Cyclopent-2-enone is one of the most important core structures in many bioactive prostaglandin derivatives and sesquiterpenes. Cyclopent-2-enone prostaglandin derivatives show anti-inflammatory and antiviral activity and inhibit NF-κB activation in human cell stimulated with tumor necrosis factor-α. Guaianes sesquiterpenes and pseudoguaianolides are widely obtained from many plants. These molecules exhibit strong anti-tumor, anti-inflammatory and anti diabetic activities. Here, we report the asymmetric synthesis of simple to complex various cyclopentanoid bioactive molecules from the readily available (R)-125 and (S)-134. Moreover, the first synthesis of (-)-teuclatriol was achieved with 26% overall yield from (R)-125. This method allows the facile access to obtain guaiane and pseudoguaianolide core structures without using protecting groups. Moreover, with this method we have successfully finished the total synthesis of (+)-orientalol-F and formal synthesis of englerin-A from building block (S)-134., Cyclopent-2-enon stellt eine der wichtigsten Kernstrukturen in vielen bioaktiven Prostaglandin Deriviaten und Sesquiterpenen dar. Cyclopent-2-enon Prostaglandin Derivate wirken sowohl entzündungshemmend also auch antiviral und unterdrücken eine durch Tumornekrosefaktor-α stimulierte NF-κB Aktivierung in menschlichen Zellen. Guajan-Sesquiterpene und Pseudoguaianolide können aus einer Vielfalt an Pflanzen isoliert werden. Diese Moleküle weisen eine starke entzündungshemmende, Antitumor- und Antidiabetes-Aktivität auf. Hier wird eine Methode beschrieben, welche einen vielseitigen Zugang zur asymmetrischen Synthese verschiedener einfacher bis hin zu komplexen bioaktiven Cyclopentanoide, ausgehend von den bereits verfügbaren Verbindungen (R)-125 und (S)-134,eröffnet. Die erste Synthese von (-)-Teuclatriol konnte mit einer Gesamtausbeute von 26% durchgeführt werden. Dieses Prinzip erlaubt den einfachen Zugang zu Guajan- und Pseudoguajanolid Grundstrukturen ohne den Einsatz von Schutzgruppen. Außerdem konnten unter zu Hilfenahme dieser Methode, ausgehend vom Baustein (S)-134, die Totalsynthese von (+)-Orientalol-F und die Formalsynthese von Englerin-A erfolgreich durchgeführt werden.
