.

Methamphetamine (METH) is a widespread illicit drug. If it is taken by pregnant women, it passes through the placenta and just as it affects the mother, it can impair the development of the offspring. The aim of our study was to identify candidates to investigate for changes in the gene expression in the specific regions of the brain associated with addiction to METH in rats. We examined the various areas of the central nervous system (striatum, hippocampus, prefrontal cortex) for signs of impairment in postnatal day 80 in experimental rats, whose mothers had been administered METH (5 mg/kg/day) during the entire gestation period. Changes in the gene expression at the mRNA level were determined by two techniques, microarray and real-time PCR. Results of two microarray trials were evaluated by LIMMA analysis. The first microarray trial detected either up-regulated or down-regulated expression of 2189 genes in the striatum; the second microarray trial detected either up-regulated or down-regulated expression of 1344 genes in the hippocampus of prenatally METH-exposed rats. We examined the expression of 10 genes using the real-time PCR technique. Differences in the gene expression were counted by the Mann–Whitney U -test. Significant changes were observed in the cocaine- and amphetamine-regulated transcript prepropeptide, tachykinin receptor 3, dopamine receptor D3 gene expression in the striatum regions, in the glucocorticoid nuclear receptor Nr3c1 gene expression in the prefrontal cortex and in the carboxylesterase 2 gene expression in the hippocampus of prenatally METH-exposed rats. The microarray technique also detected up-regulated expression of trace amine-associated receptor 7 h gene in the hippocampus of prenatally METH-exposed rats. We have identified susceptible genes; candidates for the study of an impairment related to methamphetamine addiction in the specific regions of the brain. [ABSTRACT FROM AUTHOR]

Ten to twenty years ago, in connection with increasing importance of information, was established new discipline, information management. There are many approaches to this discipline. One of them is based on emphasizing complex, system and systemantic approach to life cycle of requirement and includes both types of terms, technical and non-technical. It is Requirements engineering. This discipline includes elements of management, development or identification and formulation of requirements. Requirements engineering can be divided to two sub-disciplines, requirements development and requirements management. Formulation, obtaining, analysing, specifying, verification and also sharing requirements requires many specific activities ensured by huge number of people with different specialization, not only by IT engineers. All activities have to be coordinated to reach required outcome without delay or misunderstanding. Important person for requirements management is therefore the requirements analyst. Practical benefits of working with requirements are described in the presented article. The greatest problem that remains in both disciplines unsolved is mentioned at the end of the article. It is the tendency to underestimate theoretical knowledge and resistance to theory in general which leads to significant delays in the process of requirements engineering improvement.

43. Autoimmunity-Associated <bold>PTPN22</bold> Polymorphisms in Latent Autoimmune Diabetes of the Adult Differ from Those of Type 1 Diabetes Patients.

Background: A portion of adults with humoral immune changes have clinical diabetes that is initially not insulin-requiring (latent autoimmune diabetes of the adult, LADA). One of the genes strongly associated with autoimmune diabetes is PTPN22. We hypothesized that the manifestation and clinical features of LADA are linked to functional variants of PTPN22.Methods: We genotyped allelic frequencies of 1 protective and 3 risk-associated PTPN22 variants in 156 Czech LADA patients, 194 type 2 diabetes mellitus patients with LADA-like progression to insulinotherapy and 324 type 1 diabetes mellitus patients, and subsequently examined the associations of PTPN22 variants with the expression of autoantibodies and other clinical features of LADA.Results: We challenged the paradigm that stated that the PTPN22 c.1858T allele serves as a risk allele for LADA, although we confirmed its risk status in the geographically matched T1DM cohort. In contrast, the frequencies of other PTPN22 alleles (c.–1123C, c.788A and c.1970-852C) differed significantly from the healthy controls. We confirmed gender-related differences in the frequency of some PTPN22 polymorphisms (but not c.1858C>T) in LADA. The particular PTPN22 alleles and genotypes were associated with specific clinical features of the examined patients (autoantibodies, HbA1c and age at diagnosis of diabetes).Conclusions: The variability in PTPN22 haplotypes suggests that the genetic signature of LADA is independent and should not be considered a hybrid form of T1DM and T2DM. Further studies should elucidate the associations with clinical characteristics of the LADA patients and focus on the newly emerging types of diabetes with the disease onset in early to mid-adulthood. In this paper, Marie Černá traces the development of the women’s situation under communism in Czechoslovakia, the GDR, Hungary and Poland, by underlining that the “issue of women” was a key-point in the socialist ideology, thus leading to a progressive legislation on women’s rights. Yet, she points out that this does not affect the persistent prejudices of “smiling women and fighting men”, as well as the social and professional inequalities, especially in their institutional and structural manifestations. She concludes on the idea that the communist vision of emancipation was imposed and drummed in, following the official ideology in giving the impression that the issue of women had been resolved once and for all. The model of the "superwoman", in control of her professional work while at the same time coping with her functions in the family, was imposed during such a long time that it left no room for more general questions concerning the relationship between the sexes. This, according to the author, is one of the reasons why Czech society, including many women, keeps its distance from feminist movements even today. For most people, feminism still remains and will always remain tinged with extremism.

International audience; In this paper, Marie Černá traces the development of the women’s situation under communism in Czechoslovakia, the GDR, Hungary and Poland, by underlining that the “issue of women” was a key-point in the socialist ideology, thus leading to a progressive legislation on women’s rights. Yet, she points out that this does not affect the persistent prejudices of “smiling women and fighting men”, as well as the social and professional inequalities, especially in their institutional and structural manifestations. She concludes on the idea that the communist vision of emancipation was imposed and drummed in, following the official ideology in giving the impression that the issue of women had been resolved once and for all. The model of the "superwoman", in control of her professional work while at the same time coping with her functions in the family, was imposed during such a long time that it left no room for more general questions concerning the relationship between the sexes. This, according to the author, is one of the reasons why Czech society, including many women, keeps its distance from feminist movements even today. For most people, feminism still remains and will always remain tinged with extremism.

Background/Aims: Calcium-Sensing Receptor (CaSR) significantly affects calcium-phosphate metabolism in kidneys, and it is implicated in the pathogenesis of diabetes mellitus (DM) due to its expression in pancreatic β-cells. The role of CaSR as one of the players in pathogenesis of chronic kidney disease (CKD) has been speculated. Methods: 158 Type 2 diabetic patients divided into three groups according to occurrence and type of kidney complications, 66 nondiabetic patients CKD, and 93 healthy subjects were enrolled into the study to analyze the role of two CaSR polymorphisms (in the codon 990 and in the intron 4) in ethiopathogenesis of DM and CKD. The Type 2 diabetic groups consisted of 48 patients without any kidney abnormalities, 58 patients with diabetic nephropathy (DN), and 52 patients with nondiabetic renal disease (NDRD). The distribution of genotype and allele frequencies was studied using PCR with the TaqMan Discrimination Assay or followed by the Restriction Fragment Length Polymorphism method, respectively. Results: We have found that the intron 4 polymorphism is a risk factor for the development of DM and CKD, except DN, while the codon 990 does not show any disease association. Constantly changing market environment forces companies to make changes in their daily activities. This often leads to implementation of new tools that can optimize current business processes and to implement new ones if necessary. To decide on the use of new techniques, managers need relevant information. Information represents one of the most scarce and important asset of the company. Correct and complete information base helps managers to make the decisions that lead to future successful running of the business. One type of important decision making in the company is the decision making connected with information system selection. Suitably selected information system enables company ensuring error-free business processes and effective cooperation between the company departments. It enables to maintain current market position of the company or to improve it. This study describes the relationship between information, information management and selected information system. Main aim of the article is to assess the suitability of new information system implementation in the selected company. The article is prepared using literature review and useful information gathered by the expert interview with employee of the selected company. Risks can occur at any stage of the production process or the provision of mediation services. Information are then important basis for all decision-making processes that take place in companies. They represent one of the most scarce and important assets. Risks can be found at every step and the ability to recognize, describe and analyze them is (especially for managers and executives) very important. In order to be able to manage risks effectively, we use the help of risk analysis. It is divided into several parts, of which the first one, risk identification, represents a very difficult matter. Proper identification of risk is an essential component for the correct implementation of other parts of the risk analysis and the next steps in the process of risk management, and finally also for finding an appropriate method of risk management in conditions of a particular company. In practice, it is possible to meet many types of risks. They are usually not isolated. We can meet them as a complex of several types of risks. Among the risks that are currently often discussed (in connection with increasing importance of information and development of information technologies), are included information risks. This type of risk has been studied in relation to many areas of human activity. Accounting is a scientific discipline, whose faultless functioning in practice is significant for ensuring smooth running of the company. Better utilization and adaptation of existing information coming from managerial and financial accounting to the needs of risk management can be seen as a topic of future potential. Therefore, I believe that the analysis of including information risk management among other business activities being conducted within this area, is a topic which would be appropriate to continue to deal with in detail. This study describes the relationship between accounting and information management. The analysis is prepared more theoretically using deduction and literature review. 49. Late Rupture of Lumbar Artery as an Unusual Complication after Renal Biopsy - Case Report.

The most serious complication of renal biopsy is vascular damage with subsequent haemorrhage. To our knowledge, we present a first ever case of lumbar artery (LA) rupture accompanied by massive retroperitoneal bleeding, which developed after a significant amount of time following the biopsy itself. In a 63-year-old Caucasian female patient, a percutaneous left kidney biopsy was performed under continuous ultrasound guidance. On the fourteenth day after the procedure, she was examined for a sudden onset of left lumbar region pain. Computed tomography angiography showed a large retroperitoneal hematoma with active bleeding from the fourth left LA. Successful endovascular superselective embolization was performed immediately. The predisposing factor for the late haemorrhage could have been anticoagulation therapy, renal insufficiency and older age. Our case report highlights the need for caution, especially when performing kidney biopsy in a group of high-risk patients, particularly if they are indicated for subsequent anticoagulant therapy.
