Four new complexes [Ln(pfaa)3(H2O)2] n (Ln = Eu (1), Tb (2)) and [Ln2Cd2(pfaa)10 (phen)2(EtOH)2]·2MeCN (Ln = Eu (3), Tb (4), phen = 1,10-phenanthroline) based on pentaphorphenylacetic acid (Hpfaa) have been prepared and completely investigated. The structures of compounds 1 and 3 were studied by single crystal X-ray diffraction analysis: the isostructural complexes 1 and 2 represent 1D coordination polymer, while the isostructural complexes 3 and 4 are molecular compounds. Complexes 1–4 show strong lanthanide-centred luminescence, since the introduction of phenanthroline leads to a significant increase in the observed lifetime (τ obs) and photoluminescent quantum yield. New complexes of Eu3+ and Tb3+ with pentafluorophenylacetic acid have been studied. During synthesis under hydrothermal conditions, polymer structures are formed, while the introduction of phenanthroline and Cd2+ makes it possible to obtain molecular complexes. Five new complexes Ln(QC17)3(H2O)(Solv) (Ln = Y, Solv = H2O, Ln = Tb, Dy, Sm or Eu, Solv = EtOH) were synthesized with the acylpyrazolonato ligand QC17 bearing a long aliphatic C17H35 chain in the acyl moiety, and the crystal structure of Y(QC17)3(H2O)2 shows the three aliphatic chains from the coordinated ligands positioned in the same direction, affording plane layers built by Y(QC17)3(H2O)2 molecules connected through H-bonding interactions. The layers are stitched to each other like in “hook & loop” tapes. Luminescence of complexes was determined and the complex Tb(QC17)3(H2O)(EtOH) was immobilized on the surface of silica preprocessed using a C17H35CONH(CH2)3Si(OEt)3 reagent via hydrophobic interactions of long aliphatic chains. Luminescent properties and micromorphology of the obtained hybrid particles and hybrid films were investigated. Intensive green emission of the complex retains after grafting onto the silica surface. Inclusion of the complex on the surface of silica materials occurs as separate molecules, after the disruption of the H-bonding network present in the crystalline phase of the pure terbium sample. This work explores the generation of James Webb Space Telescope (JWSP) imagery via image-to-image translation from the available Hubble Space Telescope (HST) data. Comparative analysis encompasses the Pix2Pix, CycleGAN, TURBO, and DDPM-based Palette methodologies, assessing the criticality of image registration in astronomy. While the focus of this study is not on the scientific evaluation of model fairness, we note that the techniques employed may bear some limitations and the translated images could include elements that are not present in actual astronomical phenomena. To mitigate this, uncertainty estimation is integrated into our methodology, enhancing the translation's integrity and assisting astronomers in distinguishing between reliable predictions and those of questionable certainty. The evaluation was performed using metrics including MSE, SSIM, PSNR, LPIPS, and FID. The paper introduces a novel approach to quantifying uncertainty within image translation, leveraging the stochastic nature of DDPMs. This innovation not only bolsters our confidence in the translated images but also provides a valuable tool for future astronomical experiment planning. By offering predictive insights when JWST data are unavailable, our approach allows for informed preparatory strategies for making observations with the upcoming JWST, potentially optimizing its precious observational resources. We present a novel information-theoretic framework, termed as TURBO, designed to systematically analyse and generalise auto-encoding methods. We start by examining the principles of information bottleneck and bottleneck-based networks in the auto-encoding setting and identifying their inherent limitations, which become more prominent for data with multiple relevant, physics-related representations. The TURBO framework is then introduced, providing a comprehensive derivation of its core concept consisting of the maximisation of mutual information between various data representations expressed in two directions reflecting the information flows. We illustrate that numerous prevalent neural network models are encompassed within this framework. The paper underscores the insufficiency of the information bottleneck concept in elucidating all such models, thereby establishing TURBO as a preferable theoretical reference. Six new complexes of the ligand HQ cy (-4-(cyclohexanecarbonyl)-5-methyl-2-phenyl-2,4-dihydro-3H-pyrazol-3-one) and Ln 3+ ions with emission in the near-infrared (Nd 3+ ) or visible and near-infrared (Sm 3+ , Pr 3+ ) spectral regions were synthesized and characterized using various methods, including single crystal X-ray diffraction. The study demonstrated that both tris complexes [LnQ cy 3 (H 2 O)(EtOH)] and tetrakis-acids [H 3 O][LnQ cy 4 ] can be synthesized by varying the synthetic conditions. The photochemical properties of the complexes were investigated experimentally and theoretically using various molecular spectroscopy techniques and Judd-Ofelt theory. The objective was to quantitatively and qualitatively disclose the influence of complex stoichiometry on its luminescence properties. The study showed that the addition of an extra ligand molecule (in the tetrakis species) increased molar extinction by up to 2 times, affected the shape of photoluminescence spectra, especially of the Pr 3+ complex, and increased the quantum yield of the Sm 3+ complex by up to 2 times. The results obtained from this study provide insights into the luminescent properties of lanthanide coordination compounds, which are crucial for the design and development of novel photonic materials with tailored photophysical properties. more...

Complexes emitting in the blue spectral region are attractive materials for developing white-colored light sources. Here, we report the luminescence properties of novel coordination compounds based on the trivalent group 3, 13 metals, and the 1-phenyl-3-methyl-4-cyclohexylcarbonyl-pyrazol-5-onate (Q CH ) ligand. [M(Q CH ) 3 ] (M = Al, Ga, and In), [M(Q CH ) 3 (H 2 O)] (M = Sc, Gd, and Lu), [Lu(Q CH ) 3 (DMSO)], and [La(Q CH ) 3 (H 2 O)(EtOH)] complexes were synthesized and structurally characterized by a single-crystal X-ray diffraction study. It has been found that the luminescence quantum yields of the ligand increase by one order of magnitude upon metal coordination. A significant correspondence between the energies of the ligand's excited states and the luminescence quantum yields to the metal ion's atomic numbers was found using molecular spectroscopy techniques. The replacement of the central ion with the heavier one leads to a monotonic increase in singlet state energy, while the energy of the triplet state is similar for all the complexes. Time-resolved measurements allowed us to estimate the intersystem crossing (ISC) rate constants. It was shown that replacing the Al 3+ ion with the heavier diamagnetic Ga 3+ and In 3+ ions decreased the ISC rate, while the replacement with the paramagnetic Gd 3+ ion increased the ISC rate, which resulted in a remarkably bright and room-temperature phosphorescence of [Gd(Q CH ) 3 (H 2 O)]. more...

An analytical solution is obtained for the problem of two interacting, identical but separated spin 1/2 particles in a time-dependent external magnetic field, in a general case. The solution involves isolating the pseudo-qutrit subsystem from a two-qubit system. It is shown that the quantum dynamics of a pseudo-qutrit system with a magnetic dipole-dipole interaction can be described clearly and accurately in an adiabatic representation, using a time-dependent basis set. The transition probabilities between the energy levels for an adiabatically varying magnetic field, which follows the Landau-Majorana-Stuckelberg-Zener (LMSZ) model within a short time interval, are illustrated in the appropriate graphs. It is shown that for close energy levels and entangled states, the transition probabilities are not small and strongly depend on the time. These results provide insight into the degree of entanglement of two spins (qubits) over time. Furthermore, the results are applicable to more complex systems with a time-dependent Hamiltonian. more...

We investigate a system of two identical and distinguishable spins 1/2, with a direct magnetic dipole-dipole interaction, in an external magnetic field. Constraining the hyperfine tensor to exhibit axial symmetry generates the notable symmetry properties of the corresponding Hamiltonian model. In fact, we show that the reduction of the anisotropy induces the invariance of the Hamiltonian in the 3×3 subspace of the Hilbert space of the two spins in which S^2 invariably assumes its highest eigenvalue of 2. By means of appropriate mapping, it is then possible to choose initial density matrices of the two-spin system that evolve in such a way as to exactly simulate the time evolution of a pseudo-qutrit, in the sense that the the actual two-spin system nests the subdynamics of a qutrit regardless of the strength of the magnetic field. The occurrence of this dynamic similitude is investigated using two types of representation for the initial density matrix of the two spins. We show that the qutrit state emerges when the initial polarizations and probability vectors of the two spins are equal to each other. Patients infected by Mycoplasma genitalium are often treated empirically with the macrolide azithromycin. Macrolide resistance is becoming quite common; empirical treatment is compromised. Sequencing was initially used to detected azithromycin resistance-associated mutations. As this was laborious, qPCRs have been developed for their detection. In the present study, we describe a fast, sensitive, and specific qPCR assay that enables routine testing of M. genitalium and macrolide resistance-associated mutations in a single assay. M. genitalium positive clinical samples were used to compare (i) the commonly used MgPa assay for the detection of M. genitalium infections (MgPa qPCR), (ii) a combined 23S rRNA gene PCR/sequencing assay (Mg23S qPCR/Sequencing) to identify macrolide resistance-associated mutations, and (iii) our newly developed probe-based melt curve qPCR for simultaneous detection of M. genitalium and macrolide resistance-associated mutations (Macrolide-R/MG ELITe MGB Kit, Elitech Bothel USA in short Mg Macrolide R qPCR). Specificity of the qPCR was tested using urogenital samples that were tested positive for a range of other micro-organisms. M. genitalium was detected in 196/236 (83.1%) samples by the MgPa qPCR, versus 172/236 (72.9%) by the combined Mg23S qPCR/Sequencing, and 202/236 (85.6%) by the Mg Macrolide R qPCR. The Mg Macrolide R qPCR showed high concordance to the Mg23S qPCR/Sequencing assay (201 vs 202 could be genotyped, respectively) for the detection of the macrolide resistant mutations. None of the other urogenital pathogens were tested positive in the Mg Macrolide R qPCR, indicating specificity. The prototype demonstrated here is the first fully integrated sample-to-result diagnostic platform for performing nucleic acid amplification tests that requires no permanent instrument or manual sample processing. The multiplexable autonomous disposable nucleic acid amplification test (MAD NAAT) is based on two-dimensional paper networks, which enable sensitive chemical detection normally reserved for laboratories to be carried out anywhere by untrained users. All reagents are stored dry in the disposable test device and are rehydrated by stored buffer. The paper network is physically multiplexed to allow independent isothermal amplification of multiple targets; each amplification reaction is also chemically multiplexed with an internal amplification control. The total test time is less than one hour. The MAD NAAT prototype was used to characterize a set of human nasal swab specimens pre-screened for methicillin-resistant Staphylococcus aureus (MRSA) bacteria. With qPCR as the quantitative reference method, the lowest input copy number in the range where the MAD NAAT prototype consistently detected MRSA in these specimens was ∼5 × 10(3) genomic copies (∼600 genomic copies per biplexed amplification reaction). more...

We present a method of rapid isothermal amplification of DNA without initial heat denaturation of the template, and methods and probes for (a) real-time fluorescence detection and (b) lateral flow detection of amplicons. Isothermal strand displacement amplification (iSDA) can achieve >10(9)-fold amplification of the target sequence in <20 minutes at 49 °C, which makes it one of the fastest existing isothermal DNA amplification methods. iSDA initiates at sites where DNA base pairs spontaneously open or transiently convert into Hoogsteen pairs, i.e. "breathe", and proceeds to exponential amplification by repeated nicking, extension, and displacement of single strands. We demonstrate successful iSDA amplification and lateral flow detection of 10 copies of a Staphylococcus aureus gene, NO.-inducible l-lactate dehydrogenase (ldh1) (Richardson, Libby, and Fang, Science, 2008, 319, 1672-1676), in a clean sample and 50 copies in the presence of high concentrations of genomic DNA and mucins in <30 minutes. We also present a simple kinetic model of iSDA that incorporates competition between target and primer-dimer amplification. This is the first model that quantitates the effects of primer-dimer products in isothermal amplification reactions. Finally, we demonstrate the multiplexing capability of iSDA by the simultaneous amplification of the target gene and an engineered internal control sequence. Several 7-(hydroxy, amino, methylureido, and guanidino)alkynyl-substituted 8-aza-7-deaza- hypoxanthine analogues were investigated as potential universal nucleobases. 7-Aminobutynyl-8-aza-7-deazahypoxanthine was found to be the most promising quasi-universal nucleobase with improved hybridization and polymerase chain reaction (PCR) enhancing properties as compared to commonly used hypoxanthine (the nucleobase of inosine). It demonstrated improved ambiguity for pairing with A, T, and C bases and its base pairing properties can be summarized as follows: X:C∼X:A∼X:T > X:G. The improvement in PCR performance directly correlated with primer's Tm. Primers containing multiple 7-aminobutynyl-8-aza-7-deazahypoxanthines were successfully used without noticeable inhibition of Taq polymerase activity provided the modifications are positioned more than two bases away from the 3' end. more...

Here we describe a novel endonuclease IV (Endo IV) based assay utilizing a substrate that mimics the abasic lesions that normally occur in double-stranded DNA. The three component substrate is characterized by single-stranded DNA target, an oligonucleotide probe, separated from a helper oligonucleotide by a one base gap. The oligonucleotide probe contains a non-fluorescent quencher at the 5' end and fluorophore attached to the 3' end through a special rigid linker. Fluorescence of the oligonucleotide probe is efficiently quenched by the interaction of terminal dye and quencher when not hybridized. Upon hybridization of the oligonucleotide probe and helper probe to their complementary target, the phosphodiester linkage between the rigid linker and the 3' end of the probe is efficiently cleaved, generating a fluorescent signal. In this study, the use of the Endo IV assay as a post-PCR amplification detection system is demonstrated. Here we describe the properties of a novel class of oligonucleotide probes capable of sensitive hybridization-triggered fluorescence. These fluorogenic probes, known commercially as MGB Eclipse probes, are characterized by having a conjugated minor groove binder (MGB) ligand at the 5'-end and a fluorophore at the 3'-end. Additionally, they have an efficient quencher moiety at the 5'-end that is useful with a wide variety of fluorescent dyes. Fluorescence of the single-stranded MGB Eclipse probe is efficiently quenched by the interaction of the terminal dye and quencher groups when not hybridized. Upon hybridization to a complementary target, the MGB molecule folds into duplex and hyper-stabilizes it, allowing the use of shorter, more specific probe sequences. The 5'-MGB-quencher group also prevents nuclease digestion by Taq DNA polymerase during PCR. Because of the hybridization-triggered fluorescence and the excellent specificity imparted by the MGB, these 5'-MGB Eclipse probes have great versatility for real-time PCR applications. An open, randomized trial was performed to compare the efficacy of three Helicobacter pylori eradication regimens in children with peptic ulcer disease. A total of 106 children (5 - 15 years) were treated for 1 week with metronidazole, 30 - 40 mg/kg per day depending on age, amoxycillin, 750 mg/day, and one of three anti-secretory agents: proprietary omeprazole, 20 - 40 mg/day depending on age; generic omeprazole, 20 - 40 mg/day; or ranitidine, 150 mg twice daily. The H. pylori eradication rate was significantly higher in patients receiving proprietary omeprazole (88.9%) than in those receiving generic omeprazole (80.0%) or ranitidine (74.3%), and this was associated with a trend towards faster ulcer healing. It is concluded that triple therapy consisting of an anti-secretory agent and two antimicrobials produces effective eradication of H. pylori and ulcer healing in children with peptic ulcer disease, and that proprietary omeprazole is more effective than both ranitidine and the generic formulation used in this study. more...
